Abstract:Herein, a stable and sensitive label-free electrochemical immunosensor for detecting α-fetoprotein (AFP) was proposed based on gold nanoparticles-poly(3,4-ethylenedioxythiophene)/Prussian blue-reduced graphene oxide (AuNPs-PEDOT/PB-rGO) nanocomposite as sensing substrate. PB was used as the electron transfer mediator to provide the electrochemical signal. By utilizing rGO as the supporting material for PB, the stability and intensity of the redox signal were greatly improved. The incorporation of highly conductive AuNPs-PEDOT can further enhance the electrochemical signal and its stability. Electrochemical test showed the anodic current of AuNPs-PEDOT/PB-rGO modified electrode could remain up to 89.45% after 100 cyclic voltammetries. The assembly of PB-rGO and AuNPs-PEDOT exhibited 3D nanostructure and showed high specific surface area, which could effectively capture the antibody of AFP. With the advantages of AuNPs-PEDOT/PB-rGO nanocomposite, the prepared immunosensor showed a sensitive response to AFP in a linear range from 0.01 to 50ngmL−1 with a low detection limit of 3.3pgmL−1 (S/N=3). The proposed immunosensor also has been employed to determine AFP in human serum with satisfactory results. These excellent properties make the AuNPs-PEDOT/PB-rGO nanocomposite an ideal interface for efficient biomarkers detection.

Abstract:A renewable and magnetic electrochemiluminescence (ECL) immunosensor for alpha-fetoprotein (AFP) was fabricated by using RuL-MWNTs (Tris(2,2'-bipyridine) ruthenium(II) complex immobilized on multiwall carbon nanotubes through pi-pi stacking function between the carbon aromatic systems) doped Au (RuL-MWNTs@Au) composite as labels. Polyelectrolyte poly(dimethyldiallylammonium chloride) (PDDA) was employed to covered on the surfaces of RuL-MWNTs which made it have positive electric charge and can absorb negative colloid Au nanoparticles to fabricate RuL-MWNTs@Au through electric attraction. The primary antibody, anti-AFP was first immobilized on the Fe3O4(core)/gold(shell) nanoparticles (Fe3O4@Au, GMP) as probes. Then the antigen and the RuL-MWNTs@Au composite nanoparticles labeled Ru(bpy)(3)(2+)-conjuged secondary antibody (RuL-MWNTs@Au similar to RuLAb2) were combined successively to form RuL-MWNTs@Au similar to RuLAb2/AFP/anti-AFP-GMP sandwich-type immunocomplexes through the specific interaction. The resulting magnetic sandwich immune complexes can be absorbed on the planar screen printed carbon electrode (SPCE) due to the external magnetic field added on the bottom of it. The magnetic sandwich complexes can be easily washed away from the surface of SPCE when the magnetic field was removed which made the immunosensor reusable. The prepared RuL-MWNTs@Au composite nanoparticles own large surface area, good biocompatibility and highly effective ECL properties. The morphologies of the RuL-MWNTs@Au and its antibody labeled composite nanoparticles were characterized by transmission electronic microscope (TEM). The sensor performed high sensitivity and wide liner for detection AFP in the range of 0.01-50 ng.mL(-1) and the limit detection was 3 pg.mL(-1) (defined as S/N=3). Furthermore the sandwich immunocomplexes can be performed the rapid separation from the unbound secondary antibody quickly by magnetic field. So the immunosensor can simultaneously realize separation, enrichment and determination, with high sensitivity, which would be valuable for clinical immunoassay for AFP in human serum.

