Abstract:The high motility of synaptic weights raises the question of how the brain can retain its functionality in the face of constant synaptic remodeling. Here we used the whisker system of rats and mice to study the interplay between synaptic plasticity (motility) and the transmission of sensory signals downstream. Rats and mice probe their surroundings by rhythmically moving their whiskers back and forth. The azimuthal position of a whisker can be estimated from the activity of whisking neurons that respond selectively to a preferred phase along the whisking cycle. These preferred phases are widely distributed on the ring. However, simple models for the transmission of the whisking signal downstream predict a distribution of preferred phases that is an order of magnitude narrower than empirically observed. Here, we suggest that synaptic plasticity in the form of spike-timing-dependent plasticity (STDP) may provide a solution to this conundrum. This hypothesis is addressed in the framework of a modeling study that investigated the STDP dynamics in a population of synapses that propagates the whisking signal downstream. The findings showed that for a wide range of parameters, STDP dynamics do not relax to a fixed point. As a result, the preferred phases of downstream neurons drift in time at a non-uniform velocity which in turn, induces a non-uniform distribution of the preferred phases of the downstream population. This demonstrates how functionality, in terms of the distribution of preferred phases, can be retained not simply despite, but because of the constant synaptic motility. Our analysis leads to several key empirical predictions to test this hypothesis.

[Effect of Whisker Trimming on Behavior and Barrel Cortex of Rat].

Abstract:OBJECTIVE:To investigate the change of behavior, as well as the plasticity of somatosensory cortex after whisker trimming.METHODS:SD rats were divided into 4 groups. Group A is the normal control group; group B: bilateral vibrissotomy on the second postnatal day; group C: unilateral right vibrissotomy on the second postnatal day; group D: right unilateral whisker trimmed during 1-5 days after birth, and leave untreated after the 5th postnatal day. Their body weight, length of the left D2 whiskers was measured on the 30th postnatal day. At the same time, the changes of their behavior (including the slit-detection test, the home exploring behavior and thigmotaxis test) were also recorded on the 30th postnatal day. Cytochrome oxydase histochemistry (CO reaction)was applied to study the development and arrangement of barrel cortex.RESULTS:In the slit-detection test, control rats could find and get into the right slit very quickly. The rats in group B could get into the slit only if their noses touched the slit. The rats in group C couldn't identify the slit by right face, but if they turned their body and touched the slit with the left whiskers, they could get into the slit very quickly. The behavior of rats in group D was similar to that in group C. The time spent for finding out the right slit of the rats in group A, B, C was obviously longer than that of group A (P < 0.01, P < 0.05, P < 0.01). In the exploring behavior and thigmotaxis test, the time for left thigmotaxis, right thigmotaxis and total thigmotaxis of rats in group B was longer than that of control animals. The time for right thigmotaxis of group C was significantly shorter than that of group A (P < 0.05). Both the weight of the rats and the length of left D2 whiskers of rats in all the four groups had no significant difference. CO reaction showed that the barrels became smaller, the septum was not clear, the arrangement of the barrels was not tidy in the mice whose right whiskers were trimmed from 2-30 days after birth.CONCLUSION:Deafferentation doesn't change the body weight and length of the whiskers left. But the stimulation of whiskers is important for rodent especially in thigmotaxis and exploring behavior. Deafferentation can also induce the plastic change of barrel cortex.

Abstract:Rodents rely on their whiskers as vital sensory tools for tactile perception, enabling them to distinguish textures and shapes. Ensuring the reliability and constancy of tactile perception under varying stimulus conditions remains a fascinating and fundamental inquiry. This study explores the impact of stimulus configurations, including whisker movement velocity and object spatial proximity, on texture discrimination and stability in rats. To address this issue, we employed three distinct approaches for our investigation. Stimulus configurations notably affected tactile inputs, altering whisker vibration's kinetic and kinematic aspects with consistent effects across various textures. Through a texture discrimination task, rats exhibited consistent discrimination performance irrespective of changes in stimulus configuration. However, alterations in stimulus configuration significantly affected the rats' ability to maintain stability in texture perception. Additionally, we investigated the influence of stimulus configurations on cortical neuronal responses by manipulating them experimentally. Notably, cortical neurons demonstrated substantial and intricate changes in firing rates without compromising the ability to discriminate between textures. Nevertheless, these changes resulted in a reduction in texture neuronal response stability. Stimulating multiple whiskers led to improved neuronal texture discrimination and maintained coding stability. These findings emphasize the importance of considering numerous factors and their interactions when studying the impact of stimulus configuration on neuronal responses and behavior.

