Abstract:Biocompatible and antimicrobial wound dressings are important biomaterials for tissue adhesion and healing. Hydrogels derived from natural polyamino acids are ideal wound dressing because of their good water solubility, biocompatibility, and biodegradability. In this study, we introduce poly(amino acid)–based hydrogels as a new type of wound dressing for wound healing. The composite hydrogels were derived from water-soluble ε-polylysine and γ-poly(glutamic acid) cross-linked by 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide and N-hydroxy-succinimide. The gelation is attributed to the reaction between the amino groups of ε-polylysine and carboxyl groups of γ-poly(glutamic acid). Fourier transform infrared spectroscopy was used to characterize the composite hydrogels. The effect of the ratio of ε-polylysine and γ-poly(glutamic acid) on the gelation time, surface morphology, equilibrium swelling, elastic moduli, adhesive strength, and in vitro degradation of composite hydrogels was examined. The hydrogels showed excellent antimicrobial efficacy against Escherichia coli and Staphylococcus aureus. Indirect cytotoxicity assessment indicated that the composite hydrogels were nontoxic to the L929 cell. The potential of the composite hydrogel as wound dressing was demonstrated by applying it in the skin incision. Results demonstrated that the composite hydrogel showed superior healing effects compared with suture, fibrin glue, and compont skin adhesive, while eliciting less inflammatory response. In conclusion, antimicrobial and biocompatible ε-polylysine–γ-poly(glutamic acid) composite may have great application for wound healing.

Abstract:Development of natural protein-based hydrogels with self-healing performance and tunable physical properties has attracted increased attention owing to their wide potential not only in the pharmaceutical field, but also in wounds management. This work reports the development of a versatile hydrogel based on enzymatically-crosslinked gelatin and nanogels loaded with amoxicillin (Amox), an antibiotic used in wound infections. The transglutaminase (TGase)-crosslinked hydrogels and encapsulating nanogels were formed rapidly through enzymatic crosslinking and self-assembly interactions in mild conditions. The nanogels formed through the self-assemble of maleoyl-chitosan (MAC5) and polyaspartic acid (PAS) may have positive influence on the self-healing capacity and drug distribution within the hydrogel network through the interactions established between gelatin and gel-like nanocarriers. The physicochemical properties of the enzymatically-crosslinked hydrogels, such as internal structure, swelling and degradation behavior, were studied. In addition, the Amox release studies indicated a rapid release when the pH of the medium decreased, which represents a favorable characteristic for use in the healing of infected wounds. It was further observed through the in vitro and in vivo biocompatibility assays that the optimized scaffolds have great potential to be used as wound dressings.

Abstract:The aim of this research was to investigate the potential of ampicillin-loaded hydrogels based on polyvinyl alcohol (PVA), hyaluronic acid (HA) and kappa-carrageenan (K-Carr) as an antibiotic-releasing wound dressing. Firstly, the novel polymers (HA-g-K-Carr) were synthesized by grafting of HA onto K-Carr using 4-dimethylaminopyridine/1-(3-dimethylaminopyl)-3-ethyl-carbodiimide hydrochloride as catalyst system via esterification reaction. The characterization of the polymer structure was performed by Fourier transform infrared spectrum (FTIR), proton nuclear magnetic resonance and thermogravimetric analysis. Secondly, PVA/(HA-g-K-Carr) hydrogel with/without loading of ampicillin molecules was formed via freeze–thawing method since PVA/K-Carr and PVA/HA hydrogels were also fabricated as control groups. The hydrogels were subjected to characterizations with FTIR and X-ray diffractometer. PVA/(HA-g-K-Carr) hydrogel demonstrated the highest swelling amount and highest ampicillin release amount reaching an equilibrium value after 480 min rather than the other hydrogels. Also, PVA/(HA-g-K-Carr) hydrogel exhibited inhibition zone against Escherichia coli and Staphylococcus aureus and no cytotoxic effect for L929 cells. All the results showed that PVA/(HA-g-K-Carr) hydrogels are good candidates for wound dressing applications.
Interpenetrating Polymer Network Hydrogels Formed Using Antibiotics as a Dynamic Crosslinker for Treatment of Infected Wounds

