Abstract:<p>Serotonin is one of the important neurotransmitters in human nervous system and associated with central nervous system diseases. Herein, we have prepared a novel electrochemical aptasensor for rapid and sensitive detection of serotonin by using the pre-designed and prepared DNA aptamers. In the absence of serotonin, the electron transfer rate on the aptasensor was faster than that in the presence of serotonin due to the hairpin structure of the aptamer was loose and MB could be closer to the electrode surface. While in the presence of serotonin, the hairpin structure of the aptamer was extended and MB was far away from the electrode surface. The effect of MB labeled sites on analytical performances of the proposed aptasensors was discussed by comparing sensitivity of the aptasensors that MB labeled in the intermediate of the aptamer with that MB labeled at the 3' end of the aptamer. It was found that sensitivity of the intermediate-labeled aptasensor was much higher than the terminal-labeled aptasensor due to the Specific conformational changes before and after aptamer binding to serotonin. The developed aptasensors exhibits a rapid electrochemical response and high sensitivity for the determination of serotonin. Under the optimal experimental conditions, the linear range for serotonin concentrations by the intermediate-labeled aptasensor was 1 pM to 10 nM with a detection limit of 0.017 fM (S/N=3). Moreover, the proposed aptasensor is reusable and shows good reproducibility and selectivity for the detection of serotonin in 100-fold diluted rat cerebrospinal fluid, suggesting a good application prospect in the detection of serotonin in real samples.</p>

Monitoring the Electrochemical Responses of Neurotransmitters Through Localized Surface Plasmon Resonance Using Nanohole Array.

Abstract:In this study, a novel spectroelectrochemical method was proposed for neurotransmitters detection. The central sensing device was a hybrid structure of nanohole array and gold nanoparticles, which demonstrated good conductivity and high localized surface plasmon resonance (LSPR) sensitivity. By utilizing such specially-designed nanoplasmonic sensor as working electrode, both electrical and spectral responses on the surface of the sensor could be simultaneously detected during the electrochemical process. Cyclic voltammetry was implemented to activate the oxidation and recovery of dopamine and serotonin, while transmission spectrum measurement was carried out to synchronously record to LSPR responses of the nanoplasmonic sensor. Coupling with electrochemistry, LSPR results indicated good integrity and linearity, along with promising accuracy in qualitative and quantitative detection even for mixed solution and in brain tissue homogenates. Also, the detection results of other negatively-charged neurotransmitters like acetylcholine demonstrated the selectivity of our detection method for transmitters with positive charge. When compared with traditional electrochemical signals, LSPR signals provided better signal-to-noise ratio and lower detection limits, along with immunity against interference factors like ascorbic acid. Taking the advantages of such robustness, the coupled detection method was proved to be a promising platform for point-of-care testing for neurotransmitters.

Abstract:Herein, an efficient electrochemiluminescence energy resonance transfer (ECL-RET) system was first applied in an immunosensor for the detection of chloramphenicol (CAP). SnS2 quantum dots (SnS2 QDs) has an extremely narrow electrochemical emission spectrum, which was used as donor. Compared with conventional quantum dots, toxic-element-free SnS2 QDs exhibit superior environmental friendliness and biocompatibility. Ultrathin Ag@Au nanosheets (Ag@Au NSs) was selected as acceptor due to its excellent structure and controllable UV-vis absorption range. Furthermore, ZnO nanoflowers (ZnO NFs) was employed to modify the electrode surface to provide stable luminescent environment and support more SnS(2 )QDs. Taking these advantages, the absorption spectra of Ag@Au NSs and the ECL emission spectra of SnS2 QDs are highly matched which ensures the occurrence of ECL-RET. When the CAP analyte were absent, Ag@Au NSs was immobilized on the electrode, and its ultrathin thickness effectively shortens the path of ECL-RET to produce the weak ECL intensity. Inversely, CAP analyte would compete with coating antigen for the certain amount of antibody to remove excess Ag@Au NSs and cause relatively high ECL intensity. As a result, the proposed immunosensor performed satisfactory sensitivity with a wide linear range from 0.005 to 1000 ng(-1) and a low detection limit (1.7 pg mL(-1)).
Aptamer-based detection of serotonin based on the rapid in situ synthesis of colorimetric gold nanoparticles

