Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.
Co-exposure Impact of Nickel Oxide Nanomaterials and Bacillus subtilis on Soybean Growth and Nitrogen Assimilation Dynamics

Abstract:Nickel oxide nanoparticles (NiO-NPs) pose potential threats to agricultural production. Bacillus subtilis has emerged as a stress-mitigating microbe that alleviates the phytotoxicity caused by NiO-NPs. However, the mechanisms underlying its effectiveness, particularly in root-nodule symbiosis and biological N2-fixation (BNF), remain unclear. Here, we tested the combined exposure of NiO-NPs (50 mg kg−1) and B. subtilis on soybean (Glycine max L.) growth and BNF. Combined exposure increased root length, shoot length, root biomass, and shoot biomass by 19-26%, while Ni (200 mg kg−1) reduced them by 38--53% compared to the control. NiO-NPs at 100 and 200 mg kg−1 significantly (P < 0.05) reduced nodule formation by 16% and 58% and Nitrogen assimilation enzyme activities levels (UE, NR, HS, and GOGAT) by 13-57%. However, co-exposure with B. subtilis improved nodule formation by 22-44%. Co-exposure of NiO-NPs (200 mg kg−1) with B. subtilis increased POD, CAT, and GSH-Px activity levels by 20%, 16%, and 14% while reducing MDA (14%) and H2O2 (12%) levels compared to NiO-NPs alone. Additionally, co-exposure of NiO-NPs (100 and 200 mg kg−1) with B. subtilis enhanced the relative abundance of Stenotrophomonas, Gemmatimonas, and B. subtilis, is associated with N2-cycling and N2-fixation potential. This study confirms that B. subtilis effectively mitigates NiO-NP toxicity in soybean, offering a sustainable method to enhance BNF and crop growth and contribute to addressing global food insecurity.

Abstract:The rapid development of industrialization is causing several fundamental problems in plants due to the interaction between plants and soil contaminated with metallic nanoparticles (NPs). Numerous investigations have been conducted to address the severe toxic effects caused by nanoparticles in the past few decades. Based on the composition, size, concentration, physical and chemical characteristics of metallic NPs, and plant types, it enhances or lessens the plant growth at various developmental stages. Metallic NPs are uptaken by plant roots and translocated toward shoots via vascular system based on composition, size, shape as well as plant anatomy and cause austere phytotoxicity. Herein, we tried to summarize the toxicity induced by the uptake and accumulation of NPs in plants and also we explored the detoxification mechanism of metallic NPs adopted by plants via using different phytohormones, signaling molecules, and phytochelatins. This study was intended to be an unambiguous assessment including current knowledge on NPs uptake, accumulation, and translocation in higher plants. Furthermore, it will also provide sufficient knowledge to the scientific community to understand the metallic NPs-induced inhibitory effects and mechanisms involved within plants.
Discrepancy of Growth Toxicity of Polystyrene Nanoplastics on Soybean (Glycine max) and Mung Bean (Vigna radiata)

Abstract:Nanoplastics, as a hot topic of novel contaminants, lack extensive concern in higher plants; especially the potential impact and mechanism of nanoplastics on legume crops remains elusive. In this study, the toxicity of polystyrene nanoplastics (PS-NPs, 200 nm) with diverse doses (control, 10, 50, 100, 200, 500 mg/L) to soybean and mung bean plants grown hydroponically for 7 d was investigated at both the macroscopic and molecular levels. The results demonstrated that the root length of both plants was markedly suppressed to varying degrees. Similarly, mineral elements (Fe, Zn) were notably decreased in soybean roots, consistent with Cu alteration in mung bean. Moreover, PS-NPs considerably elevated malondialdehyde (MDA) levels only in soybean roots. Enzyme activity data indicated mung bean exhibited significant damage only at higher doses of PS-NPs stress than soybean, implying mung bean is more resilient. Transcriptome analysis showed that PS-NPs stimulated the expression of genes associated with the antioxidant system in plant roots. Furthermore, starch and sucrose metabolism might play a key role in coping with PS-NPs to enhance soybean resistance, but the MAPK pathway was enriched in mung bean. Our findings provide valuable perspectives for an in-depth understanding of the performance of plants growing in waters contaminated by nanoplastics.

