Abstract:Solid lipid nanoparticles (SLNs) are one of the extensively utilized nanocarriers in the pharmaceutical field due to their biocompatibility and biodegradability. These features of the carrier system have fuelled its use as the drug delivery system since the last three decades. This review presents different SLN preparation techniques, such as high shear homogenization, hot homogenization, cold homogenization, microemulsion-based technique, etc. The physicochemical nature of SLNs, comprising drug loading, drug release, particle size, zeta potential, stability, cytotoxicity, and cellular uptake, has been concisely discussed. The article also explains why SLNs are preferred to develop drug delivery systems in several pharmaceutical preparations. The key ingredients like lipid, surfactant/ stabilizer accompanied by co-surfactant, cryoprotectant, or charge modifiers used to fabricate SLNs are also briefly conferred. Here is an elaborate discussion of drugs that are used through various routes by the SLN carrier system and their outcome for utilization of this system. Regulatory aspects, patent aspects, and future prospects of SLN are also discussed here.

Abstract:Solid lipid nanoparticles are at the cornerstone of the swiftly growing area of medical nanotechnology, having several potential functions in drug delivery, research, clinical care, and a variety of other fields. They provide the opportunity of developing novel therapies due to their unique properties, such as small particle size and being prepared from physiological biodegradable lipids. The loading of bioactive molecules into nanocarriers is a novel drug delivery prototype employed for various drug targeting levels. Hence, SLNs hold a great promise for achieving the aim of targeted and controlled drug delivery. For this reason, they have attracted the extensive attention of scientists and researchers. This review is based on recent studies and research, and here we present advantages, disadvantages, and preparation methods, several advanced modifications, targeting strategies, and recent applications of solid lipid nanocarriers in drug delivery systems.
Preventing type 2 diabetes: making the evidence work for migrant Indian populations.

Abstract:Type 2 diabetes mellitus is a chronic and debilitating disease characterized by an inability to adequately regulate blood glucose levels. The symptoms of type 2 diabetes are associated with a reduced quality of life in the short term, whilst in the longer term the disease may lead to serious complications such as cardiovascular disease, blindness, renal failure and amputation1. The prevalence of type 2 diabetes is currently estimated to be around 6 per cent by the International Diabetes Federation, and growing2 as such it is now thought to be the fifth leading cause of mortality globally3. Data from India and elsewhere have shown that 50 per cent or more of individuals with type 2 diabetes die of cardiovascular disease and that life expectancy is substantially reduced by up to 9 years in those diagnosed at age 40 yr or more, with greater reductions in life expectancy occurring in those diagnosed at a younger age4,5. The enormous public health burden of type 2 diabetes and its associated co-morbidities are a relatively recent phenomenon; indeed type 2 diabetes was a relatively rare condition until the latter half of the 20th century. This rise was closely mirrored by a dramatic increase in industrialized environments where energy dense foods are plentiful and the link between physical activity and food procurement has been broken. Over the past few decades India has provided a good case study of this phenomenon with the rapid expansion of, and migration to, urban centres. India now heads the international league table for the highest number of subjects with type 2 diabetes, a position it is expected to maintain into the future with 80 million individuals estimated to have type 2 diabetes by 20302,6. The Indian genotype and/or culture seems to be particularly susceptible to type 2 diabetes when exposed to industrialized environments7,8. For example, the prevalence of type 2 diabetes in urban areas has been shown to be up to 5 times higher than rural settings9. Data from the Indian Diaspora also tell a revealing story. In the United Kingdom (UK), where Indian migrants and their families now form the largest ethnic minority group, the prevalence of type 2 diabetes has been estimated to be 4 6 times higher than that in the general population10. The reasons for this are incompletely understood, despite various hypotheses10. However, clearly lifestyle factors related to migration are a major causal factor. For example, data from industrialized countries have shown that migrant south Asian communities undertake around half the amount of daily physical activity and have higher levels of central obesity compared to the host white European population10,11. Given these startling figures, strategies and policies aimed at preventing type 2 diabetes in migrant Indian populations are essential and constitute a growing public health priority for India and other countries with large Indian populations.

