Abstract:We proposed a highly reproducible method that enables atomic resolution transmission electron microscopy investigations of the dynamics of nanoparticles in liquids. An ultrathin liquid layer was established as a result of formation of bubbles that was initiated and tuned by beam radiation. The migration, aggregation and rotation of palladium nanoparticles were observed.

Abstract:Objective: The mechanical properties of cells, which are the main characteristics determining their physical performance and physiological functions, have been actively studied in the fields of cytobiology and biomedical engineering and for the development of medicines. In this study, an indentation-vibration-based method is proposed to simultaneously measure the mechanical properties of cells in situ, including cellular mass (m), elasticity (k), and viscosity (c).Methods: The proposed measurement method is implemented based on the principle of forced vibration stimulated by simple harmonic force using an atomic force microscope (AFM) system integrated with a piezoelectric transducer as the substrate vibrator. The corresponding theoretical model containing the three mechanical properties is derived and used to perform simulations and calculations. Living and fixed human embryonic kidney 293 (HEK 293) cells were subjected to indentation and vibration to measure and compare their mechanical parameters and verify the proposed approach. Results: The results that the fixed sample cells are more viscous and elastic than the living sample cells and the measured mechanical properties of cell are consistent within, but not outside of the central region of the cell, are in accordance with the previous studies.Conclusion: This work provides an approach to simultaneous measurement of the multiple mechanical properties of single cells using an integrated AFM system based on the principle force vibration and thickness-corrected Hertz model. Significance: This study should contribute to progress in biomedical engineering, cytobiology, medicine, early diagnosis, specific therapy and cell-powered robots.

Abstract:An atomic force microscope (AFM) in liquid was developed. The specially designed AFM probe was composed of a tip attached to a cantilever, a cantilever-tip pedestal, a circular Plexiglas window, and a scanner which is made up of three perpendicular tube piezos. When the instrument works, the liquid level is between the upper and lower surface of the window, and a circular meniscus is established around the Plexiglas window, preventing the tip from being affected or destroyed by surface tension of the liquid. In this setup, the cantilever tip and the sample are completely immersed in fluid. Meanwhile, during the cantilever-tip scanning over the sample during measurement, this system has no restriction on the sample’s size and weight and has a high scanning speed, which can be up to 40lines∕s or 10s for a 400×400pixel image. Some experiments were carried out on the system to demonstrate the elegant performance. The results show that the AFM (i) enables the whole canitlever tip to dive into the fluid; (ii) the sample can also be totally immersed by the fluid; (iii) the size and weight of samples are not restricted, enabling the instrument to image comparatively large or heavy sample; (iv) it is capable of scanning over samples with high speed. All of these advantages indicate this AFM has a great prospect for practical applications.
The Research on Best Imaging Condition of the AFM on Cells

Abstract:Through comparing the atomic force microscope(AFM) images of the 7721 cells under different treatment conditions and observation conditions,the best imaging condition of SMMC-7721 cells in the air and buffer solution was gotten,at the same time the experimental method of imaging living cells was established.Cells were fixed by 0.5%,1%,1.5% glutaric dialdehyde,then washed by PBS three times,and observed in air circumstance with different scanning modes of AFM and scanning parameters to find the best figuring conditions in this circumstance.The living cells were cultured on the glass covered by polylysine,then put in the physiological solution and observed by the AFM directly.Compared the scanning time limit with changed the type of the circumstance solution,scanning tip and scanning frequency to get the optimized parameter.Images of the fixed cells were compared with the ones of living cells,and found very different.The living cells in the physiological solution were satiety and the edge was slick,but the edge and the surface of the fixed cells were rough and looked as fragmented.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.
A novel electrochemical atomic force microscope and its applications

Abstract:A novel electrochemical atomic force microscope (EC-AFM) operating in liquid or electrolyte is developed. The special EC-AFM probe contains a tip attached a cantilever, a laser source, a position sensitive detector and a Plexiglas glass which meanwhile can be used as a tip holder. The laser optical path in liquid also owns a special design. The cantilever and the sample are immersed in fluid completely when the system works. In addition, an open electrolytic cell has been designed which enables to change the sample's liquid environment and control the electrochemical process conveniently. Besides, when the system works, the probe tip scans over the sample surface to escape the confines of the specimen size and weight. Using the EC-AFM system, the experiment of copper plating on an iron plate in copper sulfate solution is carried out and real-time investigated. During the plating process, the AFM images of the sample surface are acquired directly in the copper sulfate with nano-scale resolution. The results show that the EC-AFM system can observe the solid-liquid interface of the electrochemical reactions and processes directly.

