Abstract:A better understanding of how microbial communities interact with their surroundings in physically and chemically heterogeneous subsurface environments will lead to improved quantification of biogeochemical reactions and associated nutrient cycling. This study develops a methodology to predict potential elevated rates of biogeochemical activity (microbial "hotspots") in subsurface environments by correlating microbial DNA and aspects of the community structure with the spatial distribution of geochemical indicators in subsurface sediments. Multiple linear regression models of simulated precipitation leachate, HCl and hydroxylamine extractable iron and manganese, total organic carbon (TOC), and microbial community structure were used to identify sample characteristics indicative of biogeochemical hotspots within fluvially-derived aquifer sediments and overlying soils. The method has been applied to (a) alluvial materials collected at a former uranium mill site near Rifle, Colorado and (b) relatively undisturbed floodplain deposits (soils and sediments) collected along the East River near Crested Butte, Colorado. At Rifle, 16 alluvial samples were taken from 8 sediment cores, and at the East River, 46 soil/sediment samples were collected across and perpendicular to 3 active meanders and an oxbow meander. Regression models using TOC and TOC combined with extractable iron and manganese results were determined to be the best fitting statistical models of microbial DNA (via 16S rRNA gene analysis). Fitting these models to observations in both contaminated and natural floodplain deposits, and their associated alluvial aquifers, demonstrates the broad applicability of the geochemical indicator based approach.

Abstract:Introduction: Dissolved organic matter (DOM) as primary and secondary energy sources can be transported via rivers to estuaries and impact coastal biogeochemical cycles. Storm-induced high discharge events can alter the dominant river flow paths and enhance leaching of shallow organic-rich soil layers, leading to elevated terrestrial DOM export. Land use/land cover (LULC) and associated anthropogenic interventions (including artificial reservoirs and agricultural irrigation) can control sources and transformation processes of exported DOM along with hydrologic factors. The relative significance of LULC, hydrological factors, and temperature variations with seasons will differ depending on geographical locations and complicate their incorporation in biogeochemical models of DOM dynamics. This study investigates the role of LULC, seasonality, and storm events on DOM concentrations and molecular composition in the Murderkill River system. Method: Surface water samples were collected seasonally and before/after storm events from 6 sites representing forested, agricultural, and developed LULC units. The DOM was characterized via parallel factor analysis of excitation-emission matrix data and electrospray ionization 21 T Fourier-transform ion cyclotron resonance mass spectrometry to determine potential DOM sources and enable the development of a conceptual model for DOM dynamics in rivers impacted by anthropogenic reservoirs. Result and Discussion: Our results suggest that storm-induced shallow and overland flow paths can increase surface-vegetation/plant-litter derived DOM based on atomic ratios associated with specific biogenic precursors (i.e., lignin, tannins, and/or oxygenated aromatic DOM), particularly in winter when autochthonous production was suppressed due to reduced temperatures. We further demonstrate that the damming effects of artificial reservoirs enhance the role of seasonal patterns of autochthonous production, disrupting storm-shunt process and stimulating significantly more bio-produced DOM export during spring and summer (i.e., tryptophan/tyrosine-like. N- and S- containing, phytoplankton-derived compounds). Collectively, these results demonstrate how artificial reservoirs alter the characteristics of DOM exported from rivers with implications for understanding carbon export and fate at river-estuary interfaces.

