Abstract:A new analytical method was developed to simultaneously determine residues of 109 pesticides (including isomers) in the foods of animal origin. Acetonitrile was selected for accelerated solvent extraction (ASE) for effectively extracting the pesticides from the fatty samples. The cleanup was performed with an automated gel permeation chromatography (GPC) cleanup system. The prepared samples were analysed with GC–MS in the selected ion monitoring mode (SIM) using one target and two qualitative ions for each analyte. Chlorpyrifos-d10 was used as an internal standard. The lowest limit of detection was 0.3μgkg−1 for some pesticides. The recoveries and relative standard deviations (RSDs) were checked by spiking untreated samples with pesticides at 0.05, 0.1 and 0.2mgkg−1. The average recoveries of most pesticides were from 62.6% to 107.8%. The precision values expressed as RSD were all ⩽20.5% (n=6). Good linearity (r⩾0.99) was observed between 0.05 and 1.5μgmL−1.

Abstract:A high performance liquid chromatography/electrospray ionization tandem mass spectrometric method has been developed for the simultaneous determination of nine carbamate pesticide residues in grains. Carbamate pesticides were extracted with acetonitrile, cleaned up with neutral-alumina column, and then analyzed by high performance liquid chromatography-electrospray (HPLC-ESI) (+) -tandem mass spectrometry (MS/MS). Sample pre-treatment and LC-MS/MS analytical conditions were optimized. The linear range was 0. 1 - 100 g/L, the correlation coefficients were 0. 9986 - 0. 9998. The average recovenies of nine carbamate pesticides in spiked grains (from 0. 001 to 0. 05 mg/Kg) ranged from 73. 40% to 102. 0%, and the relative standard deviation were between 1. 3% and 9. 9%. The results indicate that the method developed is easier, faster, more sensitive and has a better purification effect. It also demonstrats that this multiresidue analysis method can meet the requirements for simultaneous determination of many carbamate pesticides in grains for import & export inspection.
Multi-pesticide residue screening, identification, and quantification analysis in various fruits and vegetables by UHPLC-Q Exactive HRMS

Abstract:A general strategy for qualitative screening and quantitative analysis of 403 pesticides in various fruits and vegetables was developed using ultrahigh-performance liquid chromatography (UHPLC) in conjunction with a Thermo Q Exactive Focus high-resolution mass spectrometer, relying on an executable compound database comprised of the exacted mass of precursor ions, retention times, and fragment ions. Taking advantage of the powerful separation capacity of UHPLC, an Orbitrap analyzer with high sensitivity in full scan mode and elevated mass resolution of product ions in the MS/MS mode, eight pairs of isomers and fifty-seven groups of isobaric compounds were selectively identified. The method was then systematically assessed and validated for eight fruits and vegetables, in terms of screening detection limit (SDL), matrix effects, recovery, and precision over 400 pesticides. The results showed that the SDLs of 68.0-84.4% for the pesticides were less than or equal to 10 μg kg-1 in the representative matrices, recoveries in the range of 60-120% accounting for 48.6-84.4% of all the targets at three lower fortified levels of 5, 20, and 50 μg kg-1 with a precision of less than 20% while the range of overall average recoveries for the majority of the pesticides were from 82.4% to 105.1% in the as-selected matrices with RSDs between 3.9% and 7.7%. Using this method, screening data from a survey of pesticide residues in 68 practical samples across 32 different matrix types provides scientific data for the inspection and supervision of pesticide residue safety of fruits and vegetables.
High-throughput simultaneous analysis of multiple pesticides in grain, fruit, and vegetables by GC-MS/MS

