Abstract:Herein, a stable and sensitive label-free electrochemical immunosensor for detecting α-fetoprotein (AFP) was proposed based on gold nanoparticles-poly(3,4-ethylenedioxythiophene)/Prussian blue-reduced graphene oxide (AuNPs-PEDOT/PB-rGO) nanocomposite as sensing substrate. PB was used as the electron transfer mediator to provide the electrochemical signal. By utilizing rGO as the supporting material for PB, the stability and intensity of the redox signal were greatly improved. The incorporation of highly conductive AuNPs-PEDOT can further enhance the electrochemical signal and its stability. Electrochemical test showed the anodic current of AuNPs-PEDOT/PB-rGO modified electrode could remain up to 89.45% after 100 cyclic voltammetries. The assembly of PB-rGO and AuNPs-PEDOT exhibited 3D nanostructure and showed high specific surface area, which could effectively capture the antibody of AFP. With the advantages of AuNPs-PEDOT/PB-rGO nanocomposite, the prepared immunosensor showed a sensitive response to AFP in a linear range from 0.01 to 50ngmL−1 with a low detection limit of 3.3pgmL−1 (S/N=3). The proposed immunosensor also has been employed to determine AFP in human serum with satisfactory results. These excellent properties make the AuNPs-PEDOT/PB-rGO nanocomposite an ideal interface for efficient biomarkers detection.

Abstract:Early finding and diagnosis are critical for prevention and treatment of hepatocellular carcinoma (HCC). Alpha-fetoprotein (AFP) is a typical biomarker of HCC. Since AFP level can reflect the severity of HCC, it is essential to ensure the accurate detection of AFP. In this study, through a combination of the advantages exhibited by ordered mesoporous carbon (OMC)@gold nanoparticles (AuNPs) composites and AuPt-methylene blue (AuPt-MB), a disposable ultrasensitive sandwich-configuration electrochemical immunosensor for determination of AFP was designed. Characterized by excellent conductivity, highly ordered pore distribution and great surface area, OMC can be effective in promoting electron transfer and loading a large number of AuNPs. In the meantime, AuNPs can also immobilize AFP-Ab1 through Au-N bonds. As a new redox-active species, rod-like AuPt-MB demonstrates high conductivity, uniform morphology and excellent biocompatibility, which makes it capable not only to fix AFP-Ab2, but also to release electrochemical signals. A wide linearity of 10 fg mL-1-100 ng mL-1 and a low detection limit of 3.33 fg mL-1 (S/N=3) were obtained. Moreover, the proposed immunosensor exhibited acceptable selectivity, high stability and reproducibility. The excellent performance in detecting serum samples endows the proposed immunosensor with broad prospects of extensive application in the detection of disease biomarkers.

The fabrication of phosphotungstate@UIO-Au/reduced graphene oxidation for electrochemical ultrasensitive detection of alpha-fetoprotein

Abstract:As an early multi-purpose tumor marker of hepatocellular carcinoma, alpha-fetoprotein (AFP) plays a vital role in early diagnosis and treatment. To achieve the early and accurate determination of AFP, the POMOF was fabricated by embedding H3PW12O40 (PW12) into UIO-66-NH2, further immobilized on reduced GO (rGO) and fabricated an innovative POMOF nanocomposite (PW@UIO-Au/rGO) as an electrochemical immunosensor (ECI-sensor). The PW@UIO-Au/rGO achieved 17-fold signal enhancement owing to their synergistic effect, enabling PW@UIO-Au/rGO exhibit high oxidase-like catalytic activity, facilitating their sensing performance. Under optimal experimental conditions, the proposed PW@UIO-Au/rGO ECI-sensor presented excellent sensing performance over a wide range from 0.01 ng mL-1 to 500 ng mL-1 with ultra-low detection of 4.0 pg mL-1. Notably, sensing results in real serum samples were verified by the clinical enzyme-linked immunosorbent assay (ELISA) and electrochemiluminescence immunoassay (ECL) methods with excellent accuracy and consistency, indicating the excellent environmental tolerance of the proposed ECI-sensor. This work provided a promising strategy for designing feasible ultra-sensitive probe for sensing AFP in clinical test.

