Abstract:The development of a highly efficient electrochemiluminescence (ECL) emitter represents an effective strategy for enhancing the sensitivity and repeatability of ECL immunosensors. In this study, a sandwich-type ECL immunosensor with triple enhancement was developed to detect carcino-embryonic antigen (CEA). This sensor is based on a porous structure of iron-based metal-organic framework (NH 2 -MIL-88(Fe)), encapsulating the luminescent tris(2,2′-bipyridine)ruthenium (II) (Ru (bpy) 3 2+ ), Au@MoS 2 with a 3D nanoflower structure as an enhanced substrate. In this system, the MOFs framework encapsulated luminophore was realized to solve its water solubility to reach stable luminescence, as well as the triple enhancement effect based on the principle of amino catalysis, Mo 4+ /Mo 6+ active site conversion, and gold nanoparticles (Au NPs) promotion, which significantly enhanced the detection sensitivity. Furthermore, the ECL immunosensor demonstrated successful application in the highly sensitive and selective detection of CEA, achieving a detection limit of 38.9 fg mL −1 . The sensor demonstrates remarkable sensitivity, specificity, stability, repeatability, and practicality in the analysis of human serum samples. This investigation presents a highly effective approach for the ultrasensitive detection of trace proteins.

Abstract:Carcinoembryonic antigen (CEA) is often employed as a crucial tumor marker for both colon and rectal cancer. The assessment of CEA levels holds significant therapeutic importance in the diagnosis of these malignancies. In this study, an electrochemiluminescence immunosensor with a signal off was ingeniously designed with Ru@TiO 2 -MXene as the base material of the energy donor, and Pd@UiO-66-NH 2 as the marker of the energy acceptor. This immunosensor was constructed based on the strong interaction between the two components of ECL-RET. Remarkably, TiO 2 -MXene exhibits favorable film-forming properties and electrical conductivity while effectively immobilizing Ru(bpy) 3 2+ . On the other hand, Pd@UiO-66-NH 2 has excellent biocompatibility and light absorption, making it an ideal choice for use as a secondary antibody marker. The electrochemiluminescence immunosensor constructed exhibits an extensive detection range of 1 × 10 −5 ng·ml −1 to 80 ng·ml −1 and a narrow detection limit of 2.65 fg·ml −1 . The novel electrochemiluminescence immunosensor, as presented in this investigation, introduces a distinct methodology for CEA detection, carrying immense clinical significance in the field of colon and rectal cancer diagnosis.

Abstract:The concentration of carcinoembryonic antigen (CEA) is an important indicator for clinical monitoring and diagnosis of tumor. Early sensitive detection of CEA is significant for cancer diagnosis and therapy. In this study, a sensitive electrochemiluminescence (ECL) immunosensor was fabricated based on functionalized metal organic layers (MOLs) for the determination of CEA. In this immunosensor, Au nanoparticles (AuNPs) were grown on ultrathin two-dimensional MOLs via in situ reduction. AuNPs not only can accelerate the electron transfer but also can be used as the labels to conjugate with antibody. The as-prepared nanocomposite showed high ECL efficiency and can be used as an ECL emitter owing to the great ECL property of Ir-Zr-MOL. The ECL intensity of decreased linearly with the logarithm of CEA concentration in the range from 1.00 pg mL −1 to 100 ng mL −1 . And the detection limit was 0.200 pg mL −1 (S/N = 3). What's more, the as-prepared immunosensor worked well in human blood samples, showing its potential for real analysis.

