Abstract:Unlike most optical coherence microscopy (OCM) systems, dynamic speckle-field interferometric microscopy (DSIM) achieves depth sectioning through the spatial-coherence gating effect. Under high numerical aperture (NA) speckle-field illumination, our previous experiments have demonstrated less than 1 μm depth resolution in reflection-mode DSIM, while doubling the diffraction limited resolution as under structured illumination. However, there has not been a physical model to rigorously describe the speckle imaging process, in particular explaining the sectioning effect under high illumination and imaging NA settings in DSIM. In this paper, we develop such a model based on the diffraction tomography theory and the speckle statistics. Using this model, we calculate the system response function, which is used to further obtain the depth resolution limit in reflection-mode DSIM. Theoretically calculated depth resolution limit is in an excellent agreement with experiment results. We envision that our physical model will not only help in understanding the imaging process in DSIM, but also enable better designing such systems for depth-resolved measurements in biological cells and tissues.

Abstract:Nonlinear focal modulation microscopy (NFOMM) is a really simple and effective super-resolution imaging method based on phase modulation with high-intensity illumination, which can extend the effective spatial-frequency bandwidth of the imaging system and achieve a transverse resolution of similar to 60 nm (similar to lambda/10). However, multiple images under different illumination pattern is needed, which result in limited imaging speed. A novel interleaved reconstruction method is proposed to increase the frame rate without any change in the raw data of NFOMM. Since allowing easy integration with confocal microscopes, we anticipate this method will be a useful observation tool in the biological community and other research field.

Abstract:In the structured illumination microscopy (SIM) family, interferometric implementations using traditional physical gratings or advanced liquid crystal on silicon spatial light modulators are popular but have the disadvantages of expensive and limited speed. A promising alternative of a digital micromirror device (DMD) has improved the situation for its lower cost and higher speed, furthermore, it has contributed to the proposal of a simple and compact projection DMD-SIM. However, the projection DMD-SIM method is accused of erroneously measuring the resolution because of the scattering characteristic of gold beads, despite the inferior experimental resolution that is because of the low fringe contrast and the resulting low signal-to-noise ratio (SNR). As an improvement, we use a high-brightness laser for illumination to achieve higher SNR, and the accompanying problems of speckle and inhomogeneity originating from the laser’s high coherence are solved by a high-frequency phase-randomization deformable mirror in conjunction with a square-core multimode fiber, which does not limit the image acquisition speed and with fewer energy losses compared to the existing methods. Also, in terms of the problem of low SNR, we apply a parallel-iteration Richardson–Lucy deconvolution algorithm for SIM reconstruction to further improve the imaging performance based on ensuring the resolution. Based on these advances, we image fluorescent nanoparticles and other non-biological and biological fluorescent samples, and the experimental results demonstrate the improved imaging performance of our modified system.

Abstract:Most of the quantitative phase microscopy systems are unable to provide depth-resolved information for measuring complex biological structures. Optical diffraction tomography provides a non-trivial solution to it by 3D reconstructing the object with multiple measurements through different ways of realization. Previously, our lab developed a reflection-mode dynamic speckle-field phase microscopy (DSPM) technique, which can be used to perform depth resolved measurements in a single shot. Thus, this system is suitable for measuring dynamics in a layer of interest in the sample. DSPM can be also used for tomographic imaging, which promises to solve the long-existing “missing cone” problem in 3D imaging. However, the 3D imaging theory for this type of system has not been developed in the literature. Recently, we have developed an inverse scattering model to rigorously describe the imaging physics in DSPM. Our model is based on the diffraction tomography theory and the speckle statistics. Using our model, we first precisely calculated the defocus response and the depth resolution in our system. Then, we further calculated the 3D coherence transfer function to link the 3D object structural information with the axially scanned imaging data. From this transfer function, we found that in the reflection mode excellent sectioning effect exists in the low lateral spatial frequency region, thus allowing us to solve the “missing cone” problem. Currently, we are working on using this coherence transfer function to reconstruct layered structures and complex cells.
High-speed combined reflectance confocal and moxifloxacin based two-photon microscopy.

