Abstract:A viable platform for food packaging applications has evolved in the form of protein nanofibers with material properties. Through this work, a simple, extensible process of protein for amyloid fibrils formation was proposed. The addition of polyphenol was expected to improve its synergistic antibacterial properties. The isolate protein of soy and whey (SPI and WPI) were selected as model proteins with an acid-heat treatment for 5 h to prepare the nanofibrils. Protein molecules eventually lost their intrinsic structure, which was also followed by aggregation and degeneration. Atomic force microscopy was used to investigate how proteins' spherical and linear shapes changed over time. As the protein unfolded and intermolecular aggregation occurred, circular dichroism (CD) demonstrated that the protein's secondary structure altered from a-helix to β-sheet and random coils. Furthermore, the fibrous protein films' mechanical characteristics were enhanced, particularly the elongation (E) of amyloid SPI films, which increased from 33.50 % to 100.57 %. Additionally, the films' water solubility (WS) and water vapor permeability (WVP) were both decreased. In order to create an antibacterial edible film, vanillin was also added to the fibrous protein solution as a bacteriostatic agent. The results showed that the fibrous protein film demonstrated a great bacteriostatic action and retained good shape. Here we present this simple and efficient method to obtain robust, flexible films, which not only provides a strategy for films with good mechanical properties but also extends their antibacterial by adding a bacteriostatic agent to further provide a new idea for food packaging.
Preparation, Characterization, and Foaming Properties of Soy Protein Nanoparticles by the Cross-Linking Reaction Induced by Microbial Transglutaminase

Abstract:Native soy proteins have limited surface properties, but fortunately, it is expected that their foaming behaviors can be improved through the enzymatic cross-linking method. In this paper, by using the cross-linking method with microbial transglutaminase (mTG) as the catalyst, soy protein isolate (SPI), soy β-conglycinin (7S), and soy glycinin (11S) nanoparticles (NPs) were prepared in the molten state of the protein induced by dithiothreitol (DTT). We compared the structural characteristics of these nanoparticles, including particle sizes, morphologies, zeta potentials, surface hydrophobicities, changes of spatial structure, and cross-linking degrees. In addition, their foaming properties, including foamabilities, foam stabilities, and bubble diameters, were also tested. The results show that there are no significant differences in the foamabilities between the native soy proteins and their nanoparticles with similar initial foam heights. However, it is found that 11S NPs have enhanced foam stabilities with longer half-lives and smaller changes in bubble size, which is closely related to their high wettability and cross-linking degree. This study may provide references for the application of soy protein nanoparticles as new foam stabilizers to replace the traditional foaming agents in the food industry.
Sustainable Bioplastics from Amyloid Fibril-Biodegradable Polymer Blends

Abstract:Plastics waste production is a global challenging problem because its accumulation in the environment causes devastating effects on the planet's ecosystem. Sustainable and green solutions are urgently needed, and this pairs with increasingly stronger regulations combined with improved ecological awareness. This study proposes a simple, scalable, and water-based process to produce free-standing, transparent, and flexible bioplastics films by combining amyloid fibrils with biodegradable polymers as two main building blocks. Amyloid fibrils can be obtained through denaturation and self-assembly from a broad class of food proteins found in milk, soy, and egg, for example. Whey is used here as a model protein because it is the major byproduct of dairy industries, and its valorization creates a valuable opportunity to produce sustainable, biodegradable, and environmentally friendly bioplastics perfectly integrated within a circular economy. Abstract:Plant-derived proteins like rice protein are considered an ideal source for yielding amyloid fibrils due to sustainability and affordability. However, the inherent low solubility of rice protein often restricts its self-assembly capability under common fibrillization protocols. Hence, this study comprehensively investigated the effect of modification on the self-assembly behavior, protein structure and foaming properties of rice protein and its components. The modification facilitated the transformation of alpha-helix to beta-sheet, promoting fibrils formation. All modified proteins, except rice protein, exhibited significantly increased zeta potential values. AFM results revealed that the modified rice protein and globulin fibrils exhibited entangled aggregation behavior, while the glutelin fibril was notably extended, with several micrometers in length. Additionally, modification promoted the aggregation of albumin and prolamin particles to form short and straight fibrils. Amino acid analysis indicated that the content difference of amino acids with different characteristics and the change of aromatic amino acid interaction caused by modification may lead to different self-assembly behavior of the same protein. Furthermore, the foaming capability and stability of most protein fibrils were significantly improved by the modification. These findings provide valuable insights into the regulation of plant-derived protein fibrillation through modification.
Enhancement of physicochemical and techno-functional properties of soy protein isolate amyloid fibrils by moderate ultrasonic pretreatment

