Abstract:BACKGROUND Tirapazamine (TPZ), a new anti-cancer drug activated to a toxic free radical under hypoxic conditions, produces a tumor specific potentiation of cell kill by cisplatin. In the present study we discuss the mechanism and clinical potential of this effect, as well as investigate the influence of p53 mutations on the activity of TPZ. MATERIALS AND METHODS For in vitro experiments we have used mouse SCCVII tumor cells, minimally transformed mouse embryo fibroblasts (MEFs) from wild-type and p53 knockout mice, and several human NSCLC cell lines. For in vivo experiments we have used RIF-1 tumors implanted subcutaneously into C3H mice. RESULTS Prior injection of TPZ into tumor-bearing mice markedly potentiated tumor cell kill by cisplatin, but produced no effect on systemic toxicity. The maximum potentiation occurred when TPZ was injected two to three hours prior to cisplatin administration. Experiments performed with cells in vitro showed a similar synergistic interaction between the two drugs when cells were exposed to TPZ under hypoxic conditions prior to exposure to cisplatin. Experiments with MEFs from either p53 wild-type or p53-knockout mice showed no influence of p53 on the sensitivity of cells to killing by TPZ under hypoxia. A similar lack of influence of p53 on the toxicity to TPZ was obtained for a panel of NSCLC cell lines. CONCLUSIONS TPZ is a novel anticancer drug that produces tumor selective potentiation of cisplatin and carboplatin in both pre-clinical and clinical studies. The fact that the drug produces no potentiation of the systemic side effects of these drugs, or of other anticancer drugs used in combination with platinum in NSCLC, suggests that TPZ could become a useful agent in the treatment of lung cancer.
Evaluation of the Antioxidant Properties of Litchi Fruit Phenolics in Relation to Pericarp Browning Prevention.

Abstract:Phenolics were extracted from litchi fruit pericarp (LFP) tissues, purified and their antioxidant properties analyzed. LFP phenolics strongly inhibited linoleic acid oxidation and exhibited a dose-dependent free-radical scavenging activity against α,α-diphenyl-β-picrylhydrazyl (DPPH·) and hydroxyl radicals and superoxide anions. The degradation of deoxyribose by hydroxyl radicals was inhibited by phenolics acting mainly as iron ion chelators, rather than by directly scavenging the radicals. Phenolics from litchi fruit pericarp were found to display similar reducing power activity as ascorbic acid. The effect of phenolic compound treatment on pericarp browning and electrolyte leakage of litchi fruit was also evaluated and it was observed that application of exogenous litchi phenolics to harvested litchi fruit significantly prevented pericarp browning and delayed increases in electrolyte leakage. These results suggest that litchi pericarp tissue phenolics could be beneficial in scavenging free radicals, maintaining membrane integrity and, thereby inhibiting pericarp browning of litchi fruit.

Abstract:DMTU treatment activated the antioxidant system in longan pericarp. The expression levels of HPCALs related to longan browning were significantly regulated by DMTU treatment. The significant expression of transcription factors to regulate longan HPCALs were observed. Longan (Dimocarpus longan) is an important subtropical evergreen fruit tree (family Sapindaceae) in southern China and exhibits a high commercial and nutritional value. Endocarp browning of longan fruit once harvested considerably reduces the commercial value. The excessive accumulation of reactive oxygen species (ROS) can induce the rapid endocarp browning of longan fruit during storage, but the ROS‐mediated mechanism remains relatively unclear. In this investigation, a hydrogen peroxide scavenger, 1,3‐dimethylthiourea (DMTU), was used to treat longan fruit to elucidate the physiological and molecular bases of the ROS‐mediated endocarp browning of longan fruit. Application of 5 mM DMTU significantly delayed endocarp browning, inhibited respiration rate, suppressed the accumulations of H2O2 and malondialdehyde (MDA), and activated the antioxidant system of longan fruit during storage, such as 3.8 of the DMTU‐treated fruit versus 1.6 of non‐DMTU‐treated fruit of the browning index, 398 versus 346 μg kg−1 S−2 of the respiration rate, and 47.5 versus 37.8 nmol g−1 of the MDA content after 6 days of storage. Furthermore, the DMTU treatment significantly affected the expression levels of the H2O2 receptors and hydrogen peroxide‐induced Ca2+ increases (HPCA)‐LIKEs (HPCALs). Additionally, the regulatory network comprising transcription factors controlling the HPCAL expression in longan was constructed. The major transcription factors, including GATA1, GATA28, TCP20, and AGL20, may contribute to the ROS‐mediated endocarp browning of longan fruit during storage by regulating the HPCAL expression levels. This study could provide new insights into the involvement of the HPCALs in the ROS‐mediated endocarp browning of longan fruit during storage.
Effect of Hot Air Treatment on the Quality and Reactive Oxygen Species Metabolism of Cold Stored Huanghua Pear Fruits

