Abstract:A biotechnological technique that involves creating novel pesticides is the nanotechnology of natural compounds, like essential oils. With a lower threat to the environment and public health, these rational approaches provide a more sustainable choice. The present work sought to create novel nanoemulsions of ginger essential oil (G-EO) (Zingiber officinale) and basil essential oil (B-EO) (Ocimum basilicum) to control Odontotermes formosanus (O. formosanus). Stabilization of the ginger nano-emulsion (G-NE) and basil nano-emulsion (B-NE) was assessed by dynamic light scattering (DLS), zeta potential (ZP), and PH measurement, and the characterization was verified by transmission electron microscopy, UV-Vis, energy dispersive spectroscopy, and scanning electron microscopy. Furthermore, a chemical investigation utilizing gas chromatography and mass spectrometers revealed the main components of B-NE, B-EO, G-NE, and G-EO. In addition to assessing the essential oil's termiticidal abilities against O. formosanus in bulk and nanoemulsion formulations. When G-NE and G-EO were both at 17.5 mg/mL, the mortality rates were 100% and 42.87%, respectively. B-EO appeared to have a 45.33% death rate, while B-NE displayed a 100% mortality rate at a dose of 0.273 mg/mL. According to the termiticide appraisal, nanoemulsions have a greater effect on suppressing O. formosanus than essential oils. Additional research was conducted on the impact of G-NE and B-NE on the level of total protein, detoxification enzymes (carboxylesterase, acid, alkaline phosphatase, and acetylcholinesterase), and antioxidant enzyme (peroxidase). It was found that there was a significant change in biochemical activities when compared to the control. Additionally, changes in morphology and histology were seen after exposure to G-NE and B-NE. Moreover, when contrasted to the untreated group, the current study's analysis of the fungal community after exposure to B-NE revealed a high degree of diversity. This work stressed the potential of G-NE and B-NE as natural plant-derived termiticides and suggested a novel technique for sustainability and environmental friendliness in controlling O. formosanus.

Abstract:A prophenoloxidase (PPO) cDNA (OfPPO) was cloned from the Asian corn borer Ostrinia furnacalis. Sequence analysis revealed a full length transcript of the OfPPO cDNA with 2,686 bp, containing a 2,079 bp open reading frame (ORF), a 73-bp 5'-untranslated region, and a 534-bp 3'-untranslated region with a poly(A) signal. The ORF encodes a 693-amino acid polypeptide, containing two distinct copper-binding regions, a plausible thiol ester site, two proteolytic activation sites, and a conserved C-terminal region, but lacks a signal peptide sequence. Expression of the OfPPO transcript in the plasma, hemocytes, fat body and midgut was inhibited by Macrocentrus cingulum at 4 h post-parasitization (pp). In situ hybridization analysis showed that the hemocytes, especially the oenocytoids, hybridized strongly with the DNA probes of the OfPPO gene. No signal was detected in the cuticular epithelium or fat body of the parasitized larvae. Colloidal gold particles were used to visualize the PPO by immunoelectron microscopy. The time course study revealed a decrease in the labeling of the OfPPO at 4, 6, 8, 12, and 1 day pp in the larval integument and midgut parasitized by M. cingulum. We infer from time course studies of OfPPO gene expression and PO enzymatic activity that OfPPO in the integument is released from hemocytes and that the OfPPO expression was influenced at the transcriptional, translational, and then the post-translational level by parasitization challenge. Termiticidal Effects and Morpho-Histological Alterations in the Subterranean Termite (odontotermes Formosanus) Induced by Biosynthesized Zinc Oxide, Titanium Dioxide, and Chitosan Nanoparticles

