Abstract:The co-exposure of microplastics (MPs) and other contaminants has aroused extensive attention, but the combined impacts of MPs and pesticides remain poorly understood. Acetochlor (ACT), a widely used chloroacetamide herbicide, has raised concerns for its potential bio-adverse effects. This study evaluated the influences of polyethylene microplastics (PE-MPs) for acute toxicity, bioaccumulation, and intestinal toxicity in zebrafish to ACT. We found that PE-MPs significantly enhanced ACT acute toxicity. Also, PE-MPs increased the accumulation of ACT in zebrafish and aggravate the oxidative stress damage of ACT in intestines. Exposure to PE-MPs or/and ACT causes mild damage to the gut tissue of zebrafish and altered gut microbial composition. In terms of gene transcription, ACT exposure triggered a significant increase in inflammatory response-related gene expressions in the intestines, while some pro-inflammatory factors were found to be inhibited by PE-MPs. This study provides a new perspective on the fate of MPs in the environment and on the assessment of the combined effects of MPs and pesticides on organisms.

Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.
Identification of the Zeo1 Protein as a Candidate Structural Homolog of {\alpha}-Synuclein in Budding Yeast

Abstract:Human {\alpha}-synuclein (SNCA) is a 140-amino-acid protein belonging to the three-member synuclein family. It has been extensively studied due to its misfolding/aggregation in and genetic linkage to neurodegenerative diseases, especially Parkinson's disease (PD). To better understand its biology, models of SNCA toxicity have been developed in budding yeast over the past decade, which have yielded insights into the protein's modes of action in specific pathways and potential therapeutic targets. Given that the synuclein gene family is not present in yeast, an extensive homology search was undertaken to determine if any yeast protein may possess structural homology to SNCA and whose native biology may shed more light on SNCA's pathomechanism in eukaryotes. We identified Zeo1, a membrane-associated protein involved in the cell wall integrity (CWI) pathway, as a candidate structural homolog. We show that Zeo1 overexpression is toxic in yeast and, similar to SNCA, localizes to lipid membranes. A number of biochemical similarities between Zeo1 and SNCA also become apparent in light of this potential structural connection. Moreover, the yeast PKC1 gene, a kinase acting as a downstream signaling hub in the CWI pathway, rescues both SNCA- and Zeo1-induced toxicities. Using the same homology search methods that identified Zeo1, we show that Pkc1 has a hybrid structural similarity to PINK1 and PARIS, two mitochondrial PD-implicated proteins not generally linked directly to synuclein-specific pathobiology. Overall, this proof-of-concept study shows the potential utility of hitherto uncharacterized cross-species structural homologs, identified using comparative proteome-wide structure prediction algorithms, in shedding light on abstruse connections among disease-relevant proteins.
Effects of Microplastics on the Accumulation and Neurotoxicity of Methylmercury in Zebrafish Larvae.

Abstract:Microplastics (MPs) and methylmercury (MeHg) have attracted increasing attention due to ubiquitous occurrence and toxicity. This study aimed to investigate whether MPs could absorb MeHg and thus modify its bio-concentration and neurotoxicity in the zebrafish larvae (Danio rerio). The pseudo-second-order model (R-2 = 0.989) was found to be suitable for describing the adsorption kinetics of MeHg onto MPs. Compared with Freundlich and Temkin models, the Langmuir isotherm model provided a better fit with the experimental data exhibiting a maximum monolayer adsorption capacity of 54.945 mg/g. These results suggested that adsorption occurs mainly by a chemical process dominated by monolayer adsorption. MPs adsorbed MeHg to form MPs/ MeHg complex, which was ingested by zebrafish larvae, and promoted accumulation of MeHg. Thus, the presence of MPs aggravated the reduction of locomotor activity induced by MeHg, and downregulation of neuro-transmitters related genes, such as ache, gfap and scl1A3b. Metabolome analysis also revealed disrupted glutathione (GSH) metabolism upon exposure of MeHg alone and in combination with MPs, as reflected by the increased in the ratio of GSH and oxidized glutathione. These effects were also confirmed by upregulation of oxidative stress-related genes, such as sod, sod mt and gpx4a. Collectively, these results indicated that MPs could act as a carrier of MeHg and enhance its accumulation in zebrafish, thereby disrupting locomotor activity by excessive oxidative stress. This study provides a scientific basis for improving health risk assessment of environmental pollutants, particularly those potentially able to adsorb to MPs by virtue of their chemical nature.
Toxic effect of chronic exposure to polyethylene nano/microplastics on oxidative stress, neurotoxicity and gut microbiota of adult zebrafish (Danio rerio)

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.