Abstract:Over the past decade, there has been a lot of research on solid lipid nanoparticles (SLNs), also known as lipid carriers. As a new lipid-based drug delivery technology for the topical applications of numerous medicinal drugs, solid lipid nanoparticles (SLNs) have demonstrated promise. SLNs have undergone considerable research on every continent and have become effective nano-sized medication carriers. There is no doubt that nano-formulations are extremely valuable tools for drug delivery applications; the current challenge is how to optimize them to ensure that they are safe, effective and scalable, so that they can be manufactured at an industrial level and advance to clinical use. In this context, lipid nanoparticles have gained ground, since they are generally regarded as non-toxic, biocompatible and easy-to-produce formulations. However, the processes governing SLN cellular uptake and absorption by topical application, as well as the mechanism of SLN drug release, are still unclear and need further research. To make SLNs easier to apply and to improve cutaneous and transdermal administration, it is also crucial to choose the right dose form and formulation base. Additional difficulties that could prevent the clinical translation of SLNs include scaling up and regulatory authorization. Therefore, the main focus of this research is on the various pathways that SLNs use to enter cells and penetrate skin. The physicochemical characteristics of SLNs are then thoroughly discussed, including numerous formulation and dosage form parameters that may affect SLN absorption via the skin. The translational status in relation to scale-up and regulatory issues are also covered in the discussion. Researchers interested in topical uses of SLNs for the effective delivery of medications and cosmetics will find this review to be helpful.

Abstract:Solid lipid nanoparticles (SLN) are the heart of nanotechnology's tremendous development, with multiple practical benefits in drug delivery and research. Because of their size-dependent properties, solid Lipid nanoparticles can be used to create innovative medicines. Drug conversion into nanoparticles provides a new drug delivery concept that could be used for drug targeting. As a result, solid lipid nanoparticles have great promise reaching the goal of targeted and site-specific drug delivery. The goals, production techniques, advantages, limits are all discussed in this review. Scanning electron microscopy and measurement of particle size and zeta potential, dynamic light scattering are examples of appropriate analytical techniques for SLN characterization. By using novel drug formulation technologies, the drug delivery system focuses on the regulation of in vivo dynamics in order to enhance the efficacy and safety of the included pharmaceuticals. Lipids used in the development of Nano particulate dosage forms, such as fatty acids, triglycerides, vegetable oils, and their derivatives. Solid lipid nanoparticles (SLNs) are a type of lipid drug delivery method that is relatively new. They were created to solve some of the limitations with traditional drug delivery systems, such as low drug encapsulation efficiency and limited bioavailability of Biopharmaceutical Classification Systems (BCS) class II and IV medications. SLNs are made up of physiologically well tolerated substances and melt-emulsified lipids that are solid at room temperature that helps to enhance drug bioavailability ultimately results in better desired therapeutic effect.

Abstract:Nano pharmacology is considered an effective, safe, and applicable approach for drug delivery applications. Solid lipid nanoparticle (SLNs) colloids contain biocompatible lipids which are capable of encapsulating and maintaining hydrophilic or hydrophobic drugs in the solid matrix followed by releasing the drug in a sustained manner in the target site. SLNs have more promising potential than other drug delivery systems for various purposes. Nowadays, the SLNs are used as a carrier for antibiotics, chemotherapeutic drugs, nucleic acids, herbal compounds, etc. The SLNs have been widely applied in biomedicine because of their non-toxicity, biocompatibility, and simple production procedures. In this review, the complications related to the optimization, preparation process, routes of transplantation, uptake and delivery system, and release of the loaded drug along with the advantages of SLNs as therapeutic agents were discussed.

