Abstract:There are numerous past and current examples of ontology-driven projects that provide auto-generated user interfaces for managing entities and relations, each presenting its own varied and complex data model. Our Datum Proof Sheet application aims to simplify the application development landscape by building community consensus about the way basic categorical, textual and numeric datum fields should be described within the OBOFoundry community of ontologies. The proof sheet shows selected datums (grouped under the context of an OBI “data representational model” item) as form inputs on an HTML page, enabling an application ontology’s contents to be presented to end users (ranging in our case from epidemiologists to software developers) for review without necessarily having a working application to showcase them in. The basic relations and cases necessary for presenting datums in a user interface are mostly satisfied by OBI’s design, but we introduce a few extra elements to bring more clarity to datum specifications, and to provide user interface term labels and definitions that may differ from those that ontologists prefer in the “backend”. Abstract:Costs associated with subterranean termite damage and control are estimated to approach $2 billion annually in the United States alone. The Formosan subterranean termite, Coptotermes formosanus Shiraki, is one of the more economically important subterranean species. In recent years, the shortcomings associated with conventional chemical control methods have prompted policymakers and scientists to evaluate the potential for biological control of subterranean termites (C. formosanus in particular), that is, to determine the potential for natural enemies - predators, parasitoids and pathogens - to suppress termite populations. Ants are the greatest predators of termites, and may have a considerable local impact on termite populations in some areas of the world. A few parasitoids of termites are known, but their potential for regulating termite populations seems negligible. Characteristics of the colony, such as a protected, underground location (and, for the C. formosanus nest, its modular and dispersed nature), are likely to limit the impact predators and parasitoids have on subterranean termites. Thus, there seems little potential for use of these agents for subterranean termite control. For various reasons, pathogenic organisms, such as viruses, bacteria, Protozoa, nematodes and most fungi, have shown little promise for use in biological termite control. However, research suggests that strains of two well-studied, endoparasitic fungi, Beauveria bassiana and Metarhizium anisopliae, when employed in baiting schemes, may offer the potential for at least some measure of subterranean termite control, although their successful use is compromised by a number of inherent biological limitations and logistical problems that have yet to be solved. Although not strictly in the realm of classical biological control, recent studies suggest that natural products, such as ant semiochemicals and fungal metabolites (siderophores), or their synthetic analogues, eventually might find a use in termite control programmes as repellents or insecticides in wood treatments or soil applications if stable formulations can be developed.

Diversifying evolution of endo-beta-1,4-glucanase in termites (Isoptera)

Abstract:Termites are important decomposers of lignocellulose in tropical and subtropical ecosystems. The endo-beta-1,4-glucanase of termites is important for termites to decompose lignocellulose. How they evolve higher cellulose-decomposition ability remains unclear. In this paper, we investigated the adaptive evolution of termite endo-beta-1,4-glucanase using the maximum likelihood method. Phylogenetic analysis shows that C. formosanus may be the intermediate of termite evolution from lower to higher. Extensive gene duplication events were found in lower termite endo-beta-1,4-glucanase genes. In particular, MdCe12 may have originated from MdCell by relatively recent insertion of two amino acids that may influence its cellulose-decomposition ability. The adaptive evolution analyses show that endo-beta-1,4-glucanase of termites undergoes positive selection and two positively selected sites (161K and 302S) were identified in this enzyme. The two sites are predicted to be located on random coils and beta turns and exposed on the surface of the protein, suggesting that they may be involved in the proper folding and enhancing activity of endo-beta-1,4-glucanase. The findings of extensive gene duplication occurring and positive selection acting on termite endo-beta-1,4-g1ucanases suggest that endo-beta-1,4-glucanases play more important roles in the utilization of cellulosic biomass in termites than those from symbiotic microorganisms.

Abstract:The global economic impact of termites is estimated to be approximately USD 40 billion annually, and subterranean termites are responsible for about 80% of the total impact. Twenty-eight species of termites have been described as invasive, and these termites are spreading, partially due to global trade, making effective control methods essential. Termite control is complex, as is the biology and behavior of this social insect group. In the U.S., termite prevention and control (with claims of structural protection) is regulated by more than one industry (pest control and building construction), and at the federal and state levels. Termite prevention has historically relied on building construction practices that do not create conducive conditions for termite infestations, but as soil termiticides developed, heavy reliance on pesticides became the standard for termite control. The concern for human and environmental health has driven the development of termite control alternatives and regulation for products claiming structural protection. Product development has also provided unprecedented opportunities to study the biology and behavior of cryptobiotic termites. Technological advances have allowed for the re-examination of questions about termite behavior. Advances in communications via social media provide unrestricted access to information, creating a conundrum for consumers and science educators alike.