Abstract:Neural synapses are intercellular asymmetrical junctions that transmit biochemical and biophysical information between a neuron and a target cell. They are very tight, dynamic, and well organized by many synaptic adhesion molecules, signaling receptors, ion channels, and their associated cytoskeleton that bear forces. Mechanical forces have been an emerging factor in regulating axon guidance and growth, synapse formation and plasticity in physiological and pathological brain activity. Therefore, mechanical forces are undoubtedly exerted on those synaptic molecules and modulate their functions. Here we review current progress on how mechanical forces regulate receptor-ligand interactions, protein conformations, ion channels activation, and cytoskeleton dynamics and discuss how these regulations potentially affect synapse formation, stabilization, and plasticity.

Abstract:Rats generate sweeping whisker movements in order to explore their environments and identify objects. In somatosensory pathways, neuronal activity is modulated by the frequency of whisker vibration. However, the potential role of rhythmic neuronal activity in the cerebral processing of sensory signals and its mechanism remain unclear. Here, we showed that rhythmic vibrissal stimulation with short duration in anesthetized rats resulted in an increase or decrease in the amplitude of somatosensory-evoked potentials (SEPs) in the contralateral barrel cortex. The plastic change of the SEPs was frequency dependent and long lasting. The long-lasting enhancement of the vibrissa-to-cortex evoked response was side- but not barrel-specific. Local application of dl-2-amino-5-phosphonopentanoic acid into the barrel cortex revealed that this vibrissa-to-cortex long-term plasticity in adult rats was N-methyl-d-aspartate receptor-dependent. Most interestingly, whisker trimming through postnatal day (P)1-7 but not P29-35 impaired the long-term plasticity induced by 100 Hz vibrissal stimulation. The short period of rhythmic vibrissal stimulation did not induce long-lasting plasticity of field potentials in the thalamus. In conclusion, our results suggest that natural rhythmic whisker activity modifies sensory information processing in cerebral cortex, providing further insight into sensory perception.
A Critical Window for Experience-Dependent Plasticity at Whisker Sensory Relay Synapse in the Thalamus

Abstract:This study investigated the role of sensory experience in the refinement of whisker sensory relay synapses in the ventral posterior medial nucleus (VPm) of the murine thalamus. Sensory deprivation was done by whisker plucking, and synaptic connectivity was determined by whole-cell patch-clamp recording in acute slices. Sensory deprivation started at P12–P13, but not at P16, disrupted the elimination of VPm relay synapses. The majority of deprived neurons received multiple relay inputs, whereas the majority of nondeprived neurons received a single relay input. Sensory deprivation started a few days earlier at P10, however, had no effect on synapse elimination. The disruption of synapse elimination was associated with a delay in synapse maturation. The AMPA/NMDA ratio of EPSC was significantly smaller in deprived neurons. On the other hand, deprivation had no effect on the peak amplitude or decay time constant of the NMDA component, or the I–V relationship of the AMPA component, nor does it affect the paired-pulse ratio of EPSCs. The reduction in the AMPA/NMDA ratio was already evident within 24 h of whisker plucking, and the effect is associated with a reduction in the amplitude of quantal AMPA events. Thus, P12–P13 is a critical period for experience-dependent refinement at the whisker sensory relay synapse in the VPm.
An Amplitude Modulation/Demodulation Scheme for Whisker-Based Texture Perception

Abstract:Whisking rodents can discriminate finely textured objects using their vibrissae. The biomechanical and neural processes underlying such sensory tasks remain elusive. Here we combine the use of model micropatterned substrates and high-resolution videography of rats' whiskers during tactile exploration to study how texture information is mechanically encoded in the whisker motion. A biomechanical modeling of the whisker is developed, which yields quantitative predictions of the spectral and temporal characteristics of the observed whisker kinetics, for any given topography. These texture-induced whisker vibrations are then replayed via a multiwhisker stimulator while recording neuronal responses in the barrel field of the primary somatosensory cortex (S1bf). These results provide a comprehensive description of the transduction process at play during fine texture sensing in rats. They suggest that the sensory system operates through a vibratory amplitude modulation/demodulation scheme. Fine textural properties are encoded in the time-varying envelope of the whisker-resonant vibrations. This quantity is then recovered by neural demodulation, as it effectively drives the spiking-rate signal of a large fraction of S1 cortical neurons. This encoding/decoding scheme is shown to be robust against variations in exploratory conditions, such as the scanning speed or pad-to-substrate distance, thus allowing for reliable tactile discrimination in realistic conditions.

