Inhibition and Mechanism of Citral on Bacillus cereus Vegetative Cells, Spores, and Biofilms

Abstract:Bacillus cereus causes food poisoning by producing toxins that cause diarrhea and vomiting and, in severe cases, endocarditis, meningitis, and other diseases. It also tends to form biofilms and spores that lead to contamination of the food production environment. Citral is a potent natural antibacterial agent, but its antibacterial activity against B. cereus has not been extensively studied. In this study, we first determined the minimum inhibitory concentrations and minimum bactericidal concentrations, growth curves, killing effect in different media, membrane potential, intracellular adenosine triphosphate (ATP), reactive oxygen species levels, and morphology of vegetative cells, followed by germination rate, morphology, germination state of spores, and finally biofilm clearance effect. The results showed that the minimum inhibitory concentrations and minimum bactericidal concentrations of citral against bacteria ranged from 100 to 800 μg/mL. The lag phase of bacteria was effectively prolonged by citral, and the growth rate of bacteria was slowed down. Bacteria in Luria-Bertani broth were reduced to below the detection limit by citral at 800 μg/mL within 0.5 h. Bacteria in rice were reduced to 3 log CFU/g by citral at 4000 μg/mL within 0.5 h. After treatment with citral, intracellular ATP concentration was reduced, membrane potential was altered, intracellular reactive oxygen species concentration was increased, and normal cell morphology was altered. After treatment with citral at 400 μg/mL, spore germination rate was reduced to 16.71%, spore morphology was affected, and spore germination state was altered. It also had a good effect on biofilm removal. The present study showed that citral had good bacteriostatic activity against B. cereus vegetative cells and its spores and also had a good clearance effect on its biofilm. Citral has the potential to be used as a bacteriostatic substance for the control of B. cereus in food industry production.
Antibacterial Activity and Mannosylerythritol Lipids Against Vegetative Cells and Spores of Bacillus Cereus

Abstract:This study aimed to investigate the antibacterial activity and underlying mechanism of mannosylerythritol lipids (MELs) against vegetative cells and spores of Gram-positive Bacillus cereus. The minimum inhibitory concentration and minimum bactericidal concentration of MELs was 1.25 mg/mL and 2.50 mg/mL, respectively. The growth curve assay and survival rate also showed that MELs had great antibacterial effect The analysis data of intracellular constituent leakage and cell viability demonstrated MELs damaged the integrity of cell membrane. By scanning electron microscope and transmission electron microscope, the treated bacterial cells exhibited apparent morphological and ultrastructural changes, further affirming the disruption of cell membrane and the unbalance of cellular environments. Furthermore, MELs has showed good ability to inhibit the germination of B. cereus spores. The findings suggested that MELs acted its antibacterial effect primarily by means of membrane disruption, and indicating it will be alternatives to antibacterial agents and preservatives applied into food processing.
Effect of Temperatures on the Growth, Toxin Production, and Heat Resistance of Bacillus Cereus in Cooked Rice

Abstract:Bacillus cereus is capable of producing enterotoxin and emetic toxin, and Bacillus foodborne illnesses occur due to the consumption of food contaminated with endospores. The objectives of this study were to investigate the growth and toxin production of B. cereus in cooked rice and to determine the effect of temperature on toxin destruction. Cooked rice inoculated with B. cereus was stored at 15, 25, 35, and 45 degrees C or treated at 80, 90, and 100 degrees C. The results indicated that emetic toxin was produced faster than enterotoxin (which was not detected below 15 degrees C) at all the storage temperatures (15-45 degrees C) during the first 72h. Emetic toxin persisted at 100 degrees C for 2h, although enterotoxin was easily to be destroyed by this treatment within 15min. In addition, B. cereus in cooked rice stored at a warm temperature for a period was not inactivated due to survival of the thermostable endospores. These data indicate that the contaminated cooked rice with B. cereus might present a potential risk to consumers. Results from this study may help enhance the safety of such food, and provide valuable and reliable information for risk assessment and management, associated with the problem of B. cereus in cooked rice.
Control Effects of Bacillus Siamensis G-3 Volatile Compounds on Raspberry Postharvest Diseases Caused by Botrytis Cinerea and Rhizopus Stolonifer

