Abstract:Glutathione (GSH) as a biothiol is an essential peptide related to various diseases. Although multiple strategies for biothiols detection have been developed, there is increasing demand for sensors that can differentiate GSH from cysteine (Cys) and homocysteine (Hcy), owing to the similar structures and thiol groups in these amino acids. Herein, we report a novel Eu3+/LAPONITE (Lap)-based organic/inorganic hybrid material for selective detection of GSH via an "off-on" process. The fluorescence of Eu(DPA)3@Lap-Tris can be quenched by Cu2+ through photoinduced electron transfer (PET). The addition of GSH into the Eu(DPA)3@Lap-Tris/Cu2+ system induces the removal of Cu2+ from Eu(DPA)3@Lap-Tris and blocks PET, resulting in the recovery of fluorescence. This proposed assay demonstrates higher selectivity toward GSH than Cys and Hcy, and showed a detection limit of 162 nM within a linear range of 0.5-30 μM. Unlike other GSH selective sensors, this platform could be formed into a hydrogel while its sensitivity was maintained. The sensitive response to GSH in serum samples makes this platform an efficient tool for biological applications because of its ease of preparation, high selectivity, good biocompatibility, and low toxicity.

Abstract:Glutathione (GSH) exhibits antioxidant and integrated detoxification effects, providing protection to the liver. Nevertheless, in cases of liver damage or cancerization, the GSH content experiences significant fluctuations. Consequently, GSH can serve as an effective endogenous molecule for aiding the diagnosis and pathological researches of liver diseases. In this study, a GSH-responsive conductive hydrogel was prepared for the quantitative detection of GSH. Through measuring the high-frequency impedance change of the hydrogel during the reaction, the detection of GSH in a wide range (0.1-6 mM) is realized, and the sensor has high sensitivity and good anti-interference performance.

Abstract:In this paper, we report a fluorescence sensor for detecting glutathione (GSH), FITC which served as fluorescent donors, and gold nanoparticles (AuNPs, quencher)served as fluorescent acceptors. Energy transfer form FITC to AuNPs because of the electrostatic adsorption, resulting in the quenching of FITC fluorescence. After the addition of glutathione, FITC detach from the surface of gold nanoparticles and the fluorescence will restore because of the strong force of Au-S bond. Based on this principle, we construct a new method to detect GSH and give fluorescence tests in different experimental. The results revealed that there is a good linear in the range of 0.9 ~ 256mg / L in detecting the concentration of GSH, and the correlation coefficient is 0.992. We obtain a satisfying result and it can be used to detect the high concentrations of glutathione content.

Abstract:We report a novel design, based on a combination of lanthanide-doped upconversion nanoparticles and manganese dioxide nanosheets, for rapid, selective detection of glutathione in aqueous solutions and living cells. In this approach, manganese dioxide (MnO(2)) nanosheets formed on the surface of nanoparticles serve as an efficient quencher for upconverted luminescence. The luminescence can be turned on by introducing glutathione that reduces MnO(2) into Mn(2+). The ability to monitor the glutathione concentration intracellularly may enable rational design of a convenient platform for targeted drug and gene delivery.
Emission enhanced fluorometric biosensor by functionalized carbon polymer dots for glutathione detection in human real samples and molecular logic gate operation

