Abstract:A high degree of connectivity and the coordinated electrical activity of neural cells or networks are believed to be the reason that the brain is capable of highly sophisticated information processing. Likewise, the effectiveness of an animal heart largely depends on such coordinated cell activity. To advance our understanding of these complex biological systems, high spatiotemporal-resolution techniques to monitor the cell electrical activity and an ideally seamless interaction between cells and recording devices are desired. Here we present a monolithic microsystem in complementary metal oxide semiconductor (CMOS) technology that provides bidirectional communication (stimulation and recording) between standard electronics technology and cultured electrogenic cells. The microchip can be directly used as a substrate for cell culturing, it features circuitry units per electrode for stimulation and immediate cell signal treatment, and it provides on-chip signal transformation as well as a digital interface so that a very fast, almost real-time interaction (2 ms loop time from event recognition to, e.g., a defined stimulation) is possible at remarkable signal quality. The corresponding spontaneous and stimulated electrical activity recordings with neuronal and cardiac cell cultures will be presented. The system can be used to, e.g., study the development of neural networks, reveal the effects of neuronal plasticity and study cellular or network activity in response to pharmacological treatments.

Abstract:A new type of microfluidic system for biological cell manipulation, a CMOS/microfluidic hybrid, is demonstrated. The hybrid system starts with a custom-designed CMOS (complementary metal-oxide semiconductor) chip fabricated in a semiconductor foundry using standard integration circuit technology. A microfluidic channel is post-fabricated on top of the CMOS chip to provide biocompatible environment. The motion of individual biological cells that are tagged with magnetic beads is directly controlled by the CMOS chip that generates localized magnetic filed patterns using an on-chip array of micro-electromagnets. The speed and the programmability of the CMOS chip further allow for the dynamic reconfiguration of the magnetic fields, substantially increasing the manipulation capability of the hybrid system. The concept of a hybrid system is verified by simultaneously manipulating individual biological cells with microscopic resolution. A new operation protocol that exploits the fast speed of electronics to trap and move a large number of cells with less power consumption is also demonstrated. Combining the advantages of microelectronics, the CMOS/microfluidic hybrid approach presents a new model for a multifunctional lab-on-a chip for biological and medical applications.

Abstract:The closed-loop electrophysiology is gradually applied to targeted stimulation of nervous system. According to desired neural biomarkers acquired by biological signal measurement, the stimulus pattern can be automatically adjusted with the suitable closed-loop control algorithms. Moreover, the neural computing uses the numerical computing models to simulate the structures, functions and relations of the neurons and the neural networks, as the substitute of biological brain. As the prevalence of the hardware-in-loop (HIL), the HIL closed-loop electrophysiology not only bypasses the problem of biological experiments but also makes up for the insufficient efficiency and real-time performance of software digital simulation. This paper proposes a hardware experiment platform mainly made up of TMS320F28377D digital signal processor (DSP) and AMD Ryzen 5 5600X 6-Core Processor (CPU) for closed-loop electrophysiological research. The hardware platform is programable, portable, real-time and supports the repeated experiments. Closed-loop electrophysiological experiments with different kinds of control algorithms, stimulation waveforms, feedback variables or neuron models can be implemented on the platform, it may promote the development of researches in the field of closed-loop neuroscience and biomedical engineering.

Abstract:Significance A critical challenge for flexible biomedical implants is in the development of materials and structures that enable intimate coupling to biotissues with long-term stability. The results presented here address this problem through a materials and integration strategy that combines highly doped silicon nanomembranes chemically bonded to thin films of thermal silicon dioxide in a construct that simultaneously serves as a biofluid barrier and a conductively coupled biointerface. Use of this approach with various flexible electronic systems, including passive and active electrodes for electrophysiological sensing and electrical stimulation, illustrate capabilities in high-fidelity operation. Systematic accelerated lifetime studies in artificial biofluids highlight the stability of these systems for chronic operation, without electrical leakage or other forms of degradation.
A Brain-Inspired In-Memory Computing System for Neuronal Communication Via Memristive Circuits.