Abstract:Stem cells are cells characterized by their ability to differentiate into multiple types of cells. This unique property has the potential to lead to many promising solutions to human disease treatment. The stem cell differentiation process can be influenced by many factors. One of the important factors is the mechanical stimulation by modulating the extracellular matrix (ECM) elasticity. Stem cell stiffness increases as it differentiates to lower potency stem cells. This might be due to the reorganization of the cell cytoskeleton and could be confirmed by different imaging techniques. Atomic Force Microscopy (AFM) has been an ideal instrument in nanoscale imaging and mechanical property characterization because of the nature of its measurement. In this study, we applied the AFM with nanomanipulation capability to investigate mechanical properties of stem cells with and without differentiation in a time-lapse fashion. The experimental results showed that we were able to forecast the differentiation of mouse embryonic stem cells (mESC) around 24 hours after removal of Leukemia inhibitory factor (LIF). The stiffness of mESC after differentiation shows a one-fold increase over that of non-differentiated cells. Therefore the nanomechanical marker can be used as an early indicator for mESC differentiation.

Abstract:Atomic force microscope (AFM) has been widely used in the biological field owing to its high sensitivity (subnanonewton), high spatial resolution (nanometer), and adaptability to physiological environments. Nowadays, force volume (FV) and peakforce quantitative nanomechanical (QNM) are two distinct modes of AFM used in biomechanical research. However, numerous studies have revealed an extremely confusing phenomenon that FV mode has a significant difference with QNM in determining the mechanical properties of the same samples. In this article, for the case of human benign prostatic hyperplasia cells (BPH) and two cancerous prostate cells with different grades of malignancy (PC3 and DU145), the differences were compared between FV and QNM modes in detecting mechanical properties. The results show measured Young's modulus of the same cells in FV mode was much lower than that obtained by QNM mode. Combining experimental results with working principles of two modes, it is indicated that surface adhesion is highly suspected to be a critical factor resulting in the measurement difference between two modes. To further confirm this conjecture, various weight ratios of polydimethylsiloxane (PDMS) were assessed by two modes, respectively. The results show that the difference of Young's modulus measured by two modes increases with the surface adhesion of PDMS, confirming that adhesion is one of the significant elements that lead to the measurement difference between FV and QNM modes.

Abstract:A novel atomic force microscope (AFM) for large samples to be measured in liquid is developed. An innovative laser beam tracking system is proposed to eliminate the tracking and feedback errors. The open probe design of the AFM makes the operation in liquid convenient and easy. A standard 1200-lines/mm grating and a sheet of filter paper are imaged respectively in air and liquid to confirm its performance. The corrosion behavior of aluminum surface in 1-mol/L NaOH solution is further investigated by the AFM. Experimental results show that the system can realize wide range (20 x 20 (mu m)) scanning for large samples both in air and liquid, while keeping nanometer order resolution in liquid by eliminating the tracking and feedback error.

