Abstract:Cow milk (CM) allergy is a worldwide concern. Currently, few studies have been performed on the immunoreactivity of CM and fewer still on the antigenicity of CM in vivo and in vitro. In this study, we assessed the potential allergenicity of enzymatically hydrolyzed CM using in vitro ELISA and oral sensitization and challenge of BALB/c mice. Alcalase-, Protamex-, and Flavourzyme-treated CM (all from Novozymes) diminished IgE binding capacity, with greatest reductions of 56.31%, 50.62%, and 56.45%, respectively. Allergic symptoms and levels of total IgG1 were reduced, and allergic inflammation of the lung, jejunum, and spleen was relieved. Moreover, the numbers of CD8+ T and B220+ cells decreased, and the balance of CD4+ T/CD8+ T cells was effectively regulated. These findings suggest that the potential allergenicity of CM was reduced by enzymatic hydrolysis, and our research will lay a solid foundation for developing high-quality hypoallergenic CM products.

Abstract:αs1-Casein is a potential allergen to induce hypersensitivity in cow milk. We had identified αs1-casein and its epitopes in previous studies. The present study aimed to evaluate the allergic mechanism of αs1-casein in a BALB/c mouse model. The levels of specific IgE, mast cell proteinase, histamine and cytokines in sensitized mice were determined, and the clinical and pathological observation were evaluated. Results showed that the levels of specific IgE, mast cell proteinase, histamine, IL-4, IL-5, IL-10 increased significantly with a dose-dependent trend. The local alveolar septum collapsed or thickened, and lymphatic foci were produced in the spleen and thymus, and the inflammatory cells infiltrated in small intestinal mucosa mesenchyme. In conclusion, the levels of specific IgE, mast cell proteinase, histamine, IL-4, IL-5, IL-10 and some inflammatory factors could possibly serve as allergic biomarkers of αs1-casein, however, additional studies on signal transduction and gene expression are necessary in future.
Comparison of Sensitization Between Beta-Lactoglobulin and Its Hydrolysates.

Abstract:BACKGROUNDBovine beta-lactoglobulin is one of the first foreign antigens encountered by some newborn children, and it has been described as the main allergenic protein in cow's milk even when present at low concentration.OBJECTIVEEnzymatic hydrolysis has been identified to be a very effective way to reduce the sensitization of beta-lg compared to other treatments. The aim of this study was to explore whether enzymatic hydrolysis could reduce the allergenicity of native bovine beta-lg.METHODSbeta-lg was hydrolyzed by trypsin. Twenty four BALB/c mice were divided into three groups and orally sensitized by native bovine beta-lg and its hydrolysates five times at weekly intervals.RESULTSDuring the sensitization period, many serious systemic anaphylactic symptoms were observed in mice sensitized by native beta-lg compared to hydrolysates of beta-lg. Mice sensitized by hydrolysates of beta-lg showed a significantly lower spleen lymphocyte proliferation level than intact beta-lg. The beta-lg-specific IgE antibody levels in serum and intestinal fluid samples induced by native beta-lg were significantly elevated. Plasma histamine levels were also evaluated and showed the same trend as IgE. Moreover, the hydrolysates of beta-lg significantly up-regulated IFN-gamma and IL-10 production and down-regulated IL-4 and IL-5 secretions by murine splenocytes.CONCLUSIONThese results suggested that enzymatic hydrolysis could partly reduce the allergenicity of beta-lg.
Allergenicity evaluation of an extensively hydrolyzed infant formula based on cow milk protein

Abstract:Cow milk protein allergy (CMPA) is the most common food allergy in infants. Currently, hypoallergenic infant formulas on the market are mainly divided into extensively hydrolyzed whey protein formulas and extensively hydrolyzed casein formulas. There are few extensively hydrolyzed infant formulas (EHFs) with a similar protein composition to breast milk. Therefore, we developed a hypoallergenic infant formula based on extensively hydrolyzed cow milk protein (whey protein-to-casein ratio of 6 : 4) and evaluated its allergenicity in vitro and in vivo. The results showed that the antigenicity of EHF was significantly decreased. The levels of Treg and Th1 cells were increased, while the levels of Th2 cells, IgE and IgG1, plasma histamine and serum mast cell enzymes were significantly decreased. At the same time, the allergic symptoms of the jejunum and lungs of mice were relieved. This study provides a solution for the development of cow milk protein based hypoallergenic infant formulas.
A BALB/c Mouse Model for Assessing the Potential Allergenicity of Proteins: Comparison of Allergen Dose, Sensitization Frequency, Timepoint and Sex