Abstract:Mouse whisker somatosensory cortex (wS1) is a major model system to study the experience-dependent plasticity of cortical neuron physiology, morphology, and sensory coding. However, the role of sensory experience in regulating neuronal cell type development and gene expression in wS1 remains poorly understood. We assembled and annotated a transcriptomic atlas of wS1 during postnatal development comprising 45 molecularly distinct neuronal types that can be grouped into eight excitatory and four inhibitory neuron subclasses. Using this atlas, we examined the influence of whisker experience from postnatal day (P) 12, the onset of active whisking, to P22, on the maturation of molecularly distinct cell types. During this developmental period, when whisker experience was normal, ~250 genes were regulated in a neuronal subclass-specific fashion. At the resolution of neuronal types, we found that only the composition of layer (L) 2/3 glutamatergic neuronal types, but not other neuronal types, changed substantially between P12 and P22. These compositional changes resemble those observed previously in the primary visual cortex (V1), and the temporal gene expression changes were also highly conserved between the two regions. In contrast to V1, however, cell type maturation in wS1 is not substantially dependent on sensory experience, as 10-day full-face whisker deprivation did not influence the transcriptomic identity and composition of L2/3 neuronal types. A one-day competitive whisker deprivation protocol also did not affect cell type identity but induced moderate changes in plasticity-related gene expression. Thus, developmental maturation of cell types is similar in V1 and wS1, but sensory deprivation minimally affects cell type development in wS1.

Functional Dynamics and Selectivity of Two Parallel Corticocortical Pathways from Motor Cortex to Layer 5 Circuits in Somatosensory Cortex

Abstract:In the rodent whisker system, active sensing and sensorimotor integration are mediated in part by the dynamic interactions between the motor cortex (M1) and somatosensory cortex (S1). However, understanding these dynamic interactions requires knowledge about the synapses and how specific neurons respond to their input. Here, we combined optogenetics, retrograde labeling, and electrophysiology to characterize the synaptic connections between M1 and layer 5 (L5) intratelencephalic (IT) and pyramidal tract (PT) neurons in S1 of mice (both sexes). We found that M1 synapses onto IT cells displayed modest short-term depression, whereas synapses onto PT neurons showed robust short-term facilitation. Despite M1 inputs to IT cells depressing, their slower kinetics resulted in summation and a response that increased during short trains. In contrast, summation was minimal in PT neurons due to the fast time course of their M1 responses. The functional consequences of this reduced summation, however, were outweighed by the strong facilitation at these M1 synapses, resulting in larger response amplitudes in PT neurons than IT cells during repetitive stimulation. To understand the impact of facilitating M1 inputs on PT output, we paired trains of inputs with single backpropagating action potentials, finding that repetitive M1 activation increased the probability of bursts in PT cells without impacting the time dependence of this coupling. Thus, there are two parallel but dynamically distinct systems of M1 synaptic excitation in L5 of S1, each defined by the short-term dynamics of its synapses, the class of postsynaptic neurons, and how the neurons respond to those inputs.

Abstract:Sensorimotor computation integrates bottom-up world state information with top-down knowledge and task goals to form action plans. In the rodent whisker system, a prime model of active sensing, evidence shows neuromodulatory neurotransmitters shape whisker control, affecting whisking frequency and amplitude. Since neuromodulatory neurotransmitters are mostly released from subcortical nuclei and have long-range projections that reach the rest of the central nervous system, mapping the circuits of top-down neuromodulatory control of sensorimotor nuclei will help to systematically address the mechanisms of active sensing. Therefore, we developed a neuroinformatic target discovery pipeline to mine the Allen Institute's Mouse Brain Connectivity Atlas. Using network connectivity analysis, we identified new putative connections along the whisker system and anatomically confirmed the existence of 42 previously unknown monosynaptic connections. Using this data, we updated the sensorimotor connectivity map of the mouse whisker system and developed the first cell-type-specific map of the network. The map includes 157 projections across 18 principal nuclei of the whisker system and neuromodulatory neurotransmitter-releasing. Performing a graph network analysis of this connectome, we identified cell-type specific hubs, sources, and sinks, provided anatomical evidence for monosynaptic inhibitory projections into all stages of the ascending pathway, and showed that neuromodulatory projections improve network-wide connectivity. These results argue that beyond the modulatory chemical contributions to information processing and transfer in the whisker system, the circuit connectivity features of the neuromodulatory networks position them as nodes of sensory and motor integration.