Abstract:Glutathione (GSH), an active peptide, plays pivotal roles in many physiological processes and detection of GSH inside of human body is of great importance for the playing of its biological effects. Here silver-phosphorus co-doped carbonized polymer dots () were prepared via solvothermal treatment of citric acid and phytic acid in the presence of Ag + for GSH determination. The physicochemical and optical performance of the were characterized by X-ray photoelectron spectroscopy (XPS), Fourier-transform infrared (FT-IR), X-ray powder diffraction (XRD), Raman spectroscopy, transmission electron microscopy (TEM), fluorescence spectroscopy and ultraviolet–visible (UV–Vis) spectroscopy analyses. The prepared have outstanding water solubility with high monodispersity (7.81 ± 0.31 nm) and exhibited excellent optical properties with excitation-dependent emission, high photostability, pH, and ionic strength tolerance. An optimized excitation at 358 nm, the showed strong photoluminescent (PL) emission at 456 nm with a PL quantum yield (QYs) of 15.6%. Furthermore, the were used as a PL sensing platform for detection GSH in a linear range of 0–200 μM with a low limit of detection at 0.68 μM. In addition, the proposed system can construct molecular logic gates with GSH and Fe 3+ ions as the chemical inputs and PL emissions as the output. And the were successfully used for GSH determination in real samples resulting in high sensitivity and satisfactory recoveries (92.81––107.45%). More importantly, the showed low cytotoxicity at 500 μg/mL and superior cell imaging capability in HeLa cells, which offer a new path for detection and categorization of GSH in biomedical applications.

Abstract:This report outlines a highly sensitive and facile electrochemiluminescence (ECL) sensing platform based on a novel high-quantum-yield Au-nanocluster (AuNC) probe for glutathione (GSH) detection. Owing to the prominent quenching effect of GSH on the ECL of the AuNCs, the proposed ECL nanosensor showed a wide response to GSH in the ranges of 1.0 × 10-9-1.0 × 10-5M and 1.0 × 10-5-1.0 × 10-1M and a low detection limit of 3.2 × 10-10M. In addition, the proposed system exhibited good selectivity for GSH in the presence of other chemical/biological interferences. Moreover, since no further functionalization of AuNC-based sensor interface was necessary, together with the stability, high sensitivity and selectivity of the proposed nanosensor, this convenient approach was able to successfully detect GSH in both of human urine samples and blood samples with excellent recoveries, which indicated its promising application under physiological conditions. Of significant importance is that this study not only helps in gaining a better understanding of the applicability of the ECL properties of AuNCs, but also provides a new avenue for the design and development of ECL sensors based on the novel high-quantum-yield AuNC-based probe and other functional-metal-based NC probes.
[Precise and Rapid Detection of Glutathione by Using Novel Fluorescent Ag Nanoclusters]

Abstract:Glutathione (GSH) is an important three-peptide molecule, which has the functions of antioxidation and detoxification, and plays a crucial role in the fields of biology, medicine and food science. It is involved in many important biochemical reactions in cells and body fluid, and the changes of GSH content reflect the specific health problems of human body. Current methods of GSH detection are always complicated, time-consuming and expensive instrument depended, such as surface enhanced Raman spectroscopy (SERS), electrochemical analysis, high performance liquid chromatography (HPLC) and so on. The probe’s photochemical properties can be modified by the reaction between GSH and nanoclusters, which will result in the changes of fluorescence intensity and wavelength. In this paper, a new method to realize precise and rapid GSH detection is developed by using silver na-noclusters as a fluorescent probe, and simultaneously measures the probe’s fluorescence intensity and wavelength. The synthesis of the fluorescence probe reported in this paper possesses the advantages of steps-simple and pollution free, and the GSH detection method has faster response, more accurate measurement and smaller relative error over the traditional methods. The good specificity of GSH detection among other molecules with the similar structure is further proved in control group experiments by comparing the differences of their fluorescence intensities and wavelength. The measurement accuracy is fully assured due to the insensitivity of the probe to a variety of salt ions and amino acids. This technique can be further employed in the intracellular detection and imaging of GSH.

Abstract:Glutathione (GSH), as a key regulator for the redox balance of biological processes, was detected with a metal-organic framework (MOF) (UiO-67-sbdc) fluorescence sensor. The fluorescence of UiO-67-sbdc could be effectively enhanced by GSH via an electronic effect between GSH and the ligand of MOF, thus achieving the recognition of GSH. It is the first attempt to design a MOF-based fluorescent sensor to distinguish GSH from cysteine (Cys). This sensor exhibited short response time (< 120 s), suitable linear range, and high selectivity towards GSH. Further research conducted in serum also showed a quantitative and selective detection of GSH. Thus, the reported UiO-67-sbdc in this work has its application prospect in clinically judging antioxidant capacity of the body.