Abstract:Alpha-fetoprotein (AFP) has become a specific tumor marker of primary liver cancer in clinical diagnosis. In this work, we prepared worm-like platinum (WL Pt) nanomaterial via chemical etching without organic solvents and ultra-high temperature. Due to its small particle size and the formation of surface vacancies during the etching process, it had a large specific surface area, and thus exhibited superior electrocatalytic activity for the reduction of hydrogen peroxide. Combining the signal amplification based on hydrogen peroxide reduction and the specific recognition of antigen with antibody, we constructed a simple label-free electrochemical immunosensor with a sandwich-like structure. The developed electrochemical immunosensor showed a wide linear range (0.0001−100 ng mL−1), a low detection limit (0.028 pg mL−1), good selectivity and stability. Further, the immunosensor was comparable with enzyme-linked immunosorbent assay (ELISA) and had a good accuracy for AFP detection in human serum samples proving the feasibility of potential application, which is expect to become one of the most promising method in early diagnosis and clinical analysis for liver cancer.

Abstract:A nonenzymatic electrochemical immunosensor based on palladium nanoparticles conjugated reduced graphene nanosheets (Pd-GS) for sensitive detection of cancer biomarker alpha-fetoprotein (AFP) is described. Primary antibody-AFP (Ab(1)) was immobilized onto the surface of reduced graphene nanosheets (rGO) through an amidation reaction between the carboxylic acid group of the rGO and the available amine groups of Ab(1). Pd-GS which was prepared by one-spot synthesis is employed to immobilize secondary antibody (Ab(2)). The resulting Pd-GS-Ab(2) conjugate was used as a label for the immunosensor to detect AFP. The amplified immunoassay exhibits high sensitivity, wide linear rang (0.01-10 ng/mL), low detection limit (3.0 pg/mL), acceptable stability and reproducibility. And such immunosensor also shows good recovery in the assay results for AFP in human serum samples.

Abstract:The labeling-free and immobilization-free homogeneous aptamer sensor offers advantages including simple operation, low cost, and high sensitivity, demonstrating great potential in rapid detection of tumor biomarkers in biological samples. In this work, a labeling-free and immobilization-free homogeneous aptamer sensor was conveniently fabricated by combining size exclusion and charge-selective penetration of a nanochannel-modified electrode and two-dimensional (2D) nanorecognition probe which can realize selective and highly sensitive detection of alpha-fetoprotein (AFP) in serum. Vertically ordered mesoporous silica film (VMSF) with ultra-small, uniform, and vertically aligned nanochannels was easily grown on the simple, low-cost, and disposable indium tin oxide (ITO) electrode. Through π-π interaction and electrostatic force, the AFP aptamer (Apt) and electrochemical probe, tris(bipyridine)ruthenium(II) (Ru(bpy)32+), were coloaded onto graphene oxide (GO) through simple incubation, forming a 2D nanoscale recognition probe (Ru(bpy)32+/Apt@GO). Owing to the size exclusion effect of VMSF towards the 2D nanoscale probe, the electrochemical signal of Ru(bpy)32+/Apt@GO could not be detected. In the presence of AFP, the specific binding of AFP to the aptamer causes the dissociation of the aptamer and Ru(bpy)32+ from GO, resulting in their presence in the solution. The efficient electrostatic enrichment towards Ru(bpy)32+ by negatively charged VMSF allows for high electrochemical signals of free Ru(bpy)32+ in the solution. Linear determination of AFP ranged from 1 pg/mL to 1000 ng/mL and could be obtained with a low limit of detection (LOD, 0.8 pg/mL). The high specificity of the adapter endowed the constructed sensor with high selectivity. The fabricated probe can be applied in direct determination of AFP in serum.