Abstract:Serotonin, a neurotransmitter that affects brain function, is associated with cancer progression, thus making it a potential biomarker. Despite the increasing efforts and ideas for gold nanoparticle (AuNP)-based colorimetric detection over the years, preparing AuNPs and sensing targets are separate processes, and this incurs more time to operate and produces excess waste. Herein, we report a simple, sensitive, and rapid colorimetric detection method for serotonin based on the in situ formation of AuNP. When only the aptamer is present, it can prevent chloride-induced aggregation of AuNPs because it easily binds to the freshly synthesized AuNPs through its exposed bases to increase the positive charge of the AuNP surfaces. When a complex of serotonin and its aptamer is formed, this complex disturbs the adsorption between aptamers and AuNPs, resulting in reduced stability of AuNPs and easy aggregation of nanoparticles. Therefore, serotonin was measured by color change, consistent with the change in peak intensity in the UV-vis absorption spectrum. The sensor demonstrated good sensitivity with a detection limit of 1 ng/mL (5.7 nM) for serotonin, which is comparable to or better than that of other aptamer-based colorimetric detection methods, further exhibiting the requisite selectivity against possible interferents. These results serve as a basis for developing other biosensors using aptamer-mediated in situ growth of AuNPs.

Abstract:Resonance energy transfer (RET) quenching is significantly important for developing electrochemiluminescence (ECL) sensors, but RET platforms face challenges like interference from other fluorescent substances and reliance on energy transfer efficiency. This study used Zn-PTC, formed by zinc ions coordinated with perylene-3,4,9,10-tetracarboxylate, as a dual-mechanism quencher to reduce the ECL intensity of carbon nitride nanosheets (Tg-CNNSs). Co3O4/NiCo2O4 acts as a coreaction promoter, enhancing and stabilizing the luminescence of Tg-CNNSs. Zn-PTC absorbs energy from Tg-CNNSs, altering the fluorescence lifetime to confirm energy transfer, while energy-level matching demonstrates electron transfer. By leveraging both RET and electron transfer mechanisms, the designed ECL aptasensor significantly reduces signal fluctuations that may arise from a single mechanism, resulting in more stable and reliable detection outcomes. The ECL aptasensor designed for furanyl fentanyl (FUF) detection shows excellent performance with a detection limit of 5.7 × 10-15 g/L, offering new pathways for detecting FUF and other small molecules.

Abstract:Serotonin (5-hydroxytryptamine, 5-HT) is a pivotal monoamine neurotransmitter, which is widely distributed in human brain for biological, physical and psychopathological processes. The content of 5-HT can support diagnose of various diseases. To selectively detect 5-HT is very important in clinical medicine. Here, a novel microbiosensor for 5-HT is studied on acupuncture needle. Molecularly imprinted film enwrapped 5-HT was electropolymerized onto bimetallic gold/platinum (Au/Pt) nanoparticles on acupuncture needle microelectrode (ANME). Au/Pt nanostructure exhibited active sites to catalyze the oxidation of 5-HT and bind the generated polymer. 5-HT can be enwrapped by the functional monomer of pyrrole (Py) in the process of electropolymerization with suitably electroactive conformation. Comparing with interfaces of single metal or molecularly imprinted layer, synergistic microbiosensor exhibit better performance for 5-HT. 5-HT can be adsorbed and catalytically oxidized by the imprinted cavities. Under optimized conditions, the peak current linearly increases with the concentration of 5-HT from 0.03 to 500 μM, and a detection limit of 0.0106 μM is obtained. The performance of this microbiosensor is competitive with previous studies. Furthermore, the prepared microbiosensor showed effective application to analyze 5-HT in human serum and urine. Interestingly, the microbiosensor expressed the real-time monitoring ability to 5-HT from stimulated PC12 cells by K+. The microbiosensor also exhibited high selectivity, stability and reproducibility, which is promising in view of the low price, fast response and simple operation.

Abstract:The misuse of antibiotics may contaminate the environment and cause harm to human health. Therefore, rapid and accurate detection of antibiotics is essential. In this study, a novel electrochemiluminescence resonance energy transfer (ECL-RET) pair was designed using a new ECL emitter (CPM, Ce-TBAPy) as the donor and Co-MOF@AuPt as the acceptor. Moreover, a highly sensitive and specific "on-off-on" ECL aptasensor was constructed for detecting sulfadiazine (SDZ). The aptasensor exhibited a broad linear range (from 10.0 fg mL-1 to 100 ng mL-1) for the SDZ concentration, with limit of detection and limit of quantification values of 1.14 fg mL-1and 3.75 fg mL-1, respectively. The aptasensor achieved good results in spiking experiments with milk and egg samples, and successfully quantified SDZ in fish meal quality control sample. The prepared aptasensor presents great potential for food and environmental safety by detecting antibiotics.

