Abstract:Background Nuoshanyao (NSY) is a special species of Chinese yam ( Dioscorea opposita Thunb.) and shows rich functionality and superior nutritional value. The chemical components and structural properties of Nuoshanyao polysaccharide (NSYP) were investigated in his study. Results The molecular weight (Mw) of the major component in the polysaccharide was 23 kDa. NSYP was mainly composed of glucose, mannose, and galactose, and the molar ratio was 36.5:1.63:1. There might be an ordered helical conformation in NSYP, and the surface of NSYP was granular. NSYP exhibited strong scavenging capacity on the DPPH radical and chelating capacity on the ferrous ion in vitro with a rate of 94.94% and 82.49% at 4 mg/mL. Conclusion NSYP with the appropriate molecular weight range and helix structure could possess great antioxidant activity, while monosaccharide compositions may affect the activity. This work supplied important information for the potential application of NSYP in the nutritional and food industries.

Abstract:Background: Currently, balanced diet and healthy foods are attracting increasing attention due to their great health-beneficial effects. The tuber of Dioscorea (yam) is both edible and medicinal, as it contains various functional compounds with multiple pharmacological activities and disease targets. Moreover, yam tuber can serve as a vegetable or staple food due to its high starch content. Therefore, exploring the functional activities of yam tuber and its application in food and medicine industries is significant. Scope and approach: This review comprehensively summarizes the characteristics and application of yam tuber, as well as the findings about its primary nutrients, distinct functional components, extraction technologies, detection methods and biological activities, and provides some new prospects for further investigation. Besides, the development and comprehensive utilization of yam tuber products are elucidated. Finally, some future research directions for better utilization of yam tubers are suggested. Key findings and conclusions: The Dioscorea genus is distributed worldwide with a wide variety of species. Yam tuber is rich in nutrients and bioactive compounds, such as starch, protein, non-starch polysaccharides, steroid saponin, allantoin, and phenolic compounds. The most important health benefits include antioxidant activity, anti-inflammatory activity, gastrointestinal protection, gut microbiota regulation, cardiac diseases treatment, hypoglycemic activity, anti-tumor activity, and estrogen-like effect. We also summarize the applications of yam tuber in foods and medicine. Finally, the review indicates that better utilization of yam tuber is hindered by poor variety protection, limited research on its pharmacological activities, browning phenomenon, and some safety concerns.
Immunomodulatory activity and mechanism of Chinese yam polysaccharide after sulfated modification

Abstract:It is well known that yam is an important industrial crop resource. Our previous study has demonstrated that sulfated modifications (S-CYP) can improve the immunomodulatory effect of polysaccharide from Chinese yam by activating and maturating RAW264.7 cells, while its mechanism is still unknown. In this study, the underlying mechanisms of S-CYP-induced immunomodulatory effect were examined based on RNA sequencing (RNA-seq), and a co-culture system with Caco-2 cells and RAW264.7 cells. RNA-seq analysis indicated that 1764 expressed genes differentially between the control and S-CYP groups. 13 cellular component terms, 20 biological process terms, and 10 molecular function terms were identified by GO. KEGG analysis showed that MAPK signaling pathways belong to the 20 most significantly affected pathways. Furthermore, S-CYP indirectly affects the immune function of RAW264.7 in co-culture model by increasing secretion of cytokines (IL-1β, TNF-α). These results provide a theoretical basis for exploring S-CYP as an immunomodulatory agent.

Abstract:In this study, we evaluated the protective effect and molecular mechanism of a dominant phenanthrene, (6,7-dihydroxy-2,4-dimethoxyphenanthrene, CYP4), from Chinese yam peels on intestinal epithelial integrity. Three doses of Chinese yam phenolic extract (CYPE) and Chinese yam phenanthrene 4 (CYP4) were administered to BALB/c mice for 7 days before dextran sulfate sodium (DSS) treatment, with berberine hydrochloride as a positive control (PC). Results showed that both disease activity indexes (DAIs), histological damage score (HDS) and survival rate in DSS mice, were improved with preintervention of CYPE and CYP4, which exhibited better efficiency than PC. Further studies showed that administration of CYP4 downregulated the oxidative stress-associated factors, MPO and NO, and improved tight junction protein occludin. Besides, the CYP4 treatment substantially downregulated the caspase-3 expression and the apoptosis rate of intestinal epithelial cells. In addition, the CYP4 treatment ameliorated the production of inflammatory cytokines including TNF-α, IFN-γ, IL-10, and IL-23 in the colon. Furthermore, the protein expression of ERK1/2, NF-κB p65, pNF-κB, and COX-2 was suppressed in CYE4 groups as compared with that in model control (MC). These findings suggested that CHP4 could effectively inhibit the activation of NF-κB/COX-2 in an experimental UC model in vivo. It was demonstrated for the first time that CYPE and CYP4 protected intestinal mucosa from damage and prevented DSS-induced colitis in mice. CYP4 was one of the active principles obligatory for the biological effect of Chinese yam in protecting intestinal health. These findings indicated that CYP4 might be a promising and useful approach for treatment of UC in humans.