Abstract:The success of microbial termiticides in controlling termites depends on the ability of microbes to grow in different media and the functionality of the microbes as a resistant barrier or toxic bait. This study was conducted to understand the mortality rate and behaviour changes of the subterranean termite Coptotermes curvignathus Holmgren introduced with different concentrations of Serratia marcescens strain LGMS 1 and Pseudomonas aeruginosa strain LGMS 3 using wood and soil as bacterial transfer medium. In general, higher concentration of bacteria in soil caused a reduction in tunnelling activity and wood consumption and an increase in mortality. However, application on wood revealed a different outcome. Wood treated with S. marcescens of 10 6 CFU ml −1 concentration proved to be more efficient as bait than higher concentration applications as it caused a high mortality rate while still highly palatable for termites. Wood or soil treated with S. marcescens concentration higher than 10 9 CFU ml −1 creates a high toxicity and repellent barrier for termites. Pseudomonas aeruginosa of 10 9 CFU ml −1 concentrations applied on wood served as a slow-acting toxic bait. However, the ability for S. marcescens and P. aeruginosa to survive on wood is low, which made the bait unable to retain a useful level of toxicity for a long period of time and frequent reapplication is needed.

Abstract:Collective behaviors of social insects are often regulated by pheromones. In subterranean termites, some workers forage for and exploit decaying wood for new food resources while forming tunnels from their nest. Colonizing new food resources requires workers to build and disinfect tunnels and chambers inside the nest and ingest decaying wood; therefore subterranean termite colonies should have mechanisms to establish and maintain groups of workers to perform these functions. Recently, an aggregation pheromone was identified in workers of the termite Reticulitermes speratus , which induces quick attraction of nestmate workers and prolonged aggregation to the site of attraction. In this study, we extended this work to another species of Reticulitermes and identified a worker aggregation pheromone in the termite R. virginicus . GC–MS analysis and bioassays demonstrated that this pheromone consists of 3-octanone, 3-octanol, and palmitic acid and shows a colony-specific, dose-dependent attractant response but not an arrestant response. Furthermore, these pheromone components were most likely emitted from the surface of the body. This suggests that aggregation pheromone composition and function differ significantly among termite species, even within the same genus. This study advances our understanding of the regulatory mechanisms of termite aggregation behavior.

Abstract:Termites are a clade of eusocial wood-feeding roaches with > 3000 described species. Eusociality emerged ~ 150 million years ago in the ancestor of modern termites, which, since then, have acquired and sometimes lost a series of adaptive traits defining of their evolution. Termites primarily feed on wood, and digest cellulose in association with their obligatory nutritional mutualistic gut microbes. Recent advances in our understanding of termite phylogenetic relationships have served to provide a tentative timeline for the emergence of innovative traits and their consequences on the ecological success of termites. While all "lower" termites rely on cellulolytic protists to digest wood, "higher" termites (Termitidae), which comprise ~ 70% of termite species, do not rely on protists for digestion. The loss of protists in Termitidae was a critical evolutionary step that fostered the emergence of novel traits, resulting in a diversification of morphology, diets, and niches to an extent unattained by "lower" termites. However, the mechanisms that led to the initial loss of protists and the succession of events that took place in the termite gut remain speculative. In this review, we provide an overview of the key innovative traits acquired by termites during their evolution, which ultimately set the stage for the emergence of "higher" termites. We then discuss two hypotheses concerning the loss of protists in Termitidae, either through an externalization of the digestion or a dietary transition. Finally, we argue that many aspects of termite evolution remain speculative, as most termite biological diversity and evolutionary trajectories have yet to be explored.