Abstract:<p>Microplastics, as emerging pollutant, are predicted to act as carriers for organic pollutants, but the carrier role and bio-toxic effects with other pollutants in environments are poorly acknowledged. In this study, both the single and combined effects of polyethylene (PE, 10 and 40 mg/L) with the particle size of 100-150 μm and 9-Nitroanthracene (9-NAnt, 5 and 500 μg/L) on zebrafish (<em>Danio rerio</em>) had been investigated. The results illustrated that PE could be as 9-NAnt carrier to enter into zebrafish body, but significantly reduced the bioaccumulation of 9-NAnt, due to the occurrence of adsorption interactions between the simultaneous presence of both PE and 9-NAnt. After 4 days, the enzymes activity of cytochrome P4501A, acetylcholinesterase (AChE), superoxide dismutase (SOD), catalase (CAT), lactate dehydrogenase (LDH), and the abundance of malondialdehyde (MDA), lipid peroxide (LPO) responded strongly to low-dose PE exposure (10 mg/L). After 7 days exposure to PE-9-NAnt (40 mg/L), the P4501A activity increased significantly, but the activities of AChE and LDH were inhibited clearly, causing certain neurotoxicity and disorders of energy metabolism to zebrafish. The analysis of integrated biomarker response index (IBR) suggested that PE had greater bio-toxicity to zebrafish in all exposure groups after short-term exposure, but the PE-9-NAnt complex showed greater bio-toxicity after 7 days, which indicated that complex exposure of PE-9-NAnt had a delayed effect on the bio-toxicity of zebrafish. Furthermore, analysis of the intestinal microbiota exhibited that under the conditions of the exposure group with 9-NAnt, the relative abundance of the five dominant bacterial phyla (Proteobacteria, Firmicutes, Fusobacteriota, Bacteroidota and Verrucomicrobiota) changed greatly. Overall, this study confirmed that PE could carry 9-NAnt into fish causing bioaccumulation, but in the case of coexisting exposures, PE reduced 9-NAnt bioaccumulation, suggesting that microplastics with other emerging pollutants in chronic toxicity are probably next objects in future works.</p>

Abstract:Microplastics (MPs) can adsorb heavy metals and induce combined toxicity to aquatic organisms. However, the combined effects on the gut-liver and gut-brain axes are yet to be fully comprehended. This study investigated the combined effects of polystyrene microplastics (PS-MPs) with two concentrations (20 and 200 μg/L) and three sizes (0.1, 10, and 250 μm) and Pb (50 μg/L) on zebrafish through gut-liver and gut-brain axes. The results showed that the combined exposure of 0.1 μm PS-MPs and Pb resulted in the most significant changes to the community diversity of gut microbiota. The combined exposures of PS-MPs (0.1 μm and 250 μm PS-MPs) and Pb significantly down-regulated expression of zo-1 and occludin but increased the lipopolysaccharide content in zebrafish liver compared to the PS-MPs or Pb alone exposure groups, indicating impaired gut barrier function. Subsequent studies showed that combined exposure of PS-MPs (0.1 μm and 250 μm) and Pb induced liver inflammation through the TLR4/NF-κB pathway. Moreover, all exposure groups had an impact on the expression of genes related to bile acid metabolism (cyp7a1, fgf19, abcb11b, and slc10a2) and neurotransmitters (tph1a, tph2, pink, and trh). The findings of this study provide new evidence on the combined effects of MPs and heavy metals, which are significant for their hazard identification and risk assessment.

