Abstract:Abstract Improved sperm motility is necessary for successful sperm passage through the female genital system, efficacious fertilization, and a greater probability of pregnancy. By stimulating the mitochondrial respiratory chain, low-level laser photobiomodulation has been shown to increase sperm motility and velocity. The respiratory chain in mitochondria is the primary site of action for cytochrome c oxidase because it can absorb light in the visible and infrared ranges. The present study aimed to investigate the effects of red laser 650 nm, near infrared laser (NIR) 980 nm, and combination of both on human spermatozoa motility and DNA integrity at different doses. An in-vitro controlled trial was performed in Al Zahraa university hospital laboratory using thirty fresh human semen specimens. Samples were exposed to red laser 650 nm, near infrared laser (NIR) 980 nm, and combination of both for various irradiation times. Sperm motility for the test and control aliquots was assessed as recommended in the manual of WHO-2021. Sperm chromatin integrity was evaluated using the Sperm Chromatin Structure Assay. Results revealed almost 70%, 80% and 100% increase in the total motility after 3 min of the 650-nm, 980-nm and the combined laser irradiation, respectively. Additionally, the Sperm Chromatin Dispersion assay was carried out on sperm heads utilizing human sperm DNA fragmentation, demonstrating that none of the three laser types had any discernible effects. What problem does this paper attempt to address?
The problem that this paper attempts to solve is to improve the motility of human sperm by using red - light laser (650 nm) and near - infrared laser (980 nm) as well as irradiation with a combination of the two, and to evaluate the potential impact of these treatments on sperm DNA integrity. Specifically, the research aims to: 1. **Improve sperm motility**: The research observes the impact of different doses of red - light laser, near - infrared laser and their combined irradiation on the motility of human sperm. The improvement of sperm motility is crucial for successfully passing through the female reproductive system, effective fertilization and increasing the probability of pregnancy. 2. **Evaluate sperm DNA integrity**: In addition to evaluating the improvement of sperm motility, the research also evaluates the impact of laser irradiation on sperm DNA integrity through Sperm Chromatin Structure Assay (SCSA) and Sperm Chromatin Dispersion (SCD) tests to ensure the safety of the treatment. ### Research background - **Infertility problems**: Approximately one - sixth of couples worldwide face infertility problems, and about half of them are caused by male factors. Common male infertility causes include low sperm count, abnormal sperm morphology and decreased sperm motility. - **Mitochondrial function**: The mid - piece of sperm contains mitochondria, which are the main source of energy for flagellar movement. Therefore, stimulating mitochondria can improve sperm motility. - **Photobiomodulation**: Low - level laser photobiomodulation (PBM) can improve sperm motility and speed by stimulating cytochrome c oxidase (Ccox) in the mitochondrial respiratory chain. ### Research methods - **Sample collection**: The research conducted experiments on 30 fresh human semen samples in the laboratory of Zahraa Hospital, Cairo University. - **Laser irradiation**: The samples were respectively exposed to red - light laser at 650 nm, near - infrared laser at 980 nm and their combination, and the irradiation time was 1, 2, 3, 4, 5 minutes respectively. - **Evaluation indicators**: The sperm motility was evaluated using the method recommended by WHO 2021 manual, and the sperm DNA integrity was evaluated through SCD test. ### Main results - **Significantly improved sperm motility**: After 3 minutes of irradiation with 650 nm laser, 980 nm laser and combined laser, the total motility increased by approximately 70%, 80% and 100% respectively. - **No significant impact on DNA integrity**: The results of SCD test showed that none of the three laser irradiations had a significant impact on sperm DNA integrity. ### Conclusion The research results show that irradiation with red - light laser, near - infrared laser and their combination can significantly improve the motility of human sperm without causing negative impacts on sperm DNA integrity. This provides a new non - invasive method for treating male infertility.
