Abstract:Histone deacetylases (HDACs) have been identified as therapeutic targets due to their regulatory function in chromatin structure and organization. Here we analyzed the therapeutic effect of LBH589, a class I-II HDAC inhibitor, in acute lymphoblastic leukemia (ALL). In vitro, LBH589 induced dose-dependent antiproliferative and apoptotic effects, which were associated with increased H3 and H4 histone acetylation. Intravenous (i.v.) administration of LBH589 in immunodeficient BALB/c-RAG2γc mice in which human-derived T and B-ALL cell lines were injected induced a significant reduction in tumor growth. Using primary ALL cells, a xenograft model of human leukemia in BALB/c-RAG2γc mice was established, allowing continuous passages of transplanted cells to several mouse generations. Treatment of mice engrafted with T or B-ALL cells with LBH589 induced an in vivo increase in the acetylation of H3 and H4, which was accompanied with prolonged survival of LBH589-treated mice in comparison with those receiving Vincristine and Dexametasone. Notably, the therapeutic efficacy of LBH589 was significantly enhanced in combination with Vincristine and Dexametasone. Our results demonstrate the therapeutic activity of LBH589 in combination with standard chemotherapy in pre-clinical models of ALL and suggest that this combination may be of clinical value in the treatment of patients with ALL.

Abstract:Emerging contaminants (ECs), which are present in water bodies, could cause global environmental and human health problems. These contaminants originate from various sources such as hospitals, clinics, households, and industries. Additionally, they can also indirectly enter the water supply through runoff from agriculture and leachate from landfills. ECs, specifically Pharmaceutical and personal care products (PPCPs), are causing widespread concern due to their contribution to persistent water pollution. Traditional approaches often involve expensive chemicals and energy or result in the creation of by-products. This study developed a practical and environmentally-friendly method for removing PPCPs, which involved combining and integrating various techniques. To implement this method, it was necessary to establish and used a field simulator based on the real-life scenario. Based on the data analysis, the average removal rates of COD, TP, TN, and NH 4 + -N were 57%, 59%, 63%, and 73%, respectively. the removal rate of PPCPs by CCWs was found to be 82.7% after comparing samples that were not treated by constructed wetlands and those that were treated. Combined constructed wetlands (CCWs) were found to effectively remove PPCPs from water. This is due to the combined action of plant absorption, absorption, and biodegradation by microorganisms living in the wetlands. Interestingly, the wetland plant reed had been shown to play an important role in removing these pollutants. Microbial degradation was the most important pathway for PPCPs removal in CCWs. Carbamazepine was selected as a typical PPCP for analysis. In addition, the microbial community structure of the composite filler was also investigated. High-throughput sequencing confirmed that the dominant bacteria had good adaptability to PPCPs. This technique not only reduced the potential environmental impact but also served as a foundation for further research on the use of constructed wetlands for the treatment of PPCPs contaminated water bodies and its large-scale implementation.

Abstract:The removal of 12 pharmaceuticals and personal care products (PPCPs) in two full-scale wastewater treatment plants (WWTPs) and a tertiary treatment system was studied. The ecological risks of effluents from both secondary and tertiary treatment systems as well as excess sludge were evaluated. Primary treatment and ultraviolet light disinfection showed limited ability to remove most selected PPCPs. The combination of an anaerobic process and triple-oxidation ditches can eliminate DEET better than the anaerobic/anoxic/oxic process. Adsorption to sludge played a key role in the removal of triclocarban. Multistage constructed wetlands as a tertiary treatment efficiently removed caffeine and ibuprofen from wastewater and could decrease the risk of partial selected PPCPs. Selected PPCPs residues in excess sludge generally produced higher risks to the ecological environment than effluents from WWTPs.
Enhanced Degradation of Bisphenol A and Ibuprofen by an Up-Flow Microbial Fuel Cell-Coupled Constructed Wetland and Analysis of Bacterial Community Structure