Abstract:In recent years, emerging research has revealed that microplastics (MPs) can pollute the air. Cellulose and plastic fibers have been found in the lungs of patients with lung cancer. indicating that MPs can "travel" through the atmosphere into the respiratory tract. Due to the hydrophobicity and relatively large surface area of MPs, the biotoxicity of MPs is greatly enhanced by their adsorption and enrichment in persistent organic pollutants (POPs). With the rapid development of the economy, emissions from coal burning and automobile exhaust have made polycyclic aromatic hydrocarbon (PAHs) one of the key pollutants in urban air that cannot be ignored. Epidemiological studies have shown that PAH exposure is associated with the development of respiratory diseases, such as bronchitis and asthma, in children. The aim of this study was to investigate the inflammatory effects of co-exposure to different size polystyrene MPs (PS-MPs) and benzo[a]pyrene (BaP) on adult zebrafish gills and bronchial-related cells to provide basic information for assessment of the potential health risks of combined exposure to the human respiratory system. PS-MPs (with size of 0.5 and 5 mu m) and BaP were co-administered to embryos for 7 d, adult zebrafish for 14 d. and HBE cells and HBMSCs for 48 h. The relative expression levels of inflammatory cytokine genes in the different exposure groups were determined by real-time polymerase chain reaction (PCR). The ingestion of microplastics into fish bodies and cells was observed by stereoscope and confocal microscopy, respectively. Result shows that microplastics of two sizes could be ingested by zebrafish embryos. When exposed only to 0.5-mu m PS-MPs, relative expression of the TNF-alpha, IL-6, and IL-8 genes was significantly increased compared with that in the control; relative expression of the IFN-gamma gene was largely increased in the gills of zebrafish in 5-pin PS-MP-exposed group. Moreover, relative expression of the IFN-gamma, IL-6 and IL-8 genes in the group co-exposed to BaP and 0.5-mu m PS-MPs was higher than that in BaP group. In addition. 0.5-mu m PS-MPs could be ingested by both cell lines. Upon exposure to 0.5gm PS-MPs alorw, relative expression of genes TNF-alpha, IL-6. and IL-8 was considerably increased compared with that in the control HBE cells. In HBSMCs, upon exposure to 0.5-mu m PS-MPs alone, relative expression of the TNF-alpha, IL-8. and IL-1 beta genes was substantially increased compared with that in the control group. Compared with that following exposure to BaP alone. relative expression of the TNF-gamma and IL-6 genes in HBSMCs in 0.5-mu m PS-MPs and BaP co-exposure group was remarkably increased, indicating a possible combined effect; however, co-exposure of 5-mu m PS-MPs and BaP weakened the proinflammatory function of BaP and especially influenced relative expression of the TNF-alpha and IL-8 genes. In addition. none of the co-exposure groups of HBE cells showed obvious combined effects. The corollary is that particle size may affect the ingestion of PS-MPs in cells. In addition, the size-dependent effects of PS-MPs influence the accumulation of organic pollutants and their biotoxicity.

Abstract:One of the most pressing global environmental issues is the widespread abundance and distribution of microplastics (MPs). MPs can act as vectors for other contaminants in the environment making these small plastic particles hazardous for ecosystems. The presence of MPs in aquatic environments may pose threats to aquatic organisms that ingest them. This study examined effects of abamectin (ABM) and polyethylene terephthalate (PET) MP fragments on histopathological and enzymatic biomarkers in zebrafish ( Danio rerio ). Zebrafish were exposed for 96 h to pristine PET-MPs at concentrations of 5 mg/L and 10 mg/L, ABM alone at 0.006 mg/L, and the same concentration of ABM in the presence of PET-MPs in aquaria. Histopathological analysis revealed tissue content changes in liver and kidney in the presence of ABM individually and in combination with MPs. Results of enzymatic analysis showed that MPs increased the bioavailability and toxicity of pesticides due to inhibition of catalase (CAT) and acid phosphatase (ACP) enzymes. However, MPs did not affect the toxicity of ABM for glutathione s-transferase (GST) enzyme. Despite the inhibition of acetylcholinesterase (AChE) in MPs or ABM treatments, and some neurotoxicity, no change in activity of this enzyme and neurotoxicity was observed in the combined MPs and ABM treatments, although toxicity effects of MPs and ABM on zebrafish require more detailed studies.