Abstract:A multi-component quantitative GC-MS/MS method for monitoring and inspecting agricultural products was developed for the simultaneous determination of 365 pesticide residues in grain, beans, fruit and vegetables. Twenty-six pesticides that have been authorised in foreign countries but not in Korea were included in the newly adapted Korean positive list system. The method was validated by analysing spiked samples at three concentrations (0.01, 0.05, and 0.1 mg/kg) for brown rice, soy beans, mandarin orange, potato and green pepper matrice, which represented grain, beans, fruit, and vegetables. More than 95% of 365 compounds satisfied the validation criteria of 70–120% recovery and ≤20% relative standard deviation obtained through the proposed simultaneous multi-component analysis. Twenty-two types of pesticides were detected in 76 food samples, whose concentrations were below the maximum residue limits. Among the newly added pesticides, lambda-cyhalothrin, an analogue of cyhalothrin, was detected in peaches. The method can be applied to the analysis and inspection of pesticide residues in domestic and imported agricultural products for safety control.
Simultaneous Analysis of 272 Pesticides in Agricultural Products by the QuEChERS Method and Gas Chromatography with Tandem Mass Spectrometry

Abstract:The aim of this study is to develop a rapid and accurate method for simultaneous analysis of multi-residue pesticides and conduct pesticide monitoring in agricultural products produced by the production and distribution stage in Korea. The representative agricultural products were selected as brown rice, soybean, potato, mandarin, and green pepper and developed using gas chromatography with tandem mass (GC-MS/MS) for the analysis of 272 pesticide residues. The experimental samples were extracted by the QuEChERS-EN method and then cleaned up by using d-SPE, including MgSO4 and primary secondary amine (PSA) sorbents. The established method was validated in accordance with Codex CAC-GL/40, and the limit of quantitation (LOQ) was determined to be 0.01 mg/kg. A total of 243 pesticides satisfied the guidelines in five samples at three levels with values of 60 to 120% (recovery) and ≤45% (coefficient of variation, CV). The remaining 29 pesticides did not satisfy the guidelines, and these pesticides are expected to be used as a screening method for the routine inspection of agricultural products. As a result of analyzing 223 agricultural products in South Korea by applying the simultaneous analysis method, none of the detected levels in the samples exceeded the standard values based on maximum residue limits (MRLs). The developed method in this study will be used to inspect residual pesticides in agricultural products, and it is anticipated to contribute to the distribution of safe agricultural products to consumers.

Simultaneous Determination of Multi-Component Mycotoxin Contaminants in Foods and Feeds by Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry.

Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.

Abstract:A new method for screening and identification 420 pesticide residues in fruits and vegetables by ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry (UPLC-Q-TOF/MS) were developed. The samples were extracted with acetonitrile/acetic acid (99:1, V/V ), and clean-up by SinChERS-Nano (single-step, cheap, effective, rugged, safe, nano) column, determined by UPLC-Q-TOF/MS. The accurate mass database and MS/MS database which contains 420 pesticides were established, the automatic retrieval of detection results was carried on according to the accurate mass, retention time, isotope ratio, ion fragment information, and so on. Method verification was performed on leeks samples. The results showed that 420 pesticides had good linearity in the range of 0.1–100 μg/L, and the correlation coeffificients ( R 2 ) was greater than 0.990. The limits of detections (LODs) and limits of quantifications (LOQs) of 420 pesticides were in range of 0.05–2.0 and 0.1–5.0 μg/L, respectively. The average spike recoveries at 3 levels were 70.1 % to 119.7 %, and the relative standard deviations (RSD) were lower than 20 % ( n = 6). With this method, a survey of pesticide residues was conducted for 110 samples of 10 different fruits and vegetables, which provided scientific data for ensuring pesticide residue safety of the fruits and vegetables consumed daily by the public. This method was simple, sensitive and accurate, and could be used for rapid screening of 420 pesticide residues in fruits and vegetables.