Abstract:Glutathione (GSH) plays a crucial role in various physiological processes. Therefore, efficient detection of GSH is of great importance. In this study, we present a new and highly efficient ratiometric fluorescent biosensor through one-pot synthesis of coupling the self-assembly of polysaccharide (alginate dialdehyde, ADA)-dipeptide (diphenylalanine, FF) and in situ reduction of gold nanoparticles (Au NPs) simultaneously encapsulating a dye (5-aminofluorescein, FI-NH2) inside to form ADA-Au-FF-dye nanospheres. Because the fluorescence of the dye can be quenched by Au NPs and linearly recovered following the introduction of different GSH concentrations owing to nanometal surface energy transfer on and off, this new ADA-Au-FF-dye hybrid nanosphere-based sensor allows label-free and ratiometric detection of intracellular GSH and other biothiols in a wide linear range, which is of great significance for cancer diagnostics and therapy.

Abstract:A convenient, fluorescent dual-channel chemosensor on the basis of bis(3-pyridylmethyl)amine-functionalized carbon quantum dots (BPMA-CQDs) nanoprobe was constructed, and it can discriminatively detect glutathione from its analogues cysteine and homocysteine based on two distinctive strategies. Two distinct fluorescence responses of BPMA-CQDs probe to Cu(II) and Ag(I) were identified and further employed to achieve selective detection of Cu(II) and Ag(I) respectively. Based on the BPMA-CQDs/Cu(II) conjugate, discriminative detection of GSH was achieved in terms of correlation between the amounts of GSH and fluorescence recovery. The addition of GSH into BPMA-CQDs/Cu(II) system induces the reduction of Cu(II) to Cu(I), which could efficiently block PET process resulting in the following fluorescence recovery. Based on the BPMA-CQDs/Ag(I) conjugate, GSH assay could also be established on the basis of fluorescence response to GSH. The introduction of GSH into the preceding system triggers the competitive coordination to Ag(I) between BPMA and GSH, and silver ions are finally taken away by GSH from the probe, where the fluorescence is restored to its original weak state. Both of the detection strategies can achieve discriminative detection of GSH from Cys and Hcy. The assays showed good stability and repeatability, and covered a broad linear range of up to 13.3μM with a lowest detection limit of 42.0nM. Moreover, both of them were utilized to monitor GSH level in live cells.

Abstract:Glutathione (GSH) plays a critical role in biological defense system and is associated with numerous human pathologies. However, it still remains a challenge for fluorescent detection of GSH over cysteine (Cys) and homocysteine (Hcy) because of their similar structures. In this work, MnO2 nanosheets can efficiently quench the fluorescence of gold nanoclusters (Met-AuNCs) prepared by blending methionine and HAuCl4 owing to their superior absorption capability. However, GSH can reduce MnO2 nanosheets into Mn2+ which leads to the fluorescence recovery of Met-AuNCs. More intriguingly, GSH can dramatically and selectively enhance the fluorescence intensity of Met-AuNCs. Hence, a low background, ultrasensitive fluorescent detection of GSH was obtained with a detection limit of 68 nM. Moreover, the assay has been successfully used for GSH detection in human serum samples and cellular imaging with high selectivity over Cys and Hcy. (C) 2021 Elsevier B.V. All rights reserved.

