Abstract:Objective To study the effects and mechanism of Kang Fuyan on inflammation and fertility improvement in mouse models with Chlamydia trachomatisgenital tract infection.Methods The mice were randomly divided into blank group,model group,Azithromycin group and Kang Fuyan goups in high,middle and low dosage respectively.Medicine was used to treat model mice for 1/3/5 weeks.Utems tissue was taken to analyze CD1 lb and MPO expression level.Fertility was observed for 9 weeks continuously after medicine treatment.Results The uterus swelling and hydromelia were observed in model mice and relieved after medicine treatment.Compared with blank group,the expression level of CD1 lb and MPO were significantly higher in model group (P ＜ 0.05).Compared with model group,the expression level of CD 11 b and MPO were significant reduced (P＜0.05) in Kang Fuyan group.The blank group had the shortest birth time.Kang Fuyan goup could shorten the birth time,but had no significant difference compared with model group.The infertility rate of model group came up to 12.5％.Compared with model group,Reproductive number of Kang Fuyan goup significantly increased (P＜0.05).Conclusions Kang Fuyan could promote the recovery of uterus morphology and improve fertility.The mechanism may be associated with inhibiting the expression of PMN CD1 lb and MPO,reducing adhesion and infiltration of neutrophils in tissues and cells,so as to weaken the PMN inflammatory reaction after pathogen infection and relieve infected tissues.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Clinopodium chinense (Benth.) O. Kuntze (C. chinense) is a Chinese herbal medicine used in treating gynecological hemorrhagic diseases for hundreds of years. Flavonoids are one kind of the major components in C. chinense. The flavonoids of C. chinense (TFC) have a vital role in treating endometritis but the underlying therapeutic mechanisms of TFC against endometritis have been rarely reported.AIM OF THE STUDY: To elucidate the therapeutic effect and possible mechanisms of TFC against lipopolysaccharide (LPS)-induced endometritis in vivo and LPS-induced primary mouse endometrial epithelial cells (MEECs) injury in vitro.MATERIALS AND METHODS: The holistic phytochemicals of the TFC and TFC-contained serum were screened and identified using UPLC-Q-TOF-MS. The model of endometritis was established by intrauterine injection of LPS (5 mg/mL) into female BALB/c mice, and the model mice were treated with TFC for 7 days. The value of MPO was measured by Myeloperoxidase assay kit, the pathological changes in the endometrium were evaluated using H&E staining and transmission electron microscope (TEM), the secretions of IL-18, IL-1β and TNF-α were determined by ELISA kits, the mRNA expressions of IL-18, IL-1β and TNF-α were determined by RT-PCR assay, and the protein levels of TLR4, IKBα, p-IKBα, p65, p-p65, caspase-1, ASC, NLRP3 and GSDMD were measured by Western blot. Subsequently, MEECs were isolated from the uterus of pregnant female mice, injured by LPS for 24 h and incubated with the TFC-contained serum. Finally, cell viability, LDH release, hoechst 33342/PI staining, immunofluorescence staining, scanning electron microscope observation, ELISA assay, RT-PCR detection and Western blot analysis were carried out to further validate the therapeutic effect and the underlying mechanisms of TFC.RESULTS: A total of 6 compounds in the plasma of mice after being intragastric administrated of TFC were identified. The results in vivo showed that TFC significantly reduced MPO value and alleviated pathological injury of the endometrium. Furthermore, TFC significantly decreased the serum IL-18, IL-1β and TNF-α levels, and the mRNA levels of IL-18, IL-1β and TNF-α. TFC also inhibited the expressions of TLR4, p-IKBα, p-p65, caspase-1, ASC, NLRP3 and GSDMD. Besides, compared with the model group in MEECs cells, TFC-contained serum prevented pyroptosis, decreased the levels of IL-18 and IL-1β, and inhibited the mRNA expressions of IL-18, IL-1β and GSDMD. TFC-contained serum also reversed the activation of NLRP3 inflammasome caused by nigericin, and restrainted the translocation of NF-κB into nuclear.CONCLUSIONS: TFC protects mice endometritis from the injury of LPS via suppressing the activation of NLRP3 inflammasome and pyroptosis, the underlying mechanisms of which were related to restraining the TLR4/NF-κB/NLRP3 pathway activation.