Co-exposure to tire wear particles and nickel inhibits mung bean yield by reducing nutrient uptake

Abstract:Soil and terrestrial contamination with microplastics and nanoplastics has been discussed extensively, while tire wear particles (TWPs) have been largely overlooked. We investigated the root-surface interactions and growth response of mung bean (Vigna radiata L.) plants exposed to tire wear particles (TWPs) (0.05, 0.1, and 0.25% w/w) and nickel sulfate (50 and 100 mg kg-1 NiSO4) alone and in co-exposure scenarios for the full life cycle (105 days) under soil conditions. The results show that TWPs adhered to the root surface and reduced the water and nutrient uptake by the plant, particularly at higher concentrations of TWPs (0.25% w/w), without any observed organic contaminant accumulation in the root tissue. TWPs alone at 0.01, 0.1, and 0.25% (w/w) decreased mung bean yield by 11, 28, and 52%, respectively. Co-exposure to TWPs at 0.01, 0.1 and 0.25% w/w with 100 mg kg-1 NiSO4 decreased yield by 73, 79 and 88%, respectively. However, co-exposure to TWPs at 0.01 and 0.1% w/w with 50 mg kg-1 NiSO4 enhanced the yield by 32% and 7%, respectively. These changes in yield and nutritional aspects appear to be linked to Ni's regulatory influence on mineral homeostasis. Moreover, exposure to NiSO4 at 100 mg kg-1 increased Ni uptake in the root, shoot, and grain by 9, 26, and 20-fold, respectively as compared to the unamended control; this corresponded to increased antioxidant enzyme activity (10-127%) as compared to the control. TWPs caused blockages, significantly reducing plant yield and altering nutrient dynamics, highlighting emerging risks to plant health.

Abstract:The excessive use of nickel (Ni) in manufacturing and various industries has made Ni a serious pollutant in the past few decades. As a micronutrient, Ni is crucial for plant growth at low concentrations, but at higher concentrations, it can hamper growth. We evaluated the effects of Ni concentrations on nitrate (NO3−) and ammonium (NH4+) concentrations, and nitrogen metabolism enzyme activity in rice seedlings grown in hydroponic systems, using different Ni concentrations. A Ni concentration of 200 μM significantly decreased the NO3− concentration in rice leaves, as well as the activities of nitrate reductase (NR), nitrite reductase (NiR), glutamine synthetase (GS), and glutamate synthetase (GOGAT), respectively, when compared to the control. By contrast, the NH4+ concentration and glutamate dehydrogenase (GDH) activity both increased markedly by 48% and 46%, respectively, compared with the control. Furthermore, the activity of most active aminotransferases, including glutamic pyruvic transaminase (GPT) and glutamic oxaloacetic transaminase (GOT), was inhibited by 48% and 36%, respectively, in comparison with the control. The results indicate that Ni toxicity causes the enzymes involved in N assimilation to desynchronize, ultimately negatively impacting the overall plant growth.

