Abstract:A new analytical method was developed to simultaneously determine residues of 109 pesticides (including isomers) in the foods of animal origin. Acetonitrile was selected for accelerated solvent extraction (ASE) for effectively extracting the pesticides from the fatty samples. The cleanup was performed with an automated gel permeation chromatography (GPC) cleanup system. The prepared samples were analysed with GC–MS in the selected ion monitoring mode (SIM) using one target and two qualitative ions for each analyte. Chlorpyrifos-d10 was used as an internal standard. The lowest limit of detection was 0.3μgkg−1 for some pesticides. The recoveries and relative standard deviations (RSDs) were checked by spiking untreated samples with pesticides at 0.05, 0.1 and 0.2mgkg−1. The average recoveries of most pesticides were from 62.6% to 107.8%. The precision values expressed as RSD were all ⩽20.5% (n=6). Good linearity (r⩾0.99) was observed between 0.05 and 1.5μgmL−1.

Abstract:A high performance liquid chromatography/electrospray ionization tandem mass spectrometric method has been developed for the simultaneous determination of nine carbamate pesticide residues in grains. Carbamate pesticides were extracted with acetonitrile, cleaned up with neutral-alumina column, and then analyzed by high performance liquid chromatography-electrospray (HPLC-ESI) (+) -tandem mass spectrometry (MS/MS). Sample pre-treatment and LC-MS/MS analytical conditions were optimized. The linear range was 0. 1 - 100 g/L, the correlation coefficients were 0. 9986 - 0. 9998. The average recovenies of nine carbamate pesticides in spiked grains (from 0. 001 to 0. 05 mg/Kg) ranged from 73. 40% to 102. 0%, and the relative standard deviation were between 1. 3% and 9. 9%. The results indicate that the method developed is easier, faster, more sensitive and has a better purification effect. It also demonstrats that this multiresidue analysis method can meet the requirements for simultaneous determination of many carbamate pesticides in grains for import & export inspection.
[Co-occurrence of physical and psychological violence among dating adolescents in Recife, Brazil: prevalence and associated factors].

Abstract:The scope of this study was to assess the prevalence of physical and psychological violence among dating adolescents from Recife, Brazil, to identify associated factors and the co-occurrence of both types of violence. 302 adolescents aged between 15 and 19 years in public and private schools who were in a relationship in the last year filled out the questionnaire. Univariate and multivariate logistic regression statistical analysis were performed and the sample weight and the complex sample design were entered. The prevalence was 19.9% of physical violence 82.8% of psychological violence and 18.9% for the co-occurrence of both. Adolescents who have experienced violence in the community and who were in dating relationships of more than one year duration were more likely to perpetrate psychological violence. For the co-occurrence of physical and psychological violence, the variables that increased the chance of perpetration were suffering physical violence from the father, violence among siblings, physical violence and the perpetration of verbal violence in previous relationships. The conclusion reached is that psychological violence and the co-occurrence of physical and psychological violence have a distinct dynamics from physical violence in dating relationships.

Abstract:A new multi-residue method for determination of pesticide residues in a wide variety of fruit and vegetables, using the National Food Administration (NFA) ethyl acetate extraction and determination by means of LC-MS/MS, is presented. The method includes pesticides normally detected by LC-UV or LC-fluorescence such as benzimidazoles, carbamates, N-methylcarbamates and organophosphorus compounds with an oxidisable sulphide group as well. After extraction with ethyl acetate, the extract is concentrated and an aliquot of the extract is evaporated to dryness and redissolved in methanol before injection on LC-MS/MS. The method has been validated for 57 different pesticides and metabolites. Representative species from different commodity groups were chosen as matrices in order to study the influence from different matrices on recoveries. The fortification levels studied were 0.01-0.5 mg kg(-1). Matrix effects were tested for all matrices by means of standard addition to blank extracts. The matrix effect, expressed as signal in solvent compared to signal in matrix, was in general found to be small. The obtained recoveries are, with a few exceptions, in the range 70-100%. The proposed method is quick and straightforward and no additional clean-up steps are needed. The method can be used for the analysis of all 57 pesticides in one single determination step at 0.01 mg kg(-1).