Abstract:In this paper, we developed a sensitive UPLC-MS/MS method to determine pesticide residues in plant matrices (corn, fresh corn, fresh corn stover, old corn stover, and corn silage) and animal matrices (beef, fat, milk, milk fat, kidney, liver, and cow stomach) quantitatively. Twenty-seven pesticides were extracted with acetonitrile from all plant and animal matrices separately and purified with a mixture of primary secondary amine (PSA) and graphitized carbon black (GCB) or octadecylsilane (C 18 ). The average recoveries of these compounds ranged from 60.7% to 118.2%, and the relative standard deviations were less than 20.0%. The limit of quantitation for all compounds was 0.01 mg kg −1 (for cyhalothrin and beta cypermethrin the LOQ was 0.02 mg kg −1 ). The establishment of multi-residue analysis methods for a variety of matrices can be used as a database for future method research. The results of this study are essential for calculating the transfer of pesticide residues from feed to animal products and for monitoring food safety, which will protect people's health and safety.

Abstract:A simple, rapid, and sensitive multi-residue method has been developed for the determination of 205 pesticides commonly used for crop protection. Pesticide residues were extracted from samples with acetonitrile and a mini-solid phase extraction filter. No additional concentration or clean-up steps were necessary. Analyses were performed via large volume gas chromatography injection using a stomach-shaped inlet liner. The developed method was linear over the range assayed (5–50 μg L−1), with determination coefficients >0.99. The limits of quantification were in the range 0.0005–0.6 mg kg−1 and the limits of detection were between 0.00015 and 0.2 mg kg−1. The majorities of them were lower than the maximum residue set by the Japanese government in “Positive System for Agricultural Chemical Residues in Food” directive in apple. This method was applied to determine pesticide levels in vegetables, fruits, and beans.

Abstract:A new method for the analysis of pesticides in agricultural soils by using liquid chromatography coupled with a high-resolution quadrupole time-of-flight mass spectrometer (LC-QTOF-MS) has been developed. Twenty-four pesticides including herbicides, insecticides or fungicides have been studied. The linearity of the external calibration of the pesticides obtained from levels of concentration ranging from 0.010 to 400 µg/Lgives satisfactory results with the correlation coefficients higher than 0.99. The quantification limits of the equipment range from 0.010 to 1.250 µg/L and correspond to the lowest standard in the calibration range for which the residual deviation is less than 60%. An experimental design has been used to optimise the parameters of pressurised-liquid extraction (PLE) of pesticides in soils. The optimised conditions have been found by using methanol as extraction solvent at a temperature of 80°C, a pressure of 150 Bars and 2 extraction cycles of 5 minutes. The mean recoveries of pesticides (Mean) are higher than 72% with a relative standard deviation(RSD) less than 21%. These results demonstrate the good efficiency of PLE for the extraction of pesticides in agricultural soils of different nature. The LC-QTOF-MS is a sensitive, linear and robust instrument for the quantification of pesticides in unpurified soil extracts by external calibration. This analytical tool is not subjected to matrix effects of soil extracts.

Abstract:The emerging ungovernable application of pesticides in rice farming has attracted public concerns as these hazardous chemicals leave long-lasting environmental impacts and cause severe health effects. Here, an optimized analytical method was proposed for the measurement of 656 pesticide residues in rice samples collected in Vietnam. We utilized chromatography with tandem mass spectrometry systems (UPLC-MS/MS and GC-MS/MS) combined with a modified quick, easy, cheap, effective, rugged, and safe (QuEChERS) extraction method and adopted a mixed-mode SPE clean-up method for the analysis. The results showed that a total of 341 and 315 compounds were determined by UPLC- and GC-MS/MS, respectively. Usage of 10 mL MeCN, 5 mL H2O and 1% glacial acetic acid as extraction solvent outperformed other mixtures in purifying the analytes from the sample matrix. Besides, pressure swing adsorption connected to a C18 cartridge with C18 placed on top exhibited remarkably more extracted compounds of high recovery which resulted in 299 and 318 compounds with recovery ranging from 70 to 120% in GC- and UPLC-MS/MS, respectively. Our optimized protocols also resulted in maximal limits of quantification of 10 μg Kg−1 in both MS methods with repeatability and reproducibility less than 20%. Application of validated method on 20 rice samples collected in Hanoi, Vietnam showed that 14 samples were contaminated with at least one pesticide, and insecticide was the most detected group. Overall, the compliance of all method validation parameters to SANTE/12682/2019 Guideline demonstrates that this protocol can be employed for the effective management of Vietnam’s rice in accordance with international requirements.

