Abstract:Objective: This study examined the effectiveness of an outpatient programme on the leisure lifestyle of high-functioning young adults living at home or at an independent setting. Methods: A pre-test–post-test control group design was used. Participants (n = 12) completed self-reports on (a) need for leisure support, (b) leisure engagement and (c) satisfaction with leisure lifestyle. The programme consisted of cognitive-behavioural techniques. Results: Significant within-group changes for the experimental group (n = 7) were found, indicating decreases in ‘need for leisure support’, more regular leisure engagement patterns and an increase in ‘leisure satisfaction’ following the programme. Between-group statistics reveal medium and large effect sizes for decreases in need for leisure support and a medium effect size for increase in ‘leisure satisfaction’, all in favour of the experimental group. Results regarding ‘leisure engagement’ were less clear. Conclusion: The preliminary programme was effective in improving leisure lifestyle and suggestions for future research are discussed.

Abstract:The common liver fluke, Fasciola hepatica, is a trematode parasite found worldwide, typically with a focal distribution due to its requirement for suitable climatic and environmental conditions to complete its lifecycle. Bovine fasciolosis causes suboptimal production and economic losses, including liver condemnation at slaughter. The lack of reliable diagnostic methods is a disadvantage to the increasing demand for surveillance and control. The aim of this study was to evaluate the diagnostic accuracy of bulk tank milk (BTM) antibody testing and aggregated abattoir registrations (AAR) of liver fluke as herd-level tests for F. hepatica infection using Bayesian latent class models. Data from the abattoirs in 2019-2021 and BTM, sampled in the winter of 2020/2021, were collected from 437 herds on the southwest coast of Norway. The BTM samples were analysed with the SVANOVIR® F. hepatica-Ab ELISA test, with results given as an optical density ratio (ODR), and later dichotomized using the recommended cut-off value from the test manufacturer (ODR ≥0.3). Based on the BTM ELISA test, 47.8% of the herds tested positive. The AAR test was defined as the herd-level proportion of female slaughtered animals registered with liver fluke infection during the study period. For this test, three cut-offs were used (a proportion of 0.05, 0.1 and 0.2). The herds were split into two subpopulations ("Coastal" and "Inland"), which were expected to differ in true prevalence of F. hepatica infection based on climate-related and geographical factors. The diagnostic accuracies of both tests were estimated using Bayesian latent class models with minimally informative priors. Post-hoc analysis revealed that the maximum sum of sensitivity (Se) and specificity (Sp) of the tests was achieved with a herd-level proportion of ≥0.1 registered with liver fluke as the AAR test. Using this cut-off, the median estimate for the diagnostic accuracy of the BTM ELISA was 90.4% (84.0-96.2 95% Posterior Credible Interval (PCI)) for Se and 95.3% (90.6-100% PCI) for Sp, while the median estimate of Se for AAR was 87.5% (81.4-93.1% PCI) and the median estimate of Sp for AAR was 91.0% (85.2-96.5% PCI). The cut-off evaluation of the SVANOVIR® F. hepatica-Ab ELISA test for BTM confirmed the manufacturer's recommended cut-off of ODR ≥0.3 to denote positive and negative herds. This study suggests that AAR and BTM ELISA test can be used as herd-level tools to monitor liver fluke infection, so that appropriate interventions against infection can be implemented as necessary.

Abstract:A comparison has been made of the sequence of the tyrosine-containing tryptic peptide derived from the purified, unfractionated, lysine-rich histone complement of the calf thymus, rat thymus and calf liver. Two tyrosine-containing peptides, one of which is a fragment of the other, have been obtained from each tissue. The major tyrosine-containing peptide was isolated by a single electrophoretic step in about a 50%, yield. The results indicate that the sequence of amino acids around the single tyrosine present in the lysine-rich molecule is constant from tissue to tissue. However, this sequence varies among the molecular species comprising the lysine-rich histone complement of a single tissue. Within a tissue, a conservative amino acid replacement in the residue penultimate to tyrosine (from the amino terminus) has been detected. The sequence of the tyrosine-containing region was found to be Lys-Ala-Leu-Ala-AlaGly-Gly-Tyr-Asp-Val-Glu-Lys.
Goats are more susceptible to Haemonchus contortus infection than sheep under similar experimental settings

