Abstract:Polyglycolic acid (PGA) is an emerging biodegradable plastic material. Together with polylactic acid (PLA), PGA is considered a suitable alternative to conventional plastics and has been widely used in biomedical and food packaging industries. However, degradable plastics continue to face the drawbacks of harsh degradation environment and long degradation time, and may harm the environment and the human body. Therefore, our study focused on assessing the effects of degradable microplastics PGA and PLA on the development and neurobehavior of zebrafish. The results showed that PGA and PLA had little effect on 3-10 hpf embryos. However, developmental stunting was observed in a100 mg/L PGA and PLA-exposed group at 24 hpf. In addition, PGA and PLA exposure decreased the survival and hatching rates, increased wakefulness, and reduced sleep in zebrafish. This indicates that PGA and PLA may affect the circadian behavior of zebrafish by affecting the brain-derived neurotrophic factor (BDNF). Therefore, our results suggest that PGA and PLA exposure induces developmental toxicity, reduces voluntary locomotion, induces of anxiety-like behaviors, and impairs sleep/wake behaviors in zebrafish larvae. This also suggests that the potentially toxic effects of degradable plastics cannot be ignored and that the biological effects of PGA require further research.

Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.

Abstract:Plastic nanoparticles formed from both daily use of plastics and their wastes have emerged as a potential health and environmental hazard. It is necessary to study the biological process of nanoplastics in ecological risk assessment. To address this concern, we quantitatively investigated the accumulation and depuration of polystyrene nanoplastics (PSNs) in the tissues of zebrafish after the aquatic exposure using a quantitative method based on matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Via the PSNs-spiked freshwater, zebrafish were exposed to three different concentrations of PSNs for 30 days, followed by 16 days of depuration. The results showed that the amounts of PSNs accumulated in zebrafish tissues were in the following order: intestine > liver > gill > muscle > brain. The uptake and depuration of PSNs in zebrafish both followed pseudo-firstorder kinetics. It was revealed that the bioaccumulation was concentration, tissue and time dependent. When the PSNs concentration is low, the steady state might take longer time (or not occur) than that of a high concentration. After 16 days of depuration, there were still some PSNs present in the tissues particularly in the brain, where it might take 70 days or more to remove 75 % of PSNs. Overall, this work offers important knowledge on the bioaccumulation of PSNs, which may be useful for future studies into the health hazards of PSNs in aquatic environments.
Early-life exposure to five biodegradable plastics impairs eye development and visually-mediated behavior through disturbing hypothalamus-pituitary-thyroid (HPT) axis in zebrafish larvae

Abstract:Biodegradable plastics have been commonly developed and applied as an alternative to traditional plastics, which cause environmental plastic pollution. However, biodegradable plastics still present limitations such as stringent degradation conditions and slow degradation rate, and may cause harm to the environment and organisms. Consequently, in this study, zebrafish was used to evaluate the effects of five biodegradable microplastics (MPs), polyglycolic acid (PGA), polylactic acid (PLA), polybutylene succinate (PBS), polyhydroxyalkanoate (PHA) and polybutylene adipate terephthalate (PBAT) exposure on the early development, retina morphology, visually-mediated behavior, and thyroid signaling at concentrations of 1 mg/L and 100 mg/L. The results indicated that all MPs induced decreased survival rate, reduced body length, smaller eyes, and smaller heads, affecting the early development of zebrafish larvae. Moreover, the thickness of retinal layers, including inner plexiform layer (IPL), outer nuclear layer (ONL), and retinal ganglion layer (RGL) was decreased, and the expression of key genes related to eye and retinal development was abnormally altered after all MPs exposure. Exposure to PBS and PBAT led to abnormal visually-mediated behavior, indicating likely affected the visual function. All MPs could also cause thyroid system disorders, among which alterations in the thyroid hormone receptors (TRs) genes could affect the retinal development of zebrafish larvae. In summary, biodegradable MPs exhibited eye developmental toxicity and likely impaired the visual function in zebrafish larvae. This provided new evidence for revealing the effects of biodegradable plastics on aquatic organism development and environmental risks to aquatic ecosystems.

