Abstract:Background Nuoshanyao (NSY) is a special species of Chinese yam ( Dioscorea opposita Thunb.) and shows rich functionality and superior nutritional value. The chemical components and structural properties of Nuoshanyao polysaccharide (NSYP) were investigated in his study. Results The molecular weight (Mw) of the major component in the polysaccharide was 23 kDa. NSYP was mainly composed of glucose, mannose, and galactose, and the molar ratio was 36.5:1.63:1. There might be an ordered helical conformation in NSYP, and the surface of NSYP was granular. NSYP exhibited strong scavenging capacity on the DPPH radical and chelating capacity on the ferrous ion in vitro with a rate of 94.94% and 82.49% at 4 mg/mL. Conclusion NSYP with the appropriate molecular weight range and helix structure could possess great antioxidant activity, while monosaccharide compositions may affect the activity. This work supplied important information for the potential application of NSYP in the nutritional and food industries.
Antifatigue effects and antioxidant activity in polysaccharide fractions from Chinese yam bulbils

Abstract:Polysaccharides are the principal component in Chinese yam (Dioscorea opposita Thunb.) bulbils. The properties and antifatigue of polysaccharides from yam bulbils (PYB) were identified and compared. Their molecular weights (PYB-1 and PYB-2) were approximately 145 and 11 kDa, respectively, with active β-configurations. Meanwhile, the antifatigue activities of PYBs were tested in mice via exhaustive swimming tests (EST). The EST results indicated that PYB-1 and PYB-2 significantly prolonged swimming time in mice (p < .05). Associated with this increase was a rise in hepatic glycogen content and antioxidant enzyme (superoxide dismutase (SOD), glutathione peroxidase (GSH-Px)) activity, along with a decline in blood urea nitrogen, lactic acid, and malondialdehyde levels. The results showed that molecular weight might contribute to the antifatigue effects of PYBs. Additionally, antioxidant tests showed that PYB-1 had stronger free-radical scavenging activity than PYB-2. Taken together, the findings indicated that PYBs exhibited effective antifatigue and antioxidant activities providing additional evidence supporting the use of PYBs as functional food ingredients for relieving fatigue.

Abstract:Background: Currently, balanced diet and healthy foods are attracting increasing attention due to their great health-beneficial effects. The tuber of Dioscorea (yam) is both edible and medicinal, as it contains various functional compounds with multiple pharmacological activities and disease targets. Moreover, yam tuber can serve as a vegetable or staple food due to its high starch content. Therefore, exploring the functional activities of yam tuber and its application in food and medicine industries is significant. Scope and approach: This review comprehensively summarizes the characteristics and application of yam tuber, as well as the findings about its primary nutrients, distinct functional components, extraction technologies, detection methods and biological activities, and provides some new prospects for further investigation. Besides, the development and comprehensive utilization of yam tuber products are elucidated. Finally, some future research directions for better utilization of yam tubers are suggested. Key findings and conclusions: The Dioscorea genus is distributed worldwide with a wide variety of species. Yam tuber is rich in nutrients and bioactive compounds, such as starch, protein, non-starch polysaccharides, steroid saponin, allantoin, and phenolic compounds. The most important health benefits include antioxidant activity, anti-inflammatory activity, gastrointestinal protection, gut microbiota regulation, cardiac diseases treatment, hypoglycemic activity, anti-tumor activity, and estrogen-like effect. We also summarize the applications of yam tuber in foods and medicine. Finally, the review indicates that better utilization of yam tuber is hindered by poor variety protection, limited research on its pharmacological activities, browning phenomenon, and some safety concerns.
The Protective Effects of Chinese Yam Polysaccharide Against Obesity-Induced Insulin Resistance