Abstract:Antibacterial hydrogels, particularly antibiotic‐loaded hydrogels, are promising wound dressing materials for treatment of bacteria‐infected wound. However, it is challenging to achieve sustained release of antibiotics from hydrogels through physical encapsulation of the antibiotics. Herein, we reported an interpenetrating polymer network P(AA‐co‐HEMA)Gen hydrogel with double crosslinking formed by free radical polymerization of 2‐hydroxyethyl methacrylate (HEMA) and acrylic acid (AA), while using the antibiotic gentamicin (Gen) as the dynamic physical crosslinker. Gentamicin was incorporated into the hydrogel networks via electrostatic interaction between the carboxyl groups of poly(acrylic acid) and the amino groups of gentamicin, which led to pH‐responsive drug release and a significant increase in mechanical strength (i.e., elastic modulus, viscous modulus and compressive modulus). More importantly, the hydrogels with optimal compositions demonstrated long‐lasting antibacterial activity against both Gram‐positive bacteria (Staphylococcus aureus) and Gram‐negative bacteria (Escherichia coli) over 28 days. The in vivo studies that were conducted in a S. aureus‐infected full‐thickness skin wound model demonstrated that the double crosslinking hydrogels loaded with gentamicin eliminated bacteria in the wounds more effectively and significantly accelerated wound healing as compared to 3M dressing and the control without any treatment. Taken together, this antibiotic‐loaded interpenetrating polymer network hydrogel is potentially a promising wound dressing material for the treatment of bacteria‐infected wound. Abstract:The preparation of hydrogels based on biopolymers like collagen and gum arabic gives a chance to provide novel options that can be used in biomedical field. Through a polymeric semi-interpenetration technique, collagen-based polymeric matrices can be associated with gum arabic while controlling its physicochemical and biological properties. To create novel hydrogels with their potential use in the treatment of wounds, the semi-interpenetration process, altering the concentration (0–40% by wt) of gum arabic in a collagen matrix is explored. The ability of gum arabic to create intermolecular hydrogen bonds in the collagen matrix enables the development of semi-interpenetrating polymeric networks (semi-IPN)-based hydrogels with a faster gelation time and higher crosslinking. Amorphous granular surfaces with linked porosity are present in matrices with 30% (by wt) of gum arabic, enhancing the storage modulus and thermal degradation resistance. The hydrogels swell to very high extent in hydrolytic and proteolytic environments, good hemocompatibility, and suppression of growth of pathogens like E. coli, and all it is enhanced by gum arabic included them, in addition to enabling the controlled release of ketorolac. The chemical composition of theses semi-IPN matrices have no deleterious effects on monocytes or fibroblasts, promoting their proliferation, and lowering alpha tumor necrosis factor (α-TNF) secretion in human monocytes.

Abstract:The in situ injectable hydrogel system offers a widespread range of biomedical applications in prompt chronic wound treatment and management, as it provides self-healing, maintains a moist wound microenvironment, and offers good antibacterial properties. This study aimed to develop and evaluate biopolymer-based thermoreversible injectable hydrogels for effective wound-healing applications and the controlled drug delivery of meropenem. The injectable hydrogel was developed using the solvent casting method and evaluated for structural changes using proton nuclear magnetic resonance, Fourier transforms infrared spectroscopy, thermogravimetric analysis, and scanning electron microscopy. The results indicated the self-assembly of hyaluronic acid and kappa-carrageenan and the thermal stability of the fabricated injectable hydrogel with tunable gelation properties. The viscosity assessment indicated the in-situ gelling ability and injectability of the hydrogels at various temperatures. The fabricated hydrogel was loaded with meropenem, and the drug release from the hydrogel in phosphate buffer saline (PBS) with a pH of 7.4 was 96.12%, and the simulated wound fluid with a pH of 6.8 was observed to be at 94.73% at 24 h, which corresponds to the sustained delivery of meropenem. Antibacterial studies on P. aeruginosa, S. aureus, and E. coli with meropenem-laden hydrogel showed higher zones of inhibition. The in vivo studies in Sprague Dawley (SD) rats presented accelerated healing with the drug-loaded injectable hydrogel, while 90% wound closure with the unloaded injectable hydrogel, 70% in the positive control group (SC drug), and 60% in the negative control group was observed (normal saline) after fourteen days. In vivo wound closure analysis confirmed that the developed polymeric hydrogel has synergistic wound-healing potential.