Abstract:This study aims to demonstrate that an up-flow microbial fuel cell-coupled constructed wetland (UCW-MFC) can effectively treat synthetic wastewater that contains a high concentration of pharmaceutical and personal care products (PPCPs, 10 mg L−1 level), such as ibuprofen (IBP) and bisphenol A (BPA). A significant decline in chemical oxygen demand (COD) and ammonia nitrogen (NH4+-N) removal was observed when BPA was added, which indicated that BPA was more toxic to bacteria. The closed circuit operation of UCW-MFC performed better than the open circuit mode for COD and NH4+-N removal. Similarly, the removal rates of IBP and BPA were increased by 9.3% and 18%, respectively, compared with the open circuit mode. The majority of PPCPs were removed from the bottom and anode layer, which accounted for 63.2–78.7% of the total removal. The main degradation products were identified. The removal rates of IBP and BPA decreased by 14.6% and 23.7% due to a reduction in the hydraulic detention times (HRTs) from 16 h to 4 h, respectively. Electricity generation performance, including voltage and maximum power density, initially increased and then declined with a decrease in the HRT. Additionally, both the current circuit operation mode and the HRT have an impact on the bacterial community diversity of the anode according to the results of high-throughput sequencing. The possible bacterial groups involved in PPCP degradation were identified. In summary, UCW-MFC is suitable for enabling the simultaneous removal of IBP and BPA and successful electricity production.

Abstract:This study evaluated the reutilization of waste materials (scrap tires, sewage sludge, and wood chips) to remove volatile organic compounds (VOCs) benzene/toluene/ethylbenzene/xylenes/trichloroethylene/cis-1,2-dichloroethylene (BTEX/TCE/cis-DCE), plasticizer di(2-ethylhexyl) phthalate (DEHP), and pharmaceutically active compound carbamazepine from artificially contaminated water. Different hybrid removal processes were developed: (1) 300 mg/L BTEX + 20 mg/L TCE + 10 mg/L cis-DCE + tires + Pseudomonas sp.; (2) 250 mg/L toluene + sewage sludge biochar + Pseudomonas sp.; (3) 100 mg/L DEHP + tires + Acinetobacter sp.; and (4) 20 mg/L carbamazepine + wood chips + Phanerochaete chrysosporium. For the hybrid process (1), the removal of xylenes, TCE, and cis-DCE was enhanced, resulted from the contribution of both physical adsorption and biological immobilization removal. The hybrid process (2) was also superior for the removal of DEHP and required a shorter time (2 days) for the bioremoval. For the process (3), the biochar promoted the microbial immobilization on its surface and substantially enhanced/speed up the bioremoval of toluene. The fungal immobilization on wood chips in the hybrid process (4) also improved the carbamazepine removal considerably (removal efficiencies of 61.3 ± 0.6%) compared to the suspended system without wood chips (removal efficiencies of 34.4 ± 1.8%). These hybrid processes would not only be promising for the bioremediation of environmentally concerned contaminants but also reutilize waste materials as sorbents without any further treatment.

Abstract:The occurrence and elimination of 19 micro-organic pollutants including endocrine disrupting chemicals (EDCs) and pharmaceuticals and personal care products (PPCPs) in a full-scale anaerobic/anoxic/aerobic-membrane bioreactor process was investigated. The investigated process achieved over 70% removal of the target EDCs and 50%–100% removal of most of the PPCPs, with influent concentration ranging from ng/L to μg/L. Three PPCPs, carbamazepine, diclofenac and sulpiride were not well removed, with the removal efficiency below 20%. A rough mass balance suggests that the targets were eliminated through sludge-adsorption and/or biodegradation, the former of which was particularly significant for the removal of hydrophobic compounds. The two-phase fate model was employed to describe the kinetics of sludge-adsorption and biodegradation. It was found that the fast sludge adsorption (indicated by mass-transfer rates greater than 10 for most compounds) is responsible for the rapid decline of the aqueous concentration of the targets in the first compartment of the treatment process (i.e. in the anaerobic tank). In contrast, the slow biodegradation proved to be the rate-determining step for the entire degradation process, and the rates are generally positively related to the dissolved oxygen level. On the other hand, this study showed that the removal rates of most targets can reach a quasi-plateau in 5 h under aerobic conditions, indicating that hydraulic retention time of ca. 5 h in aerobic tanks should be sufficient for the elimination of most targets.
Removal of pharmaceuticals and personal care products by two-stage biofiltration for drinking water treatment