Abstract:Brain-inspired approaches can efficiently analyze the activities of biological neural networks and solve computationally hard problems with energy efficiencies unattainable with von Neumann architectures, indicating a significant improvement in the understanding of neuronal communications and functionalities. Here, we present a brain-inspired multimodal signal processing system with organic memristor arrays that can potentially integrate the signal sensory, storage, and computation. To facilitate the multimodal signal processing system design, we used four components. First, we present a multimodal signal sensory module mainly responsible for multimodal (iconic, echoic, olfactory, muscular, and gustatory) signal collection, fusion, and storage. Second, a high-density cross-point memristive synapse array is constructed after fabrication of the albumin protein memristor to realize the dense connectivity between layers of computing, data storage, and communication. Third, considering the structure and function of the brain region, we demonstrate a general learning module for hierarchy learning, which can recognize and imagine multimodal information. Finally, the necessary peripheral circuit module (consisting of winnerless competition function circuit, analog-to-digital converter, digital-to-analog converter, pulse modulator, etc.) is designed. Notably, our system can capture massive amounts of data every second and perform in situ processing of multimodal signals. This study is expected to help achieve the deep integration of nano materials into neuromorphic computing systems and energy-efficient integrated circuits.
Nanoelectronics-biology frontier: From nanoscopic probes for action potential recording in live cells to three-dimensional cyborg tissues.

Abstract:Semiconductor nanowires configured as the active channels of field-effect transistors (FETs) have been used as detectors for high-resolution electrical recording from single live cells, cell networks, tissues and organs. Extracellular measurements with substrate supported silicon nanowire (SiNW) FETs, which have projected active areas orders of magnitude smaller than conventional microfabricated multielectrode arrays (MEAs) and planar FETs, recorded action potential and field potential signals with high signal-to-noise ratio and temporal resolution from cultured neurons, cultured cardiomyocytes, acute brain slices and whole animal hearts. Measurements made with modulation-doped nanoscale active channel SiNW FETs demonstrate that signals recorded from cardiomyocytes are highly localized and have improved time resolution compared to larger planar detectors. In addition, several novel three-dimensional (3D) transistor probes, which were realized using advanced nanowire synthesis methods, have been implemented for intracellular recording. These novel probes include (i) flexible 3D kinked nanowire FETs, (ii) branched intracellular nanotube SiNW FETs, and (iii) active silicon nanotube FETs. Following phospholipid modification of the probes to mimic the cell membrane, the kinked nanowire, branched intracellular nanotube and active silicon nanotube FET probes recorded full-amplitude intracellular action potentials from spontaneously firing cardiomyocytes. Moreover, these probes demonstrated the capability of reversible, stable, and long-term intracellular recording, thus indicating the minimal invasiveness of the new nanoscale structures and suggesting biomimetic internalization via the phospholipid modification. Simultaneous, multi-site intracellular recording from both single cells and cell networks were also readily achieved by interfacing independently addressable nanoprobe devices with cells. Finally, electronic and biological systems have been seamlessly merged in 3D for the first time using macroporous nanoelectronic scaffolds that are analogous to synthetic tissue scaffold and the extracellular matrix in tissue. Free-standing 3D nanoelectronic scaffolds were cultured with neurons, cardiomyocytes and smooth muscle cells to yield electronically-innervated synthetic or 'cyborg' tissues. Measurements demonstrate that innervated tissues exhibit similar cell viability as with conventional tissue scaffolds, and importantly, demonstrate that the real-time response to drugs and pH changes can be mapped in 3D through the tissues. These results open up a new field of research, wherein nanoelectronics are merged with biological systems in 3D thereby providing broad opportunities, ranging from a nanoelectronic/tissue platform for real-time pharmacological screening in 3D to implantable 'cyborg' tissues enabling closed-loop monitoring and treatment of diseases. Furthermore, the capability of high density scale-up of the above extra- and intracellular nanoscopic probes for action potential recording provide important tools for large-scale high spatio-temporal resolution electrical neural activity mapping in both 2D and 3D, which promises to have a profound impact on many research areas, including the mapping of activity within the brain.