Abstract:Background: Chinese yam polysaccharide (CYP), a bioactive compound of Chinese yam, has been proven to exhibit many pharmacological effects, such as anti-inflammatory, anti-oxidation and immune-modulation activities. However, little is known about its effect on modulating blood fat level and its protective effect against insulin resistance. Aim: The present study aims to evaluate the protective effect against obesity-induced insulin resistance and the anti-hyperlipidemia activity of CYP in vivo. Methods: In this study, we fed C57BL/6 mice with high fat diet for 14 consecutive weeks to create a diet-induced obesity model. To validate this model, we performed the insulin tolerance test by measuring the levels of blood glucose at different time points. Blood samples were collected by orbital vein pricking. Ingredients such as total cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) in blood plasma were detected by auto biochemistry analyzer. The levels of IL-1 beta, IL-10 and leptin were quantified by ELISA kits. The content of MMP-3 and NF-kappa B (P65) was determined by western-blot. Results: Our results showed that CYP is able to reduce body weight of mice and ameliorate insulin resistance. In addition, CYP is able to lower the level of LDL and total cholesterol significantly. Mechanically, our immunohistochemistry experiments demonstrated that CYP is able to decrease the levels of IL-1 beta, IL-10 and leptin in serum and the expression of MMP-3 and NF-kappa B (P65) in white adipose tissue. Conclusions: Our in vivo study showed that CYP is able to improve the sensitivity of insulin and exert protective effect against hyperlipidemia.

Abstract:The aims of this study are to investigate the antioxidant and antitumor activities of the water and ethanol extracts isolated from Chinese yam(Dioscorea opposite Thunb.) flesh (CYF) and peel (CYP) and the effective compounds. It was found that all peel portions have a better effect on reactive oxygen (ROS) scavenging assay than meat portions, especially for the water extract of Chinese yam peel (CYP-W). Its IC50 values for hydroxyl radical (OH center dot) scavenging assay (744.25 +/- 3.46 mu g/mL) and for 1,1-diphenyl-2-picrylhydrazyl scavenging assay (374.85 +/- 6.78 mu g/mL) were both lower than that of yam flesh (CYF-W). Furthermore, the antitumor property of yam peel was more effective than that of yam flesh (CYF-W) on mouse models, with tumor inhibition rates were 47.92% and 27.41% for Ehrlich Ascites Tumor (EAC) model and 40.44% and 24.22% for H22 hepatocarcinoma tumor (H22) model. Meanwhile, extracts of peel showed higher allantoin, total flavonoids, and total phenolics contents than extracts of flesh. In conclusion, this study demonstrated that CYP-W exerted better antitumor activity than flesh extracts and the scavenging ROS effects were also significantly higher in the CYP-W in vitro. Moreover, the data indicated that allantoin may play an important role on antioxidative and antitumor capacity in yam peel.
Physical properties and antioxidant activity of mucilage from Wencheng Chinese yam(Dioscorea opposita Thunb.)

Abstract:In this study, the components, stability, apparent viscosity, and antioxidant activity of Wencheng Chinese yam（Dioscorea opposita Thunb.） were determined. The contents of polysaccharide and protein are（14.99±1.27）% and（57.16±7.24）%, respectively, and the content of Mg reached（2 654±44） mg/kg. Under the condition of acid-base and higher temperature, the particle size of mucilage decreased and the stability was improved. The viscosity of mucilage solution increased with the increase in concentration, and the viscosity increased significantly at 60-80 ℃, showing obvious pseudoplasticity and shear refinement. Meanwhile, mucilage had a good scavenging ability of hydroxyl radical and DPPH free radicals, and the scavenging rates at 4 mg/mL were 75.7% and 67.45%, respectively. Thus, Wencheng Chinese yam mucilage can be considered as an antioxidant and thickener, which lays a foundation for its application in the food industry.

Abstract:A sulfated Chinese yam polysaccharide (S-CYP) was obtained from non-starch Chinese yam polysaccharide (CYP) by chlorosulfonic acid-pyridine (CSA-Pyr) method. The physicochemical characteristics and immunological activity of S-CYP were evaluated in this study. Results showed that sulfated modification changed the physicochemical properties of Chinese yam polysaccharide. Body weights and thymus index of the mice in polysaccharides group were increased compared with the cyclophosphamide (Cy)-induced immunosuppressed mice in model group, and could restore the splenomegaly that caused by Cy. It also showed that S-CYP could enhance the immunomodulatory activity on splenic lymphocytes since it could increase the proliferation of splenic lymphocytes and effectively cooperate with ConA to induce splenic lymphocytes differentiation into T lymphocytes. Meanwhile, the number of CD3+CD4+ and CD3+CD8+ T lymphocytes was elevated, the CD4+/CD8+ ratio was restored in polysaccharides group. The levels of splenic lymphocytes cytokines (interleukin (IL)-1β, tumor necrosis (TNF)-α), and the levels of immunoglobulin (IgG and IgM) secretion in serum were also increased by S-CYP significantly. These findings demonstrate that CYP and S-CYP can serve as a promising immunomodulator with potential application in functional food and nutraceutical industry.

