Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.

Abstract:This study examines how temperature influences the response of Japanese tree frogs (Dryophytes japonicus) to microplastic (MP) pollution, assessing whether temperature can regulate the harmful effects of MPs on their life history and the dispersal of MPs across habitats. This analysis aims to understand the ecological and physiological ramifications of MP pollution. Our results demonstrated an ontogenetic transfer of MP particles across amphibian metamorphosis, possibly allowing and facilitating the translocation of MPs across ecosystems. Temperature did not significantly affect the translocation of aquatic MPs to land. However, high temperatures significantly reduced mortality and hindlimb deformities caused by MPs, thereby mitigating their harmful impact on amphibian life histories. Importantly, our study found that MPs cause hindlimb deformities during amphibian metamorphosis, potentially linked to oxidative stress. Additionally, MP exposure and ingestion induced a plastic response in the morphology of the digestive tract and changes in the fecal microbiome, which were evident at high temperatures but not at low temperatures. The effects of MPs persisted even after the frogs transitioned to the terrestrial stage, suggesting that MPs may have complex, long-term impacts on amphibian population sustainability. Our results enhance the understanding of the intricate environmental challenges posed by MPs and underscore the significant role of temperature in ectotherms regarding ontogenetic impacts and pollutant interactions.

Abstract:The first known totally noncontact optoacoustic measurement in a flame, using a pulsed laser to excite an acoustic pulse and observing the transient deflections of cw probe beams to detect the acoustic propagation, is described. All laser beams are coplanar, parallel, suitably displaced, and focused inside the flame. Abstract:Small plastic debris (0.1 μm-5 mm) or microplastics (MPs) have become major pollutants of aquatic ecosystems worldwide and studies suggest that MPs exposure can pose serious threats to human and wildlife health. However, to date the potential biological impacts of MPs accumulating in low amount in tissues during early life remains unclear. Here, for a more realistic assessment, we have used environmentally representative, mildly weathered, polyethylene terephthalate microplastics (PET MPs), cryomilled (1-100 μm) and fluorescently labelled. We leveraged the amphibian Xenopus laevis tadpoles as an animal model to define the biodistribution of PET MPs and determine whether exposure to PET MPs induce perturbations of antiviral immunity. Exposure to PET MPs for 1-14 days resulted in detectable PET MPs biodistribution in intestine, gills, liver, and kidney as determined by fluorescence microscopy on whole mount tissues. PET MPs accumulation rate in tissues was further evaluated via a novel in situ enzymatic digestion and subsequent filtration using silicon nanomembranes, which shows that PET MPs rapidly accumulate in tadpole intestine, liver and kidneys and persist over a week. Longer exposure (1 month) of tadpoles to relatively low concentration of PET MPs (25 μg/ml) significantly increased susceptibility to viral infection and altered innate antiviral immunity without inducing overt inflammation. This study provides evidence that exposure to MPs negatively impact immune defenses of aquatic vertebrates.

Abstract:In recent years, much research has focused on studying the damage caused by microplastics to the ecological environment and human health. Indeed, MPs are often consumed by shellfishes and zooplanktons due to their similarity in size to POM (particular organic matter). Especially in zooplankton, the accumulation of MPs in the body affects the reproductive system and the growth rate of juveniles. Moreover, toxins derived from MPs are continuously accumulated in predators of zooplankton and impact the whole ecosystem across the food chain. In this work, we found that even though MPs were internalized by and adherent around Daphnia magna, there were no significant differences in the survival rate of their adults and offspring. However, the population of ovigerous adults under high MPs exposure for 7 days decreased significantly, suggesting an extension of the days of sexual maturity in D. magna. The removal of MPs after 7 days’ MPs treatment resulted in an increase in D. magna juveniles and neonates which indicated their growth was reduced or inhibited in the MPs environment. Overall, the uptake of MPs led to negative effects on population reproduction and the growth of offspring in D. magna.