Abstract:The present study was to investigate the possibility of using the BALB/c mouse as an animal model for assessing the potential allergenicity of proteins.Specific IgE and IgG1 against ovalbumin were induced by dosing BALB/c mice via intraperitoneal injection (absence of adjuvant). The effects of various allergen doses (5mg, 0.5mg or 0.05mg OVA), sensitization times (twice or five times), timepoints (day 14 or day 28) and sex (male or female) were studied. IL-4, IFN-γ, OVA-specific IgE and IgG1 were measured by enzyme-linked immunosorbent assay (ELISA).A general finding was that mice treated with 0.05mg OVA had the highest OVA-specific IgE and IgG1, statistically significant higher specific IgE and IgG1 were observed in groups sensitized five times than twice, OVA-specific IgE and IgG1 on day 28 were statistically higher than day 14, and higher IL-4 was observed in OVA-allergic mice than control mice.These results demonstrate that the BALB/c mouse model treated with 0.05mg OVA intraperitoneally on days 0, 3, 6, 9, 12 might be used for further experiments. OVA-specific IgE and IgG1 should be detected on day 28. Further studies including reproducibility and other conditions were required before using the BALB/c mouse model for assessing the potential allergenicity of proteins.
A Method for Screening Proteases That Can Specifically Hydrolyze the Epitope AA83-105 of Αs1-Casein Allergen.

Abstract:Milk protein hydrolysates are common in infant formula, but some of them retain allergenicity due to incomplete hydrolysis of the epitopes for milk allergens. This study explored a method for screening proteases that could specifically hydrolyze the epitope of αs1-casein allergen. Firstly, the αs1-casein epitope AA83-105 was synthesized by the solid-phase synthesis method. Then, after purification and identification, the complete antigen was prepared through coupling with bovine serum albumin (BSA) and was used to raise monoclonal antibodies (mAb) in BALB/c mice. Additionally, an indirect competitive-enzyme-linked immunosorbent assay (icELISA) was established. The mAb raised against αs1-casein protein was used as a control. The results showed that the purity of the synthetic epitope was >90%, and the coupling rate with BSA was 6.31. The mAb subtype is IgG1, with a titer of 1:320,000. The mAb reacted specifically with αs1-casein but did not cross-react with soybean protein. The linear regression equation of the competitive inhibition curve was y = −9.22x + 100.78 (R2 = 0.9891). The detection limit of icELISA method was more sensitive, and the method showed good accuracy and repeatability. The amounts of antigen residues in papain protease hydrolysates were relatively small, and the epitope fragment was detected in papain hydrolysate via mass spectrometry. This study provides ideas and methods for screening proteases that specifically hydrolyze the epitopes of milk allergens and also provides a superior foundation for the development of an advanced hypoallergenic formula.
Dietary Linolenic Acid Increases Sensitizing and Eliciting Capacities of Cow’s Milk Whey Proteins in BALB/c Mice

Abstract:α-Lactalbumin (BLA) and β-lactoglobulin (BLG) are the major whey proteins causing allergic reactions. Polyunsaturated fatty acids (PUFAs) stand among the extrinsic factors of the food matrix that can bind BLA and BLG and change their bioactivities, but their contribution to change the allergenic properties of these proteins has not been investigated. Here, we aimed to determine how PUFAs influence BLA and BLG to sensitize and trigger allergic responses in BALB/c mice. First, tricine–SDS–PAGE and spectroscopic assays identified that α-linolenic acid (ALA, as a proof-of-concept model) can induce BLA and BLG to form cross-linked complexes and substantially modify their conformation. Then, BALB/c mice (n = 10/group) were orally sensitized and challenged with BLA and BLG or ALA-interacted BLA and BLG, respectively. Allergic reactions upon oral challenge were determined by measuring clinical allergic signs, specific antibodies, levels of type-1/2 cytokines, the status of mast cell activation, and percentage of cell populations (B and T cells) in different tissues (PP, MLN, and spleen). Overall, systemic allergic reaction was promoted in mice gavage with ALA-interacted BLA and BLG by disrupting the Th1/Th2 balance toward a Th2 immune response with the decreased number of Tregs. Enhanced induction of Th2-related cytokines, as well as serum-specific antibodies and mast cell activation, was also observed. In this study, we validated that ALA in the food matrix promoted both the sensitization and elicitation of allergic reactions in BALB/c mice.