Abstract:Recently, nanoparticles have been widely used in agricultural pest control as a secure substitute for pesticides. However, the effect of nanoparticles on controlling the subterranean termite Odontotermes formosanus (O. formosanus) has not been studied yet. Consequently, this study aimed to evaluate the effectiveness of some nanomaterials in controlling O. formosanus. The results showed that zinc oxide nanoparticles (ZnONPs), titanium dioxide nanoparticles (TiO2NPs), and chitosan nanoparticles (CsNPs) biosynthesized using the culture filtrate of Scedosporium apiospermum (S. apiospermum) had an effective role in controlling O. formosanus. Moreover, the mortality rate of O. formosanus after 48 h of treatment with ZnONPs, TiO2NPs, and CsNPs at a 1000 µg/mL concentration was 100%, 100%, and 97.67%, respectively. Furthermore, using ZnONPs, TiO2NPs, and CsNPs on O. formosanus resulted in morpho-histological variations in the normal structure, leading to its death. X-ray diffraction, UV-vis spectroscopy, Fourier transform infrared spectroscopy, scanning electron microscopy, dynamic light scattering, energy dispersive spectroscopy, and the Zeta potential were used to characterize the biosynthesis of ZnONPs, TiO2NPs, and CsNPs with strong activity against O. formosanus termites. Overall, the results of this investigation suggest that biosynthesized ZnONPs, TiO2NPs, and CsNPs have enormous potential for use as innovative, ecologically safe pesticides for O. formosanus control.

Abstract:Fungus-cultivating termites (Macrotermitinae) possess an elaborate strategy of lignocellulose digestion. It involves a lignocellulose-degrading fungal symbiont (genus Termitomyces), a diverse gut microbiota and a characteristic labour division in food processing. In this study, using pyrotag sequencing and electron microscopy, we analysed the bacterial microbiota in the hindgut of Odontotermes formosanus and its fungus comb to investigate the spatial organization, establishment and temporal succession of the bacterial communities colonizing specific microhabitats. Our results document strong differences between the communities at the hindgut epithelium and the luminal fluid of newly moulted, young and old worker termites. The differences in community structure were consistent with the density, morphology and spatial distribution of bacterial cells and the pools of microbial metabolites in the hindgut compartment, underlining that both gut development and the age-specific changes in diet affect the composition and functional role of their gut microbiota. These findings provide strong support for the concept that changes in diet and gut environment are important determinants of community structure because they create new niches for microbial symbionts.
OfGNBP silencing enhances the toxicity of Serratia marcescens Bizio (SM1) to Odontotermes formosanus (Shiraki)

Abstract:The immunity of insects plays a vital role in their survival. Our experiments found that lipopolysaccharide (LPS) and glucono-δ-lactone (GDL) could influence the virulence of Serratia marcescens Bizio (SM1) to Odontotermes formosanus (Shiraki) by affecting the immunity. Gram-negative binding proteins (GNBPs) are an important pattern recognition proteins that play a crucial role in the innate immune system. Therefore, two OfGNBPs were cloned in O. formosanus. The expression of OfGNBPs was significantly changed by LPS,SM1 and GDL, not prick. In addition, the immune-related gene expression, the phenoloxidase activity and antibacterial activity of donor termites and recipient termites were significantly induced by SM1. Furthermore, the knockdown of OfGNBP by RNA interference reduced not only individual immunity but also social immunity in O. formosanus, which increased the virulence of SM1 to O. formosanus. Importantly, dsOfGNBP alone also had good control effect on O. formosanus. In summary, we concluded that dsOfGNBPs are important termite immunosuppressants.

Abstract:Science Progress, Volume 107, Issue 1, January-March 2024. Termites cause a serious menace to wooden structures all over the world. They rely mostly on entozoic fauna residing in their hindgut for the digestion of cellulosic and hemicellulosic materials. One of the ways to control termites is through their gut symbionts. The present study was designed to characterize the hindgut bacteria isolated from Odontotermes obesus and Heterotermes indicola. Furthermore, the growth inhibitory effect of eight tropical plant extracts was investigated to find out potential control agents for these bacterial isolates. The characterization of bacteria was carried out based on their morphology, Gram staining, biochemical and amplification of 16SrRNA gene. Amplified products were sequenced to confirm their relationship with bacterial isolates from termites of other regions. The growth inhibitory effect of ethanolic leaf extracts of eight plants was evaluated in an invitro agar well diffusion method. Qualitative and quantitative phytochemical analysis of the most effective plant was carried out to learn about bioactive agents. The results confirmed the presence of five bacteria from each termite species. The Bacillus cereus, Escherichia coli, and Lysinibacillus fusiformis were common to both termites whereas Lysinibacillus xylanilyticus and Lysinibacillus macrolides were found in O. obesus only and H. indicola harbor Bacillus subtilis and Shigella sonnei in addition to common three ones. Among the plant extracts of Carica papaya, Eucalyptus camaldulensis, Osmium basilicum, Grevillea robusta, Eucalyptus globulus, Pongamia pinnata, Mentha longifolia, and Melia azedarach, the G. robusta > E. camaldulensis > O. basilicum were found to have growth inhibitory effects with increasing concentrations from 100 to 2000 μg/mL. The biodiversity of the bacterial fauna is important for the biological control of termites. Leaf extracts of these medicinal plants can be used to control termite infestation in an environment-friendly manner to save huge economic loss.

