Early Detection and Classification of Tobacco Leaves Inoculated with Tobacco Mosaic Virus Based on Hyperspectral Imaging Technique

Abstract:Tobacco mosaic virus (TMV) is a severe worldwide disease and capable of greatly reducing tobacco quality and yield. It is vital to identify the tobacco disease inoculated with TMV at the early stage of infection for preventing and managing plant diseases. The main contribution of this paper is a procedure for the early detection and classification of tobacco disease with different machine-learning algorithms based on hyperspectral imaging technique. Images from healthy leaves and leaves inoculated with TMV for a period of 7 days after inoculation were acquired per two days by a hyperspectral imaging system with the wavelength region of 380-1,023 nm. Spectral reflectance of pre-defined region of interest (ROI) from hyperspectral images was extracted by ENVI software. Successive projections algorithm (SPA) were used and evaluated for effective wavelengths (EWs) selection. In addition, different machine-learning algorithms i.e. support vector machine (SVM), back propagation neural network (BPNN), extreme learning machine (ELM), least squares support vector machine (LS-SVM), partial least squares-discrimination analysis (PLS-DA), linear discriminant analysis (LDA) and random forest (RF) were developed to quantitatively detect and classify disease stages using EWs. Most classification models showed acceptable results, with the identification rate was over 85%. The discrimination between healthy tobacco leaves and diseased leaves (healthy, 2 days post infection (DPI), 4 DPI, 6 DPI) resulted in classification accuracies up to 95% with BPNN and ELM models. Hence, the overall results clearly demonstrated that hyperspectral imaging has the potential as a fast and non-invasive method with machine-learning algorithms for presymptomatic detection of the plant diseases.
Discriminate Copper Contamination of Fresh Tobacco Leaf Based on Laser Induced Breakdown Spectroscopy

Abstract:Copper is a essential trace element for tobacco. But excessive copper content damages the quality of cigarette and harms the health of smokers. In order to discriminate copper contamination of tobacco plants, a method for identifying fresh tobacco leaf under copper contamination stress was developed by laser-induced breakdown spectroscopy (LIBS) combining with chemometrics methods. 50 tobacco plants were averagely divided into 5 groups. The plants belonging to group 2, 3, 4, 5 were supplied with nutrient solution and copper sulfate solution at different copper ion concentrations of 25 mg L-1, 50 mg L-1, 75 mg L-1, 100 mg L-1, respectively. As reference, the 10 plants of group 1 were only supplied with nutrient solution. After 45 days, 3 leaves from each tabacco plant were selected for LIBS measurements. Principle component analysis (PCA) was used to determine important variables. 18 wavelength variables corresponding to C, N, O, S, Fe, Mg, Ca were seclected. The models for discriminating fresh tobacco leaves under copper contamination stress were built by linear discriminant analysis (LDA) and support vector machine (SVM). The models on the selected wavelength variables performed well. The classification accuracies of LDA model and SVM model were 72% and 82%, respectively. The result indicated the feasibility of copper contamination determination of fresh tobacco leaves using LIBS. This research introduced a new fast heavy metal contamination detection method for fresh plant. It is potential for future portable and online detection instruments or sensors for plant growing monitoring.

Abstract:The quality and flavor of tea leaves are significantly influenced by chemical composition, with the content of free amino acids serving as a key indicator for assessing the quality of Tencha. Accurately and quickly measuring free amino acids during tea processing is crucial for monitoring and optimizing production processes. However, traditional chemical analysis methods are often time-consuming and costly, limiting their application in real-time quality control. Hyperspectral imaging (HSI) has shown significant effectiveness as a component detection tool in various agricultural applications. This study employs VNIR-HSI combined with machine learning algorithms to develop a model for visualizing the total free amino acid content in Tencha samples that have undergone different processing steps on the production line. Four pretreating methods were employed to preprocess the spectra, and partial least squares regression (PLSR) and least squares support vector machine regression (LS–SVR) models were established from the perspectives of individual processes and the entire process. Combining competitive adaptive reweighted sampling (CARS) and variable iterative space shrinkage approach (VISSA) methods for characteristic band selection, specific bands were chosen to predict the amino acid content. By comparing modeling evaluation indicators for each model, the optimal model was identified: the overall model CT+CARS+PLSR, with predictive indicators Rc2 = 0.9885, Rp2 = 0.9566, RMSEC = 0.0956, RMSEP = 0.1749, RPD = 4.8021, enabling the visualization of total free amino acid content in processed Tencha leaves. Here, we establish a benchmark for machine learning-based HSI, integrating this technology into the tea processing workflow to provide a real-time decision support tool for quality control, offering a novel method for the rapid and accurate prediction of free amino acids during tea processing. This achievement not only provides a scientific basis for the tea processing sector but also opens new avenues for the application of hyperspectral imaging technology in food science.

