Abstract:Copper-based nanoparticles (NPs) display a strong potential to replace copper salts (e.g., CuSO4) for application in agricultures as antimicrobial agents or nutritional amendments. Yet, their effects on crop quality are still not comprehensively understood. In this study, the Cu contents in soybeans grown in soils amended with Cu NPs and CuSO4 at 100-500 mg Cu/kg and the subsequent effects on the plant physiological markers were determined. The Cu NPs induced 29-89% at the flowering stage (on Day 40) and 100-165% at maturation stage (on Day 100) more Cu accumulation in soybeans than CuSO4. The presence of particle aggregates in the root cells with deformation upon the Cu NP exposure was observed by transmission electron microscopy. The Cu NPs at 100 and 200 mg/kg significantly improved the plant height and biomass, yet significantly inhibited at 500 mg/kg, compared to the control. In leaves chlorophyll-b was more sensitive than chlorophyll-a and carotenoids to the Cu NP effect. The Cu NPs significantly decreased the root nitrogen and phosphorus contents, while they significantly increased the leaf potassium content in comparison with control. Our results imply that cautious use of Cu NPs in agriculture is warranted due to relatively high uptake of Cu and altered nutrient quality in soybeans.
Effects of iron oxide nanoparticles on the mineral composition and growth of soybean (Glycine max L.) plants

Abstract:Nano-fertilization of agricultural systems is emerging as an innovative and unique strategy for agronomic fortification. In this study, a laboratory experiment was conducted to evaluate the physiological performance of soybean (Glycine max L.) exposed to either foliar or soil amendments of Fe2O3 nanoparticles (nFe2O3; 15, 30, and 60 mg/pot), fulvic acid-coated nFe2O3 (nFe2O3-FA; 60 mg nFe2O3/pot), and Fe-EDTA during an eight-week growing period. The experimental results demonstrated that none of the treatments produced toxicity stress or growth disorders. Amendments of nFe2O3 and nFe2O3-FA remarkably enhanced chlorophyll content, plant biomass, and root developmental indices. Fe-EDTA, a conventional fertilizer, did not provide satisfactory results compared with the other treatments. Regarding the supply of iron (Fe), foliar amendment provided 2–4 times higher shoot Fe concentration than soil application. Foliar amendments of nFe2O3-FA exclusively stimulated biological nitrogen fixation, which was primarily reflected in the formation of root nodules. This finding might be associated with the binary effect of the fulvic-Fe supply as well as enhanced potassium and zinc absorption in the plant than that of selective Fe penetration via negatively charged complexes. Further analysis of soil pH showed no evidence for facilitated Fe uptake via rhizosphere acidification. Conclusively, soybean responded better to the foliar amendment of nFe2O3-FA than nFe2O3 alone. This result suggests a novel perspective on the potential application of natural nanomaterial coating agents for nano-fertilization. From a practical perspective, it is necessary to consider the phosphorus (P) interaction and availability in the soil under nFe2O3 amendment, since the treatments substantially decreased P concentration in the soybean shoots, and thus a possible deficiency in plants under the critical pH value (below 6 and between 8–8.5) would be expected.

Abstract:Titanium dioxide nanoparticles (TiO2 NPs) are among the most commonly used nanomaterials and are most likely to end up in soil. Therefore, it is pertinent to study the interaction of TiO2 NPs with soil microorganisms. The present in vitro broth study evaluates the impacts of low‐dose treatments (0, 1.0, 5.0, 10.0, 20.0, and 40.0 mg L−1) of TiO2 NPs on cell viability, morphology, and plant growth promoting (PGP) traits of rhizobia isolated from mung bean root nodule. Two types of TiO2 NPs, that is, mixture of anatase and rutile, and anatase alone were used in the study. These TiO2 NPs were supplemented in broth along with a multifunctional isolate (Bradyrhizobium sp.) and two reference cultures. The exposure of TiO2 (anatase+rutile) NPs at low concentrations (less than 20.0 mg L−1) enhanced the cell growth, and total soluble protein content, besides improving the phosphate solubilization, Indole‐3‐acetic acid (IAA) production, siderophore, and gibberellic acid production. The TiO2 (anatase) NPs enhanced exopolysaccharide (EPS) production by the test rhizobial cultures. The radical scavenging assay was performed to reveal the mode of action of the nano‐TiO2 particles. The study revealed higher reactive oxygen species (ROS) generation by the TiO2 (anatase) NPs as compared with TiO2 (anatase+rutile) NPs. Exposure to TiO2 NPs also altered the morphology of rhizobial cells. The findings suggest that TiO2 NPs could act as promoters of PGP traits of PGP bacteria when applied at appropriate lower doses.