Abstract:Post-harvest diseases of raspberries affect their storage and subsequently the ability to provide consumers with fresh products. The antifungal activity of microbial volatile organic compounds (MVOCs) against both pre and postharvest diseases has previously been demonstrated, but the control of raspberry postharvest diseases caused by Botrytis cinerea and Rhizopus stolonifer using MVOCs has not been extensively studied. We have thus investigated the effects of Bacillus siamensis G-3 volatile organic compounds (VOCs) on controlling the diseases caused by B. cinerea and R. stolonifer. Bioassays of the VOCs of B. siamensis G-3 exhibited significant control effects (89.4%) on the two pathogens. The VOCs composition was studied with solid phase micro extraction and GC/mass spectrometry (SPME-GC-MS), and 22 VOCs, different from the controls, were identified. Among these compounds, only 2, 6-di-tert-butyl-4-methylphenol (BHT), and 2,4-di-tert-butylphenol (2,4-DTBP) had significant control effects on the pathogens (*P < 0.05), 52.38 and 93.33%, respectively, compared with the controls in vivo. With the 2,4-DTBP treatments, the raspberries could be stored for 20 d at 0 degrees C, with the disease rate maintained at similar to 10%. In addition, we have reported for the first time, that 2,4-DTBP may be a better preservative than BHT and could be used as a bio-fumigant for controlling raspberry postharvest diseases caused by B. cinerea and R. stolonifer in vivo.

Abstract:Bacillus cereus (B. cereus) functions as a probiotic in animals, but the underlying mechanisms remain unclear. We aim to evaluate the protective effects and definite mechanism by which orally administered B. cereus prevents D-galactosamine (D-GalN)-induced liver injury in rats. Twenty-one Sprague–Dawley rats were equally assigned into three groups (N = 7 animals per group). B. cereus ATCC11778 (2 × 109 colony-forming units/ml) was administered to the B. cereus group via gavage, and phosphate-buffered saline was administered to the positive control (PC) and negative control (NC) groups for 2 weeks. The PC and B. cereus groups received 1.1 g/kg D-GalN via an intraperitoneal injection to induce liver injury. The blood, terminal ileum, liver, kidney and mesenteric lymph nodes (MLNs) were collected for histological examinations and to evaluate bacterial translocation. Liver function was also determined. Fecal samples were collected for deep sequencing of the 16S rRNA on an Illumina MiSeq platform. B. cereus significantly attenuated D-GalN-induced liver injury and improved serum alanine aminotransferase (ALT) and serum cholinesterase levels (P < 0.05 and P < 0.01, respectively). B. cereus modulated cytokine secretion, as indicated by the elevated levels of the anti-inflammatory cytokine interleukin-10 (IL-10) in both the liver and plasma (P < 0.05 and P < 0.01, respectively) and the substantially decreased levels of the cytokine IL-13 in the liver (P < 0.05). Pretreatment with B. cereus attenuated anoxygenic bacterial translocation in the veins (P < 0.05) and liver (P < 0.05) and upregulated the expression of the tight junction protein 1. The gut microbiota from the B. cereus group clustered separately from that of the PC group, with an increase in species of the Ruminococcaceae and Peptococcaceae families and a decrease in those of the Parabacteroides, Paraprevotella, and Desulfovibrio families. The potential probiotic B. cereus attenuated liver injury by restoring the gut flora balance and enhancing the intestinal barrier function.

Abstract:Controlling harmful algal blooms with algicidal bacteria is thought to be an efficient and eco-friendly way but lack of comprehensive studies from theory to practice limited the field application. Here we presented a purple bacterial strain Duganella sp. A3 capable of killing several harmful algae, including Heterosigma akashiwo , a world-wide fish-killing microalga. A bioactivity-guided purification and identification approach revealed the major algicidal compound of A3 as the pigment violacein, which was never reported for its algicidal potential before. Violacein rapidly disrupted cell permeability, caused long-term oxidative stress, but mildly affected algal photosystem, which might explain its highly species-specific activity against unarmored H. akashiwo . To explore the application potential of violacein, a fermentation optimization approach combing single-factor and multi-factor experiments was conducted to increase the violacein yield, which finally reached 0.4199 g/L just using a simple medium formula beneficial for compound purification. Finally, taking advantages of the physical and chemical stabilities, we successfully developed the novel application of violacein as a sustained-releasing and easy-to-preserve algicidal agent using alginate-acacia-gum-chitosan encapsulation, which paved the path for its future application in controlling H. akashiwo bloom.