Abstract:Background: Carcinoembryonic antigen (CEA) is a significant glycosylated protein, and the unusual expression of CEA in human serum is used as a tumor marker in the clinical diagnosis of many cancers. Although scientists have reported many ways to detect CEA in recent years, such as electrochemistry, photoelectrochemistry, and fluorescence, their operation is complex and sensitivity is average. Therefore, finding a convenient method to accurately detect CEA is significance for the prevention of malignant tumors. With high sensitivity, quick reaction, and low background, electrochemiluminescence (ECL) has emerged as an essential method for the detection of tumor markers in blood. Results: In this work, a "signal on-off" ECL immunosensor for sensitive analysis of CEA ground on the ternary extinction effects of CuFe2O4@PDA-MB towards a self-enhanced Ru(dcbpy)32+ functionalized metal-organic layer [(Hf)MOL-Ru-PEI-Pd] was prepared. The high ECL efficiency of (Hf)MOL-Ru-PEI-Pd originated from the dual intramolecular self-catalysis, including intramolecular co-reaction between polyethylenimine (PEI) and Ru (dcbpy)32+. At the same time, loading Pd NPs onto (Hf)MOL-Ru-PEI could not only improve the electron transfer ability of (Hf)MOL-Ru-PEI, but also provide more active sites for the reaction of Ru(dcbpy)32+ and PEI. In the presence of CEA, CuFe2O4@PDA-MB-Ab2 efficiently quenches the excited states of (Hf)MOL-Ru-PEI-Pd by PDA, Cu2+, and methylene blue (MB) via energy and electron transfer, leading to an ECL signal decrease. Under optimal conditions, the proposed CEA sensing strategy showed satisfactory properties ranging from 0.1 pg mL-1 to 100 ng mL-1 with a detection limit of 20 fg mL-1. Significance: The (Hf)MOL-Ru-PEI-Pd and CuFe2O4@PDA-MB were prepared in this work might open up innovative directions to synthesize luminescence-functionalized MOLs and effective quencher. Besides, the ECL quenching mechanism of Ru(dcbpy)32+ by MB was successfully explained by the inner filter effect (ECL-IFE). At last, the proposed immunosensor exhibits excellent repeatability, stability, and selectivity, and may provide an attractive way for CEA and other disease markers determination.
Surface-enhanced electrochemiluminescence combined with resonance energy transfer for sensitive carcinoembryonic antigen detection in exhaled breath condensates

Abstract:The detection of biomarkers in exhaled breath condensates (EBCs) is regarded as a promising non-invasive diagnostic approach. However, the ultralow concentration of biomarkers in EBCs is a great challenge. Herein, a sensitive dual signal amplification strategy was developed based on surface-enhanced electrochemiluminescence (SEECL) combined with resonance energy transfer (RET). Gold nanoparticles-functionalized graphite-like carbon nitride nanohybrids (Au-g-C3N4 NHs) could be used as an energy transfer donor because of the good overlap between its emission peak and the absorption peak of tris(2,2'-bipyridine)ruthenium dichloride (Ru(bpy)3Cl2) at 460 nm. Gold-silicon dioxide core-shell nanoparticles doped with Ru(bpy)32+(Au@SiO2-Ru) were employed as an energy transfer acceptor emitting at 620 nm. Moreover, the signals at 620 nm emitted by Ru (bpy)32+ were enhanced by 5 times, attributed to the localized surface plasmon resonance (LSPR) of gold nanoparticles (Au NPs). The detection of carcinoembryonic antigen (CEA) was performed by using two aptamers as the recognition unit; whereby aptamer 1 (Apt1) was modified on the surface of Au-g-C3N4 NHs, and aptamer 2 (Apt2) was banded on the surface of Au@SiO2-Ru. In the presence of CEA, a sandwich structure was formed between Au-g-C3N4 NHs-Apt1-CEA and Apt2-Au@SiO2-Ru, which resulted in an ultrasensitive detection of CEA. The proposed electrochemiluminescence sensor showed a wide linear relationship with the CEA concentration in the range from 1.0 pg mL-1 to 5.0 ng mL-1, with a limit of detection (LOD) of 0.3 pg mL-1. Finally, the practicality of the proposed sensor was demonstrated to detect CEA in EBCs, and the obtained results were in good agreement with the enzyme-linked immunosorbent assay (ELISA) method.