Abstract:The micro-volume analysis and specific detection are both essential requirements in the field of chemical sensing and biological testing. Membrane prefiltration can be used to improve the selectivity and accu-racy of detection. But for traditional porous membrane filtration, it is difficult to achieve the transmem-brane transport of micro-volume liquid due to the influence of lateral diffusion on membrane surface. Herein, we studied the focused transmembrane transport of micro-volume liquid in the porous polyethersulfone membrane with asymmetric (Janus) surface wettability. The hydrophilic layer (poly-dopamine) and hydrophobic layer (fluoropolymer) were deposited with controllable thickness by dip-coating and roller-assisted liquid printing. The micro-volume liquid focusing effect was verified by exper-iments such as visual wetting circle and fluorescent tracer. The liquid focusing effect of as-prepared Janus membrane was integrated with glucose test strip in the application of micro-volume liquid biosensing. Compared with conventional porous membrane, detected signal amplitude and response time were improved 7.5x and 2.7x, respectively. In summary, this research studied the dynamics of liquid transport through Janus membrane and provides a new strategy for microfluidic detection applications through balancing detection volume, time and selectivity by the advantage of micro-volume liquid focusing effect. Abstract:A novel atomic force microscope (AFM) operating in liquid is described in this article. The specially designed AFM probe involves a tip attached to a cantilever, a tip holder, and a circular Plexiglas window. When the probe dives into the fluid, a circular meniscus is established around the Plexiglas window, preventing the tip from being affected or destroyed by surface tension of the liquid. In this setup, the whole scanning probe and the sample can completely dive into fluid. Meanwhile, the probe tip scans over the sample surface when the instrument works. These advantages enable the instrument to scan comparatively large or heavy samples with a high speed. The highest scan rate is about 30 lines/s or 14 s for a 400 x 400-pixel, 3 x 3 Pin image. Using the new AFM, we carry out in-situ investigation of the formation processes of porous alumina during electrochemical anodic oxidation. A lead ring and an aluminum foil serve as cathode and anode, respectively. They are entirely immersed in the bath electrolyte, which is oxalic acid solution. During anodic oxidation, the AFM images of the sample surface are successively acquired without elevating the sample out of the solution. Experiments reveal that electrochemical reactions take place soon after the power supply is switched on, and with the progression of anodization, nanostructures of porous alumina gradually occur on the aluminum substrate, finally yielding ordered arrays of nanopores. As a typical example of applications, the results of this work show that the new AFM is an ideal and powerful tool for in-situ observation and study of materials or samples in aqueous solutions. Micropore-array Confining Assisted Young's Modulus Measurement of Suspended Cells Using Atomic Force Microscopy

Abstract:Mechanical properties of cells (e.g. Young’s modulus) reflect cells physiology or pathology, thus making them indexes to evaluate the state of cells. Atomic force microscopy (AFM) is a widely applied platform to characterize cell Young’s modulus. Commonly, AFM measurements were performed on adherent cells, for suspended cells are prone to slide during the measurements. However, for the similarity to in vivo state and preclusion of ‘bottom-effect’ artefact, to measure on suspended cells is important. Here we propose a new strategy to perform AFM characterizations on suspended cells. Assisted by a micropore array, we confined suspended cells to immobilize them, facilitating the force-displacement recording to extract accurate values of Young’s modulus. The measured Young’s modulus of Huh7 cells by the proposed method (1.34 kPa) was consistent with that obtained by the previously reported semi-trypsinization method (1.58 kPa). Furthermore, this new design is capable of measuring mechanical properties of rare cells from large-volume samples in liquid biopsy for practical clinical applications.

Abstract:Tumors have posed a serious threat to human life and health. Researchers can determine whether or not cells are cancerous, whether the cancer cells are invasive or metastatic, and what the effects of drugs are on cancer cells by the physical properties such as hardness, adhesion, and Young's modulus. The atomic force microscope (AFM) has emerged as a key important tool for biomechanics research on tumor cells due to its ability to image and collect force spectroscopy information of biological samples with nano-level spatial resolution and under near-physiological conditions. This article reviews the existing results of the study of cancer cells with AFM. The main foci are the operating principle of AFM and research advances in mechanical property measurement, ultra-microtopography, and molecular recognition of tumor cells, which allows us to outline what we do know it in a systematic way and to summarize and to discuss future directions.

Abstract:Atomic force microscopy (AFM) has been emerging as a multifunctional molecular tool in nanobiology and nanomedicine. This review summarizes the recent advances in AFM study of living mammalian cells at the single-molecule and single-cell levels. Besides nanoscale imaging of cell membrane structure, AFM-based force measurements on living cells are mainly discussed. These include the development and application of single-molecule force spectroscopy to investigate ligand–receptor binding strength and dissociation dynamics, and the characterization of cell mechanical properties in a physiological environment. Molecular manipulation of cells by AFM to change the cellular process is also described. Living-cell AFM study offers a new approach to understand the molecular mechanisms of cell function, disease development and drug effect, as well as to develop new strategies to achieve single-cell-based diagnosis.
Application of Magnetic AC Mode AFM on the in Situ Imaging of Living Cells

Abstract:The Magnetic AC mode AFM(MAC mode AFM),a novel type of tapping mode, was employed to in situ image living cells of 3T3 cell line.In this study, two types of cantilevers,i.e.0.95 N/m and 0.03 N/m,were used in both MAC mode and Contact mode.The membrane surface of the living cells and the fixed cells was studied too.The results suggest that MAC mode AFM allows us to acquire a high resolution imaging of living cells by using soft cantilevers.It does little harm to living cells during AFM scanning.But contact mode AFM is fit to in situ study of living stress fibers.The membrane surface of the living cells is distincted from that of the fixed cells.The true status can be learned more if the living cells were imaged in physiological solution.