Abstract:Engineered nanoparticles (NPs) are considered potential agents for agriculture as fertilizers and growth enhancers. However, their action spectrum differs strongly, depending on the type of NP, its concentrations, and plant species per se, ranging from growth stimulation to toxicity. This work aimed to investigate effects of iron oxide (Fe3O4) NPs on growth, photosynthesis, respiration, antioxidant activity, and leaf mineral content of wheat plants. Wheat seeds were treated with NP for 3 h and plants were grown in the soil at two light intensities, 120 and 300 μmol (photons) m-2·s-1, followed by physiological assessment at several time points. High NP treatment (200 and 500 mg·L-1) enhanced plant growth, photosynthesis and respiration, as well as increasing the content of photosynthetic pigments in leaves. This effect depended on both the light intensity during plant growth and the age of the plants. Regardless of concentration and light intensity, an effect of NPs on the primary photochemical processes was not observed. Seed treatment with NP also led to increased activity of ascorbate peroxidase and reduced malondialdehyde (MDA) content in roots and leaves. Treatment with Fe3O4 also led to noticeable increases in the leaf Fe, P, and K content. It is concluded that iron oxide (Fe3O4)-based NP could enhance plant growth by improving photosynthetic performance and the availability of Fe and P.
Developmental and Reproductive Effects of Iron Oxide Nanoparticles in Arabidopsis thaliana

Abstract:Increasing use of iron oxide nanoparticles in medicine and environmental remediation has led to concerns regarding exposure of these nanoparticles to the public. However, limited studies are available to evaluate their effects on the environment, in particular on plants and food crops. Here, we investigated the effects of positive (PC) and negative (NC) charged iron oxide (Fe₂O₃) nanoparticles (IONPs) on the physiology and reproductive capacity of Arabidopsis thaliana at concentrations of 3 and 25 mg/L. The 3 mg/L treated plants did not show evident effects on seeding and root length. However, the 25 mg/L treatment resulted in reduced seedling (positive-20% and negative-3.6%) and root (positive-48% and negative-negligible) length. Interestingly, treatment with polyethylenimine (PEI; IONP-PC coating) also resulted in reduced root length (39%) but no change was observed with polyacrylic acid (PAA; IONP-NC coating) treatment alone. However, treatment with IONPs at 3 mg/L did lead to an almost 5% increase in aborted pollen, a 2%-6% reduction in pollen viability and up to an 11% reduction in seed yield depending on the number of treatments. Interestingly, the treated plants did not show any observable phenotypic changes in overall size or general plant structure, indicating that environmental nanoparticle contamination could go dangerously unnoticed.
CeO2 nanoparticle dose and exposure modulate soybean development and plant-mediated responses in root-associated bacterial communities

Abstract:Agricultural soils are increasingly undergoing inadvertent and purposeful exposures to engineered CeO 2 nanoparticles (NPs), which can impact crops and root-associated microbial communities. However, interactions between NP concentration and exposure duration on plant-mediated responses of root-associated bacterial communities are not well understood. Soybeans seedlings were grown in soil with uncoated NPs added at concentrations of 0, 1 or 100 mg kg −1 . Total soil exposure durations were either 190 days, starting 106 days before planting or 84 days with NP amendments coinciding with planting. We assessed plant development, bacterial diversity, differential abundance and inferred functional changes across rhizosphere, rhizoplane, and root tissue compartments. Plant non-monotonic dose responses were mirrored in bacterial communities. Most notably, effects were magnified in the rhizoplane under low-dose, short-exposures. Enriched metabolic pathways were primarily related to biosynthesis and degradation/utilization/assimilation, rather than responses to metals or oxidative stress. Our results indicate that plant-mediated bacterial responses were greater than direct NP impacts. Also, we identify needs for modeling non-monotonic legume stress responses that account for coinfection with mutualistic and parasitic bacteroids. Our findings provide new insights regarding effects of applications of soil amendments such as biosolids containing NPs or nano-enabled formulations used in cultivation of legumes and other crops.