Abstract:Due to differences in their feeding behavior, sheep and goats are often assumed to respond differently to helminth infections. The present study compared Haemonchus contortus infection profile between sheep and goats under the same experimental setting. Experimental infection was conducted using a randomized block design in four groups of intact sheep (InfSH and ConSH) and goats (InfG, and ConG). Groups InfSH and InfG ( N = 7 each) received 10,000L3 of H. contortus whereas the control groups ConSH and ConG ( N = 7 each) remained uninfected. Faecal egg counts and PCV were measured from Day 0 to day 56 post infection (PI). On day 56 PI, animals were humanely slaughtered and abomasal contents were recovered to measure worm burden, worm length and in utero egg count. The findings show that: (1) Haemonchus infected animals showed an increase in FEC starting from day 21 PI, (2) progressive reduction in PCV was registered from day 7 PI and continued to the end of the experiment while this remained at pre-infection levels in control groups, (3) FEC was much higher ( P < 0.001) and PCV was significantly lower ( P < 0.05) in infected goats than in infected sheep, (4) at necropsy, total worm burden with worm establishment rates of 63% and 28.87% were registered respectively for infected goats and sheep with significant difference ( P < 0.05), (5) Female worms were significantly longer ( P < 0.05) in InfG (22.8(± 1.2) compared to InfSH (20.5 ± 0.67 mm) while (6) Mean worm fecundity was 974.8 ± 239.4 and 1162.5 ± 89.4 respectively for groups InfSH and InfG with no significant difference ( P = 0.07), and (7) Parasite traits such as worm burden, FEC and female worm length were well correlated in sheep whereas few such patterns were observed in goats. In conclusion, under the same experimental infection, Arsi-Bale goats are more susceptible to H. contortus infection than Arsi-Bale sheep and hence deserve special attention when they are forced to live on grazing rather than browsing.

Ethnic-specific body mass index cut-off points for overweight and obesity in girls.

Abstract:AIM To develop ethnic-specific body mass index (BMI) cut-off points for overweight and obesity in girls from New Zealand's five major ethnic groups. METHODS A total of 1676 girls (41% European, 21% Pacific Island, 15% East Asian, 13% Māori, and 11% South Asian) aged 5-16 years participated in this study. BMI was determined from height and weight, and body fat percentage (%BF) was obtained from hand-to-foot bioelectrical impedance measurements. Using stepwise multiple regression, a series of ethnic-specific BMI cut-off points were developed that corresponded to the equivalent %BF of European girls at the BMI reference values provided by the International Obesity TaskForce (IOTF). RESULTS The adjusted cut-off points for overweight and obesity ranged from an average of 3.3 and 3.8 kg.m(-2) (respectively) lower than the IOTF standards in South Asian girls to 1.5 and 1.9 kg.m(-2) higher in Pacific Island girls. CONCLUSION We conclude that the ethnic-specific BMI cut-off points developed in this study are more appropriate than universal definitions of overweight and obesity for predicting excess adiposity in New Zealand girls.
Prevalence and risk factors related to spp. transmission in Northern Vietnam: A cross-sectional study in multiple hosts

Abstract:Background : Fascioliasis, caused by Fasciola hepatica and Fasciola gigantica, is a global veterinary problem in livestock and an emerging zoonotic disease in various countries. Here we present prevalence estimates of Fasciola spp. in all hosts involved in the life cycle and identified risk factors associated with Fasciola transmission in northern Vietnam. Methodology/Principal findings : We conducted a cross-sectional survey in a community in Nghe An province where fascioliasis is endemic and inhabitants commonly consume raw vegetables. Applying a simple random and cluster sampling approach, we collected 1,137 stool and 1,390 blood samples from 1,396 human participants. From 664 buffaloes and cattle, we collected 656 fecal and 534 blood samples. We also collected 330 lymnaeid snails and 233 water plant samples. Human and animal fecal samples were examined by copro-microscopy, while blood samples were screened by ELISA to detect Fasciola serum antibodies. We examined infections in snails using PCR and metacercariae on water plants deploying an in-house technique. Descriptive analysis and logistic regression models were applied to estimate the prevalence of and risk factors for Fasciola infections. While the prevalence of Fasciola infections was very low in humans (0% by copro-microscopy; 0.07% by ELISA), it was high in animals (52% by copro-microscopy; 54% by ELISA). In the multivariable analysis, age was the only factor associated with Fasciola infections in animals. Fasciola could not be detected in any of the sampled water plants or lymnaeid snails.. Conclusion/Significance: This study indicated a high prevalence of Fasciola infections in animals and a very low prevalence in humans in a rural community in northern Vietnam. It is recommended to implement a control program to reduce the infection rate in buffaloes and cattle. Furthermore, health education activities for livestock owners should be carried out in Fasciola endemic areas.