Abstract:Biopolymer-based hydrogels with sustained drug release capability and antibacterial activity have exhibited great potential in clinical application in drug delivery and wound healing. In this study, a new type of composite wound dressing hydrogel aiming at avoiding wound infection was developed through embedding drug loaded gellan gum microspheres (GMs) into a doubly crosslinked hydrogel, which was constructed by Schiff-base crosslinking of oxidized gellan gum (OG) (pre-crosslinked by calcium ion) and carboxymethyl chitosan (CMCS). The gelation time, swelling index, degradation rate and mechanical properties of the blank hydrogel was optimized by varying the ratios of CMCS/OG (w/w) with fixed OG/calcium (w/w) ratio. The best overall performance of the hydrogel was obtained when CMCS/OG is 16/7 (w/w), with a 139 s gelation time, swelling index remained above 30 after swelling equilibrium, 100.5% degradation rate on the seventh day, and 8.8 KPa compressive modulus. After being embedded with cargo-loaded GMs, the aforementioned performance of the blank hydrogel was improved, and the sustained release of cargoes (antibacterial drugs, tetracycline hydrochloride and silver sulfadiazine) was observed. Moreover, the excellent antibacterial activity of the composite hydrogel was also demonstrated in vitro. These results support the bioactive composite hydrogel can be employed as a promising injectable scaffold for promoting wound regeneration and drug delivery.

Abstract:The current study aimed to produce a material that has dual effects of healing and anti-inflammatory activity. For this purpose, a κ-carrageenan/polyacrylamide film loaded with cetrimide (κ-CAR/PAAm/CI) was developed using the manual casting technique. Definite concentrations of κ-CAR and AAm were heated at 80 °C for 2 h, and CI and glycerol were added. The solution was cast without using an initiator or crosslinker. The reaction of the sulfonic acid group -SO3H of κ-CAR with the –CONH2 group of PAAm lead to the formation of a sulfonamide (–SO2NH–) group. The characteristics of the produced films were investigated based on FT-IR, TGA, the contact angle, and mechanical properties. An improvement in the thermal stability of the κ-CAR/PAAm/CI2 film containing 1.5% CI was achieved, compared to the film with 0.5% CI (κ-CAR/PAAm/CI1). The contact angle measurement proved that the films were hydrophobic, enhanced by increasing the CI content. The tensile strength and elongation percent values are considered adequate for materials used in wound care. The κ-CAR/PAAm/CI2 (1.5% CI) film showed superior antimicrobial activity against P. aeruginosa, moderate activity against S. aureus, and low activity against E. coli. The κ-CAR/PAAm/CI2 film effectively inhibited heat-induced hemolysis and showed wound contraction activity at a level of 100% after 19 days of excision wound treatment. The prepared films may offer a promising approach for the development of effective wound dressings.
Antibacterial Broad-Spectrum Dendritic/Gellan Gum Hybrid Hydrogels with Rapid Shape-Forming and Self-Healing for Wound Healing Application

