Effect of Leachate Recycle and Inoculation on Microbial Characteristics of Municipal Refuse in Landfill Bioreactors

Abstract:Activity development of key groups of enzymes involved in municipal refuse decomposition was measured in laboratory landfill bioreactors with and without leachate recycling and inoculation for about 210 days. The results showed that the enzymes (amylase, protease, cellulase, lipase and pectinase) were present in fresh refuse but at low values and positively affected by leachate recycling and refuse inoculation. The total average of cellulase activity in digesters D3 operated with leachate recycling but no inoculation, D4 and D5 operated with leachate recycling and inoculation was much higher than that in digesters D1 and D2 without leachate recycling and inoculation by 88%-127%, 117%-162% and 64%-98%. The total average of protease activity was higher in digester D4 than that in digesters D1, D2, D3 and D5 by 63%, 39%, 24% and 24%, respectively, and the positive effect of leachate recycling and inoculation on protease activity of landfilled refuse mainly was at the first two months. The total average of amylase activity was higher in digesters D3, D4 and D5 than that in digesters D1 and D2 by 83%-132%, 96%-148% and 81%-129%. During the early phase of incubation, the stimulatory effect of inoculation on lipase activity was measured, but refuse moisture was the main factor affecting lipase activity of landfilled refuse. The inoculation, initial and continuous inoculation of microorganisms existing in leachate, was the mainly stimulatory factor affecting pectinase activity of landfilled refuse.
Effect of Inoculation with Effective Microorganisms and Leachate Recycle on Degradation of Municipal Refuse

Abstract:Biodegradation of waste in landfill is a slow process requiring decades for completion. Accelerated degradation of municipal refuse in modulated landfill environments may alleviate or eliminate pollution to the land, water and air. In this work, nineteen effective microorganisms (EMs) were isolated from old landfill refuse by enrichment culturing techniques and used for the inoculum of municipal refuse. The preliminary experiments demonstrate that a combination of EMs inoculation in landfill with leachate recycle resulted in increased rates of decomposition and faster process stability. The concentrations of COD, VFA and SO42- in digester with EMs inoculation and leachate recycle decreased more rapidly than others. Gas production from digester with EMs inoculation and leachate recycle commenced around 32 days, which is a week shorter than with leachate recycle only. And peak cumulative gas production was obtained much earlier in digester with EMs inoculation and leachate recycle (150 days) compared to 180 days with leachate recycle only. Moreover, in the first two months, the rate of settlement in digester with EMs inoculation and leachate recycle was more rapid than others.

Abstract:China generates over 100 million tons of food waste annually, leading to significant environmental pollution and health risks if not managed properly. Converting FW into a high-value-added platform molecule, lactic acid (LA), through fermentation offers a promising approach for both waste treatment and resource recovery. This paper presents a comprehensive review of recent advancements in LA production from FW, focusing on pure strains fermentation and open fermentation technologies, metabolic mechanisms, and problems in fermentation. It also assesses purification methods, including molecular distillation, adsorption, membrane separation, precipitation, esterification and hydrolysis, solvent extraction, and in-situ separation, analyzing their efficiency, advantages, and disadvantages. However, current research encounters several challenges, including low LA yield, low optical purity of L-(+)-LA, and difficulties in the separation and purification of LA. The integration of in-situ separation technology coupled with multiple separation methods is highlighted as a promising direction for future advancements.

Abstract:An annual production of about 500 million tons of household food waste (HFW) has been documented, resulting in significant implications for human health and the environment in the absence of appropriate treatment. The anaerobic fermentation of HFW in an open system offers the potential to recover high value-added products, lactic acid (LA), thereby simultaneously addressing waste treatment and enhancing resource recovery efficiency. Most of LA fermentation studies have been conducted under mesophilic and thermophilic conditions, with limited research on the production of LA through anaerobic fermentation under hyperthermophilic conditions. This study aimed to produce LA through anaerobic fermentation from HFW under hyperthermophilic conditions (70 ± 1 °C), while varying pH values (5.0 ± 0.1, 7.0 ± 0.1, and 9.0 ± 0.1), and compare the results with LA production under mesophilic (35 ± 1 °C) and thermophilic (52 ± 1 °C) conditions. The findings of this study indicated that the combination of hyperthermophilic conditions and a neutral pH (pH7_70) yielded the highest concentration of LA, measuring at 17.75 ± 1.51 g/L. The mechanism underlying the high yield of LA at 70 °C was elucidated through the combined analysis of organics dissolution, enzymes activities, and 16S rRNA microbiome sequencing.