Long‐term Stability of Axonal Boutons in the Mouse Barrel Cortex

Abstract:ABSTRACT Many lines of evidence indicate that postsynaptic dendritic spines are plastic during development and largely stable in adulthood. It remains unclear to what degree presynaptic axonal terminals undergo changes in the developing and mature cortex. In this study, we examined the formation and elimination of fluorescently‐labeled axonal boutons in the living mouse barrel cortex with transcranial two‐photon microscopy. We found that the turnover of axonal boutons was significantly higher in 3‐week‐old young mice than in adult mice (older than 3 months). There was a slight but significant net loss of axonal boutons in mice from 1 to 2 months of age. In both young and adult barrel cortex, axonal boutons existed for at least 1 week were less likely to be eliminated than those recently‐formed boutons. In adulthood, 80% of axonal boutons persisted over 12 months and enriched sensory experience caused a slight but not significant increase in the turnover of axonal boutons over 2–4 weeks. Thus, similar to postsynaptic dendritic spines, presynaptic axonal boutons show remarkable stability after development ends. This long‐term stability of synaptic connections is likely important for reliable sensory processing in the mature somatosensory cortex. © 2015 Wiley Periodicals, Inc. A dendritic mechanism for balancing synaptic flexibility and stability

Abstract:Biological and artificial neural networks learn by modifying synaptic weights, but it is unclear how these systems can retain previous knowledge and also acquire new information. Here we show that cortical pyramidal neurons can solve this plasticity-versus-stability dilemma by differentially regulating synaptic plasticity at distinct dendritic compartments. Oblique dendrites of adult mouse layer 5b cortical pyramidal neurons selectively received monosynaptic thalamic input, integrated linearly, and— surprisingly—lacked synaptic potentiation. In contrast, basal dendrites, which do not receive thalamic input, exhibited conventional NMDA receptor-mediated supralinear integration and synaptic potentiation. Congruently, spiny synapses on oblique branches showed low structural plasticity in vivo. A selective decline in NMDAR activity and expression at synapses on oblique dendrites was controlled by an experience-dependent critical period. Our results demonstrate a new biological mechanism for how single neurons can safeguard a set of inputs from ongoing plasticity by altering synaptic properties at distinct dendritic domains.

Synaptic Elimination and Protection after Minimal Injury Depend on Cell Type and Their Prelesion Structural Dynamics in the Adult Cerebral Cortex

Abstract:The axonal and synaptic mechanisms underlying dysfunction and repair of the injured CNS are poorly understood. Unresolved issues include to what degree, when, and how the surviving neurons degenerate and the extent of synaptic remodeling both along the severed axon and in the nearby area. One of the main reasons is the lack of tools to study the complex asynchronous and dynamic features of individual lesioned axon responses in the intact brain. To address these issues, we combined two-photon microscopy and laser microsurgery to image the real-time reorganization of cortical circuitry at synaptic resolution for periods of up to 1 year in the brain of living mice. Injured cortical axons were eliminated proximally through a two-phase retraction process, which continued for at least 3 months postlesion and was independent of the presence of scar tissue. Remarkably, axons which later attempt to regenerate in both the mature and juvenile brain retracted less, raising the possibility that targeting retraction may improve the chances of axon regrowth after axotomy. Comparing prelesion and postlesion dynamics on the same axons over several days and weeks revealed that, although synapse formation rates were unaffected, boutons on injured axons were either rapidly and persistently lost, or extremely resistant, depending on cell-type and their prelesion structural dynamics. Our data suggest a lasting deficiency in synaptic output on surviving injured cortical axons and a surprising difference in the vulnerability of synaptic boutons after axotomy, which depend on cell-type and their recent history.
Whisker deprivation triggers a distinct form of cortical homeostatic plasticity that is impaired in the Fmr1 KO

Abstract:Mouse models of Fragile X Syndrome (FXS) have demonstrated impairments in excitatory and inhibitory sensory-evoked neuronal firing. Homeostatic plasticity, which encompasses a set of mechanisms to stabilize baseline activity levels, does not compensate for these changes in activity. Previous work has shown that impairments in homeostatic plasticity are observed in FXS, including deficits in synaptic scaling and intrinsic excitability. Here, we aimed to examine how homeostatic plasticity is altered in vivo in an Fmr1 KO mouse model following unilateral whisker deprivation (WD). We show that WD in the wild type leads to an increase in the proportion of L5/6 somatosensory neurons that are recruited, but this does not occur in the KO. In addition, we observed a change in the threshold of excitatory neurons at a later developmental stage in the KO. Compromised homeostatic plasticity in development could influence sensory processing and long-term cortical organization.

