The value of RGB is carefully corrected and linked to the concentration of ASs with a standard deviation of <10% in the 10–1000 ppb (part per billion) range. In order to avoid the interference arising from inorganic anions (such as those found in tap water and groundwater), the water sample is pre-treated either by solid-phase extraction (SPE), which takes ∼30 min, or by dual liquid-phase extraction (dual-LPE, developed by us), which takes ∼5 min. In the latter case, the organic phase of the first LPE (equilibrium with water sample) is transferred and subjected to a second LPE (equilibrium with Milli-Q water) to remove any potential background interference. In the meantime, SPE can also pre-concentrate ASs at 100–1000 times (in volume) to benefit the sensitivity. Consequently, our smartphone app can detect PFOA spiked in tap/groundwater with an LOD of 10 ppb (∼12 nM, dual-LPE of ∼5 min), or 0.5 ppb (∼1.2 nM, SPE of ∼3 h), suggesting that it has the potential to succeed as a pre-screening tool for on-site application and in common laboratory tests.

Abstract:Accurate onsite profiling of fluoroquinolone antibiotics (FQs) is of vital significance for ensuring food safety and estimating environmental pollution. Here, we propose a smartphone-based QD ratiometric fluorescence-sensing system to precisely report the level of FQs. As a proof of concept, we chose gatifloxacin (GFLX, a typical member of FQs) as the model for the analytical target, which could effectively trigger the fluorescence color variation of QDs from bright yellow-green (∼557 nm) to blue (∼448 nm) through the photoinduced electron-transfer (PET) process, thus yielding an evident ratiometric response. Based on this, the level of GFLX can be reported within a wide linear range from 0.85 nM to 3.6 μM. Moreover, this assay owns a high sensitivity with a low detection limit of 0.26 nM for GFLX and a quick sample-to-answer monitoring time of 5.0 min, manifesting that this platform could be fully qualified for onsite requirements. Interestingly, this portable device has successfully been applied for the onsite detection of GFLX in real food (i.e., milk and drinking water) and environmental (i.e., fish-farming water) samples with acceptable results. This developed platform offers a great promise for the point-of-care detection of FQ residues in practical application with the merits of being label-free, low-cost, and rapid, thus opening a new pathway for the onsite evaluation of food safety and environmental health.

Abstract:A novel smartphone-assisted fluorescent sensor based on europium/zirconium metal-organic framework (Eu0.5/Zr0.5-MOF) was developed for the fast and sensitive determination of doxycycline (DOX) and L-arginine (Arg). After the addition of DOX, the fluorescence of Eu0.5/Zr0.5-MOF was quenched owing to the inner filter effect (IFE). When Arg was introduced into the Eu0.5/Zr0.5-MOF@DOX complex system, the fluorescence was recovered because the interaction between Arg and Eu0.5/Zr0.5-MOF@DOX weakened the IFE. Moreover, the Eu0.5/Zr0.5-MOF produced continuous fluorescence color changes for the visual measurement of DOX and Arg. The fluorescent probe for DOX and Arg offered broad linear ranges of 0.05-80 and 0.1-60 μg/mL, respectively, with detection limits as low as 2.07 and 67.5 ng/mL. The proposed method was successfully applied to monitor DOX in eggs and Arg in human serum. This work provides a powerful platform for the real-time and visual analysis of DOX and Arg in food and biological samples.

Abstract:In the last years, surface-enhanced Raman scattering based immunochromatographic assay (SERS-ICA) has been getting great attention and been applied for the detection of different target analytes. In this study, a novel SERS-ICA using Ag-MBA@SiO2-Ab as an immunoprobe has been developed for rapid, quantitative and ultrasensitive detection of a pharmaceutical diclofenac (DCF) residue in water samples. Ag, Ag@SiO2 and Ag-MBA@SiO2 nanoparticles (NPs) were synthesized and characterized. The immunoprobe was prepared by sandwiching the Raman reporter mercaptobenzoic acid (MBA) between the core-shell layers and immobilizing the monoclonal antibody (mAb) against DCF on the surface of the Ag@SiO2 NPs. Under optimal conditions, the sensitivity and limit of detection (LOD) of the SERS-ICA for DCF were found to be 9 pg mL(-1) and 0.07 pg mL(-1), respectively. The detection of the SERS-ICA for DCF was completed within 15 min. Ag-MBA@SiO2-Ab was stable within four months without significant loss of SERS intensity. The key to success of the SERS-ICA was the employment of the Ag@SiO2 NPs as the SERS substrate, which possessed gorgeous SERS enhancement capability, predominant biocompatibility and excellent stability. This SERS-ICA is potential as an ultrasensitive analytical method in point-of-care testing, clinical diagnosis, food inspecting and environmental monitoring.