Abstract:A signal amplification strategy based on a Ru(bpy) 3 2+ luminescence functionalized metal-organic framework (MOF) and double coreaction accelerators was developed for the detection of alpha-fetoprotein (AFP) by electrochemiluminescence (ECL). The high porosity and rich functional groups of MOF can improve the binding rate of biomolecules. Therefore, Ru(bpy) 3 2+ was encapsulated by the three-dimensional hierarchical porous MOF TMU-13 as the luminescence nanoreactor. The complex exhibits stable and excellent luminescence efficiency and rich carboxyl functional groups, which can support enough antibodies. In addition, the process of K 2 S 2 O 8 reduction was promoted by double coreaction accelerators, including MoS 2 grown in CoFe LDOs in situ and in situ reduction of gold nanoparticle (AuNPs) nanoflower structures (AuNPs/CoFe LDO/MoS 2 NFs), the ECL signal in the cathode was significantly increased. Under optimal conditions, the established AFP detection method exhibited a wide linear range of 10 −5 ng·mL −1 to 100 ng·mL −1 , and the detection limit was as low as 3.23 fg·mL −1 . Importantly, this strategy shows great potential for application in the field of bioanalysis.

Abstract:In this study, we demonstrated the highly sensitive detection of alpha-fetoprotein (AFP) by electrochemiluminescence (ECL) and electrochemistry (EC) based on the gated transport of the bifunctional probe (tris(1,10-phenanthroline) ruthenium (II) chloride, Ru (phen) 3 Cl 2 ) into the nanochannels of vertically ordered mesoporous silica films (VMSFs). Due to the negatively charged surface and ultrasmall pore size, VMSF displays a signal amplification effect on Ru (phen) 3 Cl 2 and is suitable for the construction of sensors with excellent sensitivity. With the linkage of (3-glycidyloxypropyl) trimethoxysilane, the anti-AFP antibody could covalently bind to the external surface of VMSF, generating a highly specific recognized sensing interface toward AFP. When AFP is presented, the formed immunocomplex hinders the diffusion of Ru (phen) 3 Cl 2 to the underlying electrode surface, resulting in a decreased ECL or EC response. The dual-mode detection of AFP is achieved with a relatively low limit of detection (0.56 fg/ml for ECL and 4.5 pg/ml for EC) and a wide linear range (10 fg/ml∼1 μg/ml for ECL and 10 pg/ml∼1 μg/ml for EC). Moreover, owing to the inherent anti-fouling property of VMSF, satisfactory results in the analysis of human serum were obtained, showing the great potential of the designed strategy in clinical diagnosis.

Abstract:A novel amperometric enzyme immunosensor with amplified sensitivity for the determination of alpha-fetoprotein (AFP) was constructed with layer-by-layer assembly of ZnSe quantum dots (ZnSe QDs)/Azure I/gold nanoparticles (nanoAu)/poly (3,4-ethylenedioxythiophene) (PEDOT) on Pt electrode. Firstly, citrate coated nanoAu was immobilized on the PEDOT polymer film, which was electrochemically synthesized in ionic liquid electrolyte of 1-butyl-3-methylimidazolium tetrafluoroborate ([bmim][BF4]). Then, Azure I was immobilized on nanoAu/PEDOT composite matrix as a redox probe and was used to immobilize ZnSe QDs. Subsequently, AFP antibody (anti-AFP) was adsorbed onto the surface of ZnSe layer. Finally, horseradish peroxidase (HRP) was employed to block sites against nonspecific binding and amplify the current signal of the antigen–antibody reaction. The modification processes were characterized by cyclic voltammetry, scanning electron microscopy. The factors influenced the performances of the proposed immunosensors were studied in detail. Because of the synergism between Azure I and nanoAu/PEDOT to facilitate electron-transfer process, and the small diameter of ZnSe QDs favorable for stabilization of biological activity to a large extent, the immunosensor displayed a high sensitivity, fast analytical time, a relatively low detection limit of 1.1fg/mL at 3 times of signal-to-noise ratio (S/N=3), and a especially broad linear response to AFP in a ranges from 5×10−5 to 250ng/mL. Moreover, the selectivity, repeatability, and stability of the proposed immunosensor were acceptable.