Co-exposure Impact of Nickel Oxide Nanomaterials and Bacillus subtilis on Soybean Growth and Nitrogen Assimilation Dynamics

Abstract:Nickel oxide nanoparticles (NiO-NPs) pose potential threats to agricultural production. Bacillus subtilis has emerged as a stress-mitigating microbe that alleviates the phytotoxicity caused by NiO-NPs. However, the mechanisms underlying its effectiveness, particularly in root-nodule symbiosis and biological N2-fixation (BNF), remain unclear. Here, we tested the combined exposure of NiO-NPs (50 mg kg−1) and B. subtilis on soybean (Glycine max L.) growth and BNF. Combined exposure increased root length, shoot length, root biomass, and shoot biomass by 19-26%, while Ni (200 mg kg−1) reduced them by 38--53% compared to the control. NiO-NPs at 100 and 200 mg kg−1 significantly (P < 0.05) reduced nodule formation by 16% and 58% and Nitrogen assimilation enzyme activities levels (UE, NR, HS, and GOGAT) by 13-57%. However, co-exposure with B. subtilis improved nodule formation by 22-44%. Co-exposure of NiO-NPs (200 mg kg−1) with B. subtilis increased POD, CAT, and GSH-Px activity levels by 20%, 16%, and 14% while reducing MDA (14%) and H2O2 (12%) levels compared to NiO-NPs alone. Additionally, co-exposure of NiO-NPs (100 and 200 mg kg−1) with B. subtilis enhanced the relative abundance of Stenotrophomonas, Gemmatimonas, and B. subtilis, is associated with N2-cycling and N2-fixation potential. This study confirms that B. subtilis effectively mitigates NiO-NP toxicity in soybean, offering a sustainable method to enhance BNF and crop growth and contribute to addressing global food insecurity.

Abstract:Nanoscale agrochemicals have been widely used in sustainable agriculture and may potentially affect the nitrogen fixation process in legume crops. The present study investigated the size-effects of copper oxide nanoparticles (CuO NPs) on nitrogen assimilation in soybean (G. max (L.) Merrill) plants, which were treated with different sizes (20 and 50 nm) of CuO NPs at low use doses (1 and 10 mg/kg) for 21 days under greenhouse conditions. The results showed that 50 nm CuO NPs significantly increased the fresh biomass more than 20 nm CuO NPs achieved at 10 mg/kg. The activities of N assimilation-associated enzymes and the contents of nitrogenous compounds, including nitrates, proteins, and amino acids, in soybean tissues were greatly increased across all the CuO NP treatments. The use doses of two sizes of CuO NPs had no impact on the Cu contents in shoots and roots but indeed increased the Cu contents in soils in a dose-dependent fashion. Overall, our findings demonstrated that both 20 and 50 nm CuO NPs could positively alter soybean growth and boost N assimilation, furthering our understanding that the application of nanoscale micro-nutrient-related agrochemicals at an optimal size and dose will greatly contribute to increasing the yield and quality of crops.

Abstract:The wide use of metal-based nanoparticles (MNPs) will inevitably lead to their release into soil, and consequently affect the quality and ecological functions of soil environments. In this study, two paddy soils with different properties were exposed to CuO NPs to evaluate the transformation of CuO NPs and their effects on soil properties and components. The results of single chemical extraction and X-ray absorption fine structure analysis showed that CuO NPs could release Cu ions once being applied into the flooding paddy soil and then progress toward the more stable forms (Cu2S and Cu(OH)(2) ). CuO NPs could change the soil properties by increasing the pH and Eh of the lower organic matter-soil rather than those of the higher organic matter-soil. Furthermore, we found that the 1000 mg/kg CuO NPs could accelerate the degradation or mineralization of the organic matter, as well as the Fe reduction process, by increasing the Fe(II) content by 293% after flooding for 60 days in the lower organic matter soil. The microbial biomass in both soils was severely inhibited by CuO NPs and the organic matter could partly mitigate the negative effects of CuO NPs.

