Abstract:It never rains in standard lab-confinements; thus we have limited understanding of animal reactions to water and wetness. To address this issue, we sprayed water on different body parts of rats and measured drying and fur temperature by thermal imaging while manipulating behavior, sensory cues and fur. Spraying water on rats resulted in fur changes (hair clumping, apex formation), grooming, shaking, and scratching. Anesthesia abolished behavioral responses, interfered with fur changes, and slowed drying. Spraying water on different body parts resulted in differential behavioral drying responses. Spraying the head resulted in grooming and shaking responses; water evaporated from the head twice as fast as water sprayed on the animal's back or belly. We observed no effect of whisker removal on post-water-spraying behavior. In contrast, local anesthesia of dorsal facial skin reduced post-water-spraying behavioral responses. Shaving of head fur drastically enhanced post-water-spraying behaviors, but reduced water loss during drying; indicating that fur promotes evaporation, acting in tandem with behavior to mediate drying. Excised wet fur patches dried and cooled faster than shaved excised wet skin. Water was sucked into distal hair tips, where it evaporated. We propose the wet-fur-heat-pump-hypothesis; fur might extract heat required for drying by cooling ambient air.

Abstract:Whiskers are highly developed tactile organs in mice. Here, we showed that mice with whisker trimming had a decreased anxiety behavior and activation of dorsomedial hypothalamus compared to control mice. Inhibition or damage of dorsomedial hypothalamus reversed the decrease of anxiety level induced by whisker trimming. These results expand the role of whiskers in regulating mouse behaviors to anxiety and suggest a novel function of dorsomedial hypothalamus. These findings indicate importance of normal sensory functions in modulating animal behavior.

Thus, a C-LTMR-spinoparabrachial pathway mediates wet dog shakes for rapid and effective removal of foreign particles from back hairy skin.

Abstract:Removing water from wet fur or feathers is important for thermoregulation in warm-blooded animals. The “wet dog shake” (WDS) behavior has been largely characterized in mammals but to a much lesser extent in birds. Although it is known that TRPM8 is the main molecular transducer of low temperature in mammals, it is not clear if wetness-induced shaking in furred and feathered animals is dependent on TRPM8. Here, we show that a novel TRPM8 agonist induces WDS in rodents and, importantly, in birds, similar to the shaking behavior evoked by water-spraying. Furthermore, the WDS onset depends on TRPM8, as we show in water-sprayed mice. Overall, our results provide multiple evidence for a TRPM8 dependence of WDS behaviors in all tested species. These suggest that a convergent evolution selected similar shaking behaviors to expel water from fur and feathers, with TRPM8 being involved in wetness sensing in both mammals and birds.

Abstract:Although the ability to sense skin wetness and humidity is critical for behavioral and autonomic adaptations, humans are not provided with specific skin receptors for sensing wetness. It has been proposed that we "learn" to perceive the wetness experienced when the skin is in contact with a wet surface or when sweat is produced through a multisensory integration of thermal and tactile inputs generated by the interaction between skin and moisture. However, the individual roles of thermal and tactile cues and how these are integrated peripherally and centrally by our nervous system is still poorly understood. Here we tested the hypothesis that the central integration of coldness and mechanosensation, as subserved by peripheral A-nerve afferents, might be the primary neural process underpinning human wetness sensitivity. During a quantitative sensory test, we found that individuals perceived warm-wet and neutral-wet stimuli as significantly less wet than cold-wet stimuli, although these were characterized by the same moisture content. Also, when cutaneous cold and tactile sensitivity was diminished by a selective reduction in the activity of A-nerve afferents, wetness perception was significantly reduced. Based on a concept of perceptual learning and Bayesian perceptual inference, we developed the first neurophysiological model of cutaneous wetness sensitivity centered on the multisensory integration of cold-sensitive and mechanosensitive skin afferents. Our results provide evidence for the existence of a specific information processing model that underpins the neural representation of a typical wet stimulus. These findings contribute to explaining how humans sense warm, neutral, and cold skin wetness.

