Abstract:Oral administration of drug is the most preferred one among the other routes for the majority of clinical applications. As compared to the parenteral method of administration, it has potential benefits such as increased patient compliance, fewer problems, and reduced treatment costs. Regardless of these factors, inadequate bioavailability owing to poor solubility or permeability limits the therapeutic effectiveness of orally given drugs. Though most current research focuses on BCS II (drugs with low solubility and high permeability), BCS III (drugs with high solubility and low permeability) also has poor oral bioavailability due to their limited permeability across lipid membranes and is usually administered through the parenteral route. The need for an oral alternative to parenteral administration has prompted a renewed focus on the development of innovative dosage forms that support the absorption of medicines that are poorly permeable through the intestinal epithelium. Because of their unique sizedependent feature in enhancing transmembrane permeability, ability to incorporate both lipophilic and hydrophilic drugs and biocompatible nature of components, the use of nanoparticles for improving drug bioavailability has been a focus of current study in the field of drug delivery in recent years. The lipidbased nanoparticle method presents a potential new avenue for manufacturing BCS Class III medicines with enhanced bioavailability, as poor permeability is the main issue for these agents. This research aims to assess the potential of lipid nanoparticles for improving the oral bioavailability of medicines with permeability-restricted oral absorption, such as pharmaceuticals in Biopharmaceutical Classification System (BCS) class III.

Abstract:Lipid based drug delivery system such as Solid lipid nanoparticle (SLN) and Nanostructured lipid carriers (NLC) are among the most promising drug delivery system used in many industries such as food, pharmaceuticals and cosmetics industries. Over the last few years, new constituents of lipids have developed and investigated for enhancement of bioavailability. The present manuscript is an attempt on solving the concerned uncertainty with efficacious peroral administration of hydrophobic drugs through fabricating new lipid formulations, NLC. NLC, the second-generation lipid carrier is usually composed of solid lipids and liquid lipids together in a system. This mixing causes depression in melting point of substrates and converts the mixture into solid form at body temperature and termed as NLC. NLC shows a high drug loading with minimum drug expulsion. The unique advantages of NLC over SLN and Lipid-drug conjugates (LDC) are increased capacity of drug loading, avoidance of drug expulsion. This manuscript gives detailed information on definitions and simple way of production methods, new approaches in formulation of NLC and it also highlights how NLC improves bioavailability of bioactive molecules through peroral route and its future perspective as a pharmaceutical carrier. Abstract:Oral bioavailability of drugs is mainly limited due to the poor aqueous solubility, enhanced chemical degradation, reduced permeation and/or first pass metabolism. Various novel delivery systems are developed for improved oral bioavailability of these drugs such as modified orals, buccal, transdermal and osmotic delivery systems. Colloidal carrier systems such as nanoparticles, lipid nanoparticles, nanoemulsions, microspheres, liposomes, resealed erythrocytes and transfersomes were also developed to enhance the oral delivery. Among these, solid lipid nanocarriers (SLNs) also gain much attention on the enhancement of oral bioavailability. SLNs are submicron sized nanoparticles and composed of solid lipid, surfactants and cosurfactants. The enhanced oral bioavailability of poorly soluble drugs from SLNs might be due to the reduced particle size, bypassed presystemic metabolism, and enhanced gastric mucosa permeability. Vast literature is available for the advantages, limitations, preparation methods, evaluation parameters and application of SLNs in different routes. This review mainly focused on list of drugs developed as SLNs and considered as an alternative approach to enhance the oral bioavailability based on pharmacokinetic as well as pharmacodyanmic parameters was discussed. Abstract:Solid lipid nanoparticles (SLN) are the heart of nanotechnology's tremendous development, with multiple practical benefits in drug delivery and research. Because of their size-dependent properties, solid Lipid nanoparticles can be used to create innovative medicines. Drug conversion into nanoparticles provides a new drug delivery concept that could be used for drug targeting. As a result, solid lipid nanoparticles have great promise reaching the goal of targeted and site-specific drug delivery. The goals, production techniques, advantages, limits are all discussed in this review. Scanning electron microscopy and measurement of particle size and zeta potential, dynamic light scattering are examples of appropriate analytical techniques for SLN characterization. By using novel drug formulation technologies, the drug delivery system focuses on the regulation of in vivo dynamics in order to enhance the efficacy and safety of the included pharmaceuticals. Lipids used in the development of Nano particulate dosage forms, such as fatty acids, triglycerides, vegetable oils, and their derivatives. Solid lipid nanoparticles (SLNs) are a type of lipid drug delivery method that is relatively new. They were created to solve some of the limitations with traditional drug delivery systems, such as low drug encapsulation efficiency and limited bioavailability of Biopharmaceutical Classification Systems (BCS) class II and IV medications. SLNs are made up of physiologically well tolerated substances and melt-emulsified lipids that are solid at room temperature that helps to enhance drug bioavailability ultimately results in better desired therapeutic effect.