Abstract:SIGNIFICANCE Mechanosignaling is vital for maintaining the structural integrity of bone under physiologic conditions. These signals activate and suppress multiple signaling cascades regulating bone formation and resorption. Understanding these pathways is of prime importance to exploit their therapeutic potential in disorders associated with bone loss due to disuse, trauma, or disruption of homeostatic mechanisms. RECENT ADVANCES In the case of cells of the bone, an impressive amount of data has been generated that provides evidence of a complex mechanism by which mechanical signals can maintain or disrupt cellular homeostasis by driving transcriptional regulation of growth factors, matrix proteins and inflammatory mediators in health and inflammation. Mechanical signals act on cells in a magnitude dependent manner to induce bone deposition or resorption. During health, physiological levels of these signals are essential for maintaining bone strength and architecture, whereas during inflammation, similar signals can curb inflammation by suppressing the nuclear factor kappa B (NF-κB) signaling cascade, while upregulating matrix synthesis via mothers against decapentaplegic homolog and/or Wnt signaling cascades. Contrarily, excessive mechanical forces can induce inflammation via activation of the NF-κB signaling cascade. CRITICAL ISSUES Given the osteogenic potential of mechanical signals, it is imperative to exploit their therapeutic efficacy for the treatment of bone disorders. Here we review select signaling pathways and mediators stimulated by mechanical signals to modulate the strength and integrity of the bone. FUTURE DIRECTIONS Understanding the mechanisms of mechanotransduction and its effects on bone lay the groundwork for development of nonpharmacologic mechanostimulatory approaches for osteodegenerative diseases and optimal bone health.

Abstract:It is promising to recover lactic acid (LA) from fermentation of food waste (FW). In this study, pH and temperatures were investigated comprehensively to find their effects on LA fermentation, and microbial analyses were used to take insight to the variation of LA production. The results showed that mesophilic fermentation benefited hydrolysis and acidification, leading to a high yield of LA, while thermophilic conditions restricted other producers at low pH, leading to a high purity of LA. Lactobacillus amylolyticus was the main LA producer under thermophilic conditions, but Thermoanaerobacterium thermosaccharolyticum boomed at pH 5.0-6.0 and it converted LA partly to butyric acid. Simultaneously, Bacillus coagulans also increased and improved the optical purity (OP) of L-LA. From a series of this study, an operational condition of pH 5.5 and temperature of 52 °C would be potentially suitable for lactate fermentation of FW with high purity of 89%, while a stable LA production with an OP of 68% was achieved at 55 °C and pH 6.0.

Abstract:Lactic acid production from food waste via fermentation is environmentally sustainable. However, the characteristics of food waste fermentation to produce lactic acid are not well understood due to the complexity of food waste. This study aims to understand the effects of key variables on the characteristics of food waste fermentation to maximize lactic acid production. Food waste was enzymatically hydrolyzed and fermented by Lactobacillus pentosus. Key fermentation variables, including nitrogenous nutrient supplementation, initial sugar concentration, and pH, were investigated in batch fermentation to unveil their effects on fermentation titer, yield, and productivity. The results showed that supplementation of 0.25% (w/v%) yeast extract and peptone to the food waste fermentation media significantly improved fermentation titer and productivity, but further increase in the supplementation level did not improve fermentation. Increasing the initial sugar concentration from 40 g/L to 100 g/L increased the fermentation titer from 41.0 g/L to 93.0 g/L and productivity from 0.34 g/L/h to 0.76 g/L/h. pH 6.0 was the optimal pH for the fermentation. At the optimal conditions, food waste fermentation resulted in the highest fermentation titer, yield, and productivity of 106.7 g/L, 1.12 g/g, and 3.09 g/L/h, respectively. The high fermentation yield of 1.12 g/g might be explained by the extra lactic acid production from unidentified compounds in food waste hydrolysates. By applying fed-batch fermentation, the lactic acid concentration reached 157.0 g/L with a yield and overall productivity of 0.92 g/g and 2.0 g/L/h, respectively. Based on the mass balance, a total of 251 kg lactic acid was produced from 1000 kg food waste. Practical Application Food waste is one of the largest municipal solid wastes in the US, and most food waste ends up in landfills, causing significant economic losses and environmental concerns. In this study, we developed a fermentation process to convert food waste into biorenewable lactic acid and demonstrated that food waste is a superior feedstock for fermentation due to its embedded nutrients. Moreover, due to the embedded nutrients in food waste, the supplementation of yeast extract and peptone to fermentation can be reduced by over 50%, which can reduce the operating cost of lactic acid fermentation on an industrial scale.

