Abstract:AbstractA rapid and facile method is reported for the detection of ascorbic acid using molybdenum disulfide quantum dots (MoS2 QDs) as a fluorescence sensor. Water-soluble and biocompatible MoS2 QDs with the maximum fluorescence emission at 506 nm have been successfully synthesized by hydrothermal method and specific detection for ascorbic acid (AA) was constructed to utilize the modulation of metal ion on the fluorescence of MoS2 QDs and the affinity and specificity between the ligand and the metal ion. The fluorescence of MoS2 QDs was quenched by the irreversible static quenching of Fe3+ through the formation of a MoS2 QDs/Fe3+ complex, while the pre-existence of AA can retain the fluorescence of MoS2 QDs through the redox reaction between AA and Fe3+. Based on this principle, a good linear relationship was obtained in the AA concentration range 1 to 150 μM with a detection limit of 50 nM. The proposed fluorescent sensing strategy was proven to be highly selective, quite simple, and rapid with a requirement of only 5 min at room temperature (RT), which is particularly useful for rapid and easy analysis. Satisfactory results were obtained when applied to AA determination in fruits, beverages, and serum samples as well as AA imaging in living cells, suggesting its great potential in constructing other fluorescence detection and imaging platforms.Graphical abstract

Abstract:A nitrogen and sulfur co-doped graphene quantum dots (N/S-GQDs) and permanganate was employed to prepare a N/S-GQDs/KMnO 4 dual-signal readout colorimetric and fluorescent probe for sensitive detection of ascorbic acid (AA). In the detection system, the addition of KMnO 4 quenched significantly the fluorescence (FL) of N/S-GQDs, meanwhile, the transparent aqueous dispersion of N/S-GQDs changed into yellow–brown with new absorption peak (maximum absorption at 338 nm). Interestingly, introduction of AA into the N/S-GQDs/KMnO 4 system could recover the quenched fluorescence and result in a decrease in the absorbance (at 338 nm) of the test solution in a concentration dependent manner, and hence demonstrated the potential practical use of the sensing system for AA detection. Based on various spectroscopic studies, it has been concluded that the static as well as inner filter effect was the main quenching mechanisms in the detection system. The dual-response sensor showed linear response in the concentration range between 0.01 and 30 μM with the limit of detection (LOD) of 8.3 nM and 100 nM, respectively, for the fluorescent and colorimetric detections. In addition, the proposed detection system has been successfully applied to the detection of AA in vitamin C tablets. Moreover, the proposed detection strategy is convenient and fast, prevents complex and tedious probe preparation procedure, and capable of visual detection of AA.

Abstract:A novel magnetic/fluorometric bimodal sensor was built from carbon dots (CDs) and MnO2. The resulting sensor was sensitive to glutathione (GSH), leading to apparent enhancement of magnetic resonance (MR) and fluorescence signals along with visual changes. The bimodal detection strategy is based on the decomposition of the CDs-MnO2 through a redox reaction between GSH and MnO2. This process causes the transformation from non-MR-active MnO2 to MR-active Mn(2+), and is accompanied by fluorescence restoration of CDs. Compared with a range of other CDs, the polyethylenimine (PEI) passivated CDs (denoted as pCDs) were suitable for detection due to their positive surface potential. Cross-validation between MR and fluorescence provided detailed information regarding the MnO2 reduction process, and revealed the three distinct stages of the redox process. Thus, the design of a CD-based sensor for the magnetic/fluorometric bimodal detection of GSH was emphasized for the first time. This platform showed a detection limit of 0.6 μM with a linear range of 1-200 μM in the fluorescence mode, while the MR mode exhibited a linear range of 5-200 μM and a GSH detection limit of 2.8 μM with a visible change being observed rapidly at 1 μM in the MR images. Furthermore, the introduction of the MR mode allowed the biothiols to be easily identified. The integration of CD fluorescence with an MR response was demonstrated to be promising for providing detailed information and discriminating power, and therefore extend the application of CDs in sensing and imaging.