Abstract:The effects of emerging contaminants micro/nanoplastics (MPs/NPs) and silver nanoparticles (Ag NPs) on health have attracted universal concern throughout the world. However, it is unclear on the combined effects of MPs/NPs and Ag NPs on the biogeochemistry cycle such as nitrogen transformation and functional microorganism in the soil. In the present study, we conducted a 45-day soil microcosm experiment with polystyrene (PS) MPs/NPs and Ag NPs to investigate their combined impact on nitrogen cycling and the bacterial community. The results showed that MPs or NPs exerted limited effects on nitrogen transformation in the soil. The combined effects of PS MPs/NPs and Ag NPs were mainly caused by the presence of Ag NPs. However, PS NPs alleviated the inhibition of anammox and denitrification induced by Ag NPs via upregulating anammox-related genes and elevating nitrate and nitrite reductase activities. PS MPs + Ag NPs treatment significantly reduced bacterial diversity. PS MPs/NPs + Ag NPs increased the relative abundances of denitrifying Cupriavidus by 0.32% and 0.06% but decreased nitrogen-fixing functional microorganisms of Microvirga (by 2.05% and 2.24%), Bacillus (by 0.16% and 0.22%), and Herbaspirillum (by 0.14% and 0.07%) at the genus level compared with Ag NPs alone. The significant downregulation of nitrogen-fixing genes (K02586, K02588, and K02591) was observed in PS MPs/NPs + Ag NPs treatment compared to Ag NPs in the nitrogen metabolism pathway. Moreover, g-Lysobacter and g-Aquimonas were identified as biomarkers in PS MPs + Ag NPs and PS NPs + Ag NPs by LEfSe analysis. Our study sheds the light that changes of functional microorganism abundances contributed to the alteration of nitrogen transformation. Taking the particle size of plastics into account will be helpful to accurately assess the combined ecological risks of plastics and nanomaterial contaminants.

Abstract:Agricultural soils are increasingly undergoing inadvertent and purposeful exposures to engineered CeO 2 nanoparticles (NPs), which can impact crops and root-associated microbial communities. However, interactions between NP concentration and exposure duration on plant-mediated responses of root-associated bacterial communities are not well understood. Soybeans seedlings were grown in soil with uncoated NPs added at concentrations of 0, 1 or 100 mg kg −1 . Total soil exposure durations were either 190 days, starting 106 days before planting or 84 days with NP amendments coinciding with planting. We assessed plant development, bacterial diversity, differential abundance and inferred functional changes across rhizosphere, rhizoplane, and root tissue compartments. Plant non-monotonic dose responses were mirrored in bacterial communities. Most notably, effects were magnified in the rhizoplane under low-dose, short-exposures. Enriched metabolic pathways were primarily related to biosynthesis and degradation/utilization/assimilation, rather than responses to metals or oxidative stress. Our results indicate that plant-mediated bacterial responses were greater than direct NP impacts. Also, we identify needs for modeling non-monotonic legume stress responses that account for coinfection with mutualistic and parasitic bacteroids. Our findings provide new insights regarding effects of applications of soil amendments such as biosolids containing NPs or nano-enabled formulations used in cultivation of legumes and other crops.

Abstract:Nanotechnology is grabbing great attention all over the world because of its stimulating use in numerous fields, and the nanosilica (nSi) and carbon nanoparticles (CNPs) application has been examined in various studies. Conversely, the nSi and CNPs combinatorial use is a new method and researched in limited literature. For this purpose, a pot experiment was conducted to examine various growth and biochemical parameters in barley (Hordeum vulgare L.) under the toxic concentration of nickel (Ni) i.e., 200 mg kg-1 which were primed with combined application of two NPs of nSi at 3 mM and CNPs i.e., 200 μM respectively. The results showed that the Ni toxicity in the soil showed a significantly (P < 0.05) declined in the growth, gas exchange attributes, sugars, AsA-GSH cycle, cellular fractionation, proline metabolism in H. vulgare. However, Ni toxicity significantly (P < 0.05) increased oxidative stress biomarkers, enzymatic and nonenzymatic antioxidants including their gene expression in H. vulgare. Although, the application of nSi and CNPs showed a significant (P < 0.05) increase in the plant growth and biomass, gas exchange characteristics, enzymatic and non-enzymatic compounds and their gene expression and also decreased the oxidative stress, and Ni uptake. In addition, individual or combined application of nSi and CNPs enhanced the cellular fractionation and decreases the proline metabolism and AsA-GSH cycle in H. vulgare. These results open new insights for sustainable agriculture practices and hold immense promise in addressing the pressing challenges of heavy metal contamination in agricultural soils.