Abstract:Glutathione (GSH) is a crucial non-protein thiol and an indispensable endogenous antioxidant. The aberrant expression of GSH in plasma and cytosol is closely related to numerous diseases, including cancer. Therefore, establishing a sensitive method for analyzing GSH has important application value for biomedical research and clinical medical detection. Herein, A method for the rapid and simple detection of GSH was proposed, which is based on an anti-etching mechanism by utilizing gold nanotetrapods (Au NTPs) and manganese dioxide nanosheets (MnO 2 NSs). In the absence of GSH, Au NTPs solution can cause a distinct color change from gray-green to red through the etching effect of MnO 2 NSs. However, in the presence of GSH, the redox reaction between GSH and MnO 2 NSs inhibits the etching of Au NTPs by MnO 2 NSs, and Au NTPs solution maintains persistent gray-green color. The colorimetric probe exhibited excellent selectivity for GSH. The limits of detection for GSH were 43.5 nM (UV–vis spectrum) and 0.25 μM (naked eyes). The sensing technique exhibited excellent linearity between wavelength shift and GSH concentration within the range of 0.25 μM–1.5 μM. The outcomes of GSH detection in actual biological samples demonstrate that this probe has the potential to be applied to GSH detection in intricate biological samples.

Abstract:We have developed sensitive detection of glutathione using the IrO2-hemin-TiO2 nanowire arrays. Single-crystalline TiO2 nanowires are synthesized by a hydrothermal reaction, followed by surface functionalization of ~3 nm thick hemin and ~1-2 nm diameter IrO2 nanoparticles. The IrO2-hemin-TiO2 nanowire arrays offer much enhanced photocurrent with ∼100% increase compared to the pristine TiO2 nanowires and allow for label-free, real-time, sensitive photoelectrochemical detection of glutathione. The sensitivity achieved is ~10 nM in buffer, comparable to or better than most of the existing glutathione detection methods. Furthermore, cell extracts containing glutathione are robustly detected, with ~8000 cells/mL for HeLa cells and ~5000 cells/mL for human embryonic kidney 293T cells. This nanowire PEC sensor assay exhibits excellent selectivity and stability, suggesting a potential detection platform for analyzing the glutathione level in biosamples.

Abstract:A novel electrochemical sensor for highly sensitive and selective detection of glutathione (GSH) was developed coupled thymine-Hg2+-thymine (T-Hg2+-T) coordination with mesoporous silica nanoparticles (MSN). Hg2+ ions could specifically interact with thymine bases to form strong and stable T-Hg2+-T complexes. And the electroactive molecules-methylene blue (MB) was sealed in the inner pores of MSN with a unique sequential thymine (T)-rich DNA in the presence of Hg2+. After the target was added, the formed duplex DNA T-Hg2+-T complex could deform into single-stranded DNA by a competitive displacement reaction, leading to the release of MB entrapped in the MSN. Thus the target-dependent amount of released MB could be conveniently monitored with an n-dodecanethiol modified screen-printed gold electrode. Using GSH as a model target, the proposed assay showed a wide detection ranging from 1.0nM to 1.0μM with a detection limit of 0.6nM. This strategy was simple and universal for various analytes with different competitive displacement ligands, possessing a great potential for convenient biotools testing and commercial application.
A ratiometric fluorescence nanosensor for glutathione detection based on spatially confined dual-emission of α-lipoic acid-modified gold nanoclusters and silicon nanoparticles

Abstract:The development of dual-emission ratiometric fluorescent probes with aggregation-induced emission enhancement (AIEE) overcomes the limitations of gold nanocluster (Au NC)-based probes, particularly their weak intrinsic fluorescence, in real-world applications. These AIEE probes also exhibit superior detection limits and enhanced sensitivity. A novel combination for the reliable fluorometric detection of glutathione (GSH) was proposed, utilizing aggregation-induced emission enhancement (AIEE) facilitated by electrostatic interaction and spatial confinement. The probe consists of a ratiometric combination of negatively charged α-lipoic acid-modified Au NCs (LA@Au NCs) and positively charged silicon nanoparticles (SiNPs). The addition of SiNPs causes aggregation of LA@Au NCs, enhancing the fluorescence of LA@Au NCs through the AIE effect under electrostatic interaction and spatial confinement. The addition of Cu 2+ quenched the emission of LA@Au NCs as a result of charge transfer. The fluorescence emissions of LA@Au NCs were restored upon the addition of GSH due to the interaction between GSH and Cu 2+ . Simultaneously, the emission signal of SiNPs remains unchanged, serving as an internal reference signal during GSH measurement. It was found that the fluorescence ratio ( F 680 / F 465 ) is directly proportional to the concentration of GSH in the range of 0.05–100 μM, with a detection limit of 1.7 nM (S/N = 3). The proposed system was applied to detect GSH in real samples, including dietary supplements, human serum, and saliva samples. This work opens new avenues for constructing novel sensors based on AIEE for detecting biomolecules.