Activity of Neutrophils Infected by Chlamydia Trachomatis is Modulated by Kang Fuyan Capsule

Abstract:Objective To explore the effects and mechanisms of Kang Fuyan capsule for the treatment of inflammation through observing the activity of polymorphonuclear neutrophils (PMN) infected by Chlamydia trachomatis (CT). Methods Rat peripheral blood PMN was separated and cultured. After infecting CT for 6 h, PMN was treated with Kang Fuyan capsule [10 ng/mL (K-10 ng group), 10 μg/mL (K-10 μg group), 1 mg/mL (K-1 mg group)] for 6 h, 12 h and 24 h. Cells and cell supernatant were collected to analyze reactive oxygen species (ROS), myeloperoxidase (MPO) and CT loads. Molecular mechanisms of Kang Fuyan capsule affecting the activity of PMN in inflammatory response caused by CT were researched by TLR2/4 agonist or inhibitors. Results PMN intracellular ROS level was increased significantly (P<0.001) after CT infecting PMN. Kang Fuyan capsule significantly inhibited the production of ROS (K-1 mg: 6 h P=0.000 03, 12 h P=0.000 001, 24 h P=0.000 003; K-10 μg: 6 h P=0.014, 12 h P=0.009, 24 h P=0.009; K-10 ng: 24 h P=0.000 5) and MPO (K-1 mg: 6 h P=0.046, 12 h P=0.004, 24 h P=0.038; K-10 μg: 12 h P=0.048, 24 h P=0.011; K-10 ng: 12 h P=0.005, 24 h P=0.017) and inhibited the CT load of cell culture medium. Compared with Kang Fuyan capsule group, TLR4 inhibitors and Kang Fuyan capsule acting on cells at the same time significantly inhibit the secretion of MPO (P=0.011); TLR2/4 agonist and Kang Fuyan capsule acting on cells at the same time significantly decreased the CT load in cell supernatant (PGN: P=0.002; LPS: P=0.000 08). Conclusion Kang Fuyan capsule regulates the sterilization ways of PMN by suppressing the production and secretion of ROS and MPO, and promoting PMN phagocytosis. But the molecular mechanisms of Kang Fuyan capsule to PMN had nothing to do with TLR2/4 and still need to be studied. Abstract:2-Deoxy-D-glucose (2-DG) was injected intravenously (300 mg/kg) into 6 conscious monkeys adapted to chronic restraint in primate chairs. In 10 experiments, chow and water were provided to the monkeys after the injection of 2-DG. In 16 experiments, chow and water were withheld. Feeding did not affect themarked hyperglycemia or the increase in plasma growth hormone or 17-hydroxycorticosteroids elicited by 2-DG, but feeding markedly increased plasma immunoreactive insulin (IRI) concentrations, probably through the release of enteric stimulant(s) of insulin secretion. Abstract:Endometriosis remains a common but challenging gynecological disease among reproductive-aged women with an unclear pathogenesis and limited therapeutic options.Numerous pieces of evidence suggest that NF-κB signaling, a major regulator of inflammatory responses, is overactive in endometriotic lesions and contributes to the onset, progression, and recurrence of endometriosis.Several factors, such as estrogen, progesterone, oxidative stress, and noncoding RNAs, can regulate NF-κB signaling in endometriosis.In the present review, we discuss the mechanisms by which these factors regulate NF-κB during endometriosis progression and provide an update on the role of NF-κB in affecting endometriotic cells, peritoneal macrophages (PMs) as well as endometriosis-related symptoms, such as pain and infertility.Furthermore, the preclinical drugs for blocking NF-κB signaling in endometriosis are summarized, including plant-derived medicines, NF-κB inhibitors, other known drugs, and the potential anti-NF-κB drugs predicted through the Drug-Gene Interaction Database.The present review discusses most of the studies concerning the multifaceted role of NF-κB signaling in endometriosis and provides a summary of NF-κB-targeted treatment in detail.
Fisetin Ameliorates the Inflammation and Oxidative Stress in Lipopolysaccharide-Induced Endometritis