Abstract:Treating wound infections is a difficult task ever since pathogenic bacteria started to develop resistance to common antibiotics. The present study develops hybrid hydrogels based on the formation of a polyelectrolyte complex between the anionic charges of dopamine-functionalized Gellan Gum (GG-DA) and the cationic moieties of the TMP-G2-alanine dendrimer. The hydrogels thus obtained can be doubly crosslinked with CaCl2 , obtaining solid hydrogels. Or, by oxidizing dopamine to GG-DA, possibly causing further interactions such as Schiff Base and Michael addition to take place, hydrogels called injectables can be obtained. The latter have shear-thinning and self-healing properties (efficiency up to 100%). Human dermal fibroblasts (HDF), human epidermal keratinocytes (HaCaT), and mouse monocyte cells (RAW 264.7), after incubation with hydrogels, in most cases show cell viability up to 100%. Hydrogels exhibit adhesive behavior on various substrates, including porcine skin. At the same time, the dendrimer serves to crosslink the hydrogels and endows them with excellent broad-spectrum microbial eradication activity within four hours, evaluated using Staphylococcus aureus 2569 and Escherichia coli 178. Using the same GG-DA/TMP-G2-alanine ratios hybrid hydrogels with tunable properties and potential for wound dressing applications can be produced.
Gallic Acid-Loaded Sodium Alginate-Based (Polyvinyl Alcohol-Co-Acrylic Acid) Hydrogel Membranes for Cutaneous Wound Healing: Synthesis and Characterization

Abstract:Traditional wound dressings often cannot treat wounds caused by bacterial infections or other wound types that are insensitive to these wound treatments. Therefore, a biodegradable, bioactive hydrogel wound dressing could be an effective alternative option. The purpose of this study was to develop a hydrogel membrane comprised of sodium alginate, polyvinyl alcohol, acrylic acid, and gallic acid for treating skin wounds. The newly developed membranes were analyzed using Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM), X-ray diffraction analysis (XRD), sol-gel fraction, porosity, mechanical strength, swelling, drug release and data modelling, polymeric network parameters, biodegradation, and antioxidation (DPPH and ABTS) and antimicrobial activity against Gram-positive and negative bacteria. The results revealed that hydrogel membranes were crosslinked successfully and had excellent thermal stability, high drug loading, greater mechanical strength, and exhibited excellent biodegradation. Additionally, the swelling ability and the porosity of the surface facilitated a controlled release of the encapsulated drug (gallic acid), with 70.34% release observed at pH 1.2, 70.10% at pH 5.5 (normal skin pH), and 86.24% at pH 7.4 (wounds pH) in 48 h. The gallic acid-loaded hydrogel membranes showed a greater area of inhibition against Pseudomonas aeruginosa, Staphylococcus aureus, and Escherichia coli bacteria as well as demonstrated excellent antioxidant properties. Based on Franz cell analyses, the permeation flux of the drug from optimized formulations through mice skin was 92 (pH 5.5) and 110 (pH 7.4) μg/cm2·h−1. Moreover, hydrogel membranes retained significant amounts of drug in the skin for 24 h, such as 2371 (pH 5.5) and 3300 μg/cm2 (pH 7.4). Acute dermal irritation tests in rats showed that hydrogel membranes were nonirritating. Hydrogel membranes containing gallic acid could be an effective option for improving wound healing and could result in faster wound healing.

pH-Switchable Antimicrobial Supramolecular Hydrogels for Synergistically Eliminating Biofilm and Promoting Wound Healing