Recently, serious concerns have been raised about potential safety risk posed by exogenous pesticide contaminants in herbal medicines. It is highly desirable to have reliable detection and determination of suspected pesticide residues for understanding of their overall quality. However, inherent metabolites abundant in herbal medicines typically make screening of trace-level pesticide residues arduous. Taking Lonicerae Japonicae Flos (LJF) as an example, the current study investigated the capability of a three-dimensional separation system, ultra high-performance liquid chromatography tandem ion mobility/quadrupole time-of-flight mass spectrometry (UPLC-IM/Q-TOF MS), for qualitative detection of numerous pesticides when suffering from complex matrix interferences. The comparison of collision gases was first performed to improve unwanted dissociations of pesticides, particularly labile ones, and nitrogen was ultimately chosen instead of argon. Further optimization of key MS parameters resulted a data-independent high-definition MS E (HDMS E ) method was developed. Then, an in-house pesticide scientific library (PSL) containing 438 pesticides was constructed following a four-step workflow. Based on multi-dimensional information in this library, different identification criteria were compared and an enhanced qualitative screening approach for 432 targeted pesticides was finally proposed and validated. By its application to 104 batches of LJFs, the approach was demonstrated effective in resolving complex matrix interferences. Moreover, collision cross section (CCS) for identification purpose was favorable to improve detection sensitivity while maintain ideal accuracy. Determination of Pesticide Residues in Fruit and Vegetables by High-Performance Liquid Chromatography–Tandem Mass Spectrometry with Multivariate Response Surface Methodology

Abstract:Multivariate response surface methodology optimization using Placket–Burman and Box–Behnken designs were respectively used for the screening and optimization of significant factors for liquid chromatography–tandem mass spectrometry. Consequently, the optimized instrument successfully improved the sample preparation protocol and the method was validated. However, modified QuEChERS dispersive solid phase extraction coupled with ionic liquid-based dispersive liquid–liquid microextraction were used for the determination of multi-pesticide residues in fruit and vegetable samples. The analysed samples were jackfruit, strawberries, cucumber, pears, and carrots. The resulting linearity range (5–400 µg/kg) and regression coefficient (>0.99) results were satisfactory. The 94.2 and 95.8% accuracy (89–138%) and precision (0–25%) results were satisfactory and within the recommended ranges (≤20%) and (70–120%), respectively. The limits of detection (0.01–0.54 µg/kg) and quantitation (0.03–1.79 µg/kg) were excellent. The matrix effects (≤−87%) for all analysed samples were not significant. The estimated measurement uncertainties (≤27%) were within the acceptable range (≤50%). Justifiably, the response surface methodology optimized instrument and sample treatment techniques were reliable and convenient for multi-pesticide residue determination in various fruits and vegetables.

Abstract:Currently, there are thousands of plant protection chemicals being allowed for use in agriculture. Abuse or improper use of pesticides lead to increase residues in products, exceeding the maximum residue limit according to the regulations. Therefore, the need for developing methods, which can monitor, detect, and identify many pesticides simultaneously is necessary. The liquid chromatography quadrupole-time-of-flight mass spectrometry (LC-QTOF-MS) method has advantage of being able to simultaneously quantify and screen many compounds at once, are being a trend in analytical techniques for analyzing and screening pesticide residues in foods. In this study, the LC-QTOF-MS method was used to quantify 186 pesticides in fruit and vegetable samples. The limit of detection (LOD) for all compounds is 3.0 µg/kg. The recovery is in the ranged of 74.4 -118%, relative standard deviation (RSD) <20%. At the same time, the method was initial researched for nontarget screening by reprocessing previously acquired data without re-injection of the samples. The list of pesticides can be screening, which are up to 500 chemicals, by comparison results with the available high resolution mass spectral library. The screening detection limit of the method (SDL) is 100 µg/kg. The method has been applied to quantify 186 pesticides, and nontarget screening analyse in 160 fruit and vegetable samples in the first 06 months of 2023.