Abstract:Using plant growth-promoting bacteria (PGPB) as biopesticides and biofertilizers is becoming an increasingly popular sustainable practice. However, there are significant limitations associated with these practices, which can be minimized through nanotechnology. This study aims to evaluate the efficiency of iron oxide nanoparticles (IONPs) in improving the efficacy of PGPB in soybeans. We assessed the physiological and biochemical parameters in soybean seedlings treated with different combinations of PGPB and IONPs. Our results showed that adding IONPs to soybean seed improves seed vigor and early seedling development, suggesting a synergy between IONPs and PGPB. The seed vigor percentage in the treatment IONP 300 mg kg −1 + PGPB was 45% higher than that in IONP 300 mg kg −1 − PGPB. The addition of IONP also improve the root length in 98%. The absence of lipid peroxidation indicates no damage to the cell membrane with the addition of IONPs. Furthermore, the low modulation of the enzymatic antioxidant system highlights the non-oxidative pattern of the treatment. This synergy between IONPs, PGPB, and soybean plants could result from the action of iron in improving the physiological functions of both the plant and the bacteria.

Abstract:In this study, the phytotoxicity of seven metal oxide nanoparticles(NPs)—titanium dioxide (nTiO2), silicon dioxide (nSiO2), cerium dioxide (nCeO2), magnetite (nFe3O4), aluminum oxide (nAl2O3), zinc oxide (nZnO) and copper oxide (nCuO)—was assessed on two agriculturally significant crop plants (maize and rice). The results showed that seed germination was not affected by any of the seven metal oxide NPs. However, at the concentration of 2000 mg·L−1, the root elongation was significantly inhibited by nCuO (95.73% for maize and 97.28% for rice), nZnO (50.45% for maize and 66.75% for rice). On the contrary, minor phytotoxicity of nAl2O3 was only observed in maize, and no obvious toxic effects were found in the other four metal oxide NPs. By further study we found that the phytotoxic effects of nZnO, nAl2O3 and nCuO (25 to 2000 mg·L−1) were concentration dependent, and were not caused by the corresponding Cu2+, Zn2+ and Al3+ ions (0.11 mg·L−1, 1.27 mg·L−1 and 0.74 mg·L−1, respectively). Furthermore, ZnO NPs (<50 nm) showed greater toxicity than ZnO microparticles(MPs)(<5 μm) to root elongation of both maize and rice. Overall, this study provided valuable information for the application of engineered NPs in agriculture and the assessment of the potential environmental risks.

Abstract:In this study, we investigated the effects of citric acid (CA) coated copper oxide nanoparticles (CuO NPs) and their application method (foliar or soil exposure) on the growth and physiology of soybean (Glycine max). After nanomaterials exposure via foliar or soil application, Cu concentration was elevated in the roots, leaves, stem, pod, and seeds; distribution varied by plant organ and surface coating. Foliar application of CuO NPs at 300 mg/L and CuO-CA NPs at 75 mg/L increased soybean yield by 169.5% and 170.1%, respectively. In contrast, foliar and soil exposure to ionic Cu with all treatments (75 and 300 mg/L) had no impact on yield. Additionally, CuO-CA NPs at 300 mg/L significantly decreased Cu concentration in seeds by 46.7%, compared to control, and by 44.7%, compared to equivalent concentration of CuO NPs. Based on the total Cu concentration, CuO NPs appeared to be more accessible for plant uptake, compared to CuO-CA NPs, inducing a decrease in protein content by 56.3% and inhibiting plant height by 27.9% at 300 mg/kg under soil exposure. The translocation of Cu from leaf to root and from the root to leaf through the xylem was imaged by two-photon microscopy. The findings indicate that citric acid coating reduced CuO NPs toxicity in soybean, demonstrating that surface modification may change the toxic properties of NPs. This research provides direct evidence for the positive effects of CuO-CA NPs on soybean, including accumulation and in planta transfer of the particles, and provides important information when assessing the risk and the benefits of NP use in food safety and security.

