Abstract:Traditional wound dressings have a limited capacity to absorb exudates, are permeable to microbes, and may adhere to wounds, which leads to secondary injuries. Hydrogels are promising alternative dressings to overcome the above challenges. In this study, we developed sodium alginate-based hydrogel films loaded with Betula utilis bark extract. These films were prepared via solvent-casting crosslinking method and evaluated for wound healing activity. Prepared films were 0.05–0.083 mm thick, flexible with folding endurance ranging from 197–203 folds, which indicates good physical properties. Optimized formulations exhibited successful loading of extract in the film matrix without any interaction as confirmed by FTIR. Maximum zone of inhibition against Gram-positive and Gram-negative bacteria was achieved by optimum formulation (B6), i.e., 19 mm and 9 mm, respectively, with > 90% scavenging activity. Furthermore, this optimum formulation (B6) was able to achieve 93% wound contraction in rats. Histograms of the optimized formulation treated group also revealed complete reepithelization of wounds. Conclusively, our extract-loaded hydrogel dressing successfully demonstrated its potential for cutaneous wound healing.

Preparation and characterization of amidated pectin-gelatin-oxidized tannic acid hydrogel films supplemented with in-situ reduced silver nanoparticles for wound-dressing applications

Abstract:Wound dressings play a crucial role in protecting injured tissues and promoting the healing process. Traditional fabrication of antibacterial wound dressings can be complex and may involve toxic components. In this study, we developed an innovative hydrogel film (AP:GE@OTA/Ag) composed of amidated pectin (AP), gelatin (GE), oxidized tannic acid (OTA) at varying concentrations, and in-situ reduced silver nanoparticles (AgNPs). FTIR and XRD analyses confirmed that crosslinking occurs via interactions between OTA quinone groups and free amino groups in AP and GE. TEM imaging demonstrated the well-dispersed AgNPs with an average particle size of 58.64 nm, while the TG measurements indicated the enhancement of the thermal stability compared to AP:GE films. The AP:GE@OTA/Ag films exhibited superior fluid uptake ability (90.96 % at 2 h), water retention capacity (91.69 % at 2 h), and water vapor transmission rate (1903.29 g/m2/day), alongside improved tensile strength (38 MPa). Additionally, these films showed excellent cytocompatibility and sustained potent antimicrobial activity against S. aureus and E. coli with low AgNPs loadings of 1.02 ± 0.13 μg/cm2. NIT-1 mouse insulinoma cells demonstrated robust proliferation when cultured with the prepared dressings. These films significantly accelerated wound repair in a skin excision model, indicating their potential clinical applications for wound healing.

Abstract:Carboxymethylated Cassia fistula gum (CCFG) and citric acid (CA) based wound healing film, (CCFG-CA) was developed using the solvent casting method. Glycerol was added as a plasticizing agent. The synthesized Carboxymethylated Cassia fistula gum cross-linked citric acid based hydrogel film (CCFG-CA) was evaluated morphologically, thermally, and structurally using FESEM, TGA, XRD and FTIR. Three essential oils (EO), rosemary ( Rosmarinus officinalis) , turmeric ( Curcuma longa) and thuja ( Thuja occidentalis L), known for antimicrobial and antioxidant activities, were loaded into the CCFG-CA film to develop essential oils loaded carboxymethylated Cassia fistula gum cross-linked citric acid based hydrogel film (CCFG-CA-EO). In vitro studies (MTT assay, disk diffusion assay, permeability tests and DPPH assay) confirm the biocompatibility, anti-oxidant and anti-microbial properties of the CCFG-CA-EO film. In vivo (wound healing studies on wistar rats and their histology) shows 99 % of wound healing and re-epithelialization in 14 days. Degradability (within 15 days), protein adsorption (12.05 μg/mL) and contact angle determination (69.43°ׄׄ ± 0.48) tests confirmed the potential of CCFG-CA-EO as an effective wound-healing material.