Abstract:Self-grooming is an innate stereotyped behavior influenced by sense and emotion. It is considered an important characteristic in various disease models. However, the neural circuit mechanism underlying sensory-induced and emotion-driven self-grooming remains unclear. We found that the ventral zona incerta (Ziv) was activated during spontaneous self-grooming (SG), corn oil-induced sensory self-grooming (OG), and tail suspension-induced stress self-grooming (TG). Optogenetic excitation of Ziv parvalbumin (PV) neurons increased the duration of SG. Conversely, optogenetic inhibition of ZivPV neurons significantly reduced self-grooming in all three models. Furthermore, glutamatergic inputs from the primary sensory cortex activated the Ziv and contributed to OG. Activation of GABAergic inputs from the central amygdala to the Ziv increased SG, OG, and TG, potentially through local negative regulation of the Ziv. These findings suggest that the Ziv may play a crucial role in processing sensory and emotional information related to self-grooming, making it a potential target for regulating stereotyped behavior.
The biology of skin wetness perception and its implications in manual function and for reproducing complex somatosensory signals in neuroprosthetics

Abstract:Our perception of skin wetness is generated readily, yet humans have no known receptor (hygroreceptor) to signal this directly. It is easy to imagine the sensation of water running over our hands or the feel of rain on our skin. The synthetic sensation of wetness is thought to be produced from a combination of specific skin thermal and tactile inputs, registered through thermoreceptors and mechanoreceptors, respectively. The present review explores how thermal and tactile afference from the periphery can generate the percept of wetness centrally. We propose that the main signals include information about skin cooling, signaled primarily by thinly myelinated thermoreceptors, and rapid changes in touch, through fast-conducting, myelinated mechanoreceptors. Potential central sites for integration of these signals, and thus the perception of skin wetness, include the primary and secondary somatosensory cortices and the insula cortex. The interactions underlying these processes can also be modeled to aid in understanding and engineering the mechanisms. Furthermore, we discuss the role that sensing wetness could play in precision grip and the dexterous manipulation of objects. We expand on these lines of inquiry to the application of the knowledge in designing and creating skin sensory feedback in prosthetics. The addition of real-time, complex sensory signals would mark a significant advance in the use and incorporation of prosthetic body parts for amputees in everyday life.NEW & NOTEWORTHY Little is known about the underlying mechanisms that generate the perception of skin wetness. Humans have no specific hygroreceptor, and thus temperature and touch information combine to produce wetness sensations. The present review covers the potential mechanisms leading to the perception of wetness, both peripherally and centrally, along with their implications for manual function. These insights are relevant to inform the design of neuroengineering interfaces, such as sensory prostheses for amputees.

Abstract:Tail wagging is a conspicuous behaviour in domestic dogs ( Canis familiaris ). Despite how much meaning humans attribute to this display, its quantitative description and evolutionary history are rarely studied. We summarize what is known about the mechanism, ontogeny, function and evolution of this behaviour. We suggest two hypotheses to explain its increased occurrence and frequency in dogs compared to other canids. During the domestication process, enhanced rhythmic tail wagging behaviour could have (i) arisen as a by-product of selection for other traits, such as docility and tameness, or (ii) been directly selected by humans, due to our proclivity for rhythmic stimuli. We invite testing of these hypotheses through neurobiological and ethological experiments, which will shed light on one of the most readily observed yet understudied animal behaviours. Targeted tail wagging research can be a window into both canine ethology and the evolutionary history of characteristic human traits, such as our ability to perceive and produce rhythmic behaviours.

Abstract:Automated detection of complex animal behaviors remains a challenging problem in neuroscience, particularly for behaviors that consist of disparate sequential motions. Grooming is a prototypical stereotyped behavior and is often used as an endophenotype in psychiatric genetics. Here, we used mouse grooming behavior as an example and developed a general purpose neural network architecture capable of dynamic action detection at human observer-level performance and operating across dozens of mouse strains with high visual diversity. We provide insights into the amount of human annotated training data that are needed to achieve such performance. We surveyed grooming behavior in the open field in 2,457 mice across 62 strains, determined its heritable components, conducted GWAS to outline its genetic architecture, and performed PheWAS to link human psychiatric traits through shared underlying genetics. Our general machine learning solution that automatically classifies complex behaviors in large datasets will facilitate systematic studies of behavioral mechanisms.