Exploring the Caste-Specific Multi-Layer Defense Mechanism of Formosan Subterranean Termites, Coptotermes formosanus Shiraki

Abstract:The survival and foraging of Coptotermesformosanus Shiraki in a microbe-rich environment reflect the adaptation of an extraordinary, sophisticated defense mechanism by the nest-mates. We aimed to explore the host pathogen interaction by studying caste-specific volatile chemistry and genes encoding the antioxidant defense of winged imagoes, nymphs, soldiers and workers of Formosan subterranean termites. Qualitative analyses of C.formosanus Shiraki performed by HS-SPME/GC-MS showed considerable variations in the chemical composition of volatile organic compounds (VOCs) and their proportions among all the castes. Winged imagoes produced the most important compounds such as naphthalene and n-hexanoic acid. The antifungal activity of these compounds along with nonanal, n-pentadecane, n-tetradecane, n-heptadecane and methyl octanoate against the conidial suspensions of Metarhiziumanisopliae and Beauveriabassiana isolates enable us to suggest that the failure of natural fungal infection in the nest is due to the antiseptic environment of the nest, which is mainly controlled by the VOCs of nest-mates. In addition, conidial germination of M.anisopliae and B.bassiana isolates evaluated on the cuticle of each caste showed significant variations among isolates and different castes. Our results showed that the conidia of M.anisopliae 02049 exhibited the highest germination on the cuticle of all the inoculated castes. Moreover, we recorded the lowest germination of the conidia of B.bassiana 200436. Caste-specific germination variations enabled us to report for the first time that the cuticle of winged imagoes was found to be the most resistant cuticle. The analysis of the transcriptome of C.formosanus Shiraki revealed the identification of 17 genes directly involved in antioxidant defense. Expression patterns of the identified antioxidant genes by quantitative real-time PCR (qPCR) revealed the significantly highest upregulation of CAT, GST, PRXSL, Cu/Zn-SOD2, TXN1, TXN2, TXNL1, TXNL2, TXNL4A and TPx genes among winged imagoes upon infection with the most virulent isolate, M.anisopliae 02049. Furthermore, soldiers showed the least expression of genes encoding antioxidant defense. Our findings indicated that the volatile chemistry of nest-mates and genes encoding antioxidant defense greatly contribute to the survival and foraging of Formosan subterranean termites in a microbe-rich habitat.

Abstract:The success of microbial termiticides in controlling termites depends on the ability of microbes to grow in different media and the functionality of the microbes as a resistant barrier or toxic bait. This study was conducted to understand the mortality rate and behaviour changes of the subterranean termite Coptotermes curvignathus Holmgren introduced with different concentrations of Serratia marcescens strain LGMS 1 and Pseudomonas aeruginosa strain LGMS 3 using wood and soil as bacterial transfer medium. In general, higher concentration of bacteria in soil caused a reduction in tunnelling activity and wood consumption and an increase in mortality. However, application on wood revealed a different outcome. Wood treated with S. marcescens of 10 6 CFU ml −1 concentration proved to be more efficient as bait than higher concentration applications as it caused a high mortality rate while still highly palatable for termites. Wood or soil treated with S. marcescens concentration higher than 10 9 CFU ml −1 creates a high toxicity and repellent barrier for termites. Pseudomonas aeruginosa of 10 9 CFU ml −1 concentrations applied on wood served as a slow-acting toxic bait. However, the ability for S. marcescens and P. aeruginosa to survive on wood is low, which made the bait unable to retain a useful level of toxicity for a long period of time and frequent reapplication is needed.