Abstract:Crude ginger has been used to treat wounds since ancient times till nowadays. The present study aimed at designing and characterizing topical hydrogel films loaded with ginger extract for wound healing in animal model. The hydrogel films were prepared using PVA and gelatin. The prepared films were evaluated for FTIR analysis, surface morphology, pH, swelling behavior, in vitro release, and % drug content. The wound-healing activity of the extract-loaded hydrogel films was compared with commercially available Silver Sulfadiazine® cream. The drug was compatible with the selected polymers and indicated the suitability of the selected polymers for preparation of topical hydrogel films. The SEM images clearly indicated porous structure of the prepared hydrogel films. Slight changes were observed in pH, ranging from 4.98 ± 0.079 in the beginning of the study to 4.9 ± 0.58 in the end. The swelling percentage after 8 h was 257.7%. The films released 78.7 ± 1.7% of the drug in 250 min The percent drug content was 97.78 ± 5% which did not change significantly during the storage period. The hydrogel films showed similar wound-healing activity as compared to the commercial product (p > 0.05; ANOVA), while greater wound-healing activity as compared to the control group (p < 0.05; ANOVA) evidenced by intensive collagen formation in histopathological analysis.

Gallic Acid-Loaded Sodium Alginate-Based (Polyvinyl Alcohol-Co-Acrylic Acid) Hydrogel Membranes for Cutaneous Wound Healing: Synthesis and Characterization

Abstract:Traditional wound dressings often cannot treat wounds caused by bacterial infections or other wound types that are insensitive to these wound treatments. Therefore, a biodegradable, bioactive hydrogel wound dressing could be an effective alternative option. The purpose of this study was to develop a hydrogel membrane comprised of sodium alginate, polyvinyl alcohol, acrylic acid, and gallic acid for treating skin wounds. The newly developed membranes were analyzed using Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM), X-ray diffraction analysis (XRD), sol-gel fraction, porosity, mechanical strength, swelling, drug release and data modelling, polymeric network parameters, biodegradation, and antioxidation (DPPH and ABTS) and antimicrobial activity against Gram-positive and negative bacteria. The results revealed that hydrogel membranes were crosslinked successfully and had excellent thermal stability, high drug loading, greater mechanical strength, and exhibited excellent biodegradation. Additionally, the swelling ability and the porosity of the surface facilitated a controlled release of the encapsulated drug (gallic acid), with 70.34% release observed at pH 1.2, 70.10% at pH 5.5 (normal skin pH), and 86.24% at pH 7.4 (wounds pH) in 48 h. The gallic acid-loaded hydrogel membranes showed a greater area of inhibition against Pseudomonas aeruginosa, Staphylococcus aureus, and Escherichia coli bacteria as well as demonstrated excellent antioxidant properties. Based on Franz cell analyses, the permeation flux of the drug from optimized formulations through mice skin was 92 (pH 5.5) and 110 (pH 7.4) μg/cm2·h−1. Moreover, hydrogel membranes retained significant amounts of drug in the skin for 24 h, such as 2371 (pH 5.5) and 3300 μg/cm2 (pH 7.4). Acute dermal irritation tests in rats showed that hydrogel membranes were nonirritating. Hydrogel membranes containing gallic acid could be an effective option for improving wound healing and could result in faster wound healing.

Abstract:Wound healing dressing based on a natural polymer of gellan gum incorporating zinc oxide nanoparticles and multiwall carbon nanotubes (GG/ZnONP + MWCNT) bionanocomposite film was fabricated via the solution casting method. The physicochemical properties of the film were characterized using X-ray diffraction (XRD), Fourier transform infrared (FTIR), and scanning electron microscopy (SEM). Moreover, the antibacterial properties of the bionanocomposite film were investigated for wound healing applications. The characterization results confirmed the reinforcement of the gellan gum (GG) matrix with zinc oxide nanoparticles (ZnONP) and multiwall carbon nanotubes (MWCNT), as an amorphous GG/ZnONP + MWCNT bionanocomposite film was obtained. SEM morphological analysis shows that the addition of ZnONP and MWCNT nanofillers changed the film microstructure into a sponge-like structure that is more suitable for fluid uptake and thus more useful for wound healing. The GG/ZnONP + MWCNT bionanocomposite film demonstrated good antibacterial activity against all strains tested. Furthermore, macroscopic analysis shows that the wound treated with GG/ZnONP + MWCNT bionanocomposite film recovered completely (100%) in 14 days, compared to pure GG film (90.76%) and negative control (77.40%). As a result, the GG/ZnONP + MWCNT bionanocomposite film could be a promising wound dressing material.