Abstract:Pesticide residues in foodstuffs can lead to several undesirable effects. A simple and high-throughput targeted screening method analyzing multi-residue pesticide in aquaculture water based on ultra-high-performance liquid chromatography-Q/Orbitrap mass spectrometry (UHPLC-Q/Orbi MS) was developed and validated. In this technique, the peaks of the compound using precursor ions were recorded by the full scan, which was used for rough quantitative analysis with single point matrix matched calibration. The qualitative identification was performed following the stringent confirmation criteria with fragment ions, retention time, and an isotopic pattern. Additionally, solid-phase extraction with an HLB (Hydrophilic/Lipophilic Balanced) column was selected to enrich and separate target pesticides from water. The screening detection limit of 33 compounds are less than 2 ng·L−1, while 26 compounds range from 2 ng·L−1 to 10 ng·L−1, 19 compounds are at the range of 10–200 ng·L−1, and the other two compounds are 200 ng·L−1 and 1000 ng·L−1. Most of the recovery results were found to be between 60~130%. Finally, the method was successfully applied to the analysis of pesticide residues in 30 water samples from aquaculture environment in Shanghai, indicating its applicability in pesticide screening for environmental monitoring.

Abstract:The consequences of climate change along with diverse food regulations and agricultural practices worldwide are complexifying the occurrence and management of chemical contaminants in food. In this context, we present an ultra-high-performance liquid chromatography high-resolution mass spectrometry (LC-HRMS) approach for the simultaneous identification and quantitation of over 1100 pesticide residues, mycotoxins, and plant toxins in cereals and fruits and vegetables. Analytical conditions were optimized to maximize the scope of the targeted molecules, the reliability of compound identification, and quantification performance within a single method. The method was further transferred and validated in another laboratory to assess its ruggedness. Validation according to the SANTE 11312/2021v2 guidelines showed that 92% and 98% of the molecules fulfill the quantification criteria at the lowest validated level in the cereals and fruits and vegetables groups, respectively. Analysis of fifteen certified reference materials led to a 96% satisfactory rate of z-scores confirming method's competitiveness. Furthermore, the occurrence of these contaminants was studied in 205 cereals and grains samples collected worldwide. The low µg/kg quantification limits make this LC-HRMS method a valuable tool to ensure compliance toward regulations and to screen for non-regulated substances for which occurrence data are crucial for an appropriate risk evaluation.
Multiresidue Analysis of Pesticides in Straw Roughage by Liquid Chromatography-Tandem Mass Spectrometry

Abstract:A multiresidue analytical method using a modification of the "quick, easy, cheap, effective, rugged, and safe" (QuEChERS) sample preparation approach combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis was established and validated for the rapid determination of 69 pesticides at different levels (1-100 ng/g) in wheat and rice straws. In the quantitative analysis, the recoveries ranged from 70 to 120%, and consistent RSDs ≤ 20% were achieved for most of the target analytes (53 pesticides in wheat straw and 58 in rice straw). Almost all of the analytes achieved good linearity with R(2) > 0.98, and the limit of validation levels (LVLs) for diverse pesticides ranged from 1 to 10 ng/g. Different extraction and cleanup conditions were evaluated in both types of straw, leading to different options. The use of 0.1% formic acid or not in extraction with acetonitrile yielded similar final outcomes, but led to the use of a different sorbent in dispersive solid-phase extraction. Both options are efficient and useful for the multiresidue analysis of targeted pesticides in wheat and rice straw samples.