Abstract:Method validation is an essential technique for ensuring the reliability and accuracy of an analytical method. This study aimed to optimize and validate a fast, reliable, and accurate method for quantitatively determining pesticide residues of diverse chemical classes in the tomato matrix. Various method performance characteristics were tested and compared with predefined criteria. Twenty-six different pesticides of diverse chemical classes were selected based on their use in tomato cultivation and the availability of reference materials. The pesticide residues in tomato samples were extracted with the QuEChERS technique with some modifications, followed by injection into an LC-MS/MS system operating in an optimized method. The validated method demonstrated reasonable specificity, as there were no interferences from matrix components at the retention times of pesticides. The calibration curves for all pesticides exhibited excellent linearities, with correlation coefficients exceeding 0.99. No significant matrix effect was observed for all pesticides in tomatoes, as the values fell within the range of ±20%. All pesticides were quantified successfully at a concentration of 5 μg/kg except for carbaryl, with an average recovery of more than 70% and a relative standard deviation of less than 20%. Similarly, measurement uncertainties were also estimated based on the validation data, and the values were found below the default limit of 50%. Subsequently, the validated method was applied to analyze 52 locally collected tomato samples. Study findings revealed that only four of the studied pesticides were detected in these samples, and their concentrations were below the maximum residue limits (500 μg/kg each for carbendazim, imidacloprid, and metalaxyl) established for tomatoes by the Government of Nepal and the Codex Alimentarius Commission.

Abstract:A multiresidue method, based on the sample preparation by solid-phase extraction cartridges and detection by ultra-high-performance liquid chromatography/time-of-flight mass spectrometry (UHPLC/TOF-MS), was used for the analysis of 60 pesticides in vegetable and fruit samples. Quantitation by UHPLC/TOF-MS is accomplished by measuring the accurate mass of the protonated molecules [M+H](+). The mass accuracy typically obtained is routinely better than 2ppm. The rates of recovery for pesticides studied were satisfactory, ranging from 74% to 111% with a relative standard deviation (RSD) of less than 13.2%, at concentrations below 10μgkg(-1). The method limit of quantification (MLOQ) for most compounds was below the MRLs established by the Food Safety Standard Authority of India and the European Union. The uncertainty was determined using repeatability, recovery and calibration curves data for each pesticide. The method illustrated is suitable for routine quantitative analyses of pesticides in food samples.

Abstract:Multivariate response surface methodology optimization using Placket–Burman and Box–Behnken designs were respectively used for the screening and optimization of significant factors for liquid chromatography–tandem mass spectrometry. Consequently, the optimized instrument successfully improved the sample preparation protocol and the method was validated. However, modified QuEChERS dispersive solid phase extraction coupled with ionic liquid-based dispersive liquid–liquid microextraction were used for the determination of multi-pesticide residues in fruit and vegetable samples. The analysed samples were jackfruit, strawberries, cucumber, pears, and carrots. The resulting linearity range (5–400 µg/kg) and regression coefficient (>0.99) results were satisfactory. The 94.2 and 95.8% accuracy (89–138%) and precision (0–25%) results were satisfactory and within the recommended ranges (≤20%) and (70–120%), respectively. The limits of detection (0.01–0.54 µg/kg) and quantitation (0.03–1.79 µg/kg) were excellent. The matrix effects (≤−87%) for all analysed samples were not significant. The estimated measurement uncertainties (≤27%) were within the acceptable range (≤50%). Justifiably, the response surface methodology optimized instrument and sample treatment techniques were reliable and convenient for multi-pesticide residue determination in various fruits and vegetables.

