Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.

Abstract:Aquatic ecosystems are among the main destination for microplastics (MPs) in the environment. MPs that enter aquatic ecosystems can contribute to pollution together with other co-existing pollutants. However, whether such pollution results in higher or lower toxicity to fish than that caused by co-existing pollutants alone remains controversial. This study aimed at closing this research gap based on 1380 biological endpoints under the background of environmental MP concentrations collected from 55 laboratory studies. Overall, MPs in co-existing pollutant solutions significantly increased the toxicity to fish. Specifically, MPs elevated negative effects on the immune system, metabolism, and oxidative damage. Subgroup analysis indicated that changes in toxicity were related to fish life stage and MP size, but not to co-existing pollutant or MP type. Meta-regression analysis indicated that changes in toxicity were not related to the logarithm of the octanol-water partition coefficient (logKow) or exposure time. Finally, the differences between laboratory research and the actual aquatic environment were discussed from four aspects: MPs, co-existing pollutants, environmental factors, and experimental objects. Our study provides a basis for further understanding the potential impact of MPs on aquatic organisms from a combined pollution perspective. Moreover, our results can provide a reference for the conservation and management of aquatic ecosystems.

Abstract:Microplastics (MPs), the emerging environmental pollutants, have attracted global attention due to the potential public health challenge and ecological security risk. Recent studies suggested liver as a vulnerable organ to MPs exposure, evidenced by abnormal hepatic lipid metabolism upon MPs intake in multiple animal species. How-ever, the specific changes of lipid metabolism in mammalian livers, as well as the underlying mechanisms, remain to be elucidated. In the present study, C57BL/6 mice were randomly assigned to normal drinking water or drinking water containing 100 mu g L-1 or 1000 mu g L-1 polystyrene (PS) MPs for 8 weeks. MPs exposure exerted no significant effect on body weight, serum triglyceride or total cholesteryl esters. However, mice showed impaired glucose tolerance and hepatic lipid deposition in response to high-dose MPs administration. Further lipidomic analysis showed significant alteration in hepatic lipid species particularly with free fatty acids (FFAs) and tri-acylglycerols (TAGs) in mice exposed to MPs. Meanwhile, the liver transcriptional profile indicated MPs exposure-induced differentially expressed genes (DEGs) were enriched in pathways of lipid metabolism and unfolded protein response. Furthermore, most altered lipid species were significantly correlated with DEGs enriched in lipid metabolic signaling. These findings provide lipidomic and transcriptional signatures of liver in response to MPs exposure, which will shed light on further understanding of the metabolic toxicity of MPs.

Abstract:The occurrence of microplastics (MPs) within aquatic ecosystems attracts a major environmental concern. It was demonstrated MPs could cause various ecotoxicological effects on microalgae. However, existing data on the effects of MPs on microalgae showed great variability among studies. Here, we performed a meta-analysis of the latest studies on the effects of MPs on photosynthesis and oxidative stress in microalgae. A total of 835 biological endpoints were investigated from 55 studies extracted, and 37 % of them were significantly affected by MPs. In this study, the impact of MPs against microalgae was concentration-dependent and size-dependent, and microalgae were more susceptible to MPs stress in freshwater than marine. Additionally, we summarized the biological functions of microalgae that are primarily affected by MPs. Under MPs exposure, the content of chlorophyll a (Chl-a) was reduced and electron transfer in the photosynthetic system was hindered, causing electron accumulation and oxidative stress damage, which may also affect biological processes such as energy production, carbon fixation, lipid metabolism, and nucleic acid metabolism. Finally, our findings provide important insights into the effects of MPs stress on photosynthesis and oxidative stress in microalga and enhance the current understanding of the potential risk of MPs pollution on aquatic organisms.

Incidence and management of biliary complications after 291 liver transplants following the introduction of transcystic stenting.