Abstract:A plethora of research has focused on the biosafety of biodegradable plastics (BPs), including their microplastic formation and additives leaching; however, relatively fewer studies have explored biodegradation products. This study aims to investigate the biological effects and chemical features of degradation products from three kinds of BPs, namely polyglycolic acid (PGA), poly (butylene adipate- co -terephthalate) (PBAT), and the blends of PGA/PBAT without the addition of additives, in a simulated small waterbody environment with extracted soil solution for three months. Results showed that exposure to the whole degradation remnants of three BPs had no lethal effects on zebrafish at the current BP environmental concentrations (from 0.24 to 12.72 mg plastic/L) in small waterbodies. However, from the calculated BPs environmental concentrations (from 0.57 to 43.82 mg plastic/L) in 2026, PGA and PGA/PBAT blends may cause adverse effects on the cardiovascular system such as heartbeat rate suppression in zebrafish embryos, and also lead to reduced body length and pericardial edema and spinal curvature in fish larvae. We further qualitatively analyzed the composition of degradation products, and quantitatively measured four dominant degradation monomers (glycolic acid (GA), adipic acid (A), 1,4-butanediol (B), and terephthalic acid (T)) in the degradation remnants. It was found that the observed toxicities were probably due to the presence of GA, A, and T monomers, and their concentrations can reach 0.776, 0.034, and 0.6 mg/L under the calculated future scenario, respectively. It is worth mentioning that either GA or T monomers at the above concentrations were found to cause suppressed heartbeat rate in zebrafish embryos. Collectively, though the degradation products of BPs are temporarily safe at current environmental concentrations, they may lead to non-negligible toxicity with increasing production and continual improper recycling and/or BP waste management.

Abstract:Pollution of microplastics (MPs) has become a global environmental issue due to the difficulty in its degradation and may cause unexpected ecological effects. Nevertheless, little is known about the potential effects of MPs on reproduction toxicity in aquatic species. In this study, adult loach (Paramisgurnus dabryanus, F0 generation) were exposed to two concentrations (1 and 10 mg/L) of polyethylene MPs (PE-MPs) for 15 or 30 days, and the toxic effects in parental loach and the offspring (F1 generation) were examined. Our results showed that PE-MPs exposure could change the indicators content of antioxidant system in the brain, liver, and gonad. PE-MPs can accumulate in the gonads, disrupt the transcription of HPG-axis related genes, alter sex hormone levels, increase cell apoptosis and gonadal pathological lesions, lead to the damage of biological characteristics of semen, and affect the reproduction in F0 generation. PE-MPs remaining in the parental gonads can be transferred to the F1 generation embryos and accumulated on the embryonic chorionic membrane, increasing mortality and malformation rates, accelerating hatching time, and decreasing hatching rate and body length. These results suggest that PE-MPs leads to a potential adverse influence on reproduction and serious impacts on population sustainability. This work provides a new perspective into the effects of MPs on reproductive damage and cross-generational effects in teleost fish, which have implications in fields of freshwater ecology and environmental toxicology.
Effects of biodegradable and conventional microplastics on the intestine, intestinal community composition, and metabolic levels in tilapia (Oreochromis mossambicus)