Abstract:Oral bioavailability of drugs is mainly limited due to the poor aqueous solubility, enhanced chemical degradation, reduced permeation and/or first pass metabolism. Various novel delivery systems are developed for improved oral bioavailability of these drugs such as modified orals, buccal, transdermal and osmotic delivery systems. Colloidal carrier systems such as nanoparticles, lipid nanoparticles, nanoemulsions, microspheres, liposomes, resealed erythrocytes and transfersomes were also developed to enhance the oral delivery. Among these, solid lipid nanocarriers (SLNs) also gain much attention on the enhancement of oral bioavailability. SLNs are submicron sized nanoparticles and composed of solid lipid, surfactants and cosurfactants. The enhanced oral bioavailability of poorly soluble drugs from SLNs might be due to the reduced particle size, bypassed presystemic metabolism, and enhanced gastric mucosa permeability. Vast literature is available for the advantages, limitations, preparation methods, evaluation parameters and application of SLNs in different routes. This review mainly focused on list of drugs developed as SLNs and considered as an alternative approach to enhance the oral bioavailability based on pharmacokinetic as well as pharmacodyanmic parameters was discussed. Abstract:Solid lipid nanoparticles (SLNs) are primarily composed of solid lipids, which thus impart to them some of the fundamental properties of these lipids, including biocompatibility, biodegradability and low-toxicity. SLNs represent a unique class of colloidal drug delivery systems that possess the advantages of both the "soft" drug carriers such as emulsions and liposomes and polymeric nanoparticles. In this review, we will provide an overview on the absorption, disposition and pharmacokinetics of SLNs. The lipidic nature, as well as the relatively small particle size, of SLNs ensures sufficient affinity with the biomembranes, and results in improved absorption by either of the oral, transdermal, pulmonary, nasal, ocular, rectal or buccal route. One special aspect of oral SLNs is the enhanced lymphatic absorption by either the chylomicron-association pathway or the M cell uptaking pathway. Intravenous SLNs are predominantly uptaken by the liver or spleen following opsonization by the complementary system. Modification of SLN surface with PEGs chains will mask the hydrophobic surface and divert SLNs to non-hepatic and non-splenic organs, while ligand-modification will achieve active targeting to specific tissues or organs. Degradation of SLNs is primarily based on the degradation of the lipids themselves by lipase. Pharmacokinetics reflects the effect of the lipidic vehicles of SLNs on in vivo disposition of the loaded drugs.

Abstract:Lipid nanoparticles based on solid matrix have emerged as potential drug carriers to improve gastrointestinal (GI) absorption and oral bioavailability of several drugs, especially lipophilic compounds. These formulations may also be used for sustained drug release. Solid lipid nanoparticle (SLN) and the newer generation lipid nanoparticle, nanostructured lipid carrier (NLC), have been studied for their capability as oral drug carriers. Biodegradable, biocompatible, and physiological lipids are generally used to prepare these nanoparticles. Hence, toxicity problems related with the polymeric nanoparticles can be minimized. Furthermore, stability of the formulations might increase than other liquid nano-carriers due to the solid matrix of these lipid nanoparticles. These nanoparticles can be produced by different formulation techniques. Scaling up of the production process from lab scale to industrial scale can be easily achieved. Reasonably high drug encapsulation efficiency of the nanoparticles was documented. Oral absorption and bioavailability of several drugs were improved after oral administration of the drug-loaded SLNs or NLCs. In this review, pros and cons, different formulation and characterization techniques, drug incorporation models, GI absorption and oral bioavailability enhancement mechanisms, stability and storage condition of the formulations, and recent advances in oral delivery of the lipid nanoparticles based on solid matrix will be discussed.