Abstract:BACKGROUND Biliary complications occur frequently after liver transplantation, and many are historically related to T tubes. Stents placed through the donor cystic duct have been used to attempt to reduce tube-related complications yet maintain access to the biliary tree. METHODS The outcomes of all liver transplant procedures performed at the University of Michigan between December 7, 1990 (when transcystic stenting was first used), and April 6, 1995, were analyzed retrospectively. Preoperative, perioperative, and postoperative variables were studied in relationship to biliary complications. The management of complications was also reviewed. RESULTS A total of 291 transplants qualified for study. The overall biliary complication rate was 25%, with no difference between the 237 patients who received transcystic stents, the 28 who received T tubes, and the 26 who received no tube. Among the complications patients experienced, 65% had stricture(s), 44% had stone or sludge formation, and 40% had a leak. Complications attributable solely to transcystic stents occurred in 4% of cases. Advanced age was the only preoperative variable associated with complications. Primary sclerosing cholangitis was associated with intrahepatic strictures, and prolonged cold ischemia time and rejection were associated with stone or sludge formation. Nonoperative management had the highest success rate for anastomotic stricture (76%) and the lowest for intrahepatic strictures (65%). Only one death was directly attributable to a biliary complication. CONCLUSION Transcystic stenting reduces the incidence of significant tube-related complications, but not the frequency of other biliary complications. Biliary complications can usually be managed percutaneously or endoscopically, although intrahepatic strictures and large, early leaks frequently require reoperation. Aggressive, early management of these complications can reduce excess mortality to less than 2%.

Abstract:Microplastics (plastic particles <5 mm; MPs) are ubiquitous in aquatic environments, and their ability to bind toxic chemicals has raised concern regarding their potentially adverse effects on ecosystems. However, the ecological impact of the MPs for aquatic organisms remains poorly understood because MPs usually pose limited toxic effects, which challenges the typical ecotoxicology tests. In this study, untargeted metabolomics was applied to determine the metabolic impact of MPs on a common freshwater microalga Microcystis aeruginosa after a 96-h exposure to polystyrene microbeads at combinations of different sizes and concentrations. Physiological parameters of the microalga were also determined. The results showed that MPs had no discernible effect on microalgal growth and photosynthesis, whereas oxidative stress and toxin production were slightly induced. The metabolomics analysis revealed variations in the global metabolic profile of microalgal cells exposed to MPs. Specially, we found that the larger rather than nano-sized MPs induced more differentially expressed metabolites. Metabolic pathway analysis reveals that the MPs disturb concrete metabolic pathways involved in amino acid synthesis, membrane formation, nitrogen storage, and antioxidant defense of the microalga. These observed metabolic impacts were probably related to the contact mode and contact probability between the microalgae and MPs. Our study suggests that omics-based approaches may provide insight and complement in quantifying MPs’ ecological impacts on the freshwater ecosystem.
Evidence-based meta-analysis of the genotoxicity induced by microplastics in aquatic organisms at environmentally relevant concentrations

Abstract:Microplastics (MPs) attract global concern due to their ubiquitous existence in aquatic environments. However, the genotoxic effect of MPs on aquatic organisms in the natural environment remains controversial. Therefore, this meta-analysis was conducted by recompiling 44 individual studies from 12 publications to determine whether MPs could induce genotoxicity in aquatic organisms at environmentally relevant concentrations (≤1 mg/L, median = 0.5 mg/L). Multiple genotoxic endpoints were involved, including the percentage of DNA in tail (TDNA%), tail length (TL), olive tail moment (OTM), and the number of micronuclei (NM), and their increases represented the biologically adverse effects (i.e. genotoxicity). The results showed that all included endpoints tended to increase after exposure to MPs, among which TDNA%, TL and NM were significantly increased by 20%, 32% and 81% compared with the control group, respectively. The overall estimate of all endpoints in the MPs-treated groups was remarkably increased by 24%, with high statistical power and no obvious publication bias, suggesting the evident genotoxicity caused by MPs. In addition, the magnitudes of MPs-induced genotoxicity were independent of selected endpoint, MP composition, morphology, exposure concentration and duration, but closely correlated with particle size, living habitat and tested species. Overall, this work provided a reference for the health risk assessment of MPs in the natural environment, contributing to our understanding the action mode of MPs at environmentally relevant concentrations.

Abstract:In recent years, microplastic (MP) pollution has been widely reported. Many studies have shown that MPs in the oceans not only exist in the water but also are eaten by plankton and may be transferred to other trophic levels via the plankton food web. This paper reviews the relationship between MPs and zooplankton, and discusses two aspects of the potential harm of MPs to zooplankton: the effects of MPs on zooplankton ingestion and the influence of MP toxicity on zooplankton. MPs are often eaten by zooplankton because they resemble prey in size, color, and buoyancy. Zooplankton ingest MPs directly or indirectly, resulting in intestinal damage, reduced ingestion, slow or delayed growth, reduced spawning, shortened lifespan, and abnormal or even fatal gene expression. MP ingestion by zooplankton also affects their behavior, reproduction, and offspring. MPs constitute an uncertain threat to zooplankton. Integration of MPs into the zooplankton community affects filtering organisms feeding. The mechanism of MP toxicity in zooplankton is not yet fully understood and more studies are needed.