Abstract:Interstitial water or pore water occupies the space between soil particles and provides "hotspots" and "fluvial networks" for microbial activities in floodplain soil. However, to date, we know very little about the microorganisms living in pore water and how they respond to environmental changes. This study aimed to understand microbial distribution and assemblage in riparian pore waters, and how they respond to water chemistry and redox gradients associated with hydrological processes. We analysed the annual changes of porewater microbial communities from the east and west banks of the White Clay Creek, a site at the Christina River Basin – Critical Zone Observatory, Pennsylvania, USA. Microbial abundances were quantified by epifluorescence microscopy and detailed community structures were characterised by high‐throughput sequencing. Water chemistry and redox gradients were also monitored and recorded, and their interactions with porewater microbiomes were analysed using correlations and multivariate analyses. Abundance of microbial cells increased during summer and late autumn. Wetland porewater microbiomes mainly contained Acidobacteria, Bacteroidetes, Nitrospirae and Proteobacteria, and microbiome structures were easily distinguishable from those in the underlying hyporheic gravel layer. Seasonal dynamics of bacterial community structure in the east and west wetlands were distinct, responding to floodplain topography and associated hydrological/geochemical processes. Iron (Fe)‐cycling bacteria (mainly Gallionellaceae and Rhodoferax spp.) dominated the porewater microbiome, and their relative abundance was significantly higher in the east than the west wetland. Furthermore, Fe‐oxidising bacteria (Gallionellaceae) were negatively correlated with Fe‐reducing bacteria (Rhodoferax spp.) at the east wetland. Microbial abundances (cell density) in pore waters showed similar seasonal patterns across stream banks, but microbial community structure did not. Microbiome assembly in pore water is correlated with water chemistry and redox gradients primarily associated with local hydrological processes. As a consequence of their significance for carbon (C) mineralisation and Fe reduction at terrestrial–aquatic interfaces, microbiomes in riparian pore waters and associated microbial activity play an essential role in C and mineral dynamics. These findings will inform future studies of the response of freshwater ecosystems to hydrological dynamics influenced by global climate change.

Isotope-based source assessment of water flowing from storm sewer systems to a receiving river during dry weather periods

Abstract:Urban stormwater management systems, particularly storm sewers, are critical for managing runoff in urban areas. These systems are designed to function during wet weather events; however, field-based observations of these systems suggest that they may also be active flow pathways in dry weather conditions, ultimately contributing to streamflow. Unlike dry weather flow in wastewater systems, storm sewer dry weather flow has not been thoroughly explored. This research used stable isotopes of oxygen and hydrogen in water to examine the sources of dry weather flow from storm sewers in a highly urban catchment. A stable isotope mixing model was applied at the outfalls of two stormwater catchments and the receiving Black Creek, located in Toronto, Canada. Findings suggest that during dry periods, storm sewers receive non-stormwater inputs from tap water, wastewater, and groundwater, along with some precipitation, and that these sources may constitute up to 19 % of Black Creek's flow at the watershed scale. Seasonal patterns in flow and water sources were observed for the Black Creek and outfalls. At one outfall, dry weather flow was predominantly from the water distribution system (i.e., tap water and/or wastewater) throughout spring, summer, and fall. In contrast, at the second outfall, groundwater dominated in spring and summer, and groundwater and water distribution were equally proportioned in fall. Black Creek baseflow comprises a dynamic mix of water sources that at times are similar to the sources observed at the stormwater outfalls. Considering these findings, future work should incorporate strategic sampling of additional outfalls, and multiple years of data collection to explore inter-annual variability in these processes and focus on replicating a similar study in other urban watersheds with different climates and/or water infrastructure design. The study findings highlight that our understanding of dry weather flow from storm sewers is relatively limited, emphasizing the need for further exploration of this phenomenon to inform urban hydrological modelling, water quality studies, and urban water management.
A Tale of Two Storms: Inter‐Storm Variability of Stable Water Isotopes in a Solute Transport Model