Abstract:Bacillus cereus is a well-known foodborne pathogen that can cause human diseases, including vomiting caused by emetic toxin, cereulide, requiring 105-108 cells per gram to cause the disease. The bacterial cells may be eliminated during processing, but cereulide can survive in most processing techniques due to its resistance to high temperatures, extreme pH and proteolytic enzymes. Herein, we reported dynamic processes of biofilm formation of four different types and cereulide production within the biofilm. Confocal laser scanning microscopy (CLSM) images revealed that biofilms of the four different types reach each stage at different time points. Among the extracellular polymeric substances (EPS) components of the four biofilms formed by the emetic B. cereus F4810/72 strain, proteins account for the majority. In addition, there are significant differences (p < 0.05) in the EPS components at the same stage among biofilms of different types. The time point at which cereulide was first detected in the four types of biofilms was 24 h. In the biofilm of B. cereus formed in ultra-high-temperature (UHT) milk, the first peak of cereulide appeared at 72 h. The cereulide content of the biofilms formed in BHI was mostly higher than that of the biofilms formed in UHT milk. This study contributes to a better understanding of food safety issues in the industry caused by biofilm and cereulide toxin produced by B. cereus.

Abstract:The use of natural preservatives has attracted considerable attention owing to their generally safe and environmentally friendly properties. In this study, we investigated the effects of the preservative A1, composed of plantaricin 163, thymol, and surfactin, on bacterial communities and storage quality of refrigerated crucian carp. A total of 522 operational taxonomic units belonging to 20 phyla and 272 genera were identified by high-throughput sequencing, showing a comprehensive coverage of bacterial composition of crucian carp. In untreated samples after spoilage, Brochothrix was the predominant genus, followed by Aeromonas and Pseudomonas. After treatment with A1, the growth of these spoilage bacteria was significantly inhibited according to high-throughput sequencing and plate counts, and Lactococcus became the most abundant organism at the end of storage. Meanwhile, compared with control samples, the shelf life of A1-treated samples extended from 3 to 12 days on the basis of the sensory evaluation and the total viable counts. Furthermore, the total volatile basic nitrogen, thiobarbituric acid, and pH values for A1-treated samples were significantly lower than that of control samples. The results indicate that preservative A1 has potential commercial application in the preservation of refrigerated crucian carp.
Preservative effects of a novel bacteriocin from Lactobacillus panis C-M2 combined with dielectric barrier discharged cold plasma (DBD-CP) on acquatic foods

Abstract:In this study, a novel bacteriocin Lactocin C-M2 produced by Lactobacillus panis C-M2, combined with dielectric barrier discharged cold plasma (DBD-CP), was used to evaluate the antibacterial effect on aquatic foods. After the purification procedures of ethyl acetate extraction, cation exchange chromatography and semi-preparative liquid phase, the stability of Lactocin C-M2 under DBD-CP environment was determined, and the preservation effect of these two joint treatments was investigated on fresh white fish samples. As revealed by LC-MS/MS and BLAST analysis, Lactocin C-M2 is a new type of class II bacteriocin, with a molecular weight of 863.52 Da and the N-terminal sequence MVKKTSAV. Application of Lactocin C-M2 showed significantly stronger inhibitory effect on bacteria than on yeasts and mold. All the tested 10 Gram positive bacteria and 3 Gram-negative bacteria, including Staphylococcus aureus, Shigella flexneri, Bacillus spp, Lactobacillus spp, Escherichia coli, Pseudomonas aeruginosa, and so on, were inhibited. Lactocin C-M2 also presented stable antibacterial activity after exposure to DBD-CP, with the 95% residual activity against Staphylococcus aureus under the 40∼80 kV voltage for 30∼180 s, indicating the possibility of synergistic application. Combined with addition of 0.9 mg/g Lactocin C-M2, the treatment of DBD-CP with voltage at 60 kV for 90 s on fresh white fish (Culter alburnus) could significantly inhibit the microbial growth, the accumulation of volatile nitrogen and histamine during the storage. Therefore, the Lactocin C-M2, used together with the DBD-CP, is effective in food preservation.