Abstract:The quest for biocompatible and antibacterial wound skin care dressings based on biopolymers has become a burgeoning focus in biomedicine. A wound care dressing of GG+SA@CUR thin film was prepared, and its properties were evaluated. FTIR analysis revealed distinct functional groups for GG, SA, and CUR. The GG+SA@CUR thin film is in an amorphous state, showing that CUR was successfully encapsulated for controlled drug release. The SEM images show the presence of CUR particles, which cause the film to have significant antibacterial properties. The inhibition zone was found to be 16.67 +/- 0.26 mm and 12.06 +/- 0.08 mm against S. aureus and E. coli. The good biocompatibility characteristics of the GG+SA@CUR thin film were proven by the high number of fibroblast cells. The scratch cells migrated faster after being treated with GG+SA@CUR film and accelerated wound healing in in vitro conditions, indicating their potential as new transdermal skin wound care.

Abstract:Traumatic multidrug-resistant bacterial infections are the most threat to wound healing. Lower extremity wounds under diabetic conditions display a significant delay during the healing process. To overcome these challenges, the utilization of protein-based nanocomposite dressings is crucial in implementing a successful regenerative medicine approach. These dressings hold significant potential as polymer scaffolds, allowing them to mimic the properties of the extracellular matrix (ECM). So, the objective of this study was to develop a nanocomposite film using dialdehyde-xanthan gum/soy protein isolate incorporated with propolis (PP) and halloysite nanotubes (HNTs) (DXG-SPI/PP/HNTs). In this protein-polysaccharide hybrid system, the self-healing capability was demonstrated through Schiff bonds, providing a favorable environment for cell encapsulation in the field of tissue engineering. To improve the properties of the DXG-SPI film, the incorporation of polyphenols found in PP, particularly flavonoids, is proposed. The synthesized films were subjected to investigations regarding degradation, degree of swelling, and mechanical characteristics. Additionally, halloysite nanotubes (HNTs) were introduced into the DXG-SPI/PP nanocomposite films as a reinforcing filler with varying concentrations of 3 %, 5 %, and 7 % by weight. The scanning electron microscope (SEM) analysis confirmed the proper embedding and dispersion of HNTs onto the DXG-SPI/PP nanocomposite films, leading to functional interfacial interactions. The structure and crystallinity of the synthesized nanocomposite films were characterized using Fourier Transform Infrared Spectrometry (FTIR) and X-ray diffraction (XRD), respectively. Moreover, the developed DXG-SPI/PP/HNTs nanocomposite films significantly improved cell growth of NIH-3T3 fibroblast cells in the presence of PP and HNTs, indicating their cytocompatibility. The antibacterial activity of the nanocomposite was evaluated against Escherichia coli (E. Coli) and Staphylococcus aureus (S. Aureus), which are commonly associated with wound infections. Overall, our findings suggest that the synthesis of DXG-SPI/PP/HNTs nanocomposite scaffolds holds great promise as a clinically relevant biomaterial and exhibits strong potential for numerous challenging biomedical applications.
Tannic acid-reinforced soy protein/oxidized tragacanth gum-based multifunctional hemostatic film for regulation of wound healing