Abstract:Recently, tris(2,2'-bipyridine)ruthenium(ii) (Ru(bpy)32+) has been widely used in electrochemiluminescence (ECL) biosensors. However, its ECL has to be triggered at high positive or negative voltage, leading to a series of undesired side effects containing electrical damage toward biomolecules and hydrogen/oxygen evolution reaction. In this work, a new low-potential-triggered Ru(bpy)32+ ECL system was reported using oxalate as coreactant with the aid of platinum nanoparticles (Pt NPs) electrodeposited on the surface of the graphene-ionic liquid composite. When cyclic scanning in a potential range from 0.3 to -0.2 V, the ECL signal appeared at onset potential of 0.05 V (vs. Ag/AgCl), and reached the maximum at about -0.15 V (vs. Ag/AgCl), which were quite lower than that in the typical Ru(bpy)32+-oxalate (C2O42−) ECL system. The possible mechanism was discussed in detail. Meanwhile, the low-potential-triggered ECL signals were strong and stable, which were further exploited to detect the carcinoembryonic antigen (CEA). The constructed ECL sensor exhibited a good linearity between the ECL signals and the logarithm of the concentrations of CEA in a range from 0.1 pg mL-1 to 100 ng mL-1 with the detection limit of 0.01 pg mL-1. In addition, the sensor showed a promising potential in clinical detection.

Abstract:Herein, a label-free electrochemiluminescence (ECL) biosensor for sensitive determination of carcinoembryonic antigen (CEA) was fabricated based on metal organic framework enriched CdS quantum dots (CdS ) and triethanolamine-modified gold nanoparticles () as bi-coreactants of Ru(bpy)32+ ECL system. The served as not only coreactant of Ru(bpy)32+ but also nanocarrier for CEA aptamer which was bonded to the through Au−S bond. After incubated in CEA solution, the ECL signals decreased due to the poor conductivity of CEA and there was a linear correlation between the ECL intensity and log(CCEA). Based on this result, a label-free ECL biosensor has been developed successfully and used to detect CEA with a detection limit of 85 fg/mL. The proposed ECL aptasensor had good analytical performance in the detection of CEA in human serum samples.

Abstract:Recently, electrochemiluminescent (ECL) immunosensors have received much attention in the field of biomarker detection. Here, a highly enhanced ECL immunosensing platform was designed for ultrasensitive detection of carcinoembryonic antigen (CEA). The surface of the glassy carbon electrode was enhanced by applying functional nanostructures such as thiolated graphene oxide (S-GO) and streptavidin-coated gold nanoparticles (SA-AuNPs). The selectivity and sensitivity of the designed immunosensor were improved by entrapping CEA biomolecules using a sandwich approach. Luminol/silver nanoparticles (Lu-SNPs) were applied as the main core of the signaling probe, which were then coated with streptavidin to provide overloading of the secondary antibody. The highly ECL signal enhancement was obtained due to the presence of horseradish peroxidase (HRP) in the signaling probe, in which the presence of H 2 O 2 further amplified the intensity of the signals. The engineered immunosensor presented excellent sensitivity for CEA detection, with limit of detection (LOD) and linear detection range (LDR) values of 58 fg mL −1 and 0.1 pg mL −1 to 5 pg mL −1 ( R 2 = 0.9944), respectively. Besides its sensitivity, the fabricated ECL immunosensor presented outstanding selectivity for the detection of CEA in the presence of various similar agents. Additionally, the developed immunosensor showed an appropriate repeatability (RSD 3.8%) and proper stability (2 weeks). Having indicated a robust performance in the real human serum with stated LOD and LDR, the engineered immunosensor can be considered for the detection and monitoring of CEA in the clinic.