Abstract:To explore the effect of thermal treatment on the quality and reactive oxygen species(ROS) metabolism of cold stored Huanghua pear,postharvest fruit samples were treated in hot air at 40℃for 12 h before storage at 1℃for 120 d followed by at 20℃for another 7 days,and changes in fruit firmness,conductivity,MDA content and superoxide anion radical generation rate,soluble protein content and ROS scavenging enzyme activities during the whole storage period were monitored.Results showed that fruit firmness,soluble protein content,and the activities of SOD,CAT and POD in hot air treated fruits were higher than control fruits,whereas opposite results were observed for soluble solid content,electrolyte leakage,MDA content and superoxide anion radical production rate.These results indicate that hot air treatment can improve the quality of Huanghua pear fruits during storage,and the mechanism might be related to the activation of antioxidant enzymes and the reduction of ROS accumulation.
Elevated CO2 Alleviates Browning Development by Modulating Metabolisms of Membrane Lipids, Proline, and GABA in Fresh-Cut Asian Pear Fruit

Abstract:Enzymatic browning is a major problem of fresh-cut pear, which severely limits the shelf life. In the current study, elevated carbon dioxide (CO2) was applied to investigate its effects on quality maintenance and browning repression. The results showed that 5% and 10 % CO2 treatment maintained quality attributes, including firmness, color, and total soluble solids. Browning development was also inhibited by elevated CO2. Elevated CO2 delayed the decrease of unsaturated fatty acid in membranes by decreasing the activities of lipoxygenase (LOX) and phospholipase D (PLD), maintained the integrity of membrane, and therefore alleviate enzymatic browning. In addition, elevated CO2 triggered the accumulation of two stress adaptor molecules, proline and γ-aminobutyric acid (GABA), by improving their biosynthesis and repressing their degradation, which finally contributed to browning repression. However, excessive concentration (15 %) showed adverse effects because of CO2 injury. Our current study suggested a potential strategy for browning repression in fresh-cut pear fruit.
Influence of 1-MCP Treatment on the Quality and Antioxidative Properties in Cold Stored ' Huanghua' Pear Fruit

Abstract:Postharvest ' Huanghua' pear fruit was treated with 1 μL/L 1-MCP for 12h, and then stored at 1 ℃ for 120d followed by ripening at 20℃ for 7d.Fruit firmness, soluble solid content,weight loss rate,electrolyte leakage, M DA content, superoxide anion radical( O2-·)production rate,soluble protein content and activity of antioxidative enzymes were determined to investigate the effect of 1-MCP treatment on the quality and antioxidative levels in fruit.Results showed that fruit firmness,soluble protein content, and activities of SOD, POD in 1-MCP treated fruit were higher than control fruit,while soluble solid content, weight loss rate, electrolyte leakage, MDA content and O2-· production rate,CAT activity were lower.These results indicated that 1-MCP treatment improved quality of ' Huanghua' pear after storage,which might be related to the enhancement of activities of antioxidative enzymes and the reduction of active oxygen accumulation.
Elevated Oxygen Atmosphere Maintains Antioxidant Capacity and Inhibits Cell Wall Metabolism in Postharvest Jujube

Abstract:Fruit of jujube (Ziziphus jujube Mill. cv. Dongzao), known as a high-quality fresh edible variety, is popular among consumers on account of its bright color, crispy texture and abundant nutrients. However, the softening of jujube fruit after harvest seriously shortens its shelf life. To examine possible treatments to reduce softening rate, we evaluated the effect on fruit quality attributes of an elevated atmospheric O2 concentration during cold storage, ultimately unraveling its biological effect on jujube cell wall metabolism and antioxidant level. Our results demonstrated that an elevated O2 atmosphere during cold storage induced both the activity of antioxidant enzymes and relative expression levels of related genes, promoting the scavenging of hydrogen peroxide and superoxide anion. By the end of storage, the activities of superoxide dismutase, peroxidase, and catalase in elevated O2-treated jujubes were 1.9, 1.3, and 1.1 times, respectively, those in the control group. In addition, elevated O2 concentration inhibited the activities of polygalacturonase, pectin methyl esterase, cellulase and the relative abundance of related transcripts, thus delaying the degradation of cellulose and hemicelluloses and reducing the conversion of insoluble pectin to water-soluble pectin and ionic-soluble pectin. These changes presumably contribute to the maintenance of the cell wall components and cell microstructure. In conclusion, an elevated atmospheric O2 concentration of 60 % during cold storage promoted antioxidant capacity and inhibited cell wall metabolism in postharvest jujube fruit, reducing postharvest softening.

