Abstract:Enzymatic browning is a major problem of fresh-cut pear, which severely limits the shelf life. In the current study, elevated carbon dioxide (CO2) was applied to investigate its effects on quality maintenance and browning repression. The results showed that 5% and 10 % CO2 treatment maintained quality attributes, including firmness, color, and total soluble solids. Browning development was also inhibited by elevated CO2. Elevated CO2 delayed the decrease of unsaturated fatty acid in membranes by decreasing the activities of lipoxygenase (LOX) and phospholipase D (PLD), maintained the integrity of membrane, and therefore alleviate enzymatic browning. In addition, elevated CO2 triggered the accumulation of two stress adaptor molecules, proline and γ-aminobutyric acid (GABA), by improving their biosynthesis and repressing their degradation, which finally contributed to browning repression. However, excessive concentration (15 %) showed adverse effects because of CO2 injury. Our current study suggested a potential strategy for browning repression in fresh-cut pear fruit.

Abstract:Enzymatic browning is a crucial reaction affecting the quality of fresh-cut fruit and vegetables. Purslane is an edible Chinese folk medicine with extensive distribution and containing a lot of polyphenols and alkaloids. However, little research on its' anti-browning effect on fresh-cut food was reported. In this study, the effectiveness of 0.05% (w/w) purslane aqueous extract treatment efficiently inhibited the activities of polyphenol oxidase (PPO), peroxidase (POD) and phenylalanine ammonia-lyase (PAL), the membrane integrity was effectively maintained, and malondialdehyde (MDA) content increases was retarded during whole storage period at 4 degrees C. Oddly, the higher purslane extract concentration, the lower endogenesis phenolic content. Additionally, thirty polyphenols and fifty-six alkaloids were found in purslane aqueous extract by LC-MS/MS. All results suggest that purslane aqueous extract is a promising nutritive anti-browning agent for fresh-cut potato.
Effect of High Carbon Dioxide Treatment on Reactive Oxygen Species Accumulation and Antioxidant Capacity in Fresh-Cut Pear Fruit During Storage

Abstract:Fresh-cut pear are popular for their nutritive, fresh and convenience. However, the oxidative stress caused by wounding aggravates quality loss. In the present study, the effects of 10 % CO2 on reactive oxygen species (ROS) metabolism and antioxidant capacity in fresh-cut pear were investigated. The results showed that 10 % CO2 effectively inhibited ROS accumulation during a 5-day storage, accompanied by reducing the content of hydrogen peroxide (H2O2) and superoxide anion radical (O2−). Furthermore, non-enzymatic antioxidant properties including free radical scavenging ability (DPPH), total antioxidant capacity (ABTS), and ferrous reducing ability (FRAP) in CO2-treated group were increased by 5.5 %, 23 % and 27 % respectively at the end of storage. Additionally, 10 % CO2 treatment enhanced the activities and genes expression levels of catalase (CAT), glutathione peroxidase (GPX), ascorbate peroxidase (APX), glutathione reductase (GR), superoxide dismutase (SOD) and peroxidase (POD), while NADPH oxidase (NOX) were inhibited. The accumulation of ascorbic acid (AsA) and reduced glutathione (GSH) were also accelerated by CO2. This study indicated that the generation of ROS was inhibited with inducing effects of genes and enzymes related to antioxidant capacity in response to 10 % CO2, which eventually alleviated oxidative damage and conserved storage quality of fresh-cut pear.
Evaluation of the Antioxidant Properties of Litchi Fruit Phenolics in Relation to Pericarp Browning Prevention.

Abstract:Phenolics were extracted from litchi fruit pericarp (LFP) tissues, purified and their antioxidant properties analyzed. LFP phenolics strongly inhibited linoleic acid oxidation and exhibited a dose-dependent free-radical scavenging activity against α,α-diphenyl-β-picrylhydrazyl (DPPH·) and hydroxyl radicals and superoxide anions. The degradation of deoxyribose by hydroxyl radicals was inhibited by phenolics acting mainly as iron ion chelators, rather than by directly scavenging the radicals. Phenolics from litchi fruit pericarp were found to display similar reducing power activity as ascorbic acid. The effect of phenolic compound treatment on pericarp browning and electrolyte leakage of litchi fruit was also evaluated and it was observed that application of exogenous litchi phenolics to harvested litchi fruit significantly prevented pericarp browning and delayed increases in electrolyte leakage. These results suggest that litchi pericarp tissue phenolics could be beneficial in scavenging free radicals, maintaining membrane integrity and, thereby inhibiting pericarp browning of litchi fruit.
Effect of Hot Air Treatment on the Quality and Reactive Oxygen Species Metabolism of Cold Stored Huanghua Pear Fruits

