Abstract:After decades of decline, the Australian sheep flock aspires to rebuild its population of breeding ewes. A successful, rebuild will rely on high pregnancy rates and number of lambs born and reared. To examine this potential, a cross-sectional study of historical ultrasound pregnancy scanning records was undertaken using records collated from two experienced sheep pregnancy scanning businesses (years 2006 to 2019) from 15,397 mobs of ewes, totalling 7,443,314 ewes. Client details were de-identified and excluded from analyses, but details describing the mobs were retained when available, such as season of mating, production zone, ewe age, and breed. The key finding was a mean pregnancy rate (ewes pregnant per ewe scanned) of 0.76 ± 0.24, with a median of 0.83. Mobs scanned to identify fetal number had a higher mean (0.84 ± 0.15) and median (0.89) pregnancy rate. The mean reproduction rate (fetuses per ewe scanned) was 1.21 ± 0.27 and the median was 1.25. Differences were observed between the factors including age, breed, season, year or production zone but all results were lower than anticipated. The unexpected findings imply a problem exists with the fertility of many Australian sheep flocks.
Improving triplet lamb survival: management practices used by commercial farmers

Abstract:Introduction: Prolificacy has become an important breeding goal in sheep farming to increase farm profitability. With the adoption of improved genetics and management practices leading to increased lambing percentages, the proportion of triplet-born lambs has also increased on farms. However, mortality rates of triplet lambs are higher than for single- and twin-born lambs, and additional management inputs may be needed to support survival. The aim of this study was to identify factors that affect management practices that are considered important for triplet lamb survival by commercial farmers from the United Kingdom (UK), the Ireland (IRE), and New Zealand (NZ). Methods: An online survey was developed and disseminated to farmers in each country, focusing on farmer demographics, flock characteristics, management practices and production outcomes. A total of 448 farmers completed the survey, from the UK (n = 168), IRE (n = 218), and NZ (n = 62). Results: Respondents had larger flocks, higher scanning and lambing percentages than the country average for the UK and IRE. The mean percentage of triplet litters born within flocks was 9%, and lambs lost between scanning and lambing were 14% for UK, 15% for IRE, and 25% for NZ respondents (P = 0.063). Overall, 60% of all respondents reported to lamb indoors and 40% lambed outdoors, however NZ farmers almost exclusively lambed outdoors, whereas UK and IRE farmers lambed in both systems (P < 0.001). NZ farmers were more likely to rear all triplet lambs with the ewe, whereas UK and IRE farmers were more likely to remove a lamb to rear by another ewe or artificially (P < 0.001). Factors that influenced triplet lamb management practices of respondents in this study were respondent country of origin, flock size, age, and gender. In general, younger respondents (P < 0.001), and female respondents (P < 0.05), were more likely to engage in management activities that were considered to promote better triplet lamb survival, compared to older and male respondents respectively. These practices were associated with better lamb survival reported by respondents but were less likely to be carried out when flock size increased (P < 0.001). Discussion: The results of this survey highlight future priorities or communication strategies needed to improve triplet lamb survival.

Abstract:Objective: Stillbirths account for a relevant proportion of total calf mortality on dairy farms. In addition, the stillbirth rate is an important criterion for assessing animal welfare. The aim of this study was to determine the relationship between the stillbirth rate and aspects concerning the management of cows ante partum and during the calving period in larger dairy herds. Material and methods: In 97 dairy farms with an average herd size of 550 cattle (≥24 months old), data concerning farm management and animal health were collected as part of a farm inspection using a questionnaire. The stillbirth rate of the respective farms was determined based on the farm's own data on the milk yield test and analyzed for possible correlations with farm management using a multivariable factor analysis. Results: A higher stillbirth rate was found on farms with particularly lean and overconditioned transit cows in the herd. A positive association to the stillbirth rate was also found for farms using a special birth monitoring system for heifers and those with more than 20% assisted calvings. There was no association with herd size. Conclusions: Regardless of herd size, factors that are related to the stillbirth rate may be determined by analyzing the farm management. The body condition of the cows is a key parameter. The introduction of special birth monitoring for heifers can probably be interpreted as a reaction to increased stillbirths in the herd. Clinical relevance: Conditioning cows before calving is an important field of action for herd managers in order to avoid stillbirths. The cows should not approach the calving period in an overly lean or over-conditioned state. When in such state, however, it is advisable for these cows to be monitored more closely in order to allow for professional assistance at the pertinent time period. Integration of body condition scoring of cows at dry-off, calving and the end of the high-yielding period, assessment of calving conditions and training of staff involved in obstetrics into veterinary herd management is warranted.
Perinatal mortality in German dairy cattle: Unveiling the importance of cow-level risk factors and their interactions using a multifaceted modelling approach

