Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.
Quantitative analysis and detection of adulteration in pork using near-infrared spectroscopy

Abstract:Authenticity is an important food quality criterion. Rapid methods for confirming authenticity or detecting adulteration are increasingly demanded by food processors and consumers. Near infrared (NIR) spectroscopy has been used to detect economic adulteration in pork. Pork samples were adulterated with liver and chicken in 10% increments. Prediction and quantitative analysis were done using raw data and pretreatment spectra. The optimal prediction result was achieved by partial least aquares(PLS) regression with standard normal variate(SNV) pretreatment for pork adulterated with liver samples, and the correlation coefficient(R value), the root mean square error of calibration(RMSEC) and the root mean square error of prediction (RMSEP) were 0.97706, 0.0673 and 0.0732, respectively. The best model for pork meat adulterated with chicken samples was obtained by PLS with the raw spectra, and the correlation coefficient(R value), RMSEP and RMSEC were 0.98614, 0.0525, and 0.122, respectively. The result shows that NIR technology can be successfully used to detect adulteration in pork meat adulterated with liver and chicken.
A Study on the Use of Near-Infrared Spectroscopy for the Rapid Quantification of Major Compounds in Tanreqing Injection

Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.
NIR spectroscopy-multivariate analysis for rapid authentication, detection and quantification of common plant adulterants in saffron (Crocus sativus L.) stigmas

Abstract:The presented work discusses the development of a rapid and precise analytical protocol using near infrared spectroscopy combined with multivariate data analysis to authenticate, detect and quantify most of the commonly encountered plant adulterants used in fraud of saffron stigmas including safflower, pomegranate fruit peel, calendula flower, paprika, curcuma, hibiscus, saffron stamens and exhaustively-extracted saffron stigmas. A Soft Independent Modelling of Class Analogies (SIMCA) model was constructed for authentication of saffron stigmas with 100% sensitivity and a Partial Least Squares-Discriminant Analysis (PLS-DA) model was successfully utilized for correct discrimination of unadulterated and intentionally adulterated saffron samples as it showed 100% sensitivity and 99% specificity. Quantitation of the amount of each individual adulterant was achieved through construction of partial least squares regression (PLSR) models accompanied by variable importance to projection (VIP) method for variable selection which revealed that bands in the spectral ranges 6000-5800 cm−1 followed by 4600–4200 cm−1 and 5400-5000 cm−1 were the most important for correct prediction with detection limits as low as 1%. The models performance was tested using internal and external validation sets indicating their reliability in providing a useful quality assessment tool for saffron in an attempt to prevent its fraud.

Abstract:Carcinogenic Sudan I has been added illegally into spices for an apparent freshness. 1 H solution and solid-state (SS) nuclear magnetic resonance (NMR) spectroscopies were applied and compared for determination of Sudan I in paprika powders (PPs). For solution NMR, PPs spiked with Sudan I were extracted with acetonitrile, centrifuged, rotor-evaporated, and re-dissolved in DMSO-d6 for spectral collection. For SSNMR, Sudan I contaminated PPs were mixed with DMSO-d6 solution and used for spectral collection. Linear regression models constructed for quantitative analyses resulted in the average accuracies for unknown samples as 98% and 105%, respectively. Limits of detection for the solution NMR and SSNMR spectrometers were 6.7 and 128.6 mg kg −1 , while the limits of quantification were 22.5 and 313.7 mg kg −1 . The overall analysis time required by both methods was similar (35 and 32 min). Both NMR techniques are feasible for rapid and accurate determination of Sudan I adulteration in PPs.

Abstract:Pericarpium citri reticulatae (PCR) has been used as a food and spice for many years and is known for its rich nutritional content and unique aroma. However, price increases are often accompanied by adulteration. In this study, two kinds of adulterants (Orange peel-OP and Mandarin Rind-MR) were identified by chromaticity analysis, FT-NIR and machine learning algorithm, and the doping concentration was predicted quantitatively. The results show that colorimetric analysis cannot completely differentiate between PCR and adulterants. Using spectral preprocessing combined with machine learning algorithms, PCR and two adulterants were successfully distinguished, with classification accuracy reaching 99.30 % and 98.64 % respectively. After selecting characteristic wavelengths, the R2 P of the adulterated quantitative model is greater than 0.99. Generally, this study proposes to use FT-NIR to study the adulteration of PCR for the first time, which fills the technical gap in the adulteration research of PCR, and provides an important method to solve the increasingly serious adulteration problem of PCR.