Abstract:Bovine fasciolosis is a parasitic disease of cattle caused by ingesting metacercariae of liver flukes from the genus Fasciola. A cross-sectional study was conducted from November 2021 to April 2022, encompassing a total of 384 cattle randomly selected from diverse locations. The study included cattle of various ages and genders, aiming to to determine the prevalence of bovine fasciolosis and its associated risk factors in and around Bahir Dar, Ethiopia. Fecal examinations were performed to detect fluke eggs, and data were analyzed using descriptive statistics in STATA Version 20.0 to summarize bovine fasciolosis prevalence. Chi-square tests assessed relationships with infection rates and risk factors, with significance set at P < 0.05. Out of 384 fecal samples analyzed, 49.21% tested positive for Fasciola eggs. The prevalence of the condition varied significantly by origin, with rates of 50% in Tikurit, 61.84% in Sebatamit, 27.65% in Latammba, and 59.37% in Kebele 11. In terms of body condition, prevalence was notably higher in cattle classified as poor (64%) compared to those in medium condition (50%) and fat cattle (26.96%), with these differences being statistically significant. Age also influenced prevalence, with young cattle having a rate of 50.38%, adults 47.33%, and old cattle 50.47%; however, these differences were not statistically significant. Sex did not significantly impact prevalence, with males at 49.73% and females at 48.73%. Additionally, local cattle exhibited a prevalence of 51.62%, whereas crossbreeds had a prevalence of 46.15%, with no significant differences observed between these groups (p < 0.05). The persistently high rates of bovine fasciolosis highlight the urgent need for comprehensive control measures. Implementing enhanced management practices to minimize cattle exposure to contaminated pastures and water sources is essential for effective prevention and control of this parasitic infection.

Abstract:Abstract Background Nature conservation with reduced drainage of pastures has been increasingly promoted in agriculture in recent years. However, moisture on pastures is a crucial factor for the development of free-living stages of many parasite species in ruminants. Hence, for the first time, we conducted a field study between 2015 and 2017 at the German North Sea coast to investigate the long-term effect of pasture rewetting (since 2004) on endoparasite infections in sheep and cattle. Methods We examined faecal samples of 474 sheep and 646 cattle from five farms in spring, summer and autumn each year for the presence of endoparasite infections. Animals were kept on conventionally drained, undrained and rewetted pastures. The association between pasture rewetting and endoparasite infection probability was analysed in generalized linear mixed models and including further potential confounders. Results Infection frequencies for gastrointestinal strongyles, Eimeria spp. and Strongyloides papillosus were significantly higher in sheep (62.9%, 31.7% and 16.7%) than in cattle (39.0%, 19.7% and 2.6%). Fasciola hepatica was detected with a frequency of 13.3% in sheep and 9.8% in cattle, while rumen fluke frequency was significantly higher in cattle (12.7%) than in sheep (3.8%). Nematodirus spp., lungworms (protostrongylids, Dictyocaulus viviparus ), Moniezia spp., Trichuris spp. and Dicrocoelium dendriticum were identified in less than 7% of samples. Co-infection with more than three endoparasite taxa was present significantly more often in sheep than in cattle. We identified significant positive correlations above 0.2 for excretion intensities between S. papillosus with strongyles, Eimeria spp. and Nematodirus spp. in sheep and between strongyles and Nematodirus spp. in cattle. Pasture rewetting had no long-term effect on endoparasite infections, neither in sheep nor in cattle. Interestingly, F. hepatica infections decreased significantly in sheep and cattle from 2015 (10.9% and 13.9%) to 2017 (1.4% and 2.1%). Conclusions Pasture rewetting for nature conservation did not increase endoparasite infection probability in ruminants in the long term. This finding should be confirmed in ongoing studies aimed at further animal welfare parameters. The rapid decrease in F. hepatica infections over 3 years may suggest climatic impact or competition with rumen flukes in addition to potential anthelmintic treatment after feedback of the results to the farmers. Abstract:Giardia duodenum (G. duodenalis) can cause giardiasis and infect a variety of hosts. So far, there have been no detailed data regarding the positive rate of G. duodenalis in sheep and goats in China. Here, a systematic literature review was carried out to investigate the epidemiology of G. duodenalis in sheep and goats in China. To perform the meta-analysis, the databases CNKI, VIP, WanFang, PubMed, Web of science and ScienceDirect were employed for screening studies related to the prevalence of G. duodenalis in sheep and goats in China. The total prevalence of G. duodenalis in sheep and goats was estimated to be 7.00% (95% CI: 4.00-10.00). In the age subgroup, the prevalence of G. duodenalis in sheep and goats of >12 months (11.29%; 95% CI: 8.08-14.97) was higher than that in sheep and goats of ≤12 months (7.57%; 95% CI: 3.95-12.24). An analysis based on seasons showed that the prevalence of G. duodenalis in sheep and goats was higher in summer (11.90%; 95% CI: 0.50-35.05) than that in other seasons. The prevalence of G. duodenalis in sheep and goats after 2016 was 8.57% (95% CI: 5.34-11.79), which was higher than others. The highest prevalence of G. duodenalis in sheep and goats was 13.06% (95% CI: 6.26-19.86) recorded in Southwestern China. The prevalence of Giardia infection in sheep (7.28%; 95% CI: 2.30-14.73) was higher than that in goats (5.43%; 95% CI: 2.73-8.98). The NOAA's National Center for Environmental Information (https://gis.ncdc.noaa.gov/maps/ncei/cdo/monthly) was used to extract relevant geoclimatic data (latitude, longitude, elevation, temperature, precipitation, humidity, and climate). By analyzing the data of each subgroup, it was shown that region, genetype, and climate were potential risk factors for giardiasis prevalence in sheep and goats. Based on the analysis of common factors and geographical factors, it is recommended to strengthen effective management measures (e.g. ventilation and disinfection in warm and humid areas) and formulate relevant policies according to local conditions. Breeders should strengthen the detection of G. duodenalis in sheep and goats, customize corresponding control measures according to the diet and living habits of sheep and goats, and strengthen the protection of sheep and lamb calves, so as to reduce the incidence rate and reduce the economic loss of China's animal husbandry.