Abstract:Plant protein can be functionally strengthened by its modification into amyloid fibrils, which has drawn growing interest. In this study, we investigated the effects of ultrasonic pretreatment on the fibrillation kinetics, physicochemical properties, and functional aspects of soy protein isolate amyloid fibrils (SPF). Firstly, soy protein isolates (SPI) were subjected to ultrasonic pretreatment by regulating time and power under neutral conditions. Subsequently, the pretreated SPI was converted into SPF through acidic heating. The results indicated that ultrasonic pretreatment significantly improved surface hydrophobicity and solubility of SPI through ultrasound's cavitation and thermal effects. Thioflavin T fluorescence assay revealed SPF40 (450 W, 40 min) had the fastest growth rate, 104.23 % higher than SPF0 (without ultrasonic pretreatment), while SPF10 (450 W, 10 min) had the highest fibril yield, 20.97 % higher than SPF0. Physicochemical analysis showed that ultrasonic pretreatment accelerated the formation of SPF from SPI by exposing the surface hydrophobic groups, lowering the zeta potential, breaking down original fibrils into shorter fibrils, and altering the secondary structure from α-helices into β-sheets. Morphological examination by transmission electron microscopy exhibited a higher maturation state, showing more rigid and straight fibrils structure in SPF10 compared to SPF0. Functionally, SPF with ultrasonic pretreatment had higher emulsifying activity than SPI and SPF0. Rheological analysis further confirmed SPF emulsions' better mechanical strength over SPI emulsions. Overall, this study demonstrates that ultrasonic pretreatment is an effective way to enhance the formation and properties of SPF, which can be further applied to developing advanced protein products and novel functional food systems.

Abstract:Soy protein isolate (SPI) has attracted considerable attention in the field of packaging technology due to its easy processability, biodegradability, and good film-forming characteristics. However, SPI-based films often suffer from inferior mechanical properties and high moisture sensitivity, thus restricting their practical application. In the present study, herein, a biobased nanocomposite film was developed by cross-linking SPI matrix from the synergistic reinforcement of cellulose nanofibers (CNF) and nano-silica (NS) particles. First, we functionalized the CNF with NS using a silane agent (KH560) as an efficient platform to enhance the interfacial interaction between SPI and CNF/NS, resulting from the epoxy-dominated cross-linking reaction. The chemical structure, thermal stability, and morphology of the resultant nanocomposite films were comprehensively investigated via Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), scanning electron microscopy (SEM), and thermogravimetric analysis (TGA). These results supported successful surface modification and indicated that the surface-tailored CNF/NS nanohybrid possesses excellent adhesion with SPI matrix through covalent and hydrogen-bonding interactions. The integration of CNF/NS into SPI resulted in nanocomposite films with an improved tensile strength (6.65 MPa), representing a 90.54% increase compared with the pristine SPI film. Moreover, the resulting composites had a significantly decreased water vapor permeation and a higher water contact angle (91.75°) than that of the unmodified film. The proposed strategy of synergistic reinforcements in the biobased composites may be a promising and green approach to address the critical limitations of plant protein-based materials in practical applications.

