Abstract:A set of chitosan/ε-polylysine (ε-PL) bionanocomposite films were prepared by a simple in situ self-assembly technique using sodium tripolyphosphate (TPP) as cross-linking agent. The physical, mechanical, structural, and antimicrobial properties of these films were investigated. Fourier infrared spectroscopy and X-ray diffraction showed that the introduction of TPP promoted the formation of hydrogen bonds and electrostatic interactions among functional groups of chitosan or ε-PL, which improved the tensile strength and decreased the water solubility, water vapor permeability and surface wettability of films. On the other hand, the incorporation of ε-PL weakened the bionanocomposite film' structure and integrity, resulting in a decrease trend of films' mechanical and barrier properties. More importantly, the bionanocomposite films exhibited excellent antimicrobial efficacy against E. coli and S. aureus by the increasing ratio of ε-PL. And ε-PL presented a sustained release from the films, which was closely related to TPP concentration. Results of this study suggested that chitosan/ε-PL films could be used as antimicrobial bio-material and have great potential in food industry.

Antimicrobial Activity of Chinese Bayberry Extract for the Preservation of Surimi

Abstract:BACKGROUND: Chemical preservatives such as sodium nitrite and potassium sorbate have been widely used to keep surimi products fresh. However, the potential harmfulness to human health cannot be ignored. This study was conducted to develop natural preservatives for the storage of Collichthys surimi. RESULTS: Among the eight Chinese traditional herbs and fruits, Chinese bayberry extract showed the greatest inhibitory effect against surimi spoilage bacteria Serratia marcescens and Pseudomonas aeruginosa. Moreover, N-butanol phase extract of bayberry (NB) showed the greatest activity among the different phases of bayberry extract. When Chinese bayberry extract was combined with tea polyphenol, an additive inhibitory effect was observed on growth of Hansenula anomala, Micrococcus luteus, Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. Our results further indicated that the shelf life of surimi products stored at room temperature can be extended when supplemented with Chinese bayberry extract. CONCLUSION: Our results suggest that Chinese bayberry extract can be used as a natural preservative for the storage of Collichthys surimi. Copyright (C) 2012 Society of Chemical Industry

Abstract:The effects of 10-MeV E-beam (0, 1, 3, 5, and 7 kGy) irradiation and vacuum packaging on extending the shelf-life of grass carp surimi stored at 4 °C were evaluated basing on the total viable counts (TVC), physiochemical 2-thiobarbituric acid reactive substance (TBARS), total base nitrogen (TVB-N), biogenic amines (BAs), texture (TPA) and color, and sensory changes in surimi samples. The results showed that comparing the control samples, the TVC and TVB-N content in surimi were significantly (p < 0.05) decreased by irradiation with different doses. Irradiation significantly (p < 0.05) inhibited the increase of putrescine (PUT), cadaverine (CAD), histamine (HIM), and tyramine (TYM) contents during storage. However, these parameters were significantly (p < 0.05) increased with storage time. After irradiation, the samples generally had higher lightness and lower a * and b * values and lower hardness and chewiness significantly (p < 0.05). Based on the sensory analysis, unfavorable ‘metal odor’ or ‘irradiated odor’ was observed in surimi irradiated at 5 and 7 kGy.
Physicochemical and antimicrobial properties of ε-polylysine/carboxymethyl chitosan polyelectrolyte complexes and their effect against spoilage microorganisms in raw pork

Abstract:This study investigated the properties of polyelectrolyte complexes (PECs) fabricated by using ε-polylysine (EPL) and N,O-carboxymethyl chitosan (NOCC) and their potential applications in raw pork. The phase behaviors of PECs in aqueous solutions are characterized by the ζ-potential, mean radius and turbidimetric measurements. Stable colloidal and soluble PEC systems can be fabricated by carefully adjusting the mass ratio of NOCC to EPL. The minimum inhibitory concentration (MIC) assay showed that the colloidal and soluble PECs could maintain the antimicrobial activity of EPL in a laboratory medium. The antimicrobial effectiveness of coatings incorporating three PECs against spoilage microorganisms of raw pork under refrigerated conditions (4 °C) was evaluated. Microbial analysis demonstrated that the bacterial counts were significantly (P < 0.05) lower in sprayed samples with tested PECs containing 0.1% (v/v) EPL, especially for PECs with the mass ratio (NOCC/EPL) at 3 than that of the control sample throughout the storage period, while there were not significantly different counts (P > 0.05) among all samples in the case of molds and yeasts.
Application of Ε-Polylysine to Keep Chilled Pork Fresh