Abstract:Perinatal mortality (PM) is a common issue on dairy farms, leading to calf losses and increased farming costs. The current knowledge about PM in dairy cattle is, however, limited and previous studies lack comparability. The topic has also primarily been studied in Holstein-Friesian cows and closely related breeds, while other dairy breeds have been largely ignored. Different data collection techniques, definitions of PM, studied variables and statistical approaches further limit the comparability and interpretation of previous studies. This article aims to investigate the factors contributing to PM in two underexplored breeds, Simmental (SIM) and Brown Swiss (BS), while comparing them to German Holstein on German farms, and to employ various modelling techniques to enhance comparability to other studies, and to determine if different statistical methods yield consistent results. A total of 133,942 calving records from 131,657 cows on 721 German farms were analyzed. Amongst these, the proportion of PM (defined as stillbirth or death up to 48 hours of age) was 6.1%. Univariable and multivariable mixed-effects logistic regressions, random forest and multimodel inference via brute-force model selection approaches were used to evaluate risk factors on the individual animal level. Although the balanced random forest did not incorporate the random effect, it yielded results similar to those of the mixed-effect model. The brute-force approach surpassed the widely adopted backwards variable selection method and represented a combination of strengths: it accounted for the random effect similar to mixed-effects regression and generated a variable importance plot similar to random forest. The difficulty of calving, breed and parity of the cow were found to be the most important factors, followed by farm size and season. Additionally, four significant interactions amongst predictors were identified: breed—calving ease, breed—season, parity—season and calving ease—farm size. The combination of factors, such as secondiparous SIM breed on small farms and experiencing easy calving in summer, showed the lowest probability of PM. Conversely, primiparous GH cows on large farms with difficult calving in winter exhibited the highest probability of PM. In order to reduce PM, appropriate management of dystocia, optimal heifer management and a wider use of SIM in dairy production are possible ways forward. It is also important that future studies are conducted to identify farm-specific contributors to higher PM on large farms.

Abstract:Lamb survival in Australian Merino sheep was investigated using survival records from 14,142 lambs born between 1975 and 1983. This data set included roll calls of live lambs at birth, 7 d, marking (30 d), and weaning (110 d), which allowed 4 binomial traits (alive or dead) to be recorded for each lamb at each time interval. The average survival to weaning was 72.4% with 23% of singles, 32% of twins, and 45% of multiple-born lambs not surviving to weaning. The timing of lamb loss was consistent across birth types; 6% died within 24 h of birth, a further 14% by 7 d, 3% between 7 and 30 d, and 8% between 30 and 110 d. Partitioning of phenotypic variation revealed that after the first postpartum week, mothering ability of Australian Merino ewes is not an important factor in lamb survival. Some ewes repeatedly lose lambs at birth and in the early postnatal period, but the intraclass correlation decreased by a factor of 10 for survival after 7 d of age (0.096 at birth and 0.100 at 7 d, falling to 0.009 at marking and 0.018 at weaning). This study examined the relationships of lamb survival with lamb birth weight and found varying relationships at each time period. Whereas birth weight had a highly significant curvilinear relationship with survival to weaning, the relationship was flatter for survival to 24 h with only small differences between average birth weights and the birth weight at which survival was optimized (-0.04, 0.28, and 0.54 kg for single-, twin-, and multiple-born lambs, respectively). This suggests that any management interventions to increase birth weight may increase the risk of death to both lamb and ewe during the lambing process due to dystocia. Among twin-born lambs there was a carryover effect of losses at birth on subsequent loss in the first week of life. Survival to 7 d of age was highly dependent on the survival of the littermate, favoring those whose littermate survived, but after the first week, this trend was reversed. Lamb birthcoat score had a small positive impact on survival only at birth.