Immobilizing Bifenthrin on Wood for Termite Control

Abstract:Termites are world-wide pests causing significant losses to annual and perennial crops, as well as damages to wooden components in buildings. Although various chemical, physical, and biological methods have been explored to prevent termite attack on wooden structures, new guiding principles are still needed for environmental protection. In this study, by combining the effective chemical control of bifenthrin and photo-immobilization technique of biomolecules, we developed chitosan as a carrier to embed bifenthrin, which was then immobilized by ultraviolet treatment on the surface of wood (Cunninghamia lanceolata). The immobilized bifenthrin embedded in the photoactive chitosan was characterized by Fourier transform infrared spectroscopy (FTIR), C.H.N analysis, ultraviolet, and fluorescence measurements. The surface structures and biological activity were examined by scanning electron microscopy (SEM), atomic force microscope (AFM), electron spectroscopy for chemical analysis (ESCA), and bioassays. The results indicated that the immobilized bifenthrin can be well protected from free and non-controlled releasing, and has a long-term stability allowing high efficiency against the termite at a dose of 2.5 mu g/cm(2). This study provides a novel and environmentally-benign technique for the termite control by photo-immobilizing the bifenthrin-embedded chitosan on the surface of C. lanceolata. A Novel Approach to Managing Invasive Termite Species Using Genetically Engineered Bacteria

Abstract:The hind gut of the Fonnosan subterranean tennite (FST) is a refuge for an array of protozoa and bacteria that fulfill important functions in the survival of their hosts. Due to their importance for FST survival these symbionts are excellent tools or targets for the control of FST. Lytic peptides were tested as protozoicides. Lytic peptides were shown to kill the tennite symbiotic protozoa and this defaunation was associated with the subsequent death of tennites. A prototype delivery system was developed using the yeast Kluyveromyces lactis and the lytic peptides Hecate and Melittin. Studies showed defaunation of tennite workers when fed with lytic peptide expressing yeast. Conditions to genetically engineer tennite-specific bacteria were investigated and the enzyme profile of the tennite gut was determined. These studies are crucial for development of a bacterial shuttle to produce toxins that are activated by enzymatic cleavage in the tennite gut and with further development, the system could be an environmentally friendly, cost effective alternative to chemical baiting systems presently used to control tennites.

Abstract:The familial Alzheimer's disease gene product amyloid β precursor protein (APP) is sequentially processed by β- and γ-secretases to generate the Aβ peptide. The biochemical pathway leading to Aβ formation has been extensively studied since extracellular aggregates of Aβ peptides are considered the culprit of Alzheimer's disease. Aside from its pathological relevance, the biological role of APP processing is unknown. Cleavage of APP by γ-secretase releases, together with Aβ, a COOH-terminalAPP intracellular domain, termed AID. This peptide has recently been identified in brain tissue of normal control and patients with sporadic Alzheimer's disease. We have previously shown that AID acts as a positive regulator of apoptosis. Nevertheless, the molecular mechanism by which AID regulates this process remains unknown. Hoping to gain clues about the function of APP, we used the yeast two-hybrid system to identify interaction between the AID region of APP and JNK-interacting protein-1 (JIP1). This molecular interaction is confirmed in vitro, in vivoby fluorescence resonance energy transfer (FRET), and in mouse brain lysates. These data provide a link between APP and its processing by γ-secretase, and stress kinase signaling pathways. These pathways are known regulators of apoptosis and may be involved in the pathogenesis of Alzheimer's disease.
Genome-wide identification of Coptotermes formosanus immune genes and their potential roles in termite control

Abstract:In recent years, the use of microbes to control termites has attracted increasing attention. It was found that pathogenic bacteria, nematodes, and fungi effectively control termites under laboratory conditions. However, their effects have not been replicated in the field, and one reason for this is the complex immune defense mechanisms of termites, which are mainly regulated by immune genes. Therefore, altering the expression of immune genes may have a positive influence on the biocontrol efficacy of termites. Coptotermes formosanus Shiraki is one of the most economically important termite pests worldwide. Currently, the large-scale identification of immune genes in C. formosanus is primarily based on cDNA library or transcriptome data rather than at the genomic level. In this study, we identified the immune genes of C. formosanus according to genome-wide analysis. In addition, our transcriptome analysis showed that immune genes were significantly downregulated when C. formosanus was exposed to the fungus Metarhizium anisopliae or nematodes. Finally, we found that injecting dsRNA to inhibit three immune genes (CfPGRP-SC1, CfSCRB3, and CfHemocytin), which recognize infectious microbes, significantly increased the lethal effect of M. anisopliae on termites. These immune genes show great potential for C. formosanus management based on RNAi. These results also increase the number of known immune genes in C. formosanus which will provide a more comprehensive insight into the molecular basis of immunity in termites.