Abstract:Biofilm infection will cause chronic inflammation and hinder the normal healing process of wound. Here, based on the self-assembly of three designed amphiphilic pentapeptides named EK, GG, and DR, pH-switchable antibacterial hydrogels with amphiphilic fiber network are used for the eradication of biofilms and the rescue of delayed healing in infected wounds. These pentapeptides-based hydrogels exhibit an acidic pH-switchable antimicrobial effect and are biocompatible at neutral pH. Additionally, supramolecular nanofiber networks with physical cross-linking with thermosensitive polymers (PNIPAm) and loaded antibacterial oregano oil are further developed. In vitro experiments indicate that the antimicrobial activity of hydrogels comes from the disassembly of acidic pH-dependent nanofiber network and activated release of pentapeptides and oregano oil, which achieves synergistic biofilm eradication. Remarkably, DR-based supramolecular hydrogel improves the healing efficiency of the full-thickness wound of skin in vivo, which is manifested by increased wound closure rate, reduced inflammatory response, faster angiogenesis, and collagen deposition in the wound, exhibiting great potential as wound dressing. Abstract:When skin comes into direct contact with the outside environment, it becomes extremely prone to injury and external factors can make wounds difficult to heal. Traditional medical dressings often cause secondary injury and are poorly resistant to infection. Hydrogels offer a promising alternative to overcome these difficulties. In this study, chitosan (CS)/gelatin (GEL)/polyvinyl alcohol (PVA) hydrogels were developed by chemical cross-linking and loaded with the drug kitasamycin (KM) for testing. The hydrogels' in vitro drug release and wound-healing properties were assessed. For 48 h, the drug release from the hydrogel in vitro persisted, which was significantly longer than the release time of the KM solution. Antimicrobial activity tests showed that the loaded KM hydrogel maintained its bacteriostatic ability at the same concentration as the KM solution, and during in vitro bacteriostatic inhibition, the duration of bacteriostatic inhibition of the KM hydrogel was significantly prolonged compared to that of the KM solution. This confirms the controlled release capability of the hydrogel. In addition, the hydrogel reduced the wound size in mice by 96 % and histopathological tests showed complete re-epithelialization of the wound. The prepared hydrogels successfully demonstrated their potential ability to control drug release and promote skin wound healing.

Abstract:We developed the interpenetrating double network composite hydrogel based on poly (vinyl alcohol) (PVA) and fish gelatin (FG) via thermal treatment and repeated freeze-thawing. A function of salicylic acid was incorporated into the hydrogel to improve its antibacterial properties. The color values, water contents, water evaporation rate, and swelling behavior were investigated. The drug-loading performance of the composite hydrogel was demonstrated by loading salicylic acid in various hydrogel systems. Moreover, the cumulative dissolution percentage of salicylic acid and the antibacterial activity of composite hydrogel were carried out. The results revealed that as FG concentration increased from 0% to 3.75% (w/v), gels changed from white to slight yellow and the swelling ratio increased from 54% to 83% (within 8 h). The presence of FG decreased the water content of gels which ranged from 86% to 89% and also decreased water evaporation rate. All gels presented the swelling index within 0.5–1.0, indicating a non-Fickian diffusion mechanism. The drug sustained dissolution behavior of pure PVA and composite hydrogel showed the same trend. Besides, the presence of the obvious bacteriostatic zones means that drug-loaded composite hydrogels have an effective antibacterial property. These results demonstrated that PVA/FG-based interpenetrating hydrogel is an appropriate biomaterial for drug-carrying wound dressing application.

Abstract:Background Open skin wounds increase the risk of infections and can compromise health. Therefore, applying medications to promote healing at the injury site is crucial. In practice, direct drug delivery is often difficult to maintain for a long time due to rapid absorption or wiping off, which reduces the efficiency of wound healing. Consequently, the development of bioactive materials with both antibacterial and wound‐healing properties is highly desirable. Methods This study synthesized liposomes loaded with glycyrrhizic acid (GA) and asiaticoside (AS) by film dispersion‐ultrasonication method, which were then incorporated into a GelMA solution and cross‐linked by ultraviolet light to form a bioactive composite hydrogel for wound dressings. Results This hydrogel is conducive to the transport of nutrients and gas exchange. Compared with GelMA hydrogel (swelling rate 69.8% ± 5.7%), the swelling rate of GelMA/Lip@GA@AS is lower, at 52.1% ± 1.0%. GelMA/Lip@GA@AS also has better compression and rheological properties, and the in vitro biodegradability is not significantly different from that of the collagenase‐treated group. In addition, the hydrogel polymer has a stable drug release rate, good biocompatibility, and an angiogenic promoting effect. In vitro experiments prove that, at concentrations of 0.5, 1, 2, and 3 mg/mL, GelMA/Lip@GA@AS can inhibit the growth of Staphylococcus aureus. Conclusion We synthesized GelMA/Lip@GA@AS hydrogel and found it possesses advantageous mechanical properties, rheology, and biodegradability. Experimental results in vitro showed that the bioactive hydrogel could efficiently release drugs, exhibit biocompatibility, and enhance angiogenesis and antimicrobial effects. These results suggest the promising application of GelMA/Lip@GA@AS hydrogel in wound‐dressing materials.

