Abstract:Acute ingestion of exogenous ketone supplements in the form of a (R)-3-hydroxybutyl (R)-3-hydroxybutyrate (R-BD R-βHB) ketone monoester (KME) can attenuate declines in oxygen availability during hypoxic exposure and might impact cognitive performance at rest and in response to moderate-intensity exercise. In a single-blind randomized crossover design, 16 males performed assessments of cognitive performance before and during hypoxic exposure with moderate exercise [2 × 20 min weighted ruck (∼22 kg) at 3.2 km/h at 10% incline] in a normobaric altitude chamber (4572 m, 11.8% O2). The R-BD R-βHB KME (573 mg/kg) or a calorie- and taste-matched placebo (∼50 g maltodextrin) were co-ingested with 40 g of dextrose before exposure to hypoxia. The R-βHB concentrations were rapidly elevated and sustained (>3 mM; P < 0.001) by KME. The decline in oxygen saturation during hypoxic exposure was attenuated in KME conditions by 2.4%-4.2% (P < 0.05) compared with placebo. Outcomes of cognitive performance tasks, in the form of the Defense Automated Neurobehavioral Assessment (DANA) code substitution task, the Stroop color and word task, and a shooting simulation, did not differ between trials before and during hypoxic exposure. These data suggest that the acute exogenous ketosis induced by KME ingestion can attenuate declining blood oxygen saturation during acute hypoxic exposure both at rest and during moderate-intensity exercise, but this did not translate into differences in cognitive performance before or after exercise in the conditions investigated. HIGHLIGHTS: What is the central question of this study? Can exogenous ketosis act as a countermeasure to declines in blood oxygen saturation and cognitive performance during acute hypoxic exposure while performing a weighted ruck exercise? What is the main finding and its importance? Abstract:Ketone supplementation has been proposed to enhance cognition during exercise. To assess whether any benefits are due to reduced cognitive fatigue during the latter portions of typical sport game action, we induced cognitive fatigue, provided a ketone monoester supplement (KME) vs. a non-caloric placebo (PLAC), and assessed cognitive performance during a simulated soccer match (SSM). In a double-blind, balanced, crossover design, nine recreationally active men (174.3 ± 4.2 cm, 76.6 ± 7.4 kg, 30 ± 3 y, 14.2 ± 5.5 % body fat, V ̇O2 max = 55 ± 5 mL·kg BM−1·min−1; mean ± SD) completed a 45-min SSM (3 blocks of intermittent, variable intensity exercise) consuming either KME (25 g) or PLAC, after a 40-min mental fatiguing task. Cognitive function (Stroop and Choice Reaction Task [CRT]) and blood metabolites were measured throughout the match. KME reduced concentrations of both blood glucose (block 2: 4.6 vs. 5.2 mM, p = 0.02; block 3: 4.7 vs. 5.3 mM, p = 0.01) and blood lactate (block 1: 4.7 vs. 5.4 mM, p = 0.05; block 2: 4.9 vs. 5.9 mM, p = 0.01) during the SSM vs. PLAC, perhaps indicating a CHO sparing effect. Both treatments resulted in impaired CRT performance during the SSM relative to baseline, but KME displayed a reduced (p < 0.05) performance decrease compared to PLAC (1.3 vs. 3.4% reduction in correct answers, p = 0.02). No other differences in cognitive function were seen. These data suggest that KME supplementation attenuated decrements in CRT during repeated, high intensity, intermittent exercise. More study is warranted to assess fully the potential cognitive/physical benefits of KME for athletes.