Abstract:Electrospun films (ESF) are gaining attention for active delivery due to their biocompatibility and biodegradability. This study investigated the impact of adding soy protein amyloid fibrils (SAFs) to ESF. Functional ESF based on SAFs/pullulan were successfully fabricated, with SAFs clearly observed entangled in the electrospun fibers using fluorescence microscopy. The addition of SAFs improved the mechanical strength of the ESF threefold and increased its surface hydrophobicity from 24.8° to 49.9°. Moreover, the ESF demonstrated antibacterial properties against Escherichia coli and Staphylococcus aureus. In simulated oral disintegration tests, almost 100% of epigallocatechin gallate (EGCG) dissolved within 4 min from the ESF. In summary, the incorporation of SAFs into ESF improved their mechanical strength, hydrophobicity, and enabled them to exhibit antibacterial properties, making them promising candidates for active delivery applications in food systems. Additionally, the ESF showed efficient release of EGCG, indicating their potential for controlled release of bioactive compounds.

Abstract:This study evaluates eleven different sources from a wide range of plants, and narrows down the study to seven species of plant proteins from which excellent amyloid fibrils can be generated. Furthermore, the possible applications of these plant based amyloid fibrils in multifunctional materials, such as the hydrogels, biodegradable plastics, and water purification membranes are demonstrated in this work. Artificial functional materials based on amyloid fibrils are proven to be a promising strategy toward functional materials. However, scaling‐up applications present sustainability concerns, as animal proteins are the main sources for fabricating amyloid fibrils. Plant‐protein‐based amyloid fibrils, a more sustainable alternative to animal proteins, are attracting increasing interests as building blocks in functional materials. Herein, 11 different sources from a wide range of plants are evaluated, and a comprehensive analysis of seven species of plant proteins, including kidney bean, black bean, cowpea, mung bean, chickpea, lentil, and pumpkin seed, with an excellent ability to form fibrils, is presented. A universal strategy for a diversity of plant protein extraction and fibrillization is applied. Flexible fibrils with a persistence length of ≈100 nm and rigid fibrils of several micrometers are discovered in 7S/8S and 11S subunits dominated protein, respectively. Structural evolution toward the β‐sheet content on these proteinaceous assemblies is characterized by thioflavin T (ThT) intensity, circular dichroism (CD) spectra, attenuated total reflectance‐Fourier transform infrared (ATR‐FTIR) spectra, and typical wide angle X‐ray scattering (WAXS) spectra. Finally, their multifunctional applications are further explored and proven that these sustainable protein amyloids demonstrate excellent performance in renewable and degradable bioplastics, and in water purification membranes for heavy metal removal.

Abstract:This study investigated the addition of cellulose nanoparticles (NC) and green extracted phenolic compounds from date palm leaves (DLE) as reinforcing and bioactive agent, respectively, into the soy protein isolate (SPI) edible films. Structural, physiochemical, optical, mechanical, barrier, thermal, and bioactive properties of the films were explored. The incorporation of NC improved the SPI film’s physiochemical, mechanical, and barrier properties but did not show effect on the antioxidant and antibacterial properties. Following the incorporation of DLE in liquid form (DLWE) the film’s antioxidant and antibacterial properties significantly increased but due to the presence of natural deep eutectic solvent (NADES) (plasticizer) all other properties were negatively affected. However, enriching the SPI-NC films with DLE in powder form (DLPE) improved the bioactive properties while maintaining the improved mechanical and barrier properties. These findings indicated that SPI-NC/DLPE films exhibited improvements in the whole performance and provided promising strategy for formulation of biodegradable packaging films. This approach is an effective strategy towards sustainable utilization of unexplored agricultural wastes (date palm leaves derived NC and phenolic compounds) for development of active and environmentally friendly food packaging material.