Abstract:The aim of this study was to compare the sensitization of αS1-CN in cow and goat's milk in a mouse model. Fifty mice were divided into Control group, adjuvant control group, cow's milk αS1-CN sensitized group, goat's milk αS1-CN sensitized group and cross sensitized group. Cow's and goat's milk αS1-CN were used to establish mice sensitization model. The results showed that, cow's milk αS1-CN had higher allergenicity than goat's milk αS1-CN, as can be seen in significantly increased s-IgE and Th2 cell-related inflammatory factors, the proportion of Th2, and the expression of Th2 cell-related transcription factors. Furthermore, the sensitization of cow's milk αS1-CN damaged the intestinal barrier of mice, caused the leakage of LPS, activated the TLR4-NFκB pathway, and thus resulted in the increase of IFN-γ. In addition, mice allergic to cow's milk αS1-CN were less sensitized to goat's milk αS1-CN.

Abstract:αs1-Casein (αs1-CN) is a major cow milk allergen, while the tertiary structure of αs1-CN and conformational epitopes of αs1-CN have not been clarified. Here, a reasonable three-dimensional structure of αs1-CN was established using ab initio methods, and hot-spot residues and epitopes were investigated by combining molecular dynamics simulation, peptides synthesis, and ELISA. Obtained results demonstrated that the binding mechanism between αs1-CN and IgG was located on three main regions: a helical structure zone (E77-Q97), the flexible loop zone (Y154-T174), and a flexible C-terminal (N190-L198), mainly connecting via hydrogen bond and ionic bonds. The hydrolysates produced by papain with lowest antigenicity (12.43%), which could considerably destroy the essential epitopes of αs1-CN confirmed by epitope synthesis, and LC-MS/MS. The results reported herein would provide novel insights into the interface interactions between αs1-CN and IgG, and prove valuable for developing hypoallergenic infant-formula and peptide vaccines for allergen-specific immunotherapy.

Abstract:Background Soybean protein is used in a number of food products but is also a common cause of food allergy. Soybean glycinin and beta-conglycinin represent up to one-third of protein in the soybean. Many reports have indicated that glycinin and beta-conglycinin have been characterized as major soybean allergens involved in food hypersensitivity.Objective To investigate oral allergy syndrome and anaphylactic reactions in BALB/c mice caused by soybean glycinin and beta-conglycinin with an intragastric feeding protocol without using an adjuvant.Methods BALB/c mice were sensitized by gavages with glycinin and beta-conglycinin, and allergen-specific IgE and IgG1 responses were studied by a passive cutaneous anaphylaxis assay. Serum histamine release and blood pressure were measured according to other methods. Epithelium and mast cell dye used the method of light microscopy.Results Sensitization with soybean allergens induced high levels of antigen-specific IgE and IgG1 and increased serum histamine in BALB/c mice. Percentiles of intact mast cell of small intestine in mice sensitized with glycinin and beta-conglyinin significantly decreased for 28 days. Degranulation of mast cells and damage of the epithelium in the small intestine of mice sensitized with globulins were observed. The level of blood pressure in sensitized mice reached a minimum at 3 h.Conclusion Soybean-specific IgE and IgG1 antibodies increased, with high levels of histamine release, severe degranulation of mast cells and damage of the epithelium of small intestine in mice sensitized with glycinin and beta-conglyinin.

Abstract:α-Lactalbumin represents one of the major allergens causing cow milk allergy.The identification of the epitopes of α-lactalbumin and the critical amino acids for its allergenicity is of great significance for understanding the mechanism of action of α-lactalbumin and developing hypoallergenic dairy products.In this study,a series of peptides were synthesized by a solid phase method for the characterization of immunological epitopes and critical amino acids.The immunoglobulin (Ig) E-binding epitopes were immunolabeled with individual sera from cow milk-allergic patients as probes by enzyme linked immunosorbent assay (ELISA).Alanine scanning of immunodominant epitopes was used to identify the critical amino acids (aa).The results showed that IgE-binding epitopes were located within the sequences of aa1-15,aa6-20,aa46-60,aa71-85 and aa101-115.Our initial data revealed that Val8,Phe9,Arg10,Tyr103,Leu105 and His107 were the critical amino acids for IgE-binding epitopes.This study will provide the necessary information to alter the cDNA to encode a protein capable of activating milk-specific T cells,but with reduced IgE-binding capacity.
Characterization of Critical Amino Acids in Linear Immunoglobulin G Epitopes of α-Lactalbumin in Cow Milk

