Abstract:Glutathione S-transferase isoenzymes (GSTs) play a critical role in detoxification pathways. Here we report the tissue distribution of four antioxidant GSTs gene in common carp, and their expression profiles. We also investigated the GSTs activity in different tissues after exposure to the agricultural chemicals atrazine (ATR), chlorpyrifos (CPF), and their mixture. Relative changes in the mRNA abundance of the GST isoforms were examined by real time PCR in liver, brain, kidney and gill of common carp. After exposure and recovery, we observed a statistically significant decrease in the GSTs activity in animals exposed to high concentrations of ATR (428 μg/L), CPF (116 μg/L), and their mixture (113 μg/L). At basal levels of tissue expression, four GSTs transcript were detected in liver, brain, kidney, and gill. High expression levels were found in all examined tissues. Transcription of some GST isoforms, GST kappa (GSTK), GST theta (GSTT) and GST rho (GSTR), decreased after exposure to CPF and ATR for the entire experimental period in both the kidney and gill. However, increased transcription of GST mu (GSTM) was observed in the kidney or gill 20 d after exposure to ATR or CPF, respectively. Transcription of both GSTT and GSTR was inhibited for the entire experimental period in the brain, kidney and gill of animals exposed to the ATR/CPF mixture, but transcription of GSTM was induced in the liver after 40 d of exposure. In summary, changes in the GSTs activity and their transcription varied within each organ and among organs of common carp after exposure to ATR, CPF, and their mixture.
Graphical and Algebraic Synthesis for PWM Methods

Abstract:Abstract The article proposes an algebraic synthesis for PWM methods, which is based on a graphical representation of the 2 levels VSI (Voltage Source Inverter). This theory can be the starting point for every PWM method representation. In order to understand the historical evolution of the PWM methods the authors propose two classification groups of the main PWM techniques. The main classification is based on the movement of the neutral point (zero-voltage VN0 evolution). The details of PWM methods presented here are also seen from the graphical representation perspective. The equivalence between the well-known SVM (Space Vector Modulation) practically DDT realized (Direct Digital Technique) and the three-phase PWM with a carrier based industrial realization is shown. The principles of the three-phase PWM and the 3rd harmonic injection PWM (THIPWM4, THIPWM6) are compared the same as the DPWM (Discontinuous PWM) methods. These classifications and equivalences between different techniques show that the number of contemporary used PWM principles can be reduced. The general complex space representation and the classifications proposed here are simple to comprehension educational tools and they simply the way towards practical implementation.
Aflatoxin B1 decreased flesh flavor and inhibited muscle development in grass carp (Ctenopharyngodon idella)

Abstract:In nature, aflatoxins, especially aflatoxin B1 (AFB1), are the common mycotoxins, which cause serious health problems for humans and animals. This paper aimed to study the effects of AFB1 on flesh flavor and muscle development of grass carp (Ctenopharyngodon idella) and its mechanism. There were 1440 individual fish in total, with 6 treatments and each treatment replicated 3 times. The 6 treatments were fed a control diet with different doses of AFB1 (0.04, 29.48, 58.66, 85.94, 110.43 and 146.92 μg/kg diet) for 60 d. AFB1 increased myofiber diameter, as well as decreased myofiber density of grass carp muscle (P < 0.05). The contents of free amino acid decreased gradually (P < 0.05) as dietary AFB1 increased in the muscle of grass carp. The levels of reactive oxygen species, malonaldehyde and protein carbonyl (PC) were increased (P < 0.05) with the dietary AFB1 increased. The levels of antioxidant enzyme (glutathione peroxidase, glutathione, glutathione reductase, total antioxidant capacity, anti-superoxide anion, and anti-hydroxyl radical) were decreased (P < 0.05) with the dietary AFB1 increased. In addition, dietary AFB1 decreased the content of collagen, and downregulated the mRNA and protein levels of transforming growth factor-β (TGF-β)/Smads signaling pathway in grass carp muscle (P < 0.05). The mRNA and protein levels of myogenic regulatory factors were downregulated in grass carp muscle (P < 0.05). Furthermore, the activities of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) were increased (P < 0.05), and the protein levels of phosphorylate-38 mitogen-activated protein kinase (p-p38MAPK), phosphorylate-c-Jun N-terminal kinase, urokinase-type plasminogen activator (uPA), MMP-2 and MMP-9 were upregulated (P < 0.05), but collagen Ⅰ, laminin β1 and fibronectin were downregulated (P < 0.05) with the dietary AFB1 increased in the muscle of grass carp. Based on the results of this study, we can draw the following conclusion: dietary AFB1 might damage flesh flavor and inhibit the muscle development through MAPK/uPA/MMP/extracellular matrix (ECM) signaling pathway in grass carp. Moreover, the recommended safe limit of AFB1 in feed is no more than 26.77 μg/kg diet according to the PC levels in grass carp muscle.
AFB1 Induced Transcriptional Regulation Related to Apoptosis and Lipid Metabolism in Liver of Chicken

