Abstract:Objective: To assess how laser light affects blood lymphocyte viability in vitro. Methods: The comparative study was conducted from November 2021 to April 2022 at the Department of Medical Physics, Mustansiriyah University, Baghdad, Iraq, and comprised blood samples collected from healthy adults with no medical history of major illnesses or history of taking medications for major diseases.Low-level laser 589nm and 532nm was used at 30 J/cm², 50 J/cm² and 70J/cm² energy intensities and three different post-exposure time; immediately, 1h and 2h after radiation exposure. The viability of normal human lymphocytic cells of the blood was noted. Data was analysed using SPSS 24. Results: The blood samples were drawn from 6 adult male voluteers. The percentage of cells that showed apoptosis post-exposure to 589nm laser was significantly lower than that following exposure to 532nm laser (p<0.05). However, the proportion of apoptotic cells was significantly higher following irradiation with 532nm at varying doses than after irradiation with 589nm (p<0.05). Conclusions: A low-level laser could promote and prevent apoptosis in human peripheral blood cells in a dose- and time-dependent manner.
Effect of 532nm and 589nm Diode-Pumped Solid-State (DPSS) Lasers with Different Doses on Concentration of Total Protein and Albumin in Vitro

Abstract:Objective: To test the effects of Low-level laser irradiation at 532nm and 589nm with different doses and exposure times on total protein and albumin in blood plasma. Methods: This experimental study was conducted from November 2021 to April 2022 at Mustansiriyah University, Baghdad, Iraq, and comprised blood samples from healthy adults. The samples were stored in tubes containing anticoagulant ethylenediaminetetraacetic acid. Photoradiation was managed using green laser 532nm and yellow laser 589nm diode-pumped solid-state lasers with different doses. For total protein test and albumin test, each sample was divided into 3 equal aliquots. The first was control without radiation, the second was irradiated by a laser at 532nm, and the third was irradiated at 589nm with 30J/cm², 50J/cm², 70J/cm² and 90 J/cm² doses of irradiation. All samples were examined by using a spectrophotometer device. Data was analysed using SPSS 25. Results: There were 24 samples from healthy adults in all.The most significant difference in total protein concentration occurred at dose 70J/cm² for both wavelengths compared to the controls (p<0.05). When this optimal dose was applied to the albumin test, a clear decrease in the albumin concentration was found for both 532nm and 589nm lasers (p<0.05). Conclusions: At wavelengths of 532nm and 589nm, the laser light induced processes that caused a reduction in total protein and albumin.
The effect of a diode-pumped solid-state laser with a wavelength of 589 nm on T-lymphocyte proliferation

Abstract:Objective: To assess how low-level laser irradiation affects T cells. Methods: The case-control interventional study was conducted from November 2021 to April 2022 after approval by the ethics review committee of the College of Medicine, Mustansiriyah University, Baghdad, Iraq, and comprised healthy individuals. Phlebotomy was used to collect 4ml of blood. A portion of each sample was exposed to 589nm laser radiation, while the other portion was used as a control. The irradiated sample was divided into whole blood and in-plate subgroups. Phytohemagglutinin was used for mitotic stimulation of T-lymphocytes, and cell proliferation assay was used to stimulate T lymphocytes measured using MTS assay (3-(4,5-dimethylthiazol-2-yl)-5- (3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium). Optical density was measured using enzyme-linked immunosorbent assay. Data was analysed using GraphPad prism software. Results: There were 35 subjects, 17(49%) males and 18 (51%) females, with an age range between 19-25 years, all without any disease or medication. Laser irradiation stimulated T-lymphocyte proliferation to levels that depended on the dose of laser used. At energy 30J/cm2 there was a significant increase in the in-plate subgroup of irradiated samples compared to controls and between the two irradiated subgroups (p<0.05). At 50J/cm2 dose, there was a significant increase in the whole blood subgroup compared to the controls (p<0.05). At 70J/cm2 dose, there was a significant difference in the whole blood subgroup compared to the controls, and between the two irradiated subgroups (p<0.05). Conclusions: The stimulation of T lymphocyte proliferation by the application of a 589nm laser could have medical applications in the field of immune system enhancement.
Effect of diode pumping solid state laser with wavelength 589 nm on gene expression of interlukine-2 and interferon-γ in human T-lymphocytes