Abstract:Contamination of drinking water with pharmaceuticals and personal care products (PPCPs) is an issue of health concerns. To effectively control the level of PPCPs in drinking water, a pilot study employing two parallel trains of two-stage biofiltration, i.e., a sand/anthracite (SA) biofilter coupled with a biologically-active granular activated carbon (GAC) post-filter contactor, was conducted as a post-treatment after coagulation in a drinking water treatment plant. Results showed the biofiltration process could effectively remove PPCPs with an average removal of 53.4%, where the GAC contactor played the dominant role to remove 48.1% of the total PPCPs. The molecular properties determined the removability of individual PPCPs, i.e., smaller molecules with simpler structure connectivity were more likely to be removed. Based on the quantitative structure-property relationships (QSPRs) analysis, a simple regression model was proposed to predict the removability of each PPCP across the biofiltration process. The drinking water equivalent level (DWEL) quotient method was developed to assess the health risks of detected PPCPs in water samples. The biofiltration process showed efficient capacity to reduce the health risks of PPCPs with an average removal of 79%, and the PPCPs in the effluents generally would not pose adverse health effects. Pearson correlation analysis explored the possible role of nitrogenous PPCPs (N-PPCPs) as the precursors of nitrogenous disinfection byproducts (N-DBPs) in drinking waters. Aromatic nitrogen in PPCPs was found to be a significant descriptor for the formation potential of trichloroacetonitrile (TCAN). In addition, it was found that pre-filter chlorination could slightly improve the biofiltration of PPCPs.
Enhancing neonicotinoid removal in recirculating constructed wetlands: The impact of Fe/Mn biochar and microbial interactions

Abstract:Neonicotinoids pose significant environmental risks due to their widespread use, persistence, and challenges in elimination. This study explores the effectiveness of Fe/Mn biochar in enhancing the removal efficiency of neonicotinoids in recirculating constructed wetlands (RCWs). Results demonstrated that incorporating Fe/Mn biochar into RCWs significantly improved the removal of COD, NH4+-N, TN, TP, imidacloprid (IMI), and acetamiprid (ACE). However, the simultaneous presence of IMI and ACE in the RCWs hindered the elimination of NH4+-N, TN, and TP from wastewater. The enhanced removal of nutrients and pollutants by Fe/Mn biochar was attributed to its promotion of carbon, nitrogen, and phosphorus cycling in RCWs, along with its facilitation of the adsorption and biodegradation of IMI and ACE. Metagenomics analysis demonstrated that Fe/Mn biochar altered the structure and diversity of microbial communities in RCWs. A total of 17 biodegradation genes (BDGs) and two pesticide degradation genes (PDGs) were identified within RCWs, with Fe/Mn biochar significantly increasing the abundance of BDGs such as cytochrome P450. The potential host genera for these BDGs/PDGs were identified as Betaproteobacteria, Acidobacteria, Nitrospiraceae, Gemmatimonadetes, and Bacillus. This study offers valuable insights into how Fe/Mn biochar enhances pesticide removal and its potential application in constructed wetland systems for treating pesticide-contaminated wastewater.

Abstract:The use of nanomaterials to enhance bioprocessing for the removal of pharmaceuticals and personal care products (PPCPs) has received increasing attention. Facing the increasing ecotoxicity effects of pharmaceuticals, carbamazepine was classified as medium to high-risk pollutants in wastewater treatment plant effluents and surface waters. However, the interactions between nanomaterials and microbial communities are unclear. This study reveals the potential and mechanism of synthesized octahedral molecular sieve and activated carbon fiber (OMS-2@ACF) composites in enhancing the aerobic microbial capacity of sequencing batch reactor (SBR) for the treatment of carbamazepine. In an SBR, a series doses of OMS-2@ACF at 0, 0.05, 0.25, and 1 g/L were introduced to the reactor to investigate their impact on the treatment of carbamazepine by activated sludge and recorded as S1, S2, S3, and S4, respectively. Furthermore, OMS-2@ACF demonstrated an enhancing effect on the carbamazepine removal performance, as the S4 reactor exhibited the most effective removal efficiencies between 67.8 % and 76.4 % of carbamazepine. Illumina high-throughput sequencing revealed that OMS-2@ACF material altered the composition of the microbial community in the SBR, identifying the Comamonas as the key genus contributing most significantly to the community response. Analysis indicated that the presence of OMS-2@ACF material increased metabolic pathways associated with contaminant degradation, resulting in improved treatment performance of the SBR. This study provides a promising method for the combination of nano-molecular sieve and activated carbon fiber during the pharmaceutical wastewater biotreatment process.