Abstract:Contamination of aflatoxins has important effect on the quality of corn seed. In this study, hyperspectral imaging technology was used to detect aflatoxin B1 (AFB1) with different concentrations (100, 50, 30, 20 and 10 μg/kg) on corn seeds. The near-infrared hyperspectral data, which were collected from the corn seed samples, were processed by image segmentation method. Then, the spectra in the range of 460–929 nm in the region of seed embryo were extracted from the seed-embryo hyperspectral image. Next, the modeling performance of two schemes under four preprocessing methods (none, multiplicative scatter correction (MSC), standard normal variate (SNV), and 5–3 smoothing), four feature wavelength extraction methods (full wavelength, principal component analysis (PCA), X-loading, and successive projection algorithm (SPA)) and three chemometrics methods (k-nearest neighbor (KNN), linear discriminant analysis (LDA), and support vector machine (SVM)) were compared. The optimal result of the testing set is in the case of “5–3 smoothing” pretreatment, SPA method, and SVM model, with 84.1%, 77.8% and 87.3%, 83.0% accuracies of the training set and testing set respectively in scheme 1 and scheme 2. The optimal accuracy of the testing set obtained by “SPA method” is 3.5% higher than in the case of “full wavelength” method, reducing the model calculations and improving the model accuracy to a certain extent. These 10 wavelengths (460.41, 491.50, 594.72, 617.60, 679.77, 726.69, 739.00, 772.43, 926.10, and 929.03 nm) obtained by “SPA method” provide the possibility for real-time online monitoring AFB1 contamination on corn seeds in the future.

Abstract:We investigated the feasibility and potentiality of presymptomatic detection of tobacco disease using hyperspectral imaging, combined with the variable selection method and machine-learning classifiers. Images from healthy and TMV-infected leaves with 2, 4, and 6 days post infection were acquired by a pushbroom hyperspectral reflectance imaging system covering the spectral range of 380–1023 nm. Successive projections algorithm was evaluated for effective wavelengths (EWs) selection. Four texture features, including contrast, correlation, entropy, and homogeneity were extracted according to grey-level co-occurrence matrix (GLCM). Additionally, different machine-learning algorithms were developed and compared to detect and classify disease stages with EWs, texture features and data fusion respectively. The performance of chemometric models with data fusion manifested better results with classification accuracies of calibration and prediction all above 80% than those only using EWs or texture features; the accuracies were up to 95% employing back propagation neural network (BPNN), extreme learning machine (ELM), and least squares support vector machine (LS-SVM) models. Hence, hyperspectral imaging has the potential as a fast and non-invasive method to identify infected leaves in a short period of time (i.e. 48 h) in comparison to the reference images (5 days for visible symptoms of infection, 11 days for typical symptoms).

Abstract:Accurately measuring tobacco harvest maturity is crucial for optimizing quality and crop management. Traditional methods heavily rely on qualitative evaluation and chemical experiments, which introduce subjectivity and inherent limitations. This study proposes a novel method that combines deep learning with proximal hyperspectral imaging (HSI) to achieve precise recognition of tobacco leaf maturity. Unlike traditional techniques, HSI captures rich spectral and spatial data, enabling comprehensive analysis. It adopts a pixel-level annotation strategy to annotate each pixel based on its maturity level, thereby preserving spectral complexity. The dataset comprises 3000 randomly extracted cubes ( 5×5×176 ) from 150 original hyperspectral images, encompassing five maturity levels. Spectral and spatial features are extracted from hyperspectral data using a three-dimensional convolutional neural network (3D-CNN) architecture. This method effectively leverages complex spectral patterns for maturity recognition. During testing, the model demonstrated an impressive average accuracy of 99.93%. Visual predictions vividly illustrate the model's proficiency in maturity recognition, affirming its practical utility. This study pioneers the integration of deep learning and hyperspectral near-end sensing technology in tobacco maturity assessment, mitigating the constraints of traditional methods, and establishing the groundwork for real-time monitoring and quality control of tobacco.