Abstract:e-Polylysine(e-PL)treatment was used to keep chilled pork fresh,and the change of total viable count(TVC)in fresh pork treated under different conditions ofe-PL concentration,accessory reagent type and soaking time during storage at 4℃in order to optimize the three treatment conditions.Along with this,the effect of optimizede-PL treatment on the sensory,microbiological and physiochemical properties of chilled pork were investigated.Treatment with 400 mg/Le-PL alone markedly inhibited the decline in sensory quality,the growth of microorganisms,the increase of pH and the accumulation of total volatile base nitrogen(TVB-N).Better fresh-keeping effect was observed whene-PL and acetic acid were used together,especially more significant inhibitory effect on microbial growth.The combined antimicrobial effects ofe-PL and acetic acid were largely affected by soaking time in a positive way.Collectively considering antimicrobial effect as well as treatment cost and efficiency,the optimale-PL concentration,accessory reagent type and soaking time were 400 mg/L,02%acetic acid and 5 min.The optimized e-PL treatment increased the shelf life of chilled pork from 2 to 8 d.These results demonstrate thate-PL will have extensive application prospects as a biological fresh-keeping agent for chilled pork.
Antimicrobial and Antibiofilm Effect of ε-Polylysine against Salmonella Enteritidis, Listeria monocytogenes, and Escherichia coli in Tryptic Soy Broth and Chicken Juice

Abstract:ε-Polylysine (ε-PL) is a safe food additive that is used in the food industry globally. This study evaluated the antimicrobial and antibiofilm activity of antibacterial peptides (ε-PL) against food poisoning pathogens detected in chicken (Salmonella Enteritidis, Listeria monocytogenes, and Escherichia coli). The results showed that minimum inhibitory concentrations (MICs) ranged between 0.031–1.0 mg/mL, although most bacterial groups (75%) showed MICs of 1.0 mg/mL. The reduction in the cell viability of pathogens due to ε-PL depended on the time and concentration, and 1/2 × MIC of ε-PL killed 99.99% of pathogens after 10 h of incubation. To confirm biofilm inhibition and degradation effects, crystal violet assay and confocal laser scanning microscopy (CLSM) were used. The biofilm formation rates of four bacterial groups (Salmonella, Listeria, E. coli, and multi-species bacteria) were 10.36%, 9.10%, 17.44%, and 21.37% at 1/2 × MIC of ε-PL, respectively. Additionally, when observed under a CLSM, ε-PL was found to induce biofilm destruction and bacterial cytotoxicity. These results demonstrated that ε-PL has the potential to be used as an antibiotic and antibiofilm material for chicken meat processing.

Abstract:To explore a novel efficient method in food preservation, edible zein coating which incorporates a polymeric chelator was developed and its preservative effect on commercially manufactured fish balls was investigated. The quality indicators of the fish ball during the storage, including sensory quality, total bacterial count, total basic volatile nitrogen (TVB-N) content, thiobarbituric acid reactive substances (TBARS), and pH value were determined. It has been demonstrated that a zein film containing a polymeric chelator significantly (P < 0.05) improves the sensory properties, retards the growth of spoilage bacteria, and decreases TVB-N and TBARS values. The increase of the microbial load of fish balls coated with antimicrobial zein during a 16-day refrigeration storage period was less than 1 log cfu/g, while the microbial load increased to about 5 log cfu/g for the control group lacking the coating treatment. Although statistically significant differences in the pH values were not observed between coated and uncoated fish balls (P > 0.05), fish balls coated with antimicrobial zein maintained a relatively stable state during storage. Zein coating which incorporates a polymeric chelator extends fish ball shelf life to 19 days at 4 °C, being more efficient than zein coating lacking a polymeric chelator (13 days). Clearly, polymeric chelator-incorporated zein film could find application in the preservation of surimi products by preventing the growth of bacteria and oxidation of fat during storage.

