Abstract:Enzymes are among the most sophisticated catalysts with unique features of high substrate affinity, unmatched catalytic efficiency and specificity. However, their practical applications are hindered by their high cost, the highly restricted catalytic conditions and their fragile nature. To solve these problems of enzymes for practical applications, biomimicry has therefore emerged as an effective approach to synthesize highly efficient biomimetic catalysts by imitating certain key features of enzymes. To tune the pore structures and modify the functional groups of metal organic frameworks (MOFs), the porosity, functionality and hydrophobic/hydrophilic pore nature of the emerging porous materials are systematically tunable, which endow them to match certain enzymatic features, including structural features, active sites, microenvironments, assistant sites and/or catalytic properties of enzymes. Compared with their enzyme counterparts, biomimetic MOF catalysts present the advantages of broadened applicable conditions and enhanced stability, and sometimes they exhibit catalytic properties that are not possessed by the native enzymes. This review will selectively illustrate biomimetic MOF catalysts, consisting of immobilized biomimetic active sites on the pore surfaces or inside the pore space of porous MOFs. We will discuss the design principles and synthetic strategies for biomimetic MOF catalysts, and the relationships between their catalytic properties and structures. The key roles of active sites, microenvironment, transmission channels and co-catalytic sites, associated with the synergistic catalysis of biomimetic MOFs, will be addressed. (C) 2018 Elsevier B.V. Abstract:Nature has served as a dominant source of inspiration in the area of chemistry, serving as prototypes for the design of materials with proficient performance. In this account, we present our effort to explore porous organic polymers (POPs) as a platform for the construction of biomimetic catalysts to enable new technologies to achieve efficient conversions. For each aspect, we firstly describe the chemical basis of nature, followed by presenting the principles and design strategies involved for functionalizing POPs along with a summary of critical requirements for materials, culminating in a demonstration of unique features of POPs. Our endeavors of using POPs to address the fundamental scientific problems related to biomimetic catalysis are then presented to show their enormous potential and capabilities for a wide range of catalytic transformations. To conclude, a personal perspective on the challenges and opportunities in this emerging field are presented.

Abstract:Utilizing covalent organic frameworks (COFs) as porous supports to encapsulate enzyme represents an advanced strategy for constructing COFs biocatalysts, which has inspired numerous interests across various applications. As the structural advantages including ultrastable covalent‐bonded linkage, tailorable pore structure, and metal‐free biocompatibility, the resultant enzyme‐COFs biocatalysts showcase functional enhancement in catalytic activity, chemical stability, long‐term durability, and recyclability. This Concept describes the recent advances in the methodological strategies for engineering the COFs biocatalysts, with specific emphasis on the pore entrapment and in situ encapsulation strategies. The structural advantages of the COFs hybrid biocatalysts for organic synthesis, environment‐ and energy‐associated applications are also canvassed. Additionally, the remaining challenges and the forward‐looking directions in this field are also discussed. We believe that this Concept can offer useful methodological guidance for developing active and robust COFs biocatalysts.