Abstract:Solid lipid nanoparticles (SLNs) are an alternate carrier system to liposomes, polymeric nanoparticles, and inorganic carriers. SLNs have attracted increasing attention in recent years for delivering drugs, nucleic acids, proteins, peptides, nutraceuticals, and cosmetics. These nanocarriers have attracted industrial attention due to their ease of preparation, physicochemical stability, and scalability. These characteristics make SLNs attractive for manufacture on a large scale. Currently, several products with SLNs are in clinical trials, and there is a high possibility that SLN carriers will quickly increase their presence in the market. A large-scale manufacturing unit is required for commercial applications to prepare enough formulations for clinical studies. Furthermore, continuous processing is becoming more popular in the pharmaceutical sector to reduce product batch-to-batch differences. This review paper discusses some conventional methods and the rationale for large-scale production. It further covers recent progress in scale-up methods for the synthesis of SLNs, including high-pressure homogenization (HPH), hot melt extrusion coupled with HPH, microchannels, nanoprecipitation using static mixers, and microemulsion-based methods. These scale-up technologies enable the possibility of commercialization of SLNs. Furthermore, ongoing studies indicate that these technologies will eventually reach the pharmaceutical market.

Abstract:Many pharmaceutically active molecules are highly lipophilic, which renders their administration and adsorption in patients extremely challenging. Among the countless strategies to overcome this problem, synthetic nanocarriers have demonstrated superb efficiency as drug delivery systems, since encapsulation can effectively prevent a molecules' degradation, thus ensuring increased biodistribution. However, metallic and polymeric nanoparticles have been frequently associated with possible cytotoxic side effects. Solid lipid nanoparticles (SLN) and nanostructured lipid carriers (NLC), which are prepared with physiologically inert lipids, therefore emerged as an ideal strategy to bypass toxicities issues and avoid the use of organic solvents in their formulations. Different approaches to preparation, using only moderate amounts of external energy to facilitate a homogeneous formation, have been proposed. Greener synthesis strategies have the potential to provide faster reactions, more efficient nucleation, better particle size distribution, lower polydispersities, and furnish products with higher solubility. Particularly microwave-assisted synthesis (MAS) and ultrasound-assisted synthesis (UAS) have been utilized in the manufacturing of nanocarrier systems. This narrative review addresses the chemical aspects of those synthesis strategies and their positive influence on the characteristics of SLNs and NLCs. Furthermore, we discuss the limitations and future challenges for the manufacturing processes of both types of nanoparticles.

Abstract:Various active compounds are easily damaged, so they need protection and must be easily absorbed and targeted. This problem can be overcome by encapsulating in the form of solid lipid nanoparticles (SLNs). Initially, SLNs were widely used to encapsulate hydrophobic (non-polar) active compounds because of their matched affinity and interactions. Currently, SLNs are being widely used for the encapsulation of hydrophilic (polar) and semipolar active compounds, but there are challenges, including increasing their entrapment efficiency. This review provides information on current research on SLNs for encapsulation and delivery systems for active and antioxidant compounds, which includes various synthesis methods and applications of SLNs in various fields of utilization. SLNs can be developed starting from the selection of solid lipid matrices, emulsifiers/surfactants, types of active compounds or antioxidants, synthesis methods, and their applications or utilization. The type of lipid used determines crystal formation, control of active compound release, and encapsulation efficiency. Various methods can be used in the SLN fabrication of active compounds and hydrophilic/hydrophobic antioxidants, which have advantages and disadvantages. Fabrication design, which includes the selection of lipid matrices, surfactants, and fabrication methods, determines the characteristics of SLNs. High-shear homogenization combined with ultrasonication is the recommended method and has been widely used because of the ease of preparation and good results. Appropriate fabrication design can produce SLNs with stable active compounds and antioxidants that become suitable encapsulation systems for various applications or uses.

Abstract:During the recent years, more attentions have been focused on lipid base drug delivery system to overcome some limitations of conventional formulations. Among these delivery systems solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs) are promising delivery systems due to the ease of manufacturing processes, scale up capability, biocompatibility, and also biodegradability of formulation constituents and many other advantages which could be related to specific route of administration or nature of the materials are to be loaded to these delivery systems. The aim of this article is to review the advantages and limitations of these delivery systems based on the route of administration and to emphasis the effectiveness of such formulations.