Abstract:Like many amphibians, wood frog (Lithobates sylvaticus) populations have likely declined or experienced local extirpations as a result of habitat alterations. Despite this, wood frogs are still present and breeding in altered landscapes, like the agricultural Prairie Pothole Region of central Canada, and are exposed to a variety of anthropogenic impacts. As tadpoles, water contamination can have negative effects on growth, development, and immune systems. To investigate the potential effects of agricultural land use on tadpole growth and immune system stress, we used boosted regression trees to model body mass, body condition, and neutrophil to lymphocyte ratios, a measure of immune stress, against 32 variables including water quality, wetland habitat, and landscape-level measures. Developmental stage strongly influenced all 3 endpoints, and body mass was negatively influenced by higher levels of total dissolved solids (>600-700 mg/L) and at the first sign of pesticide detection (>0.01 proportion pesticides detected of those screened). While correlative, these data suggest that tadpoles developing in agricultural environments may experience survival and reproductive disadvantages if they metamorphose at smaller body sizes. Given the potential impacts this can have on adult frogs and frog populations, these results provide an impetus for further field-based investigation into the effects that pesticides, and especially total dissolved solids, may have on tadpoles. Abstract:The toxic effects of nanoparticles have been increasingly investigated, but there has been limited research on amphibians, especially those of conservation value. This study examined the effects of different concentrations (0, 0.04, 0.2, 1, 5 mg/L) of polystyrene nanoplastics (PS-NPs, 80 nm) on the short-term exposure (7 d) of Andrias davidianus. Results demonstrated the concentration-dependent enrichment of PS-NPs in the intestine. Histological lesions displayed increased hepatic macrophages with cellular rupture, broken intestinal villi, decreased cuprocytes and crypt depression. Antioxidant- and inflammation-related enzyme activities were analysed, and it was found that hepatic and intestinal MDA content and CAT activity were highest in the N-1 group and SOD activity was highest in the N-0.2 group (p < 0.05). AKP activity continued to decline, and iNOS activity was highest in the N-0.2 group (p < 0.05). il-10, tgf-β, bcl-w and txnl1 were significantly downregulated in the N-0.2 group, while il-6 and il-8 were markedly upregulated in the N-0.2 group (p < 0.05). Exposing to PS-NPs decreased probiotic bacteria (Cetobacterium, Akkermansia) and increased pathogenic bacteria (Lachnoclostridium). Our results suggest that NPs exposure can have deleterious effects on salamanders, which predicts that NPs contamination may lead to continued amphibian declines. Therefore, we strongly recommend that attention be paid to amphibians, especially endangered species, in the field of NPs.

Abstract:Microplastics (MPs) are a prolific contaminant in aquatic ecosystems across the globe. Zooplankton (including holoplankton and meroplankton) play vital ecological roles in marine and freshwater ecosystems and have been shown to readily consume MPs. The present review uses 88 pieces of published literature to examine and compare the effects of MPs on survival, growth, development, feeding rate, swimming speed, reproduction, organ damage and gene expression of different groups of zooplankton, including copepods, daphnids, brine shrimp, euphausids, rotifers and the larvae of fishes, sea urchins, molluscs, barnacles, decapods and ascidians. Among the groups studied, daphnids and copepods are the most sensitive to MPs, with their feeding rate and fecundity significantly decreased at environmentally relevant MP concentrations. This might adversely affect daphnid and copepod populations in the long term. In contrast, molluscs, barnacles, brine shrimp and euphausids appear to be more tolerant to MPs. No clear impacts on survival, development time, growth or feeding rate can be observed in these zooplankton groups at any of the MP concentrations tested, suggesting that these groups might become more dominant with prolonged exposure to MP pollution. Leachates derived from MPs can induce severe abnormality in bivalve and sea urchin embryos. MPs have prominent effects on survival and fecundity of F1 offspring in bivalves, copepods and daphnids, indicating that MPs could incite transgenerational effects and drastically affect sustainability in zooplankton populations. Abstract:The prevalence of microplastics (MPs) in global aquatic environments has received considerable attention. Currently, concerns have been raised regarding reports that the adverse effect of MPs on aquatic animals in the exposure phase may not be (completely) reversed in the depuration phase. In order to provide insights into the legacy effect of MPs from the depuration phase, this study evaluated the kinetic characteristics and recovery potential of aquatic animals after the exposure to MPs. More specifically, a total of 68 depuration kinetic curves were highly fitted to estimate the retention time of MPs. It was shown that the retention time ranged from 1.26 to 3.01 days, corresponding to the egestion of 90 % to 99 % of ingested MPs. The retention time decreased with the increased retention rate. Furthermore, variables potentially affecting the retention time were ranked by the decision tree-based eXtreme Gradient Boosting (XGBoost) algorithm, suggesting that the particle size and tested species were of great importance for explaining the difference in retention time of MPs. Moreover, a biomarker profile was recompiled to determine the toxic changes. Results indicated that the MPs-induced toxicity significantly reduced in the depuration phase, evidenced by the recovery of energy reserves and metabolism, hepatotoxicity, immunotoxicity, hematological parameters, neurotoxicity and oxidative stress. However, the continuous detoxification and remarkable genotoxicity implied that the toxicity was not completely alleviated. In addition, the current knowledge gaps are also highlighted, with recommendations proposed for future research.