Abstract:Comprising periodically repeating inorganic nodes and organic linkers, metal-organic frameworks (MOFs) represent a novel class of porous molecular solids with well-defined pores and channels. Over the past two decades, a large array of organic linkers have been combined with many inorganic nodes to afford a vast library of MOFs. The synthetic tunability of MOFs distinguishes them from traditional porous inorganic materials and has allowed the rational design of many interesting properties, such as porosity, chirality, and chemical functionality, for potential applications in diverse areas including gas storage and separation, catalysis, light harvesting, chiral separation, and chemical sensing. In particular, the molecular functionality and intrinsic porosity of MOFs have rendered them attractive candidates as porous single-site solid catalysts for a large number of organic transformations. MOF catalysts offer several advantages over their homogeneous counterparts, including enhanced stability, recyclability and reusability, and facile removal of the toxic catalyst components from the organic products. Additionally, the highly ordered nature of MOFs leads to the generation of single-site solid catalysts, allowing for precise characterization of the catalytic sites through X-ray diffraction, X-ray absorption, and other spectroscopic interrogations and facilitating the elucidation of reaction mechanisms. Thus, MOF catalysis represents a fertile research area that is expected to witness continued growth in the foreseeable future. In this Account, we present our recent research progress in developing ligand-supported single-site MOF catalysts for challenging organic reactions. We present two complementary approaches to the design of ligand-supported MOF catalysts: direct incorporation of prefunctionalized organic linkers into MOFs and postsynthetic functionalization of orthogonal secondary functional groups of the organic linkers in MOFs. Monophosphine-, bipyridine-, β-diketimine-, and salicylaldimine-based ligands have been used to support both precious (Pd, Pt, Ir, Ru) and earth-abundant (Cu, Co, Fe) metals for a number of interesting catalytic reactions. The resulting MOF catalysts feature stable low-coordination species with minimum steric bulk around the active site-a feat that remains a challenge for homogeneous catalysts. For each ligand, we describe types of reactions catalyzed by the MOF in comparison with its homogeneous counterpart. In all cases, MOF catalysts outperformed their homogeneous counterparts in terms of catalyst stability, catalytic activity, and recyclability and reusability. Interestingly, several bipyridine- and salicylaldimine-ligated earth-abundant-metal-based MOF catalysts do not have homogeneous counterparts because the molecular compounds disproportionate or oligomerize to form inactive species in solution. This Account not only presents several interesting designs of ligand-supported single-site MOF catalysts but also provides illustrative examples of how site isolation in MOF catalysts shuts down deactivation pathways experienced by homogeneous systems. With precise knowledge of MOF structures and catalytically active sites, we envision the development of practically useful MOF catalysts comprising tailor-made building blocks that rationally optimize catalytic activities and selectivities.

Abstract:<p>Nanomaterial‐based enzyme‐mimetic catalysts (Enz‐Cats) have received considerable attention because of their optimized and enhanced catalytic performances and selectivities in diverse physiological environments compared with natural enzymes. Recently, owing to their molecular/atomic‐level catalytic centers, high porosity, large surface area, high loading capacity, and homogeneous structure, metal–organic frameworks (MOFs) have emerged as one of the most promising materials in engineering Enz‐Cats. Here, the recent advances in the design of MOF‐engineered Enz‐Cats, including their preparation methods, composite constructions, structural characterizations, and biomedical applications, are highlighted and commented upon. In particular, the performance, selectivities, essential mechanisms, and potential structure–property relations of these MOF‐engineered Enz‐Cats in accelerating catalytic reactions are discussed. Some potential biomedical applications of these MOF‐engineered Enz‐Cats are also breifly proposed. These applications include, for example, tumor therapies, bacterial disinfection, tissue regeneration, and biosensors. Finally, the future opportunities and challenges in emerging research frontiers are thoroughly discussed. Thereby, potential pathways and perspectives for designing future state‐of‐the‐art Enz‐Cats in biomedical sciences are offered.</p>

Abstract:Metal-organic frameworks (MOFs) for enzyme encapsulation-induced biomimetic mineralization under mild reaction conditions are commonly microporous and hydrophobic, which result in a rather high mass transfer resistance of the reactants and restrain the enzyme catalytic activity. Herein, we prepared a type of hierarchical porous and hydrophilic MOF through the biomimetic mineralization of enzymes, zinc ions, 2-methylimidazole, and lithocholic acid. The hierarchical porous structure accelerated the diffusion process of the reactants and the increased hydrophilicity conferred interfacial activity and increased the enzyme catalytic activity. The immobilized enzyme retained higher catalytic activity than the free enzyme and exhibited enhanced resistance to alkaline, organic, and high-temperature conditions. The nanobiocatalyst was reusable and showed long-term storage stability.
Highly efficient synergistic biocatalysis driven by stably loaded enzymes within hierarchically porous iron/cobalt metal-organic framework <i>via</i> biomimetic mineralization