Abstract:Microplastics (MPs) are ubiquitous in the environment, can adsorb organic contaminants (OCs) and be taken by various microorganisms and organisms, which could eventually lead to risk to humans. In this study, the phenotypic changes and metabolic profile alternations of zebrafish in the single- and co-exposure of MPs and phenanthrene (Phe) were investigated. The results showed that significantly higher tail malformation rate and edema rate in zebrafish induced by MPs can be enhanced with the co-existence of Phe. The metabolomic analysis revealed that both synergistic and antagonistic effects of MPs and Phe on the metabolic alternation of zebrafish larvae exist, since unique perturbations of metabolites or pathways were found in all of the three exposure scenarios. Based on PLS-DA, porphine, ribose, and L-glutamic acid were the most important metabolites resulting in the difference between the treated and control groups in the MP exposure, Phe exposure and co-exposure, respectively. Two dysregulated pathways namely d-glutamine and D-glutamate metabolism, and alanine, aspartate and glutamate metabolism were significantly affected in the co-exposure while not in either of the single exposure. These findings provide new insights into the toxic effects of MPs on aquatic organisms, and further studies on combined effects of MPs and OCs are suggested to be conducted.

Abstract:The co-existence of microplastics (MPs) and methamphetamine (METH) in aquatic ecosystems has been widely reported; however, the joint toxicity and associated mechanisms remain unclear. Here, zebrafish larvae were exposed individually or jointly to polystyrene (PS) and polyvinyl chloride (PVC) MPs (20 mg/L) and METH (1 and 5 mg/L) for 10 days. The mortality, behavioral functions, and histopathology of fish from different groups were determined. PS MPs posed a stronger lethal risk to fish than PVC MPs, while the addition of METH at 5 mg/L significantly increased mortality. Obvious deposition of MPs was observed in the larvae's intestinal tract in the exposure groups. Meanwhile, treatment with MPs induced intestinal deposits and intestinal hydrops in the fish, and this effect was enhanced with the addition of METH. Furthermore, MPs significantly suppressed the locomotor activation of zebrafish larvae, showing extended immobility duration and lower velocity. METH stimulated the outcome of PS but had no effect on the fish exposed to PVC. However, combined exposure to MPs and METH significantly increased the turn angle, which declined in individual MP exposure groups. RNA sequencing and gene quantitative analysis demonstrated that exposure to PS MPs and METH activated the MAPK signaling pathway and the C-type lectin signaling pathway of fish, while joint exposure to PVC MPs and METH stimulated steroid hormone synthesis pathways and the C-type lectin signaling pathway in zebrafish, contributing to cellular apoptosis and immune responses. This study contributes to the understanding of the joint toxicity of microplastics and pharmaceuticals to zebrafish, highlighting the significance of mitigating microplastic pollution to preserve the health of aquatic organisms and human beings.

Abstract:Despite extensive investigation on the toxicity of microplastics (MPs), an emerging global concern, little is known about the combined toxicity of MPs and co-occurring pollutants in aquatic environments. In this study, the combined toxicity of polystyrene MPs and sulfamethoxazole (SMZ) antibiotics was explored in zebrafish embryos in terms of the developmental, physiological, and endocrine toxicities. Exposure to PS and SMZ induced mortality (rate: 25.0 ± 7.5%) and malformation (rate: 20~35%) at multiple regions and stages of zebrafish development. Physiological toxicity was also induced as shown by the significant decrease in fetal movement (by 31.1~37.0%) and swimming frequency (by 26.9~36.8%) and the increase in heartbeat rate (by 19.0~20.9%). Finally, PS and SMZ exposure also induced extensive endocrine toxicities in zebrafish as confirmed by increases in various biomarkers including vitellogenin, 17β-estradiol, testosterone, and triiodothyronine. The combination index showed that antagonistic effects were present between PS and SMZ toxicity, which slightly decreased their combined toxicity. This study aims to further understand the combined toxicity of MPs and co-occurring pollutants in aquatic environments.