Abstract:Among different climatic zones in India, the tropical monsoon region comprises a diverse ecosystem characterized by the endemic nature of several parasites including certain emerging and re-emerging vector-borne pathogens of humans, whereas a systematic investigation of the occurrence of different parasites among domestic goats in this area is not yet explored. The goal of the present study is to explore the parasite diversity focusing on molecular identification of vector-borne hemoparasites and its health impacts on domestic goats reared in the tropical monsoon climate zone of Kerala, India. Among 227 goats presented to the Teaching Veterinary Clinical Complex (TVCC) in the monsoon months of 2023, thirty animals were recruited for the study. The animals were screened for the presence of different hemoparasites ( Anaplasma spp., Theileria spp., and Babesia spp), ectoparasites (ticks, lice, and fleas), and gastrointestinal (GI) parasites (hookworms, threadworms, tapeworms, whipworms, and coccidia). The isolated hemoparasites were further characterized by sequencing and phylogenetic analysis. The correlation studies to elucidate the association between the occurrence of different parasites and clinical manifestations (hyperthermia, pallor of mucous membrane, circulatory failure, respiratory signs, neurological instability, and GI signs), blood picture (anemia, leukopenia, thrombocytopenia), demographics (sex and age), and treatment history (hemoparasitic therapy, ectoparasiticidal application, and prophylactic deworming) were conducted. The co-infection status of these parasites was also evaluated. A substantial portion of the goats in the study group was found to be affected by vector-borne hemoparasitic diseases and their arthropod vectors or GI parasites or both. This can be attributed to the constantly warm and humid climate of the region, which is favorable for the survival and growth of different life cycle stages of these parasites and vectors. A strategic parasitic disease surveillance-cum-control program is the need of the hour for ensuring climate resilience and profitable goat farming in the region.