Abstract:One of the main reasons impeding wound healing is wound infection caused by bacterial colonization with a continuous stage of inflammation. Traditional wound treatments like gauze are being replaced by tissue adhesives with strong wet tissue adhesion and biocompatibility. Herein, a fast-crosslinking hydrogel is developed to achieve both strong antimicrobial properties and excellent biocompatibility. In this study, a simple and non-toxic composite hydrogel was prepared by the Schiff base reaction between the aldehyde group of 2,3,4-trihydroxybenzaldehyde (TBA) and the amino group of ε -Poly- L -lysine (EPL). Subsequently, a succession of experiments toward this new hydrogel including structure characterization, antimicrobial properties, cell experiment and wound healing were applied. The results of the experiments show that the EPL-TBA hydrogel not only exhibited excellent contact-active antimicrobial activities against Gram-negative bacteria Escherichia coli ( E. coil ) and Gram-positive Bacteria Staphylococcus aureus ( S. aureus ), but also inhibited the biofilm formation. More importantly, the EPL-TBA hydrogel promoted the wound healing with low cytotoxicity in vivo . These findings indicate that the EPL-TBA hydrogel has a promising use as a wound dressing in the bacterial infection prevention and wounds healing acceleration.

Investigation on Swelling of Agar‐Based Antibacterial Hydrogels for Hard‐To‐Heal Wound Dressings

Abstract:Despite a wide range of available wound treatments, hard‐to‐heal wounds still pose a challenge. Hydrogels are often used as dressings for these wounds, because they sustain moisture in the wound environment, supporting the natural healing process. However, it is still not fully understood how physicochemical properties of hydrogel matrix affect the drug release process. Thus, detailed swelling kinetics examination coupled with modeling is needed together with studies on drug release. In this regard, several hydrogels based on plant‐derived agar and modified with amikacin were investigated. The main properties of hydrogels were examined focusing on detailed swelling kinetics. Drug release was studied as microbiological activity against E. coli and S. Epidermidis strains. The obtained hydrogels were characterized by high swelling, reaching values in range from 465 to 1300%, fitting the second order kinetics mode and exhibiting the quasi‐Fickian diffusion properties. Furthermore, there was no correlation found between swelling properties and antibacterial activity against tested strains. The results confirmed that presented hydrogel materials have desirable properties for application as dressings for hard‐to‐heal wounds. The suggested compositions are a promising base for modification with other active substances (e.g., regenerative, anti‐inflammatory) and studying the broader correlation between swelling and drug release.

Abstract:Antibacterial hydrogels have emerged as a promising approach for effective wound treatment. However, despite extensive research on the fabrication of antibacterial hydrogels, it remains challenging to develop injectable, biocompatible, transparent, and mass-producible hydrogels with antibacterial properties. In this study, we successfully fabricated an antibacterial drug-loaded composite hydrogel, named CC45/OKG40/HS, through a Schiff base reaction between carboxymethyl chitosan (CC) and oxidized konjac glucomannan (OKG), followed by the encapsulation of stevioside-stabilized honokiol (HS) micelles. The CC45/OKG40/HS hydrogel exhibited several favorable properties, including a short gel time (<10 min), high water content (>92%), injectability, good adhesiveness, self-healing ability, and high transparency. In vitro experiments confirmed its excellent antibacterial properties, antioxidant activities, and high biocompatibility (no cytotoxicity, hemolysis ratio <5%). Furthermore, in vivo evaluation demonstrated that the CC45/OKG40/HS0.5 hydrogel accelerated wound healing by relieving inflammatory responses and enhancing re-epithelization. Given its feasibility for mass production, the findings showed that the CC45/OKG40/HS hydrogel has the potential as an advanced antibacterial wound dressing for commercial use.