Abstract:The maturity of tobacco leaves directly affects their curing quality. However, no effective method has been developed for determining their maturity during production. Assessment of tobacco maturity for flue curing has long depended on production experience, leading to considerable variation. In this study, hyperspectral imaging combined with a novel algorithm was used to develop a classification model that could accurately determine the maturity of tobacco leaves. First, tobacco leaves of different maturity levels (unripe, under-ripe, ripe, and over-ripe) were collected. ENVI software was used to remove the hyperspectral imaging (HSI) background, and 11 groups of filtered images were obtained using Python 3.7. Finally, a full-band-based partial least-squares discriminant analysis (PLS-DA) classification model was established to identify the maturity of the tobacco leaves. In the calibration set, the model accuracy of the original spectrum was 88.57%, and the accuracy of the de-trending, multiple scattering correction (MSC), and standard normalization variable (SNV) treatments was 91.89%, 95.27%, and 92.57%, respectively. In the prediction set, the model accuracy of the de-trending, MSC, and SNV treatments was 93.85%, 96.92%, and 93.85%, respectively. The experimental results indicate that a higher model accuracy was obtained with the filtered images than with the original spectrum. Because of the higher accuracy, de-trending, MSC, and SNV treatments were selected as the candidate characteristic spectral bands, and a successive projection algorithm (SPA), competitive adaptive reweighted sampling (CASR), and particle swarm optimization (PSO) were used as the screening methods. Finally, a genetic algorithm (GA), PLS-DA, line support vector machine (LSVM), and back-propagation neural network (BPNN) classification and discrimination models were established. The combination SNV-SPA-PLS-DA model provided the best accuracy in the calibration and prediction sets (99.32% and 98.46%, respectively). Our findings highlight the efficacy of using visible/near-infrared (ViS/NIR) hyperspectral imaging for detecting the maturity of tobacco leaves, providing a theoretical basis for improving tobacco production.

Abstract:Black tea has a long history in China, but in export trade, pesticide residues often exceed the standard. To obtain a rapid, accurate, and non-destructive identification method of pesticide residues on black tea, the fluorescence hyperspectral data of dry black tea sprayed with distilled water and six pesticides were collected in this study. The spectra were preprocessed by multiplicative scatter correction (MSC) and standard normal variate (SNV). Then the uninformative variable elimination (UVE), successive projections algorithm (SPA), competitive adaptive re-weighted sampling (CARS), UVE-SPA, and CARS-SPA were used to feature extraction. This study proposes a machine learning model composed of a one-dimensional convolutional neural network backbone (1D CNN backbone) and a random forest classifier (RF classifier) to identify pesticide residues on black tea, and the 1D CNN-RF model was compared with three other machine learning models (support vector machine, RF, and 1D CNN). The results show that MSC-CARS-SPA-1D CNN-RF is the best model for identifying pesticide residues on black tea in which the accuracy of the test set is 99.05%. Combined with fluorescence hyperspectral technology, the proposed 1D CNN-RF model has great potential in the non-destructive identification of pesticide residues on black tea.

Abstract:Tobacco is an important economic crop and the main raw material of cigarette products. Nowadays, with the increasing consumer demand for high-quality cigarettes, the requirements for their main raw materials are also varying. In general, tobacco quality is primarily determined by the exterior quality, inherent quality, chemical compositions, and physical properties. All these aspects are formed during the growing season and are vulnerable to many environmental factors, such as climate, geography, irrigation, fertilization, diseases and pests, etc. Therefore, there is a great demand for tobacco growth monitoring and near real-time quality evaluation. Herein, hyperspectral remote sensing (HRS) is increasingly being considered as a cost-effective alternative to traditional destructive field sampling methods and laboratory trials to determine various agronomic parameters of tobacco with the assistance of diverse hyperspectral vegetation indices and machine learning algorithms. In light of this, we conduct a comprehensive review of the HRS applications in tobacco production management. In this review, we briefly sketch the principles of HRS and commonly used data acquisition system platforms. We detail the specific applications and methodologies for tobacco quality estimation, yield prediction, and stress detection. Finally, we discuss the major challenges and future opportunities for potential application prospects. We hope that this review could provide interested researchers, practitioners, or readers with a basic understanding of current HRS applications in tobacco production management, and give some guidelines for practical works.