Abstract:The study of luminol electrochemiluminescence (ECL) is commonly conducted in alkaline solutions. However, many substances, especially biomolecules, are only stable in neutral or weakly acidic media. In this study, a sensitive ECL immunosensor was developed using polypyrrole-luminol (PPY-Lu) nanoparticles for the determination of alpha-fetoprotein (AFP) in a neutral buffer. Polypyrrole (PPY) nanoparticles were synthesized by chemical oxidation. Subsequently, luminol was attached to the PPY nanoparticles via electrostatic interaction, resulting in the formation of the desired nanoparticles which exhibited strong and stable electrochemiluminescence in neutral solution. Furthermore, the PPY-Lu nanoparticles have abundant amino groups suitable for immobilizing biomolecules. An ECL immunosensor for AFP was fabricated by immobilizing anti-AFP on the PPY-Lu-modified electrode surface. The electrochemiluminescence decreased when AFP was captured. The immunosensor exhibited a linear range from 1.0‚ÄČ√ó‚ÄČ10 ‚ąí11 to 1.0‚ÄČ√ó‚ÄČ10 ‚ąí6‚ÄČ mg/mL with a low detection limit of 8.5‚ÄČ√ó‚ÄČ10 ‚ąí12‚ÄČ mg/mL and excellent specificity, reproducibility, and long-term stability. This designed immunosensor provides a new method for the determination of biomarkers in clinical analysis.

Abstract:An ultrasensitive multiplexed electrochemiluminescence immunoassay method was developed for the detection of tumor markers by combining a functionalized graphene nanosheets and gold-coated magnetic Fe3O4 nanoparticles (GMPs) labeled alpha-fetoprotein (AFP) antibody (GMP similar to Ab1). The functionalized graphene nanosheets (PDDA-G) with poly (diallyldimethylammonium chloride) (PDDA) were synthesized and used to combine with luminol-capped gold nanoparticles (Lu-Au NPs). The resulting PDDA-G@Lu-Au composite displayed an enhanced capability for the immobilization of horseradish peroxidase (HRP) and signal antibody (Ab2) to realize its electrochemiluminescence immunoassay. Great signal amplification was achieved since PDDA-G could adsorb large amount of reporter molecules (Lu-Au). Besides gold nanoparticles were not only used as carriers of Ab2 and HRP but also catalyzed the ECL reaction of luminol, which further amplified the ECL signal of luminol in the presence of H2O2. GMPs as supporting material, not only been performed the rapid separation and purification of signal antibody on magnetic field, but also enhanced the fixed capacity of Ab1 to improve detection range. In addition, the magnetic probes were readily immobilized on the working electrode of screen-printed carbon electrode (SPCEs) by magnets. Under the optimized conditions, the ECL method shows a linear range of alpha-fetoprotein (AFP) from 0.002 to 20 ng mL(-1) and an extremely low detection limit of 0.2 pg mL(-1) (3 sigma). Moreover, the proposed method is also simple, stable, specific, and time-saving, avoiding the complicated process modification on the electrodes' surface, which may open a new door to ultrasensitive detection of tumor markers in clinical analysis.

Abstract:highly sensitive electrochemiluminescence immunosensor was originally proposed based on lead sulfide nanocrystals (PbS NCs) to detect alpha fetoprotein (AFP). Herein, PbS NCs, as a kind of nanoenzyme, was synthesized and applied to the luminol-hydrogen peroxide (L-H2O2) system for the first time. Additionally, PbS NCs with superior electrocatalytic activity toward H2O2 can enhance the electrochemiluminescence response of luminol. In this protocol, chitosan was utilized to immobilize PbS NCs through its film-forming characteristics, and further developed an ECL immunosensor by amination reaction combined with AFP antibodies. When the modified electrode was incubated with different concentrations of AFP antigen, the electrochemiluminescence intensity decreased obviously. In addition, under optimal conditions, the sensor can sensitively and selectively detect the alpha fetoprotein ranging from 1.0x10(-5) to 800 ng/mL with the detection limit of 3.0 fg/mL. On this basis, the proposed ECL immunosensor will open up a new way to sensitively detect AFP and exhibit its application prospect in biological analysis.