Abstract:The co-occurrence of heavy metals and microplastics (MPs) is an emerging issue that has attracted considerable attention. However, the interaction of nickel oxide nanoparticle (nano-NiO) combined with MPs in soil was poorly researched. Here, experiments were conducted to study the influence of nano-NiO (200 mg/kg) and polyethylene (PE) MPs with different concentrations (0.1, 1, and 10%) and sizes (13, 50, and 500 μm) on earthworms for 28 days. Compared to control, the damage was induced by PE and nano-NiO, which was evaluated by biomarker Integrated Biomarker Response index: version 2 (IBRv2) based on six biomarkers including SOD, POD, CAT, MDA, AChE, Na + /K + -ATPase and cellulase. The majority of the chosen biomarkers showed significant but complicated responses with increasing contaminant concentrations after 28 days of exposure. Moreover, the joint effect was assessed as antagonism by the effect addition index (EAI). Overall, this work expands our understanding of the combined toxicity of PE and nano-NiO in soil ecosystems.

Abstract:Based on previously published hydroponic plant, planktonic bacterial, and soil microbial community research, manufactured nanomaterial (MNM) environmental buildup could profoundly alter soil-based food crop quality and yield. However, thus far, no single study has at once examined the full implications, as no studies have involved growing plants to full maturity in MNM-contaminated field soil. We have done so for soybean, a major global commodity crop, using farm soil amended with two high-production metal oxide MNMs (nano-CeO 2 and -ZnO). The results provide a clear, but unfortunate, view of what could arise over the long term: ( i ) for nano-ZnO, component metal was taken up and distributed throughout edible plant tissues; ( ii ) for nano-CeO 2 , plant growth and yield diminished, but also ( iii ) nitrogen fixation—a major ecosystem service of leguminous crops—was shut down at high nano-CeO 2 concentration. Juxtaposed against widespread land application of wastewater treatment biosolids to food crops, these findings forewarn of agriculturally associated human and environmental risks from the accelerating use of MNMs.
Effects of microplastics on N 2 O production and reduction potential in crop soils of northern China

Abstract:Microplastics (MPs) pollution are found to be increasing in vegetable soils and potentially affecting N 2 O production and their associated pathways; however, its specific effects remain unclear. Here, we selected two common MPs, PE and PP at four different concentration levels of 0, 0.5, 1.5 and 3%, and conducted several incubation experiments aiming to explore soil bacterial and fungal N 2 O production. Results showed that the bacteria were the main contributors for the production of N 2 O, regardless of the absence or presence of MPs; and its contribution was decreased with increasing concentrations of PE and PP. The nosZ clade I and II genes were positively correlated with N 2 O reduction rates, indicating a combined regulation on soil N 2 O reduction. PE significantly inhibited the bacterial nitrification and denitrification, but did not affect the total N 2 O production rates; while PP significantly reduced both the bacterial and fungal N 2 O production rates. The resistance of fungal N 2 O production to MPs pollution was stronger than that of the bacterial N 2 O production. It highlights that the MPs pollution could reduce the potential of N 2 O production and reduction, and thus disturb soil nitrogen cycling system; while the inhibition on N 2 O production via bacteria and fungi varies with different types of MPs. This study is conducive to an improved and more comprehensive understanding of the ecological impacts of MPs within the agroecosystem.

Abstract:As an exogenous carbon input, microplastics (MPs), especially biodegradable MPs, may significantly disrupt soil microbial communities and soil element cycling (CNPS cycling), but few studies have focused on this. Here, we focused on assessing the effects of conventional low-density polyethylene (LDPE), biodegradable polybutylene adipate terephthalate (PBAT), and polylactic acid (PLA) MPs on rhizosphere microbial communities and CNPS cycling in a soil-soybean system. The results showed that PBAT-MPs and PLA-MPs were more detrimental to soybean growth than LDPE-MPs, resulting in a reduction in shoot nitrogen (14.05% and 11.84%) and shoot biomass (33.80% and 28.09%) at podding stage. In addition, dissolved organic carbon (DOC) increased by 20.91% and 66.59%, while nitrate nitrogen (NO– 3-N) significantly decreased by 56.91% and 69.65% in soils treated with PBAT-MPs and PLA-MPs, respectively. PBAT-MPs and PLA-MPs mainly enhanced copiotrophic bacteria (Proteobacteria) and suppressed oligotrophic bacteria (Verrucomicrobiota, Gemmatimonadota, etc.), increasing the abundance of CNPS cycling-related functional genes. LDPE-MPs tended to enrich oligotrophic bacteria (Verrucomicrobiota, etc.) and decrease the abundance of CNPS cycling-related functional genes. Correlation analysis revealed that MPs with different degradation properties selectively affected the composition and function of the bacterial community, resulting in changes in the availability of soil nutrients (especially NO– 3-N). Redundancy analysis further indicated that NO– 3-N was the primary constraining factor for soybean growth. This study provides a new perspective for revealing the underlying ecological effects of MPs on soil–plant systems.