Abstract:Here we provide a perspective concept of neurohybrid memristive chip based on the combination of living neural networks cultivated in microfluidic/microelectrode system, metal-oxide memristive devices or arrays integrated with mixed-signal CMOS layer to control the analog memristive circuits, process the decoded information, and arrange a feedback stimulation of biological culture as parts of a bidirectional neurointerface. Our main focus is on the state-of-the-art approaches for cultivation and spatial ordering of the network of dissociated hippocampal neuron cells, fabrication of a large-scale cross-bar array of memristive devices tailored using device engineering, resistive state programming, or non-linear dynamics, as well as hardware implementation of spiking neural networks (SNNs) based on the arrays of memristive devices and integrated CMOS electronics. The concept represents an example of a brain-on-chip system belonging to a more general class of memristive neurohybrid systems for a new-generation robotics, artificial intelligence, and personalized medicine, discussed in the framework of the proposed roadmap for the next decade period.

Abstract:The increased need for wearable and implantable medical devices has driven the demand for electronics that interface with living systems. Current bioelectronic systems have not fully resolved mismatches between engineered circuits and biological systems, including the resulting pain and damage to biological tissues. Here, salt/poly(ethylene glycol) (PEG) aqueous two-phase systems are utilized to generate programmable hydrogel ionic circuits. High-conductivity salt-solution patterns are stably encapsulated within PEG hydrogel matrices using salt/PEG phase separation, which route ionic current with high resolution and enable localized delivery of electrical stimulation. This strategy allows designer electronics that match biological systems, including transparency, stretchability, complete aqueous-based connective interface, distribution of ionic electrical signals between engineered and biological systems, and avoidance of tissue damage from electrical stimulation. The potential of such systems is demonstrated by generating light-emitting diode (LED)-based displays, skin-mounted electronics, and stimulators that deliver localized current to in vitro neuron cultures and muscles in vivo with reduced adverse effects. Such electronic platforms may form the basis of future biointegrated electronic systems.

Abstract:There are existing studies that successfully show the impact of corporate social responsibility (CSR) on firms’ financial performance. However, limited attention is paid to its impact on the business ecosystem. CSR could be seen as an investment for building a sustainable business ecosystem, which enhances the competitiveness of this system’s members. In that context, this study apprehends and captures the virtuous cycle of firm competitiveness. On analyzing data from interviews with seven firms, the study offers four propositions identifying the structure of the virtuous cycle linking CSR activities to firm competitiveness through the accumulation of social capital within business ecosystems. Based on those propositions, the study offers new insights into CSR research for academics and strategic planning guidelines for managers that integrate social and economic values for a sustainable business ecosystem and firm competitiveness.

Abstract:This study reports an integrated platform for simultaneous modulation and monitoring of cardiomyocyte function in vitro. Optical inputs are achieved through expression of a photoactivatable adenylyl cyclase, that when irradiated causes a reversible increase in beating rate. Bioelectronic outputs are achieved with a multi‐electrode array, which can record the beating rate and wavefront propagation characteristics with high spatiotemporal resolution. Bioelectronic medicine is emerging as a powerful approach for restoring lost endogenous functions and addressing life‐altering maladies such as cardiac disorders. Systems that incorporate both modulation of cellular function and recording capabilities can enhance the utility of these approaches and their customization to the needs of each patient. Here is report an integrated optogenetic and bioelectronic platform for stable and long‐term stimulation and monitoring of cardiomyocyte function in vitro. Optical inputs are achieved through the expression of a photoactivatable adenylyl cyclase, that when irradiated with blue light causes a dose‐dependent and time‐limited increase in the secondary messenger cyclic adenosine monophosphate with subsequent rise in autonomous cardiomyocyte beating rate. Bioelectronic readouts are obtained through a multi‐electrode array that measures real‐time electrophysiological responses at 32 spatially‐distinct locations. Irradiation at 27 μW mm−2 results in a 14% elevation of the beating rate within 20–25 min, which remains stable for at least 2 h. The beating rate can be cycled through "on" and "off" light states, and its magnitude is a monotonic function of irradiation intensity. The integrated platform can be extended to stretchable and flexible substrates, and can open new avenues in bioelectronic medicine, including closed‐loop systems for cardiac regulation and intervention, for example, in the context of arrythmias.