Abstract:Objective: To evaluate the impact of laser irradiation of human T-cell lymphocyte in culture to determine the photobiomodulatory effects of such irradiation on gene expression, and the release of growth factors and cytokines. Methods: The experimental study was conducted from November 2021 to April 2022 at the Postgraduate Anatomy Laboratory, Mustansiriyah University, Baghdad, Iraq, and comprised human blood samples T-cell lymphocytes were extracted and irradiated, followed by the extraction of ribonucleic acid to assess the effect of laser on gene expression of cytokines, interlukine-2 and interferon-gamma. A 589nm yellow light low-level laser beam from a diode pumping solid state laser was used at doses 30J/cm2, 50J/cm2 and 70J/cm2 for an exposure time of 15 minutes. Reverse transcriptase-polymerase chain reaction was used to measure gene expression. Data was analysed using GraphPad Prism 9. Results: Cellular gene expression of the mediators changed significantly in response to laser irradiation at a variety of dosage parameters, and these effects depended on the wavelength and radiation exposure (p<0.05). Conclusions: The laser light may photobiomodulate the production of cytokines by human cells in vitro.
A comparative study of 632.8 and 532 nm laser irradiation on some rheological factors in human blood in vitro

Abstract:The effects of laser irradiation with 632.8 and 532 nm on rheological properties of blood were comparatively studied in vitro. Under the irradiation condition of 30 mW, laser irradiation of blood samples using a spot diameter of 5 mm with each laser, showed promising results in the modulation of hemorheological properties. When blood samples from patients with abnormally high values of erythrocyte sedimentation rate (ESR) were irradiated, the values of ESR were lowered statistically by either of the 632.8 or 532 nm lasers. The laser irradiation reduced blood viscosities at different shear rates (10–110 S−1) for the hyper-viscosity blood samples. Laser irradiation increased the electrophoretic mobility (EPM) of erythrocytes when the values of the sample's EPM were abnormally slow. The erythrocyte deformability was enhanced by laser irradiation when the deformability of the sample from the patients was originally poor. For verifying the improvement of laser irradiation on erythrocyte deformability, the typical erythrocyte samples with poor deformability were produced by the pre-treatment of the erythrocytes with Ca2+. The deformability of these erythrocyte samples was also improved after laser irradiation. These results suggest that membrane-bound hemoglobin (Hbm) might be the initial site of the interaction, since Hbm is the main cause of poor deformability when erythrocytes were treated with Ca2+. In all experiments including ESR, blood viscosity, EPM and erythrocyte deformability, the 532 nm laser demonstrated more efficient effects on modulating rheological properties than 632.8 nm laser. This wavelength effect is consistent with the absorption spectrum of hemoglobin, reflecting that hemoglobin may be one of the action targets under laser irradiation.

Abstract:OBJECTIVE:The purpose of this study was to investigate the in vitro effects of He-Ne laser irradiation on some rheological factors of human blood, such as blood viscosity, erythrocyte deformability, and sedimentation rate.BACKGROUND DATA:The intravascular irradiation of low power laser has been applied in pre-clinical and clinical to treat various pathological processes. However, the mechanism is not fully understood so far. Especially the interaction and related mechanism between the laser and blood are unclear. In this work, by measuring the change of the main rheological factors after laser irradiation, the interaction and mechanism were explored.METHODS:A30-mW He-Ne laser was used for irradiation with a 4-5-mm-diameter beam spot on blood samples, with a fluence rate of about 150 mW/cm.(2) The irradiation time was 60 min, so the total dose of irradiation was 540 J/cm.(2) The pathological samples of blood were obtained from patients (volunteers), and each sample was divided into two tubes for irradiation and control. The blood viscosity, erythrocyte deformability, and sedimentation rate were measured after laser irradiation and compared with un-irradiated control. The blood samples with poor erythrocyte deformability were prepared by adding Ca(2+) to the normal erythrocytes of a healthy person for investigating the laser effect on erythrocyte deformability further.RESULTS:Laser irradiation reduced the erythrocyte sedimentation rate of blood samples, which had a hyper-sedimentation rate originally. The blood viscosity of samples in hyper-values was lowered by laser irradiation in all shear rates measured (10-110 S(-1)), with a relative variation of approximately 10%. The deformability of erythrocytes from pathological samples and Ca(2+)-treated samples was improved after laser irradiation.CONCLUSIONS:The positive effects of laser irradiation on improving the rheological properties of blood were demonstrated in vitro.
Comparison of the effect of wavelength 660 with 808 nm diode as a low level laser on non-surgical periodontal treatment in chronic periodontitis: a double-blind split-mouth randomized controlled clinical trial