Abstract:With recent advances in the field of tissue engineering, composite films with biocompatibility, antimicrobial properties, and wound healing properties have gained potential applications in the field of wound dressings. In this research work, composite films of soy protein (S)/oxidized tragacanth gum (G) were successfully made using the solution casting process. The metal-organic framework containing curcumin (MOF) with concentrations of 5 and 10 wt% and tannic acid (TA) with concentrations of 6 and 12 wt% were entered into the polymer film. Surface morphology with scanning electron microscope (FE-SEM), thermal stability, mechanical properties, chemical structure, antioxidant, water absorption, cell viability, antibacterial activity, and biodegradability of the prepared films were investigated in laboratory conditions. In addition, the toxicity of the films in the cell environment was investigated, and the results showed that cell growth and proliferation improved in the presence of the prepared films, especially films SG/MOF10/TA6 and SG/MOF10/TA12 due to the presence of TA and MOF containing curcumin. Also, the antibacterial activity of the films showed that the presence of tannic acid and curcumin in the structure of the films increases their ability against pathogens. According to the obtained results, the newly produced nanocomposite film (SG/MOF10/TA12) has a high potential to be used for wound dressing due to its favorable characteristics and was considered the optimal film.
A bioactive self-healing hydrogel wound-dressing based on Tragacanth gum: Structural and invitro biomedical investigations

Abstract:The design and development of wound-dressing hydrogels with desirable therapeutic effects and proper mechanical and self-healing properties are crucial in the healthcare sector. This research aims to prepare a new self-healing hydrogel based on Tragacanth, polyvinyl alcohol, and borax to be used as a wound dressing, the hydrogel was first prepared through a simple and one-pot reaction. The efficiency of the resulting product was then assessed based on the rheological and self-healing tests as well as cellular tests on a mouse fibroblast cell line (L929) including toxicity and scratch tests as well as the investigation of the expression of TGFβ1, TGFβ2, and VEGF-A gens (using Real-time PCR). The synthesized hydrogel exhibited proper mechanical strength, high self-healing features, and no toxicity (cell viability >100 %). Rheological studies indicate that hydrogels with a higher borax content (PVA: B ratio of 5:1) exhibit a higher storage modulus across all frequencies. The presence of hydrogel improved the migration of the L929 cells and scratch healing. The hydrogel also caused a significant improvement in the expression of the growth factors of the genes (P < 0.001). Therefore, it can be concluded that the prepared wound dressing can actively contribute to wound healing, opening promising potentials in medical applications.
Absorbable and biodegradable enzyme-crosslinked gelatin/alginate semi-IPN hydrogel wound dressings containing curcumin

Abstract:Effective wound management presents a substantial financial and time-related obstacle for healthcare institutions. Enhancing healthcare involves implementing innovative wound treatment methods to minimize healing time and expenses. This study is centered on the development of a non-toxic wound dressing using only two natural polymers and an enzyme. By adding 10 % wt microbial transglutaminase, the mechanical properties of the dressing were improved. This formulation increased the swelling rate by 70 %, deswelling rate by 15 %, conversion rate by 9 %, and networking rate by 20 %. Additionally, the non-toxic dressing showed a cell viability rate of 106 %. In drug delivery tests, explosive release behavior was observed, which is advantageous for open wounds. Cell staining experiments were also carried out to evaluate wound behavior in terms of collagen formation, granulation, and inflammation. The results suggest that the optimized hydrogel has great potential as a wound dressing. Its excellent absorption, antioxidant, and biocompatibility characteristics enhance tissue granulation rate and reduce wound treatment time by half compared to conventional methods, while also minimizing scarring risk. This innovative treatment, which eliminates the need for frequent changes, is beneficial for both secondary intentions and severe open wounds requiring bottom-up healing.

Abstract:For the healing process, in this study, an innovative polymeric hydrogel network including polyvinyl alcohol (PVA)/chitosan (CS)/gum tragacanth (GT) loaded with vitamin E (VE) was produced by the freeze–thaw approach. In order to investigate the characteristics of the prepared samples, Fourier transform infrared spectroscopy (FTIR) and scanning electron microscope (SEM) analyzes were performed. Also, water vapor transmission rate (WVTR), swelling ratio, gel fraction and mechanical properties were measured. Then, to observe their cytocompatibility, MTT assay and cell adhesion studies were assessed. The results of FTIR confirmed the presence of PVA, CS, GT, and VE in hydrogel films. As well as, the SEM images showed the effect of the freezing and thawing method in creating a smooth surface with small and regular pores. It was found with adding the CS and GT to PVA improves swelling ratio, gel fraction, WVTR and elongation of hydrogel films. Further, in examining the adhesion and cytotoxicity of the samples, the non‐toxic quiddity of the PVA/CS/GT hydrogel films was corroborated. In the end, the antibacterial properties revealed that the film containing GT and CS had the greatest antibacterial activity. According to the observed results, PVA/CS/GT hydrogel films loaded with VE can be good for wound healing applications.