Abstract:Reflectance confocal microscopy (RCM) is a non-invasive high-resolution optical imaging technique used in clinical settings as a diagnostic method. However, RCM has limited diagnostic ability by providing non-specific morphological information only based on reflection contrast. Various multimodal imaging techniques have been developed to compensate the limitations of RCM, but multimodal techniques are often slow in imaging speed compared to RCM alone. In this report, we combined RCM with moxifloxacin based two-photon microscopy (TPM) for high-speed multimodal imaging. Moxifloxacin based TPM used clinically compatible moxifloxacin for cell labeling and could do non-invasive cellular imaging at 30 frames/s together with RCM. Performance of the combined microscopy was characterized in the imaging of mouse skin and cornea, in vivo. Detail tissue microstructures including cells, extra-cellular matrix (ECM), and vasculature were visualized. The combined microscopy was applied to human skin cancer specimens, and both cells and ECM in the skin cancer and normal skin regions were visualized at high imaging speeds. The combined microscopy can be useful in the clinical applications of RCM by providing multiple contrasts.

Confocal Raman Microscopy with Adaptive Optics

Abstract:Confocal Raman microscopy, a highly specific and label-free technique for the microscale study of thick samples, often presents difficulties due to weak Raman signals. Inhomogeneous samples introduce wavefront aberrations that further reduce these signals, requiring even longer acquisition times. In this study, we introduce adaptive optics to confocal Raman microscopy for the first time to counteract such aberrations, significantly increasing the Raman signal and image quality. The method is designed to integrate seamlessly with existing commercial microscopes without hardware modifications. It uses a wavefront sensorless approach to derive aberrations using an optofluidic, transmissive spatial light modulator that can be attached to the microscope nosepiece. Our experimental results demonstrate the compensation of aberrations caused by artificial scatterers and mouse brain tissue, improving spatial resolution and achieving up to 3.5-fold signal enhancements. Our results provide a basis for the molecular label-free study of biological systems at greater imaging depths.

Abstract:The contrast and resolution of images obtained with optical microscopes can be improved by deconvolution and computational fusion of multiple views of the same sample, but these methods are computationally expensive for large datasets. Here we describe theoretical and practical advances in algorithm and software design that result in image processing times that are tenfold to several thousand fold faster than with previous methods. First, we show that an 'unmatched back projector' accelerates deconvolution relative to the classic Richardson-Lucy algorithm by at least tenfold. Second, three-dimensional image-based registration with a graphics processing unit enhances processing speed 10- to 100-fold over CPU processing. Third, deep learning can provide further acceleration, particularly for deconvolution with spatially varying point spread functions. We illustrate our methods from the subcellular to millimeter spatial scale on diverse samples, including single cells, embryos and cleared tissue. Finally, we show performance enhancement on recently developed microscopes that have improved spatial resolution, including dual-view cleared-tissue light-sheet microscopes and reflective lattice light-sheet microscopes.

Abstract:Stimulated Raman scattering (SRS) microscopy that is capable of both high-speed imaging and rapid spectroscopy will be advantageous for detailed chemical analysis of heterogeneous biological specimens. We have developed a system based on spectral focusing SRS technology with the integration of a rapid scanning optical delay line, which allows continuous tuning of SRS spectra by scanning a galvo mirror. We demonstrate SRS spectral measurements of dimethyl sulfoxide solution at low concentrations and multi-color imaging of rice pollens and HeLa cells with line-by-line delay tuning to reduce motion artifacts, as well as fast acquisition of SRS spectra at specific regions of interest.

Abstract:We present a multimodal in vivo skin imaging instrument that is capable of simultaneously acquiring multiphoton and reflectance confocal images at up to 27 frames per second with 256 × 256 pixel resolution without the use of exogenous contrast agents. A single femtosecond laser excitation source is used for all channels ensuring perfect image registration between the two-photon fluorescence (TPF), second harmonic generation (SHG), and reflectance confocal microscopy (RCM) images. Images and videos acquired with the system show that the three imaging channels provide complementary information in in vivo human skin measurements. In the epidermis, cell boundaries are clearly seen in the RCM channel, while cytoplasm is better seen in the TPF imaging channel, whereas in the dermis, SHG and TPF channels show collagen bundles and elastin fibers, respectively. The demonstrated fast imaging speed and multimodal imaging capabilities of this MPM/RCM instrument are essential features for future clinical application of this technique.

Abstract:Fast confocal imaging was achieved by combining remote focusing with differential spinning disk optical sectioning to rapidly acquire images of live samples at cellular resolution. Axial and lateral full width half maxima less than 5 um and 490 nm respectively are demonstrated over 130 um axial range with a 256 x 128 um field of view. A water-index calibration slide was used to achieve an alignment that minimises image volume distortion. Application to live biological samples was demonstrated by acquiring image volumes over a 24 um axial range at 1 volume/s, allowing for the detection of calcium-based neuronal activity in Platynereis dumerilii larvae.