Abstract:Reactive oxygen species are thought to partly be responsible for the hypoxia induced performance decrease. The present study evaluated the effects of a broad based antioxidant supplementation or the combined intake of alpha-ketoglutaric acid (α-KG) and 5-hydroxymethylfurfural (5-HMF) on the performance decrease at altitude. 18 healthy, well-trained males (age: 25±3 years; height: 179±6 cm; weight: 76.4±6.8 kg) were randomly assigned in a double-blind fashion to a placebo group (PL), a α-KG and 5-HMF supplementation group (AO1) or a broad based antioxidant supplementation group (AO2). Participants performed 2 incremental exercise tests to exhaustion on a cycle ergometer; the first test under normoxia and the second under hypoxia conditions (simulated altitude, FiO2=13% ~ 4 300 m). Supplementation started 48 h before the hypoxia test. Maximal oxygen uptake, maximal power output, power output at the ventilatory and lactate threshold and the tissue oxygenation index (NIRS) were measured under both conditions. Oxidative stress markers were measured before the supplementation and after the hypoxia test. Under hypoxia conditions all performance parameters decreased in the range of 19-39% with no differences between groups. A significant change from normoxia to hypoxia (p<0.001) and between groups (p=0.038) were found for the tissue oxygenation index. Post hoc test revealed significant differences between the PL and both, the AO1 and the AO2 group. The oxidative stress parameter carbonyl protein changed from normoxia to hypoxia in all participants and 4-hydroxynonenal decreased in the AO1 group only. In conclusion the results suggest that short-term supplementation with an antioxidant does not prevent the performance decrease at altitude. However, positive effects on muscle oxygen extraction, as indicated by the tissue oxygenation index, might indicate that mitochondrial functioning was actually influenced by the supplementation.

Abstract:Background A ketogenic diet (KD) induces nutritional ketosis (NS), benefits fatty acid oxidation (FAO), and favors moderate-intensity exercise capacity. The status that the body accommodates to produce and utilize ketone bodies (KB) and fatty acids as primary fuel sources is termed keto-adaptation. However, keto-adaptation requires time, while long-term KD also involves unfavored adverse effects. Exogenous ketone body (EKBs) administration has been introduced to elicit the advantages of NS. However, the direct use of EKBs fails to bring desired outcomes. We hypothesized that EKBs might only be effective during keto-adaptation. Methods Male C57BL/6J mice (n = 24) were divided into three groups: a control diet (Con, n = 8), a ketogenic diet (KD, n = 8), and a KD plus a ketone body (DL-β-Hydroxybutyric acid sodium salt, BHB) administration (KD+BHB, n = 8). After six weeks of KD administration, mice in the KD+BHB group receive BHB added into water bottles for another six weeks. Blood KB concentration is monitored throughout the experiment, while liver, gastrocnemius, and soleus mRNA are analyzed using RT-PCR. Results Both KD and KD+BHB induced and sustained NS and enhanced hepatic and muscular key genes regulating FAO. In addition, BHB administration upon keto-adaptation further increased circulating KB concentration and enhanced expressional levels of FAO-mediating genes ( ACO, HADH, ACADM , and MLYCD in the gastrocnemius muscle; ACO, HADH , and MLYCD in the soleus muscle), and energy-regulating genes ( PPARA and PPARG ) in the liver and skeletal muscle compared to a KD. Conclusion Compared to KD alone, chronic administration of KBs upon keto-adaptation increased the expression of key genes that favor FAO or maintain energy homeostasis in the liver and skeletal muscle. Instead of directly using EKBs in non-keto-adapted individuals, it is encouraged to use EKBs upon keto-adaptation status to elicit their energy-utilizing effects. Highlights This is the first report to evaluate the metabolic effects using exogenous ketone bodies on keto-adapted individuals. Administration of exogenous ketone body upon keto-adaptation furtherly increased circulating ketone bodies. Carbohydrate supplementation and exercise performance at high altitude: a randomized controlled trial

Abstract:Acute carbohydrate supplementation decreases effort perception and increases endurance exercise capacity at sea level. It also improves laboratory-based endurance performance at altitude. However, the effect of chronic carbohydrate supplementation at altitude, when acclimatization may attenuate carbohydrate effects, achieved doses are lower and metabolic effects may be different, is unknown and was therefore focused on in the present study. Forty-one members of a 22-day high altitude expedition were randomized in a double-blind design to receive either placebo or carbohydrate supplementation. Diet was manipulated with commercially available energy drinks consumed ad libitum throughout the expedition. Participants performed a mountaineering time trial at 5192 m, completed submaximal incremental exercise step tests to assess cardiovascular parameters before, during, and after the expedition, and recorded spontaneous physical activity by accelerometer on rest days. Compared to placebo, compliant individuals of the carbohydrate-supplemented group received daily an additional 3.5±1.4 g carbohydrate·kg body mass(-1). Compliant individuals of the carbohydrate supplemented group reported 18% lower ratings of perceived exertion during the time trial at altitude, and completed it 17% faster than the placebo group (both p<0.05 by t-test). However, cardiovascular parameters obtained during submaximal exercise and spontaneous physical activity on rest days were similar between the two groups (all p>0.05 by analysis of variance). This study utilized testing protocols of specific relevance to high altitude sojourners, including the highest mountaineering time trial completed to date at altitude. Chronic carbohydrate supplementation reduced ratings of perceived exertion and improved physical performance, especially during prolonged and higher intensity exercise tasks.