Gallic acid functionalized silk fibroin/gelatin composite wound dressing for enhanced wound healing

Abstract:As the incidence of chronic wounds increases, the requirements for wound dressings are rising. The specific aim of this study is to propose a novel gallic acid (GA) functionalized silk fibroin (SF) and gelatin (Gel) composite wound dressing in which GA is used as an antibacterial and wound healing substance. Via electrospinning, SF, Gel, and GA mixed solutions could be conveniently fabricated into a composite nanofiber mat (SF-Gel-GA), consisting of uniform fibers with an average diameter around 134.57 ± 84 nm. The internal mesh structure of SF-Gel-GA provides sufficient drug loading capacity, proper moisture permeability, and proper degradation rate. SF-Gel-GA presents excellent biocompatibility. NIH-3T3 fibroblast cells could adhere and spread stably on the SF-Gel-GA surface with slightly promoted proliferation. In the presence of SF-Gel-GA, the growth of both Gram-positive and Gram-negative bacteria, includingStaphylococcus aureusandPseudomonas aeruginosa, is significantly inhibited in both plate and suspension cultures. A cutaneous excisional mouse wound model proves the efficient ability of SF-Gel-GA to promote wound healing. Compared with pure SF dressing and commercial Tegaderm Hydrocolloid3Mdressing, the wound closure rate with SF-Gel-GA treatment is significantly improved. The histological assessments further demonstrate SF-Gel-GA could facilitate collagen deposition, neovascularization, and epithelialization at wound sites to promote wound healing. In conclusion, a novel SF-Gel-GA composite wound dressing with efficient wound healing activities have been developed for chronic wound treatment with broad healing potential.

Abstract:BACKGROUND: Wounds have a bad prognostic nature and excessive discharges whose regular wound dressings are ineffective. Hydrogels are the best candidates for dressing such wounds due to their high water content and ability to exchange substances. Accordingly, the purpose of this study was to make a novel hydrogel wound dressing following the integration of various findings on wound healing and the use of regenerative medicine.MATERIALS AND METHODS: Various compounds were fabricated by glycerol/chitosan/polyvinyl alcohol (PVA) and then characterized to obtain the optimal composition using several techniques, including a water vapor passage test, scanning electron microscopy, water absorption, tensile strength, biodegradability, Fourier transform infrared spectroscopy, and antibacterial test.RESULTS: The findings revealed the optimal dressing ratio. Better antibacterial activity was found for the silver nanoparticle (AgNP) dressing.CONCLUSION: Our new fabricated dressing, glycerol/chitosan/PVA hydrogel loaded with AgNPs, exhibited satisfactory wound healing properties.

Abstract:In this research, a biomonomer based on hydroxyethyl methacrylate, citric acid, and polyethylene glycol (HEMA‐CA‐PEG) was synthesized. After confirming the reaction through 1H‐NMR and FT‐IR tests and obtaining the intended monomer, a flexible hydrogel was synthesized as a wound dressing, and its mechanical properties were evaluated using tensile strength, DMTA, and rheometry tests. This synthesized hydrogel exhibited sufficient mechanical properties. To enhance the efficiency of this hydrogel wound dressing and its effectiveness in facilitating wound healing, tannic acid (TA), as an antibacterial and antioxidant substance, was incorporated into it. In the second phase, antibacterial activity and animal studies were conducted. The comparison of the CA–based hydrogel with and without TA demonstrated that the TA–containing hydrogel achieved more than 99.9% inhibition and elimination of bacteria. The use of CA–based hydrogel with TA shown in treating burns on the backs of rats caused the wound to close after 17 days, and hair growth was observed by the 21st day. This synthesized flexible hydrogel wound dressing, along with suitable mechanical properties, exceptional antibacterial activity, increased granulation tissue formation, re‐epithelialization, angiogenesis, and collagen deposition, showed good performance in wound healing.