Occurrence and fate of pharmaceuticals and personal care products in a wastewater treatment plant with Bacillus bio-reactor treatment

Abstract:Pharmaceuticals and personal care products (PPCPs) have attracted wide attention due to their environmental impacts and health risks. PPCPs released through wastewater treatment plants (WWTPs) are estimated to be 80 %. Nevertheless, the occurrence of PPCPs in the WWTPs equipped with Bacillus spec.-based bioreactors (BBR) treatment system remains unclear. In this study, sludge and waste water samples were collected during separate winter and summer sampling campaigns from a typical BBR treatment system. The results indicate that out of 58 target PPCPs, 27 compounds were detected in the waste water (0.06-1900 ng/L), and 23 were found in the sludge (0.6-7755 ng/g dw). Paraxanthine was the chemical of the highest abundance in the influent due to the high consumption of the parent compounds caffeine and theobromine. The profile for PPCPs in the wastewater and sludge exhibited no seasonal variation. Overall, the removal of target PPCPs in summer is more effective than the winter. In the BBR bio-reactor, it was found that selected PPCPs (at ng/L level) can be completely removed. The efficiency for individual PPCP removal was increased from 1.0 % to 50 % in this unit, after target specific adjustments of the process. The effective removal of selected PPCPs by the BBR treatment system is explained by combined sorption and biodegradation processing. The re-occurrence of PPCPs in the wastewater was monitored. Negative removal efficiency was explained by the cleavage of Phase II metabolites after the biotransformation process, and the lack of equilibrium for PPCPs in the sludge of the second clarifier. A compound specific risk quotient (RQ) was calculated and applied for studying the potential environmental risks. Diphenhydramine is found with the highest environmental risk in wastewater, and 15 other PPCPs show negligible risks in sewage sludge.
Highly enhanced biodegradation of pharmaceutical and personal care products in a novel tidal flow constructed wetland with baffle and plants

Abstract:Research on decentralized wastewaters deserves special focus due to the potential abundance of emerging organic pollutants including pharmaceutical and personal care products (PPCPs), which might pose serious threats to the local water bodies and even to human health. Constructed wetland (CW) is a common decentralized wastewater treatment technology, with a certain ability to eliminate PPCPs. Nonetheless, PPCPs removal in common CWs is frequently challenging, besides, the removal mechanism remains elusive. Based on our previous study, tidal flow constructed wetlands (TFCWs) is effective in nitrogen removal. Here, 3 TFCWs with different modifications (baffle, plants, both baffle and plants) were constructed to treat raw domestic sewage and specifically to evaluate the removal efficiencies and mechanism of PPCPs. 24 PPCPs including 7 antibiotics, 8 steroid hormones and 9 biocides were detected in the level of 1.10 ± 0.29 ng/L-799 ± 10.6 ng/L in the influents. Consequently, we found that modification with both baffle and plants significantly influenced the removal of PPCPs. Moreover, the highest removal rates of biocides (97.1 ± 0.29%), steroid hormones (99.8 ± 0.02%), and antibiotics (90.2 ± 1.60%) were achieved via both baffles and plants in TFCWs. Based on the mass balance analysis, microbial degradation dominated the removal of PPCPs with a percentage higher than 85.7%, followed by substrate adsorption (5.22 × 10−2-14.3%) and plant uptake (1.66 × 10−3-0.44%). Further, 16S rDNA sequencing analysis revealed that the presence of baffle and plants improve the removal efficiency of PPCPs by means of enhancing microbial diversity and changing dominant microorganisms. Moreover, Thaumarchaeota was potentially the key microorganism in the phylum level for PPCPs elimination by TFCWs through LEfSe (linear discriminant analysis (LDA) effect size) analysis. These findings provide new insights into the removal of PPCPs in CWs.