Abstract:Many semi‐arid regions lack seasonality in precipitation, which is exploited in many stable water isotope applications. We overcome those limitations by studying strongly contrasting precipitation events over the same area that offer an opportunity to track water as it moves through watersheds or engineered landscapes. Key information for flood management, such as the pre‐event water contribution to the flood hydrograph becomes ascertainable using this method. The case presented here constitutes an example of advances toward developing science‐based solutions to inform water resources management that can be explored in other semi‐arid regions. Stable isotopic methods in hydroclimate monitoring are powerful for improving water resources management, but applications are limited, especially in semi‐arid regions where such management is needed most. Here, we show that we can address shortcomings related to the lack of a seasonal signal using stable water isotopic signatures measured in precipitation over the East San Francisco Bay area, California, during two contrasting events sampled at more than 20 locations in the winter of 2023. The observed range in δ18O in the rain samples is similar for both storms. However, the distributions do not overlap—the mean air temperature and δ18O during Winter Storm Olive (February 2023) were 2°C and − 12‰, respectively, while a warm atmospheric river event (March 2023) had a mean temperature of 9°C and δ18O of −6‰, close to the long‐term average δ18O measured in local precipitation. The Winter Storm showed expected trends in δ18O related to geography (i.e., lower with greater distance inland and elevation), while the atmospheric river δ18O pattern was more spatially uniform. We use hydrometric data from a gaged watershed in the study area and isotopic signatures of rain sampled during the two storm events and apply a solute transport model (StorAge selection) with a travel‐time approach to examine predicted watershed responses and potential water tracing applications. In this virtual experiment, we find that event size exerts a strong control on the relative amounts of runoff versus pre‐event water in the stream, while uncertainty in stream hydrograph separation is related to the degree of contrast between precipitation/runoff and pre‐event water. Key to flood prediction, adaptation, and mitigation, especially in coastal urban areas, is knowledge of the contributing water sources and timing of stream flow. The strong contrast in stable isotopes between these two events, close in time and over the same area, illustrates the potential to use stable isotope signatures to track the transport and mixing of events through natural and engineered watersheds that are threatened by climate whiplash events.

Abstract:Abstract Climate change is rapidly altering hydrological processes and consequently the structure and functioning of Arctic ecosystems. Predicting how these alterations will shape biogeochemical responses in rivers remains a major challenge. We measured [C]arbon and [N]itrogen concentrations continuously from two Arctic watersheds capturing a wide range of flow conditions to assess understudied event‐scale C and N concentration‐discharge ( C‐Q ) behavior and post‐event recovery of stoichiometric conditions. The watersheds represent low‐gradient, tundra landscapes typical of the eastern Brooks Range on the North Slope of Alaska and are part of the Arctic Long‐Term Ecological Research sites: the Kuparuk River and Oksrukuyik Creek. In both watersheds, we deployed high‐frequency optical sensors to measure dissolved organic carbon (DOC), nitrate (), and total dissolved nitrogen (TDN) for five consecutive thaw seasons (2017–2021). Our analyses revealed a lag in DOC: stoichiometric recovery after a hydrologic perturbation: while DOC was consistently elevated after high flows, diluted during rainfall events and consequently, recovery in post‐event concentration was delayed. Conversely, the co‐enrichment of TDN at high flows, even in watersheds with relatively high N‐demand, represents a potential “leak” of hydrologically available organic N to downstream ecosystems. Our use of high‐frequency, long‐term optical sensors provides an improved method to estimate carbon and nutrient budgets and stoichiometric recovery behavior across event and seasonal timescales, enabling new insights and conceptualizations of a changing Arctic, such as assessing ecosystem disturbance and recovery across multiple timescales.

Abstract:Canadian Journal of Microbiology, Ahead of Print. The Grand River watershed is the largest catchment in southern Ontario. The river's northern and southern sections are influenced by agriculture, whereas central regions receive wastewater effluent and urban runoff. To characterize in-river microbial communities, as they relate to spatial and environmental factors, we conducted two same-day sampling events along the entire 300 km length of the river, representing contrasting flow seasons (high flow spring melt and low flow end of summer). Through high-throughput sequencing of 16S rRNA genes, we assessed the relationship between river microbiota and spatial and physicochemical variables. Flow season had a greater impact on communities than spatial or diel effects and profiles diverged with distance between sites under both flow conditions, but low-flow profiles exhibited higher beta diversity. High-flow profiles showed greater species richness and increased presence of soil and sediment taxa, which may relate to increased input from terrestrial sources. Total suspended solids, dissolved inorganic carbon, and distance from headwaters significantly explained microbial community variation during the low-flow event, whereas conductivity, sulfate, and nitrite were significant explanatory factors for spring melt. This study establishes a baseline for the Grand River's microbial community, serving as a foundation for modeling the microbiology of anthropogenically impacted freshwater systems affected by lotic processes.