Abstract:In the aquatic environment, microplastic particles (MP) can accumulate in microbial communities that cover submerged substrata, i.e. in periphyton. Despite periphyton being the essential food source for grazers in the benthic zones, MP transfer from periphyton to benthic biota and its ecotoxicological consequences are unknown. Therefore, in this study, we investigated the effects of 1) MP on embryonal development of freshwater gastropod Physa acuta embryos, 2) MP on adult Physa acuta individuals through dietary exposure and 3) on the MP surface properties. Embryonal development tests were carried out with spherical polyethylene MP in the size of 1–4 μm (MP). Over a period of 28 days, embryonal development and hatching rate were calculated. In the feeding experiments, periphyton was grown in the presence and absence of MP and was then offered to the adult Physa acuta for 42–152 h. The snails readily ingested and subsequently egested MP, together with the periphyton as shown by MP quantification in periphyton, snail soft body tissue and feces. No selective feeding behavior upon MP exposure was detected. The ingestion of MP had no effect on mortality, feeding and defecation rate. Yet, the reproductive output of snails, measured as the number of egg clutches and numbers of eggs per clutch, decreased after the ingestion of MPs, while the hatching success of snail embryos those parents were exposed remained unaffected. In contrast, hatching rate of snail embryos was significantly reduced upon direct MP exposure. MP optical properties were changed upon the incorporation into the periphyton and the passage through the digestive tract. Our results indicate that MP incorporated in periphyton are bioavailable to aquatic grazers, facilitating the introduction of MP into the food chain and having direct adverse effects on the grazers' reproductive fitness.

Abstract:Glyphosate’s primary metabolite, aminomethylphosphonic acid (AMPA), is the most detected pollutant in surface waters. Recent studies have raised concerns about its toxicity, yet underlying mechanisms remain poorly understood. A disruption of the gut microbiome, which plays a crucial role in host health, could mediate most of the adverse effects. We investigated the impact of AMPA exposure on the gut microbiome of spined toad tadpoles ( ). We hypothesized that AMPA could alter the gut microbiota composition and that these effects could depend on the microbiota source. We exposed tadpoles to minute concentrations of AMPA and analyzed their faecal microbiota using 16S rRNA gene sequencing as a proxy of the gut microbiota. AMPA exposure decreased the gut bacterial biomass and affected the bacterial community composition of tadpole’s faeces. Furthermore, we observed interactions between AMPA exposure and maternal body condition on the Bacteroidota and Actinobacteriota phyla abundances. This suggests a maternal effect on early-life microbial colonizers that could influence the response of the gut microbiome to AMPA. These findings highlight the importance of considering the gut microbiome when studying the effects of environmental contaminants. Further research is needed to elucidate the long-term implications of this microbiome alteration for amphibian health.

Abstract:Micro(nano)plastics (MNPs) are a growing problem as persistent environmental pollutants. Here, we investigated the impact of MNPs on microorganisms in aquatic microbial floc exposed to NPs (80 nm) and MPs (8 mu m) for 35 days. Water quality indicators were tested weekly and microbiological analyses were conducted on Day 7 and 28 after exposure. The results showed that there were significant differences in the levels of total ammonia nitrogen or nitrite between the MNPs groups and the control group, spanning from Day 7 to Day 28. For the microbial response, microbial community richness in the NPs and MPs groups were significantly increased at Day 7. Functional prediction showed that the relative abundances of bacteria associated with the "Forms Biofilms", "Potentially Pathogenic", "Plastic Degradation" and nitrogen cycle processes were significantly different after MNPs exposure. The results suggest that MNPs had no significant effect on the microbial diversity of mature microbial flocs. Findings suggest MPs could cause an increase in the relative abundance of potentially pathogenic bacteria, while NPs do not. In addition, stress associated with MNPs affected the nitrogen cycle of microorganisms, and NPs exerted greater impacts than MPs. Findings from this study further our understanding of the impact of MNPs at environmentally relevant concentrations on microorganisms in aquatic ecosystems.
Toxicity of Pb continuous and pulse exposure on intestinal anatomy, bacterial diversity, and metabolites of Pelophylax nigromaculatus in pre-hibernation