Versatile high-speed confocal microscopy using a single laser beam

Abstract:We present a new flexible high speed laser scanning confocal microscope and its extension by an astigmatism particle tracking device (APTV). Many standard confocal microscopes use either a single laser beam to scan the sample at relatively low overall frame rate, or many laser beam to simultaneously scan the sample and achieve a high overall frame rate. Single-laser-beam confocal microscope often use a point detector to acquire the image. To achieve high overall frame rates, we use, next to the standard 2D probe scanning unit, a second 2D scan unit projecting the image directly on a 2D CCD-sensor (re-scan configuration). Using only a single laser beam eliminates cross-talk and leads to an imaging quality that is independent of the frame rate with a lateral resolution of 0.235\unit{\mu m}. The design described here is suitable for high frame rate, i.e., for frame rates well above video rate (full frame) up to a line rate of 32kHz. The dwell time of the laser focus on any spot in the sample (122ns) is significantly shorter than in standard confocal microscopes (in the order of milli or microseconds). This short dwell time reduces phototoxicity and bleaching of fluorescent molecules. The new design opens further flexibility and facilitates coupling to other optical methods. The setup can easily be extended by an APTV device to measure three dimensional dynamics while being able to show high resolution confocal structures. Thus one can use the high resolution confocal information synchronized with an APTV dataset.

Fast biological imaging with quantum-enhanced Raman microscopy

Abstract:Stimulated Raman scattering (SRS) microscopy is a powerful label-free imaging technique that probes the vibrational response of chemicals with high specificity and sensitivity. High-power, quantum-enhanced SRS microscopes have been recently demonstrated and applied to polymers and biological samples. Quantum correlations, in the form of squeezed light, enable the microscopes to operate below the shot noise limit, enhancing their performance without increasing the illumination intensity. This addresses the signal-to-noise ratio (SNR) and speed constraints introduced by photodamage in shot noise-limited microscopes. Previous microscopes have either used single-beam squeezing, but with insufficient brightness to reach the optimal ratio of pump-to-Stokes intensity for maximum SNR, or have used twin-beam squeezing and suffered a 3 dB noise penalty. Here we report a quantum-enhanced Raman microscope that uses a bright squeezed single-beam, enabling operation at the optimal efficiency of the SRS process. The increase in brightness leads to multimode effects that degrade the squeezing level, which we partially overcome using spatial filtering. We apply our quantum-enhanced SRS microscope to biological samples, and demonstrate quantum-enhanced multispectral imaging of living cells. The imaging speed of 100x100 pixels in 18 seconds allows the dynamics of cell organelles to be resolved. The SNR achieved is compatible with video rate imaging, with the quantum correlations yielding a 20% improvement in imaging speed compared to shot noise limited operation.

Abstract:Rapid coherent Raman hyperspectral imaging shows great promise for applications in sensing, medical diagnostics, and dynamic metabolism monitoring. However, the spectral acquisition speed of current multiplex coherent anti-Stokes Raman scattering (CARS) microscopy is generally limited by the spectrometer integration time, and as the detection speed increases, the signal-to-noise ratio (SNR) of single spectrum will decrease, leading to a terrible imaging quality. In this Letter, we report a dual-comb coherent Raman hyperspec-tral microscopy imaging system developed by integrating two approaches, a rapid delay-spectral focusing method and deep learning. The spectral refresh rate is exploited by focusing the relative delay scanning in the effective Raman excitation region, enabling a spectral acquisition speed of 36 kHz, approximate to 4 frames/s, for a pixel resolution of 95 x 95 pixels and a spectral bandwidth no less than 200 cm(-1). To improve the spectral SNR and imaging quality, the deep learning models are designed for spectral preprocessing and automatic unsupervised feature extraction. In addition, by changing the relative delay focusing region of the comb pairs, the detected spectral wavenumber region can be flexibly tuned to the high SNR region of the spectrum. Mechanical-scan-free and multi-color super-resolution imaging with diffractive spot array illumination

Abstract:Point-scanning microscopy approaches are transforming super-resolution imaging. Despite achieving parallel high-speed imaging using multifocal techniques, efficient multi-color capability with high-quality illumination is currently lacking. In this paper, we present for the first time Mechanical-scan-free and multi-Color Super-resolution Microscopy (MCoSM) by spot array illumination, which enables mechanical-scan-free super-resolution imaging with adjustable resolution and field of view (FoV) based on spatial light modulators (SLMs). Through 100s-10,000s super-resolution spot illumination with different FoV for imaging, we demonstrate the adjustable capacity of MCoSM. MCoSM extends current spectral imaging capabilities through a time-sharing process of different color illumination with phase-shifting scanning, while retaining the spatial flexibility of super-resolution imaging with diffractive spot array illumination. To showcase the prospects for further combining MCoSM with multi-color imaging, we also perform spectral unmixing (four-colors) on images of fluorescent beads at high resolution. MCoSM provides a versatile platform for studying molecular interactions in complex samples at the nanoscale level.