Diagnosis of fasciolosis antibodies in Brazilian cattle through ELISA employing both native and recombinant antigens

Abstract:ABSTRACT Bovine fasciolosis is a parasitic disease with a global reach. Coprological based on egg detection in fecal samples and liver inspection to evaluate the presence of the parasite is currently the gold standard for diagnosing chronic fasciolosis in cattle. However, these techniques are labor-intensive and ineffective during the acute phase of the disease. Serodiagnosis using native and recombinant antigens has become an interesting alternative in efforts to identify cattle fasciolosis. We evaluated cattle from abattoir ( n = 139) and farms ( n = 500) through liver inspection and coprological examination, respectively. Our laboratory team optimized and validated enzyme-linked immunosorbent assay tests based on somatic antigen, excretory/secretory proteins, and the recombinant antigen cathepsin L-1 to detect serum antibodies against fasciolosis in cattle. For animals from abattoir, 10 were positive for fasciolosis according to liver inspection. Both Fh ES and Fh rCL-1 presented an area under the receiver operating characteristic (AUROC) curve of 0.80, with a sensitivity of 0.80 (95% CI: 0.46–0.95) and 0.70 (95% CI: 0.38–0.90) and specificity of 0.81 (95% CI: 0.73–0.87) and 0.87 (95% CI: 0.80–0.92), respectively. For those cattle from farms, 28 were positive only for fasciolosis according to coprological examination. In this scenario, Fh ES gave the best performance, with an AUROC of 0.84, sensitivity of 0.79 (95% CI: 0.60–0.90), and specificity of 0.86 (95% CI: 0.82–0.89). In conclusion, our study highlights the potential of serodiagnosis for accurately screening cattle fasciolosis. The promising sensitivity and specificity values of Fh ES when compared to liver inspection or coprological examination enhance its importance for cattle fasciolosis diagnosis. IMPORTANCE The aim of this article was to identify antibodies against fasciolosis in cattle in Brazil. The methodology was reproduced in our laboratory and applied for the first time to the Brazilian cattle herd. The antigens tested can be used as a screening test and thus speed up the diagnosis of bovine fascioliasis.

Abstract:BACKGROUND Dermabmsion of facial scars 4–8 weeks after injury frequently completely eliminates visible evidence of scar formation. However, efforts to define the cellular and structural mechanisms by which this phenomenon occurs have been limited in their success. OBJECTIVE We investigated wound healing after dermabrasive scar revision. METHODS The surgical scars of seven patients were abraded 6–8 weeks after injury. Comparative electron microscopic and immunohistochemical studies were performed on punch biopsy specimens taken before and after the dermabrasion. Ultrastructural changes in the basement membrane components and dermal structures were evaluated. Monoclonal antibody staining techniques were used to observe the presence, location, and temporal expression of tenascin, epiligrin, cadherins, and integrin subunits. RESULTS We observed: 1) an increase in collagen bundle density and size with a tendency toward unidirectional orientation of fibers parallel to the epidermal surface, 2) an upregulation of tenascin expression throughout the papillary dermis, and 3) expression of &agr;‐6/&bgr;‐4 integrin subunit on the keratinocytes throughout the stratum spinosum. CONCLUSIONS The mechanisms by which dermabrasive scar revision alters the events of primary cicatrix formation include modification of extracellular ligand expression, thereby influencing epithelial cell‐cell interaction, and reorganization of connective tissue.

Abstract:Tick-borne Apicomplexan parasites pose a significant threat to both public health and animal husbandry. Identifying potential pathogenic parasites and gathering their epidemiological data are essential for prospectively preventing and controlling infections. In the present study, genomic DNA of ticks collected from two goat flocks (Goatflock1 and Goatflock2) and one dog group (Doggroup) were extracted and the 18S rRNA gene of Babesia/Theileria/Colpodella spp. was amplified by PCR and sequenced. Phylogenetic analysis was conducted based on the obtained sequences. The differences in pathogen positive rates between ticks of different groups were statistically analyzed using the Chi-square or continuity-adjusted Chi-square test. As a result, two pathogenic Theileria (T.) luwenshuni genotypes, one novel pathogenic Colpodella sp. HLJ genotype, and two potential novel Colpodella spp. (referred to as Colpodella sp. struthionis and Colpodella sp. yiyuansis in this study) were identified in the Haemaphysalis (H.) longicornis ticks. Ticks of Goatflock2 had a significantly higher positive rate of Colpodella spp. than those from Goatflock1 (χ2=92.10; P = 8.2 × 10<sup>-22</sup>) and Doggroup (χ2=42.34; P = 7.7 × 10<sup>-11</sup>), and a significantly higher positive rate of T. luwenshuni than Doggroup (χ2=5.38; P = 0.02). However, the positive rates of T. luwenshuni between Goatflock1 and Goatflock2 were not significantly different (χ2=2.02; P = 0.16), and so as the positive rates of both pathogens between Goatflock1 and Doggroup groups (P > 0.05). For either Colpodella spp. or T. luwenshuni, no significant difference was found in prevalence between male and female ticks. These findings underscore the potential importance of Colpodella spp. in domestic animal-attached ticks, as our study revealed two novel Colpodella spp. and identified Colpodella spp. in H. longicornis for the first time. The study also sheds light on goats' potential roles in the transmission of Colpodella spp. to ticks and provides crucial epidemiological data of pathogenic Theileria and Colpodella. These data may help physicians, veterinarians, and public health officers prepare suitable detection and treatment methods and develop prevention and control strategies.