Abstract:Ozone treatment at a suitable concentration can improve the antioxidant capacity of postharvest fruits. However, few studies have examined the antioxidant bioactive compounds in ozone-treated postharvest strawberries, especially in relation to proteomics. In this study, the total phenol content (TPC), total flavonoid content (TFC), and total anthocyanin content (TAC) were used as the main antioxidant compound indicators and unlabeled proteomics was used to study the metabolism of phenylpropanoids in postharvest strawberries (Jingtaoxiang) treated with different concentrations of ozone (0, 1, 3, and 5 ppm) throughout the duration of storage. The results showed that the postharvest strawberries treated with 5 ppm ozone concentration exhibited improved accumulation of total phenols, flavonoids and anthocyanins in the antioxidant bioactive compounds, which was beneficial to the expression of phenylpropanoid metabolism-related proteins over the whole storage period compared with the other three groups. The results of proteomics were consistent with the changes in the key metabolites of phenylpropanoids, which indicated that ozone treatment at a suitable concentration aids the accumulation of TPC, TAC and TFC by promoting the key proteins associated with phenylpropanoid metabolism.

Abstract:In this study, the effects of dicyclohexylcarbodiimide (DCC) and disodium succinate (DS) treatments on the pericarp browning (PB) development in postharvest longan were elucidated by measuring PB-related phenotypes and the metabolisms of antioxidants, membrane lipids, and phenolics. Compared to the control, DCC-treated longan displayed the higher levels of PB index, CMP, ROS, PA, DAG, and SFAs, the higher activities of POD, PPO, and MLMEs including PLD, PC-PLC, PI-PLC, lipase, and LOX, and the higher expression levels of DlPPO1 , DlPOD1 , and genes encoding MLMEs. In contrast, the lower levels of chromaticity values, pigments, TP, PC, PI, and USFAs, the lower activities of RSEs including SOD, CAT, and APX, the lower expression levels of genes encoding RSEs, and a lower non-enzymatic antioxidant capacity were also noted in DCC-treated longan. These results imply that the exacerbated PB induced by DCC was associated with the activation of MLMEs by up-regulating the expression of genes encoding MLMEs, which expedited the damage of cell membrane. This membrane damage was also regulated by the accelerated accumulation of ROS triggered by DCC. The increased membrane permeability after the damage prompted an enzymatic oxidation between phenol oxidase and phenolics. However, the opposite effect occurred in the DS-treated longan.

Abstract:Longans is popular among consumers because of its rich nutritional value, but it is prone to pericarp browning as it is harvested in the high-temperature season. This work was to investigate the effects of salicylic acid (SA) on the metabolisms of reactive oxygen species (ROS) and membrane lipid and their relationship with pericarp browning of postharvest longans. 'Fuyan' longans were soaked in the solution of 0.3 g L - 1 SA for 5 min, then stockpiled at 28 °C. The results showed that, the pericarp browning index in SA-treated fruit was 42% lower than that in the control fruit at day 5 of storage. Meanwhile, SA treatment reduced the damage severity of cell membrane structure, and maintained a low level of cell membrane permeability, which was 25% lower than that in the control fruit at day 5 of storage. Additionally, compared to the control fruit, SA-treated longans displayed higher levels of pericarp L* , flavonoid, chlorophyll, carotenoid, total phenolics, and anthocyanin, higher activities of pericarp superoxide dismutase, catalase, and ascorbate peroxidase, higher values of pericarp reducing power and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, and higher amounts of pericarp ascorbic acid and glutathione, but lower levels of pericarp ROS and malondialdehyde. Moreover, SA-treated samples exhibited lower activities of pericarp phospholipase, lipase, and lipoxygenase, lower levels of pericarp phosphatidic acid and saturated fatty acids (SFA), but higher values of pericarp phosphatidylcholine, phosphatidylinositol, and unsaturated fatty acids (USFA), higher index of USFA, and higher ratio of USFA to SFA. These findings corroborated that SA delayed-browning occurrence in the pericarp of fresh longans was because SA enhanced the capacity to remove ROS, and reduced ROS accumulation and the membrane lipid degradation, thus maintaining the cellular membrane structure, reducing the degradation of pigments, and allowing the fresh longans to maintain a better appearance. Consequently, SA could be an effective means for suppressing pericarp browning and improving the commercial value of fresh longans.