Abstract:Microplastics (MPs) have received an increasing attention because of their ubiquitous presence and aquatic toxicity associated with MPs and MP-bound contaminants in the natural water. This review is to critically examine the chemical additives leached from MPs, the altered contaminant behaviors and the resulting changes in their aquatic ecotoxicity. Available data suggest that heavy metals Zn, Cr, Pb, and Cd regulated and present in plastics at the sub-mg g-1 to mg g-1 level can leach a significant amount depending on MPs size, aging, pH, and salinity conditions. MP-bound organic contaminants are primarily additive-derived (e.g., brominated diphenyl ethers, nonylphenol, and bisphenol A) at the µg g-1 to mg g-1 level, and secondarily pyrogenic and legacy origins (e.g., PAHs and PCBs) in the range of ng g-1 and mg g-1. MPs tend to have higher but more variable sorption capacities for organic compounds than metals (1.77 ± 2.34 vs. 0.82 ± 0.94 mg g-1). MPs alter the behavior of heavy metals through the electrostatic interactions and surface complexation, while the transport of additive derived organic compounds are altered primarily through hydrophobic effect as supported by a positive correlation (R2 = 0.71) between the logarithmic MPs-adsorbed concentrations and octanol/water partition coefficients (KOW) of organic compounds. MPs constitute less than 0.01% of the total mass of aquatic particulates in typical waters, but play a discernible role in the local partitioning and long-distance movement of contaminants. MPs alone exert higher toxicity to invertebrates than algae; however, when MPs co-occur with pollutants, both synergistic and antagonistic toxicities are observed depending mainly on the ingestibility of MPs, the extent of sorption, MPs as a transport vector or a sink to scavenge pollutants. We finally suggest several key areas of future research directions and needed data concerning the role of MPs in mitigating pollutant leaching, transport and risk under conditions mimicking natural and polluted waters.
Meta-analysis unravels the complex combined toxicity of microplastics and antibiotics in aquatic ecosystems

Abstract:The aquatic ecosystem, a repository for various pollutants, has been identified as a crucial zone where microplastics (MPs) serve as vectors for antibiotics, facilitating their spread. Despite this, the influence of MPs on the toxicity of antibiotics remains a topic of debate. In this study, we conduct a global meta-analysis, examining 730 datasets from 29 laboratory studies. Our findings reveal that the impact of MPs on antibiotic toxicity is highly dependent on biological response pathways, microplastic concentration, antibiotic properties, and exposure time. We observed that MPs amplify the accumulation of antibiotics in aquatic organisms, significantly heightening their adverse effects on growth, development, and immune functions. Intriguingly, MPs appear to mitigate the reproductive toxicity caused by antibiotics. A notable inverse relationship was identified between antibiotic toxicity and microplastic concentration and exposure time. Furthermore, antibiotic concentration predominantly affects growth, development, and reproductive health, whereas exposure time is critical in determining antibiotic accumulation and immune-related toxicity. These insights underscore that microplastic co-exposure can modify the toxicological profile of antibiotics. The outcomes of this research enhance our comprehensive understanding of the intricate combined effects of MPs and antibiotics on aquatic life, emphasizing the necessity for informed scientific management of these emerging contaminants.