Abstract:Regulation of AMPA receptor (AMPAR) membrane trafficking is critical for synaptic plasticity, as well as for learning and memory. However, the mechanisms of AMPAR trafficking in vivo remain elusive. Using in vivo two-photon microscopy in the mouse somatosensory barrel cortex, we found that acute whisker stimulation led to a significant increase in the intensity of surface AMPAR GluA1 subunit (sGluA1) in both spines and dendritic shafts and a small increase in spine size relative to prestimulation values. Interestingly, the initial spine properties biased spine changes following whisker stimulation. Changes in spine sGluA1 intensity were positively correlated with changes in spine size and dendritic shaft sGluA1 intensity following whisker stimulation. The increase in spine sGluA1 intensity evoked by whisker stimulation was NMDA receptor dependent and long lasting, similar to major forms of synaptic plasticity in the brain. In this study we were able to observe experience-dependent AMPAR trafficking in real time and characterize, in vivo, a major form of synaptic plasticity in the brain.

Abstract:Neural network plasticity is fundamental for learning and memory. Its abnormal change underlies some neural diseases. Measurement of the plasticity of cortex can help understand the mechanism of plasticity, and provide a quantitative way to observe the neural process of natural aging and neurodegenerative diseases, which may lead to a new approach for evaluation of anti-aging drugs and new medical treatments for neurodegenerative diseases. In this study, a systematic method was established based on whisker pairing (WP) experiment to measure the network plasticity in the barrel cortex in rat. WP experiment is a classical experiment to study the effect of innocuous bias of the flow of sensory activity from the whiskers for certain periods in awake and behaving rats on the receptive field organization in S1 barrel cortex neurons. In the experiment, one pair of adjacent whiskers D2 and D3 remained intact while others were being trimmed throughout a certain period. After that, receptive fields of single cells in the contralateral barrel were analyzed by post-stimulus time histogram after certain days of WP and compared with the controls. In the control group, response magnitudes to surrounding whiskers D1 and D3 deflection were not significantly different. However, after WP, a bias occurred in response to paired surrounding whisker D3 relative to the opposite trimmed surrounding whisker D1. In this study, by comparing the bias degree in rats in different groups after WP, a quantitative method was established to compare cortical plasticity. Example of corical plasticity comparison between adolescent and mature rats was employed in this paper to illustrate our method. The key techniques of this method such as the identification of D2 barrels, supragranular (L2-3) and barrel layer (L4) in real-time were described in details. The feasibility of this approach was further verified by compendious report of results and our previous study regarding cortical plasticity comparison between adolescent and mature rats.

Abstract:Significance In recent years, increasing effort has been made to study sensorimotor integration within the context of the animal’s body and the environment. However, it is challenging to collect neurophysiological data under naturalistic conditions, and simulations have become an important part of neuroscience research. We developed a simulation framework, WHISKiT Physics , to model the dynamics of a complete sensorimotor system—the rodent vibrissal array—operating under ethologically relevant conditions. The tight link between sensing and motor control and the discrete somatotopy throughout the sensory pathway make the vibrissal array ideal to study sensorimotor circuits. WHISKiT Physics makes this widely used model system accessible to sensory research as well as to other disciplines including information theory, reinforcement learning, and artificial intelligence.

Abstract:The cellular and synaptic mechanisms driving cell-type-specific function during various cortical network activities and behaviors are poorly understood. Here, we targeted whole-cell recordings to two classes of inhibitory GABAergic neurons in layer 2/3 of the barrel cortex of awake head-restrained mice and correlated spontaneous membrane potential dynamics with cortical state and whisking behavior. Using optogenetic stimulation of single layer 2/3 excitatory neurons we measured unitary excitatory postsynaptic potentials (uEPSPs) across states. During active states, characterized by whisking and reduced low-frequency activity in the local field potential, parvalbumin-expressing neurons depolarized and, albeit in a small number of recordings, received uEPSPs with increased amplitude. In contrast, somatostatin-expressing neurons hyperpolarized and reduced firing rates during active states without consistent change in uEPSP amplitude. These results further our understanding of neocortical inhibitory neuron function in awake mice and are consistent with the hypothesis that distinct genetically-defined cell classes have different state-dependent patterns of activity.
