Abstract:The covid-19 pandemic has highlighted ongoing concerns regarding biosafety and biosecurity procedures in global pathogen research. This includes questions about any possible role of field and laboratory research into the emergence of the SARS-COV-2 virus.1 The $125m Discovery and Exploration of Emerging Pathogens-Viral Zoonoses programme, funded by the US Agency for International Development, researching new zoonotic viruses was recently cancelled partly because of these concerns.2 This has raised broader discussion about the risks and benefits of viral surveillance and research. Limiting funds for pathogen surveillance and research compromises long term preparedness for pandemics and leaves pathogen spillover threats unmonitored. Rather than restrict scientific surveillance and research, a global safety governance framework that spans the entire pathogen value chain must be urgently developed. Abstract:Tremendous advances in the field of synthetic biology have been witnessed in multiple areas including life sciences, industrial development, and environmental bio-remediation. However, due to the limitations of human understanding in the code of life, any possible intended or unintended uses of synthetic biology, and other unknown reasons, the development and application of this technology has raised concerns over biosafety, biosecurity, and even cyberbiosecurity that they may expose public health and the environment to unknown hazards. Over the past decades, some countries in Europe, America, and Asia have enacted laws and regulations to control the application of synthetic biology techniques in basic and applied research and this has resulted in some benefits. The outbreak of the COVID-19 caused by novel coronavirus SARS-CoV-2 and various speculations about the origin of this virus have attracted more attention on bio-risk concerns of synthetic biology because of its potential power and uncertainty in the synthesis and engineering of living organisms. Therefore, it is crucial to scrutinize the control measures put in place to ensure appropriate use, promote the development of synthetic biology, and strengthen the governance of pathogen-related research, although the true origin of coronavirus remains hotly debated and unresolved. This article reviews the recent progress made in the field of synthetic biology and combs laws and regulations in governing bio-risk issues. We emphasize the urgent need for legislative and regulatory constraints and oversight to address the biological risks of synthetic biology.

Abstract:The United States is seeking to prevent four Chinese biotech companies from doing business in the country, citing them as "companies of concern" that threaten national security. The Biosecure Act, introduced in both the Senate (S.3558) in December and House of Representatives (H.R.7085) in January, would prohibit the federal government from contracting with certain biotech providers connected to foreign adversaries. The ban could extend to institutions that receive funding from the National Institutes of Health or other US government agencies, which would include almost all universities, research centers, hospitals and life science companies. The companies named in the bill are BGI Group, MGI Tech, Complete Genomics and WuXi AppTec — all Chinese-owned companies that provide instruments or services associated with the collection, sequencing or analysis of genetic information. The Senate bill's sponsor and chairman of the Senate Committee on Homeland Security & Governmental Affairs, Gary Peters (D-Mich.), said S.3558 was "critical legislation to strengthen our national security and combat risks posed by biotech companies from adversarial nations like China" and that "it is incredibly important that we advance this legislation quickly." In a press release on H.R.7085, sponsor Mike Gallagher (R-Wis.), chair of the House Select Committee on Strategic Competition between the United States and the Chinese Communist Party, said "The CCP will undoubtedly use the genetic data collected by BGI to further its malign aggression, potentially even to develop a bioweapon used to target the American people." Rachel King, CEO of industry trade group the Biotechnology Innovation Organization, responded in a letter to the Senate committee that the legislation would hamper US innovation, saying it would "do untold damage to the drug development supply chain both for treatments currently approved and on market as well as for development pipelines decades in the making."

Abstract:Israel has a long history of concern with chemical and biological threats, since several hostile states in the Middle East are likely to possess such weapons. The Twin-Tower terrorist attacks and Anthrax envelope scares of 2001 were a watershed for public perceptions of the threat of unconventional terror in general and of biological terror in particular. New advances in biotechnology will only increase the ability of terrorists to exploit the burgeoning availability of related information to develop ever-more destructive bioweapons. Many areas of modern biological research are unavoidably dual-use by nature. They thus have a great potential for both help and harm; and facilitating the former while preventing the latter remains a serious challenge to researchers and governments alike. This article addresses how Israel might best (1) prevent hostile elements from obtaining, from Israel's biological research system, materials, information and technologies that might facilitate their carrying out a biological attack, while (2) continuing to promote academic openness, excellence and other hallmarks of that system. This important and sensitive issue was assessed by a special national committee, and their recommendations are presented and discussed. One particularly innovative element is the restructuring and use of Israel's extensive biosafety system to also address biosecurity goals, with minimal disruption or delay.

Abstract:UK scientists have criticized a ban by the US administration on studies that increase the potency or transmissibility of viruses, saying that such studies were needed to increase understanding of viruses and to improve vaccines against them.The ban, announced unexpectedly by the White House late last week, came after a voluntary moratorium on such studies that ended in 2013 without resolving the argument between those who said that generating “superstrains” of viruses was too risky and those who defended them as a valuable tool, as long as appropriate precautions against release were in place. Abstract:The National Science Advisory Board for Biosecurity (NSABB)'s recommendation to restrict publication of the details of the generation of mammalian-transmissible H5N1 influenza virus is unprecedented. Dual-use considerations indicated that the potential biosecurity risks of a transmissible H5N1 virus with a possible mortality of 50% in humans outweigh the substantial benefits of open and complete scientific exchange in this case, although the benefits include potential early detection strategies for H5N1 viruses with specific genetic markers and control strategies, including development of antivirals and vaccines. It is argued that both the funding agency (the National Institute of Allergy and Infectious Diseases) and the scientists were responding to societal needs and acted entirely responsibly. These studies usher in a new era for life sciences, compelling the research community to confront important decisions: under what conditions should such research be done? How can the principle of full release of information be balanced with the moral imperative to protect the public health?