Abstract:Rapid and accurate detection of pesticide residue levels can help to prevent the harm of pesticide residue. This study used visible/near-infrared (Vis-NIR) (376–1044 nm) and near-infrared (NIR) (915–1699 nm) hyperspectral imaging systems (HISs) to detect the level of pesticide residues. Three different varieties of grapes were sprayed with four levels of pesticides. Logistic regression (LR), support vector machine (SVM), random forest (RF), convolutional neural network (CNN), and residual neural network (ResNet) models were used to build classification models for pesticide residue levels. The saliency maps of CNN and ResNet were conducted to visualize the contribution of wavelengths. Overall, the results of NIR spectra performed better than those of Vis-NIR spectra. For Vis-NIR spectra, the best model was ResNet, with the accuracy of over 93%. For NIR spectra, LR was the best, with the accuracy of over 97%, but SVM, CNN, and ResNet also showed closed and fine results. The saliency map of CNN and ResNet presented similar and closed ranges of crucial wavelengths. Overall results indicated deep learning performed better than conventional machine learning. The study showed that the use of hyperspectral imaging technology combined with machine learning can effectively detect the level of pesticide residues in grapes.

Abstract:In recent years, the estimation of tobacco field areas has become a critical component of precision tobacco cultivation. However, traditional satellite remote sensing methods face challenges such as high costs, low accuracy, and susceptibility to noise, making it difficult to meet the demand for high precision. Additionally, optical remote sensing methods perform poorly in regions with complex terrain. Therefore, Unmanned Aerial Vehicle multispectral remote sensing technology has emerged as a viable solution due to its high resolution and rich spectral information. This study employed a DJI Mavic 3M equipped with high-resolution RGB and multispectral cameras to collect tobacco field data covering five bands: RGB, RED, RED EDGE, NIR, and GREEN in Agang Town, Luoping County, Yunnan Province, China. To ensure the accuracy of the experiment, we used 337, 242, and 215 segmented tobacco field images for model training, targeting both RGB channels and seven-channel data. We developed a tobacco field semantic segmentation method based on PP-LiteSeg and deeply customized the model to adapt to the characteristics of multispectral images. The input layer’s channel number was adjusted to multiple channels to fully utilize the information from the multispectral images. The model structure included an encoder, decoder, and SPPM module, which used a multi-layer convolution structure to achieve feature extraction and segmentation of multispectral images. The results indicated that compared to traditional RGB images, multispectral images offered significant advantages in handling edges and complex terrain for semantic segmentation. Specifically, the predicted area using the seven-channel data was 11.43 m2 larger than that obtained with RGB channels. Additionally, the seven-channel model achieved a prediction accuracy of 98.84%. This study provides an efficient and feasible solution for estimating tobacco field areas based on multispectral images, offering robust support for modern agricultural management.

Abstract:A screening method using gas chromatography quadrupole time-of-flight mass spectrometry (GC-QTOF/MS) combined with mini solid-phase extraction (mini-SPE) was established for the quantification and validation of multiclass pesticide residues in tobacco. The method was quicker and easier, with sample purity higher than that obtained by traditional SPE and dispersed-SPE. Box-Behnken design, an experimental design for response-surface methodology, was used to optimize the variables affecting the target pesticide recovery. Under the optimized conditions, 92% of the pesticides showed satisfactory recoveries of 70%–120% with precision <20% at spiking levels of 50, 250, and 500 ng/g. The limits of detection and quantification for all the analyses were 0.05–29.9 ng/g and 0.20–98.8 ng/g, respectively. In addition, a screening method based on the retention time and a homebuilt high-resolution mass spectrometry database were established. Under the proposed screening parameters and at spiking levels of 50, 100, and 500 ng/g, 76.6%, 94.7%, and 99.0% multiclass pesticide residues were detected, respectively, using the workflow software. The validated method was successfully applied to the analysis of real tobacco samples. Thus, the combination of mini-SPE and GC-QTOF/MS serves as a suitable method for the quantitative analysis and rapid screening of multiclass pesticide residues in tobacco.