Abstract:This study aimed to characterize the structure of Chinese yam (Dioscoreae Rhizoma) polysaccharide (CYP) and to investigate its protective effect against H2O2-induced oxidative damage in IEC-6 cells. The chemical composition and structural characteristics of the samples were analyzed by chemical and instrumental methods, including high-performance gel permeation chromatography, high-performance anion-exchange chromatography (HPAEC), Fourier transformed infrared (FT-IR), ultraviolet (UV), and scanning electron microscopy (SEM). Antioxidant activity was evaluated by establishing a cellular model of oxidative damage. The molecular weight of CYP was 20.89 kDa. Analysis of the monosaccharide composition revealed that CYP was primarily comprised of galactose (Gal), glucose (Glu), and galacturonic acid (GalA), and the ratio between them was 28.57:11.28:37.59. Pretreatment with CYP was able to improve cell viability, superoxide dismutase (SOD) activity, and reduce intracellular reactive oxygen species (ROS) production and malondialdehyde (MDA) content after H2O2 injury. CYP also attenuated oxidative damage in cells through the mitogen-activated protein kinase (MAPK) signaling pathway. This study showed that CYP was an acidic heteropolysaccharide with a good protective effect against oxidative damage, and it thus has good prospects in food and biopharmaceutical industries.

Abstract:The effects of phenylbutazone and its metabolite, MI, on certain manifestations of rheumatoid arthritis were studied in 24 patients by a double-blind method. Grip strength and the duration of morning stiffness were influenced significantly by both the parent drug and its derivative. By such partially subjective criteria the metabolite MI had greater antirheumatic activity than phenylbutazone. Sedimentation rates and joint swelling (circumference) were not altered appreciably by either agent. Abstract:In the developed N2O model of this work, inorganic carbon (IC) effect on both ammonia oxidation and N2O production is explicitly considered, energy transportation is coupled to electron transfer and the catabolism and anabolism of ammonia oxidizing bacteria (AOB) are linked through ATP/ADP pool. The calibration and validation of the model are conducted using experimental data from a nitrification system. The model satisfactorily describes the N2O data under varying IC conditions. In addition, the model also predicts the impact of IC on N2O production via the AOB denitrification pathway and the hydroxylamine (NH2OH) oxidation pathway.