Abstract:Simple development of an electrochemiluminescence (ECL) immunosensor for convenient detection of tumor biomarker is of great significance for early cancer diagnosis, treatment evaluation, and improving patient survival rates and quality of life. In this work, an immunosensor is demonstrated based on an enhanced ECL signal boosted by nanochannel-confined Au nanomaterial, which enables sensitive detection of the tumor biomarker—carcinoembryonic antigen (CEA). Vertically-ordered mesoporous silica film (VMSF) with a nanochannel array and amine groups was rapidly grown on a simple and low-cost indium tin oxide (ITO) electrode using the electrochemically assisted self-assembly (EASA) method. Au nanomaterials were confined in situ on the VMSF through electrodeposition, which catalyzed both the conversion of dissolved oxygen (O2) to reactive oxygen species (ROS) and the oxidation of a luminol emitter and improved the electrode active surface. The ECL signal was enhanced fivefold after Au nanomaterial deposition. The recognitive interface was fabricated by covalent immobilization of the CEA antibody on the outer surface of the VMSF, followed with the blocking of non-specific binding sites. In the presence of CEA, the formed immunocomplex reduced the diffusion of the luminol emitter, resulting in the reduction of the ECL signal. Based on this mechanism, the constructed immunosensor was able to provide sensitive detection of CEA ranging from 1 pg·mL−1 to 100 ng·mL−1 with a low limit of detection (LOD, 0.37 pg·mL−1, S/N = 3). The developed immunosensor exhibited high selectivity and good stability. ECL determination of CEA in fetal bovine serum was achieved.

Abstract:An ultrasensitive enzyme-free electrochemical immunoassay was developed for detection of the fg/mL level carcinoembryonic antigen (CEA) by using a double strand DNA@Au nanoparticle (dsDNA@AuNP) tag and hexaammineruthenium(III) chloride (RuHex) as the electroactive indicator. The dsDNA@AuNP was synthesized by one-pot hybrid polymerization of dsDNA on initiator DNA modified AuNPs via hybridization chain reaction. The immunosensor was prepared by covalently cross-linking capture antibody on chitosan/AuNP nanocomposite modified glass carbon electrode. The AuNPs accelerated the electron transfer and led to high detection sensitivity. With a sandwich-type immunoreaction and a biotin-streptavidin affinity reaction, the dsDNA@AuNP tag was conjugated on the immunocomplex to bring a high amount of RuHex to the electrode surface via electrostatic interaction, resulting in an amplified electrochemical signal. Under optimal conditions, the proposed sensing platform showed a wide linear detection range from 10 fg/mL to 10 ng/mL along with a detection limit of 3.2 fg/mL for CEA. The immunosensor exhibited high sensitivity and good stability, showing a promising application in early cancer diagnosis and could be extended to sensitive electrochemical biosensing of other analytes.

Abstract:Aggregation-induced electrochemiluminescence (AIECL) combines the merits of aggregation-induced emission (AIE) and electrochemiluminescence (ECL), which has become a research hot spot in recent years. Therefore, we synthesized a novel AIE compound (Z)-3-(4-(2-butyl-1,3-dioxo-2,3-dihydro-1H-benzo[de]isoquinolin-6-yl)phenyl)-2-(4-(1,2,2-triphenylvinyl)phenyl)acrylonitrile (TPENI) with a donor-acceptor (D-A) structure, that is, a simple peripheral modification of 4-(2-butyl-1,3-dioxo-2,3-dihydro-1H-benzo[de]isoquinolin-6-yl) benzaldehyde (NI-CHO) with AIE-active tetraphenylethylene (TPE) to achieve the transition of NI-CHO from aggregation-caused quenching (ACQ) to an AIE molecule. When TPENI was in the aggregated state, the luminescence intensity was significantly enhanced due to the TPE structural unit restricting the free rotation of the intramolecular benzene ring, as well as the π-π stacking interactions of the molecules, which was conducive to the preparation of TPENI NPs as ECL materials. Satisfactorily, we found that the ECL intensity of TPENI NPs was increased by about 4.8-fold compared with that of the molecules dispersed in organic solution, and the stability reached about 1000 s. Based on the excellent ECL properties of TPENI NPs, an "on-off-on" ECL biosensor with a wider detection range (1 fg/mL to 100 ng/mL) and a lower detection limit of 0.20 fg/mL (S/N = 3) was proposed for sensitive analysis of a carcinoembryonic antigen (CEA). Overall, this work provided a new approach to the realization of AIECL and laid the foundation for the application of naphthalimide derivatives in ECL.
Proximity hybridization-regulated chemiluminescence resonance energy transfer for homogeneous immunoassay.