Abstract:This work demonstrates an effective quenching electrochemiluminescent (ECL) immunosensor based on resonance energy transfer for the sensitive detection of alpha fetoprotein (AFP). In this strategy, graphitic carbon nitride (g-C3N4) was coupled with bismuth molybdenum oxide (Bi2MoO6) to form a g-C3N4/Bi2MoO6 nanocomposite as a novel type of ECL immunosensor. The as-synthesized amino-modified g-C3N4/Bi2MoO6 nanocomposite presents strong and stable cathodic ECL activity compared to pristine g-C3N4. One plausible reason is that the synergistic effect between the g-C3N4 and Bi2MoO6 could facilitate charge transfer process and thereby enhancing the separation efficiency of electron-hole pairs. The other functional part of the immunosensor, carboxyl CoS2 nanoboxes with a broad absorption range, was rationally designed and introduced. The evidence that the absorption spectra of carboxyl CoS2 NBs overlap with ECL spectra of g-C3N4/Bi2MoO6 nanocomposite holds accountable for exceptionally weakened ECL signal. This sandwich-type immunosensor was setup based on quenching mechanism concerning amino-modified g-C3N4/Bi2MoO6 as an ECL donor and carboxyl CoS2 NBs as an ECL accepter. The strategy was optimized to achieve a convincible and sensitive detection goal for AFP with a wide quantifiable range of 0.5 pg/mL-10 ng/mL whilst a sufficiently low detection limit of 0.04 pg/mL (S/N = 3). This immunosensor shows great potential for real sample analysis with reasonable recoveries ranging from 95.5 to 99.0 %, demonstrating its high precision for AFP determination.

Abstract:A novel and effective electrochemical immunosensor for the rapid determination of α-fetoprotein (AFP) based on carbon paste electrode (CPE) consisting of room temperature ionic liquid (RTIL) N-butylpyridinium hexafluorophosphate (BPPF6) and graphite. The surface of the CPE was modified with gold nanoparticles for the immobilization of the α-fetoprotein antibody (anti-AFP). By sandwiching the antigen between anti-AFP on the CPE modified with gold nanoparticles and the secondary antibody, polyclonal anti-human-AFP labeled with horseradish peroxidase (HRP-labeled anti-AFP), the immunoassay was established. The concentration of AFP was determined based on differential pulse voltammetry (DPV) signal, which was generated in the reaction between O-aminophenol (OAP) and H2O2 catalyzed by HRP labeled on the sandwich immunosensor. AFP concentration could be measured in a linear range of 0.50–80.00ngmL−1 with a detection limit of 0.25ngmL−1. The immunosensor exhibited high sensitivity and good stability, and would be valuable for clinical assay of AFP.

Abstract:A novel electrochemical immunoassay based on nanobody heptamer for ultrasensitive detection of alpha fetoprotein (AFP) is proposed. First, AFP heptamer fusion protein (A1-C4bp α) is prepared via fusing the AFP-specific nanobody (A1) with the C-terminal fragment of C4-binding protein (C4bp α). Then A1-C4bp α heptamer with higher affinity is used as the molecule recognizing probe to capture numerous the AFP, increasing the sensitivity of the immunosensor. Moreover, AuNPs decorated zeolitic imidazolate framework () with large surface area, and abundant binding sites serves an ideal nanocarrier to increase heptamer loading. Multiwalled carbon nanotube (MWCNTs) with excellent electrical conductivity was immobilized on glassy carbon electrode (GCE) to enhance electrochemical signals. The coupling between the @MWCNTs and heptamer technologies constructed an ultrasensitive and specific immunosensor for AFP detection in the range of 0.1 to 105 pg mL−1 with a lower detection limit of 0.033 pg mL−1. The as-prepared electrochemical sensing exhibited excellent performance and could be employed to determine AFP with satisfactory results in actual serum samples, suggesting that the proposed strategy for nanobody multimerization provides an alternative scheme for enhancing sensitivity in important biomarkers detection.