Abstract:Ethnopharmacological relevance Ilex rotunda Thunb. (IR) is widely used for gastrointestinal diseases by Yao physician, and it has a better clinical curative effect on ulcerative colitis (UC). However, the main active components and mechanism of IR in the treatment of UC remain to be clarified. Aim of the study To investigate the main active components and mechanism of IR in the treatment of UC. Materials and methods Ten biological active components of IR were quantified by UPLC-MS/MS. In vitro , Caco2 cell monolayers were stimulated by lipopolysaccharide, and were treated with 10 biologically active components individually to investigate the protective role of the components of IR in mucosal barrier damage. In vivo , a mouse model of UC was induced by dextran sulfate sodium and administered with the candidate active components of IR. On day 8, the serum and colon tissue were collected for histological and molecular analysis to investigate the main active components and mechanism of IR. Results Ziyuglycoside I, ziyuglycoside II, syringin , and pedunculoside in IR reduced phenol red transmission of the monolayer, and inhibited the protein expression of oncostatin M and oncostatin M receptor in Caco2 cells. Notably, ziyuglycoside II and syringin decreased the transepithelial electrical resistance of the monolayer, and promoted the protein expression of Occludin, Claudin-1 and zonula occludens-1 (ZO-1) in Caco2 cells. In vivo , ziyuglycoside II and syringin improved the symptoms of UC mice, including body weight, disease activity score, shortening of colon length, damaging of acidic mucus layer, histopathological changes, and protein expression of Occludin, Claudin-1, and ZO-1. Pedunculoside reduced the neutrophils and inflammatory response in the UC mice. Moreover, when the combination of ziyuglycoside II, syringin and pedunculoside was used for the treatment of UC, syringin and pedunculoside enhanced the therapeutic effect of ziyuglycoside II. Finally, RNA sequencing and RT-qPCR analysis revealed that ziyuglycoside II + syringin + pedunculoside and IR coregulated up to 42.7% of genes, and mainly reduced the overexpression of C-X-C motif ligand 1(CXCL1), oncostatin M receptor (OSMR), interleukin 1 receptor type I (IL1R1), tumor necrosis factor receptor superfamily member 9 (TNFRSF9), C-X-C motif chemokine 13 (CXCL13), oncostatin M (OSM), and interleukin 6 (IL-6) in the cytokine-cytokine interaction pathways. Conclusions The combination of ziyuglycoside II, syringin, and pedunculoside protects against UC by modulating the intestinal mucosal barrier and inhibiting the cytokine-cytokine interaction pathways, and the effect is relatively equivalent to that of the water extract of Ilex rotunda Thunb.

Abstract:Ulcerative colitis (UC) is an inflammatory bowel disease that is persistent and nonspecific. There are several medications available for the treatment of UC. However, conventional UC medications have substantial adverse effects, low clinical effectiveness, and a high recurrence rate. Therefore, it is critical to discover new medicines that are both safe and effective for UC patients. Natural polysaccharides offer a wide range of pharmacological benefits, including anti-inflammatory, anti-virus, anti-tumor, anti-aging, immune enhancement, and gut flora regulation. In the therapy of UC, natural polysaccharides can modulate inflammatory factors, the immune system, and intestinal flora, and preserve the intestinal mucosa. It demonstrates a good curative effect and is of safety to use, thereby being a potential treatment for UC patients. This paper covers the structure, the pharmacological effects on UC, and the mechanisms of natural polysaccharides. Finally, limitations, challenges, and perspectives are discussed. It is hoped that the findings of this publication will inspire more natural polysaccharides research and provide a theoretical foundation for the creation of new UC medications.

Abstract:Exophiala dermatitidis and Exophiala jeanselmei share similar morphological features and have been confused with each other. To clarify the relationship between the two fungi, we conducted a deoxyribonucleic acid (DNA)‐DNA hybridization study using a dot blot method. Between E. dermatitidis and E. jeanselmei, only a very low level of DNA relatedness was seen and it was confirmed that these two fungi are distinct species based on DNA similarity. Close correspondence of DNA from the isolates of E. dermatitidis was obtained, whereas the isolates of E. jeanselmei were divided into 6 groups according to their DNA similarity and a possibility was shown that E. jeanselmei is composed of genetically heterogeneous groups. The subdivision of the species E. jeanselmei by the DNA‐DNA hybridization method was in agreement with serotyping exoantigens. This result suggests that DNA‐DNA hybridization studies provide an excellent tool for the identification and grouping of pathogenic dematiaceous fungi.

Abstract:As a medicinal and edible plant, Chinese yam (CY) can promote the enrichment of intestinal probiotics. Mucilage polysaccharides, diosgenin and taxifolin are the dominant components of CY. The purpose of this study was to investigate whether the impact of Chinese yam on gut microbiome structure and metabolism is attributable to its components. In the in vitro gastrointestinal digestion and colon fermentation system, the changes in gut microbiota composition and function were determined by 16S rRNA sequencing, and the levels of bacterial metabolites including short-chain fatty acids (SCFAs) and indole-like metabolites were detected by gas chromatography and an enzyme-linked immunoassay. The results show that CY, mucilage polysaccharides, diosgenin and taxifolin could increase the microbial diversity index. Furthermore, probiotics including Lactobacillus and Bacteroides were significantly increased, while harmful bacteria such as Escherichia and Proteus declined. CY could increase the production of SCFAs including acetic acid and butyric acid. Of note, CY and diosgenin displayed similar impacts on enhancing the abundance of Clostridium and promoting the production of indole-3-lactic acid and lactic acid. These findings provide evidence supporting Chinese yam as a natural food to regulate intestinal health. Diosgenin as a component of CY contributes mostly to the impact on regulating intestinal flora.