Abstract:Aflatoxin B1 (AFB1) leads to a major risk to poultry and its residues in meat products can also pose serious threat to human health. In this study, after feeding 165-day-old Roman laying hens for 35 days, the toxic effects of aflatoxin B1 at different concentrations were evaluated. The purpose of this study was to explore the mechanism of liver toxicosis responses to AFB1. We found that highly toxic group exposure resulted in liver fat deposition, increased interstitial space, and hepatocyte apoptosis in laying hens. Furthermore, a total of 164 differentially expressed lnRNAs and 186 differentially expressed genes were found to be highly correlated (Pearson Correlation Coefficient > 0.80, p-value < 0.05) by sequencing the transcriptome of control (CB) and highly toxic group (TB3) chickens. We also identify 29 differentially expressed genes and 19 miRNAs that have targeted regulatory relationships. Based on the liver cell apoptosis and fatty liver syndrome that this research focused on, we found that the highly toxic AFB1 led to dysregulation of the expression of PPARG and BCL6. They are cis-regulated by TU10057 and TU45776, respectively. PPARG was the target gene of gga-miR-301a-3p, gga-miR-301b-3p, and BCL6 was the target gene of gga-miR-190a-3p. In summary, highly toxic AFB1 affects the expression levels of protein-coding genes and miRNAs in the liver of Roman layer hens, as well as the expression level of long non-coding RNA in the liver, which upregulates the expression of PPARG and downregulates the expression of Bcl-6. Our study provides information on possible genetic regulatory networks in AFB1-induced hepatic fat deposition and hepatocyte apoptosis.

Abstract:Recently, residual organotin compounds have generally been recognised as relevant sources of aquatic environmental pollutants. However, the effects of these contaminants on the glutathione (GSH)-antioxidant system of fishes have not been adequately studied. In the current study, the chronic effects of tributyltin (TBT) found within antifouling paints for ships, on the GSH antioxidant system and related gene expression in the liver of juvenile common carp (Cyprinus carpio) were investigated. Fishes were exposed to sub-lethal concentrations of TBT (75 ng/L, 0.75 and 7.5 μg/L) for 15, 30 and 60 days. GSH levels and GSH-related enzymes activities, including glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione S-transferase (GST), were quantified in the fish liver. The levels of malondialdehyde were also measured as a marker of oxidative damage. In addition, the expression levels of gstp1, gr and gpx1 in common carp chronically exposed to TBT were determined. The results of the current study indicate that chronic exposure of TBT results in reactive oxygen species stress in the liver of common carp, and mRNA expression levels are more sensitive than related enzyme levels. In short, the measured GSH-related indices could potentially be used as molecular indicators for monitoring organotin compounds in the aquatic environment.