Assessment of Microbial Community Dynamics in River Bank Filtrate Using High-Throughput Sequencing and Flow Cytometry

Abstract:Surface-groundwater interactions play an important role in microbial community compositions of river bank filtrates. Surface water contaminations deriving from environmental influences are attenuated by biogeochemical processes in the hyporheic zone, which are essential for providing clean and high-quality drinking water in abstraction wells. Characterizing the flow regime of surface water into the groundwater body can provide substantial information on water quality, but complex hydraulic dynamics make predictions difficult. Thus, a bottom up approach using microbial community shifting patterns as an overall outcome of dynamic water characteristics could provide more detailed information on the influences that affect groundwater quality. The combination of high-throughput sequencing data together with flow cytometric measurements of total cell counts reveals absolute abundances among taxa, thus enhancing interpretation of bacterial dynamics. 16S rRNA high-throughput sequencing of 55 samples among six wells in a well field in Austria that is influenced by river bank filtrate within a time period of 3 months has revealed both, clear differences as well as strong similarity in microbiome compositions between wells and dates. A significant community shift from April to May occurred in four of six wells, suggesting that surface water flow regimes do affect these wells stronger than others. Triplicate sampling and subsequent sequencing of wells at different dates proved the method to be reproducible. Flow cytometric measurements of total cells indicate microbial shifts due to increased cell counts and emphasize the rise of allochthonous microorganisms. Typical freshwater bacterial lineages (Verrucomicrobia, Bacteroidetes, Actinobacteria, Cyanobacteria, Armatimonadetes) were identified as most increasing phyla during community shifts. The changes are most likely a result of increased water abstraction in the wells together with constant river water levels rather than rain events. The results provide important knowledge for future implementations of well utilization in dependency of the nearby Danube River water levels and can help drawing conclusions about the influence of surface water in the groundwater such that hygienically save and clean drinking water with a stable microbial community can be provided.

Abstract:The availability of long-term data series of natural stable isotopes of water ( 18 O and 2 H) observed in a small mountainous headwater catchment provides an opportunity to study soil water partitioning and runoff generation processes. In this study, the analysis of the isotopic composition of rainwater, soil water, subsurface stormflow, groundwater, and streamflow was carried out over a 10-year period. A one-dimensional vertical dual-continuum model was used to analyze the preferential flow of water and the transport of 18 O in a hillslope. The model, previously validated against observed hillslope discharge and isotope transport data dynamics, provided a framework for evaluating seasonal and episodal runoff regimes. For episodal simulations, the partitioning of soil water into pre-event and event water was done using virtual tracer concentrations instead of the observed 18 O contents – to maximize the contrast between pre-event and event water signatures. Simulated seasonal isotopic variations of 18 O in stormflow and soil water compared well with the observed 18 O contents. When evaluated as long-term median values, the observed isotopic composition did not suggest significantly different water in the different discharge components. However, when the flux-type weighting of isotopic signatures in hillslope stormflow, deep percolation, and root water uptake was applied, isotopically distinct waters became evident. For the selected episodes, approximately 62 % of stormflow and water transpired by plants was associated with pre-event water. The results suggested that isotopic differences between water associated with different discharge mechanisms prevail despite intense pre-event and event water mixing above the soil–bedrock interface. The modeling approach helped to explore the potential mechanisms behind the existence of isotopically distinct subsurface waters.