Abstract:Low-oral bioavailability as a consequence of low-water solubility of drugs is challenging for formulation scientists in the development of new pharmaceutical products. This review aims to highlight relevant considerations when implementing a rational strategy for the development of lipid-based oral drug delivery systems and to discuss shortcomings and challenges to the current classification of these delivery systems such as nanoemulsion, Solid lipid nanoparticle (SLN), Nanostructured lipid carriers (NLC), Self-emulsifying drug delivery system (SEDDS). Lipid-based drug delivery systems consist of a diverse group of formulations, each consisting of varying functional and structural properties that are amenable to modifications achieved by varying the composition of lipid excipients and other additives thereby facilitating the bioavailability of poorly water-soluble drugs. In addition, lipid nanoparticles may also protect the loaded drugs from chemical and enzymatic degradation and gradually release drug molecules from the lipid matrix into the blood, resulting in improved therapeutic profiles compared to free drugs. Therefore, due to their physiological and biodegradable properties, lipid molecules may decrease adverse side effects and chronic toxicity of the drug-delivery systems when compared to others of polymeric nature. Accordingly, the present review is mainly centred on the various lipid-based drug delivery system and excipients used in lipid-based drug delivery systems (LBDDS).

Abstract:Lipid nanoparticles based on solid matrix have emerged as potential drug carriers to improve gastrointestinal (GI) absorption and oral bioavailability of several drugs, especially lipophilic compounds. These formulations may also be used for sustained drug release. Solid lipid nanoparticle (SLN) and the newer generation lipid nanoparticle, nanostructured lipid carrier (NLC), have been studied for their capability as oral drug carriers. Biodegradable, biocompatible, and physiological lipids are generally used to prepare these nanoparticles. Hence, toxicity problems related with the polymeric nanoparticles can be minimized. Furthermore, stability of the formulations might increase than other liquid nano-carriers due to the solid matrix of these lipid nanoparticles. These nanoparticles can be produced by different formulation techniques. Scaling up of the production process from lab scale to industrial scale can be easily achieved. Reasonably high drug encapsulation efficiency of the nanoparticles was documented. Oral absorption and bioavailability of several drugs were improved after oral administration of the drug-loaded SLNs or NLCs. In this review, pros and cons, different formulation and characterization techniques, drug incorporation models, GI absorption and oral bioavailability enhancement mechanisms, stability and storage condition of the formulations, and recent advances in oral delivery of the lipid nanoparticles based on solid matrix will be discussed.

Abstract:Oral administration of dosage forms is convenient and beneficial in several respects. Lipid nanoparticulate dosage forms have emerged as a useful carrier system in deploying low solubility drugs systemically, particularly class II, III, and IV drugs of the Biopharmaceutics Classification System. Like other nanoparticulate delivery systems, their low size-to-volume ratio facilitates uptake by phagocytosis. Lipid nanoparticles also provide scope for high drug loading and extended-release capability, ensuring diminished systemic side effects and improved pharmacokinetics. However, rapid gastrointestinal (GI) clearance of particulate delivery systems impedes efficient uptake across the mucosa. Mucoadhesion of dosage forms to the GI mucosa results in longer transit times due to interactions between the former and mucus. Delayed transit times facilitate transfer of the dosage form across the mucosa. In this regard, a balance between mucoadhesion and mucopenetration guarantees optimal systemic transfer. Furthermore, the interplay between GI anatomy and physiology is key to ensuring efficient systemic uptake. This review captures salient anatomical and physiological features of the GI tract and how these can be exploited for maximal systemic delivery of lipid nanoparticles. Materials used to impart mucoadhesion and examples of successful mucoadhesive lipid nanoformulations are highlighted in this review.

Abstract:Solid lipid nanoparticle (SLN), nanostructured lipid carriers (NLC) and hybrid nanoparticles, have gained increasing interest as drug delivery systems because of their potential to load and release drugs from the Biopharmaceutical classification system (BCS) of class II (low solubility and high permeability) and of class IV (low solubility and low permeability). Lipid properties (e.g. high solubilizing potential, biocompatibility, biotolerability, biodegradability and distinct route of absorption) contribute for the improvement of the bioavailability of these drugs for a set of administration routes. Their interest continues to grow, as translated by the number of patents being field worldwide. This paper discusses the recent advances on the use of SLN, NLC and lipid-polymer hybrid nanoparticles for the loading of lipophilic, poorly water-soluble and poorly permeable drugs, being developed for oral, topical, parenteral and ocular administration, also discussing the industrial applications of these systems. A review of the patents filled between 2014 and 2017, concerning the original inventions of lipid nanocarriers, is also provided.

Abstract:A method to exact the electrical parameters and model the "second breakdown" action of MOSFET's under ESD (electrostatic discharge) on circuit-level, using TCAD simulation, is presented. MOSFET is one of the most important ESD protection devices, and is widely used as I/O protection device in integrated circuits. The avalanche breakdown of the MOSFET can be simulated by TCAD tools and the circuit-level model has been presented. However, accurate modeling and insightful analyzing of the "second breakdown" action, which leads to the permanent failure of the MOSFET, is rarely reported. We present an accurate macro model of the MOSFET based on deep analyzing of the physical mechanism of the "second breakdown", using TCAD simulation. This macro model owns fine convergency and accuracy which are of importance to the simulation of the ESD protection ability of the ESD protection network on circuit and system level.