Abstract:Cost-effective and user-friendly quantitation at points-of-need plays an important role in food safety inspection, environmental monitoring, and biomedical analysis. This study reports a stand-alone smartphone-based fluorospectrophotometer (the SBS) installed with a custom-designed application (the SBS-App) for on-site quantitation of pesticide using a ratiometric sensing scheme. The SBS can collect fluorescence emission spectra in the wavelength range of 380-760 nm within 5 s. A ratiometric fluorescence probe is facilely prepared by directly mixing the blue-emissive carbon nanodots (the Fe3+-specific fluorometric indicator) and red-emissive quantum dots (the internal standard) at a ratio of 11.6 (w/w). Based on the acetylcholinesterase/choline oxidase dual enzyme-mediated cascade catalytic reactions of Fe2+/Fe3+ transformation, a ratiometric fluorescence sensing scheme is developed. The practicability of the SBS is validated by on-site quantitation of chlorpyrifos in apple and cabbage with a comparable accuracy to the GC-MS method, offering a scalable solution to establish a cost-effective surveillance system for pesticide pollution.

Abstract:Immunoassays such as the enzyme-linked immunosorbent assay (ELISA) are utilized extensively for detecting protein biomarkers and small molecules in healthcare, environmental monitoring, and food analysis. Unfortunately, the current strategies for immunoassays often require sophisticated apparatus such as a microplate reader, which might not be available in resource-limited areas. To mitigate this problem, we designed a compact smartphone based-device and a multicolor response immunosensor. First, we designed a compact and cost-effective 3D-printed attachment, where a light-emitting diode was used as a light excitation source and a smartphone captured the fluorescent emission signals. Second, by combining quantum dots hybrid and chemical redox reaction, multiple color responses were displayed in the presence of the analyte at different concentrations. Third, solutions with distinct tonality could be readily distinguished by the naked eye and they were suitable for quantitative analysis using the hue-saturation-lightness color space based on a smartphone application. The versatility of the proposed sensing system was demonstrated by implementing an indirect competitive ELISA for analyzing trace drug residues in foodstuffs. The multicolor response of this sensing strategy allows us to visually quantify drug residues in foodstuffs. Moreover, the smartphone-based immunosensor can assess the exact concentration of the analyte by using a self-designed mobile application. The proposed assay provides a highly sensitive performance that the limit of detection was 0.37 ng/mL by visual detection and 0.057 ng/mL using the compact device. Due to its advantages in terms of portability, straightforward visual detection, high sensitivity, and cost effectiveness, the proposed immunosensor has great potential for applications in areas without access to laboratories or expensive infrastructure.

Abstract:Background: Carbon quantum dots (CQDs) have gained much interest recently for being efficient probes. Their cost-effectiveness, eco-friendliness, and unique photocatalytic activities made them distinctive alternatives to other luminescent approaches like fluorescent dyes and luminous derivatization. Meanwhile, delafloxacin (DLF) is a recently approved antibacterial medicine. DLF has been authorized for the treatment of soft-tissue and skin infections as well as pneumonia. Therefore, new eco-friendly, cost-effective, and sensitive tools are needed its estimation in different matrices. Results: In the proposed study, green copper and nitrogen carbon dots (Cu-N@CDs) were synthesized from a green source (plum juice with copper sulphate). Cu-N@CQDs were then characterized using multiple tools including X-ray photon spectroscopy (XPS), FTIR and UV-VIS spectroscopy, Zeta potential measurements, High-resolution transmission electron microscopy (HRTEM), and fluorescence spectroscopy. After gradually adding DLF, the developed quantum dots' fluorescence was significantly enhanced within the working range of 0.5-100.0 ng mL-1. The limits of detection and quantification were 0.08 and 0.27 ng mL-1, respectively. The accuracy of the proposed method ranged from 96.00 to 99.12 % in recovery%, when recovered from milk and plasma samples. Significance: Cu-N@CDs were utilized and validated for selectively determining DLF in several matrices including pharmaceutical forms, human plasma and in milk samples using spectrofluorimetric technique. The bio-analytical method is simple and could be used in content uniformity testing as well as in therapeutic drug monitoring in human plasma.
Nanozyme-mediated ratiometric fluorescence hydrogel for on-site detection of sulfite in food