Abstract:Microplastics (MPs) and polyhalogenated carbazoles (PHCZs) are widely detected in the aquatic environment, and their ecological risks have become a research focus. Although there is an extensive co-distribution of MPs and PHCZs, their combined toxicity to aquatic organisms is still unclear. This study investigated the toxic effects of polystyrene microplastics (PS-MPs) and 3,6-dibromocarbazole (3,6-DBCZ) on zebrafish embryos by individual/combined exposure. This study showed that individual or combined exposure of PS-MPs (10 mg/L) and 3,6-DBCZ (0.5 mg/L) could significantly increase the rate of zebrafish embryo deformity, whereas no significant effect was observed on mortality and hatching rate. Furthermore, exposure to 3,6-DBCZ or PS-MPs increased reactive oxygen species (ROS) levels in zebrafish embryos, and the resulting oxidative stress induced apoptosis. Comparably, the levels of oxidative stress and apoptosis in zebrafish embryos were significantly reduced with the combined exposure of 3,6-DBCZ and PS-MPs. These observations suggest that the combined exposure of 3,6-DBCZ and PS-MPs has an antagonistic effect on oxidative stress and apoptosis. Fluorescence PS-MPs tracing and 3,6-DBCZ enrichment analysis showed that, with the protection of chorion, the entry of PS-MPs (5 and 50 μm) into the embryonic stage (55 hpf) of zebrafish was prevented. Moreover, after exposure for 96–144 hpf, PS-MPs serve d as a carrier to promote the 3,6-DBCZ accumulation and its dioxin-like toxicity in zebrafish larvae through ingestion. Compared with 5-μm PS-MPs, 50-μm PS-MPs promoted higher accumulation and dioxin-like toxicity of 3,6-DBCZ in zebrafish larvae. These findings provide that MPs can be used as an important carrier of PHCZs, influencing their toxicity and bioaccumulation in the organisms.

Abstract:Microplastics (MPs) and Perfluorooctanoic acid (PFOA) have contaminated nearly all types of ecosystems, including marine, terrestrial and freshwater habitats, posing a severe threat to the ecological environment. However, their combined toxicity on aquatic organisms (e.g., macrophytes) remains unknown. This study investigated single and combined toxic effects of polypropylene (PP), polyethylene (PE), polyvinylchloride (PVC), polyethylene terephthalate (PET) and PFOA on Vallisneria natans (V. natans) and associated biofilms. Results showed that MPs and PFOA significantly affected plant growth, while the magnitude of the effect was associated with concentrations of PFOA and the types of MPs, and antagonistic effects were induced at combined MPs and PFOA exposure. In addition, antioxidant responses in plants, such as promoted activities of SOD and POD, as well as increased content of GSH and MDA, were triggered effectively by exposure to MPs and PFOA alone and in combination. Ultrastructural changes revealed the stress response of leaf cells and the damage to organelles. Moreover, single and combined exposure to MPs and PFOA altered the diversity and richness of the microbial community in the leaf biofilms. These results indicated that the coexistence of MPs and PFOA can induce effective defense mechanisms of V. natans and change the associated biofilms at given concentrations in the aquatic ecosystems.