Abstract:Despite growing interest in conventional microplastics (CMPs) and their toxicological effects on aquatic species, little is known about biodegradable microplastics (BMPs) and their corresponding implications for aquatic life. Here, tilapia (Oreochromis mossambicus) were semi-statically exposed for 14 days to the bio-based plastic polylactic acid (PLA, 100 μg/L, 2.52 ± 0.46 μm) and the petroleum-based plastic polyvinyl chloride (PVC, 100 μg/L, 1.58 ± 0.36 μm). The results showed that ingesting the above two types of microplastics (MPs) led to oxidative stress in the fish gut, and damage to gut tissues and organelles, and PLA resulted in more obvious gut tissue edema than PVC. Furthermore, PLA caused increased levels of gut microbiota dysbiosis and a decrease in the abundance of the genus Cetobacterium, which is linked to vitamin B-12 synthesis, whereas an opposite relationship was observed on PVC. Metabolomic analysis indicated that PVC caused a significant down-regulation of orotic acid, co-metabolite of folic acid with vitamin B-12, while PLA did not affect orotic acid, which may lead to the accumulation of folic acid in fish. The joint analysis found that MPs disturbed gut metabolism homeostasis, implying that abnormal gut microbiota metabolites may be a key mechanism for MPs to induce tissue damage and oxidative stress in the gut. Overall, this study systematically illustrates the differential toxic effects of BMPs and CMPs on tilapia through gut microbiota and metabolite interactions, which will contribute to assessing the risks of BMPs to organismal health.

Abstract:The important place of anticoagulants in medicine today makes new observations or little-known reactions to them worthy of notice. While attention of the physician is focused on the main arena of action of this group of drugs, often with anxiety, side-reactions of smaller importance may escape his notice. For this reason we are reporting the case of a child who developed a diffuse alopecia after the ingestion of a coumarin product. The experience of others shows, however, that loss of hair accompanying use of anticoagulants is common. Report of a Case A 2-year-old Negro girl was seen in our clinic for loss of hair. She had ingested a proprietary pesticide called "d-Con," whose active ingredient is a coumarin. Soon after this the child fell asleep. Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.

Abstract:Microplastics (MPs) can adsorb heavy metals and induce combined toxicity to aquatic organisms. However, the combined effects on the gut-liver and gut-brain axes are yet to be fully comprehended. This study investigated the combined effects of polystyrene microplastics (PS-MPs) with two concentrations (20 and 200 μg/L) and three sizes (0.1, 10, and 250 μm) and Pb (50 μg/L) on zebrafish through gut-liver and gut-brain axes. The results showed that the combined exposure of 0.1 μm PS-MPs and Pb resulted in the most significant changes to the community diversity of gut microbiota. The combined exposures of PS-MPs (0.1 μm and 250 μm PS-MPs) and Pb significantly down-regulated expression of zo-1 and occludin but increased the lipopolysaccharide content in zebrafish liver compared to the PS-MPs or Pb alone exposure groups, indicating impaired gut barrier function. Subsequent studies showed that combined exposure of PS-MPs (0.1 μm and 250 μm) and Pb induced liver inflammation through the TLR4/NF-κB pathway. Moreover, all exposure groups had an impact on the expression of genes related to bile acid metabolism (cyp7a1, fgf19, abcb11b, and slc10a2) and neurotransmitters (tph1a, tph2, pink, and trh). The findings of this study provide new evidence on the combined effects of MPs and heavy metals, which are significant for their hazard identification and risk assessment.
Plastic food? Energy compensation of zebrafish ( Danio rerio ) after long-term exposure to polylactic acid biomicroplastics

Abstract:The extensive use of bio-based plastics has led to their widespread distribution in the environment. However, their long-term ecological impact on aquatic animals is not well understood. In this study, adult zebrafish ( Danio rerio ) were exposed to 1000 items·L −1 of either polylactic acid (PLA) or polyethylene terephthalate (PET) microplastics (MPs), for 90 days. PLA is a typical bio-based plastic, while PET is a typical petroleum-derived plastic. The abundances of PLA and PET MPs in fish intestines were 981 ± 66 and 671 ± 151 items per fish, respectively, indicating a greater amount of PLA MP residues than PET MPs. However, the inhibitory effect of PET on fish weight was 1.8 times higher than that of PLA, suggesting energy compensation in PLA-treated zebrafish. Proliferation of Lactobacillus was observed in the fish intestines of the PLA group , indicating increased utilization capacity of intestinal flora for lactic acid production during PLA degradation. Metabolomics showed that the tricarboxylic acid pathway was up-regulated in the PLA group compared with that in the PET group, providing evidence of energy compensation. However, more ingested PLA MPs caused more significant histological damage to fish intestines than PET MPs. Therefore, the ecological risks of bio-based plastics still require attention.