Abstract:Background and purposePrevious studies have shown the effectiveness of low level lasers in non-surgical periodontal treatments as an adjunctive agent. The aim of this study is to evaluate and compare two wavelengths of 660 and 808 nm as low level lasers therapy in non-surgical periodontal treatment on periodontal clinical parameters in patients with chronic periodontitis.Materials and methodsThis randomized controlled split-mouth clinical trial study was conducted on 71 patients with chronic periodontitis with a periodontal pocket depth ≥ 5 mm or clinical attachment loss ≥ 3 mm in premolar teeth. Clinical periodontal parameters including clinical attachment loss (CAL), gingival index (GI), and pocket probing depth (PPD) were measured in premolar teeth. Scaling and root planning of teeth in the whole mouth were done in one session. Then the laser was irradiated randomly with two wavelengths of 660 nm and 808 nm in two different quadrants. Laser treatment with an energy density of 4 J/cm2 was repeated at intervals of 1, 3, and 7 days after the initial treatment. Re-evaluation of the clinical parameters was done 1, 3, and 6 months after the treatment of the patients. Data were analyzed with SPSS 26, using the Friedman, Wilcoxon, and Mann–Whitney comparative tests.ResultsPeriodontal clinical parameters improved significantly 1, 3, and 6 months after the intervention in both groups (P-value < 0.0001), and these changes were greater in the 660 wavelengths. There was a significant difference between the two groups in PPD, GI, and CAL parameters (P-value < 0.0001).ConclusionThe results of this clinical trial study showed that red laser (660 nm wavelengths) significantly improved periodontal clinical parameters compared to infrared laser (808 nm wavelengths).
Towards a mechanistic understanding of lipodystrophy and seipin functions

Abstract:CGL (Congenital generalized lipodystrophy) is a genetic disorder characterized by near complete loss of adipose tissue along with increased ectopic fat storage in other organs including liver and muscle. Of the four CGL types, BSCL2 (Berardinelli–Seip Congenital lipodystrophy type 2), resulting from mutations in the BSCL2/seipin gene, exhibits the most severe lipodystrophic phenotype with loss of both metabolic and mechanical adipose depots. The majority of Seipin mutations cause C-terminal truncations, along with a handful of point mutations. Seipin localizes to the ER and is composed of a conserved region including a luminal loop and two transmembrane domains, plus cytosolic N- and C-termini. Animal models deficient in seipin recapitulate the human lipodystrophic phenotype. Cells isolated from seipin knockout mouse models also exhibit impaired adipogenesis. Mechanistically, seipin appears to function as a scaffolding protein to bring together interacting partners essential for lipid metabolism and LD (lipid droplet) formation during adipocyte development. Moreover, cell line and genetic studies indicate that seipin functions in a cell-autonomous manner. Here we will provide a brief overview of the genetic association of the CGLs, and focus on the current understanding of differential contributions of distinct seipin domains to lipid storage and adipogenesis. We will also discuss the roles of seipin-interacting partners, including lipin 1 and 14-3-3β, in mediating seipin-dependent regulation of cellular pathways such as actin cytoskeletal remodelling.
In vitro impact of laser irradiation on platelet aggregation