Abstract:Tobacco stems are a critical component in cigarette production in China. However, accurately and promptly identifying impurities in tobacco stems remains a challenge with current deep learning methods, often failing to meet industry standards. This study introduces the Spectral-Spatial Network (SSNet), which is specifically optimized for rapid and precise detection of impurities in tobacco stems, leveraging spatial information in hyperspectral imaging. Designed to align with the high-speed requirements of industrial processing, SSNet effectively overcomes the complexities associated with analyzing hyperspectral data. The integration of spatial information particularly enhances recognition accuracy, especially in areas affected by fringe patterns. Our experiments reveal that SSNet significantly outperforms traditional methods in terms of speed and accuracy, marking a substantial advancement in the shift from manual to automated impurity detection in tobacco processing. Furthermore, SSNet's successful application highlights its potential for broader uses in various industrial image processing tasks.

Abstract:This paper used the newly developed terahertz time-domain spectroscopy (THz-TDS) to detect pesticides, especially imidacloprid, qualitatively and quantitatively in the frequency range of 0.5-1.6 THz. All together, four pesticides and three food powders, as well as polyethylene, were investigated. Different weight ratios of imidacloprid in polyethylene and in sticky rice powder were detected and analyzed with the partial least squares (PLS) regression method. Results show that all four pesticides exhibit unique absorption peaks in the terahertz range, and can thus be easily discriminated between each other and from those food powders that show similar and featureless absorption coefficients. Differences in the refractive indices can also be observed between the pesticides and food powders. Imidacloprid can be identified with its absorption fingerprints in the imidacloprid and polyethylene and imidacloprid and sticky rice powder mixtures. We have achieved the relative error of less than 5% predicting the weight ratio of imidacloprid in these two mixtures with the PLS method, according to the linear relationship found between the absorption coefficient and the weight ratio. In all, the THz-TDS technique is quite promising for the nondestructive pesticide residual detection in food safety control in the near future.

Abstract:Tobacco is a natural plant with complex compositions, the quality of tobacco leaves is directly affected by several external factors such as geographic location and growth conditions. Tobacco leaves are widely planted in China, and they cultivated in different areas, they have different styles. Different blended ratios play a decisive role in the quality of cigarettes. Thus, there is an emerging need for accurate and rapid identification of the origin of tobacco leaves. Near-infrared spectroscopy technology provides a new rapid, and convenient method to automatically evaluate tobacco areas. On this basis, we proposed the grey wolf optimizer (GWO) algorithm to optimize the performance of the support vector machine model (SVM) for the first time to identify and classify tobacco leaves from different origins. This study was conducted with 824 tobacco leaf samples from eight different origins, and 617 training set samples and 207 test set samples were obtained using Set partitioning based on joint x-y distance (SPXY). The wavelength selection methods such as Competitive adaptive reweighted sampling (CARS) and Random frog (RF) algorithms were applied to reduce spectral redundant information and screen the characteristic wavelengths in the -full spectrum of the samples, and 141 and 534 were selected from all 1 609 variables, respectively. Then they were used as the input parameters of the SVM classifier. The optimization effect of GWO on the SVM model was contrasted to the Particle swarm optimization (PSO) and Genetic algorithm (GA) optimization in the same search range. The analysis showed that the spectral variables screened by RF had a better modeling performance than CARS. Among them, the RF-GWO-SVM model achieved the best predictive performance with an accuracy of 96.62% in identifying tobacco leaves from 8 producing areas. More than that, the running time of RF-GWO-SVM was 156 and 131 min shorter than RF-PSO-SVM and RF-GA-SVM, respectively. To sum up, RF-GWO-SVM has the advantages of higher accuracy and faster convergence speed. It can be seen that GWO has a more efficient optimization capability for model parameters, and the support vector machine model optimized by GWO can be used for rapid identification of tobacco origin.
Classification of Pesticide Residues in Sorghum Based on Hyperspectral and Gradient Boosting Decision Trees