Abstract:In this study, the feasibility of promoting the lactic acid (LA) fermentation of food waste (FW) with iron tailings (ITs) addition was explored. The best LA yield was 0.91 g LA/g total sugar when 1 % ITs were added into the system. The mechanisms for promoting LA production were acidification alleviation effects and reduction equivalent supply of ITs. Furthermore, the addition of ITs promoted carbohydrate hydrolysis, and the carbohydrates digestibility reached 88.85 % in the 1 % ITs group. The ITs also affected the microbial communities, Lactococcus gradually replaced Streptococcus as the dominant genus, and results suggested that Lactococcus had a positive correlation with LA production and carbohydrate digestibility. Finally, the complex LAB in FW had significant effects on heavy metal removal from ITs, and the removal efficiency Cr, As, Pb, Cd, and Hg can reach 50.84 %, 26.72 %, 59.65 %, 49.75 % and 78.87 % in the 1 % ITs group, respectively.

Abstract:Medium-chain fatty acids (MCFAs) have drawn great attention due to their high energy density and superior hydrophobicity. Waste activated sludge (WAS) has been documented as a renewable feedstock for MCFAs production via anaerobic fermentation. However, MCFAs production from WAS depends on exogenous addition of electron donor (ED, e.g., lactate) for chain elongation (CE) bioprocess, which results in increased economic cost and limited practical application. In this study, a novel biotechnology was proposed to produce MCFAs from WAS with in-situ self-formed lactate by inoculating Yoghurt starter powder containing with Lactobacillales cultures. The batch experimental results revealed that the lactate was in-situ generated from WAS and the maximum production of MCFAs increased from 1.17 to 3.99 g COD/L with the increased addition of Lactobacillales cultures from 6✕10 7 to 2.3✕10 8 CFU/mL WAS. In continuous long-term test over 97 days, average MCFA production reached up to 3.94 g COD/L with a caproate yield of 82.74% at sludge retention time (SRT) 12 days, and the average MCFA production increased to 5.87 g COD/L with 69.28% caproate and 25.18% caprylate at SRT 15 days. A comprehensive analysis of the metagenome and metatranscriptome demonstrated that the genus of Lactobacillus and Streptococcus were capable of producing lactate from WAS and upgrading to MCFAs. Moreover, another genus, i.e., Candidatus Promineofilum , was firstly revealed that it might be responsible for lactate and MCFAs production. Further investigation of related microbial pathways and enzyme expression suggested that D-lactate dehydrogenase and pyruvate ferredoxin oxidoreductase contributed to lactate and acetyl-CoA production, which were the crucial steps for MCFAs generation and were most actively expressed. This study provides a conceptual framework of MCFAs from WAS with endogenous ED, potentially enhancing the energy recovery from WAS treatment.

Abstract:It is a promising method to recover lactic acid from food waste (FW) fermentation, but the bottleneck problem is the low yield when using mixed inoculation. In this study, laboratorial biogas slurry (LBS) and industrial biogas slurry (IBS) were used as the additive in semi-continuous FW fermentation, aiming to promote the production of lactic acid. According to the research results, the addition of LBS or IBS promoted the production of lactic acid significantly from FW, especially carbohydrate, because it increased the pH values, maintained low OPR levels, and increased microbial number and diversity in the fermentation systems. IBS performed better than LBS because of higher pH, more diverse microbial community and more functional microorganisms. The best ratio of IBS to feedstock was 0.2, and the lactic acid yield reached 0.42 g/gVSadded. An excessively high dose would alter the fermentation pathways, reduce the ratio of lactic acid.