Abstract:In this study, we investigated the antioxidant properties and the changes of molecular weight (Mw), antioxidant activity, and microstructure of Chinese yam polysaccharides (CYP-A) during in vitro digestion. Results showed that the scavenging rate of 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical of CYP-A was approximately 79% at the concentration of 6 mg/mL. Furthermore, the antioxidant ability positively correlated with the concentration of CYP-A. In addition, the Mw of CYP-A decreased (p < 0.05) after intestinal digestion. Fourier transform infrared showed that the degrees of esterification of CYP-A increased to 39.04% after 6 h of gastric digestion. Moreover, the structure of CYP-A changed after in vitro gastric digestion. The scanning electron microscope (SEM) images indicated that the surface morphology of CYP-A turned from smooth and irregular to a layered honeycomb after gastrointestinal digestion in vitro. Surprisingly, the activity of DPPH radical scavenging significantly increased (p < 0.05) in the intestinal digested samples of CYP-A, which showed a positive correlation with the concentration of CYP-A. However, the reducing power significantly decreased (p < 0.05) after in vitro intestinal digestion.

Abstract:Altered apoptosis is involved in each step of the pathogenesis of preeclampsia. While deficient apoptosis may induce a maternal immune response against the fetus, enhanced apoptosis may interfere with the process of placentation, placental ischemia and subsequently, systemic endothelial damage. Thus, treatment modalities to inhibit or acclerate apoptosis cannot be employed in early pregnancy for prevention. Later in pregnancy, it would make sense to test treatment modalities that inhibit hypoxia-mediated apoptosis in patients in whom early testing, such as abnormal Doppler of the uterine artery, indicates the initiation of preeclampsia. This, however, has first to be tested in animal models. Since multiple different mechanisms, rather than a single factor, could contribute to the development of apoptosis, further studies to clarify the signaling mechanisms of apoptosis in preeclampsia should be conducted before any investigational treatment modalities are employed.

Abstract:Intracellular polysaccharides (iPs) were separated and purified from Coriolus versicolor LH1 mycelia and characterized for their α-glucosidase inhibitory properties. Three iP fractions (iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1) were extracted, separated, and purified from LH1 mycelia using microwave extraction technology, a DEAE-Sepharose CL-6B column, a Diaion HP20 macroporous adsorption column, and a Sephadex™ G-50 gel-permeation column. The principal constituents of iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1 were saponins and polyphenoic compound mixtures. The enzyme inhibition activity, IC(50) values, of these three fractions were 1.7, 1.8, and 0.8 mg/mL, respectively. The α-glucosidase inhibitory properties were related to the presence of α-(1,4) glycosidic linkages in the polysaccharide structure and the total relative percentage of d-glucose and d-galactose in the structure of polysaccharides, other than triterpenoids.

Abstract:Herein, a β-1,3-D-glucan based yeast cell wall loaded with co-loaded nanoparticles of Rhein (RH) and Emodin (EMO), was developed for the combined treatment of ulcerative colitis (UC) by modulating gut microbiota and the Th17/Treg cell balance. This was achieved through an oral "nano-in-micro" advanced drug delivery system. Specifically, RH was grafted onto the HA chain via disulfide bonds to synthesize a reduction-sensitive carrier material and then used to encapsulate EMO to form nanoparticles with a specific drug ratio (denoted as HA-RH/EMO NPs). As anticipated, HA-RH/EMO NPs were encased within the "nests"-yeast cell wall microparticles (YPs), efficiently reach the colon and then released gradually, this occurs mainly due to the degradation of β-1,3-D-glucan by β-glucanase. Additionally, HA-RH/EMO NPs demonstrated a significant reduction-sensitive effect in GSH stimulation evaluations and a remarkable ability to target macrophages in in vitro cell uptake studies. Notably, HA-RH/EMO NYPs reduced inflammatory responses by inhibiting the PI3K/Akt signaling pathway. Even more crucially, the oral delivery and drug combination methods significantly enhanced the regulatory effects of HA-RH/EMO NYPs on gut microbiota and the Th17/Treg balance.