Abstract:Sporozoite-induced experimental infections of Haemoproteus meleagridis produced a moderate to severe myositis and significant effects on weight gain and growth in domestic turkey poults. Pathological effects occurred in both low- and high-dose infections (4,400 and 57,500 sporozoites, respectively). Low-dose birds weighed significantly less than controls at 3 wk postinfection (PI) when peripheral parasitemia reached a peak and had significantly shorter tarsometatarsal lengths at both 1 and 3 wk PI. High-dose birds were significantly lighter and smaller than control and low-dose birds throughout the course of the 8-wk study. Infected birds were not anemic in spite of high parasitemias that often exceeded 50% of circulating erythrocytes. The most serious pathological effects occurred prior to patency and were associated with development of megaloschizonts in skeletal muscle. Microscopic lesions in 4 high-dose birds that died between 19 and 22 days PI were characteristic of a severe, acute hemorrhagic myositis. Megaloschizonts were surrounded by a hemorrhagic inflammatory infiltrate composed of macrophages, heterophils, giant cells, and red blood cells. Muscle fibers adjacent to megaloschizonts were swollen, hyaline, and contained prominent calcium deposits. Other observations included enlargement of the spleen, deposition of pigment in macrophages of the lung and spleen, and secondary bacterial and fungal infections in the intestine and lungs. Necrotic and calcified muscle fibers and degenerating megaloschizonts were still present at 8 wk PI when the experiment ended. Our results demonstrated significant pathological changes in H. meleagridis-infected domestic turkeys that were associated primarily with preerythrocytic stages of development.

Abstract:In this work, a label-free electrochemical immunosensor was structured for detecting alpha fetal protein (AFP) effectively. The proposed electrochemical immunosensor was structured by gold silver platinum nanodendrites loaded on amino-functionalized MoO 2 nanosheets (AuAgPt NDs/NH 2 -MoO 2 NSs). The AuAgPt NDs with rich dendrite structure provided abundant specific surface area for effective capture antibody. MoO 2 possessed the satisfactory catalytic ability in terms of H 2 O 2 reduction and charge transfer capacity. AuAgPt NDs possessed favourable catalytic performance to H 2 O 2 reduction. Therefore, the prepared AuAgPt NDs/NH 2 -MoO 2 NSs was better than AuAgPt NDs and NH 2 -MoO 2 NSs in H 2 O 2 reduction on account of a synergistic effect. The constructed electrochemical immunosensor possessed a satisfying detection limit of 3.3 fg ml −1 and a broad detection range which was from 10 fg ml −1 to 100 ng ml −1 (S/N = 3) for the AFP detection under optimal conditions. The constructed electrochemical immunosensor possessed favourable detection performance in sensitivity, stability, selectivity and reproducibility, which indicated that it possessed a practical application in the aspect of clinical detection.

Abstract:Abstract A label-free electrochemical aptamer-based sensor has been fabricated for alpha-fetoprotein (AFP) detection. Platinum nanoparticles on carboxylated-graphene oxide (PtNPs/GO-COOH) modified screen-printed graphene-carbon paste electrode (SPGE) was utilized as an immobilization platform, and the AFP aptamer was employed as a bio-recognition element. The synthesized GO-COOH helps to increase the surface area and amounts of the immobilized aptamer. Subsequently, PtNPs are decorated on GO-COOH to enhance electrical conductivity and an oxidation current of the hydroquinone electrochemical probe. The aptamer selectively interacts with AFP, causing a decrease in the peak current of the hydroquinone because the binding biomolecules on the electrode surface hinder the electron transfer of the redox probe. Effects of aptamer concentration and AFP incubation time were studied, and the current changes of the redox probe before and after AFP binding were investigated by square wave voltammetry. The developed aptasensor provides a linear range from 3.0–30 ng mL −1 with a detection limit of 1.22 ng mL −1 . Moreover, the aptamer immobilized electrode offers high selectivity to AFP molecules, good stability, and sensitive determination of AFP in human serum samples with high recoveries.