Abstract:Phenyllactic acid (PLA) as a natural phenolic acid exhibits antibacterial activity against non-spore-forming bacteria, while the inhibitory effect against bacterial spore remained unknown. Herein, this study investigated the inactivation effect of PLA against Bacillus cereus spores. The results revealed that the minimum inhibitory concentration of PLA was 1.25 mg/mL. PLA inhibited the outgrowth of germinated spores into vegetative cells rather than germination of spores. PLA disrupted the spore coat, and damaged the permeability and integrity of inner membrane. Moreover, PLA disturbed the establishment of membrane potential due to the inhibition of oxidative metabolism. SEM observations further visualized the morphological changes and structural disruption caused by PLA. Besides, PLA caused the degradation of DNA of germinated spores. Finally, PLA was applied in milk beverage, and showed promising inhibitory effect against B. cereus spores. This finding could provide scientific basis for the application of PLA against spore-forming bacteria in food industry.
Interaction of a novel Bacillus velezensis (BvL03) against Aeromonas hydrophila in vitro and in vivo in grass carp

Abstract:This study evaluated the inhibition and interaction of Bacillus velezensis BvL03 as a probiotic agent against Aeromonas hydrophila. Strain BvL03 isolated from sediment samples of fish ponds had excellent antimicrobial activity against several fish pathogenic bacteria, especially Aeromonas, including A. hydrophila, A. veronii, A. caviae, and A. sobria. The successful amplification of lipopeptide antimicrobial chemical biosynthetic genes, including iturin family (ituA, ituB, and ituD), bacillomycin family (bacA, bacD, and bacAB), surfactin family (srfAB, srfC, and srfAA), and subtilosin family (albF and sunT) from the genome of BvL03 strain, confirmed its predominant antimicrobial activity. The challenge test suggested that BvL03 significantly decreased fish mortality when challenged with A. hydrophila, which had a cumulative mortality of 12.5% in the treatment group. Toxicity and hemolytic activity of A. hydrophila after co-cultured with BvL03 were relieved as confirmed by the cell experiments, when the initial inoculated concentration of BvL03 was 109 cfu/mL or higher. Moreover, the BvL03 strain labeled with GFP protein (BvL03-GFP) and AhX040 strain labeled with mCherry protein (AhX040-mCherry) were injected into grass carps. The fluorescence levels were monitored by using In Vivo Imaging System (IVIS), in which the green color was steadily increasing, whereas the red color was gradually weakening. Whole genome sequencing revealed that strain BvL03 possesses 15 gene clusters related to antibacterial compounds, including 5 NRPS gene clusters and 3 PKS gene clusters. These results suggested that B. velezensis BvL03 has the potential to be developed as a probiotic candidate against A. hydrophila infection in aquaculture.

Purification and characterization of a novel bacteriocin from Lactiplantibacillus plantarum Z057, and its antibacterial and antibiofilm activities against Vibrio parahaemolyticus

Abstract:Vibrio parahaemolyticus and its biofilm threaten food safety and human health. Bacteriocins are known to inhibit various foodborne bacteria and their biofilms; however, few bacteriocins with antibiofilm activity against V. parahaemolyticus have been reported. Herein, a novel plantaricin Z057 from Lactiplantibacillus plantarum Z057 was extracted with ethyl acetate, purified by Sephadex LH-20 gel chromatography and reversed-phase high-performance liquid chromatography, and identified by nano-liquid chromatography coupled with tandem mass spectrometry. Its molecular mass and amino acid composition were 1272.61 Da and A-G-G-D-G-G-H-G-G-S-I-I-L-Y, respectively. Plantaricin Z057 presented a broad antibacterial spectrum against Gram-positive and Gram-negative pathogens. Moreover, it also exhibited excellent heat tolerance, maintained antibacterial activity at pH 2.0–4.0, and was susceptible to common proteases (pepsin, trypsin, papain, proteinase K). In addition, scanning electron microscopy revealed that plantaricin Z057 destroyed membrane integrity and caused cell deformation and cytoplasmic leakage. The crystal violet staining and XTT reduction assays revealed that plantaricin Z057 markedly decreased biofilm biomass and metabolic activity of V. parahemolyticus . Fluorescence microscopy showed that plantaricin Z057 efficiently inactivated sessile cells, and damaged integrated biofilm structure by reducing the extracellular polymer content. In summary, plantaricin Z057 exhibited significant potential for controlling V. parahaemolyticus and could be used in food production as a biological preservative.

Abstract:Abstract Bacillus cereus, a spore-forming bacterium and frequent cause of food poisoning, poses a safety threat to dairy and rice industries due to its high contamination rates and ability to produce toxins like cereulide. Because of widespread presence and thermal resistance of the spores, B. cereus cannot be eliminated from the environment and may survive in processing plants. Survived spores can develop into vegetative cells, leading to a heightened risk of cereulide production in the processing environment. Both spores and vegetative cells have an ability to adhere to the surfaces of dairy plants and form biofilm, serving as the site for cereulide production and accumulation. Therefore, it is crucial for the food industry to address potential sources and pathways of B. cereus contamination and their connections to cereulide production in processing lines. In this review, the sources of contamination of B. cereus, including spores, vegetative cells, and biofilms, and analyzes their potential role in cereulide production at each stage of dairy and cooked rice processing were analyzed. In addition, the controlling methods to prevent B. cereus contamination and cereulide production in the processing lines were proposed, offering valuable insights for improving microbial risk management in the food industry.