Abstract:Purpose: Fisetin is a natural flavone of polyphenol, which widely exists in many fruits and vegetables and has many pharmacological activities. However, the mechanism involved remains largely unknown. Here, we investigate the mechanisms of fisetin on the inflammatory response and oxidative stress in LPS-induced endometritis model and bovine endometrial epithelial cell line (BEND). Methods: The function of fisetin was analyzed by network pharmacology. Effects of increasing doses of fisetin on inflammation and oxidative stress are studied in BALB/c mice with LPS-induced endometritis. The underlying mechanisms of antioxidant activity of fisetin were further explored in LPS-stimulated BEND cells. Results: The results showed that fisetin significantly alleviated LPS-induced inflammatory injury and oxidative stress both in vivo and in vitro. Further studies found that fisetin greatly inhibited the LPS stimulated TLR4 expression and nuclear translocation of nuclear factor-KB (NF-KB), thus reducing the pro-inflammatory mediators secretion. Silencing TLR4 reduced LPS-induced inflammatory responses. Moreover, we observed that fisetin evidently decreased ROS production but activated Nrf2/HO-1 pathway in LPS-stimulated BEND cells. To further explore the role of Nrf2 in fisetin-induced HO-1 protein expression, the specific siRNA was used to silence Nrf2 expression. Silencing Nrf2 abrogated the inhibitory effects of fisetin on LPS-induced pro-inflammatory cytokines TNF-alpha, IL-1 beta secretion, NADPH oxidase-4 (Nox4) and ROS production. Conclusion: In conclusion, fisetin effectively protected against LPS-induced oxidative stress and inflammatory responses which may be closely correlated to inhibition of TLR4-mediated ROS/NF-KB and activation of the Nrf2/HO-1 pathway.

Abstract:Endometritis is a serious reproductive disease in mammals that commonly results in reproductive loss and even permanent infertility. Kynurenic acid (KYNA) is the main bioactive metabolite of tryptophan degradation and exhibits neuroprotective and anticonvulsant properties. However, little is known about the role of KYNA in achieving endometritis remission. This study investigated the protective effects and mechanisms of KYNA using a mouse model of against lipopolysaccharide (LPS)-induced endometritis. The endometritis model was induced by an intrauterine injection of LPS, and KYNA was intraperitoneally injected before and two hours after LPS treatment. Twenty-four hours after LPS administration, pathological changes in uterine tissues were observed by hematoxylin- and eosin (H&E) staining. The levels of the inflammatory factors, TNF-α and IL-1β, were measured by ELISA. The myeloperoxidase (MPO) activity in uterine tissues was detected using MPO kits and immunohistochemistry. Furthermore, the expression of signaling pathway proteins and tight junction proteins occludin and ZO-1 in uterine tissues was detected by western blot. KYNA prominently inhibited uterine pathological injury and neutrophil infiltration and restricted the secretion of TNF-α and IL-1β in the uteri of subjects with endometritis. Furthermore, KYNA upregulated the levels of the tight junction proteins (TJPs)occludin and ZO-1 in the uterus. In vitro, KYNA inhibited LPS-induced TNF-α and IL-1β production, and NF-κB activation in mouse endometrial epithelial cells (mEECs). In addition, KYNA increased the expression of G protein-coupled receptor 35 (GPR35) and inhibition of GPR35 reversed the anti-inflammatory effects of KYNA. In conclusion, KYNA protected against LPS-induced endometritis by maintaining epithelial barrier permeability and suppressing proinflammatory responses via the GRP35/NF-κB signaling pathway.

Abstract:Endometritis is one of the main diseases that causes great economic losses in the dairy industry. Recent studies have shown that matrine extracted from the traditional Chinese herb Sophora flavescens is an alkaloid with a broad range of bioactivities. Here, we aimed to investigate the protective effects of matrine on Staphylococcus aureus lipoteichoic acid (LTA)-induced endometritis in mice and elucidate the possible molecular mechanisms in vitro. Histopathological changes showed that matrine remarkably attenuated the uterus injury in a mouse model of LTA-induced endometritis. qPCR and ELISA results showed that matrine dose-dependently reduced the expression of pro-inflammatory cytokines (TNF-α and IL-1β). To further elucidate the underlying mechanisms of this protective effect of matrine, LTA-stimulated bovine endometrial epithelial cells (bEECs) were employed in this study. The results demonstrated that TLR2 expression and its downstream nuclear factor (NF)-κB activation were both suppressed by matrine treatment. Furthermore, a small interference RNA targeting TLR2 gene mimicked matrine in its inhibition on LTA-induced activation of TLR2 and NF-κB. In conclusion, these findings suggest the protective effect of matrine against LTA-induced endometritis through negative regulation of TLR2-mediated NF-κB pathway.
Guizhi Fuling Capsule ameliorates endometrial hyperplasia through promoting p62-Keap1-NRF2-mediated ferroptosis