Abstract:Reproductive wastage is a major economic burden in sheep production globally, especially within Australia as livestock production systems face increased pressure from climatic variability (e.g. prolonged droughts or flooding). Sheep are sensitive to acute changes in their environment such as heat stress, which if not adequately monitored will result in significant production losses such as reproductive failure, increased parasite and worm burden, morbidity and mortality risks. Through basic and applied research in the areas of stress and reproductive physiology our team has made significant advancements in the understanding of sheep behaviour and physiological responses to acute and chronic stressors. Using minimally invasive hormone monitoring technology in combination with field based assessment of sheep health and productivity traits, our team has delivered new knowledge on how sheep react to acute environmental stress and how it impacts on sheep reproduction. In this chapter, we evaluated the fertility rates and embryo quality of Merino ewes under AI breeding program. We discuss factors such as heat stress that can impact on ewe and offspring quality.
Herd-level risk factors associated with preweaning mortality on Ontario dairy farms

Abstract:The objective of this cross-sectional, herd-level study was to identify associations between calf management practices and herd-level preweaning mortality on Ontario dairy farms. From April to August 2022, a convenience sample of 100 dairy farms from Ontario, Canada, were visited once. A questionnaire, covering farm biosecurity, calving, colostrum management, preweaning nutrition, and housing, was verbally administered at each farm visit. Furthermore, data regarding preweaning calf mortality were retrieved from each farm's herd management software or records, including the total count of calves that survived, were culled, or died during the preweaning period for the 365 d before the farm visit. Preweaning mortality was defined as the proportion of calves that died between ≥48 h and 60 d of age divided by the total number of calves alive after 48 h of age. The herd-level preweaning mortality risk on sampled farms ranged from 0 to 15.9%, with an average of 2.8% (SD = 3.8%). A multivariable Poisson regression model was used to evaluate associations between 22 explanatory variables and preweaning mortality. Factors associated with greater herd-level preweaning mortality were larger herd size; having treatment protocols for diarrhea, pneumonia or navel infection written in collaboration with a veterinarian (compared with farms with the same treatment protocols developed without a veterinarian), and the herd-veterinarian never inquiring about calf health (compared with farms where the herd veterinarian inquired sometimes). Factors associated with lower herd-level preweaning mortality were using the calving pen for sick cows, having more than 4 people working with calves, offering calves a minimum volume of ≥9 L of milk/day, and farmers with a level of formal education higher than secondary school. These results indicate that producers may be able to reduce preweaning calf mortality by providing adequate labor for calf care, offering calves sufficient volumes of milk, being proactive in communicating with their veterinary practitioners about calf health, and potentially by engaging in continuous education.

Abstract:Poor lamb survival pre-weaning is a major source of reproductive inefficiency in Australian sheep flocks. While nutrition and management options have been extensively researched and promoted to improve lamb survival, the present review focuses on the prospects for obtaining genetic gain and helps identify selection strategies for boosting such gains to improve overall reproductive efficiency in the Australian sheep industry. Estimated heritability for lamb survival using linear model analysis is low, although use of threshold models suggests that heritability could be higher, which, if true, could help explain the substantial genetic gains obtained in long-term selection experiments. Epigenetic mechanisms may hinder selection and quantitative trait-loci identification through confounding and/or masking genetic variances and co-variances. With sufficient information, these effects could be considered in genetic evaluations by identifying those components that are amenable to selection. Regarding indirect selection, finding effective criteria for improving lamb survival has proved elusive. Most measures of maternal behaviour, temperament and lambing difficulty researched are poorly correlated genetically with lamb survival. Of lamb behaviours and thermo-genic indicators studied, latency to bleat following handling by humans is moderately genetically correlated with lamb survival, as is neonatal rectal temperature. Industry application remains to be adequately explored for the more promising of these measures. Finally, in lieu of direct selection for lamb survival, there is merit in selecting for multiple-rearing ability or its equivalent, possibly with additional selection criteria for lamb survival and reproductive efficiency.

Abstract:Common health problems observed during peripartum include milk fever, mastitis, fatty liver disease, ketosis, dystocia, retained placenta, metritis, hypomagnesaemia and abomasal displacements. The increased incidence of health problems observed during the periparturient period can be partly attributed to suboptimal immune responses. Factors contributing to decreased periparturient immunity include the act of parturition itself, impaired leukocytic activity, effects of colostrogenesis and lactogenesis, and associated hypocalcemia and negative energy balance. Nutritional and other management strategies represent a relevant short-term strategy aimed at improving the health and welfare of the transitioning cow. Additionally, it is important to consider improving the health of dairy herds through the genetic selection of animals with enhanced robustness by identifying those with superior disease resistance or resilience in the face of infection. As a consequence these animals are better able to cope with the production and environmental stresses. These may provide long-term selection strategies for improving the health and welfare of the transitioning cow particularly when combined with sound management practices, allowing dairy cattle to reach their full genetic potential.