Abstract:Irritable bowel syndrome (IBS) is the most common disorder in patients seen by gastroenterologists. Twenty subjects with IBS diagnosed with the Rome criteria were treated for 12 weeks with 20-40 mg/day of paroxetine (mean dose=31 mg/day). At baseline, 10 patients had a lifetime history of an anxiety disorder, and 10 patients did not have such a history. Both groups had similar improvement in abdominal pain, constipation, diarrhea, incomplete emptying, and bloating/ abdominal distension. Paroxetine was very well tolerated.

Abstract:Pesticide residues in aquatic products are of great concern due to the risk of environmental transmission and their extensive use in aquaculture. In our work, a quick screening approach was developed for the qualitative and semi-quantitative screening of 87 pesticide residues in aquatic products. The sample preparation was investigated, including extract solvent, extract methods, buffer salts, lipid removal, cleanup materials and filter membranes for aquatic products. Samples were extracted using a modified QuEChERS procedure, and two clean-up procedures were developed for UHPLC-Q/Orbitrap MS analysis based on the fat content of the aquatic products. The screening detection limits for all studied pesticides were distributed between 1 and 500 μg/kg in the three representative matrices. Seventy-one pesticides could be analyzed with a screening limit between 1 and 25 μg/kg in grass carp and crayfish, sixty-one pesticides could be screened for limits between 1 and 50 μg/kg in crab. The accuracy results showed that recoveries ranged from 50 to 120% for 60, 56 and 52 pesticides at medium-level for grass carp, crayfish and crab, respectively. At high spiking levels, 74, 65 and 59 pesticides were recovered within the range of 50–120% for the three matrices, respectively. The relative standard deviations of most compounds in different matrices were less than 20%. With this method, the local farmed aquatic products were tested for pesticide residues. In these samples, ethoxyquinoline, prometryn and phoxim were frequently detected. The majority of these confirmed compounds did not exceed 2.00 μg/kg. A grass carp with trichlorfon at 4.87 μg/kg and two carps with ethoxyquinoline at 200 μg/kg were detected, indicating the potential dietary risk.

Abstract:Pesticide testing of foodstuffs is usually accomplished with generic wide-scope multi-residue methods based on liquid chromatography tandem mass spectrometry (LC-MS/MS). However, this approach does not cover some special pesticides, the so called "single-residue method" compounds, that are hardly compatible with standard reversed-phase (RP) separations due to their specific properties. In this article, we propose a comprehensive strategy for the integration of single residue method compounds and standard multiresidue pesticides within a single run. It is based on the use of a parallel LC column assembly with two different LC gradients performing orthogonal hydrophilic interaction chromatography (HILIC) and reversed-phase (RPLC) chromatography within one analytical run. Two sample aliquots were simultaneously injected on each column, using different gradients, being the eluents merged post-column prior to mass spectrometry detection. The approach was tested with 41 multiclass pesticides covering a wide range of physicochemical properties across several orders of log Kow (from -4 to +5.5). With this assembly, distinct separation from the void was attained for all the pesticides studied, keeping similar performance in terms of sensitivity, peak area reproducibility (<6 RSD% in most cases) and retention time stability of standard single column approaches (better than±0.1min). The application of the proposed approach using parallel HILIC/RPLC and RPLC/aqueous normal phase (Obelisc) were assessed in leek using LC-MS/MS. For this purpose, a hybrid QuEChERS (Quick, easy, cheap, effective, rugged and safe)/QuPPe (quick method for polar pesticides) method was evaluated based on solvent extraction with MeOH and acetonitrile followed by dispersive solid-phase extraction, delivering appropriate recoveries for most of the pesticides included in the study within the log Kow in the range from -4 to +5.5. The proposed strategy may be extended to other fields such as sport drug testing or environmental analysis, where the same type of variety of analytes featuring poor retention within a single chromatographic separation occurs.