Abstract:Ethnopharmacological relevance: Guizhi Fuling Capsule (GFC) is a classical traditional Chinese medicine officially recorded in Synopsis of the Golden Chamber and has long been used to treat gynecological diseases in China. However, scientific evidence for the anti-endometrial hyperplasia potential of GFC used in traditional medicine is lacking. Aim of the study: This study evaluated whether GFC protects against endometrial hyperplasia and its potential mechanism in mice. Methods and materials: We used estrogen (estradiol) to induce endometrial hyperplasia in mice. C57BL/6 mice were treated with estradiol subcutaneously for 21 days, and GFC (75 mg/kg and 150 mg/kg) was given intragastric administration from the first day of the modeling. H&E staining is used to evaluate endometrial tissue structure change. Malondialdehyde was measured to explore lipid peroxidation. Western blot, immunohistochemistry and immunofluorescence were performed to observe the expressions of GPX4, p62, Keap1 and NRF2. Results: The degree of ferroptosis in endometrial tissue of patients with endometrial hyperplasia was lower than normal endometrial tissue. In addition, ferroptosis inducer imidazole ketone erastin could improve endometrial hyperplasia in mice. Interestingly, GFC significantly alleviated endometrial hyperplasia through triggering ferroptosis. Furthermore, GFC inhibited p62-Keap1-NRF2 pathway in estradiol-induced endometrial hyperplasia model. Conclusions: GFC may attenuate estrogen-induced endometrial hyperplasia in mice through triggering ferroptosis via inhibiting p62-Keap1-NRF2 pathway. These findings suggest that GFC might act as a promising traditional Chinese medicine to treat endometrial hyperplasia.
Engeletin Alleviates Lipopolysaccharide-Induced Endometritis in Mice by Inhibiting TLR4-mediated NF-κB Activation.

Abstract:Engeletin (dihydrokaempferol 3-rhamnoside) is a flavanonol glycoside. It can be found in the skin of white grapes and white wine and is widely distributed in southeast Asia, and the leaves are used in a tea. Here, we explored the impact of engeletin against the inflammatory reaction in a lipopolysaccharide (LPS)-induced endometritis mouse model. Engeletin treatment significantly attenuated uterus damage and decreased myeloperoxidase activity. ELISA and qPCR assays showed that engeletin dose-dependently suppressed the expression of TNF-α, IL-1β, and IL-6. Molecular studies also demonstrated that the levels of iNOS, COX-2, and TLR4, along with their downstream molecules MyD88, IRAK1, TRAF6, and TAK1, were also suppressed by engeletin. In addition, engeletin treatment inhibited NF-κB signaling-pathway activation. Moreover, immunofluorescence analysis demonstrated that engeletin suppressed NF-κB-p65 nuclear translocation. These data indicated the protective action of engeletin against LPS-stimulated endometritis in mice via negative regulation of pro-inflammatory mediators via the TLR4-regulated NF-κB pathway.
IFN-τ Plays an Anti-Inflammatory Role in Staphylococcus aureus-Induced Endometritis in Mice Through the Suppression of NF-κB Pathway and MMP9 Expression.

Abstract:Interferon-tau (IFN-tau) is a type I interferon and considered as a pregnancy recognition signal in ruminants. Our previous reports have confirmed that IFN-tau has a potential anti-inflammatory effect in macrophage. However, the anti-inflammatory effect of IFN-tau on endometritis has never been reported. Thus, the aim of this study was to investigate the effects of IFN-tau in a mouse model of Staphylococcus aureus-induced endometritis. The histopathological and myeloperoxidase activity results showed that IFN-tau could protect the uterus from S. aureus damage. Enzyme-linked immunosorbent assay and quantitative real-time polymerase chain reaction results revealed that IFN-tau inhibited TNF-alpha, IL-1 beta, and IL-6 production. TLR2, involved in the S. aureus infection, was downregulated by IFN-tau and directly activated nuclear transcription factor kappa-B (NF-kappa B) pathway. Then, we measured the phosphorylation of I kappa B alpha and NF-kappa B p65 by Western blotting. Western blotting results indicated that IFN-tau inhibited the phosphorylation of I kappa B alpha and NF-kappa B p65 in the S. aureus-induced endometritis. Matrix metalloproteinase (MMP)9, which has been reported to be regulated by NF-kappa B, was also suppressed by IFN-tau, but its inhibitors, tissue inhibitor of metalloproteinases1 level, increased. All of these findings suggested that IFN-tau plays an anti-inflammatory role in S. aureus-induced endometritis by suppressing NF-kappa B pathway and MMP9 expression.
MicroRNA Let-7C Improves LPS-Induced Outcomes of Endometritis by Suppressing NF-κB Signaling