Abstract:Apply AFM to study the best situation observed the fixed cells in the air and the living cells in buffer solution in real time.Useing AFM to observe the cells treated with different fixative and the same fixative but different concentration as well as the cells without treated with any fixative.Imagine with good quality can be acquired when cells were treated with 0.5%～1% glutaraldehyde for 15 min then washed with Hanks solution.Thus,the imagine obtained from the living cells in the Hanks solution is poorer than that of cells fixed in the air,but they can be kept in the living state.Utilize the ATM to acquire imagines with high resolution of living cells,the improvement on the preparation of the cells and the observation system is necessary.

Abstract:Nanoindentation based on atomic force microscopy (AFM) can measure the elasticity of biomaterials and cells with high spatial resolution and sensitivity, but relating the data to quantitative mechanical properties depends on information on the local contact, which is unclear in most cases. Here, we demonstrate nonlocal deformation sensing on biorelevant soft matters upon AFM indentation by using nitrogen-vacancy centers in nanodiamonds, providing data for studying both the elasticity and capillarity without requiring detailed knowledge about the local contact. Using fixed HeLa cells for demonstration, we show that the apparent elastic moduli of the cells would have been overestimated if the capillarity was not considered. In addition, we observe that both the elastic moduli and the surface tensions are reduced after depolymerization of the actin cytoskeleton in cells. This work demonstrates that the nanodiamond sensing of nonlocal deformation with nanometer precision is particularly suitable for studying mechanics of soft biorelevant materials.

Abstract:Cell mechanics plays a key role in determining physical performances and physiological functions of cells, as well as the early detection of diseases and development of biomedical engineering. In this study, we utilized a combination of atomic force microscopy (AFM) and finite element method (FEM) to compare the cellular elasticity (Young's modulus) and viscoelasticity (stress-relaxation time) of living and fixed endothelial cells (ECs) across varying loading rates. The results showed that both mechanical properties of normal ECs are more sensitive to loading speed compared with fixed ECs. The Young's modulus of normal endothelial cells (ECs) exhibits an increasing trend with the growing loading rate, whereas the Young's modulus of fixed ECs is almost not affected by the loading rate. Among various viscoelastic properties of cells under varying loading rates, the long-term relaxation time, especially at a loading rate of 5μm/s, showed the most significant difference between living and fixed cells. This work comprehensively evaluated the effectiveness of using different mechanical properties to distinguish cells with different physiological characteristic. This research would improve our knowledge of single-cell mechanical behaviors and provide new ideas for distinguishing various types of cells by AFM-based cellular elastic and viscoelastic properties with varying loading rates.

Abstract:To determine the lateral detachment force for individual bacterial cells, a quantitative method using the contact mode of an atomic force microscope (AFM) was developed in this study. Three key factors for the proposed method, i.e. scan size, scan rate and cantilever choice, were evaluated and optimized. The scan size of 40×40μm2 was optimal for capturing sufficient number of adhered cells in a microscopic field and provide adequate information for cell identification and detachment force measurement. The scan rate affected the measurement results significantly, and was optimized at 40μm/s considering both force measurement accuracy and experimental efficiency. The hardness of applied cantilevers also influenced force determination. The proposed protocol for cantilever selection is to use those with the lowest spring constant first and then step up to a harder cantilever until all cells are detached. The lateral detachment force of Escherichia coli cells on polished stainless steel and a glass-slide coated with poly-l-lysine were measured as 0.763±0.167 and 0.639±0.136nN, respectively. The results showed that the established method had good repeatability and sensitivity to various bacteria/substrata combinations. The detachment force quantified by AFM (0.639±0.136nN) was comparable to that measured by the centrifugation method (1.12nN).