Abstract:Grooming is a complex innate animal behavior that can serve as a marker of physiological state of rodents under stress. Here, we analyze the impact of various experimental factors, including genetic, pharmacological and physiological, on self-grooming behavior of laboratory mice and rats. Analyses of grooming behavior assess not only the number and duration of its bouts but also its microstructure, i.e. frequency, sequence, localization, and consistency. Rodent grooming and its microstructure can serve as a sensitive marker for changes in the brain, its response to stress, and predisposition to pathological conditions simulating such human mental disorders as obsessive-compulsive disorder, autism and depression. Studying rodent self-grooming microstructure can provide valuable information about the mechanisms of brain pathogenesis, having multiple translational implications for neuroscience research.

Epilepsy and the smell of fear

Abstract:Objective: In our canine scent detection research involving a specific volatile organic compound (VOC) associated with human epileptic seizure, we began to suspect involvement of the primitive neural networks associated with production of a previously undescribed human alarm pheromone as the origin of our seizure scent. We hypothesized that if we presented fear-scented sweat to our canine seizure scent detection team, and they identified the fear scent as their seizure scent, then that would suggest that they are identical compounds. Methods: Following consent and approval, sweat samples taken from volunteers associated with the Brooke Gordon Comprehensive Epilepsy Center at Denver Health were processed by the Canine Assistants (CA) service dog team that had been imprinted to recognize the unique seizure scent from our previous study. In part one, sweat samples were collected from subjects, who had no prior history of epilepsy or seizures, under two different testing environments: watching a scary movie (It) and a neutral/comedy movie (Airplane!). In part two, a larger follow-up study utilizing fear sweat, exercise sweat, epilepsy sweat, and other distractor scents were provided in a multiple choice paradigm to better understand the inter-rater reliability of the canine responses. Results: In part one, our canine seizure scent detection team identified fear-scented sweat samples as their seizure scent in 4 of 5 study participants. There was almost perfect agreement of seizure scent detection during fear scent trials between the canine seizure scent detectors with a kappa value of 0.814 (95% CI: 0.668-0.960). In part two, (utilizing eleven different subjects) our canine scent detection team identified samples of either fear or seizure sweat with a sensitivity of 82% and a specificity of 100% (no false positives) from among the multiple choices offered. Additionally, there was 92% agreement between the members of the canine scent detection team. Significance: While this hypothesis testing study is small and deserves replication, it confirms that the Canine Assistants seizure scent detection team consistently and accurately identified fear-scented sweat as their seizure scent, implying that the VOC, menthone, is common to both conditions. This further implies that human seizure propagation and fear network circuitry may share a common anatomy, and that menthone may not only be an early seizure biomarker, but a newly described human alarm pheromone.

Abstract:The striatum comprises multiple subdivisions and neural circuits that differentially control motor output. The islands of Calleja (IC) contain clusters of densely packed granule cells situated in the ventral striatum, predominantly in the olfactory tubercle (OT). Characterized by expression of the D3 dopamine receptor, the IC are evolutionally conserved, but have undefined functions. Here, we show that optogenetic activation of OT D3 neurons robustly initiates self-grooming in mice while suppressing other ongoing behaviors. Conversely, optogenetic inhibition of these neurons halts ongoing grooming, and genetic ablation reduces spontaneous grooming. Furthermore, OT D3 neurons show increased activity before and during grooming and influence local striatal output via synaptic connections with neighboring OT neurons (primarily spiny projection neurons), whose firing rates display grooming-related modulation. Our study uncovers a new role of the ventral striatum’s IC in regulating motor output and has important implications for the neural control of grooming.