Abstract:α-Lactalbumin is one of the major allergens in cow milk. Characterizing the critical amino acids in the epitopes of α-lactalbumin is of great significance for the understanding of the role of immunoglobulin G (IgG) in milk allergy and the development of hypoallergenic dairy products. In this study, the alanine scanning of immunodominant epitopes was used to identify the critical amino acids. First, a series of peptides based on the sequence of α-lactalbumin were identified with IgG in individual sera from cow milk-allergic patients by enzyme-linked immunosorbent assay (ELISA). Then, new peptides were synthesized by sequential replacement of the amino acids acting on the epitopes with alanine, and the critical amino acids were identified with serum pool from cow milk-allergic patients by ELISA. The results showed that IgG-binding epitopes were located in the sequences of aa 6-20, aa 21-35, aa 36-50 and aa 86-100. Our data revealed Phe9, Leu15, Pro24, Trp26and His32were the critical aa in the IgG-binding epitopes.
Study on Main Allergen in Cow Milk Protein Allergy Children

Abstract:Objective:to study the species and contents of main allergen of cow's protein allergy children.Methods:cow' milk antibodies of Ig were measured with an indirect and complete enzyme-linked immunosorbent assay(ELISA).Results:the patients with CMPA showed that the levels of IgG class antibodies to αs-CN,BSA and β-LG was significantly associated with CMPA.The result of measuring specific IgG4 showed that β-LG and BSA were main allergen.Linear regression multianalysis showed thatαs-CN,CN and β-LG had significantly contribution to CMPA.Unstandardized coefficients ofαs-CN was maxim(B=-0.589),β-LG's was least(B=-0.271).Conclusion:β-lactoglobin,αs-casein,casein and bovine serum albumin were probably main antigen of children in Heilongjiang.

Abstract:Glycinin and beta-conglycinin are major soybean allergens involved in food hypersensitivity. However, the mechanism of immune responses induced by glycinin and -conglycinin has not been fully understood. Balb/c mice were oral sensitised with different doses (0.1, 1.0 and 10 mg/day) of soybean glycinin and -conglycinin for five weeks. Allergen-specific immunoglobulin (Ig), serum histamine and T-cell responses were tested to assess the allergenic activity of glycinin and -conglycinin. Mice sensitised with 0.1 or 1.0 mg/day allergens induced high levels of specific IgE, IgG1 and serum histamine compared with mice treated with saline. Furthermore, specific T-cell proliferation and significant up-regulation of interleukin (IL)-4, IL-5 and interferon (IFN)- were observed in splenocytes from mice gavaged with 0.1 or 1.0 mg/day soybean proteins. Low doses of glycinin or -conglycinin can induce allergic reactions in BALB/c mice, which might be associated with increased IgE and cytokine production.
Effects of different casein-derived peptides and their separated fractions on mouse cell immunity

Abstract:The effects of casein trypsin, pepsin, alcalase, pepsin-trypsin, papain hydrolysates and their separated fractions on the lymphoproliferation activity and Interleukin 2 (IL-2) release of mouse spleen cells were studied. Results showed that the immunoregulatory activities of trypsin, pepsin, pepsin-trypsin, and alcalase hydrolysates were much superior to that of papain hydrolysate. The peptide yield of pepsin hydrolysate was much lower than that of the others. The separated fractions of alcalase, or pepsin-trypsin hydrolysate had synergistic effect on the lymphoproliferation activity and IL-2 release of mouse spleen cells. Considering the price of alcalase was much lower than that of the others, especially animal enzymes, the use of alcalase in the in vitro production of casein hydrolysate was more practical.

Abstract:A prerequisite for systemic hyporesponsiveness to dietary antigens is their processing in the gut. This study investigated whether bovine caseins degraded by enzymes of an intestinal bacterial strain, Lactobacillus GG (ATCC 53103), could regulate the cytokine production by anti-CD3 antibody-induced peripheral blood mononuclear cells of 14 atopic patients, aged 5-29 (mean, 16) months. Purified casein up-regulated the interleukin-4 and interferon-gamma production, P = 0.008 and P = 0.008, respectively. Conversely, Lactobacillus GG-degraded casein down-regulated the interleukin-4 production, P = 0.003, with no effect on interferon-gamma. These results indicate that intestinal bacteria may modify immunomodulatory properties of native food proteins and introduce a promising tool to provide protection from potentially harmful dietary antigens at a young age.