Abstract:The effects of thiamylal on contractions induced by various mechanisms were investigated in mesenteric arteries isolated from dogs. Thiamylal (10(-4) to 10(-3) M) significantly inhibited contractions induced by KCl (20 mM) in normal media, and those induced by norepinephrine (10(-5) M) in normal and Ca(2+)-free media. Caffeine-induced contraction was significantly inhibited by thiamylal in the concentrations greater than 3 x 10(-5) M in intact fibers and 10(-5) M in chemically skinned fibers. Chemically skinned fibers that were precontracted with Ca2+ were relaxed by thiamylal in concentrations lower than those required to relax intact fibers that were precontracted with KCl (20 mM); the ED50 was 1.52 x 10(-5) M in skinned fibers and 5.50 x 10(-4) M in intact fibers. These results suggest that intracellular mechanisms are involved in thiamylal-induced vasodilatation of dog mesenteric artery.
Association of uterine health in the first lactation with transition cow health and reproductive performance in the second lactation of Holstein dairy cows.

Abstract:The objective of this study was to evaluate the effects of puerperal metritis (PM) diagnosed and treated during the early postpartum period of the first lactation on transition cow health, milk production, reproduction, and culling of dairy cows in their second lactation. Diagnosis of PM was based on fetid watery red-brown uterine discharge and rectal temperature above 39.5°C. Two farms were enrolled in this retrospective observational cohort study (Farm A and B). In both farms, the following diseases were recorded during the first 30 DIM in lactation 1 and 2: clinical hypocalcemia (CH), retained fetal membrane (RFM), PM, hyperketonemia (KET), left displaced abomasum (LDA) and clinical mastitis (MAST). Statistical analyses were performed using SPSS for Windows separately for each farm. Linear and logistic regression models were used for continuous (e.g., milk yield) and binary (e.g., disease, pregnancy per AI, pregnancy loss) outcomes, respectively. Cox proportional hazard regression models were calculated to model the time to event outcomes for culling or death during the first 60 DIM and for pregnancy within 250 d of the second lactation. The initial models contained the following variables: year of calving, month of calving, calving ease, stillbirth, twins, days open in lactation 1, 305 d milk yield in lactation 1, PM in lactation 1, and PM in lactation 2 as explanatory variables. A total of 4,834 cows (Farm A) and 4,238 cows (Farm B) in the second lactation were considered for statistical analyses. On farm A, the incidence of PM in lactation 1 and 2 was 20.1% and 11.2%, respectively. On farm B, the incidence of PM in lactation 1 and 2 was 14.4% and 8.5%, respectively. On both farms, cows with PM in their first lactation had greater odds for RFM and PM in their second lactation, while there was no association of PM in the first lactation with any other non-uterine diseases (i.e., CH, KET, LDA, and MAST) in the second lactation. Cows with PM in lactation 2 had reduced milk yield. The reduction in milk yield in second lactation was greater for cows that already experienced PM in lactation 1. On Farm A, cows with PM in their first lactation had a greater hazard for culling within 60 DIM of the second lactation; however, the same association was not present on Farm B. Cows with PM in lactation 1 had reduced pregnancy per AI at first service in the second lactation only on farm B. Cows with PM in lactation 2 had reduced pregnancy per AI at first service in the second lactation on both farms. Pregnancy loss in lactation 2 was only associated with PM in lactation 2 but not with PM in lactation 1. On both farms, cows had a reduced hazard for pregnancy in their second lactation within 250 DIM when they experienced PM in either lactation. In conclusion, PM in the first lactation had long-lasting negative consequences (i.e., risk of uterine disease and lower reproductive performance) for cows in their next lactation.

Abstract:Ligation of the superior mesenteric and the superior pancreatico-duodenal arteries just prior to alloxan treatment and their release ten minutes after treatment did not protect rats against development of alloxan diabetes. Prior incubation of alloxan in vitro for a period of ten minutes at a physiological pH value and 37° C. resulted in loss of diabetogenic activity. Identical blood flow interruption without alloxan treatment did not produce elevated blood glucose levels. After the intravenous injection of alloxan, a substance giving a fluorescence product with alloxan reagent was detected in blood over four hours. The substance remained at higher levels when animals, prior to alloxan treatment, were injected with reduced glutathione. There was also a prolonged detection of alloxan during incubation in whole blood and in buffer containing reduced glutathione. The production of alloxan diabetes following temporary interruption of arterial blood flow is discussed in the light of the present results.