Abstract:The risk of plague as a bioweapon has prompted increasing research efforts to develop plague vaccines due to its extreme virulence and the ease of its transmission. Subunit vaccines that contain F1 and LcrV antigens of Y. pestis have been tested for safety and immunogenicity, but doubts have been raised about whether subunit vaccines that engender antibody responses will protect against pneumonic plague, which requires both humeral and cellular immune responses for protection. The live, attenuated vaccine EV76, a pgm locus deficient Y. pestis strain, has been used for a long time in the Former Soviet Union and some Asian countries, but is not commercially available in the US and Europe due to safety concerns. However, the live attenuated Y. pestis vaccines are still considered to be the most effective way to prevent plague. In this review, we present our opinions about rationally creating live, safe and immunogenic Y. pestis vaccines with potential use for human based on established researches.

Abstract:Acute mountain sickness occurs due to rapid altitude ascents and/or insufficient acclimatization. Acetazolamide (AZ) is commonly prescribed for AMS prophylaxis but inhibits exercise performance. Methazolamide (MZ), an analogous drug, has similar prophylactic benefits but does not impair isolated muscle mass exercise performance in normoxia. We sought to compare whole-body exercise performance in acute hypoxia (F I O 2 = 0.15) between AZ, MZ and placebo (PLA). Fifteen healthy participants completed 5 testing visits: day 1 maximal exercise test, day 2 a familiarization, and days 3-5 were the experimental visits. Each experimental visit involved a 5-km hypoxic cycling time trial performed after a 2-day dosing protocol of either AZ (250 mg t.i.d.), MZ (100 mg b.i.d.) or PLA (t.i.d.); the order was randomized and double-blinded. Prior to exercise, capillary blood samples were taken, and maximal voluntary contractions of quadriceps were performed . AZ and MZ resulted in a partially compensated metabolic acidosis at rest compared to PLA (capillary H + 47±3, 43±2, 39±2 nmol for AZ, MZ and PLA respectively, p<0.01). Time to complete 5-km with PLA (562±32 seconds, p 0.05). Overall, both AZ and MZ impair whole-body exercise performance in acute normobaric hypoxia.

Abstract:Background: Sleeping at (simulated) altitude is highly common in athletes as an integral part of altitude training camps or sport competitions. However, it is also often feared due to proclaimed negative effects on sleep quality, thereby potentially hampering exercise recovery and next-day exercise performance. We recently showed that ketone ester (KE) ingestion beneficially impacted sleep following strenuous, late evening exercise in normoxia, and alleviated hypoxemia. Therefore, we hypothesized that KE ingestion may be an effective strategy to attenuate hypox(em)ia-induced sleep dysregulations. Methods: Eleven healthy, male participants completed three experimental sessions including normoxic training and subsequent sleep in normoxia or at a simulated altitude of 3,000m while receiving either KE or placebo post-exercise and pre-sleep. Sleep was evaluated using polysomnography, while next-day exercise performance was assessed through a 30-min all-out time trial (TT30). Physiological measurements included oxygen status, heart rate variability, ventilatory parameters, blood acid-base balance and capillary blood gases. Results: Hypoxia caused a ~3% drop in sleep efficiency, established through a doubled wakefulness after sleep onset and a ~22% reduction in slow wave sleep. KE ingestion alleviated the gradual drop in SpO2 throughout the first part of the night, but did not alter hypoxia-induced sleep dysregulations. Neither KE, nor nocturnal hypoxia affected TT30 performance, but nocturnal hypoxia hampered heart rate recovery following TT30. Conclusion: We observed that sleeping at 3,000m altitude already impairs sleep efficiency. Although this hypoxia-induced sleep disruption was too subtle to limit exercise performance, we for the first time indicate that sleeping at altitude impairs next-day exercise recovery. KE alleviated nocturnal hypoxemia whenever SpO2 values dropped below ~85%, but this did not translate into improved sleep or next-day exercise performance.