A Bidirectional Neural Interface IC With Chopper Stabilized BioADC Array and Charge Balanced Stimulator

Abstract:We present a bidirectional neural interface with a 4-channel biopotential analog-to-digital converter (bioADC) and a 4-channel current-mode stimulator in 180 nm CMOS. The bioADC directly transduces microvolt biopotentials into a digital representation without a voltage-amplification stage. Each bioADC channel comprises a continuous-time first-order ∆Σ modulator with a chopper-stabilized OTA input and current feedback, followed by a second-order comb-filter decimator with programmable oversampling ratio. Each stimulator channel contains two independent digital-to-analog converters for anodic and cathodic current generation. A shared calibration circuit matches the amplitude of the anodic and cathodic currents for charge balancing. Powered from a 1.5 V supply, the analog and digital circuits in each recording channel draw on average [Formula: see text] and [Formula: see text] of supply current, respectively. The bioADCs achieve an SNR of [Formula: see text] and a SFDR of [Formula: see text] , for better than 9-b ENOB. Intracranial EEG recordings from an anesthetized rat are shown and compared to simultaneous recordings from a commercial reference system to validate performance in-vivo . Additionally, we demonstrate bidirectional operation by recording cardiac modulation induced through vagus nerve stimulation, and closed-loop control of cardiac rhythm. The micropower operation, direct digital readout, and integration of electrical stimulation circuits make this interface ideally suited for closed-loop neuromodulation applications.
Closed-loop control of a modular neuromorphic biohybrid

Abstract:Neural networks modularity is a major challenge for the development of control circuits of neural activity. Under physiological limitations, the accessible regions for external stimulation are possibly different from the functionally relevant ones, requiring complex indirect control designs. Moreover, control over one region might affect activity of other downstream networks, once sparse connections exist. We address these questions by developing a hybrid device of a cortical culture functionally integrated with a biomimetic hardware neural network. This design enables the study of modular networks controllability, while connectivity is well-defined and key features of cortical networks are accessible. Using a closed-loop control to monitor the activity of the coupled hybrid, we show that both modules are congruently modified, in the macroscopic as well as the microscopic activity levels. Control impacts efficiently the activity on both sides whether the control circuit is an indirect series one, or implemented independently only on one of the modules. Hence, these results present global functional impacts of a local control intervention. Overall, this strategy provides an experimental access to the controllability of neural activity irregularities, when embedded in a modular organization.
Multimodal Wireless-Powered Flexible System for Closed-Loop Neuromodulation