Abstract:Biocontrol of subterranean termites is largely impeded by their social immune responses. Studies on biocontrol agents combined with natural insecticides and their possible effects on the immune defense mechanisms of termites are limited. In this study, we investigated the effects of a combined biocontrol strategy using a plant-derived insect ATPase inhibitor, α-terpineol, with the entomopathogenic nematodes (EPNs) Steinernema carpocapsae against the subterranean termite Coptotermes formosanus Shiraki. Survival assays showed that even a low lethal concentration of α-terpineol significantly increased the EPNs-induced virulence in C. formosanus. α-terpineol treatment majorly inhibited the activity of Na+- K+- ATPase, which disturbed the EPNs-induced enhancement of locomotor activity and grooming behavior in termites treated with the combined strategy. Furthermore, the combination treatment had a synergistic inhibitory effect on innate immune responses in C. formosanus, which were measured as changes in the expression of immune-related genes and activities of immune system enzymes. In conclusion, α-terpineol can weaken the immune defense of termites against EPNs at low lethal concentrations, and is a suitable non-synthetic insecticide to prove the biocontrol efficiency of EPNs on C. formosanus. This study provides a theoretical basis and technical reference for a novel biocontrol strategy that promises to overcome the problems of host immune defense in termites.

Transcriptomic dissection of termite gut microbiota following entomopathogenic fungal infection

Abstract:Termites are social insects that live in the soil or in decaying wood, where exposure to pathogens should be common. However, these pathogens rarely cause mortality in established colonies. In addition to social immunity, the gut symbionts of termites are expected to assist in protecting their hosts, though the specific contributions are unclear. In this study, we examined this hypothesis in Odontotermes formosanus , a fungus-growing termite in the family Termitidae , by 1) disrupting its gut microbiota with the antibiotic kanamycin, 2) challenging O. formosanus with the entomopathogenic fungus Metarhizium robertsii , and finally 3) sequencing the resultant gut transcriptomes. As a result, 142531 transcripts and 73608 unigenes were obtained, and unigenes were annotated following NR, NT, KO, Swiss-Prot, PFAM, GO, and KOG databases. Among them, a total of 3,814 differentially expressed genes (DEGs) were identified between M. robertsii infected termites with or without antibiotics treatment. Given the lack of annotated genes in O. formosanus transcriptomes, we examined the expression profiles of the top 20 most significantly differentially expressed genes using qRT-PCR. Several of these genes, including APOA2, Calpain-5, and Hsp70, were downregulated in termites exposed to both antibiotics and pathogen but upregulated in those exposed only to the pathogen, suggesting that gut microbiota might buffer/facilitate their hosts against infection by finetuning physiological and biochemical processes, including innate immunity, protein folding, and ATP synthesis. Overall, our combined results imply that stabilization of gut microbiota can assist termites in maintaining physiological and biochemical homeostasis when foreign pathogenic fungi invade.

Abstract:Background and Objective: Termites have been classified into morphology-physiologically distinct castes that have a broad range of diets. In this study, the candidate of wood-feeding and fungus-growing termites was chosen to examine the relation between gut morphology, the density of microorganisms and starch hydrolyzing enzyme. Materials and Methods: The gut morphologies of the soldiers of wood-feeding (Globitermes sulphureus (Haviland, 1898), Microcerotermes crassus (Snyder, 1934) and fungus-growing (Macrotermes annandalei (Silvestri, 1914) and Odontotermes feae (Wassman, 1896) termites were determined and compared with those of the workers. Results: No significant differences were observed in the morphological structures between the soldiers and workers in different species. However, the microbial cell counts in the guts of the fungus-growing termites were 3-fold higher than those in the wood-feeding termites, with the hindgut having the highest number of microbes. In the wood-feeding termites, the workers and soldiers harboured a similar number of microorganisms, whereas, the workers of the fungus-feeding termites had 3-fold higher levels of microbes than the soldiers. The enzymatic activity of α-amylase in the fungus-growing termites was dramatically higher than that in the wood-feeding termites. However, there were no differences in enzyme activity in the gut segments of the wood-feeding group and the fungus-growing group. Conclusion: Taken together, the different feeding behaviour and castes of the termites affected the density of symbionts in their gut and α-amylase enzymatic activity was modified according to the amount of nutrition in their diets. This study provided additional information on the hidden role of microorganisms in starch metabolism in the termite’s gut.