Abstract:Endometritis is a common inflammatory disease which endangers human and animal reproductive health. MicroRNA (miRNA) let-7c plays an important role in the inflammatory process; however, the regulatory underlying mechanism of let-7c in endometritis is unclear. In this study, we confirmed that let-7c was significantly reduced in LPS-induced mouse endometritis model, and overexpression of let-7c was able to effectively reduce uterine tissue damage caused by lipopolysaccharide (LPS), and then, a LPS-induced bovine endometrial epithelial cell (BEND) line was used to mimic the inflammatory model in vitro . Our data showed that overexpression of let-7c significantly reduced the expression of pro-inflammatory cytokines in BEND cells induced by LPS. Meanwhile, immunofluorescence and western blotting results showed that let-7c significantly inhibited LPS-induced phosphorylation of NF-κB, thereby inhibiting downstream pro-inflammatory cytokine expression. Taken together, our results suggested that let-7c ameliorates LPS-induced endometritis by attenuating the expression of pro-inflammatory cytokines via inhibition of the activation of NF-κB.
Curative Effect of Kangfuyan Capsule Combined with Antibiotic Treatment on Pelvic Inflammatory Disease.

Abstract:This study aims to investigate the curative effect of Kangfuyan capsule in the treatment of damp-heat and blood stasis type of pelvic inflammatory disease (PID), and its influence on serum inflammatory factors IL-6, CRP and TNF-α. A total of 83 patients with PID were randomly divided into two groups: Western medicine group (control group, n=41) received oral antibiotics (azithromycin + metronidazole) alone and the traditional Chinese medicine combined with Western medicine group (experimental group, n=42) received Kangfuyan capsule based on Western medicine therapy. Clinical efficacy between these two groups and the influence of drugs in serum inflammatory factors (IL-6, CRP and TNF-α) were compared. The total effective rate was 78.05% in the control group and 97.62% in the experimental group and difference between these two groups was statistically significant (P<0.01). The symptoms and signs in the two groups significantly improved after treatment (P<0.05) and improvement rate was significantly better in the experimental group than in the control group (P<0.05). After treatment, serum inflammatory factor levels in the two groups were significantly lower than levels before treatment (P<0.05) and improvement rate was significantly better in the experimental group than in the control group (P<0.05). Kangfuyan capsule combined with antibiotics can effectively relieve the symptoms and signs of patients, improve the efficiency of treatment, provide high safety, and does not increase adverse reactions. The possible mechanism may be that this therapy suppresses chronic PID by reducing serum inflammatory factor (IL-6, CRP and TNF-α) levels.

Abstract:The protective effects of epigallocatechin‐3‐gallate (EGCG) on lipopolysaccharide (LPS)‐induced endometritis in vivo and in vitro will be explored in this study. The endometritis model was induced in female BALB/c mice uterus by perfusion with lipopolysaccharide (LPS) and EGCG were administered at 1 h before LPS induction. The primary bovine endometrial epithelial cells (BEECs) were treated with EGCG for 1 h before LPS stimulation. Uterine histopathological changes, myeloperoxidase (MPO) activity, inflammatory cytokine levels and oxidative stress markers were determined. The extent of Bax, Bcl‐2, cleaved caspase‐3, silent information regulator transcript‐1 (SIRT1), nucleotide oligomerization domain (NOD)‐like receptor pyrin domain‐containing 3 (NLRP3), apoptosis‐associated speck‐like protein (ASC) and Caspase1 was detected by Western blot and real‐time quantitative PCR assays. The results showed that EGCG significantly reversed the LPS‐induced uterine histopathological changes, MPO activity, pro‐inflammatory cytokine levels. Additionally, EGCG decreased oxidative stress and reduced cell apoptosis by upregulating SIRT1 expression, downregulating the NLRP3 inflammasome activation. These findings indicated that EGCG exerted its greatest protective effects by blocking inflammatory responses, lowering oxidative stress, and reducing apoptosis via the SIRT1/NLRP3, making its promising candidate treatment for endometritis.