Abstract:The anaerobic co-digestion of food waste and young landfill leachate was carried out in a sequencing batch reactor. Results showed that the anaerobic co-digestion system could enter the methanogenic stage without adding other inoculum, and while the system stopped producing gas, adding activated sludge as inoculum could restart the anaerobic co-digestion process. Specially, for that anaerobic co-digestion system with acid inhibition, the addition of activated sludge will help the system enter gas-production stage which may be ascribed to it consumed the acetic acid in the system. With this novel process, the dissolved organic carbon after anaerobic co-digestion was 20.5% of that of the leachate (1915 mg/L), while the proportion of humic substances in dissolved organic matter was increased to over 90%. This preliminary study indicates that this process would have a prospective application to the high value utilization of food waste and young landfill leachate.Graphical Abstract

Abstract:A unique method to produce highly optically-active l-lactic acid and soil amendments that promote plant growth from food waste was proposed. Three Bacillus strains Bacillus subtilis KBKU21, B. subtilis N3-9 and Bacillus coagulans T27, were used. Strain KBKU21 accumulated 36.9 g/L l-lactic acid with 95.7% optical activity and 98.2% l-lactic acid selectivity when fermented at 43°C for 84 h in a model kitchen refuse (MKR) medium. Residual precipitate fraction (anaerobically-fermented MKR (AFM) compost) analysis revealed 4.60%, 0.70% and 0.75% of nitrogen (as N), phosphorous (as P2O5), and potassium (as K2O), respectively. Additionally, the carbon to nitrogen ratio decreased from 13.3 to 10.6. AFM compost with KBKU21 promoted plant growth parameters, including leaf length, plant height and fresh weight of Brassica rapa (Komatsuna), than that by chemical fertilizers or commercial compost. The concept provides an incentive for the complete recycling of food waste, contributing towards a sustainable production system.

Abstract:Landfill leachate (LL) is endowed with double roles as refractory wastewater and nutrients/energy sources due to inherent vast inorganics and organics. Bioconversion of LL’s nutrients into biohydrogen and volatile fatty acids (VFAs) via eco-friendly dark fermentation (DF) is a promising approach to simultaneously deal with environment deterioration and energy crisis, but its application is severely restricted by poor fermentative performance attributing to strong toxicity of LL and vulnerable vitality of fermentative bacteria. Herein, a novel conversion strategy was proposed by coupling microalgal photosynthesis with DF, which was capable of reclaiming nutrients and organics from LL to produce biohydrogen and VFAs relying on robust microalgae coupled with DF. Results demonstrated that microalgae grew well in 10% LL with maximum biomass concentration of 1.41 g/L. More importantly, 86.12% NH4 + and 53.00% organics were recovered from LL and stored as carbohydrates (26.4%), proteins (48.7%) and lipid (15.9%) in microalgal cells. The accumulated intracellular carbohydrate and protein were then converted into biohydrogen and VFAs via DF, producing 16.37 kJ/L of output energy with overall energy conversion efficiency of 11.76%. Transformations of macromolecular organics and possible conversion mechanism of microalgae biomass to bioenergy were detailed discussed. Together, this work may provide a promising strategy for better dealing with LL disposal.

Abstract:Anaerobic co-fermentation is a favorable way to convert agricultural waste, such as swine manure (SM) and apple waste (AW), into lactic acid (LA) through microbial action. However, the limited hydrolysis of organic matter remains a main challenge in the anaerobic co-fermentation process. Therefore, this work aims to deeply understand the impact of cellulase (C) and protease (P) ratios on LA production during the anaerobic co-fermentation of SM with AW. Results showed that the combined use of cellulase and protease significantly improved the hydrolysis during the enzymatic pretreatment, thus enhancing the LA production in anaerobic acidification. The highest LA reached 41.02 ± 2.09 g/L within 12 days at the ratio of C/P = 1:3, which was approximately 1.26-fold of that in the control. After a C/P = 1:3 pretreatment, a significant SCOD release of 45.34 ± 2.87 g/L was achieved, which was 1.13 times the amount in the control. Moreover, improved LA production was also attributed to the release of large amounts of soluble carbohydrates and proteins with enzymatic pretreated SM and AW. The bacterial community analysis revealed that the hydrolytic bacteria Romboutsia and Clostridium_sensu_stricto_1 were enriched after enzyme pretreatment, and Lactobacillus was the dominant bacteria for LA production. This study provides an eco-friendly technology to enhance hydrolysis by enzymatic pretreatment and improve LA production during anaerobic fermentation.