Abstract:Stream processing applications run continuously and have varying load. Cloud infrastructures present an attractive option to meet these fluctuating computational demands. Coordinating such resources to meet end-to-end latency objectives efficiently is important in preventing the frivolous use of cloud resources. We present a framework that parallelizes and schedules workflows of stream operators, in real-time, to meet latency objectives. It supports data- and task-parallel processing of all workflow operators, by all computing nodes, while maintaining the ordering properties of sorted data streams. We show that a latency-oriented operator scheduling policy coupled with the diversification of computing node responsibilities encourages parallelism models that achieve end-to-end latency-minimization goals. We demonstrate the effectiveness of our framework with preliminary experimental results using a variety of real-world applications on heterogeneous clusters.

Abstract:Glutathione (GSH) is a crucial non-protein thiol and an indispensable endogenous antioxidant. The aberrant expression of GSH in plasma and cytosol is closely related to numerous diseases, including cancer. Therefore, establishing a sensitive method for analyzing GSH has important application value for biomedical research and clinical medical detection. Herein, A method for the rapid and simple detection of GSH was proposed, which is based on an anti-etching mechanism by utilizing gold nanotetrapods (Au NTPs) and manganese dioxide nanosheets (MnO 2 NSs). In the absence of GSH, Au NTPs solution can cause a distinct color change from gray-green to red through the etching effect of MnO 2 NSs. However, in the presence of GSH, the redox reaction between GSH and MnO 2 NSs inhibits the etching of Au NTPs by MnO 2 NSs, and Au NTPs solution maintains persistent gray-green color. The colorimetric probe exhibited excellent selectivity for GSH. The limits of detection for GSH were 43.5 nM (UV–vis spectrum) and 0.25 μM (naked eyes). The sensing technique exhibited excellent linearity between wavelength shift and GSH concentration within the range of 0.25 μM–1.5 μM. The outcomes of GSH detection in actual biological samples demonstrate that this probe has the potential to be applied to GSH detection in intricate biological samples.

Abstract:A method is described for the determination of ascorbic acid (AA) in complex biological fluids. It based on maganese(II)-doped zinc/germanium oxide nanoparticles (Mn@ZnGe NPs) with appealing time-resolved phosphorescence (TRP). TRP can provide a background-free reporter signal in analytical methods. The absorption of AA overlaps the excitation band of Mn@ZnGe NPs at 254 nm. This reduces the intensity of fluorescence via an inner filter effect (IFE) with increasing concentration of AA. Typical experimental conditions include an emission peak at 536 nm, a delay time of 50 μs and a counting time of 2 ms. This method can detect AA in a range of 5–500 μM with a 0.13 μM limit of detection. If AA is oxidized by the enzyme AA oxidase (AAOx), dehydroascorbic acid will be formed which doesn't absorb at 254 nm. Hence, the IFE cannot occur and fluorescence is not reduced. The strategy can be used to quantify AAOx in the activity range of 1–4 U·mL−1. By using a handheld UV lamp and a smart phone with a color-scanning feature, the feasibility for visual detection and real-time/onsite quantitative scanometric monitoring of AA and AAOx is demonstrated.Schematic presentation of a fluorometric method for determination of ascorbic acid (AA) and ascorbic oxidase and a scanometric visual assay. Abstract:Glutathione (GSH) as a biothiol is an essential peptide related to various diseases. Although multiple strategies for biothiols detection have been developed, there is increasing demand for sensors that can differentiate GSH from cysteine (Cys) and homocysteine (Hcy), owing to the similar structures and thiol groups in these amino acids. Herein, we report a novel Eu3+/LAPONITE (Lap)-based organic/inorganic hybrid material for selective detection of GSH via an "off-on" process. The fluorescence of Eu(DPA)3@Lap-Tris can be quenched by Cu2+ through photoinduced electron transfer (PET). The addition of GSH into the Eu(DPA)3@Lap-Tris/Cu2+ system induces the removal of Cu2+ from Eu(DPA)3@Lap-Tris and blocks PET, resulting in the recovery of fluorescence. This proposed assay demonstrates higher selectivity toward GSH than Cys and Hcy, and showed a detection limit of 162 nM within a linear range of 0.5-30 μM. Unlike other GSH selective sensors, this platform could be formed into a hydrogel while its sensitivity was maintained. The sensitive response to GSH in serum samples makes this platform an efficient tool for biological applications because of its ease of preparation, high selectivity, good biocompatibility, and low toxicity.
Dual Colorimetric and Ratiometric Fluorescent Responses for the Determination of Glutathione Based on Fluorescence Quenching and Oxidase-Like Activity of MnO2 Nanosheets