Abstract:Aquafeeds are susceptible to contamination caused by aflatoxin B1 (AFB1). The gill of fish is an important respiratory organ. However, few studies have investigated the effects of dietary AFB1 exposure on gill. This study aimed to discuss the effects of AFB1 on the structural and immune barrier of grass carp gill. Dietary AFB1 increased reactive oxygen species (ROS) levels, protein carbonyl (PC) and malondialdehyde (MDA) contents, which consequently caused oxidative damage. In contrast, dietary AFB1 decreased antioxidant enzymes activities, relative genes expression (except MnSOD ) and the contents of glutathione (GSH) ( P < 0.05), which are partly regulated by NF-E2-related factor 2 (Nrf2/Keap1a). Moreover, dietary AFB1 caused DNA fragmentation. The relative genes of apoptosis (except Bcl-2, McL-1 and IAP ) were significantly upregulated ( P < 0.05), and apoptosis was likely upregulated through p38 mitogen-activated protein kinase (p38MAPK). The relative expressions of genes associated with tight junction complexes (TJs) (except ZO-1 and claudin-12 ) were significantly decreased ( P < 0.05), and TJs were likely regulated by myosin light chain kinase (MLCK). Overall, dietary AFB1 disrupted the structural barrier of gill. Furthermore, AFB1 increased gill sensitivity to F. columnare, increased Columnaris disease and decreased the production of antimicrobial substances ( P < 0.05) in grass carp gill, and upregulated the expression of genes involved with pro-inflammatory factors (except TNF-α and IL-8 ) and the pro-inflammatory response partly attributed to the regulation by nuclear factor κB (NF-κB), meanwhile, the anti-inflammatory factors were downregulated ( P < 0.05) in grass carp gill under the challenged with F. columnare , which was partly attributed to the target of rapamycin (TOR). These results suggested that AFB1 aggravated the disruption of the immune barrier of grass carp gill after being challenge with F. columnare . Finally, the upper limit of safety of AFB1 for grass carp, based on Columnaris disease, was 31.10 μg/kg diet.

Abstract:hile hereditary (familial) amyloidosis has probably been around for as long as recorded hisW tory, it has only been recognized as a distinct entity in the latter half of the 20h Century. The first documentation of transthyretin amyloidosis as an autosomal dominant inherited disease was in 1952 by Corino Andrade' . This disease which was named familial amyloidotic polyneuropathy (FAP) because of the predominance of peripheral neuropathy in affected individuals of Portuguese origin has grown in importance as the number of mutations in transthyretin which are associated with pathology (presently over 70) and the sytemic nature of the disease have become appreciated*. It is truly the prototype of the hereditary systemic amyloidoses and displays all of the characteristics of this group of fibril protein deposition diseases. 1) Amyloid affects many organ systems with variation fiom kindred to kindred and individual to individual. 2) The disease is adultonset; usually starting after the childbearing years, as would be expected for a fatal hereditary condition. 3) The disease is dominantly inherited, a common feature of diseases due to mutations in genes for structural proteins. 4)Anumber of mutations in the same protein may govern expression of the genetic disease. 5 ) The occurrence of amyloid deposition in old age in the absence of any genetic mutation. Transthyretin amyloidosis is only one of several forms of systemic amyloidosis that have been characterized in the last 20 years. Mutations in the gene for apolipoprotein AI cause a number of syndromes of systemic amyloidosis with restrictive cardiomyopathy the most prominent clinical feature, but with cutaneous and renal amyloidosis in some kindreds. As with transthyretin, a number of disease causing mutations of apoA 1 have been described; at least eight within the last ten years. Four mutations in the gene for fibrinogen Aa-chain are associated with renal amyloidosis. Two mutations in lysozyme have been associated with either renal or hepatic amyloidosis. Two mutations in gelsolin and one in cystatin C can cause systemic amyloid deposition. In the case of transthyretin and apolipoprotein AI, it is now known that normal protein can be associated with amyloid deposits; transthyretin in the heart, apoAI in the aorta. Whether amyloid deposits can be derived fiom normal fibrinogen Aa-chain, lysozyme, gelsolin or cystatin C has yet to be explored. Besides the hereditary systemic amyloid syndromes, several inherited forms of localized amyloidosis have been described. The most common is Alzheimer disease due to mutations in APPP, presenilin1 or presenilin-2. GertstmannStraussler-Scheinker disease and familial Cruetzfeld-Jacob diseases are due to mutations in the pnon protein. As with the systemic amyloidoses, several mutations have been found to cause the localized forms and, at least with Alzheimer disease, senile amyloid deposits fiom normal ApPP occur without obvious genetic basis. Despite the amazing amount of knowledge gained in the last two decades about these diseases, we still have little in the way of specific therapy to offer individuals and families affected with hereditary amyloidoses. We can offer DNA testing, genetic counseling and the reassurance that science is progressing at an incredible rate.
THE EFFECT OF SHORT-TERM EXPOSURE TO LOW pH/ALUMINUM ON THE ION REGULATION, RELATED ATPase ACTIVITIES AND HAEMATOLOGICAL PARAMETERS OF JUVENILE COMMON CARP