Bacteriol. 92:1099–1110. 1966.—The ribonucleic acid (RNA) contained in 70S ribosomes and in 50S and 30S subunits was hydrolyzed by pancreatic ribonuclease. A 7% amount of the RNA was removed from the 70S particle; at 10−4m magnesium concentration, a maximum of 24 and 30% of the RNA in the 50S and the 30S fractions, respectively, was removed by ribonuclease. At the two lower magnesium ion concentrations, 50S ribosomes did not lose any protein, whereas 30S ribosomes lost protein as a result of ribonuclease treatment. A number of proteins were removed from the 30S particles by ribonuclease, and these proteins were antigenically related to proteins present in 50S ribosomes. The differential effect of ribonuclease on 50S and 30S ribosomes suggested that they have structural dissimilarities.
Study on the Removal Characteristics of IBP and DCF in Wastewater by CW-MFC with Different Co-Substrates

Abstract:Pharmaceuticals and Personal Care Products (PPCPs) constitute a group of compounds that are challenging to break down and potentially pose risks to both ecosystems and human health when they accumulate in water bodies. This study established and operated small-scale constructed wetland–microbial fuel cells (CW-MFCs) continuously for 60 days, maintaining a hydraulic retention time (HRT) of 3 days. The research aimed to assess the treatment efficiency of wastewater containing Ibuprofen (IBP) and Diclofenac (DCF) using different co-substrates (glucose, sucrose, and sodium acetate) and to analyze the impact of these co-substrates on the composition of bacterial communities within the CW-MFC. After 60 days of operation, CW-MFC achieved removal rates of 89.29% for IBP and 84.10% for DCF. The elimination of IBP was primarily dependent on co-metabolic degradation processes occurring in both the anode and cathode, while DCF removal relied on anodic co-metabolism. Additionally, various co-substrates have an influence on the bacterial community diversity of the anode and cathodes. The possible bacterial groups involved in PPCP degradation were identified. In summary, Glu was identified as a more suitable co-substrate for CW-MFC in the removal of IBP and DCF, while SA as a co-substrate favored the induction and enrichment of EAB in the anodes. These findings offer valuable insights into the potential of CW-MFC for mitigating emerging contaminants.

This study assessed multi-module biochar filters (MmBFs) as onsite wastewater treatment systems (OWTSs), comprising movable modules filled with biochar to remove chemical oxygen demand (COD), nitrogen, phosphorus, and Escherichia coli ( E. coli ) in wastewater. The MmBF treats wastewater sequentially through six modules: three aerobic modules (M1-M3) for organic matter oxidation and nitrification, two anoxic modules (M4-M5) for denitrification, and an additional module (M6) for the removal of faecal bacteria using biochar and bark. The experiments ran for 381 days using three identical MmBF pilots with two distinct sampling periods, conducted under conditions relevant to OWTSs using municipal wastewater as influent. Water samples were taken from the influent, final effluent, and effluent of each module to evaluate the removal efficiency of organic matter, nitrogen, phosphorus, and E. coli . During the second sampling period, the results showed a 95 ± 2.1% removal of COD, along with a substantial removal of total inorganic nitrogen (71 ± 6.6%). However, phosphate removal was limited (3.4 ± 30.4%). E. coli removal decreased from 2.63 ± 0.93 log 10 removal in the first sampling period to 1.8 ± 0.73 log 10 removal in the second sampling period. In summary, the MmBFs showed promising potential in treating organic matter, nitrogen, and E. coli , making it an alternative option for OWTS. However, further exploration is needed to assess long-term performance, micropollutant removal, and biological activities. Design enhancements, especially for phosphorus removal are necessary.