Abstract:The review of blood optical and rheological parameters plays an important role in many medical routine diagnosis and therapeutic applications and is the best way to understand the mechanism of action of low-level laser irradiation on biological tissues. The aim of this study was to investigate the in vitro effect of laser radiation on platelet count and aggregation. Blood samples were obtained from 30 healthy volunteers; each sample was divided into four aliquots, one of them was considered as a control while the other three were exposed to three different laser doses. A wavelength of 532 nm and a low power of 100 mW were used for irradiation with a 4-mm-diameter beam spot. The irradiation times were 1.8, 3.7, and 6.2 s giving doses of irradiation 1.5, 3, and 5 J/cm2, respectively. Microsoft Excel was used for statistical analysis. Low laser irradiation induced significant changes in platelet aggregation in the presence of weak agonists such as adenosine diphosphate (ADP) (P ≤ 0.05) and epinephrine (P ≤ 0.01). Low-level laser therapy has no influence on platelet count; however, it promotes platelet aggregation in response to weak agonists, specifically ADP and epinephrine.

Abstract:Diode laser irradiation is a novel approach applied to preserve the quality of fresh commodities and minimise oxidative processes, including browning in fresh-cut products. The objective of this study was to investigate the efficacy of diode laser treatment in enhancing the quality of celtuce slices during refrigerated storage at 4 degrees C. The celtuce slices were subjected to a 660 or 450 nm diode laser for 5 min and then stored for 10 d. Both laser treatments hindered the progression of weight loss and firmness increase in the celtuce slices. The use of a 450 nm diode laser effectively prevented the development of browning and preserved the surface colour by inhibiting the increase in a* and overall colour difference values, as compared to other treatments, during storage for 10 d. The diode laser treatment suppressed the increase in oxidative stress by lowering the levels of MDA, H2O2, and O-2 while retaining a high level of DPPH scavenging activity, AsA, and antioxdant enzyme activities, including SOD, CAT, APX, and GR, in comparison to the untreated slices. The treatment enhanced chlorophylls content due to the up-regulation of LsHEMA1 gene and down-regulation of LsNYC1 gene. The treatment also inhibited factors involving enzymatic browning reaction by suppressing PAL activity, total phenols content, PPO activity and LsPPO gene expression. These suggest that the blue diode laser exposure at 450 nm for 5 min is an effective approach hindering oxidative stress and discoloration of celtuce slices during storage.
The in Vitro Effects of Low Power Laser Irradiation on Animal Erythrocyte Rheology

Abstract:Using the blood samples of animal, the in vitro effects of low power laser irradiation on erythrocyte rheology were investigated. After the deposited pig′s erythrocytes (the deformation of erythrocytes have already turned to worse) were irradiated with laser, the effects of laser irradiation on erythrocyte′s deformation were measured. When 650 nm laser was used for irradiation, the deformability of erythrocytes was improved obviously. The changing tendency is that, the erythrocyte′s deformation was enhanced with the increasing of irradiation power and then saturated around 4～5 mW. When the blood samples were irradiated by 650 nm and 632.8 nm lasers respectively with the same power (10 mW), the erythrocyte deformability in two cases were all obviously increased to the similar level. These phenomena can be explained by that, the hemoglobin in erythrocytes has the similar absorption to both lasers. Taking the mouse blood as the samples, the effects of laser irradiation on erthrocyte′s electrophoretic mobility was studied further. When irradiated by 632.8 nm laser, the electrophoretic mobility of erythrocytes was speeded also, reflecting the electric charges on the surface of erythrocytes were increased which will help to decrease the aggregation of erythrocytes. Under such low power irradiation (less than 20 mW), no morphological change was detected by the microscopy and no hemolysis was found also.

Abstract:PURPOSE: Low level laser therapy (LLLT) in the visible to near infrared spectral band (390-1100 nm) is absorption of laser light at the electronic level, without generation of heat. It may be applied in a wide range of treatments including wound healing, inflammation and pain reduction. Despite its potential beneficial impacts, the use of lasers for therapeutic purposes still remains controversial in mainstream medicine. Whilst taking into account the physical characteristics of different qualities of lasers, this review aims to provide a comprehensive account of the current literature available in the field pertaining to their potential impact at cellular and molecular levels elucidating mechanistic interactions in different mammalian models. The review also aims to focus on the integral approach of the optimal characteristics of LLLT that suit a biological system target to produce the beneficial effect at the cellular and molecular levels.METHODS: Recent research articles were reviewed that explored the interaction of lasers (coherent sources) and LEDs (incoherent sources) at the molecular and cellular levels.RESULTS: It is envisaged that underlying mechanisms of beneficial impact of lasers to patients involves biological processes at the cellular and molecular levels. The biological impact or effects of LLLT at the cellular and molecular level could include cellular viability, proliferation rate, as well as DNA integrity and the repair of damaged DNA. This review summarizes the available information in the literature pertaining to cellular and molecular effects of lasers.CONCLUSIONS: It is suggested that a change in approach is required to understand how to exploit the potential therapeutic modality of lasers whilst minimizing its possible detrimental effects.