Abstract:Abstract Background/Introduction Post-exercise ketone supplementation (PEKS) is commonly used in endurance athletes to alleviate overtraining symptoms and to stimulate recovery. The introduction of PEKS can blunt the development of symptoms associated with overtraining, suggesting a potential improvement in exercise recovery and exercise-induced cardiac adaptations. Although acute nutritional ketosis temporarily improves cardiac function at rest in healthy individuals and heart failure patients, the effects of PEKS on cardiac function during exercise and its relation to endurance performance remain to be tested. Purpose The aim of this study is to investigate the effects of PEKS on (a) endurance performance, (b) cardiac adaptations, and (c) safety, during a well-balanced 8-week training intervention to prevent overtraining. Methods Recreationally active young men (n=28) were enrolled in a double-blind, pair-matched, placebo-controlled, supervised 8-week endurance training program (Figure 1). The intervention group (n=14) received 25 grams of oral ketone ester (KE) immediately after each training session and 30 min before sleeping time, while the controls (n=14) ingested an isocaloric control drink (CON). Before (PRE) and after (POST) the training intervention, maximal oxygen uptake, endurance performance, and cardiac function during exercise were evaluated using a cardiopulmonary exercise test (CPET), a 30-minute time trial (TT30min), and a CPET combined with echocardiography (CPETecho), respectively. Left ventricular global longitudinal strain during exercise was used as a measure of contractile reserve. Results Mean power output during TT30min at POST (299±28W vs. 291±27W, p=0.0004) (Figure 2A), as well as during upright CPET (473±41W vs. 457±43W, p=0.029) was significantly higher between KE and CON. Peak oxygen consumption (KE: +208±320 mL vs. CON: +327±398 mL, p>0.05) and peak exercise cardiac output (KE: +0.82±3.28 L/min vs. CON: +1.67±1.97 L/min, p>0.05) during CPETecho increased after the training intervention, but no significant difference was observed between KE and CON (Figure 2B-C). Global longitudinal strain during exercise was similar in PEKS compared to controls. Conclusions This study is the first to demonstrate that PEK improves endurance performance in recreationally active men but does not impact cardiac function during exercise. This indicates that the positive effect of PEKS on exercise performance is not mediated by cardiac adaptations. Further research is required to elucidate the mechanisms through which PEKS improves endurance performance.

Abstract:Relative to carbohydrate (CHO) alone, exogenous ketones followed by CHO supplementation during recovery from glycogen-lowering exercise have been shown to increase muscle glycogen resynthesis. However, whether this strategy improves subsequent exercise performance is unknown. The objective of this study was to assess the efficacy of ketone monoester (KME) followed by CHO ingestion after glycogen-lowering exercise on subsequent 20 km (TT20km) and 5 km (TT5km) best-effort time trials. Nine recreationally active men (175.6 ± 5.3 cm, 72.9 ± 7.7 kg, 28 ± 5 y, 12.2 ± 3.2% body fat, VO2max = 56.2 ± 5.8 mL· kg BM−1·min−1; mean ± SD) completed a glycogen-lowering exercise session, followed by 4 h of recovery and subsequent TT20km and TT5km. During the first 2 h of recovery, participants ingested either KME (25 g) followed by CHO at a rate of 1.2 g·kg−1·h−1 (KME + CHO) or an iso-energetic placebo (dextrose) followed by CHO (PLAC + CHO). Blood metabolites during recovery and performance during the subsequent two-time trials were measured. In comparison to PLAC + CHO, KME + CHO displayed greater (p < 0.05) blood beta-hydroxybutyrate concentration during the first 2 h, lower (p < 0.05) blood glucose concentrations at 30 and 60 min, as well as greater (p 0.05) in power output nor time to complete either time trial were observed vs. PLAC + CHO. These data indicate that the metabolic changes induced by KME + CHO ingestion following glycogen-lowering exercise are insufficient to enhance subsequent endurance time trial performance.