Abstract:The integration of multimodal chemo-/bio-catalysis for efficient cascade reactions has long provided broad prospects in the field of biotechnology for ages. In this work, we describe the synthesis of a biomimetic multienzyme hybrid with hierarchically porous structure and outstanding catalytic activity via in situ encapsulation of natural enzymes in an iron-cobalt bimetallic metal-organic framework (Fe/Co-MOF, FCM). The combination of a single enzyme (glucose oxidase) or dual enzyme (beta-galactosidase and glucose oxidase) with FCM resulted in remarkable synergistic biocatalysis ability; in contrast to simple biocatalyst mixtures in solution, the prepared multienzyme hybrid resulted in 3.2-fold and 2.1-fold improvements in activity for tandem reactions, respectively. The reinforced cascade bioactivity of the multienzyme hybrid benefitted from the synergistic effect between iron/cobalt in the FCM nanozyme, the opened substrate channel between enzymes/nanozymes, and the beneficial effect provided by the hierarchical MOF pores. The enlarged pores not only provided adequate space for immobilized proteins to diffuse and reorientate in FCM with low surface energy, but also reduced the intrinsic mass transfer obstacle to increase the diffusional efficiency of reactants/intermediates. In addition, on account of the shielding effect provided by FCM, the multienzyme hybrid exhibited enhanced tolerance towards severe circumstances and excellent reusability and has been successfully applied in small molecule detection, such as glucose and lactose. The current study highlights the superiority of synergistic bioreactors integrated with the MOF nanozyme and natural enzymes, suggesting great potential for applications in sustainable biomimetic catalysis.
Constructing artificial mimic-enzyme catalysts for carbon dioxide electroreduction

Abstract:Natural bio-enzyme catalysts usually exhibit unexpected performances for many significant reactions, which are worthy of reference. Here we report artificial metal-sulfur-carbon (M-S-C) mimic-enzyme catalysts based on bionic design. The catalysts combine metal centers and functional ligands, which realize the universal fabrication of phase and adjustable dimension. The synthesized catalysts inherit the highly active and selective feature of bio-enzyme catalysts. When directly used for carbon dioxide electroreduction reaction, the Sn-S-C catalyst exhibits high selectivity for formate (Faradaic efficiency >95%), as well as a continuous stability over 120 h at a high current density of 740 mA cm −2 , greatly outperforming the reported catalysts for formate formation. The catalytic sites and pathways are probed with in-situ Fourier transform infrared (FTIR) spectra, in-situ Raman spectra and synchrotron-radiation X-ray photoelectron spectra. These results break the inherent conundrum that it is impossible to simultaneously realize activity and durability under high selectivity. Our findings offer a versatile strategy to inherit from nature and integrate different components, thus designing efficient catalysts for various challenging reactions and energy conversions via a natural sustainable way.

Abstract:Porous metal-organic frameworks (MOFs) are built from periodically alternate organic moieties and metal ions/ clusters. The unique features of the open framework structures, the high surface areas, the permanent porosity, and the appropriate hydrophilic and hydrophobic pore nature mean that MOF materials are a class of ideal host matrices for immobilization of molecular catalysts. The emerging porous materials can not only retain but are also able to enhance the catalytic functions of the single individuals. MOF catalysts have the following super characters: i) uniformly dispersed catalytic sites on the pore surfaces to improve the utility, ii) appropriate hydrophilic and hydrophobic pore nature to facilitate the recognition and transportation of reactant and product molecules, iii) a collaborative microenvironment to realize synergistic catalysis, and iv) simple separation and recovery for long-term usage. Accompanying the development of the synthetic strategies and the technologies for the characterization of MOF materials, MOF catalysis has undergone an upsurge, which has transcended the stage of opportunism. Here, the rational design and synthesis of MOF catalysts are discussed, along with the key factors of active sites, microenvironments, and transmission channels that lead to the distinct catalytic properties of MOF catalysts.

