Abstract:Food allergy (FA), triggered by specific dietary allergens, has emerged as a substantial global concern for food safety and public health. While studies have elucidated changes in immune cells and cytokines associated with allergen exposure, a comprehensive analysis of the host's metabolic features and the interaction between metabolites and the gut microbiota has not been conducted. In this study, egg allergen ovalbumin (OVA) was administered by the oral route to sensitized BALB/c mice to faithfully replicate key aspects of human FA, including severe allergic diarrhea, mast cell infiltration, and elevated levels of serum IgE, mMCPT-1, and Th2 cell hallmark cytokines (such as IL-4, IL-5, and IL-13). Furthermore, the untargeted and targeted metabolomic analyses indicated that FA mice precipitate a substantial decrease in tryptophan metabolites-indole-3-acrylic acid (IA) and indole-3-lactic acid (ILA). The integration of shotgun metagenome and metabolome data further unveiled the dysregulation of indole metabolism is related to a decline in the abundance of beneficial bacteria such as Lactobacillus and Bifidobacterium. Additionally, disruption of the tryptophan-indole derivative pathway compromises the maintenance of intestinal mucosal function through the AHR signaling pathway, manifested by decreased expression of Reg3g and IL22. Taken together, this study demonstrated that the anaphylaxis triggered by oral ingestion of food allergens can lead to disruptions in tryptophan metabolism, consequently impairing intestinal immune homeostasis.

Abstract:Food allergy is a worldwide food safety problem with increasing prevalence. Developing novel approaches for food allergy investigations is the basis for controlling food allergies. In this work, a 3-dimensional (3D) intestinal cell model was established to simulate the intestinal mucosal immune system. Gut epithelial cell line CMT93 was cultured in a transwell insert above dendritic cells (DCs) isolated from mouse spleen and stimulated by egg allergen ovalbumin (OVA), then the conditioned media of DCs was transferred to T cells isolated from mouse spleen. The allergy-related indexes of each cell type were determined by qPCR and flow cytometry. Then the TAZ gene was knocked down in the CMT93 cells and the role of the Hippo pathway in OVA-induced food allergy was investigated. The 3D intestinal cell model showed more significant and more specific allergic responses than conventional cell models and is more convenient to be manipulated than the mouse models. This model is an ideal tool for food allergy investigations and would facilitate studies in the field of intestinal mucosal immunity.

Dietary Eggshell Membrane Powder Improves Survival Rate and Ameliorates Gut Dysbiosis in Interleukin-10 Knockout Mice

Abstract:Inflammatory bowel disease (IBD) is known to be associated with compositional and metabolic changes in the gut microbiota. The aim of this study was to investigate whether dietary eggshell membrane (ESM) improves survival rate or ameliorates gut dysbiosis in a spontaneous IBD model of interleukin-10 knockout (IL10(-/-)) mice. Female C57BL/6J wild-type (WT) and IL10(-/-) mice (KO) were fed an AIN-93G basal diet or an ESM diet (KOE) for 19 weeks. Gut microbiota profiles were analyzed via 16S rRNA sequencing, and short-chain fatty acids in cecal content were analyzed with high-performance liquid chromatography. The results demonstrated that ESM supplementation significantly improved the survival rate and body composition in KO mice. Alpha diversity analysis of the microbiota revealed that ESM supplementation significantly increased gut microbial diversity, which was decreased in IL10(-/-) mice. The Firmicutes/Bacteroidetes ratio was recovered to a normal level by ESM supplementation, suggesting that ESM helps maintain the compositional balance of the gut microbiota. ESM increased relative abundance of commensal bacterial Ruminococcus and Bacteroidales S24-7 and reduced the abundance of the proinflammatory-related bacterium, Enterobacteriaceae. Additionally, ESM supplementation promoted the production of butyrate in cecal contents and downregulated the expression of proinflammatory genes, including interleukin-1 beta (Il-1 beta) and tumor necrosis factor-alpha (Tnf-alpha) in IL10(-/-) mice colon, indicating anti-inflammatory functions. These findings suggest that ESM may be used as a beneficial dietary intervention for IBD.