Abstract:Oral toxicity of 4 insecticides, chlorpyrifos, fenvalerate, abamectin and fipronil to two termites, Coptotermes formosanus and Reticulitermes flaviceps, was tested using different baits material. Trail-following activity of the toxic baits to R. Flavicps was evaluated. The oral test showed that sugarcane was more attractive bait than that of starch and pinewood. Sugarcane bait decayed by Gloeophyllum trabeum was more attractive than the fresh one. Three of the 4 insecticides tested (except fenvalerate) maintained their attraction to the termites without losing the toxicity. A 30-day field-tests with chlorpyrifos bait gave a 66.7% rate of a termite population control on a dam.
Termite digestomes as a potential source of symbiotic microbiota for lignocelluloses degradation: a review

Abstract:Termites thrive in great abundance in terrestrial ecosystems and the symbiotic gut microbiota play important roles in digestion of lignocelluloses and nitrogen metabolism. Termites are excellent models of biocatalysts as they inhabit dense microbes in their guts that produce digestive enzymes to decompose lignocelluloses and convert it to end products such as sugars, hydrogen, and acetate. Different of digestive system between lower and higher termites which lower termites dependent on their dual decomposing system, consisting of termite's own cellulases and gut's protists. Higher termites decompose cellulose using their own enzymes, because of the absence of symbiotic protists. Termite gut prokaryotes efficiently support lignocelluloses degradation. In this review, a brief overview of recent experimental works, development and commercialization is discussed. Significant progress has been made to isolate cellulolytic strains from termites and optimise the digestion efficiency of cellulose. Future perspective should emphasize the isolation of cellulolytic strains from termites, genetically modifying or immobilization of the microbes which produce the desired enzyme and thus benefits on the microbiology and biotechnology.

Abstract:Our findings explain why some toxic baiting programs might fail. To achieve successful control, efforts should be directed at reducing the availability of background food to increase the probability of encounter and uptake of toxic baits. It is important to measure and understand the role of background food on toxic baiting programs to explain variable outcomes and inform strategies for successful bait delivery. Baiting is widely used in wildlife management for various purposes, including lethal control, fertility control, disease and parasite control, and conditioned aversion programs for many invasive vertebrate species. The efficacy of baiting programs relies on the likelihood that target animals will encounter the bait, consume it and receive an appropriate dose of the active ingredient. However, there has been little focus on encounter rate of toxic baits combined with behavioural aversion, which are likely to be significant factors affecting efficacy. Following optimal foraging theory, the likelihood of an animal encountering and consuming a toxic grain bait should increase in proportion to its availability relative to background food quantity if it is neither more or less detectable or palatable. Furthermore, the probability of consuming toxic baits might also be influenced by bait aversion following ingestion of a non‐lethal dose of toxin. Using a model system of wild house mice (Mus musculus L.) in mouse‐proof enclosures in Australia, we manipulated background food, applied zinc phosphide (Zn3P2) baits and measured mouse mortality. When background food was scarce, mouse mortality was high, whereas an increasing abundance of background food led to reduced mortality. A scenario modelling random encounters and including bait aversion explained 78% of the variation in observed mortality outcomes and achieved a closer fit to the data than modelling random encounters alone. Mortality rates were predicted to be higher with a higher strength bait, which would overcome behavioural aversion. Ensuring that animals locate and consume a lethal dose of toxic bait is a critical factor for successful bait delivery and efficacy. This is particularly significant in toxic baiting programs, where sublethal doses can make animals feel sick, leading to a negative association with the bait, and the development of aversion. Synthesis and applications: Our findings explain why some toxic baiting programs might fail. To achieve successful control, efforts should be directed at reducing the availability of background food to increase the probability of encounter and uptake of toxic baits. Abstract:Abstract M iscanthus sp. is regarded as suitable biomass for different biorefinery value chains. However, due to high recalcitrance, its wide use is yet untapped. Termite is the most efficient lignocellulose degrading insect, and its success results from synergistic cooperation with its gut microbiome. Here, we investigated at holobiont level the dynamic adaptation of a higher termite Cortaritermes sp. to imposed Miscanthus diet, with a long-term objective of overcoming lignocellulose recalcitrance. We used an integrative omics approach, comprising amplicon sequencing, metagenomics and metatranscriptomics that we combined with enzymatic characterisation of carbohydrate active enzymes from termite gut Fibrobacteres and Spirochaetae. Adaptation to the new diet was evidenced by reduced gut bacterial diversity and modified gene expression profiles, further suggesting a shift towards utilisation of cellulose and arabinoxylan, two main components of Miscanthus lignocellulose. Low identity of reconstructed microbial genomes to microbes from closely related termite species, supported the hypothesis of a strong phylogenetic relationship between host and its gut microbiome. Application-wise, this makes each termite gut system an endless source of enzymes that are potentially industrially relevant. This study provides a framework for better understanding the complex lignocellulose degradation by the higher termite gut system and paves a road towards its future bioprospecting.
Colony Suppression and Possible Colony Elimination of the Subterranean Termites Coptotermes formosanus and Reticulitermes speratus by Discontinuous Soil Treatment Using a Diluent of Fipronil Suspension Concentrate