Abstract:It is well known that aquatic products such as fish and shellfish, when stored for a long period of time under inappropriate conditions, can suffer from muscle softening. This phenomenon is mainly caused by endogenous proteases, which are activated during heating and accelerates the degradation of myofibrillar proteins, directly leading to weaker gels and poorer water retention capacity. This paper reviews the changes in fish proteins during storage after death and the factors affecting protein hydrolysis. A brief overview of the extraction of protease inhibitors, polysaccharides and proteins is given, as well as their mechanism of inhibition of protein hydrolysis in surimi and the current status of their application to improve the properties of surimi.

Abstract:The quality of surimi, widely used in processed seafood, is compromised by freeze-thaw cycles, leading to protein denaturation and oxidative degradation. The objective of this study is to explore the effects of adding natural whey peptide hydrolysate (WPH) on the myofibrillar proteins of repeatedly freeze-thawed surimi. Results indicated surimi treated with 15% WPH exhibited only a 128% increase in surface hydrophobicity and a maximum peroxide value of 7.84 μg/kg, significantly lower than the control group. Additionally, salt-soluble protein content, emulsification activity, and stability decreased with the increase in freeze-thaw cycles. With a 15% WPH offering the most significant protective effect, evidenced by reductions of only 25.02%, 42.52% and 37.02% in salt-soluble protein content, emulsification activity, and stability, respectively. These outcomes demonstrate that WPH effectively reduces protein denaturation during repeated freeze-thaw processes. Future research should explore the molecular mechanisms underlying WPH's protective effects and evaluate their applicability in other food systems.

Abstract:Surimi processing industry generates huge quantities of underutilized surimi processing by-products (SPB). This study aimed to investigate the potential antioxidant-cryoprotective and gelation-enhancing effects of SPB hydrolysates on silver carp (Hypophthalmichthys molitrix) surimi based on protein oxidation, protein degradation, and gel-forming ability analysis. Results showed that untreated surimi was particularly susceptible to frozen-induced protein degradation and oxidation as well as rapid deterioration in gelation properties. Compared with 4% sucrose-added surimi, partial replacement of sucrose with 2% trypsin- and alcalase-treated SPB hydrolysates effectively delayed the oxidation of cysteine, carbonylation of amino acids, loss of Ca2+-ATPase activity, and the destroy of structural integrity of myofibrillar protein in surimi. Moreover, addition of SPB hydrolysates significantly (P < 0.05) increased the initial gelation properties and reduced the loss in gelation and water-holding capacity in surimi with prolonged frozen storage. Therefore, SPB hydrolysates could be used as natural antioxidant-cryoprotectant and gel texture enhancer in freshwater fish surimi.
Preparation of Coaxial Polylactic Acid-Propyl Gallate Electrospun Fibers and the Effect of Their Coating on Salmon Slices during Chilled Storage

Abstract:Pseudomonas fluorescens bacteria can grow well in cold-storage conditions and cause food spoilage. Quorum sensing (QS) is a biological pathway existing in a large number of microorganisms, through which bacteria regulate several of their physiological activities. A number of substances have been identified as quorum sensing inhibitors (QSIs); they can interfere with the QS system and control bacterial spoilage characteristics and production of virulence factors. In our previous study, propyl gallate at sub-minimum inhibitory concentration levels showed a potent anti-QS activity. Thus, in this study, coaxial polylactic acid-propyl gallate electrospun fibers were fabricated and their physicochemical properties were characterized. Salmon slices were coated with these electrospun fibers and the effect of this coating on the salmon slices during chilled storage was evaluated. The results showed that the electrospun fibers had a small diameter and smooth surface with no beads or other defects. The thermal stability, tensile strength, and other properties of the fibers were suitable for refrigerated storage conditions. Without inhibiting the bacterial growth in the salmon slices, the QSI-containing electrospun fibers exerted a significant inhibitory effect on the production of total volatile base nitrogen and trimethylamine. Furthermore, the deterioration of muscle tissue in the salmon slices was significantly delayed during cold storage. Quantitative analysis indicated that the electrospun fibers had a significant inhibitory effect on the bacterial spoilage ability. The results suggested that the electrospun fibers loaded with QSIs might be an effective strategy to control food spoilage and enhance the quality of aquatic food products.