Abstract:αs1-Casein is a potential allergen to induce hypersensitivity in cow milk. We had identified αs1-casein and its epitopes in previous studies. The present study aimed to evaluate the allergic mechanism of αs1-casein in a BALB/c mouse model. The levels of specific IgE, mast cell proteinase, histamine and cytokines in sensitized mice were determined, and the clinical and pathological observation were evaluated. Results showed that the levels of specific IgE, mast cell proteinase, histamine, IL-4, IL-5, IL-10 increased significantly with a dose-dependent trend. The local alveolar septum collapsed or thickened, and lymphatic foci were produced in the spleen and thymus, and the inflammatory cells infiltrated in small intestinal mucosa mesenchyme. In conclusion, the levels of specific IgE, mast cell proteinase, histamine, IL-4, IL-5, IL-10 and some inflammatory factors could possibly serve as allergic biomarkers of αs1-casein, however, additional studies on signal transduction and gene expression are necessary in future.

Abstract:Background: The pathological mechanism of the gastrointestinal forms of food allergies is less understood in comparison to other clinical phenotypes, such as asthma and anaphylaxis Importantly, high-IgE levels are a poor prognostic factor in gastrointestinal allergies. Methods: This study investigated how high-IgE levels influence the development of intestinal inflammation and the metabolome in allergic enteritis (AE), using IgE knock-in (IgEki) mice expressing high levels of IgE. In addition, correlation of the altered metabolome with gut microbiome was analysed. Results: Ovalbumin-sensitized and egg-white diet-fed (OVA/EW) BALB/c WT mice developed moderate AE, whereas OVA/EW IgEki mice induced more aggravated intestinal inflammation with enhanced eosinophil accumulation. Untargeted metabolomics detected the increased levels of N-tau-methylhistamine and 2,3-butanediol, and reduced levels of butyric acid in faeces and/or sera of OVA/EW IgEki mice, which was accompanied with reduced Clostridium and increased Lactobacillus at the genus level. Non-sensitized and egg-white diet-fed (NC/EW) WT mice did not exhibit any signs of AE, whereas NC/EW IgEki mice developed marginal degrees of AE. Compared to NC/EW WT mice, enhanced levels of lysophospholipids, sphinganine and sphingosine were detected in serum and faecal samples of NC/EW IgEki mice. In addition, several associations of altered metabolome with gut microbiome-for example Akkermansia with lysophosphatidylserine-were detected. Conclusions: Our results suggest that high-IgE levels alter intestinal and systemic levels of endogenous and microbiota-associated metabolites in experimental AE. This study contributes to deepening the knowledge of molecular mechanisms for the development of AE and provides clues to advance diagnostic and therapeutic strategies of allergic diseases.

Abstract:: Eosinophilic esophagitis (EoE) is a chronic T helper type 2 (Th2)-associated inflammatory disorder triggered by food allergens, resulting in esophageal dysfunction through edema, fibrosis, and tissue remodeling. The role of epithelial remodeling in EoE pathogenesis is critical but not fully understood. : To investigate the role of epithelial IKKβ/NFκB signaling in EoE pathogenesis using a mouse model with conditional knockout in esophageal epithelial cells ( ). : EoE was induced in mice through skin sensitization with MC903/Ovalbumin (OVA) followed by intraesophageal OVA challenge. Histological and transcriptional analyses were performed to assess EoE features. Single-cell RNA sequencing (scRNA-seq) was used to profile esophageal mucosal cell populations and gene expression changes. : /EoE mice exhibited hallmark EoE features, including eosinophil infiltration, intraepithelial eosinophils, microabscesses, basal cell hyperplasia, and lamina propria remodeling. RNA-seq revealed significant alterations in IKKβ/NFκB signaling pathways, with decreased expression of and increased expression of IKKβ negative regulators. scRNA-seq analyses identified disrupted epithelial differentiation and barrier integrity, alongside increased type 2 immune responses and peptidase activity. : Our study demonstrates that loss of epithelial IKKβ signaling exacerbates EoE pathogenesis, highlighting the critical role of this pathway in maintaining epithelial homeostasis and preventing allergic inflammation. The /EoE mouse model closely mirrors human EoE, providing a valuable tool for investigating disease mechanisms and therapeutic targets. This model can facilitate the development of strategies to prevent chronic inflammation and tissue remodeling in EoE.