Abstract:A novel amperometric enzyme immunosensor with amplified sensitivity for the determination of alpha-fetoprotein (AFP) was constructed with layer-by-layer assembly of ZnSe quantum dots (ZnSe QDs)/Azure I/gold nanoparticles (nanoAu)/poly (3,4-ethylenedioxythiophene) (PEDOT) on Pt electrode. Firstly, citrate coated nanoAu was immobilized on the PEDOT polymer film, which was electrochemically synthesized in ionic liquid electrolyte of 1-butyl-3-methylimidazolium tetrafluoroborate ([bmim][BF4]). Then, Azure I was immobilized on nanoAu/PEDOT composite matrix as a redox probe and was used to immobilize ZnSe QDs. Subsequently, AFP antibody (anti-AFP) was adsorbed onto the surface of ZnSe layer. Finally, horseradish peroxidase (HRP) was employed to block sites against nonspecific binding and amplify the current signal of the antigen–antibody reaction. The modification processes were characterized by cyclic voltammetry, scanning electron microscopy. The factors influenced the performances of the proposed immunosensors were studied in detail. Because of the synergism between Azure I and nanoAu/PEDOT to facilitate electron-transfer process, and the small diameter of ZnSe QDs favorable for stabilization of biological activity to a large extent, the immunosensor displayed a high sensitivity, fast analytical time, a relatively low detection limit of 1.1fg/mL at 3 times of signal-to-noise ratio (S/N=3), and a especially broad linear response to AFP in a ranges from 5×10−5 to 250ng/mL. Moreover, the selectivity, repeatability, and stability of the proposed immunosensor were acceptable.

Abstract:A novel electrochemical immunoassay based on nanobody heptamer for ultrasensitive detection of alpha fetoprotein (AFP) is proposed. First, AFP heptamer fusion protein (A1-C4bp α) is prepared via fusing the AFP-specific nanobody (A1) with the C-terminal fragment of C4-binding protein (C4bp α). Then A1-C4bp α heptamer with higher affinity is used as the molecule recognizing probe to capture numerous the AFP, increasing the sensitivity of the immunosensor. Moreover, AuNPs decorated zeolitic imidazolate framework () with large surface area, and abundant binding sites serves an ideal nanocarrier to increase heptamer loading. Multiwalled carbon nanotube (MWCNTs) with excellent electrical conductivity was immobilized on glassy carbon electrode (GCE) to enhance electrochemical signals. The coupling between the @MWCNTs and heptamer technologies constructed an ultrasensitive and specific immunosensor for AFP detection in the range of 0.1 to 105 pg mL−1 with a lower detection limit of 0.033 pg mL−1. The as-prepared electrochemical sensing exhibited excellent performance and could be employed to determine AFP with satisfactory results in actual serum samples, suggesting that the proposed strategy for nanobody multimerization provides an alternative scheme for enhancing sensitivity in important biomarkers detection.

Abstract:Accurate detection of tumor biomarkers in blood is crucial for diagnosing and treating tumor disease. In this study, a metal enzyme-linked immunosorbent assay (MeLISA) was fabricated for the ultrasensitive and naked-eye detection of tumor biomarker alpha-fetoprotein (AFP) in clinical serum samples. Herein, novel copper metal-organic frameworks and gold platinum nanoparticles composites (Cu-MOFs@AuPtNPs) were synthesized for the first time by in-situ method, which showed an enormous specific surface area and excellent peroxidase (POx) mimicking property. Cu-MOFs@AuPtNPs linked with antibodies targeting AFP performed as signal nanoprobe to amplify the detection signal. Additionally, the specificity of MeLISA was significantly enhanced through conventional antigen-antibody reaction and efficient blocking of non-specific sites with BSA. In optimal conditions, the sandwich-type MeLISA exhibited a wide range from 0.001 to 1000 ng mL-1 (R2 = 0.997) and a low detection limit of 0.86 pg mL-1 (S/N = 3) with acceptable stability, selectivity, and reproducibility. It was noteworthy that the suggested MeLISA performed exceptionally well in detecting clinical serum samples, which were visible to the naked eye and did not require complex platforms. To sum up, the innovative MeLISA based on Cu-MOFs@AuPtNPs provides an alternative method for early cancer diagnosis, particularly in economically backward areas where simple diagnostic apparatus is extremely desirable.