Abstract:Bacillus cereus is a prevalent foodborne pathogen that induces food poisoning symptoms such as vomiting and diarrhea. Its capacity to form spores and biofilm enables it to withstand disinfectants and antimicrobials, leading to persistent contamination during food processing. Consequently, it is necessary to develop novel and efficient antimicrobial agents to control B. cereus, its spores, and biofilms. Peptidoglycan hydrolases have emerged as a promising and eco-friendly alternative owing to their specific lytic activity against pathogenic bacteria. Here, we identified and characterized a Lysozyme-like cell wall hydrolase Lys14579, from the genome of B. cereus ATCC 14579. Recombinant Lys14579 specifically lysed B. cereus without affecting other bacteria. Lys14579 exhibited strong lytic activity against B. cereus, effectively lysing B. cereus cell within 20 min at low concentration (10 μg/mL). It also inhibited the germination of B. cereus spores and prevented biofilm formation at 12.5 μg/mL. Moreover, Lys14579 displayed good antimicrobial stability with negligible hemolysis in mouse red blood cells and no cytotoxicity against RAW264.7 cells. Notably, Lys14579 effectively inhibited B. cereus in boiled rice and minced meat in a dose-dependent manner. Furthermore, bioinformatics analysis and point mutagenesis experiments revealed that Glu-47 was the catalytic site, and Asp-57, Gln-60, Ser-61 and Glu-63 were active-site residues related with the cell wall lytic activity. Taken together, Lys14579 could be a promising biocontrol agent against vegetative cells, spores, and biofilm of B. cereus in food industry.

Effect of Bacillus velezensis on Aeromonas veronii-Induced Intestinal Mucosal Barrier Function Damage and Inflammation in Crucian Carp (Carassius auratus)

Abstract:Aeromonas veronii is an emerging aquatic pathogen causing hemorrhagic septicemia in humans and animals. Probiotic is an effective strategy for controlling enteric infections through reducing intestinal colonization by pathogens. Here we report that the consumption of Bacillus velezensis regulated the intestinal innate immune response and decreased the degree of intestinal inflammation damage caused by the A. veronii in Crucian carp. In this study, we isolated four strains of B. velezensis, named C-11, S-22, L-17 and S-14 from apparently healthy Crucian carp, which exerted a broad-spectrum antimicrobial activity inhibiting both Gram-positive and Gram-negative bacteria especially the fish pathogens. B. velezensis isolates showed typical Bacillus characteristics by endospore staining, physiological and biochemical test, enzyme activity analysis (amylase, protease, and lipase), and molecular identification. Here, Bacillus-containing dietary was orally administrated to Crucian carp for 8 weeks before A. veronii challenge. Immunological parameters and the expression of immune-related genes were measured at 2, 4, 6, 8, and 10 weeks post-administration. The results showed that B. velezensis was found to promote the increase in the phagocytic activities of peripheral blood leukocytes (PBLs) and head kidney leukocytes (HKLs), as well as the increase in interleukin 1β (IL-1β), IL-10 and tumor necrosis factor α (TNF-α) concentration of serum. Lysozyme levels (113.76 U/mL), ACP activity (25.32 U/mL), AKP activity (130.08 U/mL), and SOD activity (240.63 U/mL) were maximum (P < 0.05) in the B. velezensis C-11 treated group at 8 week. Our results showed that Crucian carp fed with the diet containing B. velezensis C-11 and S-22 developed a strong immune response with significantly higher (P < 0.05) levels of IgM in samples of serum, mucus of skin and intestine compared to B. velezensis L-17 and S-14 groups. Moreover, B. velezensis spores appeared to show no toxicity and damage in fish, which could inhabit the gut of Crucian carp. B. velezensis restrained up-regulation of pro-inflammation cytokines (IL-1β, IFN-γ, and TNF-α) mRNA levels in the intestine and head kidney at final stage of administration, and the expression of IL-10 was increased throughout the 10-week trial. A. veronii infection increased the population of inflammatory cells in the intestinal villi in the controls. In contrast, numerous goblet cells and few inflammatory cells infiltrated the mucosa in the B. velezensis groups after challenge with A. veronii. Compared with A. veronii group, B. velezensis could safeguard the integrity of intestinal villi. The highest post-challenge survival rate (75.0%) was recorded in B. velezensis C-11 group. The present data suggest that probiotic B. velezensis act as a potential gut-targeted therapy regimens to protecting fish from pathogenic bacteria infection.IMPORTANCEIn this work, four Bacillus velezensis strains isolated from apparently healthy Crucian carp, which exhibited a broad-spectrum antibacterial activity especially the fish pathogens. Administration of B. velezensis induced the enhancement of the intestinal innate immune response through reducing intestinal colonization by pathogens. The isolation and characterization would help better understand probiotic can be recognized as an alternative of antimicrobial drugs protecting human and animal health.