Abstract:In this work, a ratiometric fluorescence hydrogel nanosensor was developed by integrating a composite consisting of o-phenylenediamine (OPD), manganese dioxide nanoflakes (MnO2 NFs), and N-doped carbon dots (N-CDs) into an agarose hydrogel for sulfite detection. MnO2 NFs demonstrated intense oxidase-like activity, facilitating the conversion of non-fluorescent OPD into yellow-emissive 2,3-diaminophenazine (DAP). As a result, a significant emission peak belongs to DAP, alongside the fluorescence quenching of N-CDs through FRET. Upon interaction with sulfite, MnO2 NFs lost their oxidase-like function. This process decreased the fluorescence of DAP and restored the blue fluorescence of N-CDs, producing a typical ratiometric response, ranging from 3 nM ∼ 400 μM, with a detection limit (LOD) of 3.79 nM. Employing a smartphone, the fluorescence color change demonstrated by the hydrogel sensor was translated into quantitative data (LOD: 8.44 nM). This hydrogel sensor offers an affordable, portable, and user-friendly solution for sulfite detection and food safety monitoring.

Abstract:Exploring a rapid, sensitive, low-cost, in-situ intelligent monitoring multi-target fluorescence detection platform is important for food safety and environmental monitoring. A dual-mode ratiometric fluorescence sensing system integrated with a smartphone based on a luminescent metal-organic framework (NH 2 -MIL-53) and CdTe/Eu was developed for visual, in-situ analysis of Fe 3+ and doxycycline (DOX) in this paper. Interestingly, with increasing Fe 3+ concentration, the fluorescence sensing system exhibits dual-emission with CdTe QDs at 540 nM as the response signal and NH 2 -MIL-53 at 438 nm as the reference signal, resulting in a significant color shift of fluorescence color from blue-green to blue, with a linear range of 5- 1550 nM and a detection limit of 1.08 nM. In the presence of DOX, the blue fluorescence of NH 2 -MIL-53 and the green fluorescence of CdTe QDs were quenched respectively by the internal filtering effect and the photoelectron transfer effect. While DOX enhances the red fluorescence of Eu 3+ by the antenna effect, forming a triple-emission fluorescence sensor. The visual color of this fluorescent sensor shifted from blue green to grey to pink-white to pink to fuchsia to red as the DOX concentration increased with a detection limit of 0.11 nM. Furthermore, the developed intelligent sensing platform achieved real-time in-situ detection of Fe 3+ and DOX with detection limit of 1.47 nM and 6.43 nM, respectively. The platform was applied to detection actual samples with satisfactory results, which proved a promising application for real-time on-site food safety monitoring and human health monitoring.

Abstract:Food sources are susceptible to contamination with ochratoxin A (OTA), which is a serious threat to human health. Thus, the construction of novel, simple sensing platforms for OTA monitoring is of utmost need. Manganese-doped lead halide perovskite quantum dots encapsulated with mesoporous SiO 2 (Mn-CsPbBr 3 QDs@SiO 2 ) were prepared here and used as a ratiometric fluorescent probe for OTA. Mn-CsPbBr 3 QDs, synthesized at room temperature, exhibit dual emission with maximum wavelengths of 440 and 570 nm and, when embedded in the SiO 2 layer, produce a stable and robust photoluminescence signal. By adding OTA to the probe, emission at 440 nm increases while emission at 570 nm decreases, so a ratiometric response is obtained. Experimental variables affecting the probe signal were studied and optimized and the mechanism of sensing was discussed. This ratiometric sensor demonstrated excellent selectivity and low detection limit (4.1 ng/ml) as well as a wide linear range from 5.0 to 250 ng/ml for OTA. A simple portable smartphone-based device was also constructed and applied for the fluorescence assay. With different OTA concentrations, the multicolor transition from pink to blue under a UV lamp led to simple visual and smartphone-assisted sensing of OTA by using a color analyzing application. Satisfactory recoveries in black tea, coffee, moldy fig and flour samples confirmed the reliability of the assay. The accuracy of the probe was proved by comparison of the results with high-performance liquid chromatography (HPLC).