Abstract:Point-scanning microscopy approaches are transforming super-resolution imaging. Abstract:Abstract Fluorescence microscopy has undergone rapid advancements, offering unprecedented visualization of biological events and shedding light on the intricate mechanisms governing living organisms. However, the exploration of rapid biological dynamics still poses a significant challenge due to the limitations of current digital camera architectures and the inherent compromise between imaging speed and other capabilities. Here, we introduce sHAPR, a high-speed acquisition technique that leverages the operating principles of sCMOS cameras to capture fast cellular and subcellular processes. sHAPR harnesses custom fiber optics to convert microscopy images into one-dimensional recordings, enabling acquisition at the maximum camera readout rate, typically between 25 and 250 kHz. We have demonstrated the utility of sHAPR with a variety of phantom and dynamic systems, including high-throughput flow cytometry, cardiomyocyte contraction, and neuronal calcium waves, using a standard epi-fluorescence microscope. sHAPR is highly adaptable and can be integrated into existing microscopy systems without requiring extensive platform modifications. This method pushes the boundaries of current fluorescence imaging capabilities, opening up new avenues for investigating high-speed biological phenomena.

Abstract:Compared with two-photon point-scanning microscopy, line-scanning temporal focusing microscopy breaks the limitation on imaging rate and maintains the axial resolution, which makes it promising for various biomedical studies. However, for deep tissue imaging, it suffers from reduced axial resolution and increased background noise due to sample induced wavefront distortion. Here, we propose a spatio-spectral focal modulation technique to enhance axial resolution and background rejection by simply subtracting an aberrated image, which is induced by a spatial light modulator, from an unaberrated image. The proposed technique could improve the axial resolution by a factor of 1.3 in our implementation, verified by both simulations and experiments. Besides, we show that compared with spatial modulation alone, spatio-spectral modulation induces less peak intensity loss caused by image subtraction. We further demonstrate the performance of our technique on the enhanced axial resolution and background rejection by deep imaging of cleared mouse brains and in vivo imaging of living mouse brains.

Abstract:Laser-scanning confocal microscopy serves as a critical instrument for microscopic research in biology. However, it suffers from low imaging speed and high phototoxicity. Here we build a novel deep compressive confocal microscope, which employs a digital micromirror device as a coding mask for single-pixel imaging and a pinhole for confocal microscopic imaging respectively. Combined with a deep learning reconstruction algorithm, our system is able to achieve high-quality confocal microscopic imaging with low phototoxicity. Our imaging experiments with fluorescent microspheres demonstrate its capability of achieving single-pixel confocal imaging with a sampling ratio of only approximately 0.03% in specific sparse scenarios. Moreover, the deep compressive confocal microscope allows single-pixel imaging at the single-photon level, thus reducing the excitation light power requirement for confocal imaging and suppressing the phototoxicity. We believe that our system has great potential for long-duration and high-speed microscopic imaging of living cells.

Abstract:The ability to image at high speeds is necessary for biological imaging to capture fast-moving or transient events or to efficiently image large samples. However, due to the lack of rigidity of biological specimens, carrying out fast, high-resolution volumetric imaging without moving and agitating the sample has been a challenging problem. Pupil-matched remote focusing has been promising for high NA imaging systems with their low aberrations and wavelength independence, making it suitable for multicolor imaging. However, owing to the incoherent and unpolarized nature of the fluorescence signal, manipulating this emission light through remote focusing is challenging. Therefore, remote focusing has been primarily limited to the illumination arm, using polarized laser light to facilitate coupling in and out of the remote focusing optics. Here, we introduce a novel optical design that can de-scan the axial focus movement in the detection arm of a microscope. Our method splits the fluorescence signal into S and P-polarized light, lets them pass through the remote focusing module separately, and combines them with the camera. This allows us to use only one focusing element to perform aberration-free, multi-color, volumetric imaging without (a) compromising the fluorescent signal and (b) needing to perform sample/detection-objective translation. We demonstrate the capabilities of this scheme by acquiring fast dual-color 4D (3D space + time) image stacks with an axial range of 70 μm and camera-limited acquisition speed. Owing to its general nature, we believe this technique will find its application in many other microscopy techniques that currently use an adjustable Z-stage to carry out volumetric imaging, such as confocal, 2-photon, and light sheet variants.