Preparation of Cross-linked Chitosan Quaternary Ammonium Salt Hydrogel Films Loading Drug of Gentamicin Sulfate for Antibacterial Wound Dressing

Abstract:Hydrogels, possessing high biocompatibility and adaptability to biological tissue, show great usability in medical applications. In this research, a series of novel cross-linked chitosan quaternary ammonium salt loading with gentamicin sulfate (CTMCSG) hydrogel films with different cross-linking degrees were successfully obtained by the reaction of chitosan quaternary ammonium salt (TMCS) and epichlorohydrin. Fourier transform infrared spectroscopy (FTIR), thermal analysis, and scanning electron microscope (SEM) were used to characterize the chemical structure and surface morphology of CTMCSG hydrogel films. The physicochemical property, gentamicin sulphate release behavior, cytotoxicity, and antibacterial activity of the CTMCSG against Escherichia coli and Staphylococcus aureus were determined. Experimental results demonstrated that CTMCSG hydrogel films exhibited good water stability, thermal stability, drug release capacity, as well as antibacterial property. The inhibition zone of CTMCSG hydrogel films against Escherichia coli and Staphylococcus aureus could be up to about 30 mm. Specifically, the increases in maximum decomposition temperature, mechanical property, water content, swelling degree, and a reduction in water vapor permeability of the hydrogel films were observed as the amount of the cross-linking agent increased. The results indicated that the CTMCSG-4 hydrogel film with an interesting physicochemical property, admirable antibacterial activity, and slight cytotoxicity showed the potential value as excellent antibacterial wound dressing.

Abstract:Tissue adhesion of hydrogels plays an important role in wound healing, which can improve the efficiency of wound treatment, stop bleeding, facilitate tissue growth and wound closure. However, most non-covalent crosslinked hydrogels have weak tissue adhesion and rheological properties. Furthermore, it remains a challenge to synthesize a fully physically crosslinked hydrogel with good rheological properties without compromising its tissue adhesion strength. In this paper, a physically crosslinked hydrogel was developed from a mixture of chitosan and pullulan in different polymer volume ratios using aqueous NaOH. Fourier transform infrared spectroscopy, scanning electron microscopy, thermal analysis, rheological and lap shear tests were used to evaluate the influence of polymer volume ratios on the rheological, and tissue adhesive properties of the hydrogels. It was found that the hydrogels possessed high tissue adhesive strength ranging from 18.0 ± 0.90 to 49.0 ± 2.45 kPa and good storage moduli up to 5.157 ± 1.062 kPa. Gentamicin was incorporated into this polymer matrix and the release profile was investigated. The ratio of chitosan and pullulan to obtain hydrogels with optimum viscoelastic and tissue adhesive properties was identified to be CS/PUL 2:1. These results indicated that the synthesized hydrogels can be potential materials for biomedical applications such as medical adhesives and wound dressings.

Abstract:Prevention and management of wound infections receive a lot of attention, since the presence of micro-organisms interferes with the wound-healing process. The aim of this work was to use cyclodextrins (CDs) to endow hydrogels and gauzes with the ability to take up antiseptics and sustain their delivery for several hours. Benzalkonium chloride (BzCl) can form inclusion complexes with cross-linked CDs that regulate the release through an affinity-driven mechanism. Grafting of CDs to cotton gauzes using citric acid as the linker, at 190 °C and for 15 min, led to grafting yields of about 148%, much larger than those obtained at 180 °C or with shorter reaction times. Microbiological tests revealed that the BzCl-loaded networks can inhibit the growth of Staphylococcus epidermidis and Escherichia coli both on agar plates and in liquid medium. Furthermore, the antiseptic-loaded gauzes were able to inhibit biofilm formation by Staphylococcus aureus RN1HG pMV158GFP when applied in early stages of biofilm formation and could reduce the number of living cells in preformed biofilms grown in a chronic wound biofilm model. These findings highlight the role of CDs as main components of hydrogels and gauzes for the efficient delivery of antiseptics.