Abstract:OBJECTIVE(S): Endometritis is the inflammatory response of the uterine lining which is linked to infertility. Administration of platelet-rich plasma (PRP) represents a well-recommended strategy for the treatment of endometrium-associated infertility. In this study, we set to characterize the role and molecular mechanism of PRP intrauterine infusion in mice with endometritis.METHODS: A mouse model of endometritis was established using lipopolysaccharide (LPS). Mouse endometrial epithelial cells were obtained in primary culture. PRP-treated cells were assayed for proliferative and apoptotic activities. Moreover, iNOS expression and chemokine and inflammatory factor contents in cells were assessed using RT-qPCR and ELISA. The mice were subjected to PRP intrauterine infusion. The expression of genes related to uterine development was analyzed by qPCR and the ki-67 content and caspase-3 activation in endometrial tissues were examined by immunohistochemistry. Finally, the Nrf2/HO-1 pathway activity in tissues was examined by Western blot.RESULTS: LPS induced inflammatory cell recruitment and tissue damage in the endometrium of mice, along with significantly increased levels of inflammatory and chemokine factors. PRP significantly enhanced endometrial epithelial cell activity, decreased apoptosis, and reduced inflammatory factor secretion. In addition, PRP intrauterine infusion significantly increased the expression of genes related to uterine development, promoted tissue proliferation, decreased apoptosis, and diminished inflammatory response in endometrial tissues of mice. PRP intrauterine infusion significantly elevated Nrf2/HO-1 pathway activity in endometrial epithelial cells and tissues.CONCLUSION: PRP intrauterine infusion significantly inhibited endometrial cell injury and alleviated the inflammatory response through activating the Nrf2/HO-1 pathway.
SCM-198 Prevents Endometritis by Interrupting the CCL2-Mediated Endometrial Stromal Cell-Macrophage Interaction Loop

Abstract:Background: Endometritis is an inflammatory disease of the endometrium that is closely associated with endometrial dysfunction, a higher incidence of recurrent pregnancy loss, and an increased probability of infertility. The disordered communication induced by infections, stress, or surgical injuries may destruct the healthy endometrial milieu, leading to endometritis. SCM-198, an alkaloid active component of Leonurus japonicus Houtt, possesses various biological activities, especially anti-inflammatory effects. However, the molecular mechanism of SCM-198 in preventing endometritis has not been completely elucidated. Methods: In vitro, we evaluated the inflammatory level of LPS-stimulated human endometrial stromal cells (hESCs), macrophages or their coculture after SCM-198 treatment, via RT-PCR, ELISA, Western blot and Flow cytometry. In vivo, endometritis C57BL/6J mice stimulated by LPS were vaginally or intraperitoneally injected with SCM-198 and the uterus tissues were studied compared to untreated mice. Findings: We found that SCM-198 could prevent lipopolysaccharide (LPS)-induced endometrial inflammation in hESCs and a mouse model. SCM-198 directly attenuated LPS-induced inflammation via TLR-4/NF-κB signaling in hESCs. SCM-198 also efficiently decreased CCL2 production by endometrial stromal cells (ESCs), which inhibited the endometrial infiltration of monocytes and promoted local macrophage switching to M2 via a CCL2/CCR2 interaction. Polarized M2 macrophages protected ESCs against inflammatory damage, thereby reinforcing the therapeutic effect of SCM-198. Interpretation: Our study is the first to propose the concept that dysregulated communication between ESCs and macrophages via CCL2/CCR2 axis boosting local macrophage enrichment and proinflammation amplification is a critical mechanism of the occurrence of endometritis. SCM-198 can inhibit CCL2 production in ESCs and interrupt the CCL2-mediated ESC-macrophage interaction loop. It also provided a new entry point for the prevention and treatment of endometritis in clinical practice. Leonurine Ameliorates the Inflammatory Responses in Lipopolysaccharide-Induced Endometritis.