[Long-term development of carotid stenosis treated by endovascular means: a prospective study in a neurovascular unit].

Abstract:AIMS Even today, unlike endarterectomy, long-term development in carotid angioplasty has still not been well defined. The aim of this study is to describe both the short- and long-term development and the rate of restenosis in a consecutive series of patients with carotid stenosis who underwent endovascular treatment at the Hospital Universitario La Fe; a distinction is made between patients with and without symptoms. PATIENTS AND METHODS From 1999 to 2010, 426 patients were submitted to a carotid angioplasty (25.5% were asymptomatic patients). Clinical follow-ups were conducted in the case of 374 patients. They had annual check-ups and every year a neurosonological study was performed. The aim was to determine what complications occurred during the first 30 days, as well as the long-term complications, to establish the rate of restenosis and to evaluate possible differences between patients with and without symptoms. RESULTS The morbidity and mortality rate in the first month was 4.2% (4.4% in symptomatic patients and 3.8% in asymptomatic patients). The mean follow-up time was 55 months: 8% of patients presented strokes, 11% suffered from myocardial infarct and 24.3% died, with no significant differences between patients with or without symptoms. The rate of restenosis was 17.3% at five years, 7.5% equal to or above 50% and, of those, 1.1% were symptomatic. Five patients with significant restenosis were successfully treated with endovascular techniques. CONCLUSIONS Carotid angioplasty is an effective, safe technique in the hands of an experienced professional; our findings are good, both in the short and the long term, with a low rate of restenosis. If this latter condition occurs, vascular intervention techniques are safe and efficient.
Gastrointestinal Incomplete Degradation Exacerbates Neurotoxic Effects of PLA Microplastics via Oligomer Nanoplastics Formation

Abstract:Polylactic acid (PLA) polymer and oligomer microplastics (MPs) degrade incompletely and completely in mice's gastrointestinal tract, respectively. Incomplete degradation of PLA polymer MPs increases their bioavailability, transforming them into oligomer nanoplastics (NPs). Both types of MPs induce PD‐like neurotoxicity by causing mitochondrial calcium overload in midbrain neurons. Notably, PLA oligomer MPs provoke a more significant neurotoxic response. Biodegradable plastics, hailed for their environmental friendliness, may pose unforeseen risks as they undergo gastrointestinal degradation, forming oligomer nanoplastics. Despite this, the influence of gastrointestinal degradation on the potential human toxicity of biodegradable plastics remains poorly understood. To this end, the impact of the murine in vivo digestive system is investigated on the biotransformation, biodistribution, and toxicity of PLA polymer and PLA oligomer MPs. Through a 28‐day repeated oral gavage study in mice, it is revealed that PLA polymer and oligomer microplastics undergo incomplete and complete degradation, respectively, in the gastrointestinal tract. Incompletely degraded PLA polymer microplastics transform into oligomer nanoplastics, heightening bioavailability and toxicity, thereby exacerbating overall toxic effects. Conversely, complete degradation of PLA oligomer microplastics reduces bioavailability and mitigates toxicity, offering a potential avenue for toxicity reduction. Additionally, the study illuminates shared targets and toxicity mechanisms in Parkinson's disease‐like neurotoxicity induced by both PLA polymer and PLA oligomer microplastics. This involves the upregulation of MICU3 in midbrains, leading to neuronal mitochondrial calcium overload. Notably, neurotoxicity is mitigated by inhibiting mitochondrial calcium influx with MCU‐i4 or facilitating mitochondrial calcium efflux with DBcAMP in mice. These findings enhance the understanding of the toxicological implications of biodegradable microplastics on human health.