Abstract:The 3s-3p excitation of spin-polarized Na atoms by Ne/sup +/ ions has been studied for impact energies E/sub lab/ = 200 eV to 6 keV, i.e., in the adiabatic regime. The total excitation cross section and the three Stokes polarization parameters of the fluorescence light have been measured. The linear polarization of the light shows a preferential excitation of the chemically bondm/sub l/chemically bond = 1 magnetic substates. The circular polarization probes the spin orientation of the excited 3p state. At the highest impact energies investigated the experimental data are compatible with conservation of spin orientation during the collision. With decreasing impact energy, the spin polarization of the final 3p state becomes smaller than the spin polarization of the initial 3s state. This apparent spin depolarization is attributed to the exchange interaction between the Na valence electron and the unfilled Ne/sup +/ 2p/sup 5/ core in the quasimolecule formed during the collision.

Abstract:Serotonin (SRT), one among the neurotransmitters, is mainly responsible for physiological functions in human body. The effect of changes and intake of SRT can influence higher impact of peoples, thus sensing such compound with more effective and simple technique will be an extreme significance. Zeolite imidazole framework among MOF was constructed with the correlation of multiwall carbon nanotubes (ZIF-67/MWCNT) as sensor material via simple co-precipitation method. The rationally designed ZIF-67/MWCNT was evaluated for physico-chemical properties analyzed with XRD, XPS and the morphological structures were identified with FESEM. Cyclic voltammetry (CV) and amperometric (IT) technique was initiated for electrochemical detection of SRT. The ZIF-67/MWCNT exhibited a dodecahedron shape and larger surface area as agreed from BET analysis. The electrochemical results obtained at the interfacial site of ZIF-67/MWCNT proved to be more competent with excellent selectivity. SRT was electrochemically detected in human blood serum and urine as real sample analysis. The linear range was 0.049 μM to 800 μM with a lower LOD-7 nM. Moreover, the attained sensitivity was 5.613 µA µM−1 cm2 with excellent repeatability and reproducible behavior. The highly conducting matrix ZIF-67/MWCNT with abundantly accessible sites and higher surface area has gained significant results towards SRT detection.

Abstract:Serotonin (5-HT) is an important neurotransmitter and also a biomarker closely related to a variety of neurodegenerative diseases. In this work, a dual-emissive europium-copper cluster, as an artificial ultra-small-sized luminescent enzyme (clusterzyme), was specially designed and synthesized for the catalysis and sensing of 5-HT. This clusterzyme is catalysis and luminescence-sensing all-in-one, not only highly tyrosinase-active but also ultra-sensitive for 5-HT, and can sense 5-HT as low as 0.5 nM through the enhanced luminescence of Eu 3+ . The enzymatic detection for 5-HT is outstandingly specific even if in the presence of similar neurotransmitters such as dopamine (DA), epinephrine (EP) and nomethylephrine (NE). The prepared clusterzyme was found to be able to catalyze the oxidation of 5-HT like tyrosinase to generate a neurotoxic product of tryptamine-4, 5-dione. Mechanism of catalysis and sensing of the clusterzyme for 5-HT have been proposed and demonstrated. Such a luminescent clusterzyme can be used to replace the combination of natural enzymes and chromogenic agents currently widely used in enzyme-based assays, also offering a promising prospect for measuring neurotransmitters in situ and visually due to the unique advantage of its ultra-small-size allowing passage through the blood-brain barrier.