Abstract:This study aimed to characterize the structure of Chinese yam (Dioscoreae Rhizoma) polysaccharide (CYP) and to investigate its protective effect against H2O2-induced oxidative damage in IEC-6 cells. The chemical composition and structural characteristics of the samples were analyzed by chemical and instrumental methods, including high-performance gel permeation chromatography, high-performance anion-exchange chromatography (HPAEC), Fourier transformed infrared (FT-IR), ultraviolet (UV), and scanning electron microscopy (SEM). Antioxidant activity was evaluated by establishing a cellular model of oxidative damage. The molecular weight of CYP was 20.89 kDa. Analysis of the monosaccharide composition revealed that CYP was primarily comprised of galactose (Gal), glucose (Glu), and galacturonic acid (GalA), and the ratio between them was 28.57:11.28:37.59. Pretreatment with CYP was able to improve cell viability, superoxide dismutase (SOD) activity, and reduce intracellular reactive oxygen species (ROS) production and malondialdehyde (MDA) content after H2O2 injury. CYP also attenuated oxidative damage in cells through the mitogen-activated protein kinase (MAPK) signaling pathway. This study showed that CYP was an acidic heteropolysaccharide with a good protective effect against oxidative damage, and it thus has good prospects in food and biopharmaceutical industries.

Abstract:Yam (Dioscorea spp.), known as an edible and medicinal tuber crop in China, has been used historically for the treatment of diabetes, diarrhea, asthma, and other ailments in traditional Chinese medicine. Moreover, it has been consumed as starchy food for thousands of years in China. Modern phytochemistry and pharmacological experiments have been proved that non-starch polysaccharide is one of the main bioactive substances of yam. Many studies have been focused on the isolation and identification of polysaccharides and their bioactivities of Chinese yam. However, due to the difference in the variety of raw materials and the method of polysaccharides extracting, the structure and biological activity of the obtained polysaccharides also differ. It has been demonstrated that Chinese yam polysaccharide has various important biological activities, such as hypoglycemia, immunomodulatory, antioxidant, and antitumor activities. This paper is aimed at summarizing previous and current references of the isolation processes, structural features and bioactivities of yam polysaccharides. The review will serve as a useful reference material for further investigation and application of yam polysaccharides in functional foods and medicine fields.

Abstract:Aim To study the structure and antitumor activity of polysaccharide from Chinese yam. Methods The tube root of Chinese yam was extracted with water, from the extract, the polysaccharide of RDPS-I was separated and purified using Sevag deprotein, DEAE-cellulose and Sephadex G-100 column chromatography. Its structure was researched by PC, GC, IR, partial hydrolysis, periodate oxidation, Smith degradation, methylation analysis, 1HNMR and 13 CNMR analysis etc. The antitumor activity in vivo also studied with animal experiments. Results RDPS-I was found to be composed of Glc, Man and Gal, in a molar ratio of 1∶0.4∶0.1, contained a backbone composed of α- D -(1,3)-Glcp and, a short branch of α- D -(1→2)-Manp-β- D -1)-Galp is attached to the main chain, the polysaccharide had a molecular weight of 41 000 , [α] 20 D= +188.4° (c 0.80, H 2O) and [η]=16.48×10 -3 ( mL·g -1 ). RDPS-I could significantly inhibit the cancer cell line of melanoma B16 and Lewis lung cancer in mice in vivo . Conclusion RDPS-I is a heteropolysaccharide of Glc, Man and Gal has significant antitumor activity.