A Pre-Workout Supplement of Ketone Salts, Caffeine, and Amino Acids Improves High-Intensity Exercise Performance in Keto-Naïve and Keto-Adapted Individuals

Abstract:Background: Acute ingestion of ketone supplements alters metabolism and potentially exercise performance. No studies to date have evaluated the impact of co-ingestion of ketone salts with caffeine and amino acids on high intensity exercise performance, and no data exists in Keto-Adapted individuals.Methods: We tested the performance and metabolic effects of a pre-workout supplement containing beta-hydroxybutyrate (BHB) salts, caffeine, and amino acids (KCA) in recreationally-active adults habitually consuming a mixed diet (Keto-Naïve; n = 12) or a ketogenic diet (Keto-Adapted; n = 12). In a randomized and balanced manner, subjects consumed either the KCA consisting of ∼7 g BHB (72% R-BHB and 28% S-BHB) with ∼100 mg of caffeine, and amino acids (leucine and taurine) or Water (control condition) 15 minutes prior to performing a staged cycle ergometer time to exhaustion test followed immediately by a 30 second Wingate test.Results: Circulating total BHB concentrations increased rapidly after KCA ingestion in KN (154 to 732 μM) and KA (848 to 1,973 μM) subjects and stayed elevated throughout recovery in both groups. Plasma S-BHB increased >20-fold 15 minutes after KCA ingestion in both groups and remained elevated throughout recovery. Compared to Water, KCA ingestion increased time to exhaustion 8.3% in Keto-Naïve and 9.8% in Keto-Adapted subjects (P < 0.001). There was no difference in power output during the Wingate test between trials. Peak lactate immediately after exercise was higher after KCA (∼14.9 vs 12.7 mM).Conclusion: These results indicate that pre-exercise ingestion of a moderate dose of R- and S-BHB salts combined with caffeine, leucine and taurine improves high-intensity exercise performance to a similar extent in both Keto-Adapted and Keto-Naïve individuals.

Abstract:Background: Previous studies have reported that sodium bicarbonate ingestion may enhance high-intensity exercise performance and cause severe gastrointestinal distress. However, enteric-coated sodium bicarbonate may reduce gastrointestinal symptoms of sodium bicarbonate after oral administration. This remains to be confirmed. This study aimed to verify the effects of serial and acute enteric-coated sodium bicarbonate supplementation on anaerobic performance, physiological profile, and metabolomics in healthy young men. Methods: Healthy young males (n = 12) ingested 0.2 g/kg body mass of enteric-coated sodium bicarbonate (ES) in serial enteric-coated sodium bicarbonate (SES, continuous ES supplementation for 5 days) and acute enteric-coated sodium bicarbonate (AES, acute ES supplementation before exercise) or a placebo (PL) in a randomized crossover design. After each supplement protocol, the participants completed four Wingate anaerobic tests (WAT). The first three Wingate tests (testing anaerobic capacity) were performed with a 5-min passive recovery between each. After the third Wingate test, participants were required to complete a 50-min recovery followed by a fourth WAT test (testing the recovery of anaerobic capacity after 50-min intervals). Blood lactate (BLA), heart rate (HR), and ratings of perceived exertion (RPE) were measured in all conditions during the test, as was the subjective gastrointestinal-symptoms assessment questionnaire (GSAQ). Mean power (MP) and peak power (PP) were recorded after four WATs. Urine samples were collected before the test and 50 min after the 3rd WAT. Results: Serial enteric-coated sodium bicarbonate supplementation improved anaerobic capacity in the third bout of WATs, as observed based on an increase in mean power (SES vs. PL (613 ± 57 vs. 542 ± 64 W), P = 0.024) and peak power (SES vs. PL (1,071 ± 149 vs. 905 ± 150 W), P = 0.016). Acute ES supplementation did not affect anaerobic capacity. The occurrence of gastrointestinal symptoms after enteric-coated sodium bicarbonate supplementation was minimal and no difference compared to placebo in the current study. In particular, serial enteric-coated sodium bicarbonate supplementation had no gastrointestinal side effects before the test. The AES and SES groups had a trivial effect on blood lactate compared to the PLA group. There was no significant difference in HR and RPE among the three groups. Based on targeted metabolomics analysis, the 50 min after the third WAT, the levels of lactate (P < 0.001), L-Malic acid (P < 0.05), and oxaloacetate (P < 0.05) were significantly higher in the SES group than in the PL group. Compared with the AES group, the levels of lactate and fumarate in the SES group were significantly increased (P < 0.05). Conclusions: Our study indicates that serial enteric-coated sodium bicarbonate supplementation positively improves anaerobic performance among healthy young men. However, acute ingestion of enteric-coated sodium bicarbonate did not improve anaerobic exercise performance. Either with serial or acute supplementation doses, enteric-coated sodium bicarbonate produced fewer gastrointestinal symptoms and no difference compared to placebo, especially with no gastrointestinal side effects after serial supplementation. Serial and acute supplementation of enteric-coated sodium bicarbonate might tend to promote lactate clearance. Furthermore, serial enteric-coated sodium bicarbonate ingestion may cause changes in the metabolism of lactate, L-Malic acid, oxaloacetate, and fumarate 50 min after exercise, which presumably may promote the tricarboxylic acid cycle and lactate clearance.
Acute ketone supplementation in the absence of muscle glycogen utilization: Insights from McArdle disease