Abstract:CYP 2C8, which carries out the oxidative metabolism of at least 5% of clinical drugs, has attracted increasing attention in recent years. New drugs (substances), inducers and inhibitors of CYP 2C8 have been developed and the drug metabolism has been investigated to understand the clinical role of CYP2C8. The cases of CYP2C8 genetic polymorphisms linked to diseases have increased and have been investigated. Herein, important progress in these areas has been reviewed with an emphasis on drug metabolism. Polymorphisms, diseases related to CYP2C8, some important drugs (substances) and inhibitors are reviewed and discussed.
Epsilon-poly-l-lysine alleviates brown blotch disease of postharvest Agaricus bisporus mushrooms by directly inhibiting Pseudomonas tolaasii and inducing mushroom disease resistance

Abstract:The natural antimicrobial peptide, epsilon-poly-l-lysine (ε-PL), is widely acknowledged as a food preservative. However, its potential in managing bacterial brown blotch disease in postharvest edible mushrooms and the associated mechanism remain unexplored. In this study, concentrations of ε-PL ≥ 150 mg L-1 demonstrated significant inhibition effects, restraining over 80% of growth and killed over 99% of Pseudomonas tolaasii (P. tolaasii). This inhibition effect occurred in a concentration-dependent manner. The in vivo findings revealed that treatment with 150 mg L-1 ε-PL effectively inhibited P. tolaasii-caused brown blotch disease in Agaricus bisporus (A. bisporus) mushrooms. Plausible mechanisms underlying ε-PL's action against P. tolaasii in A. bisporus involve: (i) damaging the cell morphology and membrane integrity, and increasing uptake of propidium iodide and leakage of cellular components of P. tolaasii; (ii) interaction with intracellular proteins and DNA of P. tolaasii; (iii) inhibition of P. tolaasii-induced activation of polyphenol oxidase, elevation of antioxidative enzyme activities, stimulation of phenylpropanoid biosynthetic enzyme activities and metabolite production, and augmentation of pathogenesis-related protein contents in A. bisporus mushrooms. These findings suggest promising prospects for the application of ε-PL in controlling bacterial brown blotch disease in A. bisporus.
Postharvest quality maintenance of wax apple and guava fruits by use of a fermented broth of an ε-poly-l-lysine-producing Streptomyces strain

Abstract:ε-Poly- l -lysine (ε-PL) is a natural antimicrobial polymer with significant inhibitory activity against a broad spectrum of microorganisms, and nowadays used widely as a preservative in the food industry. In the present study, ε-PL broth was obtained from Streptomyces ahygroscopicus GIM8 fermentation in a nutrient-limited liquid medium. The in vitro antifungal activity of the broth against fruit pathogens Penicillium expansum and Colletotrichum gloeosporioides was investigated, and its usage for postharvest storage of two highly perishable fruits wax apple and guava was evaluated. Results showed that ε-PL concentration in the broth reached 0.61 g/L, and the nutrition level of the broth was low. The antifungal activity of ε-PL broth was comparable to that of the aqueous solution of ε-PL under the same concentration. Immersion with the diluted broth (200 mg/L ε-PL) markedly delayed the decline in the quality of postharvest wax apple and guava fruits during storage, and the decay incidences were also greatly decreased as compared to their respective controls (distilled water immersion). A further investigation demonstrated that the ε-PL broth immersion induced an increase in the activity of defense-related enzymes peroxidase and polyphenol oxidase in the two fruits during storage. The present study proved that the fermentation broth of ε-PL could be used as a promising alternative to high purity ε-PL and synthetic fungicides for preserving fruits at postharvest stage.

Comparative study of a liposome and emulsion system with cinnamon essential oil on the quality and proteolysis of refrigerated minced pork

Abstract:Essential oils have been recognized for their strong antibacterial property, making them an innovative approach for preserving meat. However, their chemical instability and direct impact on meat proteins limit their application. To overcome these limitations, various loading systems have been explored. This study aimed to compare the effect of cinnamon essential oil (CEO) loaded in a liposome and emulsion system on the proteolysis of minced pork and to evaluate the advantages of each delivery system in preventing microorganism-induced quality deterioration of meat. Minced pork treated with CEO-liposomes exhibited lower pH, total volatile basic nitrogen (TVB-N), and total viable count (TVC) values than CEO-emulsions and provided better protection against microorganisms. SDS-polyacrylamide gel electrophoresis (PAGE) analysis confirmed that CEO-liposome was more effective in protecting proteins from degradation. Moreover, CEO-liposome produced lower amount of bitter amino acids and harmful biogenic amines. Antibacterial mechanisms indicated that CEO-liposome exhibited a stronger inhibitory effect against major spoilage bacteria in meat products by increasing cell membrane permeability. The membrane damage was further supported by an increase in conductivity and the leakage of nucleic acids. Compared to the CEO-emulsion system, CEO-liposome emerged as an effective preservative for minced pork. These results provided important theoretical support for using a bioactive compound delivery system to prevent microorganism-induced quality deterioration in meat.