Abstract:Maternal behaviour is important for lamb survival, as ewes perform many behaviours that affect the chances of a lamb surviving. Collecting maternal behaviour data directly at lambing is time-consuming and not considered suitable for acquiring the large volumes of data that would be required for using as selection criteria within commercial breeding flocks. The aim of this study was to investigate if a simple scoring system is heritable and assesses the expression of behaviours that reduce the probability of lamb mortality. Ewe behaviour was scored on a 3-point Maternal Assistance Score (MAS): (1) the ewe shows a high level of maternal interest (assumed if no intervention required); (2) the ewe shows limited interest in her lamb; and (3) the ewe shows no interest in her lamb. A total of 19 453 MAS were collected over 12 years, across 24 farms (including both indoor and outdoor lambing systems) and 12 different breed lines that make up the Innovis breeding programme. Ewe parity, breed, number of lambs carried, flock, lambing batch, lambing day within flock and pre-mating weight all had a significant effect on MAS (P < 0.05). The maternal assistance score was shown to be heritable (h2 = 0.05) and repeatable (0.10), positively genetically correlated to lambing difficulty (rg = 0.29) and amount of assistance the lamb required to suckle from the ewe (rg = 0.88), and negatively genetically correlated with the number of lambs successfully reared (rg = 0.49). This study shows that an easy-to-measure score can be used by shepherds with large breeding flocks, based on whether the ewe requires further assistance to support her lamb rearing. The score could be used in breeding programmes to select for lamb rearing ability in the future and potentially lead to an improvement in lamb welfare through a reduction in mortality.
Uterine tone influences fertility of Merino ewes following laparoscopic artificial insemination

Abstract:Artificial insemination (AI) plays a critical role in facilitating rapid genetic and production gains within the sheep industry. However, variable rates of AI success remain a concern for the industry and a barrier to adoption. Furthermore, the degree to which female factors influence the success of intrauterine laparoscopic AI rather than natural mating remains unknown. As such, this study investigates the effect of several factors collected during the time of AI, on the success of intrauterine laparoscopic AI. Data was generously donated by artificial breeding companies and stud breeders during routine commercial AI operations. AI data was collected over 3 breeding seasons during commercial AI programs (N = 24 programs) using Merino ewes (N = 24,700). Sire ID (N = 253), time of AI following progesterone removal (approx. 43-59 h post removal), uterine tone and intra-abdominal fat (both scored 1-5) as well as age of the ewe were all recorded at the time of AI. Transcutaneous ultrasound subsequently determined pregnancy rate approximately 55 days post-AI. A multivariate regression analysis was performed and revealed pregnancy success to increase when semen was inseminated into a ewe with a uterine tone score of 4 or 5 (P < 0.001). The remaining factors fell short of significance within the multivariate model. An interclass coefficient variation matrix was also used to determine the proportion of variation contributed to AI success by random factors allocated in the model; site, sire, AI date and breeding season (45.99 %, 29.94 %, 15.15 % and 8.92 %, respectively). These results highlight the influence of uterine tone on ewe fertility following laparoscopic AI, but also that program location and the sire used can further modify this influence on pregnancy rate. Abstract:In recent years, various developments have advanced the field of optical sensors based on single-photon avalanche diodes. In this contribution we present two sensors that were designed in 0.35μm CMOS technology. A silicon photomultiplier achieves a fill factor of 68 % at 50 μm pixel pitch and allows improved functionality by cointegration of application-specific readout electronics. A time-gated line sensor with gating times down to 1.5ns and serial digital output was designed based on optimized pixel geometry suitable for spectroscopy applications. Planned further improvements of sensors based on single-photon avalanche diode technology, including on-chip temperature compensation, are presented.

Abstract:Twelve polymorphic microsatellite loci were isolated for the bonefish, Albula vulpes using a polymerase chain reaction‐based procedure. The number of alleles ranged from two to 23 (mean = 8.8) in 37 specimens from south Florida. Observed and expected heterozygosities ranged from 0.07 to 0.77 (mean = 0.42) and from 0.07 to 0.84 (mean = 0.48), respectively. There were no significant departures from Hardy–Weinberg equilibrium and no evidence of genotypic disequilibrium between any pair of loci. In a cross‐amplification test, all markers yielded appropriately sized alleles for specimens of the provisional Albula sp. B and 11 of the 12 loci amplified for those of Albula glossodonta.