Abstract:To explore the effect of thermal treatment on the quality and reactive oxygen species(ROS) metabolism of cold stored Huanghua pear,postharvest fruit samples were treated in hot air at 40℃for 12 h before storage at 1℃for 120 d followed by at 20℃for another 7 days,and changes in fruit firmness,conductivity,MDA content and superoxide anion radical generation rate,soluble protein content and ROS scavenging enzyme activities during the whole storage period were monitored.Results showed that fruit firmness,soluble protein content,and the activities of SOD,CAT and POD in hot air treated fruits were higher than control fruits,whereas opposite results were observed for soluble solid content,electrolyte leakage,MDA content and superoxide anion radical production rate.These results indicate that hot air treatment can improve the quality of Huanghua pear fruits during storage,and the mechanism might be related to the activation of antioxidant enzymes and the reduction of ROS accumulation.

Abstract:Postharvest ' Huanghua' pear fruit was treated with 1 μL/L 1-MCP for 12h, and then stored at 1 ℃ for 120d followed by ripening at 20℃ for 7d.Fruit firmness, soluble solid content,weight loss rate,electrolyte leakage, M DA content, superoxide anion radical( O2-·)production rate,soluble protein content and activity of antioxidative enzymes were determined to investigate the effect of 1-MCP treatment on the quality and antioxidative levels in fruit.Results showed that fruit firmness,soluble protein content, and activities of SOD, POD in 1-MCP treated fruit were higher than control fruit,while soluble solid content, weight loss rate, electrolyte leakage, MDA content and O2-· production rate,CAT activity were lower.These results indicated that 1-MCP treatment improved quality of ' Huanghua' pear after storage,which might be related to the enhancement of activities of antioxidative enzymes and the reduction of active oxygen accumulation.

Abstract:Browning and lignification often occur in fresh-cut apple processing, leading to quality deterioration and limiting the shelf life of products. In this study, 0.8% (v/v) phytic acid was used to improve the quality and shelf life of fresh-cut apples. From the results, the browning was inhibited by the phytic acid treatment and the browning index (BI) of the control fruit was 1.62 times that of phytic acid treatment at 2 d of storage. The lignin content in phytic acid-treated fruit significantly decreased at 2, 4, and 6 d of storage compared to the control. Phytic acid treatment also reduced H2O2 and malonaldehyde (MDA) contents, which may indicate lighter membrane damage to apples. Compared with the control, the polyphenol oxidase (PPO) and peroxidase (POD) activities decreased while superoxide dismutase (SOD) and catalase (CAT) activities increased in phytic acid-treated fruit. Consistent with the lignin content, the activities of phenylpropane metabolism-related enzymes phenylalanine ammonia-lyase (PAL), cinnamate 4-hydroxylase (C4H), and 4-coumarate: CoA ligase (4CL) were inhibited by phytic acid treatment. In conclusion, phytic acid alleviated the browning and lignification of fresh-cut apples by reducing PPO and POD activities, maintaining cell membrane integrity, and inhibiting phenylpropane metabolism.

Abstract:Enzymatic browning is the main cause of quality loss in minimally processed fresh-cut apples and this study was aimed to control it by evaluating the use of a treatment with α-lipoic acid and ascorbic acid on Golden Delicious apples. Also hexanal, an effective natural enhancer of apple odor, was added to the treatment. At the end of the preservation period, colorimetric analysis demonstrated that α-lipoic acid exerts an anti-browning effect comparable with that of ascorbic acid about 60 times more concentrated and, moreover, chemical assays demonstrated its synergistic interaction with this substance. The application of the “antibrowning mix” does not substantially change the volatile profile of the apple slices while the addition of hexanal (0.076 mmol per liter of air) improves it primarily with an increased production of hexyl acetate (about 86% of the average aromatic component), without modifying the fruit respiration.