Abstract:Shrimp is a common seafood that is often processed into culinary powder by using only the abdomen of the shrimp. Some producers are, however, suspected to produce the powder using immature shrimps which are illegal to fish, while some include the head regardless of standard procedures that the thorax (head) and other parts may contain sharp stones and objects that can be hazardous upon consumption. Methods for detecting such adulteration are technical, expensive, and time-consuming. Near infrared spectroscopy (NIRS) is a rapid analytical tool that can be explored for such analysis. This study aimed to develop models for detecting shrimp powder adulteration using a handheld spectrophotometer in tandem chemometrics and multiple spectral preprocessing. Using linear discriminant analysis, shrimp powder adulterated with milled immature shrimps at 0, 5, 10, 20, 30, 40, 50, and 100% w/w could be classified with an average cross-validation accuracy of 93.10%, while a higher average cross-validation accuracy of 98.63% was obtained for classifying milled shrimp head in shrimp powder. The models confirmed suspected fraudulent activities of shrimp powder adulteration in some Ghanaian markets. Using partial least squares regression and multiple spectral preprocessing, authentic shrimp powder was predicted with R 2 CV's up to 0.996, RMSECV's lower than 1.25 g/100 g and RPD lower than 3.1. The handheld spectrophotometer brings an added advantage of remote analysis for adaptation of the developed models.

Abstract:Food adulteration imposes a significant health concern on the community. Being one of the key ingredients used for spicing up food dishes. Red chilli powder is almost used in every household in the world. It is also common to find chilli powder adulterated with brick powder in global markets. We are amongst the first research attempts to train a machine learning-based algorithms to detect the adulteration in red chilli powder. We introduce our dataset, which contains high quality images of red chilli powder adulterated with red brick powder at different proportions. It contains 12 classes consists of 0%, 5%, 10%, 15%, 20%, 25%, 30%, 35%, 40%, 45%, 50%, and 100% adulterant. We applied various image color space filters (RGB, HSV, Lab, and YCbCr). Also, extracted features using mean and histogram feature extraction techniques. We report the comparison of various classification and regression models to classify the adulteration class and to predict the percentage of adulteration in an image, respectively. We found that for classification, the Cat Boost classifier with HSV color space histogram features and for regression, the Extra Tree regressor with Lab color space histogram features have shown the best performance.

Abstract:Near infrared spectroscopy combined with chemometrics was investigated for the fast determination of total organic carbon (TOC) and soluble solids contents (SSC) of Tanreqing injection intermediates. The NIR spectra were collected in transflective mode, and the TOC and SSC reference values were determined with Multi N/C UV HS analyzer and loss on drying method. The samples were divided into calibration sets and validation sets using the Kennard–Stone (KS) algorithm. The Dixon test, leverage and studentized residual test were studied for the sample outlier analysis. The selection of wavebands, spectra pretreated method and the number of latent variables were optimized to obtain better results. The quantitative calibration models were established with 3 different PLS regression algorithms, named linear PLS, non-linear PLS and concentration weighted PLS, and the net result was defined as the average of the predicted values of the different calibration models. The overall results indicated that the presented method is more powerful than single multivariable regression method, characterized by higher mean recovery rate (MRR) of the validation set, and can be used for the rapid determination of TOC and SSC values of Tanreqing injection intermediates, which are two important quality indicators for the process monitoring.

Abstract:Food adulteration involving the illegal addition of dyes to foodstuffs has become an alarming issue in recent years. This study developed and validated a high-performance liquid chromatography (HPLC)-DAD (diode array detector) method for the simultaneous determination of nine azo dyes (Butter Yellow, Sudan Orange G, Para Red, Sudan I, Sudan II, Sudan III, Sudan IV, Sudan Red 7B, and Scarlet 808). Moreover, a qualitative analysis method using liquid chromatography-tandem mass spectrometry was developed to more accurately identify peaks detected in HPLC-DAD. The calibration curve represented good linearity (r2 ≥ 0.9998) over the measured concentration range of 0.5-25 mg/kg. limit of detection and limit of quantification were 0.01-0.04 and 0.04-0.12 mg/kg, respectively. Accuracy and precision were 96.0-102.6 and 0.16-2.01 (relative standard deviation%), respectively. Additionally, the measurement uncertainty and HorRat value were estimated. Several Curcuma longa L. distributed in Korea were collected and monitored for azo dye contaminants. PRACTICAL APPLICATION: The proposed HPLC-DAD method represents a significant advancement in the field, offering a reliable means of quantifying azo dyes and identifying their presence even at trace levels in adulterated turmeric. This not only contributes to ensuring the safety and integrity of turmeric products but also establishes precedent for robust analytical techniques in addressing food safety challenges.