Abstract:Amphibians are often exposed to pulse pollution due to fluctuational inputs of pollutants in water. Traditional ecotoxicology research is mainly performed under constant exposure conditions, which is not consistent with the true environmental pollution. Frogs are sensitive to changes in water pollutants in pre-hibernation. Thus, to understand the toxicity difference to continuous and pulse exposure in environmental concentrations of Pb (100 μg/L), Pelophylax nigromaculatus adults were exposed to short-term treatments (8 days) in pre-hibernation. Individual mortality, intestinal anatomical structure, bacterial diversity, and metabolites were measured in a control group (CON), a Pb continuous treatment group (CEPb) and a Pb pulse treatment group (PEPb). The results showed that PEPb significantly increased individual mortality, compared to the control group and CEPb. PEPb induced pathological changes in the small intestinal tissues, such as mucosal erosion, swollen and distorted villi, large vacuoles, and the proliferation of goblet cells. In addition, PEPb altered the structure and diversity of intestinal bacteria, resulting in an increase in some pathogenic bacteria (e.g. Bacteroides and Ruminococcus) and a decrease in beneficial bacteria (e.g. Cetobacterium and Akkermansia). Both CEPb and PEPb significantly changed intestinal metabolites and metabolic pathways. Moreover, PEPb has a significant effect on the metabolism of amino acids by increasing the content of 5-Aminopentanoic acid, cis-4-Hydroxy-l-proline, Glycocholic acid, l-Alanine, and l-Isoleucine. We concluded that PEPb may lead to intestine impairment of P. nigromaculatus in pre-hibernation by inducing intestinal structural integrity destruction, bacterial imbalance, and metabolic dysfunction, resulting in a significant increase in mortality. The study provides new insights for understanding the intestinal responses of frogs to pulse metal exposure.

Abstract:Genetic instability is promoted by unusual sequence arrangements and DNA structures. Hairpin DNA structures can form from palindromes and from triplet repeats, and they are also intermediates in V(D)J recombination. We have measured the genetic stability of a large palindrome which has the potential to form a one-stranded hairpin or a two-stranded cruciform structure and have analyzed recombinants at the molecular level. A palindrome of 15.3 kb introduced as a transgene was found to be transmitted at a normal Mendelian ratio in mice, in striking contrast to the profound instability of large palindromes in prokaryotic systems. In a significant number of progeny mice, however, the palindromic transgene is rearranged; between 15 and 56% of progeny contain rearrangements. Rearrangements within the palindromic repeat occur both by illegitimate and homologous, reciprocal recombination. Gene conversion within the transgene locus, as quantitated by a novel sperm fluorescence assay, is also elevated. Illegitimate events often take the form of an asymmetric deletion that eliminates the central symmetry of the palindrome. Such asymmetric transgene deletions, including those that maintain one complete half of the palindromic repeat, are stabilized so that they cannot undergo further illegitimate rearrangements, and they also exhibit reduced levels of gene conversion. By contrast, transgene rearrangements that maintain the central symmetry continue to be unstable. Based on the observed events, we propose that one mechanism promoting the instability of the palindrome may involve breaks generated at the hairpin structure by a hairpin-nicking activity, as previously detected in somatic cells. Because mammalian cells are capable of efficiently repairing chromosome breaks through nonhomologous processes, the resealing of such breaks introduces a stabilizing asymmetry at the center of the palindrome. We propose that the ability of mammalian cells to eliminate the perfect symmetry in a palindromic sequence may be an important DNA repair pathway, with implications regarding the metabolism of palindromic repeats, the mutability of quasipalindromic triplet repeats, and the early steps in gene amplification events.
Effects of microplastics on key reproductive and biochemical endpoints of the freshwater microcrustacean Daphnia magna

Abstract:Human activities have directly impacted the environment, causing significant ecological imbalances. From the different contaminants resulting from human activities, plastics are of major environmental concern. Due to their high use and consequent discharge, plastics tend to accumulate in aquatic environments. There, plastics can form smaller particles (microplastics, MPs), due to fragmentation and weathering, which are more prone to interact with aquatic organisms and cause deleterious effects, including at the basis of different food webs. This study assessed the effects of two microplastics (polyethylene terephthalate, PET; and polypropylene, PP; both of common domestic use) in the freshwater cladoceran species Daphnia magna. Toxic effects were assessed by measuring reproductive traits (first brood and total number of offspring), and activities of biomarkers involved in xenobiotic metabolism (phase I: cytochrome P-450 isoenzymes CYP1A1, 1A2 and 3A4; phase II/conjugation: glutathione S-transferases; and antioxidant defense (catalase)). Both MPs showed a potential to significantly reduce reproductive parameters in D. magna. Furthermore, PET caused a significant increase in some isoenzymes of CYP450 in acutely exposed organisms, but this effect was not observed in chronically exposed animals. Similarly, the activity of the antioxidant defense (CAT) was significantly increased in acutely exposed animals, but not in chronically exposed organisms. This pattern of effects suggests a possible mechanism of long-term adaptation to the presence of the tested MPs. In conclusion, the herein tested MPs have shown the potential to induce deleterious effects on D. magna mainly observed in terms of the reproductive outcomes. Changes at the biochemical level seems transient and are not likely to occur in long term, environmentally exposed crustaceans.
Antitumor activities and immunomodulatory effects of ginseng neutral polysaccharides in combination with 5-fluorouracil.