Abstract:The polysaccharide of yam was extracted by hot water method and purified by column chromatography. The physicochemical properties of Chinese yam polysaccharide were analyzed by UV, IR, GPC, 1D-NMR and 2D-NMR spectra. The results showed that Chinese yam polysaccharide had alpha-D-Gluc-(1 -> 4) glycoside bond, and the C2 hydroxyl group was replaced by ethoxyl group. The average molecular weight was determined to be 7.28 x 104. It showed that The scavenging effect of yam polysaccharide on hydroxyl radicals was similar to VC. The sulfated polysaccharide (S-P), phosphorylated polysaccharide (P-P), carboxymethylated polysaccharide (C-P) and acetylated polysaccharide (A-P) were identified by IR and NMR. The results showed that P and its derivatives showed good antioxidant activity. Especially, their scavenging ability to hydroxyl radicals reached the level of VC. This laid a theoretical foundation for the development of yam polysaccharide-related foods.

Antioxidant Properties of Polysaccharides from Dioscorea Opposite Thunb Roots

Abstract:Crude polysaccharides (CYPP) were prepared from a variety of Chinese yam, Dioscora opposite Thunb, by boiling-water extraction and ethanol precipitation. Purified polysaccharides (CYP) were obtained from crude polysaccharides after removing protein and decoloring. Antioxidant activities of rude and purified polysaccharides were investigated using various in vitro systems, including reducing power, DPPH and hydroxyl radical scavenging activity, and lipid peroxidation inhibiting ability. Polysaccharides showed good antioxidant activity. At 5 mg/mL, the polysaccharides showed scavenging abilities on hydroxyl radicals were as high as 96.32%-97.28%. Furthermore, the inhibitory rates of lipid spontaneous peroxidation were 66.17%-69.14% at the polysaccharide concentration of 5 mg/mL. In the assay of reducing power, scavenging ability on DPPH radical and inhibitory activities on lipid peroxidation induced by Fe2+-H2O2, CYPP showed higher activity than that of CYP. On the contrary, in the assay of scavenging effect on hydroxyl radical and inhibiting ability on lipid spontaneous peroxidation, CYP showed higher activity than that of CYPP. The results suggest that the polysaccharides from Chinese yam have direct and potent antioxidant activities.

Abstract:Yam (Dioscorea) is a tuber crop cultivated for food security, revenue, and medicinal purposes. It has been used to treat diabetes, asthma, diarrhea, and other diseases. The main active ingredients in yam, polysaccharides, are regarded to be the important reason for its widespread applications. Now, a comprehensive review of research developments of yam polysaccharides (YPs) was presented to explore their prospects. We outlined the structural characteristics, biological activities, structure-activity relationships, and potential applications. Around 13 neutral components and 17 acidic components were separated. They exhibited various bioactivities, including immunomodulatory, hypoglycemic, hypolipidemic, antioxidant, gastrointestinal protective, anti-fatigue, and senile disease treatment activities, as well as prebiotic effect. Structure-activity relationships illustrated that unique structural properties, chemical modifications, and carried biopolymers could influence the bioactivities of YPs. The potential applications in medicine, food, and other fields have also been summarized.

Abstract:It is well known that yam is an important industrial crop resource. Our previous study has demonstrated that sulfated modifications (S-CYP) can improve the immunomodulatory effect of polysaccharide from Chinese yam by activating and maturating RAW264.7 cells, while its mechanism is still unknown. In this study, the underlying mechanisms of S-CYP-induced immunomodulatory effect were examined based on RNA sequencing (RNA-seq), and a co-culture system with Caco-2 cells and RAW264.7 cells. RNA-seq analysis indicated that 1764 expressed genes differentially between the control and S-CYP groups. 13 cellular component terms, 20 biological process terms, and 10 molecular function terms were identified by GO. KEGG analysis showed that MAPK signaling pathways belong to the 20 most significantly affected pathways. Furthermore, S-CYP indirectly affects the immune function of RAW264.7 in co-culture model by increasing secretion of cytokines (IL-1β, TNF-α). These results provide a theoretical basis for exploring S-CYP as an immunomodulatory agent.