Abstract:Food allergy is an important health problem that affects human quality of life and socioeconomic development, and its treatment requires improvement. Intestinal flora dysbiosis is closely associated with food allergies. A sensitised mouse model was established by the intraperitoneal injection of ovalbumin (OVA). The mice were randomly divided into four groups: control, model, high-dose (H), and low-dose (L) inulin. The mice were administered water containing different concentrations of inulin four weeks before the OVA injection. Body weight changes were monitored. After the last OVA injection, the mice were scored for allergic reactions. The levels of total immunoglobulin E (IgE) and diamine oxidase (DAO) in the serum and secretory IgA (sIgA) in the small intestinal mucus were measured, and 16S rRNA sequencing of the faecal flora was performed to evaluate microbial parameters. The intestinal flora biomarkers, correlations between them, and biochemical indicators were analysed. Inulin treatment had no effect on the body weight of OVA-sensitised mice but attenuated allergic reactions and intestinal injury in mice. Compared with the control group, the model group had significantly higher levels of serum DAO and IgE and significantly lower levels of intestinal mucus IgA. IgA levels in the intestinal mucus of mice treated with inulin prior to OVA sensitisation were higher than those in non-inulin-treated OVA-sensitised mice. Furthermore, analysis of operational taxonomic units showed that inulin treatment decreased the abundance of Alloprevotella, Rikenellaceae RC9, Eubacterium siraeum, and Eubacterium xylanophilum, and increased the abundance of Blautia and Lachnospiraceae. Serum DAO levels were positively associated with Eubacterium siraeum, Alloprevotella, Eubacterium xylanophilum, and Odoribacter and negatively associated with Blautia, Tyzzerella, Alistipes, Desulfovibrionaceae, and Ruminococcaceae UCG005. In addition, IgE levels were positively associated with Eubacterium siraeum, Alloprevotella, Eubacterium xylanophilum, Odoribacter, and Citrobacter and negatively associated with Blautia, unclassified Ruminococcaceae, and Alistipes. IgA exhibited significant positive correlation with Blautia, norank_f_Eubacterium coprostanoligenes, Alistipes, norank Desulfovibrionaceae, Muribaculum, and Ruminococcaceae U C G 005 and significant negative correlation with Eubacterim siraeum, Eubacterium xylanophilum, Odoribacter, and Citrobacter. Inulin exerts a protective effect against food allergies in mice, which is partially mediated by alterations in the gut microbiota.

Abstract:In order to explore the in vivo anti-food allergy activity of Lactobacillus.sakei subsp.sakei-fermented Eucheuma spinosum polysaccharides F1-ESP-3 (Earlier reports). The OVA-induced food allergy mouse model was established by ascites immunization and gavage. The weight, temperature, incidence of diarrhea, levels of allergic mediators and inflammatory factors in serum of mice were analyzed. We analyzed the differentiation of mice spleen lymphocytes and the proportion of sensitized mast cells by Flow Cytometry. The intestinal barrier status of mice was analyzed by intestinal pathological tissue sections and microbiota sequencing. The results showed that F1-ESP-3 could alleviate the food allergy symptoms of mice, such as hypothermia and loose stool; Levels of OVA specific immunoglobulin E, mast cell protease and histamine in the serum of sensitized mice and the proportion of dendritic cells and mast cells in mouse spleen were significantly reduced; In addition, F1-ESP-3 can protect the intestinal barrier and further improve the intestinal microenvironment of food allergic mice by regulating the abundance of Bacteroidetes and Firmicutes. F1-ESP-3 can further improve the intestinal microenvironment of food allergic mice by up regulating the levels of Lachnospiraceae, and may affecting the signal pathways such as NOD like receptor, MAPK, I kappa B and antigen processing and presentation.