Abstract:Aquatic acidification has been becoming a major issue in recent years. Concomitant with aquatic acidification, Aluminum (Al) toxicity also pose threat to the fish. The purpose of the present study was to assess the combined effect of aquatic acidification and Al on ion concentrations in plasma, ATPase activities in liver and kidney, and health status of fish. Juvenile common carp were exposed to solutions with different combined concentrations of pH (7, 5) and Al (0, 800 mu g L-1) for 1, 3, 9 days. We found that 9 days exposure to Al increased the levels of Na+ and Cl-. The K+ level significantly increased in low pH and low pH/Al treatments after exposure for 1 and 9 days. Furthermore, the activities of Na+/K+-ATPase and Ca2+-ATPase in liver were significantly decreased after exposure for 3 and 9 days, which indicated that the liver played an important role in coping with the stress. Besides, the increase of RBC and decrease of WBC also reconfirmed the toxic effect of low pH and Al on common carp. These results provide some evidence on understanding the impact of low pH and Al on the health status of fish.

Abstract:Aflatoxin B1 (AFB1) is kind of a common mycotoxin in food and feedstuff. Aquafeeds are susceptible to contamination of AFB1. In teleost fish, the spleen and head kidney are key immune organ. Moreover, the fish skin is a critical mucosal barrier system. However, there was little study on the effects of dietary AFB1 on the immune response of these immune organs in fish. This study aimed to explore the impacts of oral AFB1 on the immune competence and its mechanisms in the skin, spleen, and head kidney of grass carp. Our work indicated that dietary AFB1 reduced antibacterial compounds and immunoglobulins contents, and decreased the transcription levels of antimicrobial peptides in grass carp immune organs. In addition, dietary AFB1 increased the transcription levels of pro-inflammatory cytokines and reduced the transcription levels of anti-inflammatory cytokines in the grass carp immune organs, which might be regulated by NF-κB and TOR signaling, respectively. Meanwhile, we evaluated the content of AFB1 in the grass carp diet should not exceed 29.48 μg/kg diet according to the levels of acid phosphatase and lysozyme. In summary, dietary AFB1 impaired immune response in grass carp skin, spleen, and head kidney.

Species-specific differences in DNA damage sensitivity at early developmental stage: A comparative study of sterlet (Acipenser ruthenus) and common carp (Cyprinus carpio)

Abstract:DNA damage in embryos shapes the development of an organism. Understanding life stage-specific differences between fish species is essential for ecological risk assessment measures. We explored DNA damage sensitivity in two nonmodel fish species, sterlet (Acipenser ruthenus) and common carp (Cyprinus carpio). Embryos of these species were exposed to a model genotoxicant, camptothecin (CPT), during cleavage (2-cell) stage and gastrulation. Results revealed a species-specific DNA damage sensitivity only at cleavage stage. 3 nM CPT caused lethality in sterlet embryos while carp embryos hatched normally. Multiple nuclear abnormalities were observed in sterlet embryos by early gastrula stage. However, carp embryos exhibited nuclear abnormalities and DNA fragmentation at neurula stage only when exposed to 7 nM CPT. Moreover, increased expression of tp53 in carp embryos at gastrula stage suggests activation of apoptosis mechanism. These findings suggest that carp embryos activate DNA damage response more efficiently than sterlet embryos at same developmental stage.