Water Level Fluctuations Modulate the Microbiomes Involved in Biogeochemical Cycling in Floodplains

Abstract:Drastic changes in hydrological conditions within floodplain ecosystems create distinct microbial habitats. However, there remains a lack of exploration regarding the variations in microbial function potentials across the flooding and drought seasons. In this study, metagenomics and environmental analyses were employed in floodplains that experience hydrological variations across four seasons. Analysis of functional gene composition, encompassing nitrogen, carbon, and sulfur metabolisms, revealed apparent differences between the flooding and drought seasons. The primary environmental drivers identified were water level, overlying water depth, submergence time, and temperature. Specific modules, e.g., the hydrolysis of β-1,4-glucosidic bond, denitrification, and dissimilatory/assimilatory nitrate reduction to ammonium, exhibited higher relative abundance in summer compared to winter. It is suggested that cellulose degradation was potentially coupled with nitrate reduction during the flooding season. Phylogenomic analysis of metagenome-assembled genomes (MAGs) unveiled that the Desulfobacterota lineage possessed abundant nitrogen metabolism genes supported by pathway reconstruction. Variation of relative abundance implied its environmental adaptability to both the wet and dry seasons. Furthermore, a novel order was found within Methylomirabilota, containing nitrogen reduction genes in the MAG. Overall, this study highlights the crucial role of hydrological factors in modulating microbial functional diversity and generating genomes with abundant nitrogen metabolism potentials.
Longitudinal Analysis of Urban Stormwater Microbiome and Resistome from Watersheds with and without Green Infrastructure using Long-Read Sequencing

Abstract:Since stormwater conveys a variety of contaminants into water bodies, green infrastructure (GI) is increasingly being adopted as an on-site treatment solution in addition to controlling peak flows. The purpose of this study was to identify differences in microbial water quality of stormwater in watersheds retrofitted with GI vs. those without GI. Considering stormwater is recently recognized as a contributor to the antibiotic resistance (AR) threat, another goal of this study was to characterize changes in the microbiome and collection of AR genes (resistome) of urban stormwater with season, rainfall characteristics, and fecal contamination. MinION long-read sequencing was used to analyze stormwater microbiome and resistome from watersheds with and without GI in Columbus, Ohio, United States, over 18 months. We characterized fecal contamination in stormwater via culturing Escherichia coli and with molecular microbial source tracking (MST) to identify sources of fecal contamination. Overall, season and storm event (rainfall) characteristics had the strongest relationships with changes in the stormwater microbiome and resistome. We found no significant differences in microbial water quality or the microbiome of stormwater in watersheds with and without GI implemented. However, there were differences between the communities of microorganisms hosting antibiotic resistance genes (ARGs) in stormwater from watersheds with and without GI, indicating the potential sensitivity of AR bacteria to treatment. Stormwater was contaminated with high concentrations of human-associated fecal bacterial genes, and the ARG host bacterial community had considerable similarities to human feces/wastewater. We also identified 15 potential pathogens hosting ARGs in these stormwater resistome, including vancomycin-resistant Enterococcus faecium (VRE) and multidrug-resistant Pseudomonas aeruginosa . In summary, urban stormwater is highly contaminated and has a great potential to spread AR and microbial hazards to nearby environments. This study presents the most comprehensive analysis of stormwater microbiome and resistome to date, which is crucial to understanding the potential microbial risk from this matrix. This information can be used to guide future public health policy, stormwater reuse programs, and urban runoff treatment initiatives.

Abstract:Urbanization and the persistent environmental changes present a major challenge for urban freshwaters and availability of water for humans and wildlife. In order to increase understanding of urban ecohydrology, we investigated the variability of planktonic bacteria and benthic diatoms - as two key biological indicators - coupled with insights from hydrochemistry and stable water isotopes across four urban streams characterized by different dominant water sources in Berlin, the German capital, over a period of one year (2021-2022). DNA metabarcoding results show that substantial spatio-temporal variability exists across urban streams in terms of microbial diversity and richness, with clear links to abiotic factors and nutrient concentrations. Bacterial communities showed clear distinction between effluent-impacted and non-effluent impacted streams as well as clear seasonal turnover. In-stream benthic diatom assemblages also showed robust seasonal variation as well as high species diversity. Our multiple-tracer approach is relevant for emerging questions regarding the increased use of treated effluent to supplement declining baseflows, the assessment of stream restoration projects and the impact of storm drainage and surface pollution on aquatic ecosystem health. eDNA analysis allows analysis of spatial and temporal patterns not feasibly studied with traditional analyses of macroinvertebrates. This can ultimately be leveraged for future water resource management and restoration planning and monitoring of urban freshwater systems across metropolitan areas.