Abstract:A dual colorimetric and ratiometric fluorescent assay is constructed for the detection of glutathione (GSH) based on MnO2 nanosheets (MnO2 NS). Amplex Red (AR) and curcumin ( CUR) fluorescence-pairing probes have opposite responses to MnO2 NS with fluorescence quenching and oxidase-like property. MnO2 nanosheets can largely quench the fluorescence of curcumin. However, the fluorescence of oxidized AR (oxAR) was obviously raised via the catalytic oxidation reaction of no fluorescent AR. The color of the solution gradually changes from yellow to orange. However, MnO2 nanosheets lose the fluorescence quenching capacity and oxidase-like property after the addition of GSH. Therefore, the fluorescence intensity of curcumin shows an obvious increase and the intensity of oxAR a decrease, accompanied by the restoration of the solution color from orange to yellow. For MnO2 nanosheets, the linear relationship between the absorbance ratio of the two substrates and the GSH concentration (50-15 000 nM) is obtained with the detection limit of 16 nM. Besides, the ratiometric fluorescent method has a good linear range from 10 to 5000 nM for GSH. The detection limit is as low as 4.2 nM. Further, this method can not only be used for the determination of GSH in an aqueous solution but also shows a high responsive ability to GSH in human serum. This result indicates its potential in bioanalysis and disease diagnosis.

Abstract:We report a novel design, based on a combination of lanthanide-doped upconversion nanoparticles and manganese dioxide nanosheets, for rapid, selective detection of glutathione in aqueous solutions and living cells. In this approach, manganese dioxide (MnO(2)) nanosheets formed on the surface of nanoparticles serve as an efficient quencher for upconverted luminescence. The luminescence can be turned on by introducing glutathione that reduces MnO(2) into Mn(2+). The ability to monitor the glutathione concentration intracellularly may enable rational design of a convenient platform for targeted drug and gene delivery.

Abstract:g-C3N4/BSA@MnO2 fluorescent off-on probe was constructed for sensitive and selective detection of ascorbic acid (AA). BSA@MnO2 nanoparticles using bovine serum albumin (BSA) as a template were anchored on the surface of graphite-phase carbon nitride nanosheets (g-C3N4 NSs) via electrostatic attraction to synthesize the g-C3N4/BSA@MnO2 nanocomposites. The fluorescence of g-C3N4 NSs was quenched by BSA@MnO2 nanoparticles (NPs) via fluorescence resonance energy transfer (FRET) and recovered in the presence of AA via the redox reaction between AA and MnO2. The nanocomposites therefore realized the fluorescent off–on detection of AA. Asatisfactory linear range of 0–120 μM and a limit of detection of 133 nM were obtained. The nanoprobe was applied for the detection of AA in food and drug samples as well as the fluorescent images of exogenous AA in Raw cells. The results showed that the nanoprobe possessed excellent properties for AA detection and broadened the biochemical analysis applications of g-C3N4-based nanomaterials.