Effects of Sub-Minimum Inhibitory Concentrations of Bacteriocin BM173 on Listeria Monocytogenes Biofilm Formation

Abstract:Listeria monocytogenes is a significant foodborne pathogen that can form biofilms on various food processing surfaces, thereby enhancing resistance to disinfectants and exacerbating harm to human health. Previous studies have indicated that bacteriocin BM173 exhibits antibacterial and antibiofilm activities. In the current study, our aim was to assess the inhibitory mode of action of sub-inhibitory concentrations (SICs, 1/32 × MIC and 1/16 × MIC) of BM173 on the biofilm formation L. monocytogenes. Crystal violet staining assay revealed that SICs of BM173 significantly inhibit L. monocytogenes biofilm formation. Furthermore, the results of swimming motility assay, plate count, ruthenium red staining, and scanning electron microscopy (SEM) revealed that SICs of BM173 could effectively reduce the movement, cell adhesion, and exopolysaccharide (EPS) production of L. monocytogenes, thereby inhibiting biofilm formation. Real-time quantitative PCR analyses further demonstrated that SICs of BM173 down-regulated the expression of biofilm-associated genes, including those encoding adhesion, virulence factors, and quorum sensing. Additionally, SICs of BM173 effectively reduced the biofilm formation of L. monocytogenes on the surfaces of three food-grade materials (glass, stainless steel, and silicone) at 4 and 25 °C. These outcomes suggest that BM173 holds great potential for development as a promising food preservative for application in the food industry.
Effects of Bacillus on Water Quality, Growth and Antioxidant Activity of Intestinal Mucosa of Ctenopharyngodon Idellus

Abstract:A 45d culture experiment was conducted to study the effect of Bacillus on growth, antioxidant activity of intestinal mucosa of grass carp （ Ctenopharyngodon idellus） and water quality. 450 grass carps with initial average weight of （51.0 ± 2.3） g were randomly divided into 3 groups （control group, treatment group 1 and 2 ; three rep- licates for each group and 50 fish per replicate）. Control group was fed with the basal diet, Group 1 were fed the basal diet supplemented with Bacillus complex （ proportion of mixture of B. subtilis and B. amyloliquefaciens ： 1 ： 1 ; 105 cfu/g） , and Group 2 were fed the basal diet supplemented with B. subtilis only （105efu/g）. The results indi- cated that compared with the control group, water nitrite nitrogen level in Group 1 and Group 2 significantly de- creased from the 21^st day to the end of the experiment （except on the 35th day for group 1, P 〈0.01 ） ; and ammo- nium nitrogen level in group 2 also decreased significantly on the 21^st and 35^th day. No significant differences were found for water pH and nitrate nitrogen level among the three groups. Supplementation of Bacillus also decreased the death rate （ P 〈 0.05 ） and improved the growth performance of grass carps. Compared with the control group, WGR of Group 1 and Group 2 increased by 52.93% （P 〈0.01 ） and 21.78% （P 〈0.05）, and ADFI increased by 44.44% （P 〈0.01 ） and 16.67% （P 〈0.05）, respectively; and compared with Group 2, supplementation of complex Bacillus increased both WGR and ADFI of grass carp significantly （25.58% and 23.81% , respectively, P 〈 0.01 ）. Further study showed that supplementation of Bacillus improved partial antioxidant capacity of intestinal mucosa of grass carp, as was evidenced by increased activities of superoxide dismutase （SOD） and catalase （CAT）. it can be concluded that, Bacillus supplementation in basal diets can improve water quality and antioxi- dant capacity of intestinal mucosa of grass carp, and further improve the growth performance and decrease the death rate of grass carp.