Abstract:Storm events can mobilize nitrogen species from landscapes into streams, exacerbating eutrophication and threatening aquatic ecosystems as well as human health. However, the transport pathways and storm responses of different nitrogen forms remain elusive. We used high-frequency chemical and isotopic sampling to partition sources of stormwater runoff and determine transport pathways of multiple nitrogen forms in an agricultural catchment. Macroinvertebrate Community Responses to Disturbance in a Fragmented River With Contrasting Legacies of Alteration

Abstract:Flow is a critical factor determining the riverine ecosystem structure and function. Widespread hydrologic alteration, however, has impacted the ecological integrity of rivers in ways that are not well understood, including responses of biological communities to increasingly frequent and severe climatic disturbances. Our study compared the responses of invertebrate communities on woody debris to large flooding and extreme drought in two highly contrasting segments of an impaired low‐gradient river. The upstream segment, which according to previous research has higher α‐diversity and production of large‐bodied and sensitive invertebrates, maintained higher flows and longitudinal connectivity throughout the 4‐year study. Communities in this upper segment resembled one another among sites (lower spatial turnover) but experienced greater temporal shifts in composition associated with hydrological disturbances. Conversely, invertebrate communities in the highly altered downstream segment, which is impaired by reduced flow, sedimentation, and hypoxia, were composed of smaller‐bodied and pollution‐tolerant taxa with lower α‐diversity. Unlike the upper segment, communities were patchily distributed among sites (higher spatial turnover), which made it more difficult to detect system‐wide temporal variation in composition throughout the study. Our study underscores the benefit of including measures of connectivity and spatial heterogeneity when assessing the ecological integrity of lotic systems. Understanding the system‐wide response to disturbances across longer time frames can help better predict and mitigate the impacts of climate change on ecosystem integrity in degraded rivers.

More Than a First Flush: Urban Creek Storm Hydrographs Demonstrate Broad Contaminant Pollutographs

Abstract:Stormwater runoff clearly impacts water quality and ecological health of urban receiving waters. Subsequent management efforts are often guided by conceptual models of contaminant "first flushes", defined by disproportionate concentrations or mass loads early in the storm hydrograph. However, studies examining the dynamics of contaminant transport and receiving water hydrology have primarily focused on "traditional" stormwater contaminants and point sources, with less evaluation of chemically complex nonpoint pollution sources. Accordingly, we conducted baseflow and storm sampling in Miller Creek, a representative small, urban watershed in the Puget Sound region (WA, USA). We comprehensively characterized organic contaminant profiles and dynamics via targeted quantification of 35 stormwater-derived chemicals, complementary nontarget HRMS analyses, and surrogate chemical metrics of ecological health. For quantified analytes, total daily baseflow loads were 0.8-3.4 g/day and storm event loads were ∼80-320 g/storm (∼48 h interval), with nine contaminants detected during storms at >500 ng/L. Notably, urban creek "pollutographs" were much broader than relatively sharp storm hydrographs and exhibited transport-limited (rather than mass-limited) source dynamics, with immediate water quality degradation during low-intensity precipitation and continued mobilization of contaminant mass across the entire hydrograph. Study outcomes support prioritization of source identification and focused stormwater management efforts to improve water quality and promote ecosystem function in small urban receiving waters.
Quantitative fecal pollution assessment with bacterial, viral, and molecular methods in small stream tributaries

