Abstract:Enantioselective trifluoromethylation of aromatic ketones promoted by the cinchona alkaloid-derived ammonium bromide and sodium hydride was described. A series of trifluoromethyl-substituted aryl alcohols could be obtained in up to 82% ee with 98% yield under mild conditions. A possible catalytic cycle was also presented.

Abstract:An atom and step economy cascade trifluoromethylation/cyclization of unactivated alkene with Langlois reagent as a CF3 source is described. A variety of polycyclic quinazolinones were successfully synthesized in 52–81% yields under transition metal‐ and oxidant‐free conditions. The Langlois reagent used in this strategy as a CF3 reagent possesses the advantages of bench‐stablity, cost‐effectivity and high‐efficiency. Additionally, gram‐scale reaction, broad substrate scope and good functional group tolerance demonstrated the synthetic usefulness of this protocol.

Abstract:The development of graphene oxide–based heterogeneous materials with an economical and environmentally–friendly manner has the potential to facilitate many important organic transformations but proves to have few relevant reported reactions. Herein, we explore the synergistic role of catalytic systems driven by graphene oxide and visible light that form nucleophilic alkoxyl radical intermediates, which enable an anti-Markovnikov addition exclusively to the terminal alkenes, and then the produced benzyl radicals are subsequently added with N–methylquinoxalones. This photoinduced cascade radical difunctionalization of olefins offers a concise and applicable protocol for constructing alkoxyl–substituted N–methylquinoxalones.

Abstract:A mild and simple protocol for the synthesis of alpha-trifluoromethylated ethanone oximes from aryl-substituted ethylenes, tert-butyl nitrite, and the Langlois reagent was developed under transition metal-free conditions. In this three-component reaction, tert-butyl nitrite was employed not only as the oxidant but also as the oxime source. A range of alpha-trifluoromethylated ethanone oximes were obtained, indicating the wide substrate scope of the reaction.

Abstract:An iron‐catalyzed direct C3‐H radical trifluoromethylthiolation of quinoxalin‐2(1H)‐ones has been developed. This protocol use earth‐abundant FeSO4·7H2O as the catalyst, and AgSCF3 as the trifluoromethylthiolating reagent to provide various SCF3‐containing quinoxalin‐2(1H)‐one derivatives in 26‐88% yields. Preliminary mechanistic investigations indicate that the reaction proceeds through a radical pathway.

Abstract:Traditional alkyne carbotrifluoromethylation protocols lead to the production of trifluoromethylated alkenes, while new reaction modes remain to be explored. Herein, a visible light induced reductive alkyltrifluoromethylation of ynones with the Langlois reagent is developed, providing a direct and diastereoselective access to highly functionalized trifluoromethylated cyclopentanones at room temperature. Mechanistic studies indicate a radical cascade involving the regioselective trifluoromethylation of C-C triple bonds, 1,5-HAT, 5-endo-trig closure, single electron transfer reduction, and protonation.

Abstract:Herein, we describe a novel electrochemistry facilitated one-pot multicomponent cascade annulation and bi-functionalization reaction that enables the efficient synthesis of highly functionalized 3,4-dihydroquinazolinones from readily available starting materials, realizing indirect C(sp3)-H trifluoromethylation of aromatic ketones. This methodology achieves two goals in one simple reaction, the direct synthesis of quinazolinones and the concurrent introduction of functional groups. This transformation has good compatibility with common functional groups and the scaleup electrolysis can be easily performed.

Abstract:Direct vicinal difunctionalization of alkynes through a catalyst-free four-component reaction of Togni's reagent, alkynes, sulfur dioxide, and hydrazines at room temperature is developed, which generates (E)-3,3,3-trifluoroprop-1-ene-1-sulfonohydrazides in good yields. This one-pot transformation works efficiently under extremely mild conditions without any catalysts and additives. The reaction process proceeds through trifluoromethylation of alkynes and aminosulfonylation via insertion of sulfur dioxide with excellent stereoselectivity and regioselectivity. Additionally, this reaction shows a broad substrate scope with good functional group tolerance. A plausible mechanism is proposed, supported by DFT calculations.

This method provides a variety of quinoxalines in good to high yields under a simple and mild reaction conditions with molecular O2 as a terminal oxidant. Preliminary mechanistic studies show that a novel reaction mechanism is involved in the current reaction, which is different from the previous reported dicarbonyl compound mechanism via the oxidation of the alkyne.

Abstract:A general electrohemical strategy for the combined trifluoromethylation/C(sp2)-H functionalization using Langlois' reagent as the CF3 source under oxidant-free conditions was developed. Using Mn salts as the redox mediator, this method provides an efficient and sustainable means to access a variety of functionalized heterocycles bearing a CF3 moiety. Detailed mechanistic studies are consistent with the formation of CF3-bound high oxidation state Mn species, suggesting a transition-metal-mediated CF3 transfer mechanism for this trifluoromethylation/C(sp2)-H functionalization process.