Abstract:Natural antibacterial agents with antimicrobial properties have a broad potential to prevent bacterial from forming biofilms adhesion in marine environments. ε-Polylysine (E-PL) consist of homomeric polymer with 25–30 lysine residues with stability, nontoxicity, and biodegradability. ε-Polylysine is a natural cationic antibacterial peptide that can resist microbial forming biofilm adhesion. The current study investigated the action of E-PL against Pseudomonas aeruginosa biofilm isolated from a marine environment. Crystal violet staining was used to examine the effects of E-PL on the formation and destruction of mature biofilms. Scanning Electron and fluorescence microscopy revealed that E-PL treatment damaged the biofilm structure and affected the secretion of extracellular polymers. The CCK8 colorimetric assay showed that E-PL also decreased the metabolic activity and motility of biofilm bacteria. QPCR and transcriptome analysis revealed that E-PL affected biofilm formation and transcriptional regulation by downregulating genes involved in flagellar synthesis (flgE, PA4651, pilW), chemotaxis transduction (PA1251, PA4951, PA4788), biofilm biosynthesis (pelC, pelD, pslK, plsM), transcriptional regulation (PA3973, PA3508, PA0268), phenazine biosynthesis (phzM, phzH, phzS), and electron transfer (PA5401, PA5400, PA3492). This study used multiple methods to identify the mechanism of E-PL action against biofilm, informing the design of novel biofilm treatment methods.

Abstract:In this study, a novel bacteriocin Lactocin C-M2 produced by Lactobacillus panis C-M2, combined with dielectric barrier discharged cold plasma (DBD-CP), was used to evaluate the antibacterial effect on aquatic foods. After the purification procedures of ethyl acetate extraction, cation exchange chromatography and semi-preparative liquid phase, the stability of Lactocin C-M2 under DBD-CP environment was determined, and the preservation effect of these two joint treatments was investigated on fresh white fish samples. As revealed by LC-MS/MS and BLAST analysis, Lactocin C-M2 is a new type of class II bacteriocin, with a molecular weight of 863.52 Da and the N-terminal sequence MVKKTSAV. Application of Lactocin C-M2 showed significantly stronger inhibitory effect on bacteria than on yeasts and mold. All the tested 10 Gram positive bacteria and 3 Gram-negative bacteria, including Staphylococcus aureus, Shigella flexneri, Bacillus spp, Lactobacillus spp, Escherichia coli, Pseudomonas aeruginosa, and so on, were inhibited. Lactocin C-M2 also presented stable antibacterial activity after exposure to DBD-CP, with the 95% residual activity against Staphylococcus aureus under the 40∼80 kV voltage for 30∼180 s, indicating the possibility of synergistic application. Combined with addition of 0.9 mg/g Lactocin C-M2, the treatment of DBD-CP with voltage at 60 kV for 90 s on fresh white fish (Culter alburnus) could significantly inhibit the microbial growth, the accumulation of volatile nitrogen and histamine during the storage. Therefore, the Lactocin C-M2, used together with the DBD-CP, is effective in food preservation.

Abstract:Solid-fermentation is a promising way of producing novel surimi-based food. In the present study, changes in physicochemical characteristics, protein hydrolysis, and textual properties of surimi during the fermentation, including pehtze preparation, salting, and ripening stages, were studied. The protease produced by Actinomucor elegans led to continuous hydrolysis of surimi protein, reflected from the high value of protease activity which reached 69.34 U/g after pehtze preparation stage, the decrease of protein bands intensity in sodium dodecyl sulfate polyacrylamide gel electrophoresis image especially for bands of 200, 43, and 36 KDa, and the increasing contents of free amino acids and trichloroacetic acid-soluble nitrogen. Hardness, springiness, and cohesiveness of the surimi all decreased during pehtze preparation stage, increased after salting, and decreased again after ripening, while adhesiveness kept increasing until the ripening stage started. Scanning electron microscopy images showed a dense and soft microstructure of surimi after ripening. The final surimi product showed high overall acceptability and no Enterobacteriaceae was detected.