Abstract:This article presents a wirelessly powered closed-loop neuromodulation system, containing both implantable and wearable parts. The implantable part is an implantable neuromodulator tag that contains a customized application specific integrated circuit (ASIC) for neurostimulation and recording, an off-the-shelf near-field communication (NFC) proximity integrated circuit card (PICC) chip, and one coupling coil for communication and power transferring. There is a wireless monitoring device, an NFC reader, electroencephalogram (EEG) electrodes, and near-infrared spectrum (NIRS) modules in wearable part. Intracranial neural recorder and wearable EEG/NIRS modules are able to provide multimodal biological information, such as local field potentials (LFPs), EEG, and cerebral tissue oxygen saturation (StO(2)) for algorithms to propose a more precise closed-loop stimulation strategy for better treatment effect. The ASIC chip is fabricated in a 0.18-mu m CMOS technology. The stimulator has a limited size of 0.027 mm(2). Its stimulation current ranges from 0 to 0.6 mA with a bandwidth of 10 Hz-8 kHz, which is able to support precise neuromodulation. The recording channel features a bandwidth of 0.5-100 Hz with a noise floor of 0.93 mu V-rms. The entire neuromodulator tag has a small footprint of 20 x 24 x 1.4 mm, and its weight is only 0.25 g.
Response of C fibre nociceptors in the anaesthetized monkey to heat stimuli: estimates of receptor depth and threshold.

Abstract:1. Responses to ramped or stepped temperature stimuli were obtained from fifty‐three cutaneous C fibre mechano‐heat nociceptors (CMHs) in the hairy skin of the pentobarbitone‐morphine anaesthetized monkey. A three‐layer heat transfer model was developed to describe the temperature distribution within the skin and to estimate receptor depth and heat threshold. 2. Surface heat threshold, defined as the surface temperature when the first action potential occurs, increased as: (a) the rate of temperature rise for the ramped stimuli increased from 0.095 to 5.8 degrees C s‐1; (b) the duration of stepped heat stimuli decreased from 30 to 1 s; and (c) the base temperature of stepped heat stimuli decreased from 38 to 35 degrees C. These results suggest that the heat threshold for CMHs is determined by the temperature at the depth of the receptor. 3. Receptor depth estimates from responses to ramped stimuli ranged from 20 to 570 microns with a mean of 201 microns. The estimated mean receptor heat threshold was 40.4 +/‐ 2.2 degrees C (+/‐ S.D.). No correlation was observed between depth and thermal or mechanical threshold. The average receptor depth and threshold, estimated from the responses to stepped heat stimuli, were 150 microns and 40.2 degrees C, respectively. 4. We conclude that: (a) the receptor endings of CMHs occur in the epidermis and dermis; (b) temperature at the level of the receptor determines threshold; (c) temperature at the receptor ending is much lower than skin surface temperature at threshold; and (d) the tight distribution of receptor heat thresholds suggests a uniform transducer mechanism for heat in CMHs.
A quantitative study of the attitudes of members of a black developing community towards their dogs and veterinary services.

Abstract:To further the study of human-companion animal interactions in black developing communities, the clients (n = 94) of a university-based animal hospital, serving primarily a developing community, were interviewed. The interviews covered demographics, socio-economic aspects, perceptions, values and attitudes of clients towards their dogs. Regular and new clients were compared. A profile of the hospital's black clientele was also constructed. The study showed that the majority of black clients value their dogs for the sake of security and protection. Attitudes towards pets parallelled improvements in socio-economic status. Most clients felt positive towards existing veterinary services. The results of this study support and highlight the value of a previous observational study and could prove to be invaluable in designing, implementing and planning socially relevant animal health services.

Abstract:Continuous monitoring of tissue microphysiology is a key enabling feature of the organ-on-chip (OoC) approach for in vitro drug screening and disease modeling. Integrated sensing units are particularly convenient for microenvironmental monitoring. However, sensitive in vitro and real-time measurements are challenging due to the inherently small size of OoC devices, the characteristics of commonly used materials, and external hardware setups required to support the sensing units. Here we propose a silicon-polymer hybrid OoC device that encompasses transparency and biocompatibility of polymers at the sensing area, and has the inherently superior electrical characteristics and ability to house active electronics of silicon. This multi-modal device includes two sensing units. The first unit consists of a floating-gate field-effect transistor (FG-FET), which is used to monitor changes in pH in the sensing area. The threshold voltage of the FG-FET is regulated by a capacitively-coupled gate and by the changes in charge concentration in close proximity to the extension of the floating gate, which functions as the sensing electrode. The second unit uses the extension of the FG as microelectrode, in order to monitor the action potential of electrically active cells. The layout of the chip and its packaging are compatible with multi-electrode array measurement setups, which are commonly used in electrophysiology labs. The multi-functional sensing is demonstrated by monitoring the growth of induced pluripotent stem cell-derived cortical neurons. Our multi-modal sensor is a milestone in combined monitoring of different, physiologically-relevant parameters on the same device for future OoC platforms.