Abstract:For the widely used Au nanoparticles (AuNPs)-based colorimetric probe, AuNPs generally change from the dispersion to the aggregation state and corresponding colors turn from red to blue concomitantly. In previous studies, there are few probes based on the anti-aggregation of AuNPs though anti-aggregation of AuNPs is preferable to aggregation to achieve higher selectivity. In this manuscript, a fast and simple but sensitive and selective sensor suitable for on-site and real-time detection of glutathione (GSH) has been developed based on the anti-aggregation of AuNPs. The sensor has a LOD of 8 nM and excellent selectivity toward GSH by a factor of 200-fold or more relative to natural amino acids as well as homocysteine (Hcys) and glutathione disulfide (GSSG). The dynamic range of the sensor can be tuned simply by adjusting the amount of aggregation agent used.
A fluorescent probe for turn‐on sensing of glutathione using red emitting gold nanoclusters

Abstract:Schematic representation depicting the fluorescent sensing of GSH. The probe was developed by the addition of cystine (Cystine‐AuNCs) and further extended for the turn‐on sensing of GSH. The probe achieved sensitive detection of GSH with a low Limit of Detection (LoD) of 0.43 μM. Glutathione (GSH), an oxidative stress biomarker plays a major role in different pathological processes and lead to a vast number of diseases including cancer. Biologically, cystine uptake is a fundamental factor in maintaining GSH levels in cells; however, the interaction between cystine and GSH in biological systems is least explored in developing sensors for GSH. In the present work, we have explored this interaction as the fundamental factor in "turn‐on" sensing of GSH from body fluids using a fluorescent gold nanocluster platform. The experimental results revealed that the probe displayed appreciable performance for GSH over a linear range from 0.2 mM to 1.3 mM with a limit of detection (LoD) of 0.43 μM. The probe also revealed sensitive as well as selective response towards GSH even in the presence of possible co‐existing interferants and provided satisfactory recovery percentage of 94.17 % to 108.12 % from real serum samples. These findings of the present study can be extended in future applications of sensing GSH from clinical samples.

Abstract:Abnormal glutathione (GSH) is associated with various diseases, like Parkinson's, cancer, and cardiovascular disease. The efficient detection of GSH has been the research focus for many years. However, the current methods remain a grand challenge to substantially enhance the detection sensitivity and selectivity of GSH, owing to the unavoidable influence of other molecules containing free sulfhydryl groups on detecting GSH. Herein, a series of sterically and electronically modulated Au(I)-N-heterocyclic carbene (NHC) complexes were synthesized and applied for GSH detection. We demonstrated that the Au(I)-NHC complexes exhibited peroxidase-like activity, which could catalyze the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2 to produce blue oxidized TMB. Significantly, the catalytic activity could be enhanced by introducing the electron-withdrawing moieties on the NHC backbone. As a proof-of-concept application, the Au(I)-NHC complexes catalyzed TMB + H2O2 system is designed as a reliable colorimetric assay for sensitive detection of GSH, which displayed a low detection limit of 0.14 μM. The present findings provide an efficient strategy to discriminate GSH, which can remarkably extend the potential applications of Au(I)-NHC complexes as novel colorimetric biosensors and also opened promising avenues for more powerful biosensors that operate upon other metal-NHC complexes.