Abstract:Food adulteration occurs globally, in many facets, and affects almost all food commodities. Adulteration is not just a crucial economic problem, but it may also lead to serious health problems for consumers. Turmeric (Curcuma longa) is a world-class spice commonly contaminated with various chemicals and colors. It has also been used extensively in many Asian curries, sauces, and medications. Different traditional approaches, such as chemical and physical methods, are available for detecting adulterants in turmeric. These approaches are rather time-consuming and inaccurate methods. Therefore, it is of utmost importance to identify the adulterants in turmeric accurately and instantly. A cloud-based system was developed to detect adulteration in adulterated turmeric. The dataset consists of spectral images of turmeric with tartrazine-colored rice flour adulterant. Adulterants in weight percentages of 0%, 5%, 10%, and 15% were mixed with turmeric. A convolutional neural network (CNN) was implemented to detect adulteration, which achieved 100% accuracy for training and 94.35% accuracy for validation. The deep CNN (DCNN) models, namely, VGG16, DenseNet201, and MobileNet, were implemented to detect adulteration. The proposed CNN model outperforms DCNN models in terms of accuracy and layers. The CNN model is deployed to the platform as a service (PaaS) cloud. The deployed model link can be accessed using a smartphone. Uploading the adulterated turmeric image to a cloud link can analyze and detect adulteration.

Abstract:Turmeric essential oil (TEO), derived from Turmeric (Curcuma longa L.) herb, is one of several natural products which has seen a rise in demand in recent years. This rising demand increases the tendency for TEO to be adulterated with inferior or less expensive alternatives, such as vegetable oils and synthetic compounds – a practice that has significant economic and organoleptic consequences. Adulteration of highly valued essential oils such as TEOs with other inexpensive vegetable oils, especially at very low concentrations poses a significant challenge as the latter has no apparent effect on the gas chromatographic profile of the adulterated TEOs. Therefore, the overall goal of this study was to develop a facile, convenient, affordable, rapid, and direct analytical method for the authentication of TEO using Fourier transform infrared spectroscopic and chemometric methods, namely, partial least squares (PLS) and principal component regression (PCR). To accomplish this goal, TEO was first adulterated with sunflower (SFO) and soybean (SBO) oil in varying concentrations. For the PLS model, the best predictive performance in both the training and testing sets was observed at 3, 2 and 4 components for TEO, SFO and SBO, respectively. Further, the PLS model was more accurate than the PCR in the prediction of TEO in the training, testing, as well as unknown sets, with minimum root mean square error values of 1.2% and maximum R2=0.999. The developed PLS model could potentially be applied for the rapid and facile detection of vegetable oil adulterants in TEO, a valuable approach to assess the authenticity of such a highly valued commodity.

Abstract:This study aimed to evaluate the potential of near infrared spectroscopy (NIRS) as a fast and non-destructive tool for detecting and quantifying different adulterants in fresh and frozen-thawed minced beef. Partial least squares regression (PLSR) models were built under cross validation and tested with different independent data sets, yielding determination coefficients (R(P)(2)) of 0.96, 0.94 and 0.95 with standard error of prediction (SEP) of 5.39, 5.12 and 2.08% (w/w) for minced beef adulterated by pork, fat trimming and offal, respectively. The performance of the developed models declined when the samples were in a frozen-thawed condition, yielding R(P)(2) of 0.93, 0.82 and 0.95 with simultaneous augments in the SEP of 7.11, 9.10 and 2.38% (w/w), respectively. Linear discriminant analysis (LDA), partial least squares-discriminant analysis (PLS-DA) and non-linear regression models (logistic, probit and exponential regression) were developed at the most relevant wavelengths to discriminate between the pure (unadulterated) and adulterated minced beef. The classification accuracy resulting from both types of models was quite high, especially the LDA, PLS-DA and exponential regression models which yielded 100% accuracy. The current study demonstrated that the VIS-NIR spectroscopy can be utilized securely to detect and quantify the amount of adulterants added to the minced beef with acceptable precision and accuracy.