Abstract:Microplastics (MPs) are a prolific contaminant in aquatic ecosystems across the globe. Zooplankton (including holoplankton and meroplankton) play vital ecological roles in marine and freshwater ecosystems and have been shown to readily consume MPs. The present review uses 88 pieces of published literature to examine and compare the effects of MPs on survival, growth, development, feeding rate, swimming speed, reproduction, organ damage and gene expression of different groups of zooplankton, including copepods, daphnids, brine shrimp, euphausids, rotifers and the larvae of fishes, sea urchins, molluscs, barnacles, decapods and ascidians. Among the groups studied, daphnids and copepods are the most sensitive to MPs, with their feeding rate and fecundity significantly decreased at environmentally relevant MP concentrations. This might adversely affect daphnid and copepod populations in the long term. In contrast, molluscs, barnacles, brine shrimp and euphausids appear to be more tolerant to MPs. No clear impacts on survival, development time, growth or feeding rate can be observed in these zooplankton groups at any of the MP concentrations tested, suggesting that these groups might become more dominant with prolonged exposure to MP pollution. Leachates derived from MPs can induce severe abnormality in bivalve and sea urchin embryos. MPs have prominent effects on survival and fecundity of F1 offspring in bivalves, copepods and daphnids, indicating that MPs could incite transgenerational effects and drastically affect sustainability in zooplankton populations. Abstract:Microplastics (MPs) are a class of pollutants that can persist for centuries, entering global ecosystems in large amounts. Previous studies have focused on the toxicological effects of MPs on single species or population scales from a narrow ecotoxicological perspective, while the systemic effects of MPs on the structure and function of global terrestrial and aquatic ecosystems are still unknown. In this review, we describe the fate and trophic transfer of MPs in ecosystems, which determines the possible scenarios of their interaction with ecosystems, highlighting the alteration of ecosystem structure and function by MPs. The presence of MPs in ecosystems changes the physicochemical properties of the abiotic environment and the habitat of organisms; these particles then directly or indirectly interact with organisms, leading to toxicological and ecological effects. Importantly, MPs, as a new global pressure, likely affect essential biogeochemical processes such as carbon and nitrogen processes. In addition, MPs affect energy flow by interfering with processes such as energy fixation and transfer in primary producers, energy acquisition, consumption and distribution in consumers, and energy metabolism in decomposers. The effects of MPs on information exchange alter the growth, development, and reproduction of organisms, as well as intraspecific and interspecific relationships. At the same time, as vectors for other global pollutants and pathogens, MPs influence their global transport patterns and pose a significant ecological risk. Early understanding of the toxic costs of MPs from the perspective of ecosystem structure and function is essential to maintain the homeostasis and stability of global ecosystems.

Abstract:In recent years, much research has focused on studying the damage caused by microplastics to the ecological environment and human health. Indeed, MPs are often consumed by shellfishes and zooplanktons due to their similarity in size to POM (particular organic matter). Especially in zooplankton, the accumulation of MPs in the body affects the reproductive system and the growth rate of juveniles. Moreover, toxins derived from MPs are continuously accumulated in predators of zooplankton and impact the whole ecosystem across the food chain. In this work, we found that even though MPs were internalized by and adherent around Daphnia magna, there were no significant differences in the survival rate of their adults and offspring. However, the population of ovigerous adults under high MPs exposure for 7 days decreased significantly, suggesting an extension of the days of sexual maturity in D. magna. The removal of MPs after 7 days’ MPs treatment resulted in an increase in D. magna juveniles and neonates which indicated their growth was reduced or inhibited in the MPs environment. Overall, the uptake of MPs led to negative effects on population reproduction and the growth of offspring in D. magna.

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.
Potential toxicity of microplastics on vertebrate liver: A systematic review and meta-analysis

Abstract:Microplastics (MPs) pollution is emerging as a significant environmental concern, threatening human and animal health. Liver, as an important organ, plays an important role in the metabolism and detoxification of pollutants. Many studies have suggested that the liver is a potential target organ for MPs. However, the extent and consequences of the impacts of MPs on the liver reported in studies remain inconsistent. We categorized vertebrates into fish, mammals, and birds and performed a meta-analysis to comprehensively examine the effects of MPs on the liver. Hedges' g values were calculated to evaluate effect sizes. To further explore the sources of heterogeneity among the studies, we conducted subgroup analyses focusing on life stage, MPs type, MPs size, and exposure duration. Additionally, we normalized the MPs concentrations and conducted meta-regression analyses to explore the relationship between MPs concentrations and their hepatotoxic effects. A total of 118 studies were included in this meta-analysis. By quantifying 19 indicators, the results showed that MPs could damage the liver by altering liver morphology, inducing oxidative stress, producing intracellular toxicity, altering biotransformation processes, and disturbing lipid metabolism. Intracellular toxicity, followed by oxidative stress, had the greatest impact. Organisms are more sensitive to MPs under the following conditions: longer exposure duration, smaller MPs sizes, and earlier life stages. As the concentration of MPs increases, the levels of several liver indicators, including catalase, glutathione S-transferase, reactive oxygen species, and alkaline phosphatase, progressively increase. This study provides a comprehensive understanding of the effects of MPs on the liver and suggests the underlying mechanisms of MPs hepatotoxicity.
Metabolic impacts of polystyrene microplastics on the freshwater microalga Microcystis aeruginosa