Abstract:A new method for the analysis of pesticides in agricultural soils by using liquid chromatography coupled with a high-resolution quadrupole time-of-flight mass spectrometer (LC-QTOF-MS) has been developed. Twenty-four pesticides including herbicides, insecticides or fungicides have been studied. The linearity of the external calibration of the pesticides obtained from levels of concentration ranging from 0.010 to 400 µg/Lgives satisfactory results with the correlation coefficients higher than 0.99. The quantification limits of the equipment range from 0.010 to 1.250 µg/L and correspond to the lowest standard in the calibration range for which the residual deviation is less than 60%. An experimental design has been used to optimise the parameters of pressurised-liquid extraction (PLE) of pesticides in soils. The optimised conditions have been found by using methanol as extraction solvent at a temperature of 80°C, a pressure of 150 Bars and 2 extraction cycles of 5 minutes. The mean recoveries of pesticides (Mean) are higher than 72% with a relative standard deviation(RSD) less than 21%. These results demonstrate the good efficiency of PLE for the extraction of pesticides in agricultural soils of different nature. The LC-QTOF-MS is a sensitive, linear and robust instrument for the quantification of pesticides in unpurified soil extracts by external calibration. This analytical tool is not subjected to matrix effects of soil extracts.

Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.

Abstract:BACKGROUND Comparing the relative effectiveness of interventions on specific outcomes across trials can be problematic due to differences in the choice and definitions of outcome measures used by researchers. We sought to identify a minimum set of outcome measures for evaluating models of maternity care from the perspective of key stakeholders. METHODS A 3-round, electronic Delphi survey design was used. Setting was multinational, comprising a range of key stakeholders. Participants consisted of a single heterogeneous panel of maternity service users, midwives, obstetricians, pediatricians/neonatologists, family physicians/general practitioners, policy-makers, service practitioners, and researchers of maternity care. Members of the panel self-assessed their expertise in evaluating models of maternity care. RESULTS A total of 320 people from 28 countries expressed willingness to take part in this survey. Round 1 was completed by 218 (68.1%) participants, of whom 173 (79.4%) completed round 2 and 152 (87.9%) of these completed round 3. Fifty outcomes were identified, with both a mean value greater than the overall group mean for all outcomes combined (x=4.18) and rated 4 or more on a 5-point Likert-type scale for importance of inclusion in a minimum data set of outcome measures by at least 70 percent of respondents. Three outcomes were collapsed into a single outcome so that the final minimum set includes 48 outcomes. CONCLUSIONS Given the inconsistencies in the choice of outcome measures routinely collected and reported in randomized evaluations of maternity care, it is hoped that use of the data set will increase the potential for national and international comparisons of models for maternity care. Although not intended to be prescriptive or to inhibit the collection of other outcomes, we hope that the core set will make it easier to assess the care of women and their babies during pregnancy and childbirth.
Deep learning enabled miniature mass spectrometer for rapid qualitative and quantitative analysis of pesticides on vegetable surfaces

Abstract:Excessive pesticide use poses a significant threat to food safety. Rapid on-site detection of multi-target pesticide residues in vegetables is crucial due to their widespread distribution and limited shelf life. In this study, a rapid on-site screening method for pesticide residues on vegetable surfaces was developed by employing a miniature mass spectrometer. A direct pretreatment method involves placing vegetables and elution solution into a customized flexible ziplock bag, allowing thorough mixing, washing, and filtration. This process effectively removes pesticide residues from vegetable surfaces with minimal organic solvent usage and can be completed within 2 min. Moreover, this study introduced a deep learning algorithm based on a one-dimensional convolutional neural network, coupled with a feature database, to autonomously discriminate detection outcomes. By combining full scan MS and tandem MS analysis methods, the proposed method achieved a qualitative recognition accuracy of 99.62%. Following the qualitative discrimination stage, the target pesticide residue and internal standard can be simultaneously isolated and fragmented in the ion trap, thus enabling onsite quantitative analysis and warning. This method achieved a quantitative detection limit of 10 mu g/kg for carbendazim in cowpea. These results demonstrate the feasibility of the proposed analytical system and strategy in food safety applications.
Validation and Simultaneous Monitoring of 311 Pesticide Residues in Loamy Sand Agricultural Soils by LC-MS/MS and GC-MS/MS, Combined with QuEChERS-Based Extraction