Abstract:In this study, we demonstrated the highly sensitive detection of alpha-fetoprotein (AFP) by electrochemiluminescence (ECL) and electrochemistry (EC) based on the gated transport of the bifunctional probe (tris(1,10-phenanthroline) ruthenium (II) chloride, Ru (phen) 3 Cl 2 ) into the nanochannels of vertically ordered mesoporous silica films (VMSFs). Due to the negatively charged surface and ultrasmall pore size, VMSF displays a signal amplification effect on Ru (phen) 3 Cl 2 and is suitable for the construction of sensors with excellent sensitivity. With the linkage of (3-glycidyloxypropyl) trimethoxysilane, the anti-AFP antibody could covalently bind to the external surface of VMSF, generating a highly specific recognized sensing interface toward AFP. When AFP is presented, the formed immunocomplex hinders the diffusion of Ru (phen) 3 Cl 2 to the underlying electrode surface, resulting in a decreased ECL or EC response. The dual-mode detection of AFP is achieved with a relatively low limit of detection (0.56 fg/ml for ECL and 4.5 pg/ml for EC) and a wide linear range (10 fg/ml∼1 μg/ml for ECL and 10 pg/ml∼1 μg/ml for EC). Moreover, owing to the inherent anti-fouling property of VMSF, satisfactory results in the analysis of human serum were obtained, showing the great potential of the designed strategy in clinical diagnosis.

Abstract:The conventional electrochemical detection strategy for alpha-fetoprotein (AFP) is limited by the antigen-antibody (Ag-Ab) reactions and suffers from low sensitivity and poor reproducibility due to the inconsistency of Ab-modified electrodes. Herein, we designed and explored a sandwich-type electrochemical sensor for highly sensitive detection of AFP based on aptamer (Apt)-AFP-Ab interaction mode with silver@gold (Ag@Au) core-shell nanoparticles (NPs) as a signal amplifier. AuNPs were electrodeposited onto MXene (Ti3C2TX)-modified glassy carbon electrode (GCE) to get AuNPs/MXene/GCE and further used as the signal amplification substrate. The tetrahedral DNA-linked AFP aptamers were immobilized onto AuNPs/MXene/GCE surface via Au-S bonds and used as the sensing and recognition platform for AFP capturing. Ag@AuNPs with core-shell structures were synthesized, characterized, and bound with Ab as detection elements by catalyzing H2O2 reduction. In the presence of AFP, a stable Apt-AFP-Ab sandwich structure was formed owing to the high affinities of aptamer and Ab toward the target AFP. The catalytic current produced by H2O2 reduction increased linearly with the logarithm of AFP concentration from 5 × 10-4 ng/mL to 1 × 105 ng/mL, accompanied by a low detection limit (1.6 × 10-4 ng/mL). Moreover, the novel sandwich-type electrochemical sensor shows high sensitivity, outstanding selectivity, and promising performance in the analysis of actual samples, displaying a broad application prospect in bioanalysis.

Abstract:A label-free method for the determination of alpha-fetoprotein (AFP) was successfully developed by graphene oxide (GO)-based electrochemical aptasensor. This aptasensor was constructed by covalently immobilizing NH2functionalized AFP-specific aptamer on GO with plenty of carboxylic groups. Cyclic voltammetry (CV) and electrochemical impedance spectra (EIS) analysis were carried out to investigate the practicability of the fabrication procedures. Fourier transform infrared spectra (FTIR), Raman, atomic force microscopy (AFM) and scanning electron microscope (SEM) were performed to indicate the changes of the sensing interface. CV was used to detect the signal change of the aptasensor. Peak current of CVs changed before and after incubating the aptasensor with different concentration of AFP solution. The changes of peak current were proportional to the AFP concentration, with a wide linear range of 0.01-100 ng mL(-1), a low detection limit of 3 pg(-1) and good specificity. The proposed simple and cost-effective label-free strategy is promising for the determination of clinical biomarkers such as AFP.