Effect of Pulsed Low-Level Lasers on Adult versus Neonatal Human Retinal Pigment Epithelial Cells: An in-vitro Study

Abstract:Purpose: To investigate the short-term effects of low-level lasers (LLLs; also known as low-power laser therapy) on the structure, genetic, and phenotype of cultured human retinal pigment epithelial (hRPE) cells from both adult and neonatal sources. Methods: Cultivated adult and neonatal hRPE cells were irradiated with two types of LLL (630 nm and 780 nm), 1 min daily for five consecutive days. Results: An increase in doubling time was observed in 630 nm-irradiated adult hRPE cells (P = 0.032). The gene expression profile revealed increased expression of retinoid isomerohydrolase RPE65 (RPE65) (P < 0.01 for 630 nm laser, P < 0.001 for 780 nm laser) and nestin (NES) (P < 0.01 for 630 nm laser) in neonatal hRPE cells, upregulation of RPE65 (P < 0.001 for 780 nm laser) and paired box 6 (PAX6) (P < 0.001 for 780 nm laser) genes in adult hRPE cells, and reduced expression of actin alpha 2 (ACTA2) in 780 nm-irradiated adult hRPE cells (P < 0.001). Except the significant increase of α -SMA in 780 nm-irradiated neonatal hRPE cells, no significant change was noted in the expressions of other investigated proteins. Conclusion: Short-term irradiation of neonatal and adult hRPE cells with LLLs may induce multipotency at the transcriptional level. Irradiation of neonatal hRPE cells with LLLs can be associated with increased risk of myofibroblastic transformation; however, adult hRPE cells irradiated with the 780 nm laser have minimal risk of myofibroblastic differentiation. It seems that the 780 nm laser may be a promising option for future photobiomodulation in retinal degenerations in adults.
Improving human sperm motility via red and near-infrared laser irradiation: in-vitro study

Abstract:Abstract Improved sperm motility is necessary for successful sperm passage through the female genital system, efficacious fertilization, and a greater probability of pregnancy. By stimulating the mitochondrial respiratory chain, low-level laser photobiomodulation has been shown to increase sperm motility and velocity. The respiratory chain in mitochondria is the primary site of action for cytochrome c oxidase because it can absorb light in the visible and infrared ranges. The present study aimed to investigate the effects of red laser 650 nm, near infrared laser (NIR) 980 nm, and combination of both on human spermatozoa motility and DNA integrity at different doses. An in-vitro controlled trial was performed in Al Zahraa university hospital laboratory using thirty fresh human semen specimens. Samples were exposed to red laser 650 nm, near infrared laser (NIR) 980 nm, and combination of both for various irradiation times. Sperm motility for the test and control aliquots was assessed as recommended in the manual of WHO-2021. Sperm chromatin integrity was evaluated using the Sperm Chromatin Structure Assay. Results revealed almost 70%, 80% and 100% increase in the total motility after 3 min of the 650-nm, 980-nm and the combined laser irradiation, respectively. Additionally, the Sperm Chromatin Dispersion assay was carried out on sperm heads utilizing human sperm DNA fragmentation, demonstrating that none of the three laser types had any discernible effects. A novel technique for reference point generation to aid in intraoral scan alignment.