Abstract:We assessed the efficacy of a discontinuous soil treatment using a diluent of fipronil suspension concentrate in controlling colonies of Coptotermes formosanus and Reticulitermes speratus. In-ground monitoring stations were installed at Isogi Park and Kindai University, and individual termites inhabiting the stations were collected for four or six years to determine the numbers and locations of colonies present in test areas before and after the discontinuous soil treatment. Microsatellite genotyping indicated that two C. formosanus and two R. speratus colonies in the test area at Isogi Park and five R. speratus colonies in the test area at Kindai University were active and that their territories fluctuated every year. One of the two C. formosanus colonies at Isogi Park and one of the five R. speratus colonies at Kindai University were subjected to discontinuous soil treatments with fipronil and were strongly affected by the treatment at the colony level, resulting in the suppression and possible elimination of colonies. Termite activity of the fipronil-treated colony of C. formosanus was detected within one week after the discontinuous soil treatment and was not found for more than two years (28 months), while termite activity of the fipronil-treated colony of R. speratus was detected within four days and three weeks after the discontinuous soil treatment and was not detected thereafter for three years. Fipronil residue analysis showed that workers of C. formosanus moved at least 28 m and that workers of R. speratus moved 6 m from the treated soil locations for up to three weeks.

Abstract:With the rapid development of the electronic information industry in recent years, electronic products are being updated faster and faster, and e-waste recycling has become a common problem around the world. Firstly, this article contrasts recycling at home and abroad using the predicament of Midea Corp. Based on a closed-loop supply chain with the system dynamics method, a model is constructed and simulated. In this model, the collection point coverage rate is introduced to adjust the e-waste recycling rate dynamically. Aiming at a recycling mode dominated by the third party of the closed-loop supply chain, the article mainly discusses the impact on the sales rate and market share of the recycling model by third-party enterprises and compares the total revenue of all supply chains. Simulation results show that the model is more effective and optimal than the traditional recycling model.

Abstract:BACKGROUND: Termites are worldwide pests causing considerable damage to agriculture, forestry and buildings. While various approaches have been tried to eliminate termite populations, the relevant toxicants are associated with certain risks to the environment and human health.RESULTS: In this study, to combine the merits of effective chemical control by bifenthrin and a drug photoimmobilisation technique, silk fibroin was used as a carrier to embed bifenthrin, which was then photoactively immobilised by ultraviolet treatment on the surface of wood (cellulose). The immobilised bifenthrin embedded in the photoactive silk fibroin was characterised by Fourier transform infrared spectroscopy (FTIR), ultraviolet absorption spectroscopy (UV), fluorescence measurement and CHN analysis. The surface structures and biological activity were examined by scanning electron microscopy (SEM), atomic force microscopy (AFM), electron spectroscopy for chemical analysis (ESCA) and bioassays respectively.CONCLUSIONS: The results indicate that the embedded and immobilised bifenthrin has been very well protected from free release and has a long-term stability allowing slow release with a high efficiency against termites at a low dose of 1.25 mu g cm(-2). This study provides a novel and environmentally benign technique for termite control by photoimmobilising silk-fibroin-embedded bifenthrin on the surface of materials that are otherwise easily attacked by termites.