Abstract:Microplastics (plastic particles < 5 mm; MPs) are ubiquitous in aquatic environments but their potential adverse ecological effects on biota remain poorly understood. This is in part because in typical ecotoxicology tests the toxic effects of MPs were found to be limited. To capture the potential find-scale effects of MPs on freshwater organisms, we employed ultra-performance liquid chromatography-tandem mass spectrometry based untargeted metabolomics to investigate the metabolic impact of polystyrene microbeads microplastics (PS-MPs) of different sizes (0.1, 1, 10, 100 μm) and concentrations (1, 10, 100 mg/L) on a common freshwater microalga, Microcystis aeruginosa , after a 96-h exposure test. The phenotype-based results illustrated that while PS-MPs had no discernible effects on microalgal growth and photosynthesis, both oxidative stress and microcystin production were slightly increased. Metabolomics analysis revealed that the PS-MPs altered the global metabolic profile of the microalga. Specially, PS-MPs of larger size and higher concentration induced a larger number of differentially expressed metabolites. The PS-MPs significantly disturbed metabolisms involved in amino acid synthesis, membrane formation, nitrogen storage, and antioxidant defense of the microalga, consistent with the phenotypic observations. These results suggested several perturbed metabolic pathways, especially arginine-related pathways, as the mechanism. Our study showed that the insights provided by metabolomics-based approaches can enhance assessments of the ecological impacts of MPs on freshwater organisms.

Abstract:Microplastics (MPs) are abundantly present in aquatic environments, where the phytoplankton-microalgae, are now inevitably bound to a long-term coexistence with them. While numerous studies have focused on the toxicological effects of high-concentration MPs exposure, there remains controversy over whether and how MPs affect microalgae at environmentally relevant concentrations. This study aims to draw conclusions that narrow the gap from 52 studies with varying results. Overall, MPs can inhibit growth and photosynthesis, induce oxidative damage, from which microalgae can recover after an appropriate period. Cyanobacteria exhibit greater vulnerability than chlorophyta. The relative size of MPs to algal cells potentially governs their coexistence behavior, thereby altering the mechanisms of impact. Pristine MPs may increase the production of extracellular polymeric substances (EPS) and microcystins (MCs), while aged MPs have the opposite effect. Additionally, relevant factors are systematically discussed, offering insights for future research.
Microplastics: A tissue-specific threat to microbial community and biomarkers of discus fish (Symphysodon aequifasciatus)

Abstract:As detriments in aquatic environments, microplastics (MPs) have been commonly studied on organisms, but tissue-scale effects of MPs were poorly understood. Discus fish (Symphysodon aequifasciatus), herewith, were exposed to polystyrene MPs (0/20/200 μg/L) for 28 d. We found that MPs significantly inhibited growth performance. MPs were observed in skin, gill and intestine after 14/28-d exposure. MPs bioaccumulation was independent of exposure time, but increased with MPs concentrations. Microbial community diversity of fish gill, but not skin and intestine, in MPs treatments was significantly increased. Bacterial community of MP-treated skin and gill were obviously separated from control. Skin dominant phyla changed from Actinobacteriota to Proteobacteria and Firmicutes. Proteobacteria gradually occupied dominance in gill after exposure. Furthermore, MPs-induced skin oxidative stress was demonstrated by the activation of superoxide dismutase and catalase. Skin malondialdehyde also increased and showed significant correlations with four bacterial phyla, e.g., Proteobacteria. Gill Na+/K+-ATPase activity decreased, strongly correlating to microbial community changes caused by MPs. Intestinal digestive enzymes activity (pepsin, lipase and α-amylase) reduced, revealing correlation with bacterial community especially Fibrobacterota. These results suggest a tissue-specific effect of MPs to microbial community and biomarkers in aquatic organism.

Abstract:Microplastics (MPs) can be ingested by a variety of species and mainly accumulate in the gut. However, the consequences of MPs exposure in the gut are largely unknown. Here we evaluated the impacts of MPs exposure in zebrafish gut. Animals were experimentally exposed to polystyrene MPs (5-μm beads; 50 μg/L and 500 μg/L) for 21 days and monitored for alterations in tissue histology, enzymatic biomarkers, gut microbiome and metabolomic responses. Inflammation and oxidative stress were observed in the zebrafish gut after exposed to MPs. Furthermore, significant alterations in the gut microbiome and tissue metabolic profiles were observed, with most of these were associated with oxidative stress, inflammation and lipid metabolism. This study provides evidence that MPs exposure causes gut damage as well as alterations in gut metabolome and microbiome, yielding novel insights into the consequences of MPs exposure.