Abstract:In this study, nanocomposite active films were fabricated containing silver nanoparticles (SNPs) embedded within soy protein isolate (SPI)/Persian gum (PG) matrices. The physical, mechanical, and antibacterial properties of these composite films were then characterized. In addition, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction (XRD) were used to provide information about the microstructure, interactions, and crystallinity of the films. Pure SPI films had poor physicochemical attributes but the addition of PG (0.25, 0.5, or 1 wt%) improved their water vapor permeability, mechanical properties, and water solubility (WS). The moisture content (MC) of the films decreased after the introduction of PG, which was attributed to fewer free hydroxyl groups to bind to the water molecules. Our results suggest there was a strong interaction between the SPI and the PG and SNPs in the films, suggesting these additives behaved like active fillers. Optimum film properties were obtained at 0.25% PG in the SPI films. The addition of PG (0.25%) and SNPs (1%) led to a considerable increase in tensile strength (TS) and a decrease in elongation at break (EB). Furthermore, the incorporation of the SNPs into the SPI/PG composite films increased their antibacterial activity against pathogenic bacteria (Escherichia coli and Staphylococcus aureus), with the effects being more prominent for S. aureus. Spectroscopy analyses provided insights into the nature of the molecular interactions between the different components in the films. Overall, the biodegradable active films developed in this study may be suitable for utilization as eco-friendly packaging materials in the food industry.
Tannic acid-loaded halloysite clay grafted with silver nanoparticles enhanced the mechanical and antimicrobial properties of soy protein isolate films for food-packaging applications

Abstract:Poor mechanical and water barrier properties restrict the applicability of protein-based packaging in high-moisture food commodities. In this study, a nanocomposite (Nc) of tannic acid-loaded halloysite nanotubes grafted with silver nanoparticles was developed as a reinforcement agent to enhance the functional properties of soy protein isolate (SPI)-based films. The 5%- and 10%-Nc incorporation in SPI films significantly (p < 0.05) improved the tensile strength (by 1.94–3.14 fold) and thermal stability (by 5–7%) and reduced water vapor permeability (by 1.28–1.30 fold), oxygen permeability (by 2.72–3.84 fold), and water solubility (by 6.3–9.5 fold). The 5%- and 10% Nc/SPI films showed antioxidant properties (by scavenging 18%−28% of DPPH) and exhibited a 3–5 log10 reduction in multi-drug resistant S. aureus and S. Typhimurium contamination in chicken breast fillets. Although 10% Nc/SPI exhibited silver migration to chicken fillets beyond the acceptable limit contributing to color change, 5% Nc/SPI films enhanced the shelf-life of fillets for 4 days by facilitating a controlled pH, lipid oxidation, and microbial growth with acceptable levels of silver migration. In conclusion, the multifunctionality of Nc in SPI films enables its potential to be an alternative to non-biodegradable and non-eco-friendly polymers.

Abstract:Soy protein based materials are of great interest because of the merits of biocompatibility, biodegradability, renewability, etc. However, the poor mechanical properties and high water sensitivity limit their further application in many fields. In this paper, we tried to overcome these shortcomings through a slight chemical modification of the polypeptide chains of soy protein. 31P NMR and solid state 13C CP/MAS NMR spectroscopy confirmed that the diethoxy phosphoryl groups were successfully grafted onto soy protein chains with a molar grafting ratio of 0.15–1.18%, which almost did not change the nature of soy protein. The isoelectric point and rheological behavior of the modified soy protein sample varied with the grafting ratio, indicating that the tertiary structure of the protein was changed after phosphoryl modification. The FTIR spectra of the modified soy protein suggested that the increase of β-sheet conformation from the slight chemical modification could be the reason for the change of the globular structure of soy protein. Finally, we obtained a robust soy protein film as expected, and we did not use any crosslinking agent and plasticizer that were almost unavoidable in the previous studies reported in the literature. The tensile strength and the elongation at break of our soy protein films were 35 ± 5 MPa, 2.5 ± 0.5% in the dry state, and 3.8 ± 1.5 MPa, 125 ± 5% in the wet state, respectively. We believe that the method we developed in this communication provides a practical approach to improve the mechanical properties and broaden the applications of natural soy protein based materials.