Abstract:Longans is popular among consumers because of its rich nutritional value, but it is prone to pericarp browning as it is harvested in the high-temperature season. This work was to investigate the effects of salicylic acid (SA) on the metabolisms of reactive oxygen species (ROS) and membrane lipid and their relationship with pericarp browning of postharvest longans. 'Fuyan' longans were soaked in the solution of 0.3 g L - 1 SA for 5 min, then stockpiled at 28 °C. The results showed that, the pericarp browning index in SA-treated fruit was 42% lower than that in the control fruit at day 5 of storage. Meanwhile, SA treatment reduced the damage severity of cell membrane structure, and maintained a low level of cell membrane permeability, which was 25% lower than that in the control fruit at day 5 of storage. Additionally, compared to the control fruit, SA-treated longans displayed higher levels of pericarp L* , flavonoid, chlorophyll, carotenoid, total phenolics, and anthocyanin, higher activities of pericarp superoxide dismutase, catalase, and ascorbate peroxidase, higher values of pericarp reducing power and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, and higher amounts of pericarp ascorbic acid and glutathione, but lower levels of pericarp ROS and malondialdehyde. Moreover, SA-treated samples exhibited lower activities of pericarp phospholipase, lipase, and lipoxygenase, lower levels of pericarp phosphatidic acid and saturated fatty acids (SFA), but higher values of pericarp phosphatidylcholine, phosphatidylinositol, and unsaturated fatty acids (USFA), higher index of USFA, and higher ratio of USFA to SFA. These findings corroborated that SA delayed-browning occurrence in the pericarp of fresh longans was because SA enhanced the capacity to remove ROS, and reduced ROS accumulation and the membrane lipid degradation, thus maintaining the cellular membrane structure, reducing the degradation of pigments, and allowing the fresh longans to maintain a better appearance. Consequently, SA could be an effective means for suppressing pericarp browning and improving the commercial value of fresh longans.

Abstract:Litchi fruit contains abundant polyphenols and is susceptible to browning after harvest. Herein the combined treatments of malic acid (MA) and lycopene (LYC) to delay the development of browning in litchi fruit stored at room temperature (25°C) and low temperature (4°C) was investigated. The results showed that the pericarp browning could be alleviated, and the increase of malondialdehyde (MDA) content and relative leakage rate was retarded by the combined MA and LYC during storage. As compared to control, the content of pericarp anthocyanins, flavonoids, and the total phenols maintained higher levels; and the decrease of antioxidant activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity and reducing power were slowed down in treated fruit. The enzyme activity of polyphenol oxidase (PPO) and peroxidase (POD) related to oxidation of polyphenols were depressed by the combined treatments. Furthermore, correlation analysis revealed that the content of phenols in the pericarp negatively affected the changes in the browning index, and was positively related to the DPPH radical scavenging capacity. Taken together, the combined treatments of MA and LYC exhibited potential effects in delaying the pericarp browning of litchi fruit by maintaining the content of polyphenols, antioxidant activity, and membrane integrity.

Abstract:Rapid pericarp browning is the leading factor for quality deterioration and marketing decline of litchi fruit. In the present study, litchi fruit was treated with phenolic compounds, proanthocyanidin, p-coumalic acid, chlorogenic acid, apple polyphenol, and tea polyphenol, before being stored at 20°C for 10 d. Phenolic compound treatment delayed pericarp browning and weight loss, reduced respiration rate, and maintained high total acid levels and vitamin C content in litchi fruit. Subsequently, the key antioxidant characteristics were determined to evaluate the effects of phenolic compounds on the antioxidant system during pericarp browning. The phenolic compound-treated fruit exhibited higher levels of antioxidant enzymes, anthocyanins, total phenols, flavonoids, and antioxidant capacity, as well as a lower superoxide radical production rate and polyphenol oxidase and peroxide activities during whole-fruit storage than the control. Of the five treatments, proanthocyanidin application produced the most significant effects; therefore, this may be the best method for inducing antioxidant system-related factors, enhancing antioxidant capacity, and delaying pericarp browning in litchi.

Abstract:The performance of p-Anisaldehyde (PAA) for preserving pitaya fruit quality and the underpinning regulatory mechanism were investigated in this study. Results showed that PAA treatment significantly reduced fruit decay, weight loss and loss of firmness, and maintained higher content of total soluble solids, betacyanins, betaxanthins, total phenolics and flavonoids in postharvest pitaya fruits. Compared with control, the increase in hydrogen peroxide (H2O2) content and superoxide anion (O2•−) production was inhibited in fruit treated with PAA. Meanwhile, PAA significantly improved the activity of antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT). Moreover, PAA-treated pitaya fruit maintained higher ascorbic acid (AsA) and reduced-glutathione (GSH) content but lower dehydroascorbate (DHA) and oxidized glutathione (GSSG) content, thus sustaining higher ratio of AsA/DHA and GSH/GSSG. In addition, activities of ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR) and dehydrogenation ascorbic acid reductase (DHAR), as well as the expression of HpSOD, HpPOD, HpCAT, HpAPX, HpGR, HpDHAR and HpMDHAR, were enhanced after PAA treatment. The findings suggest that postharvest application of PAA may be a reliable method to control postharvest decay and preserve quality of harvested pitaya fruit by enhancing the antioxidant potential of the AsA-GSH cycle and activating an antioxidant defense system to alleviate reactive oxygen species (ROS) accumulation.