Abstract:Saffron, one of the most expensive spices around the world, is highly vulnerable to economic adulteration in its powder form. Plant materials and artificial colorants are commonly used as adulterants to reduce saffron concentration as well as maintaining color strength in powdered saffron products. However, current analytical methods lack the ability to test samples accurately onsite. Therefore, the objective of this study is to develop a fast screening and quantification method combining thin-layer chromatography (TLC) and Raman spectroscopy for saffron powder analysis, including quantifying the main colorant crocin content and identifying possible adulterants out of a lab setting. A droplet of aqueous extract of saffron powder was deposited on a TLC plate, exhibiting a uniform yellow round pattern on the TLC plate. The content of crocin was analyzed using the ISO 3632 method and Raman spectroscopy separately. A quantitative model based on Raman spectra was established and validated using partial least square (PLS) with R of 0.9955 and 0.9929 for calibration and validation of the PLS plot, respectively. Possible adulterants such as powders of safflower, turmeric powder, red 40 (allura red), and yellow 5 (tartrazine) were mixed with saffron powder at various degrees. Their water extracts exhibited unique patterns under bright ambient light and UV light that can be discriminated from that of pure saffron samples through visual observation and principal component analysis (PCA) based on the L*a*b* values. We also estimated the lowest adulteration degrees that can be detected were 2.59% for Allura red, 4.15% for tartrazine, 31.01% for safflower, and 41.98% turmeric, respectively, using the PLS analysis. Further identification was evaluated using Raman spectroscopy, which provided fingerprint spectra for all adulterants except safflower. Combining TCL pattern and Raman spectroscopy, samples with identical optical properties to pure saffron but adulterated with multiple adulterants were analyzed and successfully detected. This study demonstrates the potential practical application of this method in rapid analysis of saffron quality and adulteration.

Abstract:The rising demand for cocoa powder has resulted in an upsurge in market prices, leading to the emergence of adulteration practices aimed at achieving economic benefits. This study aimed to detect and quantify cocoa powder adulteration using visible and near-infrared spectroscopy (Vis-NIRS). The adulterants used in this study were powdered carob, cocoa shell, foxtail millet, soybean, and whole wheat. The NIRS data could not be resolved using Savitzky-Golay smoothing. Nevertheless, the application of a random forest and support vector machine successfully classified the samples with 100% accuracy. Quantification of adulteration using partial least squares (PLS), Lasso, Ridge, elastic Net, and RF regressions provided R2 higher than 0.96 and root mean square error <2.6. Coupling PLS with the Boruta algorithm produced the most reliable regression model (R2 = 1, RMSE = 0.0000). Finally, an online application was prepared to facilitate the determination of adulterants in the cocoa powder.
Evaluating the Use of Vibrational Spectroscopy to Detect the Level of Adulteration of Cricket Powder in Plant Flours: The Effect of the Matrix

Abstract:Edible insects have been recognised as an alternative food or feed ingredient due to their protein value for both humans and domestic animals. The objective of this study was to evaluate the ability of both near- (NIR) and mid-infrared (MIR) spectroscopy to identify and quantify the level of adulteration of cricket powder added into two plant proteins: chickpea and flaxseed meal flour. Cricket flour (CKF) was added to either commercial chickpea (CPF) or flaxseed meal flour (FxMF) at different ratios of 95:5% w/w, 90:10% w/w, 85:15% w/w, 80:20% w/w, 75:25% w/w, 70:30% w/w, 65:35% w/w, 60:40% w/w, or 50:50% w/w. The mixture samples were analysed using an attenuated total reflectance (ATR) MIR instrument and a Fourier transform (FT) NIR instrument. The partial least squares (PLS) cross-validation statistics based on the MIR spectra showed that the coefficient of determination (R2CV) and the standard error in cross-validation (SECV) were 0.94 and 6.68%, 0.91 and 8.04%, and 0.92 and 4.33% for the ALL, CPF vs. CKF, and FxMF vs. CKF mixtures, respectively. The results based on NIR showed that the cross-validation statistics R2CV and SECV were 0.95 and 3.16%, 0.98 and 1.74%, and 0.94 and 3.27% using all the samples analyzed together (ALL), the CPF vs. CKF mixture, and the FxMF vs. CKF mixture, respectively. The results of this study showed the effect of the matrix (type of flour) on the PLS-DA data in both the classification results and the PLS loadings used by the models. The different combination of flours (mixtures) showed differences in the absorbance values at specific wavenumbers in the NIR range that can be used to classify the presence of CKF. Research in this field is valuable in advancing the application of vibrational spectroscopy as routine tools in food analysis and quality control.