Abstract:Chemiluminescence resonance energy transfer (CRET) and the proximity ligation assay have been widely used in design of sensors for the bioanalysis. Here, a wash-free and homogeneous strategy was proposed to detect carcino-embryonic antigen (CEA) based on proximity hybridization-regulated CRET. The Cy5 demonstrated strong chemiluminescence (CL) via the oxidation of TCPO in the presence of H2O2 and energy transfer between excited TCPO and Cy5. Graphene oxide (GO) as an excellent quencher was used to produce the “Signal off” mode that little CL emission was observed through CRET between GO and the Cy5-labelled DNA3. Once CEA was introduced, the target-induced proximity hybridization occurred to form a proximate complex, which inhibited the CRET by preventing GO from absorbing Cy5-labelled DNA3. Furthermore, taking advantage of nicking endonuclease Nt.BbvCI for in situ recycling, the signal could be further amplified for highly sensitive CL detection. Our results showed that this strategy enabled a specific response to CEA with a detection range of 5 orders of magnitude, along with a detection limit of 3.2pg mL−1. Apart from its easy operation, high sensitivity and acceptable accuracy, the proposed method needed only 0.3μL of sample, indicating its great opportunity for commercial application.
A bimetallic metal-organic framework with high enzyme-mimicking activity for an integrated electrochemical immunoassay of carcinoembryonic antigen

Abstract:Metal-organic frameworks (MOFs) show excellent catalytic activity and have been widely applied in diagnosis of diseases and tumors. However, current assay methods usually involve cumbersome configurations and complicated procedures, which inhibit their practical applications. Hence, a Cu-Ni MOF/carbon printed electrode (CPE)-based integrated electrochemical immunosensor was constructed for highly sensitive and efficient determination of carcinoembryonic antigen (CEA). First, highly conductive carbon ink was screen-printed onto a polyethylene terephthalate substrate to manufacture a CPE. Afterward, an aminated Cu-Ni MOF was prepared by a typical solvothermal strategy and modified on the CPE. Owing to its excellent peroxidase activity, the Cu-Ni MOF can catalytically oxidize hydroquinone using hydrogen peroxide, which greatly amplifies the peak current signal. Then the formation of an immune complex inhibited the catalytic activity of the MOF, thus enabling the quantitative determination of CEA content with a wide linear range of 0.5 pg mL-1-500 ng mL-1 and a low detection limit of 0.16 pg mL-1. Furthermore, the Cu-Ni MOF/CPE-based integrated portable electrochemical immunosensor also showed satisfactory performance in the detection of CEA in clinical serum samples with excellent accuracy, showing great potential for application in point-of-care disease diagnosis.

Abstract:Herein, an innovative sandwich-type electrochemical immunosensor based on multi-order Rubik's cube-type platinum nickel nanocubes and Au NPs/cPDA NTs was used for the ultrasensitive detection of carcinoembryonic antigen (CEA). In this process, an appropriate amount of uniformly dispersed gold nanoparticles (Au NPs) was effectively immobilized on nitrogen riched carbonized polydopamine nanotubes (cPDA NTs) through Au-N bonds, avoiding the stacking and agglomeration of Au NPs, thereby enhancing the conductivity. Subsequently, multi-order Rubik's cube-type platinum nickel nanocubes (PtNi NCs), composed of numerous small interlaced nanocubes, was successfully prepared to immobilize the secondary antibodies (Ab 2 ) effectively. Due to its inherent hierarchical architectures and high density of active sites, PtNi NCs exhibited excellent mass and electron transfer performance towards the reduction of hydrogen peroxide (H 2 O 2 ) for amplifying the current signal to detect CEA. The rationally designed immunosensor presented a broad linear ranging from 50 fg mL -1 to 100 ng mL -1 and a low limit of detection of 17.78 fg mL -1 (S/N = 3) for CEA determination under optimal conditions. Furthermore, the as-fabricated immunosensor exhibited excellent selectivity, reproducibility and stability, which can be extended to accurate and sensitive detection of other biomarkers.