Abstract:Soil can be contaminated by pesticide residues through agricultural practices, by direct application or through spray-drift in cultivations. The dissipation of those chemicals in the soil may pose risks to the environment and human health. A simple and sensitive multi-residue analytical method was optimized and validated for the simultaneous determination of 311 active substances of pesticides in agricultural soils. The method involves sample preparation with QuEChERS-based extraction, and determination of the analytes with a combination of GC-MS/MS and LC-MS/MS techniques. Calibration plots were linear for both detectors over the range of five concentration levels, using matrix-matched calibration standards. The obtained recoveries from fortified-soil samples ranged from 70 to 119% and from 72.6 to 119% for GC-MS/MS and LC-MS/MS, respectively, while precision values were <20% in all cases. As regards the matrix effect (ME), signal suppression was observed in the liquid chromatography (LC)-amenable compounds, which was further estimated to be negligible. The gas chromatography (GC)-amenable compounds showed enhancement in the chromatographic response estimated as medium or strong ME. The calibrated limit of quantification (LOQ) value was 0.01 μg g-1 dry weight for most of the analytes, while the corresponding calculated limit of determination (LOD) value was 0.003 μg g-1 d.w. The proposed method was subsequently applied to agricultural soils from Greece, and positive determinations were obtained, among which were non-authorized compounds. The results indicate that the developed multi-residue method is fit for the purpose of analyzing low levels of pesticides in soil, according to EU requirements.
Development of a Microwave-Assisted Extraction Protocol for the Simultaneous Determination of Mycotoxins and Pesticide Residues in Apples by LC-MS/MS

Abstract:The possible presence of contaminants, pesticide residues and mycotoxins, in agricultural commodities is a critical issue for food safety, causing great concern. In this work, a simple and rapid analytical method employing liquid chromatography–tandem mass spectrometry (LC-MS/MS) was developed for the simultaneous determination of pesticide residues and mycotoxins in apples. Microwave-assisted extraction (MAE) was used for sample preparation. The MAE protocol was optimized after evaluating the effects of the following extraction parameters: (a) extraction solvent, (b) acidic environment, (c) temperature, and (d) extraction time. The multiresidue MAE-LC-MS/MS method was validated for linearity, accuracy (bias and precision), limits of detection (LODs), limits of quantification (LOQs), and matrix effect. The validation of the method was carried out according to the SANTE/12682/2019 document. The method demonstrated good linearity with R2 ≥ 0.99, acceptable accuracy in the recovery rate range 70–116%, acceptable interassay precision with RSD% ≤ 20, and low LODs and LOQs in the ranges 0.005–0.015 μg/g and 0.01–0.03 μg/g, respectively. Matrix effects were observed only for the 25% of the analytes. The performance of the MAE-LC-MS/MS method was compared to that of the QuEChERS sample preparation method, and the MAE-LC-MS/MS method proved to be rapid and effective.

Abstract:Antibiotics used in livestock husbandry can contaminate the environment through animal excretions and aquaculture wastewater. Absorption of these antibiotics by vegetables may pose health risks to humans through the food chain. However, research on detecting multi-antibiotics in vegetables remains limited. This study aims to develop a reliable and sensitive method to quantify 72 antibiotics from six classes in vegetable samples using modified QuEChERS and UPLC-MS/MS methods. The QuEChERS method was optimized by investigating extraction solvents, salting-out agents, and adsorbents. The percentage recovery of the 72 analyte antibiotics from spiked vegetable samples ranged between 61.5% and 117.8%. Intra-day and inter-day precision were below 12.1% and 15.4%, respectively. The limits of detection and quantification were 0.010–0.232 µg.kg−1 and 0.032–0.772 µg.kg−1, respectively. The method demonstrated a weak matrix effect for 61.1% of the tested compounds. Applying the developed method to analyze 248 vegetable samples, 15 antibiotics were detected, ranging from 0.010 µg.kg−1 to 561.0 µg.kg−1 fresh weight. Notably, bean sprouts showed the presence of multiple antibiotics, suggesting the possible misuse of antibiotics in their production.