Abstract:Collective behaviors of social insects are often regulated by pheromones. In subterranean termites, some workers forage for and exploit decaying wood for new food resources while forming tunnels from their nest. Colonizing new food resources requires workers to build and disinfect tunnels and chambers inside the nest and ingest decaying wood; therefore subterranean termite colonies should have mechanisms to establish and maintain groups of workers to perform these functions. Recently, an aggregation pheromone was identified in workers of the termite Reticulitermes speratus , which induces quick attraction of nestmate workers and prolonged aggregation to the site of attraction. In this study, we extended this work to another species of Reticulitermes and identified a worker aggregation pheromone in the termite R. virginicus . GC–MS analysis and bioassays demonstrated that this pheromone consists of 3-octanone, 3-octanol, and palmitic acid and shows a colony-specific, dose-dependent attractant response but not an arrestant response. Furthermore, these pheromone components were most likely emitted from the surface of the body. This suggests that aggregation pheromone composition and function differ significantly among termite species, even within the same genus. This study advances our understanding of the regulatory mechanisms of termite aggregation behavior.

Anti-Termitic Activity of Three Plant Extracts, Chlorpyrifos, and a Bioagent Compound (Protecto) against Termite Microcerotermes eugnathus Silvestri (Blattodea: Termitidae) in Egypt

Abstract:A trend towards environmentally friendly chemicals for use in termite management has been occurring globally. This study examined three naturally occurring plant extracts from Lavandula latifolia (Spike lavender), Origanum vulgare (Marjorum), and Syzygium aromaticum (Clove) against the termite Microcerotermes eugnathus. Plant extract results were compared to two commercially used termite pesticides, the bio-insecticide, Bacillus thuringiensis var. kurstaki (Protecto 9.4% WP) and Dursban (Chlorpyrifos 48%). Gas chromatography-mass spectrometry (GC-MS) analysis was used to identify the main compounds in the three plant extracts. The main compounds in Lavandula Latifolia were linalool (21.49%), lavandulol (12.77%), β-terpinyl acetate (10.49%), and camphor (9.30%). Origanum vulgare extract contained thymol (14.64%), m-cymene (10.63%), linalool (6.75%), and terpinen-4-ol (6.92%) as main compounds. Syzygium aromaticum contained eugenol (99.16%) as the most abundant identified compound. The extract of O. vulgare caused the highest termite death rate, with an LC50 of 770.67 mg/L. Exposure to lavender extract showed a high death rate with an LC50 of 1086.39 mg/L. Clove extract did not show significant insecticidal activity with an LC50 > 2000 mg/L. Significant termiticide effects were found, with LC50 values of 84.09 and 269.98 mg/L for soldiers and workers under the application of Dursban and Protecto, respectively. The LC50 values reported for nymphs were <120, <164.5, and 627.87 mg/L after exposure to Dursban, Protecto, and O. vulgare extract, respectively. The results of the study show that some of the extracts have low toxicity compared to the bioagent and Dursban, and may show promise as natural termiticides, particularly as extracts from O. vulgare.

Abstract:Some insect herbivores sequester plant secondary metabolites (PSMs) for their own defense, raising the interesting possibility that grazing herbivores are defended by combinations of PSMs from different plant species. In this study, we tested the hypothesis that the grazing caterpillar, Grammia incorrupta, deters the ant, Aphaenogaster cockerelli, by eating a mixture of plants containing iridoid glycosides (IGs) and those containing pyrrolizidine alkaloids (PAs), and that this deterrence is greater than that attained by eating either plant alone. This hypothesis was tested against the non-mutually exclusive hypothesis that mixing plants containing PAs with those containing IGs improves growth performance. Caterpillar survival and growth were measured on three experimental diets: a PA plant, an IG plant, and a mixture of the two. We measured the degree of deterrence associated with these, and an additional experimental diet devoid of PSMs at naturally occurring A. cockerelli nests. Caterpillars fed both plants gained more mass than those fed either plant alone, but took longer to develop. These differences were not caused by diet-based variation in growth efficiency, but by eating more food when offered the mixed-plant diet relative to single-plant diets. The mixed diet was shown to provide deterrence to ants, whereas caterpillars fed single-plant diets were not significantly more deterrent than caterpillars that had eaten the PSM-free diet. We hypothesize that enhanced defense results from increased food consumption in response to multiple plant species, perhaps leading to greater PSM sequestration. Through this mechanism, bottom-up and top-down effects may mutually reinforce the grazing dietary strategy.