Abstract:Capacity and ability of Lactiplantibacillus plantarum and Lacticaseibacillus rhamnosus probiotic strains isolated from some human and food sources were evaluated for modulating and regulating the immune system against hypersensitivity type 1 (dependent on IgE). In this study, given the probiotic properties of the strains and the use of mouse models, the strains were orally administered (1×109 cfu/ml) during 45 days with gavage needles. Levels of total IgE antibodies, cytokines TGF-β, INF-ɤ, and IL-4 were measured by ELISA and compared with control groups. In addition, the nasopharyngeal lavage (total and differential count) was evaluated. Among the strains, Lactiplantibacillus plantarum a7 and Lacticaseibacillus rhamnosus M1 had the highest suppression of IL-4 cytokine production in splenocytes from ovalbumin-sensitized mice in vitro. Also, a7 and M1 had a significant (P < 0.0001) effect on reducing total IgE levels compared to the control group (OVA). Further, Lactiplantibacillus plantarum M8, a7 and M1 enhanced the production of INF-ɤ and TGF-β cytokines. Examination of nasal lavage results revealed that the total cell count in the a7, M1 and Lacticaseibacillus rhamnosus RHM groups and eosinophil cells in Lactiplantibacillus plantarum LF57, a7, and M1 significantly decreased compared to the control group. It was observed oral feeding of the isolated strains had a beneficial effect on reducing the total count on Plate Count Agar (PCA) and Mac Conkey Agar in mice feces during 7 days. M1 and a7 showed the highest level of Lactic Acid Bacteria at 7.28 and 6.9 Log cfu/g on day14, respectively. In conclusion, the animals that received the strains M1 and a7 (isolated from cow's milk and infant's small intestine) had the highest count of lactic acid bacteria in the mice gastrointestinal tract on day 14 (7.28 and 6.9 Log cfu/g), respectively. In addition, the mentioned strains can modulate immune system of mice by suppressing IL-4 production, increasing INF-ɤ and TGF-β cytokines, and reducing total IgE levels.
Inulin has a beneficial effect by modulating the intestinal microbiome in a BALB/c mouse model

Abstract:Food allergy is an important health problem that affects human quality of life and socioeconomic development, and its treatment requires improvement. Intestinal flora dysbiosis is closely associated with food allergies. A sensitised mouse model was established by the intraperitoneal injection of ovalbumin (OVA). The mice were randomly divided into four groups: control, model, high-dose (H), and low-dose (L) inulin. The mice were administered water containing different concentrations of inulin four weeks before the OVA injection. Body weight changes were monitored. After the last OVA injection, the mice were scored for allergic reactions. The levels of total immunoglobulin E (IgE) and diamine oxidase (DAO) in the serum and secretory IgA (sIgA) in the small intestinal mucus were measured, and 16S rRNA sequencing of the faecal flora was performed to evaluate microbial parameters. The intestinal flora biomarkers, correlations between them, and biochemical indicators were analysed. Inulin treatment had no effect on the body weight of OVA-sensitised mice but attenuated allergic reactions and intestinal injury in mice. Compared with the control group, the model group had significantly higher levels of serum DAO and IgE and significantly lower levels of intestinal mucus IgA. IgA levels in the intestinal mucus of mice treated with inulin prior to OVA sensitisation were higher than those in non-inulin-treated OVA-sensitised mice. Furthermore, analysis of operational taxonomic units showed that inulin treatment decreased the abundance of Alloprevotella, Rikenellaceae RC9, Eubacterium siraeum, and Eubacterium xylanophilum, and increased the abundance of Blautia and Lachnospiraceae. Serum DAO levels were positively associated with Eubacterium siraeum, Alloprevotella, Eubacterium xylanophilum, and Odoribacter and negatively associated with Blautia, Tyzzerella, Alistipes, Desulfovibrionaceae, and Ruminococcaceae UCG005. In addition, IgE levels were positively associated with Eubacterium siraeum, Alloprevotella, Eubacterium xylanophilum, Odoribacter, and Citrobacter and negatively associated with Blautia, unclassified Ruminococcaceae, and Alistipes. IgA exhibited significant positive correlation with Blautia, norank_f_Eubacterium coprostanoligenes, Alistipes, norank Desulfovibrionaceae, Muribaculum, and Ruminococcaceae U C G 005 and significant negative correlation with Eubacterim siraeum, Eubacterium xylanophilum, Odoribacter, and Citrobacter. Inulin exerts a protective effect against food allergies in mice, which is partially mediated by alterations in the gut microbiota.