Abstract:In this work, a ratiometric fluorescent nanoplatform for the detection of ascorbic acid (AA) was constructed based on the Ag2S quantum dots (QDs) and multifunctional hydroxyl cobalt oxide nanoflakes (CoOOH NFs). Ag2S QDs can be assembled on the surface of CoOOH NFs by electrostatic adsorption, resulting in the quenching of the NIR fluorescence emission of Ag2S QDs at 680 nm effectively through the inner filter effect (IFE). o-Phenylenediamine (OPD), a common substrate of oxidase-like (OXD) mimic, is rapidly oxidized into the fluorescent product of 2,3-diaminophenazine (DAP) with the appearance of an emission peak at 575 nm under the catalysis of CoOOH NFs. After AA was added, the fluorescence emission of DAP declined because of the decline in the OXD-like activity of CoOOH NFs due to the transformation of Co2+. Simultaneously, Ag2S QDs were released, accompanied by the recovery of red fluorescence. These two fluorescent signals can be excited at the same excitation wavelength, simplifying the detection procedure. Using F575/F680 as the readout, the quantification of AA can be realized with the linear range and detection limit of 0.2 μM-20 mM and 0.014 μM, respectively. The ratiometric fluorescence sensor can be effectively used to determine the content of AA in real samples such as juice and serum. This work integrates the in-situ formation of the fluorescent species via the catalysis of the nanozyme and the redox reaction to destroy the CoOOH NFs nanozyme as well as the two dimensional nanoflake induced turn-off-on strategy for Ag2S QDs, which provides a specific strategy for the selective detection of AA and may offer a reliable approach for the construction of other biosensing platforms.

Abstract:Glutathione (GSH) plays an important role in cells, which is an essential endogenous antioxidant. Here, we have developed a new detection platform to analyze GSH levels. In our system, fluorescent polydopamine (PDA) nanoparticles, as signal indicators, were obtained by oxidation through cobalt oxyhydroxide (CoOOH) nanosheets. When CoOOH was present, CoOOH could quickly oxidize dopamine to fluorescent PDA nanoparticles. However, once GSH existed, CoOOH nanosheets were decomposed into Co2+, and oxidation between CoOOH and dopamine was prevented with weaker fluorescence occurring. Thus, we could realize detection of the GSH concentration according to the decreased fluorescence value of the fluorescent polydopamine. This method provides a fast, simple, high sensitivity and desirable selectivity platform for GSH monitoring.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Ascorbic acid (AA) detection in biological sample and food sample is critical for human health. Herein, a MnO2 nanosheet (MnO2-NS)-based ratiometric fluorescent nanosensor has been developed for high sensitive and specific detection of AA. The MnO2-NS presents peroxidase-like activity and can oxidize non-fluorescent substrate of o-phenylenediamine (OPDA) into fluorescent substrate, presenting maximum fluorescence at 568 nm (F568). If MnO2-NS is premixed with AA, the MnO2-NS is then decomposed as Mn2+ by AA, decreasing the fluorescent intensity of F568. Meantime, AA is oxidized as dehydroascorbic acid (DHAA), which can react with OPDA to generate fluorescent substrate. A new fluorescence response is found at 425 nm (F425). The dual fluorescent responses can be excited with a universal excitation wavelength, simplifying the detection procedure. With F425/F568 as readout, limit of detection for AA reaches as low as 10.0 nM. Satisfactory recoveries are found for AA detection in serum and diverse beverages. The ratiometric strategy significantly eliminates false-negative and false-positive results, providing a cost-effective, rapid, and reliable way for AA detection in real sample.

A Non-Oxidation Reduction Strategy for Highly Selective Detection of Ascorbic Acid by “On-off-on” Photoluminescence Switches

Abstract:Great progress was made to detect ascorbic acid (AA) based on the photoluminescent nanoprobes/oxidant complexes by using the reducibility of AA. However, some reducing substances including cysteine and gluta-thione disturbed the accuracy of AA assay. Herein, a non-oxidation reduction regulation strategy was fabricated to construct a novel "on-off-on" photoluminescence model for highly selective determination of AA. Initially, copper nanoclusters capsulated into zeolitic imidazolate framework-8 (CuNCs@ZIF-8) was achieved. And then inorganic phosphate ions quenched the emission of CuNCs@ZIF-8 owing to the decomposition of ZIF-8 shell, rendering the signal in "turn-off" state. The linear range for phosphate ions was 1.0-60 mu M with LOD of 0.35 mu M. Interestingly, upon introducing AA into CuNCs@ZIF-8/phosphate, the emission of CuNCs@ZIF-8 recovered possibly due to the electronegativity improvement of metal core and surface rigidified through hydrogen bond interaction, and the system signal was "turned-on". The LOD of AA was as low as 51 nM in a good linear range from 0.1 to 40 mu M. Furthermore, CuNCs@ZIF-8 were used for AA detecting in tablets, beverages, and fresh fruits with the satisfactory recoveries from 96% to 104%. More importantly, a paper-based analytical device containing CuNCs@ZIF-8/phosphate were evaluated for the visual sensing of AA in food safety.
Characterization of thermal transport in multiwalled carbon nanotube using FIB irradiation