Abstract:A facile label-free electrochemiluminescence (ECL) aptasensor, based on the ECL of cadmium sulfidegraphene (CdS-GR) nanocomposites with peroxydisulfate as the coreactant, was designed for the detection of carcinoembryonic antigen (CEA). Tripetalous CdS-GR nanocomposites were synthesized through a simple onepot solvothermal method and immobilized on the glassy carbon electrode surface. L-Cystine (L-cys) could largely promote the electron transfer and enhance the ECL intensity. Gold nanoparticles (AuNPs) were assembled onto the L-cys film modified electrode for aptamer immobilization and ECL signal amplification. The aptamer modified with thiol was adsorbed onto the surface of the AuNPs through a Au-S bond. Upon hybridization of the aptamer with the target protein, the sequence could conjugate CEA to form a Y architecture. With CEA as a model analyte, the decreased ECL intensity is proportional to the CEA concentration in the range of 0.01-10.0 ng mL(-1) with a detection limit of 3.8 pg mL(-1) (SIN = 3). The prepared aptasensor was applied to the determination of CEA in human serum samples. The recoveries of CEA in the human serum samples were between 85.0% and 109.5%, and the RSD values were no more than 3.4%.

Abstract:For the diagnosis and therapy of small cell lung cancer (SCLC), the accurate and sensitive determination of neuron-specific enolase (NSE) content is crucial. This work outlines a dual-quenching electrochemiluminescence resonance energy transfer (ECL-RET) immunosensor based on the double quenching effects of iron base metal organic frameworks (FeMOFs) loaded with small sized CuO nanoparticles (FeMOFs-sCuO) towards CoPd nano -particles (CoPdNPs) enhanced porous g-C3N4 (P-C3N4-CoPdNPs). To be specific, we prepared a porous g-C3N4 (P-C3N4) which has a rich porous structure, and significantly increased the specific surface area and the number of reaction sites of P-C3N4. Meanwhile, the CoPdNPs were loaded onto P-C3N4 to improve the ECL luminescence property of P-C3N4/K2S2O8 system through acting as a coreaction accelerator. In addition, the ultraviolet-visible (UV-vis) absorption spectra of FeMOFs and small sized CuO nanoparticles (sCuO) showed considerable overlap with the ECL emission spectra of P-C3N4 appropriately. Therefore, FeMOFs with high specific surface area were prepared and well combined with sCuO to effectively dual-quenching the ECL emission of P-C3N4 based on resonance energy transfer. Hence, a new type ECL-RET couple made up of P-C3N4-CoPdNPs (donor) and FeMOFs-sCuO (acceptor) were developed for the first time. A certain amount of P-C3N4-CoPdNPs, Ab1, BSA, NSE were modified layer by layer onto the electrode surface. Then FeMOFs-sCuO-Ab2 bioconjugates was incubated through the immune recognition binding. As a result, a sandwich-type ECL biosensor was manufactured suc-cessfully for NSE immunoassay. Under optimal experimental conditions, the limit of detection (LOD) and the limit of quantitation (LOQ) of the prepared ECL sensor for NSE analysis was 20.4 fg mL-1 and 7.99 fg mL-1, respectively, with the relative standard deviation (RSD) of 1.68%. The linear detection range was 0.0000500-100 ng mL-1. The studied immunosensor had satisfactory sensitivity, specificity and reproducibility, manifesting the suggested sensing strategy might offer a good technical means and theoretical basis for the sensitivity analysis of NSE and has a potential application in clinical diagnosis analysis.