Abstract:Nanoplastics (NPs) have emerged as global environmental pollutants with concerning implications for sustainable agriculture. Understanding the underlying mechanisms of NPs toxicity and devising strategies to mitigate their impact is crucial for crop growth and development. Here, we investigated the nanoparticles of zinc oxide (nZnO) to mitigate the adverse effects of 80 nm NPs on fragrant rice. Our results showed that optimized nZnO (25 mg L −1 ) concentration rescued root length and structural deficits by improving oxidative stress response, antioxidant defense mechanism and balanced nutrient levels, compared to seedlings subjected only to NPs stress (50 mg L −1 ). Consequently, microscopy observations, Zeta potential and Fourier transform infrared (FTIR) results revealed that NPs were mainly accumulated on the initiation joints of secondary roots and between cortical cells that blocks the nutrients uptake, while the supplementation of nZnO led to the formation of aggregates with NPs, which effectively impedes the uptake of NPs by the roots of fragrant rice. Transcriptomic analysis identified a total of 3973, 3513 and 3380 differentially expressed genes (DEGs) in response to NPs, nZnO and NPs+nZnO, respectively, compared to the control. Moreover, DEGs were significantly enriched in multiple pathways including biosynthesis of secondary metabolite, phenylpropanoid biosynthesis, amino sugar and nucleotide sugar metabolism, carotenoid biosynthesis, plant-pathogen interactions, MAPK signaling pathway, starch and sucrose metabolism, and plant hormone signal transduction. These pathways could play a significant role in alleviating NPs toxicity and restoring fragrant rice roots. Furthermore, metabolomic analysis demonstrated that nZnO application restored 2-acetyl-1-pyrroline (2-AP) pathways genes expression, enzymatic activities, and the content of essential precursors related to 2-AP biosynthesis under NPs toxicity, which ultimately led to the restoration of 2-AP content in the leaves. In conclusion, this study shows that optimized nZnO application effectively alleviates NPs toxic effects and restores both root structure and aroma production in fragrant rice leaves. This research offers a sustainable and practical strategy to enhance crop production under NPs toxicity while emphasizing the pivotal role of essential micronutrient nanomaterials in agriculture.

Abstract:Interest has been generated in nanotechnology application in agricultural and food systems and understanding ecotoxicity of nanoparticles (NPs) is imperative for ecosystem health and food safety. This study compared the dose-effects of newly synthesized polysuccinimide NPs (PSI-NPs) with mull-walled carbon nanotubes (MWCNTs), nano-Ag and nano-TiO2, on soil microbiological and biochemical processes. The effects of NPs varied largely with the type and dose of NPs as well as soil properties. Of the tested NPs, PSI-NPs had no significant perturbance, while the rest of NPs decreased microbial biomass C and N (MBC and MBN), stimulated the microbial metabolic quotient (MMQ), and inhibited dehydrogenase (DA), urease (UA), and phosphatase (PA) activities. Significant dose-effects of MWCNTs, nano-Ag and nano-TiO2 occurred, and the influences were more intensive at higher application rate (500 ppm). Irrespective of NP type and dose, the effects were severer in Spodosol soil than Alfisol soil. Principle component analysis (PCA) also confirmed that most of the measured indicators were not affected by PSI-NPs, partly affected by MWCNTs, but intensively influenced by nano-Ag and nano-TiO2. These results highlight the advantages of PSI-NPs over the other NPs for agricultural applications and could facilitate the development of guidelines in regulating NPs application in agroecosystems.