Abstract:To investigate the anti-alpha s1-casein allergy mechanism of two tea-derived polyphenols, epigallocatechin (EGC) and epigallocatechin gallate (EGCG), BALB/c mice were sensitized and challenged with alpha s1-casein and nutritional intervention was given by EGC and EGCG during the sensitization provocation phase. The main evaluation indexes used were levels of mast cell proteases, histamine, and specific antibody immunoglobulin E (IgE), as well as cytokine secretion and pathological observation. The results showed that both EGC and EGCG significantly reduced levels of mast cell protease, histamine, specific IgE antibodies, and Th2 cytokines in allergic mice. The histopathology results showed that both EGC and EGCG markedly reduced the degree of lesions in the intestine, thymus, spleen, and lung. The conclusions from this study can provide a theoretical basis for the mechanism by which tea polyphenols regulate food allergens.(c) 2023 Beijing Academy of Food Sciences. Abstract:Abstract House dust mites (HDM) are one of the important factors of airway allergic diseases, HDM allergens can be detected in the human gut mucosa, which induces local inflammation and increases intestinal epithelial permeability. This study tests a hypothesis that HDM contribute to the development of OVA (ovalbumin)-induced intestinal allergy. The serum levels of IgE against HDM in patients with food allergy were detected with UniCAP100 (Pharmacia, Uppsala, Sweden); a mouse model of food allergy was developed with OVA and HDM as the specific antigens. Compared to healthy controls, patients with food allergy have higher levels of serum HDM-specific IgE. Compared to food allergy alone groups, the levels of HDM-specific IgE in patients with food allergy and asthma or allergic rhinitis were significantly higher. In mouse models, we found that HDM/OVA induced allergy-like symptoms, lower body temperature, and lower body weight. The levels of IgE, IgG1, mMCP-1 (mouse mast cell protease-1), IL-4 and IL-5 in the HDM and HDM + CT (cholera toxin) groups were higher than the control groups, and the levels of IgE, IgG1, IL-4 and IL-5 in the HDM, OVA and HDM + OVA groups were higher than the control groups. The pathological changes of intestinal tissues in the HDM and HDM + CT/the HDM, OVA and HDM + OVA groups were more severe, more eosinophil infiltration than the control groups. Moreover, exposure to HDM induced intestinal barrier dysfunction, and facilitated the development of intestinal allergy in mice. In conclusion, HDM exposure enhances immune responses to OVA-induced food allergy.

Abstract:Background/Objectives: Cow's milk allergy is one of the most common food allergies in children, and its pathomechanism is still under investigation. Recently, an increasing number of studies have linked food allergy to intestinal barrier dysfunction. The present study aimed to investigate changes in the intestinal microenvironment during the development of β-lactoglobulin (β-lg) allergy under conditions of early intestinal dysfunction. Methods: BALB/c mice received intraperitoneal β-lg with Freund's adjuvant, followed by oral β-lg while receiving dextran sulphate sodium salt (DSS) in their drinking water (0.2% w/v). The immunized group without DSS and the groups receiving saline, oral β-lg, or DSS served as controls. Results: The study showed that the immunization effect was greater in mice with mild intestinal barrier dysfunction. Although DSS did not affect the mice's humoral response to β-lg, in combination with β-lg, it significantly altered their cellular response, affecting the induction and distribution of T cells in the inductive and peripheral tissues and the activation of immune mediators. Administration of β-lg to sensitized mice receiving DSS increased disease activity index (DAI) scores and pro-inflammatory cytokine activity, altered the distribution of claudins and zonulin 1 (ZO-1) in the colonic tissue, and negatively affected the balance and activity of the gut microbiota. Conclusions: The research model used appears attractive for studying food allergen sensitization, particularly in relation to the initial events leading to mucosal inflammation and the development of food hypersensitivity.