Abstract:Stream water quality can be impacted by a myriad of fecal pollution sources and waste management practices. Identifying origins of fecal contamination can be challenging, especially in high order streams where water samples are influenced by pollution from large drainage areas. Strategic monitoring of tributaries can be an effective strategy to identify conditions that influence local water quality. Water quality is assessed using fecal indicator bacteria (FIB); however, FIB cannot differentiate sources of fecal contamination nor indicate the presence of disease-causing viruses. Under different land use scenarios, three small stream catchments were investigated under 'wet' and 'dry' conditions (Scenario 1: heavy residential; Scenario 2: rural residential; and Scenario 3: undeveloped/agricultural). To identify fecal pollution trends, host-associated genetic targets HF183/BacR287 (human), Rum2Bac (ruminant), GFD (avian), and DG3 (canine) were analyzed along with FIB (Escherichia coli and enterococci), viral indicators (somatic and F+ coliphage), six general water quality parameters, and local rainfall. Levels of E. coli exceeded single sample maximum limits (235 CFU/100 mL) in 70.7 % of samples, enterococci (70 CFU/100 mL) in 100 % of samples, and somatic coliphage exceeded advisory thresholds (600 PFU/L) in 34.1 % of samples. The detection frequency for the human-associated genetic marker was highest in Scenario 3 (50 % of samples) followed by Scenario 2 (46 %), while the ruminant-associated marker was most prevalent in Scenario 1 (64 %). Due to the high proportion of qPCR-based measurements below the limit of quantification, a Bayesian data analysis approach was applied to investigate links between host-associated genetic marker occurrence with that of rainfall and fecal indicator levels. Multiple trends associated with small stream monitoring were revealed, emphasizing the role of rainfall, the utility of fecal source information to improve water quality management. And furthermore, water quality monitoring with bacterial or viral methodologies can alter the interpretation of fecal pollution sources in impaired waters.

Abstract:Biological assemblages in streams are influenced by hydrological dynamics, particularly in non‐perennial systems. Although there has been increasing attention on how drying impacts stream organisms, few studies have investigated how specific characteristics of drying and subsequent wetting transitions influence biotic responses via resistance and resilience traits. Here, we characterized how hydrologic metrics, including those quantifying drying and wetting transitions as well as dry and wet phases, alter diversity and composition of three aquatic assemblages in non‐perennial streams in southern California: benthic macroinvertebrates, soft‐bodied algae and diatoms. We found that flow duration prior to sampling was correlated with variation in macroinvertebrate and soft‐bodied algal assemblage composition. The composition and richness of diatom assemblages, however, were predominantly influenced by the drying start date prior to sampling. Contrary to other studies, the duration of the dry phase prior to sampling did not influence the composition or richness of any assemblage. Although our study was conducted within a region in which each assemblage experienced comparable environmental conditions, we found no single hydrologic metric that influenced all assemblages in the same way. The hot‐summer Mediterranean climate of southern California likely acts as a strong environmental filter, with taxa in this region relying on resistance and resilience adaptations to survive and recolonize non‐perennial streams following wetting. The different responses of algal and diatom assemblages to hydrologic metrics suggest greater resilience to drying and wetting events, particularly for primary producers. As drying and wetting patterns continue to change, understanding biodiversity responses to hydrologic metrics could inform management actions that enhance the ecological resilience of communities in non‐perennial streams. In particular, the creation and enhancement of flow regimes in which natural timing and duration of dry and wet phases sustain refuges that support community persistence in a changing environment.

Abstract:Antecedent hydrological conditions are recorded through the evolution of dissolved lithium isotope signatures (δ7 Li) by juxtaposing two storm events in an upland watershed subject to a Mediterranean climate. Discharge and δ7 Li are negatively correlated in both events, but mean δ7 Li ratios and associated ranges of variation are distinct between them. We apply a previously developed reactive transport model (RTM) for the site to these event‐scale flow perturbations, but observed shifts in stream δ7 Li are not reproduced. To reconcile the stability of the subsurface solute weathering profile with our observations of dynamic stream δ7 Li signatures, we couple the RTM to a distribution of fluid transit times that evolve based on storm hydrographs. The approach guides appropriate flux‐weighting of fluid from the RTM over a range of flow path lengths, or equivalently fluid residence times. This flux‐weighted RTM approach accurately reproduces dynamic storm δ7 Li‐discharge patterns distinguished by the antecedent conditions of the watershed.