Abstract:CRISPR, a powerful gene-editing technology, is revolutionizing the life sciences and medical research. The technology has also become democratized. Costs to use CRISPR are low and decreasing, kits are available to make the use of CRISPR straightforward, and there is a rapidly growing scientific literature describing CRISPR methodologies and novel applications. However, like other powerful advances in the life sciences, CRISPR raises biosecurity concerns: it could be misused for harm, and it lowers technical barriers to biological weapons development. This essay describes the history and dissemination of CRISPR as genome-editing techniques have become widespread, outlines potential biosecurity concerns, and recommends actions governments and scientists may take to reduce biosecurity risks. While it is not possible to eliminate biosecurity risks from the misuse of biotechnologies, including CRISPR, steps can be taken to increase security while allowing this powerful technology to remain widely available for beneficent purposes.

Abstract:If the attacking force has a vaccine before the attack, will inflict a crippling damage on the enemy. Thus, virus emerges as potential danger since they can be used in the development of such weapons. The reported of a neurotropic strain influenza A virus (IAV) induced the conversion of normal prion protein (PrPC) into infectious prion protein (PrPSc) as well as formation of infectious prions bring out the danger of a type of prion/virus as unprecedented infectious pathogens that cause fatal neurodegenerative diseases. Due to their singular characteristics a virus inducer of prion disease can be a stealth advantage for the attacker if he has an effective vaccine. Therefore, lethal prion/virus can be developed by malicious researchers who could use it to attack political enemies.

Abstract:Transparency was not always a desired aspect of medicine or STEM (science, technology, engineering, and math) research. In the late 1940s, the Nuremberg Code heralded a new era of informed patient consent, research subject protection, and the view that the public had a stake in emerging technology and should have some knowledge and input into the directions of scientific research. This understanding intensified in the United States with the very public discussions leading to the promulgation of the NIH Guidelines for Recombinant DNA Research in the 1970s. The way in which oversight of recombinant DNA research was handled is still the exception rather than the rule. Starting in the 1990s, various terror incidents led to the enactment of statutes and issuance of regulations that undermined the ability of scientists and research institutions to self-regulate and in some cases to disseminate information freely. This essay explores how the scientific community got to this status quo, and how it could regain some measure of control despite competing needs for transparency and security, so that research critical to biosecurity is supported rather than impeded.

Abstract:Since the September 11, 2001, terrorist attacks, concern for the future direction of biological research has expanded to include not only issues involving the welfare of mankind, but also the more immediate and personal concern of individual scientists in the life sciences community. For many investigators in the life sciences, the remainder of their careers will be impacted by concerns about bioterrorism and issues associated with dual-use research. Awareness of these issues will be critical for all professional groups and affiliated organizations as they navigate the growing call for more federal rules and regulations. Veterinarians are not immune to the coming changes in science; all researchers risk criminal sanctions if they violate the USA Patriot Act or the Bioterrorism Act of 2002. Compliance with these regulations will be necessary, not just because of the potential legal ramifications, but because establishing and maintaining public trust is a never-ending requirement for the future of scientific research.

Abstract:In response to the outbreak of COVID-19 that has been sweeping the world, scientists reconstructed the SARS-CoV-2 rapidly using a synthetic genomics platform, in order to accelerate therapeutics and vaccine development. However, given the dual-use nature of this technology, there exists a high biosecurity risk. This paper points out the potential risks of the engineering SARS-CoV-2 virus and puts forward 6 questions to this work. The authors emphasize that the two basic values of safety/security and intellectual freedom of research must be considered evenly. From the perspective of responsible development of biotechnology, this paper calls for a careful assessment to the risks of the technology, replacing risky technologies with safe ones. The risks of publication also need to be strictly assessed. The authors believe, in addition to enhancing the "self-government" and self-discipline of scientists and scientific communities, government supervision must be reinforced, laws and regulations should be improved, and global regulation framework ought to be constructed.

Abstract:Significance The recent emergence and spread of zoonotic viruses, including Ebola virus and severe acute respiratory syndrome coronavirus 2, demonstrate that animal-sourced viruses are a very real threat to global public health. Virus discovery efforts have detected hundreds of new animal viruses with unknown zoonotic risk. We developed an open-source risk assessment to systematically evaluate novel wildlife-origin viruses in terms of their zoonotic spillover and spread potential. Our tool will help scientists and governments assess and communicate risk, informing national disease prioritization, prevention, and control actions. The resulting watchlist of potential pathogens will identify targets for new virus countermeasure initiatives, which can reduce the economic and health impacts of emerging diseases.