Abstract:Recently, the use of nanopesticides has shown significant efficacy in the control of many pests. However, the effect of nanopesticides, especially nanoemulsions, on suppressing termites, Odontotermes formosanus (Shiraki, 1909) ( O. formosanus ), has not been studied yet. Therefore, this study aimed to produce nanoemulsions of the essential oils of eucalyptus ( Eucalyptus globulus Labill; E-EO) and nutmeg ( Myristica fragrans Houtt; N-EO) to suppress O. formosanus . The analysis of eucalyptus nanoemulsion (E-NE) and nutmeg nanoemulsion (N-NE) was confirmed by using UV-Vis, dynamic light scattering, zeta potential, transmission electron microscopy, scanning electron microscopy, and energy dispersive spectroscopy. In addition, chemical analysis by Gas Chromatography with a mass spectrometer (GC-MS) exhibited the major constituents of E-NE and N-NE. The principal chemical components of E-NE included D-limonene, eucalyptol, 1,5-cyclooctadiene,3,4-dimethyl, benzene, and 1-methyl-3-(1 methylethyl)-, while the main constituents in N-NE were cyclohexane,1-methylene-4-(1 methylethenyl)-, eucalyptol, and L-. alpha. -terpineol. The mortality rates were 100% and 99.53%, respectively, after 24 hours of treatment with a concentration of 140 mg/mL, compared to 23.43% and 43.55%, respectively, from E-EO and N-EO treatment. These results refer to the essential oils’ nanoemulsion as far more effective than the essential oils themselves. Furthermore, the effects of E-NE and N-NE on detoxification enzymes such as acetylcholinesterase, carboxylesterase, acid and alkaline phosphatase were investigated, as well as total protein concentrations, and the results have been found to be significantly increasing or decreasing in comparison with control. Besides, histological and morphological alterations found post exposure to E-NE and N-NE were shown. Overall, the results from this study clearly indicate that the nanopesticide-formulated nanoemulsions may have great potential to be used as novel, environmentally safe insecticides for controlling O. formosanus .

Abstract:Olfaction is critical for survival because it allows animals to look for food and detect pheromonal cues. Neuropeptides modulate olfaction and behaviors in insects. While how the neuroregulation of olfactory recognition affects foraging behavior in termites is still unclear. Here, we analyzed the change after silencing the olfactory co-receptor gene (Orco) and the neuropeptide Y gene (NPY), and then investigated the impact of olfactory recognition on foraging behavior in Odontotermes formosanus under different predation pressures. The knockdown of Orco resulted in the reduced Orco protein expression in antennae and the decreased EAG response to trail pheromones. In addition, NPY silencing led to the damaged ability of olfactory response through downregulating Orco expression. Both dsOrco- and dsNPY-injected worker termites showed significantly reduced walking activity and foraging success. Additionally, we found that 0.1 pg/cm trail pheromone and nestmate soldiers could provide social buffering to relieve the adverse effect of predator ants on foraging behavior in worker termites with the normal ability of olfactory recognition. Our orthogonal experiments further verified that Orco/NPY genes are essential in manipulating termite olfactory recognition during foraging under different predation pressures, suggesting that the neuroregulation of olfactory recognition plays a crucial role in regulating termite foraging behavior.

Abstract:Although subterranean termites live within soil, little attention has been paid on the potential interaction among subterranean termites and soil microbes. Herein, we conducted different choice tests to investigate aggregation and tunneling behaviors of Coptotermes formosanus Shiraki responding to soil/sand treated with conidia of seven soil fungi, Trichoderma longibrachiatum Rifai, Trichoderma koningii Oud., Trichoderma hamatum (Bon.) Bain., Trichoderma atroviride Karsten, Trichoderma spirale Indira and Kamala, Trichoderma harzianum Rifai, and Trichoderma viride Pers. ex Fries. In aggregation-choice test, soil treated with nearly all Trichoderma species tested (except T. koningii) significantly increased termite aggregation compared with untreated soil. In tunneling-choice tests, termites produced significantly larger tunnels in sand treated with T. longibrachiatum or T. koningii than that in untreated sand. We hypothesized that Trichoderma species could benefit termites by protecting them from infection of the entomopathogenic fungus Metarhizium anisopliae (Metschn) Sorokin, and three Trichoderma species that attracted termites (T. longibrachiatum, T. atroviride, and T. harzianum) were tested. The antagonism tests showed that the three Trichoderma species suppressed growth of M. anisopliae. Also, the median lethal time (LT50) of termites exposed to both Trichoderma species and M. anisopliae was significantly longer than termites exposed to M. anisopliae alone. Interestingly, though significantly fewer termites aggregated in soil treated with M. anisopliae conidia compared with untreated soil, M. anisopliae conidia mixed with T. longibrachiatum or T. harzianum were no longer repellent to termites. Our results showed that the fungi in the genus Trichoderma (1) exerted generally attractive effects on termites, (2) protected termites from the infection of entomopathogenic fungus, and (3) altered pathogen-avoiding behaviors of termites. Future studies will be required to understand the mechanisms underlying these newly discovered effects.