Abstract:Sensory Adaptation (SA) is a prominent aspect of how neurons respond to sensory signals, ubiquitous across species and modalities. However, SA depends on the activation state of the brain and the extent to which SA is expressed in awake, behaving animals during active sensation remains unclear. Here, we addressed this question by training head-fixed mice to detect an object using their whiskers and recording neuronal activity from barrel cortex whilst simultaneously imaging the whiskers in 3D. We found that neuronal responses decreased during the course of whisker-object touch sequences and that this was due to two factors. First, a motor effect, whereby, during a sequence of touches, later touches were mechanically weaker than early ones. Second, a sensory encoding effect, whereby neuronal tuning to touch became progressively less sensitive during the course of a touch sequence. The sensory encoding effect was whisker-specific. These results show that SA does occur during active whisker sensing and suggest that SA is fundamental to sensation during natural behaviour.

Effect of friction on vibrotactile sensation of normal and dehydrated skin

Abstract:Background: Vibrotactile sensation mediated is highly dependent on surface mechanical and frictional properties. Dehydration of skin could change these properties. Objectives: To investigate the relationship between friction and vibrotactile sensation of normal and dehydrated skin. Methods: Vibrations were firstly measured during surface exploration using a biomimetic sensor. Piglet skin was used as human skin model to study frictional properties for both normal and dehydrated skin using an atomic force microscope on nanoscale and a pin-on-disk tribometer on macroscale. Effect of vibrational frequency on friction and vibrotactile perception was also observed on nano and macro scale for normal and dehydrated skin. Results: The result indicated that dehydrated skin was less sensitive than normal skin. The coefficient of friction of dehydrated skin is smaller than that of normal skin on both nano and macro scale. The coefficient of friction increases as increasing scanning frequencies. Conclusion: There is a positive correlation between coefficient of friction and vibrotactile sensation on nanoscale and macroscale.
Preliminary Study of Scent Rolling in Captive Wolves (Canis lupus L. 1758) in Response to Olfactory Enrichment

Abstract:Scent rolling, a behaviour observed in various large carnivores like wolves, entails the animal lowering its chin and neck towards a scent, followed by rubbing the head, neck, shoulders, and back into it. This behaviour is prevalent among wolves exposed to diverse scents, though its exact purpose remains uncertain. In this study, captive wolves at Osijek Zoo responded differently to odours during olfactory enrichment sessions. In the initial year of this study, the highest level of interest, evidenced by both the frequency of responses and scent-rolling behaviour, was noted when the wolves encountered odours such as curry and rosemary, along with deer/mouflon and rat faeces. While certain odours, such as llama faeces and deer/mouflon faeces, garnered longer durations of interest in the second year of study, others, like guinea pig faeces and oregano, elicited shorter responses. Female wolves demonstrated a higher level of engagement with scents compared with males, particularly through scent rolling behaviour, which was exclusively observed in females during the second year of this study. Interestingly, certain odours did not trigger scent rolling, suggesting selective preferences. On the other hand, sheep's wool induced the longest duration of scent rolling, and a lack of significant differences in behaviour was observed between morning and afternoon sessions. Despite the existence of multiple hypotheses put forward to explain the causation of scent rolling in wolves, it seems to be elicited by unfamiliar odours.

Abstract:Sensory integration theory predicts natural selection should favour adaptive responses of animals to multiple forms of information, yet empirical tests of this prediction are rare, particularly in free-living mammals. Studying indirect predator cues offers a salient opportunity to inquire about multimodal risk assessment and its potentially interactive effects on prey responses. Here we exposed California ground squirrels from two study sites (that differ in human and domestic dog activity) to acoustic and/or olfactory predator cues to reveal divergent patterns of signal dominance. Olfactory information most strongly predicted space use within the testing arena. That is, individuals, especially those at the human-impacted site, avoided coyote urine, a danger cue that may communicate the proximity of a coyote. By contrast, subjects allocated less time to risk-sensitive behaviours when exposed to acoustic cues. Specifically, although individuals were consistent in their behavioural responses across trials, ‘quiet coyotes’ (urine without calls) significantly increased the behavioural reactivity of prey, likely because coyotes rarely vocalize when hunting. More broadly, our findings highlight the need to consider the evolution of integrated fear responses and contribute to an emerging understanding of how animals integrate multiple forms of information to trade off between danger and safety cues in a changing world.