Rainfall-induced changes in aquatic microbial communities and stability of dissolved organic matter: Insight from a Fen river analysis

Abstract:Microbial communities are pivotal in aquatic ecosystems, as they affect water quality, energy dynamics, nutrient cycling, and hydrological stability. This study explored the effects of rainfall on hydrological and photosynthetic parameters, microbial composition, and functional gene profiles in the Fen River. Our results demonstrated that rainfall-induced decreases in stream temperature, dissolved oxygen, pH, total phosphorus, chemical oxygen demand, and dissolved organic carbon concentrations. In contrast, rainfall increased total dissolved solids, salinity, and ammonia-nitrogen concentrations. A detailed microbial community structure analysis revealed that Cyanobacteria was the dominant microbial taxon in the Fen River, accounting for approximately 75% and 25% of the microalgal and bacterial communities, respectively. The abundance of Chlorophyta and Bacillariophyta increased by 47.66% and 29.92%, respectively, whereas the relative abundance of Bacteroidetes decreased by 37.55% under rainfall conditions. Stochastic processes predominantly affected the assembly of the bacterial community on rainy days. Functional gene analysis revealed variations in bacterial functions between sunny (Sun) and rainy (Rain) conditions, particularly in genes associated with the carbon cycle. The 3-oxoacyl-[acyl-carrier-protein] reductase gene was more abundant in the Fen River bacterial community. Particular genes involved in metabolism and environmental information processing, including the acetyl-CoA C-acetyltransferase (atoB), enoyl-CoA hydratase (paaF), and branched-chain amino acid transport system gene (livK), which are integral to environmental information processing, were more abundant in Sun than the Rain conditions. In contrast, the phosphate transport system gene, the galactose metabolic gene, and the pyruvate metabolic gene were more abundant in Rain. The excitation-emission matrix analysis with parallel factor analysis identified four fluorescence components (C1-C4) in the river, which were predominantly protein- (C1) and humic-like (C2-C4) substances. Rainfall affected organic matter production and transport, leading to changes in the degradation and stability of dissolved organic matter. Overall, this study offers insight into how rainfall affects aquatic ecosystems.

Abstract:Lotic surface water sites (e.g. creeks) are important resources for localized agricultural irrigation. However, there is concern that microbial contaminants within untreated surface water may be transferred onto irrigated soil and crops. To evaluate this issue, water samples were collected between January 2017 and August 2018 from a freshwater creek used to irrigate kale and radish plants on a small farm in the Mid-Atlantic, United States. In addition, on one sampling date, a field survey was conducted in which additional water (creek source and point-of-use) and soil samples were collected to assess the viral and bacterial communities pre- and post- irrigation. All samples were processed for DNA extracts and shotgun sequenced on the Illumina HiSeq platform. The resulting metagenomic libraries were assembled de novo and taxonomic and functional features were assigned at the contig and peptide level. From these data, we observed that Betaproteobacteria (e.g. Variovorax) dominated the water, both at the source and point-of-use, and Alphaproteobacteria (e.g. Streptomyces) dominated both pre- and post-irrigated soil. Additionally, in the creek source water there were variations in the abundance of the dominant bacterial genera and functional annotations associated with seasonal characteristics (e.g. water temperature). Antibiotic resistance genes and virulence factors were also identified in the creek water and soil, with the majority specific to their respective habitat. Moreover, an analysis of clustered regularly interspaced short palindromic repeat (CRISPR) arrays showed the persistence of certain spacers through time in the creek water, as well as specific interactions between creek bacteriophages and their hosts. Overall, these findings provide a more holistic picture of bacterial and viral composition, dynamics, and interactions within a freshwater creek that can be utilized to further our knowledge on its suitability and safety for irrigation.