Abstract:Solid lipid nanoparticle (SLN), nanostructured lipid carriers (NLC) and hybrid nanoparticles, have gained increasing interest as drug delivery systems because of their potential to load and release drugs from the Biopharmaceutical classification system (BCS) of class II (low solubility and high permeability) and of class IV (low solubility and low permeability). Lipid properties (e.g. high solubilizing potential, biocompatibility, biotolerability, biodegradability and distinct route of absorption) contribute for the improvement of the bioavailability of these drugs for a set of administration routes. Their interest continues to grow, as translated by the number of patents being field worldwide. This paper discusses the recent advances on the use of SLN, NLC and lipid-polymer hybrid nanoparticles for the loading of lipophilic, poorly water-soluble and poorly permeable drugs, being developed for oral, topical, parenteral and ocular administration, also discussing the industrial applications of these systems. A review of the patents filled between 2014 and 2017, concerning the original inventions of lipid nanocarriers, is also provided.

Abstract:Solid Lipid Nanoparticles (SLNs) have gained significant attention in recent years as a promising delivery system for drugs targeting the Central Nervous System (CNS) via the Nose-To-Brain (NTB) route. The unique characteristics of SLNs, such as their small particle size, high stability, and ability to encapsulate lipophilic drugs, make them suitable for crossing the Blood Brain Barrier (BBB) and achieving targeted delivery to the brain. This has led to the development of SLNs-based formulations of drugs for neurological disorders such as Alzheimer's disease and Parkinson's disease, which are being evaluated in preclinical and clinical studies. Overall, the recent advances in SLN technology have improved these nanoparticles' stability, drug loading capacity and BBB crossing ability, making them a promising delivery system for NTB drug delivery. SLNs are composed of a solid lipid core surrounded by a surfactant coating, which allows for the encapsulation of hydrophilic and hydrophobic drugs. Additionally, we will also highlight the current challenges and future perspectives of using SLNs for NTB delivery of CNS therapeutics. Overall, this review aims to provide a comprehensive overview of the current state of the art in using SLNs for NTB delivery and to encourage further research in this field.

Abstract:Despite all the advances seen in recent years, the severe adverse effects and low specificity of conventional chemotherapy are still challenging problems regarding cancer treatment. Nanotechnology has helped to address these questions, making important contributions in the oncological field. The use of nanoparticles has allowed the improvement of the therapeutic index of several conventional drugs and facilitates the tumoral accumulation and intracellular delivery of complex biomolecules, such as genetic material. Among the wide range of nanotechnology-based drug delivery systems (nanoDDS), solid lipid nanoparticles (SLNs) have emerged as promising systems for delivering different types of cargo. Their solid lipid core, at room and body temperature, provides SLNs with higher stability than other formulations. Moreover, SLNs offer other important features, namely the possibility to perform active targeting, sustained and controlled release, and multifunctional therapy. Furthermore, with the possibility to use biocompatible and physiologic materials and easy scale-up and low-cost production methods, SLNs meet the principal requirements of an ideal nanoDDS. The present work aims to summarize the main aspects related to SLNs, including composition, production methods, and administration routes, as well as to show the most recent studies about the use of SLNs for cancer treatment.

Abstract:Lipid based drug delivery system such as Solid lipid nanoparticle (SLN) and Nanostructured lipid carriers (NLC) are among the most promising drug delivery system used in many industries such as food, pharmaceuticals and cosmetics industries. Over the last few years, new constituents of lipids have developed and investigated for enhancement of bioavailability. The present manuscript is an attempt on solving the concerned uncertainty with efficacious peroral administration of hydrophobic drugs through fabricating new lipid formulations, NLC. NLC, the second-generation lipid carrier is usually composed of solid lipids and liquid lipids together in a system. This mixing causes depression in melting point of substrates and converts the mixture into solid form at body temperature and termed as NLC. NLC shows a high drug loading with minimum drug expulsion. The unique advantages of NLC over SLN and Lipid-drug conjugates (LDC) are increased capacity of drug loading, avoidance of drug expulsion. This manuscript gives detailed information on definitions and simple way of production methods, new approaches in formulation of NLC and it also highlights how NLC improves bioavailability of bioactive molecules through peroral route and its future perspective as a pharmaceutical carrier.