Abstract:Rapid pericarp browning is the leading factor for quality deterioration and marketing decline of litchi fruit. In the present study, litchi fruit was treated with phenolic compounds, proanthocyanidin, p-coumalic acid, chlorogenic acid, apple polyphenol, and tea polyphenol, before being stored at 20°C for 10 d. Phenolic compound treatment delayed pericarp browning and weight loss, reduced respiration rate, and maintained high total acid levels and vitamin C content in litchi fruit. Subsequently, the key antioxidant characteristics were determined to evaluate the effects of phenolic compounds on the antioxidant system during pericarp browning. The phenolic compound-treated fruit exhibited higher levels of antioxidant enzymes, anthocyanins, total phenols, flavonoids, and antioxidant capacity, as well as a lower superoxide radical production rate and polyphenol oxidase and peroxide activities during whole-fruit storage than the control. Of the five treatments, proanthocyanidin application produced the most significant effects; therefore, this may be the best method for inducing antioxidant system-related factors, enhancing antioxidant capacity, and delaying pericarp browning in litchi.

Antioxidant activity of thymol essential oil and inhibition of polyphenol oxidase enzyme: a case study on the enzymatic browning of harvested longan fruit

Abstract:Abstract Background Enzymatic browning and microbial decay are the primary concerns that limit the postharvest life of longan fruit. These factors can be effectively prevented by sulfur dioxide (SO 2 ) fumigation; however, due to the safety and regulatory issues of SO 2 , other alternatives must be tested. In this study, antioxidant and antimicrobial activities of thymol were determined against the pericarp browning and decay of longan fruit. A simple, cost-effective method was designed for its controlled release. Thymol vapors were obtained from the slurry prepared from 5 g of thymol in 5 mL of distilled water in a 180-mL glass jar, hermetically sealed and allowed for 24 h to accumulate the vapors in the headspace. Fruits were packed in polyethylene packages and fumigated with thymol through a septum. Non-fumigated fruits served as control and all the packages were stored at 25 ± 2 °C for 8 days. Results Thymol significantly ( P ≤ 0.05) retarded pericarp browning (BI), delayed the decay incidence (DI) and maintained high color values of longan pericarp. Thymol also retained high total phenolic (TPC) and total flavonoid (TFC) contents, inhibited polyphenol oxidase (PPO) and peroxidase (POD) activities than those in control. A high coefficient of correlation of PPO with BI ( r = 0.86), L* ( r = − 0.94), weight loss ( r = 0.93), TPC ( r = − 0.77), TFC ( r = − 0.80), DI ( r = 0.92) and many other quality and color parameters indicated the antioxidant efficacy of thymol. Longer shelf life of 8 days with good quality attributes was obtained in thymol-treated fruits than 6 days in control. Conclusion Thymol could be effectively used as a natural antioxidant for a wide range of fruits. Graphic abstract

Abstract:Litchi fruit contains abundant polyphenols and is susceptible to browning after harvest. Herein the combined treatments of malic acid (MA) and lycopene (LYC) to delay the development of browning in litchi fruit stored at room temperature (25°C) and low temperature (4°C) was investigated. The results showed that the pericarp browning could be alleviated, and the increase of malondialdehyde (MDA) content and relative leakage rate was retarded by the combined MA and LYC during storage. As compared to control, the content of pericarp anthocyanins, flavonoids, and the total phenols maintained higher levels; and the decrease of antioxidant activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity and reducing power were slowed down in treated fruit. The enzyme activity of polyphenol oxidase (PPO) and peroxidase (POD) related to oxidation of polyphenols were depressed by the combined treatments. Furthermore, correlation analysis revealed that the content of phenols in the pericarp negatively affected the changes in the browning index, and was positively related to the DPPH radical scavenging capacity. Taken together, the combined treatments of MA and LYC exhibited potential effects in delaying the pericarp browning of litchi fruit by maintaining the content of polyphenols, antioxidant activity, and membrane integrity.