Abstract:This research aimed to investigate the allergic reaction of C3H/HeJ mice after sensitization with ovalbumin (OVA) without any adjuvant and to analyze the association between intestinal microbiota and allergy-related immune cells in mesenteric lymph nodes (MLN). The allergic responses of C3H/HeJ mice orally sensitized with OVA were evaluated, and immune cell subsets in spleen and MLN and cytokines were also detected. The intestinal bacterial community structure was analyzed, followed by Spearman correlation analysis between changed gut microbiota species and allergic parameters. Sensitization induced a noticeable allergic response to the gavage of OVA without adjuvant. Increased levels of Th2, IL-4, CD103 + CD86 + DC, and MHCII + CD86 + DC and decreased levels of Th1, Treg, IFN-γ, TGF-β1, and CD11C + CD103 + DC were observed in allergic mice. Furthermore, families of Lachnospiraceae , Clostridiaceae_1 , Ruminococcaceae , and peprostreptococcaceae , all of which belonging to the order Clostridiales , were positively related to Treg and CD11C + CD103 + DC, while they were negatively related to an allergic reaction, levels of Th2, CD103 + CD86 + DC, and MHCII + CD86 + DC in MLN. The family of norank_o_Mollicutes_RF39 belonging to the order Mollicutes_RF39 was similarly correlated with allergic reaction and immune cells in MLN of mice. To sum up, allergic reactions and intestinal flora disturbances could be induced by OVA oral administration alone. The orders of Clostridiales and Mollicutes_RF39 in intestinal flora are positively correlated with levels of Treg and CD11C + CD103 + DC in MLN of mice.

Abstract:Trichloroethylene (TCE) is a major occupational hazard and environmental contaminant that can cause multisystem disorders in the form of occupational medicamentosa-like dermatitis. Development of dermatitis involves several proinflammatory cytokines, but their role in TCE-mediated dermatitis has not been examined in a well-defined experimental model. In addition, few animal models of TCE sensitization are available, and the current guinea pig model has apparent limitations. This study aimed to establish a model of TCE-induced skin sensitization in BALB/c mice and to examine the role of several key inflammatory cytokines on TCE sensitization. The sensitization rate of dorsal painted group was 38.3%. Skin edema and erythema occurred in TCE-sensitized groups, as seen in 2,4-dinitrochlorobenzene (DNCB) positive control. Trichloroethylene sensitization-positive (dermatitis [+]) group exhibited increased thickness of epidermis, inflammatory cell infiltration, swelling, and necrosis in dermis and around hair follicle, but ear painted group did not show these histological changes. The concentrations of serum proinflammatory cytokines including tumor necrosis factor (TNF)-α, interferon (IFN)-γ, and interleukin (IL)-2 were significantly increased in 24, 48, and 72 hours dermatitis [+] groups treated with TCE and peaked at 72 hours. Deposition of TNF-α, IFN-γ, and IL-2 into the skin tissue was also revealed by immunohistochemistry. We have established a new animal model of skin sensitization induced by repeated TCE stimulations, and we provide the first evidence that key proinflammatory cytokines including TNF-α, IFN-γ, and IL-2 play an important role in the process of TCE sensitization.