Abstract:A fluorescent probe was fabricated for the determination of sparfloxacin. The nanocomposite probe was constructed by incorporating nitrogen-doped graphene quantum dots, zinc oxide decorated carbon foam and magnetite into a matrix formed from a molecularly imprinted polymer. The fabricated nanoprobe was characterized and the fabrication and determination conditions were optimized to obtain the highest sensitivity. The determination of sparfloxacin was based on the decreasing of emission response of the nanocomposite probe when the target analyte rebinds to the inherent recognition sites of the fluorescent probe. Under the best conditions, the quenching of fluorescence was linear in the range of 0.10–100.0 μg L −1 (R 2 = 0.991) with the limit of detection being 0.10 μg L −1 . The nanocomposite probe exhibited high specificity with an imprinting factor of 7.93. This optosensor was used to determine sparfloxacin in milk and good recoveries were observed ranging from 93.0 % to 100.8 % with RSDs lower than 5 %. The results also agreed with the results obtained using a chromatographic method but the developed method has higher sensitivity, at lower cost and faster determination procedure. In addition, the developed optosensor can be simply modified for highly selective determination of other trace levels of target compounds by changing the template with minor changes in determination conditions.

Sorption-Fluorimetric Determination of Quinolones in Waste and Natural Waters with a Smartphone

Abstract:A simple, affordable, and safe (green chemistry) method is proposed for determining 17 quinolone antibiotics in waste and natural waters by the sorption-fluorimetric method using a smartphone as an analyte signal recorder. The method is based on static sorption of quinolones by silica gel from aqueous solutions. When the sorbate is irradiated with ultraviolet light (365 nm), blue or turquoise fluorescence is observed, the intensity of which is measured with a smartphone. The values of the basic components of the RGB colorimetric system are used as an analytical signal, followed by the calculation of the final color. The limits of detection and quantification are 0.0007–0.004 and 0.002–0.01 μg/mL, respectively, for all analyzed analytes. The range of the determined contents is 0.002 (0.01)–0.2 μg/mL. A procedure for the determination of quinolones in waste and natural waters is proposed. The relative standard deviation of the analysis results does not exceed 0.2.

Abstract:The massive use of cyfluthrin pesticide in agriculture has brought growing concerns on the food safety due to its chemical stability, easy-to-accumulation, and several harmful effects on human health. Design of a rapid, reliable and portable analytical method for on-site monitoring cyfluthrin is of great significance. Although reported assays showed high sensitivity and selectivity to cyfluthrin, these intensity-based outputs suffered from the interference from a variety of analyte-independent factors, i.e. the instrumental parameters, which often led to the poor accuracy and repeatability of detection results. In this work, we reported a novel host-guest supramolecular probe by using albumin as the host and solvatofluorochromic flavonoid as the guest, for the first time, successfully realizing ratiometric determination of cyfluthrin. This probe was capable of detecting cyfluthrin with the fast response (10 s), high sensitivity (LOD similar to 70 ppb) and selectivity, good anti-interference performance, and remarkably distinct emission color change from orange to green. Moreover, the portable paper-based test strips could be facilely obtained and utilized for ratiometric determination cyfluthrin pesticide in food samples, such as apple, tomato, cucumber. The signal outputs could be captured and analyzed by using a smartphone, demonstrating the application potential of this probe in on-site food analysis. We expected this work may open up a novel avenue for on-site determination of pesticides with the simple testing apparatuses.