Abstract:The influence of petroleum-based microplastics (MPs) on phytoplankton has been extensively studied, while research on the impact of biodegradable MPs, derived from alternative plastics to contest the environmental crisis, remains limited. This study performed a 63 days co-incubation experiment to assess the effect of polylactic acid MPs (PLA-MPs) on the growth, physiology, and carbon utilization of M. aeruginosa and the change in PLA-MPs surface properties. The results showed that despite PLA-MPs induced oxidative stress and caused membrane damage in M. aeruginosa , the presence of PLA-MPs (10, 50, and 200 mg/L) triggered significant increases (p < 0.05) in the density of M. aeruginosa after 63 days. Specifically, the algal densities upon 50 and 200 mg/L PLA-MPs exposure were increased by 20.91% and 36.31% relative to the control, respectively. Meanhwhile, the reduced C/O ratio on PLA-MPs surface and change in PLA-MPs morphological characterization, which is responsible for substantially increase in the aquatic dissolved inorganic carbon concentration during the co-incubation, implying the degradation of PLA-MPs; thus, provided sufficient carbon resources that M. aeruginosa could assimilate. This was in line with the declined intracellular carbonic anhydrase content in M. aeruginosa . This study is the first attempt to uncover the interaction between PLA-MPs and M. aeruginosa , and the finding that their interaction promotes the degrading of PLA-MPs meanwhile favoring M. aeruginosa growth will help elucidate the potential risk of biodegradable MPs in aquatic environment.

Polystyrene Nanoplastics Toxicity to Zebrafish: Dysregulation of the Brain–Intestine–Microbiota Axis

Abstract:In animal species, the brain-gut axis is a complex bidirectional network between the gastrointestinal (GI) tract and the central nervous system (CNS) consisting of numerous microbial, immune, neuronal, and hormonal pathways that profoundly impact organism development and health. Although nanoplastics (NPs) have been shown to cause intestinal and neural toxicity in fish, the role of the neurotransmitter and intestinal microbiota interactions in the underlying mechanism of toxicity, particularly at environmentally relevant contaminant concentrations, remains unknown. Here, the effect of 44 nm polystyrene nanoplastics (PS-NPs) on the brain-intestine-microbe axis and embryo-larval development in zebrafish (Danio rerio) was investigated. Exposure to 1, 10, and 100 μg/L PS-NPs for 30 days inhibited growth and adversely affected inflammatory responses and intestinal permeability. Targeted metabolomics analysis revealed an alteration of 42 metabolites involved in neurotransmission. The content of 3,4-dihydroxyphenylacetic acid (DOPAC; dopamine metabolite formed by monoamine oxidase activity) was significantly decreased in a dose-dependent manner after PS-NP exposure. Changes in the 14 metabolites correlated with changes to 3 microbial groups, including Proteobacteria, Firmicutes, and Bacteroidetes, as compared to the control group. A significant relationship between Firmicutes and homovanillic acid (0.466, Pearson correlation coefficient) was evident. Eight altered metabolites (l-glutamine (Gln), 5-hydroxyindoleacetic acid (5-HIAA), serotonin, 5-hydroxytryptophan (5-HTP), l-cysteine (Cys), l-glutamic acid (Glu), norepinephrine (NE), and l-tryptophan (l-Trp)) had a negative relationship with Proteobacteria although histamine (His) and acetylcholine chloride (ACh chloride) levels were positively correlated with Proteobacteria. An Associated Network analysis showed that Firmicutes and Bacteroidetes were highly correlated (0.969). Furthermore, PS-NPs accumulated in the gastrointestinal tract of offspring and impaired development of F1 (2 h post-fertilization) embryos, including reduced spontaneous movements, hatching rate, and length. This demonstration of transgenerational deficits is of particular concern. These findings suggest that PS-NPs cause intestinal inflammation, growth inhibition, and restricted development of zebrafish, which are strongly linked to the disrupted regulation within the brain-intestine-microbiota axis. Our study provides insights into how xenobiotics can disrupt the regulation of brain-intestine-microbiota and suggests that these end points should be taken into account when assessing environmental health risks of PS-NPs to aquatic organisms.