Effect of Exogenous Oxalic Acid Treatment on Ripening and Preservation of Annona squamosa L. Fruits during Postharvest Storage

Abstract:The objective of this study is to elucidate the effect of exogenous oxalic acid on ripening,softening and preservation process of postharvested sugar apple(Annona squamosa L.) and provide a new method to preserve sugar apple fruits.After being dipped in 5 mmol/L oxalic acid solution for 20 min,sugar apple fruits(Annona squamosa L.cv.African Pride) were stored at 15 ℃,and the physiological parameters related to delaying ripening,softening and cracking process were measured.The results showed that oxalic acid treatment decreased the softening rate and delayed the increase of total soluble solids(TSS).Compared with the control,the cracking rate of the treatment group was reduced and the activities of both pectin methylesterase(PE) and polygalacturonase(PG) were significantly inhibited.Oxalic acid treatment could promote the activities of superoxide dismutase(SOD) and peroxidase(POD) at the later period of storage,and decrease the loss of soluble protein and vitamin C significantly.In addition,oxalic acid treatment could also decrease polyphenol oxidase(PPO) activity and malondialdehyde(MDA) content.Correlation analysis showed that the physiological parameters of PE activity and PG activity were significantly correlated with the softening and cracking process.Therefore,exogenous oxalic acid treatment can delay the ripening and senescence rate of sugar apple fruits.

Abstract:'Zaosu' pear fruit is prone to yellowing of the surface and softening of the flesh after harvest. This work was performed to assess the influences of L-glutamate treatment on the quality of 'Zaosu' pears and elucidate the underlying mechanisms involved. Results demonstrated that L-glutamate immersion reduced ethylene release, respiratory intensity, weight loss, brightness ( L* ), redness ( a* ), yellowness ( b* ), and total coloration difference (ΔE); enhanced ascorbic acid, soluble solids, and soluble sugar contents; maintained chlorophyll content and flesh firmness of pears. L-glutamate also restrained the activities of neutral invertase and acid invertase, while enhancing sucrose phosphate synthetase and sucrose synthase activities to facilitate sucrose accumulation. The transcriptions of PbSGR1 , PbSGR2 , PbCHL , PbPPH , PbRCCR , and PbNYC were suppressed by L-glutamate, resulting in a deceleration of chlorophyll degradation. L-glutamate concurrently suppressed the transcription levels and enzymatic activities of polygalacturonases, pectin methylesterases, cellulase, and β-glucosidase. It restrained polygalacturonic acid trans-eliminase and pectin methyl-trans-eliminase activities as well as inhibited the transcription levels of PbPL and Pbβ-gal . Moreover, the gene transcriptions and enzymatic activities of arginine decarboxylase, ornithine decarboxylase, S-adenosine methionine decarboxylase, glutamate decarboxylase, γ-aminobutyric acid transaminase, glutamine synthetase along with the PbSPDS transcription was promoted by L-glutamate. L-glutamate also resulted in the down-regulation of PbPAO , PbDAO , PbSSADH , PbGDH , and PbGOGAT transcription levels, while enhancing γ-aminobutyric acid, glutamate, and pyruvate acid contents in pears. These findings suggest that L-glutamate immersion can effectively maintain the storage quality of 'Zaosu' pears via modulating key enzyme activities and gene transcriptions involved in sucrose, chlorophyll, cell wall, and polyamine metabolism.

The results showed that 1% L -cysteine pre-cut treatment for 15 min could significantly inhibit the browning and extend the shelf life of fresh-cut apple slices. Pre-cut L -cysteine treatment was found to significantly inhibit polyphenol oxidase (PPO) activity and its gene expression, and increased superoxide dismutase (SOD) and catalase (CAT) activity as well as DPPH radical scavenging rate. In addition, pre-cut L -cysteine treatment improved the content of total and individual phenolics, particularly the content of chlorogenic acid and epicatechin in pulp. Pre-cut L -cysteine treatment also increased the cysteine, glutamic acid, serine and leucine level in pulp and peel. Therefore, pre-cut L -cysteine treatment is an effective method to control the browning of fresh-cut apple by inhibiting PPO activity, improving antioxidant ability and regulating the synthesis of phenolics and free amino acids.