Abstract:A simple dual-colour fluorescent nanoprobe has been designed composed of blue and yellow emission carbon quantum dots (CQDs). This system is inexpensive and easy to operate and was successfully employed for on-site measurements based on a smartphone app. The designed nanoprobe exhibited increased selectivity for Cr(VI), leading to a double stable response of the two CQDs. The dual-emission nanoprobe showed blue-violet fluorescence upon UV irradiation, and the fluorescent emission peaks were located at 418 nm and 552 nm. The blue light emission of CQDs was quenched with increasing Cr(VI) concentration due to the inner filter effect, whereas the yellow light emission was enhanced due to the aggregation-induced emission effect. The different responses of the dual emissions to Cr(VI) resulted in a fluorescent colour variation, thus enabling facile macroscopic visualization. With a smartphone, the change in the fluorescence colour could be observed more apparently than that of a single fluorescence nanoprobe, and the response increased linearly so that the nanoprobe could be applied to instantaneous measurements. Furthermore, the dual-emission nanoprobe was successfully employed for analysing food and water samples. Accurate concentrations were obtained by constructing a calibration plot using a fluorescence spectrometer and a smartphone app; the recoveries were 81.6% to 107.7%, and the relative standard deviation was below 3.6%. Therefore, this smartphone-integrated dual-emission detection system is promising as a new portable method for the on-site measurement of Cr(VI) ions.

Abstract:A simple smartphone-integrated ratiometric fluorescent sensing system for visual determination of copper ions (Cu2+) and fluoroquinolones (FQs) was developed based on carbon dots (CDs) which were synthesized through the high-temperature pyrolysis of citric acid. In this system, with the fluorescence resonance energy transfer effect between CDs and 2,3-diaminophenazine (oxOPD), the detection of Cu2+ and ciprofloxacin (CIP, an example for FQs) was realized. Cu2+ catalyzes the oxidation of OPD to form oxOPD with yellow fluorescence, resulting in the quenching of CDs. In addition, CIP can inhibit the catalytic activity of Cu2+ and induce the recovery of CDs fluorescence. Under the excitation of 400 nm, the changes of CDs fluorescence at 472 nm and oxOPD fluorescence at 556 nm were monitored. The detection results showed that the sensing system exhibited good selectivity and sensitivity to Cu2+ and CIP with the limit of detection of 2.32 × 10−8 mol L−1 and 0.2 ng mL−1, respectively. In addition, a smartphone was developed as a portable analyzer to capture the change of fluorescence color and quickly analyze the concentration of Cu2+ and CIP. The proposed smartphone-based sensing platform has satisfactory sensitivity, and it has application prospects for detecting Cu2+ and FQs in food safety monitoring.

Abstract:A sensitive, miniaturized, and low-cost method combining pipette tip solid-phase microextraction and smartphone-based fluorescent detection has been developed for the determination of total sulfonamides in water samples. Sulfonamides antibiotics (SAs) are contaminants commonly found in water matrices, leading to antibiotic-resistant bacteria and risks to human health and the environment. Thus, its real-time monitoring is essential to the risk assessment, and the subsequent management of any water supply. Sample preparation consisted of extraction/preconcentration of SAs using graphene nanoplatelets packed inside a pipette tip, followed by fluorescent derivatization using fluorescamine inside the microplate reader, both 3D printed. Subsequently, a 3D printed detection platform that houses monochromatic LED strips as radiation source and a smartphone as detector have been used for determination total SAs. Digital image processing was based on the RGB colour model using image J software with its Readplate plugin and the green intensity channel was used as analytical signal due to its higher sensitivity. Several factors that affect the extraction efficiency and detection have been optimized. Under the optimized conditions, good linearity for SAs studied were obtained in a range of 10–60 μg L −1 with r ≥ 0.990 and limits of detection between 2.5–3.1 μg L −1 for a sample volume of 10 mL. The recoveries of sulfamethoxazole (as a model compound to express total SAs) spiked in diverse water matrices were tested at two different levels showed good recoveries from 94% to 102% with RSD ≤ 7.6%. The results obtained with the proposed method were satisfactorily compared with those obtained with a conventional spectrofluorometer (P ≥ 0.13). This easy-to-operate system features a simple extraction procedure (up to 20-fold enrichment), excellent sensitivity and precision, which is very useful and practical for on-site analysis. Furthermore, three greenness evaluation methodologies (GAPI, AGREE, and AGREEprep) were used to assess the environmental friendliness of the proposed method, demonstrating its superior performance compared to previously published HPLC and spectrophotometric methods.