The fate and impact of Co3O4 nanoparticles in the soil environment: Observing the dose effect of nanoparticles on soybeans

Abstract:The widespread presence and distribution of metal-based nanoparticles (NPs) in soil is threatening crop growth and food security. However, little is known about the fate of Co3O4 NPs in the soil-soybean system and their phytotoxicity. The study demonstrated the effects of Co3O4 NPs on soybean growth and yield in soil after 60 days and 140 days, and compared them with the phytotoxic effects of Co2+. The results showed that Co3O4 NPs (10-500 mg/kg) had no significant toxic effect on soybeans. Soil available Co content was significantly increased under 500 mg/kg Co3O4 NPs treatment. Compared with Co2+, Co3O4 NPs mainly accumulated in roots and had limited transport to the shoots, which was related to the particle size, surface charge and chemical stability of Co3O4 NPs. The significant accumulation of Co3O4 NPs in roots further led to a significant decrease in root antioxidant enzyme activity and changes in functional gene expression. Co3O4 NPs reduced soybean yield after 140 days, but interestingly, at specific doses, it increased grain nutrients (Fe content increased by 17.38% at 100 mg/kg, soluble protein and vitamin E increased by 14.34% and 16.81% at 10 mg/kg). Target hazard quotient (THQ) assessment results showed that consuming soybean seeds exposed to Co3O4 NPs (≥100 mg/kg) and Co2+ (≥10 mg/kg) would pose potential health risks. Generally, Co3O4 NPs could exist stably in the environment and had lower environmental risks than Co2+. These results help to better understand the environmental behavior and plant effect mechanisms of Co3O4 NPs in soil-plant systems.

The effect of ZnO, TiO2, and Fe2O3 NP on corn (Zea mays L.), common beans (Phaseolus vulgaris L.), nanobioremediation of polycyclic aromatic hydrocarbons (PAH), and soil organisms from agricultural or forest soils was studied at laboratory, greenhouse, and land level. The samples were analyzed by X-ray diffraction (XRD), field emission scanning electron microscopy with X-ray energy dispersion spectrometry (FESEM-EDS), scanning electron microscopy with energy-dispersive X-ray spectroscopy (SEM-EDS) gas chromatography (GC), ultra-performance liquid chromatography coupled to mass spectrometry (UPLC-MS) and Fourier transform infrared spectrometry with attenuated total reflectance (FTIR-ATR). ZnO-NP did not harm the mycorrhizal root colonization but, the presence of ZnO-NP decreased the degradation of PAH. The synthesis of metabolites from corn was more affected by the PAH than by ZnONP. FTIR spectra showed that NP affected the synthesis of compounds from specific functional groups in common bean plants. Fe2O3-NP were attached to the body of forestsoil organisms and significantly increased the concentration of Fe in their body, while TiO2-NP changed the morphological tissue of roots and stems of common bean as witnessed by micrographs of longitudinal and cross-sections. The NP used in this research significantly changed some response variables on the experiments carried-out at laboratory, greenhouse, and land level.
Physiological and metabolic responses of maize (Zea mays) plants to Fe3O4 nanoparticles

Abstract:Fe3O4 nanoparticles (NPs), as representative magnetic materials, have been widely used in the industrial and biomedical sectors, and their environmental impacts must be evaluated for their sustainable use. In this study, the interactions between Fe3O4 NPs and maize plants were investigated by a combination of phenotypic and metabolic approaches. Maize plants (Zea mays) were grown in soil treated with Fe3O4 NPs at 0, 50 and 500 mg/kg for 4 weeks. Fe3O4 NPs had no impact on plant biomass or photosynthesis. However, root length of maize plant significantly increased, with decreased malondialdehyde (MDA) level, indicating the positive effects on root development and membrane integrity. Inductively coupled plasma optical emission spectrometry (ICP-OES) revealed that Fe3O4 NPs resulted in a significant Fe accumulation in roots, instead of leaves. In addition, 500 mg/kg Fe3O4 NPs significantly promoted dehydrogenase enzyme activity by 84.9%. Metabolomics revealed that maize root metabolomes were re-programmed by Fe3O4 NPs exposure. Metabolic pathways associated with antioxidant and defence were inactivated by Fe3O4 NPs, indicating the protective role of Fe3O4 NPs for microbes and plant roots. Taken together, the results indicate a limited impact of environmental Fe3O4 NPs on plant growth. Taken together, the results of this study offer new insights into the molecular mechanisms by which maize responds to Fe3O4 NP exposure.