Abstract:Toxoplasmosis, caused by the protozoan parasite Toxoplasma gondii , is a globally distributed zoonotic infection with significant implications for human and animal health. This study investigated the prevalence of T. gondii infection in a population of beef cattle at three different stages of their productive lifespan and examined the impact of T. gondii serological status on blood parameters. A commercial beef fattening unit in Italy was the setting for this research, which involved a biosecurity assessment upon cattle arrival, blood sampling at three time points and Toxoplasma -specific serological testing using indirect fluorescent antibody tests (IFAT). Results revealed a dynamic pattern of T. gondii seropositivity in cattle, with an initial prevalence of 30.6% at arrival (T0) that increased to 44.6% at 14 days (T1) and then decreased slightly to 39.3% at slaughter after 5 months (T2). Interestingly, seroconversion was observed during the study, indicating ongoing infections, and antibody waning occurred in some animals. In terms of blood parameters, seropositive cattle exhibited significantly lower mean corpuscular volume (MCV) and a higher neutrophil–lymphocyte (N/L) ratio, suggesting an activation of the innate immune response. Furthermore, cattle with higher antibody titres displayed higher neutrophil counts. However, all blood parameters with a statistical significance were within the reference range. This study provides for the first time a longitudinal investigation on the serological status for T. gondii in naturally exposed beef cattle. These findings provide valuable insights into the clinico-pathological aspects of natural T. gondii exposure in cattle and underscore the importance of monitoring and managing T. gondii infection in livestock production systems.

Abstract:Background: The consumption of raw or undercooked meat, especially pork, and offal containing infective tissue cysts is suspected to be a significant route of infection with Toxoplasma gondii. Although the use of "animal-friendly pig production systems" ensuring direct contact with the natural environment offers ethical benefits, it limits the ability to ensure animal health; it may also increase the probability of infections by pathogens such as T. gondii, and thus their entry into the food chain. This study determines the seroprevalence of T. gondii in pigs from different housing systems and farms with different hygiene standards in Poland, as well as among pigs of different age groups from farms with high hygiene standards. In total 760 pig serum samples were examined for the presence of specific antibodies using the PrioCHECK® Toxoplasma Ab porcine commercial ELISA test (Prionics, Switzerland). Results: Test results with PP ≥ 20% were regarded as positive, as indicated by the manufacturer. Antibodies to T. gondii were found in 193 of 760 (25.4%) tested sera. Regarding different housing systems, antibodies were found in 117 pigs: of these, 52.6% (61/116) were from organic farms, 40.9% (47/115) from farms with low hygiene standards, 5.4% (9/167) from farms with high hygiene standards and 0% (0/40) from a farm with a high level of biosecurity. Regarding age groups, antibodies were found in 76 animals on farms with high hygiene standards: 11.1% (7/63) were pigs younger than 3 months, 0% (0/60) aged 3-4 months, 12.3% (7/57) aged 5-6 months (final fattening stage) and 43.7% (62/142) were sows aged 9 months and older. Conclusions: Antibodies to T. gondii were most often found in pigs from organic and low-hygiene farms, as well as in pigs aged 9 months and older. Meat derived from seropositive animals can pose a potential source of infection for humans. As maternal antibodies to T. gondii can be present in the blood of piglets aged up to 3-4 months, serological examination is unjustified in piglets up to this age.