Abstract:The analysis of pesticide residues in food is nowadays an increasingly important task. Quality control has to be very strict in order to safeguard the consumers’ health. One of the most important goals of food quality assurance is testing food for residues and contaminants. Among chemical hazards, the contamination of food with pesticides has been characterized as a significant source of many serious diseases. Consumption of food containing pesticide residues may cause cancer, malformations, and damage to the endocrine, nervous, and immune system. In order to assure human food safety, The European Community has established maximum residue limits (MRL) of pesticides permitted in products of animal or vegetable origin that are intended for human consumption. In the EU, Regulation (EC) No. 396/2005 of the European Parliament and Council on pesticide residues established the levels of pesticide residues allowed in food and feed. To ensure the compliance of marketed food commodities with the law on food safety, sensitive and reliable analytical methods for the determination of pesticide residues are required. Multiresidue liquid chromatography-mass spectrometry methods (LC-MS) are widely recognized as an ideal, highly specific, and extremely sensitive technique for testing food products. This review discusses LC-MS approaches applied most widely to pesticide residue analysis over the last few years. The main ionization methods and MS detectors used as analytical tools in quantification and/or identification/confirmation of pesticide residues in food are presented.

Abstract:A liquid chromatography-mass spectrometry (LC-MS) method was established for the simultaneous determination of carbamate and organophosphorus pesticides (OPPs) in fruits and vegetables by dispersive solid phase extraction using acetonitrile as extraction solvent and primary secondary amine (PSA) as sorbent instead of octadecyl bonded silica (ODS). Four common representative samples (tomato, apple, carrot and cabbage) were selected from the supermarket in order to validate the method by recovery and precision. The obtained recoveries were, with a few exceptions, in the range of 80%-110% with RSDs less than 8%; and limits of detection ranged from 0.5 to 35 μg/kg in tomato. In addition, the method was applied to pesticides residue detection in vegetables and fruits obtained from local markets.

Abstract:The widespread use of pesticides and their consequential presence in the environment is a growing concern due to the harmful health effects associated with pesticide exposure. For clinical and toxicology laboratories, a method for simultaneously determining these compounds and their metabolic products in body fluids, such as blood and urine, is important. In the present study, a rapid, sensitive and simultaneous LC-QToF-MS method for detecting multiclass pesticides and metabolites in blood and urine samples has been developed and validated. Four sample preparation procedures, protein precipitation and three different variants of QuEChERS-based extraction were evaluated to find a suitable, simple, and effective sample pretreatment technique. The final optimized sample preparation method (acetonitrile; 400 μl, MgSO4; 40 mg and NaCl; 10 mg) was validated for accuracy, precision, matrix effect, recovery, stability, carryover, and dilution integrity. Analyte recoveries ranged from 75.40 to 113.54 % while accuracy was evaluated in the range of 71.41-108.26 % and precision (%RSD) in the range of 0.01 %-16.85 %. The limit of quantification (LOQ) for all compounds was established in the range of 0.82-7.05 ng mL-1. The developed reliable, robust, and sensitive method was successfully applied for the quantification of target pesticides and metabolites in human blood and urine samples. Evaluated samples resulted in detection of eleven analytes (seven pesticides and four metabolites).

Abstract:Abstract A method for detection, quantitation, and confirmation of more than 100 pesticides by gas chromatography (GC) with ion trap mass spectrometry (MS/MS) has been developed. The sensitivity of this method for many analytes is equal to or lower than those of selective GC detectors such as flame photometric detectors and electrolytic conductivity detectors. Using MS/MS, very low detection limits and good confirmation (1 precursor ion and 2 or more product ions) are achieved simultaneously. The entire list of pesticides is screened with 2 injections per sample. Samples are introduced onto the column by a temperature-programmed cold injection to maximize response. Each pesticide is run with its own unique set of parameters, which fragment the compound, retaining only the precursor ion. This ion is then refragmented to create a product spectrum. The selectivity of MS/MS gives a very clean spectrum, making compound identification and confirmation clear, even with a relatively dirty food matrix. If care is taken to maintain the injection port and guard column, this method can reliably identify and confirm more than 100 pesticides at the low parts-per-billion range.