Abstract:A neutral polysaccharide fraction (WGPN) prepared from Panax ginseng C.A. Meyer by hot water extraction and DEAE-cellulose chromatography was tested for its anticancer activity alone and in combination with 5-fluorouracil (5-FU) in Sarcoma-180 (S180) tumor-bearing mice by intragastric administration. WGPN alone inhibited S180 tumor growth in a bell-shaped dose-response curve, and the combination with 5-FU showed a synergistic effect. Studies of various immunological activities in S180-bearing mice revealed that WGPN stimulated the proliferation of lymphocytes, increased natural killer cell cytotoxicity, enhanced the phagocytosis and nitric oxide production by macrophages, and increased the level of tumor necrosis factor-alpha in serum. In combination with 5-FU, WGPN mitigated damage to the immune system caused by 5-FU in S180-bearing mice. These results suggest that WGPN might be a potential adjuvant for chemotherapeutic drugs.
Effects of Microplastics on Immune Responses of the Yellow Catfish Pelteobagrus fulvidraco Under Hypoxia

Abstract:Compared with marine organisms, research on microplastics (MPs) in freshwater organisms is still less although MPs have been widely found in the freshwater ecosystem. Hypoxia is a ubiquitous issue in freshwater aquaculture, and under such scenarios, the toxic effects of MPs on typical aquaculture fish need to be clarified. In this study, we studied the effects of MPs (polystyrene) on specific growth rate (SGR), hypoxia-inducible factor-1α (HIF-1α), tumor necrosis factor-α (TNF-α), interleukin-8 (IL-8), and interferon (IFN) in the yellow catfish ( Pelteobagrus fulvidraco ) under hypoxic conditions. After 15 days of exposure, the SGR was not affected by MPs or hypoxia. MPs significantly increased the expressions of HIF-1α and TNF-α but inhibited the expression of IFN at high concentration MPs under normoxia. However, hypoxia significantly inhibited the expression of IL-8 and TNF-α under high MP concentration and low MP concentration, respectively. In addition, MPs had significant concentration-dependent inhibitory effects on IFN under hypoxia. Surprisingly, a positive correction between HIF-1α and TNF-α was found in fish. Although hypoxia might alleviate the effects of MPs with low concentrations, the interaction of hypoxia and MPs aggravated the negative effects of MPs on immune factors at high concentration MPs. This study provided new insight into the complex effects of hypoxia and MPs on aquatic organisms, and future studies should focus on the cellular pathways of immune cells in fish. Given that MPs could induce the immune response in fish, considerations should be paid to the impacts of MPs on freshwater aquaculture, and hypoxia should be taken into consideration when evaluating the effects of MPs.

Abstract:The occurrence of microplastics (MPs) in aquatic ecosystems and their ability to absorb hydrophobic pollutants, such as persistent organic pollutants (POPs), is currently a significant concern. MPs, which are the main breakdown product of plastics, have been frequently detected in the environment, posing serious threats to organisms' health. One particular pollutant, 2,2',4,4'-tetrabromodiphenyl ether (BDE-47), is a dominant congener of PBDEs and is highly toxic to organisms. However, there is limited knowledge regarding the exposure of marine fishes to PBDEs through MPs and their combined toxic effects. In this study, the embryo toxicity of Hexagrammos otakii was conducted to investigate the combined effects of MPs and BDE-47. The results showed that MPs and BDE-47 co-exposure had detrimental effects on embryonic development, such as reduced hatchability, increased mortality, decreased heart rate, and body malformation. Moreover, the combined toxicity of these substances appeared more pronounced harmful effects compared to exposure to BDE-47 alone. Histopathological examination revealed that co-exposure can cause greater damage to hatching glands and yolk. The enrichment of Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways included phagosome, metabolism of xenobiotics by cytochrome P450, TCA cycle, and Wnt signaling pathway, which are closely related to embryonic growth. BDE-47 and MPs may activate the Wnt signaling pathway to affect the normal development of embryos. Our results suggest that MPs and BDE-47 exposure may cause growth disorders in the early life stages of H.otakii, leading to abnormal embryonic development. All these results will contribute to the further study of the ecological risk assessment and toxicity of MPs and organic pollutant mixtures in marine fish.