Abstract:Alternative materials to replace the synthetic plastics are being developed using agro‐based sources, but despite their promising results, these materials still need to have their performance improved to be used in packaging. Blending different molecules can combine the properties of each polymer resulting in the overall performance improvement of the material. In the present study, collagen, whey protein, and methylcellulose were investigated in the development of individual films and blends. They were produced by the casting technique and evaluated for their packaging‐related characteristics (including mechanical, barrier, and color properties), microstructure, thermal properties, and biodegradability in soil. Methylcellulose films presented excellent technological properties, such as total solubility in water, high tensile strength (15.78 MPa), transparency (30.4%), and good barrier to water vapor (0.43 g·mm/h·m2·kPa). In the blends with collagen and whey protein, it was responsible by the increase in tensile strength, barrier, and thermal properties. Collagen films presented the highest elongation at break (101.4%), while whey protein films showed lower solubility in water (28.3%). All the samples tested were completely biodegraded in 10 days in soil. Potential applications for the materials developed include soluble sachets for powdered foods due to the high solubility of some samples, as well as oil containers and capsules for instant coffee machines.

Robust design of starch composite nanofibrous films for active food packaging: Towards improved mechanical, antioxidant, and antibacterial properties

Abstract:Developing efficient and biodegradable packaging films is of paramount significance owing to the scarcity of petroleum based resources. However, their applications in food packaging are limited due to their poor mechanical properties and inadequate biological activities. This study proposes a novel approach to develop the starch composite nanofibrous films (SNFs/TA/Fe3+) consisting of starch, tannic acid, and Fe3+ using the temperature-assisted electrospinning method. The addition of TA resulted in a decrease in the rate of thermal degradation, indicating an improvement in the thermal stability of SNFs. However, the incorporation of TA or TA/Fe3+ showed only a slight impact on the internal structure of SNFs. SNFs/TA/Fe3+ loaded with 0.1 wt% of Fe3+ demonstrated a significantly higher tensile strength compared to SNFs and those loaded with TA alone. The presence of TA enhances the antioxidant activity of SNFs, and the robust SNFs/TA/Fe3+ exhibited comparable antioxidant activity to SNFs/TA. However, the SNFs/TA/Fe3+ showed a reduction in antibacterial activity, possibly due to the high valence state of the metal ions. Overall, these findings highlighted that a simple electrospinning method was used to produce SNFs/TA/Fe3+ resulted in improved mechanical properties and antioxidant activity, offering a new strategy for the development of active food packaging using SNFs.
Exploring the effect of corn starch/pea protein blending on the physicochemical and structural properties of biopolymer films and their aging resistance

Abstract:This study investigated the potential effect of blending corn starch and pea protein isolate in various ratios (100:0, 70:30, 50:50, 30:70, and 0:100) on the aging properties of biodegradable films. Unlike previous research, the focus was on the often-overlooked aspect of film aging. Fourier-transform infrared spectroscopy and X-ray diffraction demonstrated the physical blending of corn starch and pea protein, along with chemical bonding and conformational changes. The optical and microstructural properties showed the formation of smooth, homogeneous films with good compatibility between the polymers. The water resistance, barrier, and mechanical properties corresponding to the intrinsic nature of protein polymers showed a minimized fluctuations in film properties as film ages, with a reduction of at least twice when protein is added. Remarkably, the blend with a ratio of 30:70 demonstrated the most stable properties during aging. These results demonstrated that blending the pea protein isolate was favorable for delaying the retrogradation and recrystallization of corn starch films. Understanding how these blends influence the aging characteristics of films is not only a novel contribution to the scientific community but also holds practical significance, potentially opening a potential for applications in various industries.
Nanoparticle and Dialdehyde Polysaccharide Synergistically Improving the Physicochemical Properties of Soy Protein Isolate Films Enriched with Lingonberry Extract