Abstract:The effect of the alpha 1 adrenoreceptor antagonist doxazosin on low density lipoprotein (LDL) receptor activity, has been studied in cultured skin fibroblasts. Doxazosin at a concentration of 10(-5) M significantly increased the LDL receptor activity. The possible clinical implications of this observation are discussed.

Abstract:The expansion of nanotechnology raises concerns about the consequences of nanomaterials in plants. Here, the effects of nanoparticles (NPs; 100–500 mg/kg) on processes related to micronutrient accumulation were evaluated in bean (Phaseolus vulgaris) exposed to CuO NPs, a mixture of CuO and ZnO (CuO:ZnO) NPs, and in CuO NP-exposed plants colonized by a root bacterium, Pseudomonas chlororaphis O6 (PcO6) in a sand matrix for 7 days. Depending on exposure levels, the inhibition of growth by CuO NPs was more apparent in roots (10–66 %) than shoots (9–25 %). In contrast, CuO:ZnO NPs or root colonization with PcO6 partially mitigated growth inhibition. At 500 mg/kg exposure, CuO NPs increased soluble Cu in the growth matrix by 23-fold, relative to the control, while CuO:ZnO NPs increased soluble Cu (26-fold), Zn (127-fold) and Ca (4.5-fold), but reduced levels of Fe (0.8-fold) and Mn (0.75-fold). Shoot accumulations of Cu (3.8-fold) and Na (1-fold) increased, while those of Fe (0.4-fold), Mn (0.2-fold), Zn (0.5-fold) and Ca (0.5-fold) were reduced with CuO NP (500 mg/kg) exposure. CuO:ZnO NPs also increased shoot Cu, Zn and Na levels, while decreasing that of Fe, Mn, Ca and Mg. Root colonization reduced shoot uptake of Cu and Na, 15 and 24 %, respectively. CuO NPs inhibited ferric reductase (up to 49 %) but stimulated cupric (up to 273 %) reductase activity; while CuO:ZnO NPs or root colonization by PcO6 altered levels of ferric, but not copper reductase activity, relative to CuO NPs. Cu ions at the level released from the NPs did not duplicate these effects. Our findings demonstrate that in addition to the apparent phytotoxic effects of NPs, NP exposure may also have subtle impacts on secondary processes such as metal nutrition.

Abstract:Titanium dioxide nanoparticles are used in different processes, derived from this their presence in wastewater is common, concentrating in residual sludge. These residues are used as agricultural soil improvers, being a source of crop exposure. In this study, the effect of TiO2-NPs (450, 900, and 1800 mg L-1) on the germination of alfalfa, tomato, and pepper seeds was evaluated. The germination parameters were not different (p > 0.05) except for, the seed vigor index in alfalfa and pepper, and the mean germination time in tomato. The germination index was below 80% in 450 mg L-1 in tomato, which showed moderate phytotoxicity. Morphological modifications with differences (p < 0.05) were found in the three crops, mainly in the root. In tomatoes, the length of the main root, root hairs, and the width of the root tip were reduced but increased the width of the main root, piliferous zone, and the length of root hairs were. For alfalfa, root length and number of secondary roots augment. However, the stem, root tip width, and root villi were reduced. Finally, for pepper, the length, and width of the root and the piliferous zone were modified. Additionally, a concerning trend has been observed in the length of root hairs. TiO2-NPs affected germination and morphology in alfalfa, tomato and pepper seeds differently. Tomato was most negatively affected, with reduced root length and width. Alfalfa showed mixed effects, with positive impacts on some parameters but negatives on others. Pepper seeds responded positively overall, with improved germination and root length, despite some impacts on root morphology.