Bamboo leaf extract treatment alleviates the surface browning of fresh-cut apple by regulating membrane lipid metabolism and antioxidant properties

Abstract:Background: The effect of bamboo leaf extract (BLE) on controlling the browning of fresh-cut apple stored at 4 °C was investigated. Browning index, H2 O2 content, O2 - production rate, malondialdehyde (MDA) contents, total phenolic content (TPC) and soluble quinone content (SQC), the activities of polyphenol oxidase (PPO), peroxidase (POD), lipoxygenase (LOX), superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), DPPH (2,2-diphenyl-2-picryl-hydrazyl) and ABTS [2,2-azinobis(3-ethylbenzothiazoline- 6-sulfonic acid)] radical scavenging activities, and the expression of genes related to browning were all investigated. Results: BLE effectively alleviated the surface browning of fresh-cut apple, accompanied by a reduction in SQC, LOX activity, H2 O2 , O2 - production rate and MDA accumulation. Furthermore, BLE treatment enhanced the TPC, enzymatic (SOD, CAT, APX and POD) and non-enzymatic antioxidant activities. Principal component analysis and Pearson correlation analysis found the browning inhibition by BLE is not through the reduction of phenolic substrates and PPO activity. Conclusion: BLE controls the browning of fresh-cut apple by increasing the antioxidant capacity to scavenge ROS, which could alleviate oxidative damage and maintain the membrane integrity. © 2023 Society of Chemical Industry.
Transient production of H2O2 and NO induced by ascorbic acid coincides with promotion of antioxidant enzyme activity and reduction of pericarp browning of harvested longan fruit

Abstract:Ascorbic acid (AsA) plays an important role in the protection against oxidative stress in plants. Although the underlining mechanism is yet to be fully elucidated, it is believed that hydrogen peroxide (H2O2) and nitric oxide (NO) play a key role as a signaling agent. This study investigated the relationship between exogenously applied AsA and the activation of antioxidant defense through these signaling molecules, which coincided with reduced pericarp browning of harvested longan (Dimocarpus longan Lour. cv. Daw). It was found that the level of H2O2 in longan pericarp increased immediately after treatment with 2.5 mM AsA, reaching maximum 9 h afterward. The expression of nicotinamide adenine dinucleotide phosphate oxidase (NOX) and the activity of NOX simultaneously peaked after the AsA treatment and remained so for the next 9 h. NO content, on the other hand, did not rise until H2O2 peak was reached, attaining the highest level 3 h later. The change in NO content correlated well with the increase in nitric oxide synthase (NOS) gene expression and enzyme activity. Interestingly, the AsA treatment differently altered gene expression and activities of some vital antioxidant enzymes including superoxide dismutase, catalase, ascorbate peroxidase and glutathione peroxidase during the first 4 d of storage. With the exception of catalase, the treatment positively affected both the expression and the activity. Catalase activity remained constant for the first 9 h before rising afterward. These changes in the activity of the antioxidant enzymes were closely associated with the reduction in pericarp browning. These results suggested that H2O2 and NO generation triggered by AsA may activate the antioxidant defense mechanism in longan pericarp. This, in turn, helps overcome subsequent stressed-induced H2O2 and NO production, thereby reducing the pericarp browning.

Abstract:The plant resistance elicitor Benzo (1,2,3)-thiadiazole-7-carbothioic acid S-methyl ester (BTH) can enhance disease resistance of harvested fruit. Nonetheless, it is still unknown whether BTH plays a role in regulating fruit senescence. In this study, exogenous BTH treatment efficiently delayed the senescence of postharvest pitaya fruit with lower lipid peroxidation level. Furthermore, BTH-treated fruit exhibited lower hydrogen peroxide (H2O2) content, higher contents of reduced ascorbic acid (AsA) and reduced glutathione (GSH) levels and higher ratios of reduced to oxidized glutathione (GSH/GSSG) and ascorbic acid (AsA/DHA), as well as higher activities of ROS scavenging enzymes, including superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), peroxidase (POD) and glutathione reductase (GR) in comparison with control fruit. Moreover, BTH treatment enhanced the activities of phenylpropanoid pathway-related enzymes, including cinnamate-4-hydroxylase (C4H), phenylalanine ammonia-lyase (PAL) and 4-coumarate/coenzyme A ligase (4CL) and the levels of phenolics, flavonoids and lignin. In addition, BTH treatment upregulated the expression of HuSOD1/3/4, HuCAT2, HuAPX1/2 and HuPOD1/2/4 genes. These results suggested that application of BTH delayed the senescence of harvested pitaya fruit in relation to enhanced antioxidant system and phenylpropanoid pathway.