Abstract:Endometritis is the inflammation of the endometrium that is associated with lower conception rates, increased intervals from calving to first service, and more culls for failure to conceive, which leads to serious economic losses in the dairy industry. Leonurine, a natural active compound of Leonurus cardiaca, has been proved to possess various biological activities. However, there is still no study about its anti-inflammatory effects on LPS-induced endometritis. The present study aimed to demonstrate the underlying mechanism responsible for the anti-inflammatory effects of leonurine on LPS induced endometritis in mice and in bovine endometrial epithelial cells (bEECs). The results of pathological section displayed that leonurine alleviated LPS induced uterine injury. qRT-PCR and ELISA experiments suggested that leonurine inhibited the expression levels of TNF-α and IL-1β in uterus tissues and bEECs. Molecular studies showed that TLR4 expression and nuclear factor (NF)-κB activation were both inhibited by leonurine treatment. These results suggested that the therapeutic effects of leonurine on LPS-induced endometritis in mice and bEECs may act by inhibiting the expression of TLR4 and its downstream mediated NF-κB pathway. Accordingly, leonurine may serve as an effective drug in preventing and treating LPS induced endometritis.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacological relevance</h3><p>Ermiao fang (EMF) is a traditional Chinese medicinal herbal formula from ancient times and recorded in the pharmacopeia of the People's Republic of China. It is composed of two typical Chinese herbal medicines, <em>Cortex Phellodendri</em> (Huangbai), the bark of <em>Phellodendron chinensis Schneid.</em> (Rutaceae), and <em>Rhizoma Atractylodis</em> (Cangzhu), the rhizome of <em>Atractylodes lancea</em> (Thunb.) DC. (Compositae). EMF has been clinically used for the treatment of endometritis for many years in China.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Aim of the study</h3><p>This study was aimed to identify the active ingredients, potential targets, and mechanism of action of EMF for the treatment of endometritis.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3><p>In this research, the pharmacological effects of EMF on endometritis were first evaluated by establishing a rat model of endometritis. A network pharmacology-based analytical strategy was then used to predict its targets and signaling pathways. An endometritis-related protein target and compound database was built for EMF. The compounds in EMF and those absorbed into the blood were identified by ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF/MS). High-throughput virtual screening and molecule docking methods were used to predict the protein targets of EMF. The surface plasmon resonance analysis (SPR) method was used to validate the affinity between the compound and proteins. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis was used to predict the related pathways. Western blotting analysis was used to evaluate the expression of key proteins in the related pathways.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The animal study showed that EMF could reduce uterine inflammation in rats with endometritis. Then, an ingredient database including 187 compounds and a protein target database including 836 proteins were constructed. Twenty-four compounds in EMF were identified by UHPLC-Q-TOF/MS, among which eight compounds were present in rat plasma after an oral administration of EMF. Afterward, 39 potential target proteins were predicted by the high-throughput screening method, and 20 of them were selected after further screening using molecular docking. Subsequently, an ingredient-target network was constructed, and the target proteins were classified into the NF-κB and MAPK signal pathways by KEGG pathway enrichment analysis. Finally, the affinity between the active ingredients and the target proteins was verified by SPR. The Western blotting analysis showed that EMF significantly inhibited the elevated NF-κB and MAPK pathway proteins in rats with endometritis.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3><p>EMF exhibited a significant pharmacological effect on rats with endometritis. Network pharmacology analysis revealed that eight compounds were absorbed into the blood after oral administration and interacted with 20 targets. Western blotting analysis indicated that EMF exerted anti-inflammatory effects by inhibiting the NF-κB and MAPK signaling pathway proteins in the treatment of endometritis.</p>

Abstract:Objective To study the effect of kangfuyan capsule on the rabbit model of sequelae of pelvic inflammatory disease(PID).Methods 19 PID rabbit models were divided into four groups:KC group was given kangfuyan capsule treatment alone(four rabbits),AT group was treated with antibiotic(five rabbits),AK group was treated with antibiotic and kangfuyan capsule(five rabbits)and control group(non-intervention).The blood routine,body weight,levels of TNF-a and TGF-β1 were determined.Laparoscopic biopsy of the uterine and tube applied regularly to evaluate the effects.Results Kangfuyan capsule alone could inhibit inflammation but could not fully cure the acute pelvic inflammatory disease.Antibiotic treatment achieved satisfied effect in acute PID episode but not in chronic phase.Antibiotic plus Chinese medicine kangfuyan capsule improved the treatment effect significantly both in acute and chronic phase of PID.Conclusion Kangfuyan capsule has antibacterial effect during the acute phase,when given with antibiotic,it can improve the treatment effect of PID.