Abstract:5-Hydroxytryptamine (5-HT) is an important neurotransmitter, and its content in the human body is of great significance to human health. In this study, an l-cysteine-terminated triangular silver nanoplate loaded on a MXene (two-dimensional transition metal carbide or nitride) (Tri-AgNP/l-Cys/MXene) electrochemical sensor was used to detect 5-HT. As an electrically active amino acid with a sulfhydryl group, l-cysteine (l-Cys) forms a more stable Ag-S bond with silver nanoparticles, which can selectively substitute trisodium citrate (TSC) in TSC-capped triangular silver nanoplates (Tri-Ag-NP/TSC). Due to the good conductivity, biocompatibility, and large surface area, MXenes provide a good platform for loading Tri-AgNP/l-Cys. Under optimized conditions, the concentration range for detecting 5-HT with the sensor is 0.5-150 μM, and the limit of detection (LOD) is 0.08 μM (S/N = 3). For detecting 5-HT in actual serum samples, the sensor also showed a good recovery rate (95.38-102.3%), and the relative standard deviation was 2.2-3.4%.

Abstract:The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for the synthesis of green- to near-infrared-emitting CdSeTe/CdS/ZnS QDs with time- and component-tunable photoluminescence was proposed. And, on the basis of the adjustable optical properties of both CdSeTe/CdS/ZnS QDs and GNRs, excellent overlap between donor emission and acceptor absorption can be obtained to ensure effective ECL-ET quenching, thus improving the sensing sensitivity. This method represents a novel approach for versatile detection of biomolecules at low concentrations.

Abstract:In the present work, electrogenerated chemiluminescence (ECL) of luminol was investigated in neutral condition at a gold electrode in the presence of silicon quantum dots (SiQDs). The results revealed that SiQDs can not only greatly enhance luminol ECL, but also act as energy acceptor to construct a novel ECL resonance energy transfer (ECL-RET) system with luminol. As a result, strong anodic ECL signal was obtained in neutral condition at the bare gold electrode, which is suitable for biosensing application. Lysozyme exhibited apparent inhibiting effect on the ECL-RET system, based on which an ECL aptasensor was fabricated for the sensitive detection of lysozyme. The proposed method showed high sensitivity, good selectivity, and wide linearity for the detection of lysozyme in the range of 5.0×10-14-5.0×10-9gmL-1 with a detection limit of 5.8×10-15gmL-1 (3σ). The results suggested that as-proposed luminol/SiQDs ECL biosensor will be promising in the detection enzyme.
Dual-quenching electrochemiluminescence system based on resonance energy transfer from gold dendrite@polypyrrole core–shell nanoparticles enhanced g-C3N4 to ZnONFs@PDA-sCuO for procalcitonin immunosensing

Abstract:This work presents a dual-quenching electrochemiluminescence resonance energy transfer (ECL-RET) immunosensor on account of the double quenching effects of polydopamine coated ZnO nanoflowers (ZnONFs@PDA) loaded with small-sized copper oxide nanoparticles (sCuO), named ZnONFs@PDA-sCuO, towards gold dendrite@polypyrrole core–shell nanoparticles (AuD@PPy) enhanced g-C3N4 (g-C3N4-AuD@PPy) for sensitive analysis of procalcitonin (PCT). To be specific, AuD@PPy with core-shell structure were loaded onto g-C3N4 to enhance the emission performance of g-C3N4. In addition, the ultraviolet absorption spectra of ZnONFs@PDA and sCuO showed considerable overlap with the ECL emission spectra of g-C3N4 appropriately. So sCuO and ZnONFs@PDA were combined and designed as an efficient dual-quencher of ECL luminescence of g-C3N4 by RET interaction. The biosensor showed superior linear detection range from 0.00005 to 50 ng mL−1, with a sensitive detection limit of 17.2 fg mL−1 (S/N = 3). It is worthy to note that a new type ECL-RET couple made up of g-C3N4-AuD@PPy (donor) and ZnONFs@PDA-sCuO (acceptor) was developed to construct a sandwich ECL biosensor for PCT detection. The studied immunosensor had satisfactory sensitivity, specificity and reproducibility, indicating the proposed sensing method could provide a good technical means and theoretical basis for the diagnosis of serious diseases.