Abstract:In the drive toward green and sustainable methodologies for chemicals manufacturing, biocatalysts are predicted to have much to offer in the years to come. That being said, their practical applications are often hampered by a lack of long-term operational stability, limited operating range, and a low recyclability for the enzymes utilized. Herein, we show how covalent organic frameworks (COFs) possess all the necessary requirements needed to serve as ideal host materials for enzymes. The resultant biocomposites of this study have shown the ability boost the stability and robustness of the enzyme in question, namely lipase PS, while also displaying activities far outperforming the free enzyme and biocomposites made from other types of porous materials, such as mesoporous silica and metal-organic frameworks, exemplified in the kinetic resolution of the alcohol assays performed. The ability to easily tune the pore environment of a COF using monomers bearing specific functional groups can improve its compatibility with a given enzyme. As a result, the orientation of the enzyme active site can be modulated through designed interactions between both components, thus improving the enzymatic activity of the biocomposites. Moreover, in comparison with their amorphous analogues, the well-defined COF pore channels not only make the accommodated enzymes more accessible to the reagents but also serve as stronger shields to safeguard the enzymes from deactivation, as evidenced by superior activities and tolerance to harsh environments. The amenability of COFs, along with our increasing understanding of the design rules for stabilizing enzymes in an accessible fashion, gives great promise for providing "off the shelf" biocatalysts for synthetic transformations.

Abstract:Natural enzymes are crucial catalysts that not only control over the biotransformation in intracellular environments with high efficiency but also are widely harnessed in vitro for diverse applications, among which are used as a core element to engineer biosensor attributing to their high sensitivity and specificity in recognition. However, the vulnerable conformation of a native enzyme arises a series of unavoidable issues associated with lifetime and cost effectiveness, greatly limiting their large-scale applications. The insight into the enzyme mimics using programmable and structurally stable nanomaterial, known as nanozyme, offers an efficient scheme to circumvent the obstacles of natural enzymes, yet, render the biocatalytic functions. In the various categories of nanozymes, porous organic frameworks (POFs) including metal organic frameworks (MOFs), covalent organic frameworks (COFs) and hydrogen-bonded organic frameworks (HOFs) arise enormous attention due to their unique features of ultrahigh surface area, high porosity, and intercommunicating channels. Most importantly, POFs have unambiguous yet predictable topologies, which are structurally favorable for mimicking the active sites as well as microenvironments of natural enzymes. In this review, we overview the advances of POFs nanozymes in which different oxidoreductase- and hydrolase-mimicking activities are involved, with special emphasis on harnessing these diverse activities for conceiving next-generation biosensor targeting micromolecules, metal ions, peptides and proteins, nucleic acids and circulating tumor cells (CTCs), etc. Finally, the current challenges and future perspectives on the design and biosensing applications of POFs nanozymes are discussed. We believe that this review can offer new insight into the access of enzyme mimics with user-defined bioactivity and high stability, and may inspire more interesting biosensing applications.

Ordered Mesoporous CoO/CeO2 Heterostructures with Highly Crystallized Walls and Enhanced Peroxidase-Like Bioactivity