Abstract:Background & Aims Ketone supplementation is gaining popularity. Yet, its effects on exercise performance when muscle glycogen cannot be used remain to be determined. McArdle disease can provide insight into this question, as these patients are unable to obtain energy from muscle glycogen, presenting a severely impaired physical capacity. We therefore aimed to assess the effects of acute ketone supplementation in the absence of muscle glycogen utilization (McArdle disease). Methods In a randomized cross-over design, patients with an inherited block in muscle glycogen breakdown (i.e., McArdle disease, n=8) and healthy controls (n=7) underwent a submaximal (constant-load) test that was followed by a maximal ramp test, after the ingestion of a placebo or an exogenous ketone ester supplement (30 g of D-beta hydroxybutyrate/D 1,3 butanediol monoester). Patients were also assessed after carbohydrate (75 g) ingestion, which is currently considered best clinical practice in McArdle disease. Results Ketone supplementation induced ketosis in all participants (blood [ketones]=3.7±0.9 mM) and modified some gas-exchange responses (notably increasing respiratory exchange ratio, especially in patients). Patients showed an impaired exercise capacity (-65% peak power output (PPO) compared to controls, p<0.001) and ketone supplementation resulted in a further impairment (-11.6% vs. placebo, p=0.001), with no effects in controls (p=0.268). In patients, carbohydrate supplementation resulted in a higher PPO compared to ketones (+21.5%, p=0.001) and a similar response was observed vs. placebo (+12.6%, p=0.057). Conclusions In individuals who cannot utilize muscle glycogen but have a preserved ability to oxidize blood-borne glucose and fat (McArdle disease), acute ketone supplementation impairs exercise capacity, whereas carbohydrate ingestion exerts the opposite, beneficial effect.

Abstract:Stress relaxation in three varieties of cherries was studied using the penetration test with a cylindrical pin, 4 mm in diameter, having a flat tip. One half of the fruit was modified by partial skinning, since experiments show that skin approximately doubles the apparent modulus of elasticity of the cherries. The skin also obscures a part of the fruit's distinctiveness, and affects the experimental results, even if a part of the fruit has been removed. A thermal activation theory with the residual part of stress was used to evaluate the stress relaxation experiments, and yielded results very similar to those previously obtained for vegetable flesh. Real consistency was observed mainly for residual stress and activation volume. It was observed that the relation between parameters Ct and log bt, and the initial relative slope of the relaxation curve provides information on fruit firmness.