Abstract:Background: Zinc (Zn) fertilization has been reported to improve the quality and storability of many fruits, but the mechanism had not been systematically studied. In this study, the effect of preharvest 0.2% zinc sulfate (ZnSO4 ) spray on the storability of longan fruits was investigated. Results: The preharvest ZnSO4 spray did not significantly influence the quality but increased the Zn content in longan pericarp by 12.5-fold. More importantly, the treatment significantly reduced the rotting rate, pericarp browning, and aril breakdown of longan fruits stored at room temperature and low temperature. Physiological and biochemical results indicated that the treatment resulted in higher water retention capacity and inhibited the degradation of cellulose, pectin, flavonoid, and phenolics of longan pericarp at the late stages of storage. Consistent with these results, the activity of cellulase, polygalacturonase, polyphenol oxidase, and lipoxygenase was significantly inhibited in the ZnSO4 -treated longan pericarp at the late stages of storage. Conclusion: Higher Zn content in the ZnSO4 -treated longan pericarp might help to enhance the resistance against microbial infection, inhibit the hydrolysis of cell wall components, and thus effectively protected the cell wall components, maintained the cellular compartmentation, and prevented the phenolics and flavonoid from degradation by browning-related enzymes. © 2018 Society of Chemical Industry.
Preparation of Vanillin-Taurine Antioxidant Compound, Characterization, and Evaluation for Improving the Post-Harvest Quality of Litchi

Abstract:Litchi's post-harvest pericarp browning is one of the main constraints that drastically affect its visual attributes and market potential. Therefore, the vanillin-taurine Schiff base (VTSB) compound prepared from natural compounds of vanillin and taurine exhibited higher DPPH-radical-scavenging invitro antioxidant activity than vanillin. VTSB first-time report to mitigate the postharvest browning of litchi fruit. In this study, litchi fruits were dipped in 0.3 mM (based on pre-experiment) VTSB solution and stored at 25 ± 1 °C for six days to examine their effects on browning and postharvest quality. Fruit treated with VTSB had lower levels of browning degree (BD), browning index (BI), weight loss, soluble quinone (SQ), relative electrolyte leakage (REL), and malondialdehyde (MDA) than control fruit. Additionally, total anthocyanins and phenolic concentrations, Total soluble solids (TSS), and 2,2-diphenyl-1-picrylhydrazyl-free radical scavenging activity (DPPH-RSA) were preserved higher in VTSB-treated litchi fruit. The levels of Ascorbate peroxidase (APX), Superoxide dismutase (SOD), and Catalase (CAT) were higher in treated fruit, whereas polyphenol oxidase (PPO) and Peroxidase (POD) were decreased during the postharvest period. This study suggested that VTSB would be very useful for different post-harvest problems in the fruit and vegetable industry.

Abstract:An excessive ROS accumulation in harvested longan (Dimocarpus longan Lour.) fruit could promote the pulp breakdown and lower the fruit quality. To solve this problem, the influence of propyl gallate, one kind of ROS scavenger, on the ROS metabolism in pulp of postharvest longans and its connection with the breakdown occurrence of longan pulp were investigated. Contrasted to control samples, the propyl gallate-treated group manifested a lower index of pulp breakdown, MDA content, and production rate of O2−., but higher gene expression levels of DlSOD, DlCAT and DlAPX, higher SOD, CAT and APX activities, higher levels of AsA, flavonoid, GSH and total phenolics, and higher ability of scavenging free radicals. These results revealed that propyl gallate inhibited longan pulp breakdown occurrence, which was because propyl gallate enhanced the ROS scavenging ability and reduced ROS generation and accumulation in longan pulp. These activities involved alleviating the oxidative damage of cell membrane and the peroxidation of membrane lipid in longan pulp, which maintained the structural integrity of longan pulp cell membrane and suppressed the pulp breakdown occurrence in postharvest longan fruit.