Abstract:OBJECTIVE When using a completely digital workflow on larger prosthetic cases it is often difficult to communicate to the laboratory or chairside Computer Aided Design and Computer Aided Manufacturing system the provisional prosthetic information. The problem arises when common hard tissue data points are limited or non-existent such as in complete arch cases in which the 3D model of the complete arch provisional restorations must be aligned perfectly with the 3D model of the complete arch preparations. In these instances, soft tissue is not enough to ensure an accurate automatic or manual alignment due to a lack of well-defined reference points. A new technique is proposed for the proper digital alignment of the 3D virtual model of the provisional prosthetic to the 3D virtual model of the prepared teeth in cases where common and coincident hard tissue data points are limited. Clinical considerations: A technique is described in which fiducial composite resin dots are temporarily placed on the intraoral keratinized tissue in strategic locations prior to final impressions. These fiducial dots provide coincident and clear 3D data points that when scanned into a digital impression allow superimposition of the 3D models. CONCLUSIONS Composite resin dots on keratinized tissue were successful at allowing accurate merging of provisional restoration and post-preparation 3D models for the purpose of using the provisional restorations as a guide for final CLINICAL SIGNIFICANCE: Composite resin dots placed temporarily on attached tissue were successful at allowing accurate merging of the provisional restoration 3D models to the preparation 3D models for the purposes of using the provisional restorations as a guide for final restoration design and manufacturing. In this case, they allowed precise superimposition of the 3D models made in the absence of any other hard tissue reference points, resulting in the fabrication of ideal final restorations.
Effect of Low-Level Diode Laser and Red Light-Emitting Diode on Survival and Osteogenic/Odontogenic Differentiation of Human Dental Pulp Stem Cells

Abstract:Background: This investigation set out to compare the impacts of low-level diode laser (LLDL) and red light-emitting diode (LED) on the survival of human dental pulp stem cells (hDPSCs) and osteogenic/odontogenic differentiation. Methods and materials: In this ex vivo experimental study, the experimental groups underwent the irradiation of LLDL (4 J/cm2 energy density) and red LED in the osteogenic medium. Survival of hDPSCs was assessed after 24 and 48 h (n = 9) using the methyl thiazolyl tetrazolium (MTT) assay. The assessment of osteogenic/odontogenic differentiation was conducted using alizarin red staining (ARS; three repetitions). The investigation of osteogenic and odontogenic gene expression was performed at two time points, specifically 24 and 48 h (n = 12). This analysis was performed utilizing real-time reverse-transcription polymerase chain reaction (RT-PCR). The groups were compared at each time point using SPSS version 24. To analyze the data, the Mann-Whitney U test, analysis of variance, Tukey's test, and t-test were utilized. Results: The MTT assay showed that LLDL significantly decreased the survival of hDPSCs after 48 h, compared with other groups (p < 0.05). The qualitative results of ARS revealed that LLDL and red LED increased the osteogenic differentiation of hDPSCs. LLDL and red LED both upregulated the expression of osteogenic/odontogenic genes, including bone sialoprotein (BSP), alkaline phosphatase (ALP), dentin matrix protein 1 (DMP1), and dentin sialophosphoprotein (DSPP), in hDPSCs. The LLDL group exhibited a higher level of gene upregulation (p < 0.0001). Conclusions: The cell survival of hDPSCs was reduced, despite an increase in osteogenic/odontogenic activity. Clinical relevance: Introduction of noninvasive methods in regenerative endodontic treatments.

Abstract:Objective To observe the change of hemorheological factors and viscosity in blood via nasal low level laser radiation (NLLLR),and to assess ith efficacy and safety.Methods Eighty-four subjects with hemorheological disorder were divided into trial or control groups randomly by 1∶1 enveloped sequence. The trial subjects were irradiated by 635 nm red diode laser device for 30 minutes per day for 15 days,meanwhile the control ones received pseudo-laser irradiation in the same time. The blood and urine samples were collected at pre-and post-irradiation and 10 days after the irradiation. Results The main parameter values about erythrocyte aggregation and erythrocyte deformability in trial group were 10.27±1.11 and 4.18±0.43 after NLLLR,respectively,which were remarkably lower than that of the baseline (11.44±1.59 and 4.48±0.52 ,P<0.01 and P<0.01). The values likewise had 95% CI 1.17(0.80-1.54)and 0.30(0.18-0.42),respectively. The efficacy of NLLLR,however,did not maintain after 10 days,for no significant difference of erythrocyte aggregation and erythrocyte deformability between 2 groups was found (P>0.05). The results had indicated that NLLLR had a weak effect on plasma viscosity,total cholesterol,triglycerin,and glucose as well (P>0.05). Blood pressure,heart rate,routine tests,hepatic and renal functions and nasoscope were within normal range during the trial course in 2 groups. Conclusions NLLLR could be a useful and safe technique to improve erythrocyte aggregation and erythrocyte deformability,and prevent the occurrence of hemorheological disorder.