Abstract:An ultrasensitive and renewable electrochemiluminescence (ECL) immunosensor was developed for the detection of tumor markers by combining a newly designed trace tag and streptavidin-coated magnetic particles (SCMPs). The trace tag (RuL@SiO2-Au similar to RuL-Ab2) was prepared by loading Ru(bpy)(3)(2+)(RuL)-conjuged secondary antibodies (RuL-Ab2) on RuL@SiO2 (RuL-doped SiO2) doped Au (RuL@ SiO2-Au). To fabricate the immunosensor, SCMPs were mixed with biotinylated AFP primary antibody (Biotin-Ab1), AFP, and RuL@SiO2-Au similar to RuL-Ab2 complexes, then the resulting SCMP/Biotin-Ab1/AFP/RuL@SiO2-Au similar to RuL-Ab2 (SBAR) sandwich-type immunocomplexes were absorbed on screen printed carbon electrode (SPCE) for detection. The immunocomplexes can be easily washed away from the surface of the SPCE when the magnetic field was removed, which made the immunosensor reusable. The present immunosensor showed a wide linear range of 0.05-100 ng mL(-1) for detecting AFP, with a low detection limit of 0.02 ng mL(-1) (defined as S/N = 3). The method takes advantage of three properties of the immunosensor: firstly, the RuL@SiO2-Au similar to RuL-Ab2 composite exhibited dual amplification since SiO2 could load large amount of reporter molecules (RuL) for signal amplification. Gold particles could provide a large active surface to load more reporter molecules (RuL-Ab2). Accordingly, through the ECL response of RuL and tripropylamine (TPA), a strong ECL signal was obtained and an amplification analysis of protein interaction was achieved. Secondly, the sensor is renewable because the sandwich-type immunocomplexes can be readily absorbed or removed on the SPCE's surface in a magnetic field. Thirdly, the SCMP modified probes can perform the rapid separation and purification of signal antibodies in a magnetic field. Thus, the present immunosensor can simultaneously realize separation, enrichment and determination. It showed potential application for the detection of AFP in human sera.

Abstract:The conventional electrochemical detection strategy for alpha-fetoprotein (AFP) is limited by the antigen-antibody (Ag-Ab) reactions and suffers from low sensitivity and poor reproducibility due to the inconsistency of Ab-modified electrodes. Herein, we designed and explored a sandwich-type electrochemical sensor for highly sensitive detection of AFP based on aptamer (Apt)-AFP-Ab interaction mode with silver@gold (Ag@Au) core-shell nanoparticles (NPs) as a signal amplifier. AuNPs were electrodeposited onto MXene (Ti3C2TX)-modified glassy carbon electrode (GCE) to get AuNPs/MXene/GCE and further used as the signal amplification substrate. The tetrahedral DNA-linked AFP aptamers were immobilized onto AuNPs/MXene/GCE surface via Au-S bonds and used as the sensing and recognition platform for AFP capturing. Ag@AuNPs with core-shell structures were synthesized, characterized, and bound with Ab as detection elements by catalyzing H2O2 reduction. In the presence of AFP, a stable Apt-AFP-Ab sandwich structure was formed owing to the high affinities of aptamer and Ab toward the target AFP. The catalytic current produced by H2O2 reduction increased linearly with the logarithm of AFP concentration from 5 × 10-4 ng/mL to 1 × 105 ng/mL, accompanied by a low detection limit (1.6 × 10-4 ng/mL). Moreover, the novel sandwich-type electrochemical sensor shows high sensitivity, outstanding selectivity, and promising performance in the analysis of actual samples, displaying a broad application prospect in bioanalysis.
Multifunctional Nanolabels Based on Polydopamine Nanospheres for Sensitive Alpha Fetoprotein Electrochemical Detection