Abstract:Drug delivery via the oral route has always been challenging for poorly soluble drugs. Acid-induced hydrolysis, enzymatic degradation, and poor mucosal absorbency remain the primary hiccups for effective oral delivery of medications. With the advent of nanotechnology, nanostructured lipid carriers (NLCs) have emerged as a promising delivery carrier that can circumvent gastrointestinal tract (GIT) barriers hindering the solubility and bioavailability of such drugs. These NLCs can efficiently transport drug moieties across intestinal membranes shielding medications from intestinal pH and enzymatic degradation. Because they are composed of lipidic materials, they can be easily absorbed or taken up by various pathways such as transcellular absorption, paracellular transport, and M-cell uptake. Such mechanisms not only improve the absorption and solubility of drugs but also augment bioavailability and residence time and may bypass first-pass metabolism. This review explores the diverse applications of nanostructured lipid carriers (NLCs) in oral drug delivery for various medical conditions, shedding light on their current regulatory status, including FDA-approved options and those in pre/clinical stages. The review also features patented NLC formulations. It provides valuable insights into how NLCs can be harnessed for effective oral drug delivery and outlines recent advancements in optimizing their performance to tackle gastrointestinal barriers, thus opening new possibilities for NLCs in future pharmaceutical applications.

Abstract:Solid lipid nanoparticles (SLNs) are one of the extensively utilized nanocarriers in the pharmaceutical field due to their biocompatibility and biodegradability. These features of the carrier system have fuelled its use as the drug delivery system since the last three decades. This review presents different SLN preparation techniques, such as high shear homogenization, hot homogenization, cold homogenization, microemulsion-based technique, etc. The physicochemical nature of SLNs, comprising drug loading, drug release, particle size, zeta potential, stability, cytotoxicity, and cellular uptake, has been concisely discussed. The article also explains why SLNs are preferred to develop drug delivery systems in several pharmaceutical preparations. The key ingredients like lipid, surfactant/ stabilizer accompanied by co-surfactant, cryoprotectant, or charge modifiers used to fabricate SLNs are also briefly conferred. Here is an elaborate discussion of drugs that are used through various routes by the SLN carrier system and their outcome for utilization of this system. Regulatory aspects, patent aspects, and future prospects of SLN are also discussed here.

Abstract:Numerous drugs have emerged to treat various diseases, such as COVID-19, cancer, and protect human health. Approximately 40% of them are lipophilic and are used for treating diseases through various delivery routes, including skin absorption, oral administration, and injection. However, as lipophilic drugs have a low solubility in the human body, drug delivery systems (DDSs) are being actively developed to increase drug bioavailability. Liposomes, micro-sponges, and polymer-based nanoparticles have been proposed as DDS carriers for lipophilic drugs. However, their instability, cytotoxicity, and lack of targeting ability limit their commercialization. Lipid nanoparticles (LNPs) have fewer side effects, excellent biocompatibility, and high physical stability. LNPs are considered efficient vehicles of lipophilic drugs owing to their lipid-based internal structure. In addition, recent LNP studies suggest that the bioavailability of LNP can be increased through surface modifications, such as PEGylation, chitosan, and surfactant protein coating. Thus, their combinations have an abundant utilization potential in the fields of DDSs for carrying lipophilic drugs. In this review, the functions and efficiencies of various types of LNPs and surface modifications developed to optimize lipophilic drug delivery are discussed.

Abstract:Solid lipid nanoparticles (SLNs) are primarily composed of solid lipids, which thus impart to them some of the fundamental properties of these lipids, including biocompatibility, biodegradability and low-toxicity. SLNs represent a unique class of colloidal drug delivery systems that possess the advantages of both the "soft" drug carriers such as emulsions and liposomes and polymeric nanoparticles. In this review, we will provide an overview on the absorption, disposition and pharmacokinetics of SLNs. The lipidic nature, as well as the relatively small particle size, of SLNs ensures sufficient affinity with the biomembranes, and results in improved absorption by either of the oral, transdermal, pulmonary, nasal, ocular, rectal or buccal route. One special aspect of oral SLNs is the enhanced lymphatic absorption by either the chylomicron-association pathway or the M cell uptaking pathway. Intravenous SLNs are predominantly uptaken by the liver or spleen following opsonization by the complementary system. Modification of SLN surface with PEGs chains will mask the hydrophobic surface and divert SLNs to non-hepatic and non-splenic organs, while ligand-modification will achieve active targeting to specific tissues or organs. Degradation of SLNs is primarily based on the degradation of the lipids themselves by lipase. Pharmacokinetics reflects the effect of the lipidic vehicles of SLNs on in vivo disposition of the loaded drugs.

Abstract:Lipid nanoparticles are prepared with solid fatty acids or esters, which is widely concerned in drug delivery due to their good biocompatiblity and safety. The oral bioavailability of poor-soluble drugs, peptides and proteins are very low because of their poor solubility, permeability or stability. However, lipid nanoparticles as their carrier can improve the oral bioavailability significantly, which enhance the application or investigation of lipid nanoparticles as oral drug delivery systems.