Abstract:To address the challenges posed by chemical methods for detecting pesticide residues in sorghum, such as complicated sample preparation and prolonged detection periods, this study presents a rapid and nondestructive detection approach based on hyperspectral imaging (HSI) technology. A group of sorghum without pesticide residues and three groups uniformly sprayed with pesticides were used in this study. Firstly, support vector machine (SVM) classification models were built using spectral data preprocessed with Savitzky–Golay (SG), discrete wavelet transform (DWT), and standard normal variate (SNV) methods, respectively, and SNV was determined to be the best preprocessing method. Secondly, the gradient boosting decision tree (GBDT) algorithm, principal component analysis (PCA), and the successive projections algorithm (SPA) were respectively used to extract feature wavelengths. Pesticide residue identification models based on full and feature wavelengths were then respectively established using backpropagation neural network (BPNN), SVM, and partial least squares discriminant analysis (PLS‐DA). The results show that the BPNN model developed using the feature wavelengths obtained from GBDT was the best for identification of pesticide residues, with an accuracy of 97.8% for both the training and testing sets. Finally, visualization of pesticide residue species in sorghum was achieved using the optimal model. This study demonstrates that utilizing HSI in conjunction with the GBDT‐BPNN model is an effective, rapid, and nondestructive method for identifying pesticide residues in sorghum.

Trace Identification and Visualization of Multiple Benzimidazole Pesticide Residues on Toona sinensis Leaves Using Terahertz Imaging Combined with Deep Learning

Abstract:Molecular spectroscopy has been widely used to identify pesticides. The main limitation of this approach is the difficulty of identifying pesticides with similar molecular structures. When these pesticide residues are in trace and mixed states in plants, it poses great challenges for practical identification. This study proposed a state-of-the-art method for the rapid identification of trace (10 mg·L-1) and multiple similar benzimidazole pesticide residues on the surface of Toona sinensis leaves, mainly including benzoyl (BNL), carbendazim (BCM), thiabendazole (TBZ), and their mixtures. The new method combines high-throughput terahertz (THz) imaging technology with a deep learning framework. To further improve the model reliability beyond the THz fingerprint peaks (BNL: 0.70, 1.07, 2.20 THz; BCM: 1.16, 1.35, 2.32 THz; TBZ: 0.92, 1.24, 1.66, 1.95, 2.58 THz), we extracted the absorption spectra in frequencies of 0.2-2.2 THz from images as the input to the deep convolution neural network (DCNN). Compared with fuzzy Sammon clustering and four back-propagation neural network (BPNN) models (TrainCGB, TrainCGF, TrainCGP, and TrainRP), DCNN achieved the highest prediction accuracies of 100%, 94.51%, 96.26%, 94.64%, 98.81%, 94.90%, 96.17%, and 96.99% for the control check group, BNL, BCM, TBZ, BNL + BCM, BNL + TBZ, BCM + TBZ, and BNL + BCM + TBZ, respectively. Taking advantage of THz imaging and DCNN, the image visualization of pesticide distribution and residue types on leaves was realized simultaneously. The results demonstrated that THz imaging and deep learning can be potentially adopted for rapid-sensing detection of trace multi-residues on leaf surfaces, which is of great significance for agriculture and food safety.

Abstract:Efficient, rapid, and non-destructive detection of pesticide residues in fruits and vegetables is essential for food safety. The visible/near infrared (VNIR) and short-wave infrared (SWIR) hyperspectral imaging (HSI) systems were used to detect different types of pesticide residues on the surface of Hami melon. Taking four pesticides commonly used in Hami melon as the object, the effectiveness of single-band spectral range and information fusion in the classification of different pesticides was compared. The results showed that the classification effect of pesticide residues was better by using the spectral range after information fusion. Then, a custom multi-branch one-dimensional convolutional neural network (1D-CNN) model with the attention mechanism was proposed and compared with the traditional machine learning classification model K-nearest neighbor (KNN) algorithm and random forest (RF). The traditional machine learning classification model accuracy of both models was over 80.00%. However, the classification results using the proposed 1D-CNN were more satisfactory. After the full spectrum data was fused, it was input into the 1D-CNN model, and its accuracy, precision, recall, and F1-score value were 94.00%, 94.06%, 94.00%, and 0.9396, respectively. This study showed that both VNIR and SWIR hyperspectral imaging combined with a classification model could non-destructively detect different pesticide residues on the surface of Hami melon. The classification result using the SWIR spectrum was better than that using the VNIR spectrum, and the classification result using the information fusion spectrum was better than that using SWIR. This study can provide a valuable reference for the non-destructive detection of pesticide residues on the surface of other large, thick-skinned fruits.