Abstract:Multifunctional nanolabels with both facile signal amplification and high selectivity are essential for the sensitive determination of the vital tumor marker alpha fetoprotein (AFP). Here, we prepared an immunosensing nanolabel (Ab2-PDA-AF) for convenient AFP detection by grafting the detection antibody (Ab2) and the electroactive redox probe aminoferrocene (AF) onto polydopamine (PDA) nanospheres through Michael addition and Schiff base reactions. The electrochemical signal is dually amplified by the Ab2-PDA-AF nanolabels when the nanolabels are selectively bound to the electrode. The electrochemical signal can be first amplified by oxidizing AF of the Ab2-PDA-AF on the electrode. The signal can be further amplified by the redox reaction of PDA nanospheres with the linked AF/AF+ on the nanospheres. More importantly, the integration of AF-PDA-Ab2 favors a quick electron transfer between AF and PDA nanospheres. Redox probes of AF/AF+ are bonded to the nanolabels instead of dispersing in an electrolyte solution, thus decreasing the electron-transfer resistance and increasing the current signal. The interfacial kinetic analysis demonstrates that the charge-transfer resistance is decreased by 80% by using Ab2-PDA-AF nanolabels. The sensor displays a low detection limit of ca. 0.01 pg/mL, as well as excellent selectivity, reproducibility, and stability. The linear calibration plot of the AFP immunosensor has been obtained in a range of 0.5–1000.0 pg/mL. The capability of the immunosensor has been verified by implementing the detection of AFP in human serum samples.

Abstract:A nonenzymatic electrochemical immunosensor based on palladium nanoparticles conjugated reduced graphene nanosheets (Pd-GS) for sensitive detection of cancer biomarker alpha-fetoprotein (AFP) is described. Primary antibody-AFP (Ab(1)) was immobilized onto the surface of reduced graphene nanosheets (rGO) through an amidation reaction between the carboxylic acid group of the rGO and the available amine groups of Ab(1). Pd-GS which was prepared by one-spot synthesis is employed to immobilize secondary antibody (Ab(2)). The resulting Pd-GS-Ab(2) conjugate was used as a label for the immunosensor to detect AFP. The amplified immunoassay exhibits high sensitivity, wide linear rang (0.01-10 ng/mL), low detection limit (3.0 pg/mL), acceptable stability and reproducibility. And such immunosensor also shows good recovery in the assay results for AFP in human serum samples.

Abstract:The labeling-free and immobilization-free homogeneous aptamer sensor offers advantages including simple operation, low cost, and high sensitivity, demonstrating great potential in rapid detection of tumor biomarkers in biological samples. In this work, a labeling-free and immobilization-free homogeneous aptamer sensor was conveniently fabricated by combining size exclusion and charge-selective penetration of a nanochannel-modified electrode and two-dimensional (2D) nanorecognition probe which can realize selective and highly sensitive detection of alpha-fetoprotein (AFP) in serum. Vertically ordered mesoporous silica film (VMSF) with ultra-small, uniform, and vertically aligned nanochannels was easily grown on the simple, low-cost, and disposable indium tin oxide (ITO) electrode. Through π-π interaction and electrostatic force, the AFP aptamer (Apt) and electrochemical probe, tris(bipyridine)ruthenium(II) (Ru(bpy)32+), were coloaded onto graphene oxide (GO) through simple incubation, forming a 2D nanoscale recognition probe (Ru(bpy)32+/Apt@GO). Owing to the size exclusion effect of VMSF towards the 2D nanoscale probe, the electrochemical signal of Ru(bpy)32+/Apt@GO could not be detected. In the presence of AFP, the specific binding of AFP to the aptamer causes the dissociation of the aptamer and Ru(bpy)32+ from GO, resulting in their presence in the solution. The efficient electrostatic enrichment towards Ru(bpy)32+ by negatively charged VMSF allows for high electrochemical signals of free Ru(bpy)32+ in the solution. Linear determination of AFP ranged from 1 pg/mL to 1000 ng/mL and could be obtained with a low limit of detection (LOD, 0.8 pg/mL). The high specificity of the adapter endowed the constructed sensor with high selectivity. The fabricated probe can be applied in direct determination of AFP in serum.