Abstract:Costs associated with subterranean termite damage and control are estimated to approach $2 billion annually in the United States alone. The Formosan subterranean termite, Coptotermes formosanus Shiraki, is one of the more economically important subterranean species. In recent years, the shortcomings associated with conventional chemical control methods have prompted policymakers and scientists to evaluate the potential for biological control of subterranean termites (C. formosanus in particular), that is, to determine the potential for natural enemies - predators, parasitoids and pathogens - to suppress termite populations. Ants are the greatest predators of termites, and may have a considerable local impact on termite populations in some areas of the world. A few parasitoids of termites are known, but their potential for regulating termite populations seems negligible. Characteristics of the colony, such as a protected, underground location (and, for the C. formosanus nest, its modular and dispersed nature), are likely to limit the impact predators and parasitoids have on subterranean termites. Thus, there seems little potential for use of these agents for subterranean termite control. For various reasons, pathogenic organisms, such as viruses, bacteria, Protozoa, nematodes and most fungi, have shown little promise for use in biological termite control. However, research suggests that strains of two well-studied, endoparasitic fungi, Beauveria bassiana and Metarhizium anisopliae, when employed in baiting schemes, may offer the potential for at least some measure of subterranean termite control, although their successful use is compromised by a number of inherent biological limitations and logistical problems that have yet to be solved. Although not strictly in the realm of classical biological control, recent studies suggest that natural products, such as ant semiochemicals and fungal metabolites (siderophores), or their synthetic analogues, eventually might find a use in termite control programmes as repellents or insecticides in wood treatments or soil applications if stable formulations can be developed.

Abstract:In recent years, the use of microbes to control termites has attracted increasing attention. It was found that pathogenic bacteria, nematodes, and fungi effectively control termites under laboratory conditions. However, their effects have not been replicated in the field, and one reason for this is the complex immune defense mechanisms of termites, which are mainly regulated by immune genes. Therefore, altering the expression of immune genes may have a positive influence on the biocontrol efficacy of termites. Coptotermes formosanus Shiraki is one of the most economically important termite pests worldwide. Currently, the large-scale identification of immune genes in C. formosanus is primarily based on cDNA library or transcriptome data rather than at the genomic level. In this study, we identified the immune genes of C. formosanus according to genome-wide analysis. In addition, our transcriptome analysis showed that immune genes were significantly downregulated when C. formosanus was exposed to the fungus Metarhizium anisopliae or nematodes. Finally, we found that injecting dsRNA to inhibit three immune genes (CfPGRP-SC1, CfSCRB3, and CfHemocytin), which recognize infectious microbes, significantly increased the lethal effect of M. anisopliae on termites. These immune genes show great potential for C. formosanus management based on RNAi. These results also increase the number of known immune genes in C. formosanus which will provide a more comprehensive insight into the molecular basis of immunity in termites.

Abstract:There is a growing risk of pollinators being exposed to multiple fungicides due to the widespread use of fungicides for plant protection. A safety assessment of honeybees exposed to multiple commonly used fungicides is urgently required. Therefore, the acute oral toxicity of the ternary mixed fungicide of ABP (azoxystrobin: boscalid: pyraclostrobin = 1:1:1, m/m/m) was tested on honeybees (Apis cerana cerana), and its sublethal effect on foragers' guts was evaluated. The results showed that the acute oral median lethal concentration (LD50) of ABP for foragers was 12.6 μg a.i./bee. ABP caused disorder of the morphological structure of midgut tissue and affected the intestinal metabolism; the composition and structure of the intestinal microbial community was perturbed, which altered its function. Moreover, the transcripts of genes involved in detoxification and immunity were strongly upregulated with ABP treatment. The study implies that exposure to a fungicide mixture of ABP can cause a series of negative effects on the health of foragers. This work provides a comprehensive understanding of the comprehensive effects of common fungicides on non-target pollinators in the context of ecological risk assessment and the future use of fungicides in agriculture.