Abstract:This study investigated the effect of acidic electrolyzed water (AEW) on the inhibition of pulp breakdown of postharvest longan fruit. The harvested longan fruit were immersed in AEW (pH 2.5 and available chlorine concentration 80 mg L −1 ) for 10 min, then air-dried and stored at 25 °C. It was revealed that the AEW treatment reduced the cell membrane permeability and breakdown incidence of pulp by modulating membrane lipid metabolism. Compared to the untreated fruit, AEW treatment significantly suppressed the activities of lipoxygenase (LOX), lipase, and phospholipase D (PLD), down-regulated the expression of DlPLD1, DlPLD2, Dllipase1, Dllipase2, DlLOX1 and DlLOX2 , and reduced the hydrolysis of phosphatidylinositol (PI) and phosphatidylcholine (PC) and the accumulation of phosphatidic acid (PA). In addition, a lower saturated fatty acids (SFAs) proportion, but a higher unsaturated fatty acids (USFAs) proportion, the index of unsaturated fatty (IUFA), and unsaturated to saturated fatty acids ratio (U/S) were found in AEW-treated longan fruit. These findings indicated that AEW treatment inhibited the activities of membrane lipid metabolism-related enzymes (LOX, lipase, and PLD) and their expressions of associated genes, and altered the cell membrane lipid composition, which was conductive to maintaining the integrity of cell membrane, ultimately alleviating the progress of pulp breakdown in harvested longan fruit. Consequently, AEW treatment may be a reliable method to reduce the occurrence of pulp breakdown and improve the storability of harvested longan fruit.

Abstract:Background: The effect of bamboo leaf extract (BLE) on controlling the browning of fresh-cut apple stored at 4 °C was investigated. Browning index, H2 O2 content, O2 - production rate, malondialdehyde (MDA) contents, total phenolic content (TPC) and soluble quinone content (SQC), the activities of polyphenol oxidase (PPO), peroxidase (POD), lipoxygenase (LOX), superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX), DPPH (2,2-diphenyl-2-picryl-hydrazyl) and ABTS [2,2-azinobis(3-ethylbenzothiazoline- 6-sulfonic acid)] radical scavenging activities, and the expression of genes related to browning were all investigated. Results: BLE effectively alleviated the surface browning of fresh-cut apple, accompanied by a reduction in SQC, LOX activity, H2 O2 , O2 - production rate and MDA accumulation. Furthermore, BLE treatment enhanced the TPC, enzymatic (SOD, CAT, APX and POD) and non-enzymatic antioxidant activities. Principal component analysis and Pearson correlation analysis found the browning inhibition by BLE is not through the reduction of phenolic substrates and PPO activity. Conclusion: BLE controls the browning of fresh-cut apple by increasing the antioxidant capacity to scavenge ROS, which could alleviate oxidative damage and maintain the membrane integrity. © 2023 Society of Chemical Industry.

Abstract:DMTU treatment activated the antioxidant system in longan pericarp. The expression levels of HPCALs related to longan browning were significantly regulated by DMTU treatment. The significant expression of transcription factors to regulate longan HPCALs were observed. Longan (Dimocarpus longan) is an important subtropical evergreen fruit tree (family Sapindaceae) in southern China and exhibits a high commercial and nutritional value. Endocarp browning of longan fruit once harvested considerably reduces the commercial value. The excessive accumulation of reactive oxygen species (ROS) can induce the rapid endocarp browning of longan fruit during storage, but the ROS‐mediated mechanism remains relatively unclear. In this investigation, a hydrogen peroxide scavenger, 1,3‐dimethylthiourea (DMTU), was used to treat longan fruit to elucidate the physiological and molecular bases of the ROS‐mediated endocarp browning of longan fruit. Application of 5 mM DMTU significantly delayed endocarp browning, inhibited respiration rate, suppressed the accumulations of H2O2 and malondialdehyde (MDA), and activated the antioxidant system of longan fruit during storage, such as 3.8 of the DMTU‐treated fruit versus 1.6 of non‐DMTU‐treated fruit of the browning index, 398 versus 346 μg kg−1 S−2 of the respiration rate, and 47.5 versus 37.8 nmol g−1 of the MDA content after 6 days of storage. Furthermore, the DMTU treatment significantly affected the expression levels of the H2O2 receptors and hydrogen peroxide‐induced Ca2+ increases (HPCA)‐LIKEs (HPCALs). Additionally, the regulatory network comprising transcription factors controlling the HPCAL expression in longan was constructed. The major transcription factors, including GATA1, GATA28, TCP20, and AGL20, may contribute to the ROS‐mediated endocarp browning of longan fruit during storage by regulating the HPCAL expression levels. This study could provide new insights into the involvement of the HPCALs in the ROS‐mediated endocarp browning of longan fruit during storage.