A Mouse Model of Atopic March Induced by Epicutaneous Immunization with House Dust Mite Allergen: a Preliminary Study

Abstract:Objective To establish a mouse model of atopic march induced by epicutaneous sensitization and nasal challenge with dust mite allergen.Methods C57BL/6J mice were subjected to repeated epicutaneous sensitization with dust mite allergen or saline and then challenged with intranasal administration of mite allergen to induce airway inflammation.The mouse model was evaluated in terms of pathology of skin and lung,quantitative mRNA expression of inflammatory molecules,count of inflammatory cells in bronchoalveolar lavage fluid,and airway hyperresponsiveness.Results Mite allergen sensitized mice exhibited typical pathological changes of eczematous dermatitis and significantly higher expression levels of Th2 cytokines in lesions while Thl cytokines remained unchanged in comparison with normal controls.The number of total cells,lymphocytes and eosinophils in the bronchial alveolar lavage fluid were significantly higher in model mice than in the controls.Pulmonary histopathology showed peribronchial infiltration of eosinophils.The expression levels of Th2 cytokine mRNA in lung tissue also significantly increased.Methacholine dose-dependently induced increase in the lung resistance and decrease in the dynamic compliance of the lungs in mouse model.Conclusion In this study,we use epicutaneous sensitization and nasal challenge to successfully establish the mite allergen induced atopic march model.
Effects of Stimulating Food on Allergic Contact Dermatitis in BALB/c mice

Abstract:OBJECTIVE To study the effects of diet fawu on allergic contact dermatitis(ACD)in mice models and to explore the possible mechanism.METHODS Mice models of ACD was established and the effects of mutton,bamboo shoots,yellow wine,green pepper,sea shrimp on inflammatory reaction and related cytokines was observed respectively at 48hafter elicitation.RESULTS Compared with the control group,mutton,yellow wine,green pepper groups can significantly increase the ear swelling and the number of infiltrating inflammator cells in mice models(P<0.05),bamboo shoots,sea shrimp groups showed no significant difference(P>0.05).The ear weight changes of each group except mutton showed no significant difference compared with the control group(P>0.05);mutton,yellow wine,green pepper groups can markedly increase expression level of IFN?γ,IL-17,IL-2in peripheral blood(P<0.05),bamboo shoots group decreased(P<0.05),no significant effect of shrimp group(P>0.05).CONCLUSION Mutton,green peppers,yellow wine can aggravate ACD in mice possibly by upregulation the expressions of IFN?γ,IL-17,IL-2.Bamboo shoots and sea shrimp have no effect on mice ACD.

Abstract:Background: Mucosal mast cells (MC) are key players in IgE-mediated food allergy (FA). The evidence on the interaction between gut microbiota, MC and susceptibility to FA is contradictory. Objective: We tested the hypothesis that commensal bacteria are essential for MC migration to the gut and their maturation impacting the susceptibility to FA. Methods: The development and severity of FA symptoms was studied in sensitized germ-free (GF), conventional (CV), and mice mono-colonized with L. plantarum WCFS1 or co-housed with CV mice. MC were phenotypically and functionally characterized. Results: Systemic sensitization and oral challenge of GF mice with ovalbumin led to increased levels of specific IgE in serum compared to CV mice. Remarkably, despite the high levels of sensitization, GF mice did not develop diarrhea or anaphylactic hypothermia, common symptoms of FA. In the gut, GF mice expressed low levels of the MC tissue-homing markers CXCL1 and CXCL2, and harbored fewer MC which exhibited lower levels of MC protease-1 after challenge. Additionally, MC in GF mice were less mature as confirmed by flow-cytometry and their functionality was impaired as shown by reduced edema formation after injection of degranulation-provoking compound 48/80. Co-housing of GF mice with CV mice fully restored their susceptibility to develop FA. However, this did not occur when mice were mono-colonized with L. plantarum. Conclusion: Our results demonstrate that microbiota-induced maturation and gut-homing of MC is a critical step for the development of symptoms of experimental FA. This new mechanistic insight into microbiota-MC-FA axis can be exploited in the prevention and treatment of FA in humans.