Oligomer-Nanoparticle Release from PLA Bioplastics Catalyzed by Gut Enzymes and Its Acute Inflammatory Effect

Abstract:Abstract Although the risks of microplastics in environmental exposure and human health are being increasingly studied, little is known about the behavior of “eco-friendly” bioplastics in humans, especially their effects on our gastrointestinal tract. Here we demonstrate that enzymatic hydrolysis of bio-based polylactic acid (PLA) microplastics rapidly generates an excess of nanoplastic particles by competing for triglyceride-degrading lipase during gastrointestinal processes. These tiny nanoparticles are oligomers formed by hydrophobic-driven self-aggregation, and upon exposure the oligomers and their associated nanoparticles can bioaccumulate in in vitro and several in vivo organs, including the liver, intestine, and even in the brain. Severe intestinal damage and inflammation are also observed, the toxic effect of which is mostly pronounced from hydrolyzed oligomer products. Furthermore, the oligomers’ potential protein target screening using large scale pharmacophore model reveals that oligomers can interact with matrix metallopeptidase 12 protein (MMP12), which is further validated using protein binding assay. A close mechanistic study reveals high binding affinity of oligomers to the catalytic zinc ion finger domain, leading to MMP12 inactivation and mediating the adverse bowel inflammatory effect following PLA oligomer exposure. Since biodegradable plastics are highly proposed as one solution for the global plastic problem, understanding the gastrointestinal fate and toxicity of bioplastics, will provide ground-breaking data on bioplastics as a substantial risk to human health.

Abstract:Weathered microplastics (MPs) exhibit different physicochemical properties compared to pristine MPs, thus, their effects on the environment and living organisms may also differ. In the present study, we investigated the gut-toxic effects of virgin polypropylene MPs (PP) and UV-weathered PP MPs (UV-PP) on zebrafish. The zebrafish were exposed to the two types of PP MPs at a concentration of 50 mg/L each for 14 days. After exposure, MPs accumulated primarily within the gastrointestinal tract, with UV-PP exhibiting a higher accumulation than PP. The ingestion of PP and UV-PP induced gut damage in zebrafish and increased the gene expression and levels of enzymes related to oxidative stress and inflammation, with no significant differences between the two MPs. Analysis of the microbial community confirmed alterations in the abundance and diversity of zebrafish gut microorganisms in the PP and UV-PP groups, with more pronounced changes in the PP-exposed group. Moreover, the Kyoto Encyclopedia of Genes and Genomes pathway analysis confirmed the association between changes in the gut microorganisms at the phylum and genus levels with cellular responses, such as oxidative stress, inflammation, and tissue damage. This study provides valuable insights regarding the environmental impact of MPs on organisms.

Abstract:In recent years, everyone has recognized microplastics as an emerging contaminant in aquatic ecosystems. Polypropylene is one of the dominant pollutants. The purpose of this study was to examine the effects of exposing zebrafish (Danio rerio) to water with various concentrations of polypropylene microplastics (11.86 ± 44.62 μm), including control (0 mg/L), group 1 (1 mg/L), group 2 (10 mg/L), and group 3 (100 mg/L) for up to 28 days (chronic exposure). The bioaccumulation of microplastics in the tract was noted after 28 days. From the experimental groups, blood and detoxifying organs of the liver and brain were collected. Using liver tissues evaluated the toxic effects by crucial biomarkers such as reactive oxygen species, anti-oxidant parameters, oxidative effects in protein & lipids, total protein content and free amino acid level. The study revealed that the bioaccumulation of microplastics in the organisms is a reflection of the oxidative stress and liver tissue damage experienced by the group exposed to microplastics. Also, apoptosis of blood cells was observed in the treated group as well as increased the neurotransmitter enzyme acetylcholine esterase activity based on exposure concentration-dependent manner. The overall results indicated bioaccumulation of microplastics in the gut, which led to increased ROS levels. This consequently affected antioxidant biomarkers, ultimately causing oxidation of biomolecules and liver tissue injury, as evidenced by histological analysis. This study concludes that chronic ingestion of microplastics causes considerable effects on population fitness in the aquatic environment, as well as other ecological complications, and is also critical to understand the magnitude of these contaminants' influence on ichthyofauna.