Abstract:The toxicological interactions of microplastics (MPs) and heavy metals have been paid much attention in aquatic organism. The mechanisms are not fully clear, particularly in fish early life stages. To the end, zebrafish embryos were exposed to 500 μg/L MPs, 5 μg/L cadmium (Cd), and their combination for 30 days. Body weight, adsorption characteristics of Cd onto MPs, Cd accumulation, oxidative stress, apoptosis, and growth hormone/insulin-like growth factor-I (GH/IGF) axis were examined. Exposure to MPs and Cd alone reduced body weight, which was aggravated by co-exposure. An increase in reactive oxygen species (ROS) levels was observed in larvae exposed to Cd or MPs + Cd, suggesting an induction of oxidative stress. Lipid peroxidation levels were not affected by exposure to MPs and Cd alone but dramatically enhanced by co-exposure, which may be explained by the reduction of total antioxidant capacity (TAOC) and activity levels of Mn-superoxide dismutase (Mn-SOD) and catalase (CAT) after co-exposure. Increased apoptotic cells were observed in the vertebral body of larvae exposed to Cd, the esophagus of larvae exposed to MPs, and both organs of larvae exposed to MPs + Cd, which was further confirmed by changes in the activities of Caspase-3, Caspase-8 and Caspase-9. PCR array on the transcription of genes related to growth, oxidative stress and apoptosis was examined, showing that the combined exposure resulted in greater magnitude of changes than MPs and Cd alone. The results indicate that MPs can enhance the negative effects of Cd on growth, oxidative damage and apoptosis in early life stages of zebrafish. However, the adsorption of Cd onto MPs was not observed and the combined exposure did not increase the Cd content in larvae compared to the single Cd exposure, implying that vector role of MPs in Cd uptake is negligible.

Abstract:Although the combined effects of microplastics (MPs) and other organic pollutants have raised increasing attention, the impacts of polymer types on the biological effects (e.g., bioaccumulation and toxicity) of the mixtures are still unclear. This study aimed to evaluate the influence of different polymer types of MPs including polyethylene (PE), polypropylene (PP), and polyvinyl chloride (PVC) on the adsorption, accumulation, and toxic effects of triclosan (TCS) in zebrafish. As a result, all three types of MPs could adsorb TCS and PP-MPs has the highest adsorption capacity for TCS (1.18 mg/g). Compared with the TCS alone, MPs changed the distribution of TCS in tissues and increased the accumulation of TCS in the liver and gut following the order of TCS + PP > TCS + PVC > TCS + PE. Compared with individual TCS and PP-MPs, after co-exposed for 28 days, TCS + PP significantly aggravated oxidative stress and lipid peroxidation in the liver as well as enhanced neurotoxicity in the brain. Moreover, TCS + PP disturbed the metabolism in the liver and MPs contributed more to the metabolic disorders. The upregulated lipid metabolites (e.g., sphingosine and L-palmitoylcarnitine) and downregulated carbohydrate metabolites (e.g., sucrose) could be potential targets for future risk assessment of MPs combined with other pollutants.

Abstract:Microplastic particles (MPs) are widely distributed in the environment. The high surface ratio of MPs make them effective transmission mediums for many toxic pollutants. The combined toxicity of MPs and heavy metals have received increasing attention in recent years. In this study, effects of MPs (100 μg/L) on the toxicity of low (15 μg/L) and high (150 μg/L) concentrations of cadmium (Cd) to zebrafish were evaluated based on a 10-day subacute exposure. The survival rate, growth, antioxidant capacity, reactive oxygen species (ROS) accumulation, histology and Cd biological enrichment in different tissues were investigated with the objective to understand the effect and mechanism of MPs on Cd toxicity to zebrafish. The results showed that the effect of MPs on Cd toxicity mainly depended on the concentration of Cd. MPs significantly enhanced the toxicity of low concentrations of Cd (LCd), including lower antioxidant enzyme activities, higher ROS levels, more severe tissue damage, inhibited growth rate and lower survival rate. However, the effects of MPs on the toxicity of high concentrations of Cd (HCd) were exactly opposite to LCd. Cd enrichment analysis showed that MPs could significantly increase LCd accumulation in intestine, gill, skin and muscle tissues, while decrease the enrichment of HCd in liver, intestine, gill and muscle tissues. Free Cd in the exposure water was significantly decreased by MPs in the HCd and MPs combined exposure group. These results suggest that effect of MPs on Cd toxicity to zebrafish depending on Cd concentration, MPs can increase the enrichment of LCd in zebrafish and enhance its toxicity, but can decrease the enrichment of HCd in zebrafish and attenuate its toxicity. The present study will broaden our understanding of the interaction between MPs and heavy metals.