Abstract:Blastocystis spp., Enterocytozoon bieneusi, and Giardia duodenalis are three common zoonotic intestinal parasites that cause severe diarrhea and enteric diseases. Leizhou black goats are characterized by a high reproductive rate, fast growth, and good meat quality, making them one of the pre-eminent goat breeds in China. Goats are reportedly common reservoirs of these three intestinal pathogens, but no information on their prevalence or genotypic distributions in black goats in Guangdong Province, China, is available. A total of 226 fecal samples were collected from goats in Zhanjiang city and genomic DNA was extracted from them. The presence of the three pathogens was detected using nested PCR targeting the sequences encoding SSU rRNA (Blastocystis spp.), the internal transcribed spacer of rRNA (ITS; E. bieneusi), as well as beta-giardin, glutamate dehydrogenase, and triosephosphate isomerase (G. duodenalis). All PCR products were sequenced to determine the species and genotypes of the organisms. The total prevalence rates of Blastocystis spp., E. bieneusi, and G. duodenalis were 33.63% (76/226), 17.70% (40/226), and 24.78% (56/226), respectively. Four subtypes of Blastocystis spp. were detected: ST5 (n = 6), ST10 (n = 50), ST14 (n = 14), and ST21 (n = 6). Among them, ST10 was the dominant genotype, accounting for 65.79% of strains, followed by the genotypes ST14 (18.42%), zoonotic ST5 (7.89%), and ST21 (7.89%). Four genotypes of E. bieneusi were detected: CHG3 (n = 32), CM21 (n = 4), CHG1 (n = 2), and ET-L2 (n = 2). Among these, CHG3 was the dominant genotype. Assemblage E (n = 54) and concurrent assemblages A and E (n = 2) were identified in the G. duodenalis-positive goats using multilocus genotyping. Blastocystis spp., E. bieneusi, and G. duodenalis infections were common in Leizhou black goats, all of which have zoonotic genotypes, indicating the potential risk of zoonotic transmission. Our results provide basic data for the prevention and control of these three intestinal pathogens. Further studies are required to better understand their genetic characteristics and zoonotic potential in Guangdong Province.

Abstract:This monograph presents complex data on Fascioloides magna from all aspects of its research (general information, distribution, spectrum of hosts) and summarizes the latest information on molecular structure of informative genes which were recently applied in resolving taxonomy and biogeography of this veterinary important parasite. The giant liver fluke, Fascioloides magna, is important liver parasite of free-living and domestic ruminants. Due to its biology, distribution, medical impact, and invasive character, this liver fluke attracts attention of wide spectrum of specialists – veterinary doctors, hunters and farmers, as well as scientists. The parasite utilizes wide range of free living and domestic ruminants as definitive hosts, with various pathological impacts ranging from moderate infections towards lethal effects. Fascioloides magna is of North American origin where it occurs in five enzootic regions. It was introduced to Europe along with its deer hosts in 19th century and it has established three permanent natural foci. The giant liver fluke represents an outstanding model for studying the origin, spatial distribution, migratory routs, and invasion processes of introduced species.

Abstract:Small ruminant lentivirus (SRLV) infections are spread in the flocks of sheep and goats all over the world, causing economic loss. Although only a fraction of infected animals develop disease symptoms, all of them may shed the virus, causing uncontrolled spread of the infection. Antibodies against the virus can be detected in the blood of infected animals and are the main marker of infection. Additionally, in most infected animals, proviral DNA can also be detected, but at different levels. Due to the lack of treatment or vaccines, the most effective strategy to prevent SRLV infections are control programmes introduced by several countries based on the elimination of seropositive individuals from the flock. An alternative approach, which has currently become the rationale, is an identification of host factors which may predispose certain individuals or breeds to resistance or susceptibility to small ruminant lentivirus infection. In our work, attention was paid to goats of the Carpathian breed infected with SRLV. Available RNA-seq results from the blood of 12 goats with a determined level of SRLV proviral load were used to analyse single nucleotide polymorphisms (SNPs) by the variant calling method. Six SNPs within five genes (POU2AF1, BCAT2, TMEM154, PARP14, UBASH3A) were selected for genotyping to determine their association with the level of small ruminant lentivirus proviral DNA in a group of 60 goats. Interestingly, in seronegative individuals, only the TT genotype of the PARP14 gene was observed, while the TMEM154 CC genotype was found only in seropositive goats. Both genes may be considered potential markers for resistance/susceptibility to SRLV infection. In contrast, polymorphisms identified in POU2AF1 and UBASH3A genes seemed to be deleterious for respective protein functions; therefore, these genes are less likely to be recognised as resistance/susceptibility markers of SRLV infection.