Abstract:An effective and precise electrochemiluminescence resonance energy transfer (ECL-RET), including the efficient regulation over the proximity of a donor and an acceptor and the reliable stimuli responsive as well as the avoidance of undesirable probes leakage, etc., is significant for the development of an accurate and sensitive ECL detection method; yet, the current literature in documentation involves only a limited range of such ECL-RET systems. Herein, we propose an ECL-RET strategy with dually quenched ultralow background signals and a dual-stimuli responsive, accurate signal output for the ultrasensitive and reliable detection of anatoxin-a (ATX-a). The dual quenching is accomplished by an integrated ECL-RET probe of metal organic frameworks (MOFs) encapsulated into Ru(bpy)32+ (Ru-MOF) (donor) coated with silver nanoparticles (AgNPs) shell (acceptor 1) and close proximity with DNA-ferrocene (Fc) (acceptor 2). Multistimuli responsive DNAzyme facilitated the accurate signal switch by both target ATX-a and hydrogen peroxide (H2O2). Because of the specific recognition of the aptamer toward ATX-a, an intricate design of the DNA sequence enabled the exposure of the Ag+-dependent DNAzyme sequence and H2O2 in situ generated Ag+ triggering a catalytic cleavage reaction to freely release the two ECL-RET energy acceptors, thus switching the ECL signal significantly and achieving ultrasensitive detection. It is noteworthy that AgNPs are key in this ECL-RET strategy, serving both as the gate-keepers for avoiding ECL probes leakage and also the ECL energy acceptors, and mostly importantly serving as the redox substrate for the subsequent DNAzyme catalytic signal switch. The proposed ECL-RET aptasensor for ATX-a detection displayed splendid monitoring performance with a quite low detection limit of 0.00034 mg mL–1. Synergistic Multieffect Catalytic Amplified Cathodic Electrochemiluminescence Biosensor via Target Binding-Induced Aptamer Conformational Changes for the Ultrasensitive Detection of Synthetic Cathinone

Abstract:Signal amplification is a powerful approach to increasing the detection sensitivity of electrochemiluminescence (ECL). Here, we developed synergistic multieffect catalytic strategies based on CuCo2O4 nanorod combination of Ag NPs as coreaction accelerators to fabricate an efficient covalent organic framework (PTCA-COF)-based ternary ECL biosensor. Concretely, the high redox reversibility of Co3+/Co2+ and Cu2+/Cu+ would constantly promote the decomposition of S2O82- for ECL emission. Meanwhile, the introduction of Ag NPs with excellent electrocatalytic activity further realized multiple amplification of the ECL signal. Furthermore, the good hydrogen evolution reaction (HER) ability of Ag@CuCo2O4 nanorods could accelerate the proton transmission rate of the system to amplify ECL behavior. In the presence of the target synthetic cathinone 4-chloroethcathinone (4-CEC) as the quenching ECL signal-response probe, the Ferrocene (Fc)-labeled aptamer folded into the conformationally limited stem-loop structure, bringing Fc near the ECL luminophore and resulting in quenched ECL emission. The quenching effect was connected with target-induced aptamer conformational changes and consequently reflected the target concentration. Under optimum conditions, the proposed biosensor realized a highly sensitive assay for 4-CEC with a large dynamic range from 1.0 × 10-12 to 1.0 × 10-6 g/L and a detection limit as low as 2.5 × 10-13 g/L. This study integrated multiple amplification strategies for efficient ECL enhancement, which provided a novel approach to constructing highly bioactive and sensitive sensors.

Abstract:The organic luminophores have inspired widespread interest in electrochemiluminescence (ECL). Herein, a novel rod-like metal-organic framework was formed by chelating Zn ion with 9,10-di(p-carboxyphenyl)-anthracene (DPA), defined as Zn-MOF for simplicity. In this proposal, the prepared Zn-MOF was first used as a powerful organic luminophore with low trigger potential, thus developing a competitive ECL immunoassay for ultrasensitive detection of 5-fluorouracil (5-FU) with 1,4-diazabicyclo[2.2.2]octane (D-H 2 ) as the coreactant. The absorption spectrum of cobalt oxyhydroxide (CoOOH) nanosheets and the ECL emission spectrum of Zn-MOF could be highly matched, which ensured the occurrence of resonance energy transfer (RET). For that, ECL-RET was applied in the assembly strategy of the ECL biosensor, and Zn-MOF was used as the energy donor and CoOOH nanosheets as the acceptor. Taking advantage of the luminophore and ECL-RET, the immunoassay can be used for ultra-sensitive quantitative detection of 5-fluorouracil. The proposed ECL-RET immunosensor showed satisfactory sensitivity and accuracy with a wider linear range from 0.001 to 1000 ng/mL, and a lower detection limit (0.52 pg/mL). Hence, it is worth believing that this strategy can pave a bright research direction for the detection of 5-FU or other biological small molecules.