Abstract:Red blood cell (RBC) storage solutions have evolved significantly over the past decades to optimize the preservation of cell viability and functionality during hypothermic storage. This comprehensive review provides an in-depth analysis of the effects of various storage solutions and conditions on critical RBC parameters during refrigerated preservation. A wide range of solutions, from basic formulations such as phosphate-buffered saline (PBS), to advanced additive solutions (ASs), like AS-7 and phosphate, adenine, glucose, guanosine, saline, and mannitol (PAGGSM), are systematically compared in terms of their ability to maintain key indicators of RBC integrity, including adenosine triphosphate (ATP) levels, morphology, and hemolysis. Optimal RBC storage requires a delicate balance of pH buffering, metabolic support, oxidative damage prevention, and osmotic regulation. While the latest alkaline solutions enable up to 8 weeks of storage, some degree of metabolic and morphological deterioration remains inevitable. The impacts of critical storage conditions, such as the holding temperature, oxygenation, anticoagulants, irradiation, and processing methods, on the accumulation of storage lesions are also thoroughly investigated. Personalized RBC storage solutions, tailored to individual donor characteristics, represent a promising avenue for minimizing storage lesions and enhancing transfusion outcomes. Further research integrating omics profiling with customized preservation media is necessary to maximize post-transfusion RBC survival and functions. The continued optimization of RBC storage practices will not only enhance transfusion efficacy but also enable blood banking to better meet evolving clinical needs.
Cellular and Biochemical Effects of Combined X-Ray Radiation and Storage on Whole Blood

Abstract:Background: Evaluating the impact of ionizing radiation on stored blood is relevant since blood banks are major assets in emergency conditions such as radiation incident/attack. This study aimed to fill our knowledge gap of combined radiation and storage effects on blood. Methods: Blood collected from 16 anesthetized rats was anticoagulated, aliquoted into storage bags, and assigned to 8 groups using protocols combining storage (1-day vs 3-day 4oC) plus irradiation (75 Gy vs 0 Gy - control). Bags were positioned inside an X-ray irradiator (MultiRad-350). Complete blood count, differential white blood cell count, biochemistry, and hemostasis were analyzed (≥7 bags/group). Results: Na+, bicarbonate, glucose, and pH significantly reduced, while K+, Cl-, and lactate increased by storage. Coagulation measures were not significantly altered after radiation. White blood cell count and most cell types were numerically reduced after radiation, but changes were statistically significant only for monocytes. No significant alterations were noted in aggregation or rotational thromboelastometry parameters between irradiated and control. Conclusions: Evaluating cellular/biochemical parameters aids in assessing stored blood adequacy after radiation. Data suggest that fresh or cold-stored blood can sustain up to 75 Gy without major critical parameter changes and may remain suitable for use in critically ill patients in military/civilian settings.
Visible Raman excitation laser induced power and exposure dependent effects in red blood cells

Abstract:We present results of Raman spectroscopic studies carried out on optically trapped red blood cells with Raman excitation wavelength in Q-band region of the hemoglobin (Hb) absorption spectrum. The results obtained suggest that when exposed to the Raman excitation laser the RBCs get deoxygenated due to photo-dissociation of oxygen from hemoglobin. For smaller exposure durations (5 s) the level of deoxygenation increases with an increase in power. However, for longer exposure durations the deoxygenated hemoglobin in the cells gets irreversibly oxidized to form a low spin ferric derivative of hemoglobin. The rate of oxidation depends upon the initial level of deoxygenation; higher the initial level of deoxygenation, higher is the rate of oxidation. However, the RBCs deoxygenated via oxygen deprivation (i.e. N2 purging) were found to be very stable against any laser induced effect. These observations suggests that in case of laser induced deoxygenation of RBCs the free oxygen generated by photo-dissociation acts as the oxidizing agent and leads to oxidative damage of the RBCs.