Abstract:Mesenchymal stem cells are multipotent cells resident in the bone marrow throughout adulthood which have the capacity to differentiate into cartilage, bone, fat, muscle, and tendon. A number of monoclonal antibodies raised against human MSCs have been shown to react with surface antigens on these cells in vitro. A protein of molecular mass 92 kDa was immunoprecipitated using the SH-2 monoclonal antibody. This was purified and identified by peptide sequencing analysis and mass spectrometry as endoglin (CD105), the TGF-beta receptor III present on endothelial cells, syncytiotrophoblasts, macrophages, and connective tissue stromal cells. Endoglin on MSCs potentially plays a role in TGF-beta signalling in the control of chondrogenic differentiation of MSCs and also in mediating interactions between MSCs and haematopoietic cells in the bone marrow microenvironment.

Abstract:Introduction: Recent research has suggested that medium-chain triglyceride (MCT) supplementation may increase fat oxidation (FatOx) during aerobic exercise, sparing muscle glycogen and, perhaps, enhancing performance. As both MCT and caffeine (CAF) are theorized to elicit these effects, this pilot study’s purpose was to compare the physiological responses of their combined supplementation during submaximal cycling exercise. Methods: Eight aerobically trained males (mean±SD; age 23.6±4.4 years; body mass 82.3±15.8 kg; height 180.9±8.7 cm) completed one aerobic capacity (VO2peak) test and three 45-min exercise trials at 60% VO2peak. Blinded and counterbalanced, one-hour prior to each trial, participants consumed: MCT+CAF (20 mL + 100 mg), long-chain triglycerides (LCT)+CAF (40 mL + 100 mg), or CAF (100 mg). Oxygen consumption (VO2), carbon dioxide output (VCO2), and minute ventilation (VE) were measured, subsequently calculating respiratory exchange ratio (RER), energy expenditure (EE), carbohydrate oxidation (CarbOx), and FatOx. Results: No significant differences between conditions were observed for average VO2 (p=0.474; η2=0.101), VE (p=0.323; η2=0.149), (RER (p=0.323; η2=0.149), EE (p=0.474; η2=0.101), CarbOx (p=0.274; η2=0.169), or FatOx (p=0.478; η2=0.100) or for total EE (p=0.474; η2=0.101), CarbOx (p=0.274; η2=0.169), or FatOx (p=0.478; η2=0.100). Conclusions: Co-ingestion of MCT+CAF didn’t produce any significantly different physiological responses compared to co-ingestion of LCT+CAF or the CAF control.

Abstract:Findings from a recent 70 day bedrest investigation suggested intermittent exercise testing in the control group may have served as a partial countermeasure for skeletal muscle size, function, and fiber-type shifts. The purpose of the current study was to investigate the metabolic and skeletal muscle molecular responses to the testing protocols. Eight males (29±2y) completed muscle power (6x4 sec; peak muscle power: 1369±86W) and VO 2 max (13±1min; 3.2±0.2L/min) tests on specially designed supine cycle ergometers during two separate trials. Blood catecholamines and lactate were measured pre, immediately post, and 4h postexercise. Muscle homogenate and muscle fiber-type specific (myosin heavy chain (MHC) I and MHC IIa) mRNA levels of exercise markers ( myostatin, IκBα, myogenin, MuRF-1, ABRA, RRAD, Fn14, PDK4) and MHC I, IIa, and IIx were measured from vastus lateralis muscle biopsies obtained pre and 4h postexercise. The muscle power test altered (p≤0.05) norepinephrine (+124%), epinephrine (+145%), lactate (+300%), and muscle homogenate mRNA ( IκBα, myogenin, MuRF-1, RRAD, Fn14). The VO 2 max test altered (p≤0.05) norepinephrine (+1394%), epinephrine (+1412%), lactate (+736%), and muscle homogenate mRNA ( myostatin, IκBα, myogenin, MuRF-1, ABRA, RRAD, Fn14, PDK4). In general, both tests influenced MHC IIa muscle fibers more than MHC I with respect to the number of genes that responded and the magnitude of response. Both tests also influenced MHC mRNA expression in a muscle fiber-type specific manner. These findings provide unique insights into the adaptive response of skeletal muscle to small doses of exercise and could help shape exercise countermeasures for astronauts and Earth-based populations (e.g., aging individuals).