Abstract:With increasing requirements of replacing the high-cost and unstable natural enzymes, the rational design of highly efficient inorganic mimetic enzymes (nanozymes), as one of the most important bioactive materials, has attracted more and more attention in recent years. In our work, using the solvent evaporation induced co-assembly strategy with high-molecular weight poly(ethylene oxide)-block-polystyrene (PEO-b-PS) diblock copolymer as the template and two metal salts as the co-precursors in one-pot system, we synthesized ordered mesoporous heteroatom-doped CeO2 with high surface area, bimodal pore sizes and highly crystallized walls. The composite materials were then constructed as the mimetic enzymes to investigate their peroxidase-like catalytic performance and detect H2O2 and glucose as bioactive materials in diverse systems including complex biological samples. It was demonstrated that mesoporous 5%CoO/CeO2 catalyst was the best peroxidase mimic, following the typical Michaelis-Menten kinetics and exhibiting the excellent peroxidase-like catalytic performance toward 3,3',5,5'-tetramethylbenzidine (TMB) with comparable K-m and V-m values to horseradish peroxidase (HRP) enzyme. A highly sensitive and selective colorimetric assay was developed to detect H2O2 and glucose with detection limits of 0.88 and 19.6 mu M, respectively. The promising catalytic behavior of mesoporous 5%CoO/CeO2 as the peroxidase- like bioactive catalyst could be ascribed to its interconnected large mesopores, rich exposed surface defects and oxygen vacancies introduced by the heterojunction interface of ultrasmall CeO2 and CoO nanocrystals within pore walls. Abstract:Metalloporphyrins are the active sites in monooxygenases that oxidize a variety of substrates efficiently and under mild conditions. Researchers have developed artificial metalloporphyrins, but these structures have had limited catalytic applications. Homogeneous artificial metalloporphyrins can undergo catalytic deactivation via suicidal self-oxidation, which lowers their catalytic activity and sustainability relative to their counterparts in Nature. Heme molecules in protein scaffolds can maintain high efficiency over numerous catalytic cycles. Therefore, we wondered if immobilizing metalloporphyrin moieties within porous metal-organic frameworks (MOFs) could stabilize these structures and facilitate the molecular recognition of substrates and produce highly efficient biomimetic catalysis. In this Account, we describe our research to develop multifunctional porphyrinic frameworks as highly efficient heterogeneous biomimetic catalysts. Our studies indicate that porous porphyrinic frameworks provide an excellent platform for mimicking the activity of biocatalysts and developing new heterogeneous catalysts that effect new chemical transformations under mild conditions. The porous structures and framework topologies of the porphyrinic frameworks depend on the configurations, coordination donors, and porphyrin metal ions of the metalloporphyrin moieties. To improve the activity of porous porphyrinic frameworks, we have developed a two-step synthesis that introduces the functional polyoxometalates (POMs) into POM-porphyrin hybrid materials. To tune the pore structures and the catalytic properties of porphyrinic frameworks, we have designed metalloporphyrin M-H8OCPP ligands with four m-benzenedicarboxylate moieties, and introduced the secondary auxiliary ligands. The porphyrin metal ions and the secondary functional moieties that are incorporated into porous metal-organic frameworks greatly influence the catalytic properties and activities of porphyrinic frameworks in different reactions, such as the oxidation of alkylbenzenes, olefins, and hexane and the photo-oxygenation of 1,5-dihydroxynaphthalene and sulfides. The porphyrin metal ions and the secondary auxiliary sites in the pores can work together synergistically to enhance the catalytic activities of porphyrinic frameworks. Compared with their homogeneous counterparts, the activities and stabilities of the heterogeneous porphyrinic frameworks are remarkable: the immobilization of metalloporphyrins onto the pore surfaces of MOFs not only prevents their suicidal self-oxidation but also allows them to activate inert substrate molecules, such as cyclohexane. Moreover, because the bulky molecules cannot easily access the active sites inside the pores of porphyrinic frameworks, these porous materials demonstrate interesting size-selective catalytic properties toward substrates.

Abstract:A substantial development in nanoscale materials possessing catalytic activities comparable with natural enzymes has been accomplished. Their advantages were owing to the excellent sturdiness in an extreme environment, possibilities of their large-scale production resulting in higher profitability, and easy manipulation for modification. Despite these advantages, the main challenge for artificial enzyme mimetics is the lack of substrate selectivity where natural enzymes flourish. This review addresses this vital problem by introducing substrate selectivity strategies to three classes of artificial enzymes: molecularly imprinted polymers, nanozymes (NZs), and DNAzymes. These rationally designed strategies enhance the substrate selectivity and are discussed and exemplified throughout the review. Various functional mechanisms associated with applying enzyme mimetics in biosensing and bioassays are also given. Eventually, future directives toward enhancing the substrate selectivity of biomimetics and related challenges are discussed and evaluated based on their efficiency and convenience in biosensing and bioassays.