Abstract:The nervous system poses a grand challenge for integration with modern electronics and the subsequent advances in neurobiology, neuroprosthetics, and therapy which would become possible upon such integration. Due to its extreme complexity, multifaceted signaling pathways, and ∼1 kHz operating frequency, modern complementary metal oxide semiconductor (CMOS) based electronics appear to be the only technology platform at hand for such integration. However, conventional CMOS-based electronics rely exclusively on electronic signaling and therefore require an additional technology platform to translate electronic signals into the language of neurobiology. Organic electronics are just such a technology platform, capable of converting electronic addressing into a variety of signals matching the endogenous signaling of the nervous system while simultaneously possessing favorable material similarities with nervous tissue. In this review, we introduce a variety of organic material platforms and signaling modalities specifically designed for this role as "translator", focusing especially on recent implementation in in vivo neuromodulation. We hope that this review serves both as an informational resource and as an encouragement and challenge to the field.

A Biomimetic, SoC-Based Neural Stimulator for Novel Arbitrary-Waveform Stimulation Protocols

Abstract:Novel neural stimulation protocols mimicking biological signals and patterns have demonstrated significant advantages as compared to traditional protocols based on uniform periodic square pulses. At the same time, the treatments for neural disorders which employ such protocols require the stimulator to be integrated into miniaturized wearable devices or implantable neural prostheses. Unfortunately, most miniaturized stimulator designs show none or very limited ability to deliver biomimetic protocols due to the architecture of their control logic, which generates the waveform. Most such designs are integrated into a single System-on-Chip (SoC) for the size reduction and the option to implement them as neural implants. But their on-chip stimulation controllers are fixed and limited in memory and computing power, preventing them from accommodating the amplitude and timing variances, and the waveform data parameters necessary to output biomimetic stimulation. To that end, a new stimulator architecture is proposed, which distributes the control logic over three component tiers - software, microcontroller firmware and digital circuits of the SoC, which is compatible with existing and future biomimetic protocols and with integration into implantable neural prosthetics. A portable prototype with the proposed architecture is designed and demonstrated in a bench-top test with various known biomimetic output waveforms. The prototype is also tested in vivo to deliver a complex, continuous biomimetic stimulation to a rat model of a spinal-cord injury. By delivering this unique biomimetic stimulation, the device is shown to successfully reestablish the connectivity of the spinal cord post-injury and thus restore motor outputs in the rat model.

Abstract:Presented is the design, fabrication, and evaluation of 72-channel microcircuit electrode arrays (biochips) built to interface neurons and computers. Contributions include a detailed mathematical model of the electrode/electrolyte interface and subsequent electrode design optimization (in terms of maximum signal-to-noise ratio). Electrode impedance measurements were obtained with the electrodes submerged in electrolytic neural solution. Large sample results indicate a 7% discrepancy between theoretical and measured impedances. In addition to the engineering efforts, biological cell culturing techniques were developed to enable the growth of synaptically interconnected, electrically excitable cells on the biochip. Both dissociated invertebrate neurons and clonal mammalian cell systems were employed. Together these contributions enabled microvolt signals to be recorded from individual neurons grown and synaptically interconnected on the biochip surface. Also recorded were neuronal responses to on-chip stimulation. Consequently, the biochip technology presented represents a step towards a long-term, noninvasive, multisite electrical stimulation and recording capability necessary for extracting the mechanisms governing the way living neural networks connect, integrate, and self-organize.