Abstract:Bioconversion of nutrients and energy from landfill leachate (LL) to biohydrogen and volatile fatty acids (VFAs) using dark fermentation (DF) is a promising technique for developing a sustainable ecosystem. However, poor performance of DF caused by vulnerable fermentative bacteria vitality and strong LL toxicity significantly hinder its commercialization. Herein, an integrated technique linking microalgae photosynthesis and DF was proposed, in which mixed microalgae were applied to robustly reclaim nutrients and chemical oxygen demand (COD) from LL. Then, microalgae biomass was fermented into biohydrogen and VFAs using the DF process. Underlying synergistic mechanisms of the interaction of Scenedesmus obliquus and Chlorella vulgaris resulting from the functioning of extracellular polymeric substances (EPS) were discussed in detail. For better absorption of nutrients from LL, the mixed microalgae secreted obviously more EPS than pure microalgae, which played vital roles in the assimilation of cellular nutrients by forming more negative zeta potential and secreting more tyrosine-/tryptophan-family proteins in EPS. Besides, mixed microalgae produced more intracellular proteins and carbohydrates than the pure microalgae, thereby providing more feedstock for DF and achieving higher energy yield of 10.80 kJ/L than 6.64 kJ/L that was obtained when pure microalgae were used. Moreover, the energy conversion efficiency of 7.75% was higher for mixed microalgae than 4.77% that was obtained for pure microalgae. This work may inspire efficient disposal of LL and production of bioenergy, together with filling the knowledge gaps of synergistic mechanisms of dual microalgal interactions.

Abstract:Fermentation processes have been shown to be a good approach to food waste (FW) management. Among the commodities that can be bioproduced by using FW as an organic substrate and exploiting its biodegradability, there is lactic acid (LA). LA has gained the interest of research because of its role in the production of polylactic acid plastics. In this study, the influence of the HRT (2–5 days) used during the fermentation of the liquid fraction (∼12–13 g COD/L) of FW on LA yield and concentration was investigated. Moreover, the changes in the chemical composition (in terms of carbohydrates and organic metabolites concentration) of the influent occurring in the feeding tank were monitored and its influence on the downstream fermentation process was examined. High instability characterized the reactor run with the optimal production yield obtained on day 129 at an HRT 2 days with 0.81 g COD/g COD. This study shows the importance of the fluctuating composition of FW, a very heterogeneous and biologically active substrate, for the LA fermentation process. The non-steady state fermentation process was directly impacted by the unstable influent and shows that a good FW storage strategy has to be planned to achieve high and constant LA production. Abstract:Introduction: Lactic acid (LA) production from fossil resources is unsustainable owing to their depletion and environmental concerns. Thus, this study aimed to optimize the production of LA by Lactobacillus casei in a cultured medium containing fruit wastes (FWs) from agro-industries and second cheese whey (SCW) from dairy production, supplemented with maize steep liquor (MSL, 10% v/v) as the nitrogen source. Methods: The FWs were selected based on seasonal availability [early summer (early ripening peach), full summer (melon), late summer (pear), and early autumn (apple)] and SCW as annual waste. Small-scale preliminary tests as well as controlled fermenter experiments were performed to demonstrate the potential of using various food wastes as substrates for LA fermentation, except for apple pomace. Results and discussion: A 5-cycle repeated batch fermentation was conducted to optimize waste utilization and production, resulting in a total of 180.56 g/L of LA with a volumetric productivity of 0.88 g/L∙h. Subsequently, mechanical filtration and enzymatic hydrolysis were attempted. The total amount of LA produced in the 5-cycle repeated batch process was 397.1 g/L over 288 h, achieving a volumetric productivity of 1.32 g/L∙h. These findings suggest a promising biorefinery process for low-cost LA production from agri-food wastes.

Abstract:The global market for lactic acid is witnessing growth on the back of increasing applications of lactic acid for manufacturing polylactic acid. Indeed, the lactic acid market is expected to reach 9.8 billion US dollars by 2025. The new concept of meta-fermentation has been proposed in recent years as an alternative to fermentation with pure cultures, due to multiple advantages such as lower susceptibility to contamination, no need for sterilization of culture media or lower raw material costs. However, there are still challenges to overcome to increase the conversion efficiency, decrease formation of by-products and facilitate fermentation control. In this context, the purpose of the study was to develop a robust meta-fermentation process to efficiently produce lactic acid from the OFMSW, stable at pre-industrial scale (1500 L). To maximize lactic acid production, operating conditions (pH, HRT) were modified, and a novel bioaugmentation strategy was tested.