Abstract:Enzymes are natural catalysts with high catalytic activity, substrate specificity and selectivity. Their widespread utilization in industrial applications is limited by their sensitivity to harsh reaction conditions and difficulties relating to their removal and re-use after the reaction is complete. These limitations can be addressed by immobilizing the enzymes in solid porous supports. Covalent organic frameworks (COFs) are ideal candidate carriers because of their good biocompatibility, long-term water stability and large surface area. In post-synthetic immobilization, the enzyme is added to an existing COF; this has had limited success because of enzyme leaching and pore blockage by enzymes that are too large. Direct-immobilization methods—building the COF around the enzyme—allow tailored incorporation of proteins of any size and result in materials with lower levels of leaching and better mass transport of reactants and products. This protocol describes direct-immobilization methods that can be used to fabricate enzyme@COF (@ = engulfing) biocomposites with rationally programmed structures and functions. If COF construction requires harsh reaction conditions, the enzyme can be protected by using a removable metal-organic framework. Alternatively, a direct in situ approach, in which the enzyme and the COF monomers assemble under very mild conditions, can be used. Examples of both approaches are described: enzyme@COF-42-B/43-B capsules (enzymes including catalase, glucose oxidase, etc.) with ZIF-90 or ZPF-2 as protectors, and lipase@NKCOF-98/99 via in situ direct-immobilization methods (synthesis timing: 30–100 min). Example assays for physical and functional characterization of the COF and enzyme@COF materials are also described.

Abstract:Metal-organic frameworks (MOFs), constructed by metal ions/clusters and organic linkers, featuring crystalline porous structures, have been intensively employed as templates/precursors for the synthesis of diverse porous materials including porous carbons, metal-based compounds (such as metal oxides/carbides/phosphides) and their composites. The large surface area, high porosity and excellent tailorability of MOFs can be well inherited to their derivatives, making MOF-derived porous materials very promising for catalytic applications. Herein, we systematically summarize the versatile synthetic strategies to fabricate MOF-derived porous materials and give an overview on their recent progress on organic heterogeneous catalysis, photocatalysis and electrocatalysis. Finally, the challenges and prospects related to MOF-derived porous materials for catalysis are also discussed. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:Metalloporphyrins are the active sites in monooxygenases that oxidize a variety of substrates efficiently and under mild conditions. Researchers have developed artificial metalloporphyrins, but these structures have had limited catalytic applications. Homogeneous artificial metalloporphyrins can undergo catalytic deactivation via suicidal self-oxidation, which lowers their catalytic activity and sustainability relative to their counterparts in Nature. Heme molecules in protein scaffolds can maintain high efficiency over numerous catalytic cycles. Therefore, we wondered if immobilizing metalloporphyrin moieties within porous metal-organic frameworks (MOFs) could stabilize these structures and facilitate the molecular recognition of substrates and produce highly efficient biomimetic catalysis. In this Account, we describe our research to develop multifunctional porphyrinic frameworks as highly efficient heterogeneous biomimetic catalysts. Our studies indicate that porous porphyrinic frameworks provide an excellent platform for mimicking the activity of biocatalysts and developing new heterogeneous catalysts that effect new chemical transformations under mild conditions. The porous structures and framework topologies of the porphyrinic frameworks depend on the configurations, coordination donors, and porphyrin metal ions of the metalloporphyrin moieties. To improve the activity of porous porphyrinic frameworks, we have developed a two-step synthesis that introduces the functional polyoxometalates (POMs) into POM-porphyrin hybrid materials. To tune the pore structures and the catalytic properties of porphyrinic frameworks, we have designed metalloporphyrin M-H8OCPP ligands with four m-benzenedicarboxylate moieties, and introduced the secondary auxiliary ligands. The porphyrin metal ions and the secondary functional moieties that are incorporated into porous metal-organic frameworks greatly influence the catalytic properties and activities of porphyrinic frameworks in different reactions, such as the oxidation of alkylbenzenes, olefins, and hexane and the photo-oxygenation of 1,5-dihydroxynaphthalene and sulfides. The porphyrin metal ions and the secondary auxiliary sites in the pores can work together synergistically to enhance the catalytic activities of porphyrinic frameworks. Compared with their homogeneous counterparts, the activities and stabilities of the heterogeneous porphyrinic frameworks are remarkable: the immobilization of metalloporphyrins onto the pore surfaces of MOFs not only prevents their suicidal self-oxidation but also allows them to activate inert substrate molecules, such as cyclohexane. Moreover, because the bulky molecules cannot easily access the active sites inside the pores of porphyrinic frameworks, these porous materials demonstrate interesting size-selective catalytic properties toward substrates.