Abstract:Folic acid (FA) is a vitamin essential for human physiology; thus, rapid, accurate, and sensitive methods for its detection and quantification in different biological samples are necessary. A fluorescent ratiometric nanoprobe (the complex composed of quantum dots and carbon dots) based on carboxyl-modified quantum dots (QDs) and amino-modified graphene-based carbon dots (CDs) has been developed for the visual determination of folic acid (FA). The nanoprobe is the composition in which QDs were covalently bonded with CDs. Red and blue fluorescence of nanoprobe can be selectively quenched and enhanced by FA, respectively. Thus, the nanoprobe is capable to generate clearly visible fluorescence color change from pink to blue upon exposure to UV light. Depending on the color change and its intensity, FA can be identified, and their concentrations can be determined. The sensor prepared based on the nanoprobe demonstrated good selectivity and sensitivity toward FA with exceptionally low detection limit equal to 0.039 μM. We also prepared a paper-based sensor to demonstrate convenient and simple approach to visually detect FA. Thus, the nanoprobe demonstrated feasibility of rapid real-time quantitative FA detection; considering its simple, effective, and practicality, expectedly it can be used for practical application.

Abstract:Measurement of fluorescence emission for quantitative analysis is always based on the traditional spectrofluorometer, which limits the onsite detection approach. Thus, an alternative device should be developed for fulfilling the analysis outside the laboratory. Therefore, a low-cost, portable, and low-energy consumption fluorescence reader-based smartphone device was developed. A UV-LED was used to construct the fluorescence device-based smartphone as a low-power excitation light source. The smartphone camera was used as a detector for detecting photons from the fluorescence emission process of the fluorescence probe and connected to a digital image platform. A transparent acrylic with orange and yellow colors was demonstrated as a filter for reducing the interference from the light source intensity. The obtained digital image was converted to RGB intensity by a custom-designed smartphone application. The N,S-doped carbon nanodots (N,S-CDs) were introduced to be a good fluorescence indicator for determining trace quantities of Mn2+ in cosmetics. The approach demonstrated high selectivity and sensitivity, detecting and quantifying analytes at 1-5 μM concentrations. Furthermore, the method's detection limit of 0.5 μM reflects its capacity to detect trace amounts of the target analyte. The Mn2+ in cosmetic products was successfully analyzed by this device with high accuracy comparable with the results from ICP-OES.

Abstract:The UIO‐66⊃FL/RhB‐based hydrogel beads are constructed for visual recognition of fluoroquinolone (FQs), and a portable smartphone‐based sensing platform is built for the sensitive detection of ofloxacin. The UIO‐66⊃FL/RhB establishes a promising strategy for the simple and accurate discrimination of FQs. Development of new multi‐ratiometric fluorescence probes has been a popular research field. This approach provides richer and more accurate information for the recognition of different substances which have similar chemical and physical properties. In this work, a ratiometric fluorescence sensor UIO‐66⊃FL/RhB is successfully constructed by incorporating fluorescein (FL) and rhodamine B (RhB) to the UIO‐66. With the addition of fluoroquinolones (FQs), the prepared UIO‐66⊃FL/RhB sensor can be used for the 2D emission‐fingerprint map, the linear discriminant analysis (LDA) and hierarchical clustering analysis (HCA) in recognition the presence of each FQs. The fluorescence probe offers a broad linear range (0‐8 μM) and a low detection limit (3.7 nM) in sensing ofloxacin. In addition, a smartphone‐assisted portable sensing platform and a portable UIO‐66⊃FL/RhB hydrogel test material are constructed successfully for screening before fluorescence detection. This study proposes a novel multi‐ratiometric fluorescence probe, which can recognize different FQs. Besides, the UIO‐66⊃FL/RhB‐based hydrogel beads are construction for screening before the detection of FQs, which established a promising strategy for the simple and accurate discrimination of FQs.