Abstract:The wound dressings fabricated by polymers and oregano essential oil (OEO) can be very effective as a hydrogel. The current study has been focused on fabricating the hydrogel membranes of oregano oil encapsulated as an antibacterial agent into sodium alginate (SA) solution by solvent casting method and then evaluated the antibacterial, antioxidant activity, and physicochemical performance of SA/OEO-based polymeric membranes. The polymeric interactions, surface morphology, water absorption capability, thermal stability, and encapsulation efficiency were investigated by FT-IR, SEM, swelling ratio, DSC, and encapsulation efficiency. The percentage encapsulation efficiency of essential oil was 40.5%. FTIR validated the presence of molecular interaction between individual components. SEM images showed a rough and porous appearance for hydrogel membranes. Moreover, DSC showed that the fabricated membranes were thermally stable. The inclusion of more content OEO decreased swelling ratios. The antioxidant test was carried out by DPPH assay and antibacterial test through disc diffusion method against microbes. The results revealed that membranes containing the highest content of OEO had more excellent antioxidant and antibacterial efficacy. Therefore, the polymeric membranes of sodium alginate loaded with oregano essential oil can be employed as an effective wound-healing candidate.

Abstract:Fabricating a fibrous well-ordered wound dressing for accelerating full-thickness wounds is a desirable treatment vector. Here, through modifications in the material extrusion device and adding a pneumatic-based injection, a material extrusion method for gelatin was introduced with the ability to fabricate 3D structure with repeat layers to support cell activity for the under layer. Furthermore, in the upper layer, the co-electrospinning of PU with gelatin was designed to simultaneously exploit the oxygen permeability and mechanical stability of PU with regenerative properties and collagen-like structure of gelatin. Moreover, zinc oxide nanoparticles (ZnO) was added into the 3D-printed under layer to synergistically benefit from the antibacterial properties of ZnO and the excellent biocompatibility of gelatin. The controllable porosity of the under layer, enabled through the additive manufacturing method, was adjusted to mimic the extracellular matrix of natural tissue with around (127.28 ± 20.70) μm pore size after swelling with smooth fibers. S. aureus, E. coli, Bacillus subtilis, and Pseudomonas with inhibition zone diameters at ∼ 2.14 cm and ∼ 1.96 cm, ∼ 4.01 cm, and ∼ 2.24 cm, respectively. Moreover, the scaffold showed great biocompatibility toward fibroblast cells after 7 days of cell culture with ∼ 89 % cell viability.

Abstract:Keeping in view the progressive future prospectus of carbohydrate research, herein, the potential of bioactive polysaccharides aloe vera (AV) and sterculia gum (SG) was explored to develop network hydrogel wound dressings. Both polysaccharides, acemannan of AV gel and glucuronic-galacturonic acid of SG accelerate wound healing process. The antibiotic drug cefuroxime encapsulated dressings were designed by graft-copolymerization with carbopol. The copolymers were characterized by SEM, AFM, FTIR, C13 NMR, XRD, TGA-DTG, DSC and swelling studies. The slow diffusion of cefuroxime drug exhibited non-Fickian mechanism and release of drug was occurred with a first order kinetic model. Polymer-bio membrane interactions indicated mucoadhesion nature. The clot formation was reduced during polymer-blood interactions indicated bio-compatible nature of the dressings moreover dressings were found antioxidant in nature. The porous dressings were O2 permeable and microbial impermeable. The hydrogel dressings revealed antimicrobial activity against P. aeruginosa. Degree of antimicrobial activity enhanced in case of hydrogel wound dressings loaded with cefuroxime. Overall, the results of these biochemical assays, sustained release of antibiotic drug and inherent wound healing potential of AV-SG indicated the use of hydrogels as dressing materials for better wound healing.