Abstract:A water-soluble polysaccharide named CYP-1 was isolated from Chinese yam. CYP-1 was characterized as a mannoglucan having a backbone consisting predominately of 1,4-α-linked Glcp branched at O-2, O-3, and O-6 position by t-α-linked Manp with a molecular weight of 2.86 kDa. CYP-1 could inhibit the overproduction of pro-inflammatory cytokines (such as TNF-α and IL-1β) in LPS-induced RAW 264.7 cells and DSS-induced colitis mice. Oral administration of CYP-1 dramatically alleviated colonic pathological damage, suppressed the activation of colonic inflammatory signaling pathways (such as NF-κB and NLRP3 inflammasome), recovered the mRNA expression of junctional proteins (such as ZO-1, claudin-1, occludin, and connexin-43), and modulated the gut microbiota by decreasing the abundances of Alistipes, Helicobacter, and an unidentified Enterobacteriaceae, in DSS-induced colitis mice. Overall, the present study elucidated that a new polysaccharide structure CYP-1 from Chinese yam and its therapeutic potential as a prebiotic for the prevention of inflammatory bowel disease.
Study of anti-fatigue activity of polysaccharide from fruiting bodies of Armillaria gallica

Abstract:Fatigue is a common physiological response that is closely related to energy metabolism. Polysaccharides, as excellent dietary supplements, have been proven to have a variety of pharmacological activities. In this study, A 23.007 kDa polysaccharide from Armillaria gallica (AGP) was purified and performed structural characterization, including analysis of homogeneity, molecular weight and monosaccharide composition. Methylation analysis is used to analyze the glycosidic bond composition of AGP. The mouse model of acute fatigue was used to evaluate the anti-fatigue effect of AGP. AGP-treatment improved exercise endurance in mice and reduced fatigue symptoms caused by acute exercise. AGP regulated the levels of adenosine triphosphate, lactic acid, blood urea nitrogen and lactate dehydrogenase, muscle glycogen and liver glycogen of acute fatigue mice. AGP affected the composition of intestinal microbiota, the changes of some intestinal microorganisms are correlated with fatigue and oxidative stress indicators. Meanwhile, AGP reduced oxidative stress levels, increased antioxidant enzyme activity and regulated the AMP-dependent protein kinase/nuclear factor erythroid 2-related factor 2 signaling pathway. AGP exerted an anti-fatigue effect through modulation of oxidative stress, which is related to intestinal microbiota.
Effect of in vitro simulated gastrointestinal digestion on the antioxidant activity, molecular weight, and microstructure of polysaccharides from Chinese yam

Abstract:In this study, we investigated the antioxidant properties and the changes of molecular weight (Mw), antioxidant activity, and microstructure of Chinese yam polysaccharides (CYP-A) during in vitro digestion. Results showed that the scavenging rate of 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free radical of CYP-A was approximately 79% at the concentration of 6 mg/mL. Furthermore, the antioxidant ability positively correlated with the concentration of CYP-A. In addition, the Mw of CYP-A decreased (p < 0.05) after intestinal digestion. Fourier transform infrared showed that the degrees of esterification of CYP-A increased to 39.04% after 6 h of gastric digestion. Moreover, the structure of CYP-A changed after in vitro gastric digestion. The scanning electron microscope (SEM) images indicated that the surface morphology of CYP-A turned from smooth and irregular to a layered honeycomb after gastrointestinal digestion in vitro. Surprisingly, the activity of DPPH radical scavenging significantly increased (p < 0.05) in the intestinal digested samples of CYP-A, which showed a positive correlation with the concentration of CYP-A. However, the reducing power significantly decreased (p < 0.05) after in vitro intestinal digestion.

Abstract:The aim of this study was to develop whether the polysaccharides extracted from H.fusiformis have the anti-fatigue effect on mice. It was important for the further development of H. fusiformis. The weight,loading swimming time,the activities of blood SOD,the content of blood MDA,liver glycogen,muscle lactic acid, the content of blood urea nitrogen were measured after 15 days through being given different doses(50、150mg/mL·d) of the polysaccharides to mice. The result showed there was no significant difference on the body weights between control and polysaccharides fed mice(p>0.05). However,the loading swim time(p<0.01), the activity of SOD(p<0.01) and the concentration of liver glycogen(p<0.01) were enhanced,while the concentration of MDA (p<0.01),muscle lactic acid(p<0.01、0.05),and urea in blood(p<0.01)were decreased. In conclusion,the polysaccharides extracted from H. fusiformis do had anti-fatigue effect on mice.