Abstract:The study aims to understand the occurrence and removal of 20 emerging contaminants (ECs) in each unit process of a sequencing batch reactor-based wastewater treatment plant (WWTP) and explore the potential of biological activated carbon (BAC) for the treatment of residual ECs and organic matter in the secondary effluent. Analgesic-acetaminophen, anti-inflammatory drug-ibuprofen, and stimulant-caffeine were detected at high concentrations in the influent. Most of the removal was observed in the biological treatment stage in the SBR basins. The mass load of the ECs was 2.93 g/d in the secondary effluent and 0.4 g/d in the final sludge, while the total removal of the mass load of ECs till the secondary treatment stage was 93.22%. 12 of the 20 ECs were removed by more than 50%, while carbamazepine (negative removal), sulfamethoxazole, and trimethoprim were removed by less than 20%. As a polishing step and to remove residual ECs, two BAC units were studied for 11,000 bed volumes (324 days). Packed column studies on granular activated carbon were conducted, and GAC development to BAC was monitored. SEM and FTIR were used to confirm and characterize the BAC. The BAC appeared to be more hydrophobic than the GAC. The BAC removed 78.4% and 40% of the dissolved ECs and organic carbon at an optimum EBCT of 25 min. Carbamazepine, sulfamethoxazole, and trimethoprim were removed by 61.5, 84, and 52.2%, respectively. Parallel column tests revealed adsorption as an important mechanism for the removal of positively charged compounds. The results indicate that the BAC is an effective tertiary/polishing technique for removing organic and micropollutants in the secondary wastewater effluent.

Abstract:Recovery of phosphorus (P) from wastewater has led to growing public concern considering its scarcity and future availability as well as its detrimental environmental impacts. However, the recovered P is inevitably contaminated with co-existing antibiotics like tetracycline (TC) and sulfamethazine (SMT) which will pose serious risks to the health of human and animals after being spread to the environment. In this study, we propose a novel scheme that can recover P from synthetic wastewater and at the same time degrade the co-existing antibiotics. To achieve such a goal, a series of biochar (BC) were prepared from calcination of waste sludge and were used both as the adsorbent for P recovery and as the catalyst for peroxymonosulfate (PMS) activation and antibiotic degradation. Results showed that the sludge source (i.e. Sm: municipal sludge, Sp: paper mill sludge), calcination atmosphere (i.e. air-deficient, N2, vacuum) and temperature (i.e. 600 and 800 °C) exhibited significant influence on P adsorption capacity. Generally, the BC calcined in N2 showed better P uptake, and increase of calcination temperature from 600 °C to 800 °C could further improve P uptake. Though BCp-N-600 (prepared from Sp in N2 at 600 °C) showed faster and higher P uptake (56.3 mg/g) than its counterpart BCm-N-600 (33.2 mg/g), BCm-N-600 showed stronger catalytic activity and more stable performance in the complex pollutant system (P + SMT). It was proposed that P was recovered primarily through the chemisorption and precipitation mechanism, while SMT was nearly completely degraded primarily by the ROS generated from PMS activation.

Abstract:A novel composite biochar (NCB) was produced from the pyrolysis of co-fermentation products of sewage sludge, food wastes and rice straw, and exhibited higher nitrogen and phosphorus adsorption capacity due to the larger surface area (14.7 m2 g-1) and higher Ca content (51753.7 mg kg-1) than single rice straw biochar. The addition of NCB was then investigated in a non-aerated vertical baffled flow constructed wetland (VBFCW) for contaminated water treatment. The VBFCW with NCB addition significantly improved CODMn, NH4+-N, TN and TP removal efficiencies of 83.3 ± 5.3%, 95.9 ± 3.4%, 28.0 ± 4.0% and 59.5 ± 11.8%, respectively, at a hydraulic retention time (HRT) of 3 d. In addition, the TN and TP removal rates at a decreased HRT of 2 d were much higher than those at an HRT of 3 d without NCB addition. The presence of NCB in the VBFCW system enhanced nutrient adsorption and improved the enrichment of bacteria for organic and nitrogen removal mainly including the genera Bacillus and Lactococcus.