Abstract:The combination of proteins and anthocyanins showed great potential in constructing smart films, but their weak interactions led to poor mechanical properties in the resulted films. This study enhanced the physicochemical properties of lingonberry extract (LE)-enriched soy protein isolate (SPI) smart active films by incorporating dialdehyde konjac glucomannan (DKGM) as a cross-linker and quercetin-loaded shellac nanoparticles (SNPs-Qu) as fillers. The films' chemical structure, thermal properties, surface and cross-sectional morphology were analyzed using FT-IR, DSC, and SEM, respectively. Additionally, the light-shielding properties, mechanical properties, water vapor permeability, and antioxidant properties of the films were systematically studied. The results indicated considerable interactions among film components. The combination of DKGM with SNPs-Qu significantly enhanced UV absorption while reducing moisture content from 35.07% to 24.84% and water vapor transmission from 0.63 to 0.41 (mm.g.m(-2).h(-1).KPa-1) of the films. Importantly, DKGM positively affected thermal stability and mechanical properties whereas SNPs-Qu improved internal structure and the antioxidant properties of the films significantly. Notably, film color shifted from red to yellow-green with rising pH levels effectively tracking pork filling freshness status. Overall, the smart active film constructed from SPI, LE, DKGM, and SNPs-Qu exhibited excellent physicochemical properties alongside pH-sensitive qualities-promising applications in smart food packaging.
Plant Protein-Based Films for Food Packaging Applications

Abstract:The use of plastics for packaging across many industries has become a standard, however, in recent years there has been an increase in demand for more eco-friendly packaging alternatives. In this vein, the use of plant proteins in film formation has been studied extensively.1-2 Due to the strong mechanical properties obtained through the use of plant-based proteins, the biodegradability of such materials, and their economic efficiency, these proteins are of substantial interest in the effort to replace current synthetic packaging films.3 -5 In our work, we focus on combining both experimental and computational techniques in order to develop protein-based films for food packaging applications. By combing the two techniques, we aim to better understand the interactions of plant-based proteins with selected plasticizing modifiers in order to develop films with optimal mechanical properties. In our work we compare the interactions of selected modifiers with soy protein and zein protein from corn. In this regard, we apply various computational techniques, including protein-ligand docking6-7 and molecular modeling methods8-9 to assess the interactions and then compare our finding with experimental data1,3,10-11. By modeling how choice plasticizing modifiers interact differently with each protein, we aim to better formulate our films in order to achieve mechanical properties that compete with those of current synthetic packaging systems. Improved structure-stability and packaging characters of crosslinked collagen fiber-based film with casein, keratin and SPI

Abstract:Background: To improve the structure-stability and packing characters of collagen fiber, we manufactured crosslinked collagen fiber (CColF)-based edible films using transglutaminase (TGase). Then we made a comparison on structure-stability and packing characteristics among the CColF-based films loaded with casein (CN), keratin (KRT) and soy protein isolate (SPI), respectively. Results: Observed from scanning electron microscopy (SEM), the CColF loaded with CN, KRT and SPI showed some unique morphology of the additional proteins. The CColF-protein films performed better packing characteristics including barrier properties, mechanical properties and thermal-stability properties, compared with CColF films. Importantly, with 500 g kg-1 CN (of CColF) addition, CColF-based films possessed a greater thermal stability than the other films judged from differential scanning calorimetry (DSC). Meanwhile, the CColF loaded with 100 g kg-1 CN provided a higher value of tensile strength (TS) and the CColF loaded with 100 g kg-1 KRT showed a higher value in elongation-at-break (EAB) than the other films. Conclusion: In conclusion, the collagen fiber-based edible films with better structure-stability and packing characteristics for food packaging was obtained which could be an advantage to promote the development of the application of collagen in packing products. © 2019 Society of Chemical Industry.