Abstract:The development of nanotechnology and the widespread application of metal-based nanoparticles (MNPs) in various consumer products have released a large number of nanoparticles into soil environment, especially agricultural systems. The accumulation of MNPs in the soil has threatened the soil health and plant growth. This review uses Ag NPs and ZnO NPs which have been widely used in commercial products as examples, combined with other kinds of MNPs (such as TiO 2 , CuO, CeO 2 , etc. ) to discuss the interplay of MNPs with plant rhizosphere microenvironment. Plant rhizosphere properties, such as soil pH, organic matter, microorganisms, or rhizosphere exudates, can affect the environmental behaviors of MNPs. Further MNPs can alter rhizosphere soil chemistry and microbial properties which in turn affect the MNP behaviour and biological effects. Meanwhile, MNPs can cause damage to roots or be absorbed by plant roots to induce oxidative stress in plant roots, affecting plant root growth and nutrient uptake. Finally, given the inadequacies of current research on MNPs and soil–plant systems, several future research priorities are recommended.
Size Effects of Copper Oxide Nanoparticles on Boosting Soybean Growth Via Differentially Modulating Nitrogen Assimilation

Abstract:Nanoscale agrochemicals have been widely used in sustainable agriculture and may potentially affect the nitrogen fixation process in legume crops. The present study investigated the size-effects of copper oxide nanoparticles (CuO NPs) on nitrogen assimilation in soybean (G. max (L.) Merrill) plants, which were treated with different sizes (20 and 50 nm) of CuO NPs at low use doses (1 and 10 mg/kg) for 21 days under greenhouse conditions. The results showed that 50 nm CuO NPs significantly increased the fresh biomass more than 20 nm CuO NPs achieved at 10 mg/kg. The activities of N assimilation-associated enzymes and the contents of nitrogenous compounds, including nitrates, proteins, and amino acids, in soybean tissues were greatly increased across all the CuO NP treatments. The use doses of two sizes of CuO NPs had no impact on the Cu contents in shoots and roots but indeed increased the Cu contents in soils in a dose-dependent fashion. Overall, our findings demonstrated that both 20 and 50 nm CuO NPs could positively alter soybean growth and boost N assimilation, furthering our understanding that the application of nanoscale micro-nutrient-related agrochemicals at an optimal size and dose will greatly contribute to increasing the yield and quality of crops.
Physiological Effects of Nanoparticulate ZnO in Green Peas (pisum Sativum L.) Cultivated in Soil

Abstract:The toxicological effects of zinc oxide nanoparticles (ZnO NPs) in plants are still largely unknown. In the present study, green pea (Pisum sativum L.) plants were treated with 0, 125, 250, and 500 mg kg(-1) of either ZnO NPs or bulk ZnO in organic matter enriched soil. Corresponding toxicological effects were measured on the basis of plant growth, chlorophyll production, Zn bioaccumulation, H2O2 generation, stress enzyme activity, and lipid peroxidation using different cellular, molecular, and biochemical approaches. Compared to control, all ZnO NP concentrations significantly increased (p ≤ 0.05) root elongation but no effects were observed in the stem. Whereas all bulk ZnO treatments significantly increased both root and stem length. After 25 days, chlorophyll in leaves decreased, compared to control, by ~61%, 67%, and 77% in plants treated with 125, 250, and 500 mg kg(-1) ZnO NPs, respectively. Similar results were found in bulk ZnO treated plants. At all ZnO NP concentrations CAT was significantly reduced in leaves (p ≤ 0.05), while APOX was reduced in both roots and leaves. In the case of bulk ZnO, APOX activity was down-regulated in the root and leaf and CAT was unaffected. At 500 mg kg(-1) treatment, the H2O2 in leaves increased by 61% with a twofold lipid peroxidation, which would be a predictive biomarker of nanotoxicity. This study could be pioneering in evaluating the phytotoxicity of ZnO NPs to green peas and can serve as a good indicator for measuring the effects on ZnO NPs in plants grown in organic matter enriched soil.