Abstract:Thermal conductance of an individual multiwalled carbon nanotube was measured as a function of its length. Focused ion beam was used to shorten a 4.8 micrometer-long nanotube to 2.4, 1.2, 0.6, and 0.3-micrometer-long specimens on a hot-film sensor. As the nanotube is shorten, the conductance decreases more than expected by the diffusive thermal conduction theory. We treated two nanotubes of 64nm and 85nm diameters, both of which showed quasi-ballistic phonon transport. This is the first experiment to quantitatively investigate the contribution of phonons with long free paths in multiwalled carbon nanotube. The principle of thermal measurement, amorphous carbon induced by the ion beam, and considerable errors are also explained.

Glutathione (GSH) is one of the most abundant bioethanol antioxidants in living cells. Here, a fluorescent probe based on MnO2 nanosheets and sulfur quantum dots (SQDs) was fabricated. Because of the synergistic effect of IFE and FRET, the fluorescence from SQDs was quenched by MnO2 nanosheets. In the presence of GSH, the fluorescence of SQDs could be recovered because of the reduction of MnO2 nanosheets by GSH. The method can detect GSH in the concentration range of 5 ~ 1000 μM with the detection limit as low as 1.26 μM. This quick, easy, and cost‐effective sensor could be used for the quantification of GSH in serum samples and the imaging of GSH in Escherichia coli O157:H7.

Abstract:Glutathione (GSH), as a key regulator for the redox balance of biological processes, was detected with a metal-organic framework (MOF) (UiO-67-sbdc) fluorescence sensor. The fluorescence of UiO-67-sbdc could be effectively enhanced by GSH via an electronic effect between GSH and the ligand of MOF, thus achieving the recognition of GSH. It is the first attempt to design a MOF-based fluorescent sensor to distinguish GSH from cysteine (Cys). This sensor exhibited short response time (< 120 s), suitable linear range, and high selectivity towards GSH. Further research conducted in serum also showed a quantitative and selective detection of GSH. Thus, the reported UiO-67-sbdc in this work has its application prospect in clinically judging antioxidant capacity of the body.
“Turn-Off-On” Fluorescence Switching of Ascorbic Acid-Reductive Silver Nanoclusters: a Sensor for Ascorbic Acid and Arginine in Biological Fluids

Abstract:In this report, a novel one-step chemical reduction method was reported for synthesis of water-soluble and stable fluorescent glutathione-templated silver nanocluster (GSH-Ag NCs) with ascorbic acid as an environmental-friendly reducing agent. On the basis of an oxidoreduction-induced fluorescence quenching mechanism, the prepared GSH-Ag NCs found to act as a cheap, non-toxic and highly sensitive “turn-off” fluorophore for ascorbic acid (AA). Furthermore, the fluorescence of the fluorophore/AA system could be recovered through addition of arginine (Arg), which made the system function as a highly selective “turn-on” sensor for arginine. Therefore, a “turn-off-on” switch sensor was proposed for detection of AA and Arg. Under optimized conditions, the probe gives a fluorescent response that is linear in the 2–300 μM concentration range of AA, with a detection limit of 0.1 μM. The probe for Arg, in turn, has a linear range in the 10–180 μM concentration range, and the limit of detection is 0.5 μM. In addition, the developed method showed great accuracy when employed to detect AA and Arg in human urine and serum, which shows its great potential in biological molecular recognition applications.