Abstract:This study devised a label-free electrochemical immunosensor for the quantitative detection of alpha-fetoprotein (AFP). 3-Polythiophene acetic acid (3-PTAA) nanoparticles were anchored onto a few-layer graphene (FLG) nanosheet, and the resulting nanocomposite was utilized as the immunosensor platform. The AFP antibody (anti-AFP) was immobilized on 3-PTAA@FLG via a covalent interaction between the amine group of anti-AFP and the carboxylic group of 3-PTAA via ethyl-3-(3-dimethyl aminopropyl) carbodiimide (EDC)/N-hydroxysuccinimide (NHS) coupling. FLG is largely responsible for providing electrochemical signals, whereas 3-PTAA nanoparticles are well-known for their ability to be compatible with biological molecules in neutral aqueous solutions. Moreover, the carboxyl group present in 3-PTAA effectively binds anti-AFP through EDC/NHS conjugation. Owing to good dispersibility and higher surface area of 3-PTAA, it is very convenient for casting the polymer directly on the electrode substrate followed by immobilization of anti-AFP. Thus, it is feasible to regulate the activity of AFP proteins and control the spatial distribution of the immobilized anti-AFP proteins. The electrochemical sensing performance was assessed via cyclic voltammetry and electrochemical impedance spectroscopy. For an increase in the bioconjugate concentration, the results demonstrated a surge in charge-transfer resistance and a consequent decline in the current response. This approach effectively detected AFP at an extended dynamic range of 0.0001-250 ng/mL with a detection limit of 0.047 pg/mL. Furthermore, the sensing capacity of the immunosensor for AFP detection has been demonstrated to be steady in real human serum cultures. Our approach exhibits good electrochemical performance in terms of reproducibility, selectivity, and stability, which would surely impart budding applications in the clinical diagnosis of several other tumor markers.

Abstract:The detection of alpha-fetoprotein-L3 (AFP-L3) is of great importance for hepatocellular carcinoma (HCC) diagnosis, but remains unsatisfactory owing to poor sensitivity and complex operating steps. In this work, a simple and sensitive method has been proposed for the detection of AFP-L3. Firstly, biotinylated Lens culinaris agglutinin-integrated silver nanoparticles (B-LCA-AgNPs) is fabricated. Owing to the specific interaction between Lens culinaris agglutinin and AFP-L3, AFP-L3 can be detected directly through the electrochemical signal readout of AgNPs, avoiding separated steps used in clinical practice. Furthermore, after the recognition between B-LCA-AgNPs and AFP-L3, large amount of AgNPs can be gathered at the binding site through avidin-biotin interactions, which can amplify the signal. Therefore, detection of AFP-L3 can be sensitively achieved. Moreover, compared with the other approaches, this new method has a better linear correlation (25–15,000pg/mL) and a lower detection limit (12pg/mL). Also, the new method developed in this work has been demonstrated to have good stability and reproducibility for the assay of AFP-L3 in human serum samples, so, it may hold a great application prospect in clinical diagnostics.

Abstract:The study of luminol electrochemiluminescence (ECL) is commonly conducted in alkaline solutions. However, many substances, especially biomolecules, are only stable in neutral or weakly acidic media. In this study, a sensitive ECL immunosensor was developed using polypyrrole-luminol (PPY-Lu) nanoparticles for the determination of alpha-fetoprotein (AFP) in a neutral buffer. Polypyrrole (PPY) nanoparticles were synthesized by chemical oxidation. Subsequently, luminol was attached to the PPY nanoparticles via electrostatic interaction, resulting in the formation of the desired nanoparticles which exhibited strong and stable electrochemiluminescence in neutral solution. Furthermore, the PPY-Lu nanoparticles have abundant amino groups suitable for immobilizing biomolecules. An ECL immunosensor for AFP was fabricated by immobilizing anti-AFP on the PPY-Lu-modified electrode surface. The electrochemiluminescence decreased when AFP was captured. The immunosensor exhibited a linear range from 1.0‚ÄČ√ó‚ÄČ10 ‚ąí11 to 1.0‚ÄČ√ó‚ÄČ10 ‚ąí6‚ÄČ mg/mL with a low detection limit of 8.5‚ÄČ√ó‚ÄČ10 ‚ąí12‚ÄČ mg/mL and excellent specificity, reproducibility, and long-term stability. This designed immunosensor provides a new method for the determination of biomarkers in clinical analysis.