Abstract:An ultrasensitive and renewable electrochemiluminescence (ECL) immunosensor was developed for the detection of tumor markers by combining a newly designed trace tag and streptavidin-coated magnetic particles (SCMPs). The trace tag (RuL@SiO2-Au~RuL-Ab2) was prepared by loading Ru(bpy)32+(RuL)-conjuged secondary antibodies (RuL-Ab2) on RuL@SiO2 (RuL-doped SiO2) doped Au (RuL@SiO2-Au). To fabricate the immunosensor, SCMPs were mixed with biotinylated AFP primary antibody (Biotin-Ab1), AFP, and RuL@SiO2-Au~RuL-Ab2 complexes, then the resulting SCMP/Biotin-Ab1/AFP/RuL@SiO2-Au~RuL-Ab2 (SBAR) sandwich-type immunocomplexes were absorbed on screen printed carbon electrode (SPCE) for detection. The immunocomplexes can be easily washed away from the surface of the SPCE when the magnetic field was removed, which made the immunosensor reusable. The present immunosensor showed a wide linear range of 0.05–100 ng mL–1 for detecting AFP, with a low detection limit of 0.02 ng mL–1 (defined as S/N = 3). The method takes advantage of three properties of the immunosensor: firstly, the RuL@SiO2-Au~RuL-Ab2 composite exhibited dual amplification since SiO2 could load large amount of reporter molecules (RuL) for signal amplification. Gold particles could provide a large active surface to load more reporter molecules (RuL-Ab2). Accordingly, through the ECL response of RuL and tripropylamine (TPA), a strong ECL signal was obtained and an amplification analysis of protein interaction was achieved. Secondly, the sensor is renewable because the sandwich-type immunocomplexes can be readily absorbed or removed on the SPCE’s surface in a magnetic field. Thirdly, the SCMP modified probes can perform the rapid separation and purification of signal antibodies in a magnetic field. Thus, the present immunosensor can simultaneously realize separation, enrichment and determination. It showed potential application for the detection of AFP in human sera.

Abstract:Herein, an aptamer-luminol modified magnetic graphene oxide and copper-based MOF composite was prepared and used to build a novel target-triggered "turn on" chemiluminescence (CL) sensor for alpha-fetoprotein (AFP) detection. Magnetic graphene oxide (MGO) was functionalized with the complementary sequence of the AFP aptamer (cDNA), and then MGO-cDNA was linked to aptamer modified luminol (Apt-luminol) through the complementary base pairing effect. The functionalized magnetic graphene oxide (MGO-cDNA/Apt-luminol) was prepared as a specific magnetic separation and signal switch material. ZnONPs-Au@CuMOFs shows excellent catalytic performance and was used as a catalyst for the luminol-H2O2 reaction. AFP will specifically recognize and bind to Apt on MGO-cDNA/Apt-luminol when AFP is present, which causes luminol release and triggers the CL reaction. The released luminol encounters ZnONPs-Au@CuMOFs and produces strong CL intensity. Therefore, a novel target-triggered "turn on" CL method with high selectivity and sensitivity for detecting AFP has been established. The linear range and detection limit were 1.0 × 10-4-50 ng mL-1 and 4.2 × 10-5 ng mL-1, respectively. The sensor also exhibited good selectivity, reproducibility and stability, and was finally used for AFP detection in serum samples.