Abstract:Aflatoxin B1 (AFB1) is a ubiquitous mycotoxin that causes oxidative damage in various organs. At present, the research studies on AFB1 are primarily focused on its effects on the terrestrial environment and animals. However, its toxicity mechanism in aquatic environments and aquatic animals has not been largely explored. Thus, in this study, zebrafish was used as a model to study the toxicity mechanism of AFB1 on the liver of developing larvae. The results showed that AFB1 exposure inhibited liver development and promoted fat accumulation in the liver. Transcriptome sequencing analysis showed that AFB1 affected liver redox metabolism and oxidoreductase activity. KEGG analysis showed that AFB1 inhibited the expression of gsto1 , gpx4a , mgst3a , and idh1 in the glutathione metabolizing enzyme gene pathway, resulting in hepatic oxidative stress. At the same time, AFB1 also inhibited the expression of acox1 , acsl1b , pparα , fabp2, and cpt1 genes in peroxidase and PPAR metabolic pathways, inducing hepatic steatosis and lipid droplet accumulation. Antioxidant N-Acetyl- l -cysteine (NAC) preconditioning up-regulated gsto1 , gpx4a and idh1 genes, and improved the AFB1-induced lipid droplet accumulation in the liver. In summary, AFB1 induced hepatic oxidative stress and steatosis, resulting in abnormal liver fat metabolism and accumulation of cellular lipid droplets. NAC could be used as a potential preventative drug to improve AFB1-induced fat accumulation.

Abstract:Ochratoxin A (OTA) is a common hazardous food contaminant that seriously endangers human and animal health. However, limited study is focused on aquatic animal. This research investigated the multiple biotoxicity of OTA on spleen (SP) and head kidney (HK) in grass carp and its related mechanism. Our data showed that, dietary supplemented with OTA above 1209 μg/kg caused histopathological damages by decreasing the number of lymphocytes and necrotizing renal parenchymal cells. Meanwhile, OTA caused oxidative damage and reduced the isoforms mRNAs transcripts of antioxidant enzymes (e.g., GPX1, GPX4, GSTO) partly due to suppressing NF-E2-related factor 2 (Nrf2). OTA triggered apoptosis through mitochondria and death receptor pathway potentially by p38 mitogen-activated protein kinase (p38MAPK) activation. Besides, OTA exacerbated inflammation by down-regulation of anti-inflammatory factor (e.g., IL-10, IL-4) and up-regulations of pro-inflammatory factors (e.g., TNF-α, IL-6), which could be ascribed to signaling meditation of Janus kinase / signal transducer and activator of transcription (JAK/STAT). Additionally, the safe upper limits of OTA were estimated to be 677.6 and 695.08 μg/kg based on the immune-related indexes (C3 contents in the SP and LZ activities in the HK, respectively). Our study has provided a wide insight for toxicological assessment of feed pollutant in aquatic animals.

Abstract:Aflatoxin B1 (AFB1) not only causes significant losses in livestock production but also poses a serious threat to human health. It is the most carcinogenic among known chemicals. Pigs are more susceptible to AFB1 and experience a higher incidence. However, the molecular mechanism of the toxic effect of AFB1 remains unclear. In this study, we used assay for transposase-accessible chromatin using sequencing (ATAC-seq) and RNA-seq to uncover chromatin accessibility and gene expression dynamics in PK-15 cells during early exposure to AFB1. We observed that the toxic effects of AFB1 involve signaling pathways such as p53, PI3K-AKT, Hippo, MAPK, TLRs, apoptosis, autophagy, and cancer pathways. Basic leucine zipper (bZIP) transcription factors (TFs), including AP-1, Fos, JunB, and Fra2, play a crucial role in regulating the biological processes involved in AFB1 challenge. Several new TFs, such as BORIS, HNF1b, Atf1, and KNRNPH2, represent potential targets for the toxic mechanism of AFB1. In addition, it is crucial to focus on the concentration of intracellular zinc ions. These findings will contribute to a better understanding of the mechanisms underlying AFB1-induced nephrotoxicity and offer new molecular targets.
Effect of aflatoxin B1, benzo[a]pyrene, and methapyrilene on transcriptomic and epigenetic alterations in human liver HepaRG cells

Abstract:The increasing number of man-made chemicals in the environment that may pose a carcinogenic risk highlights the need for developing reliable time- and cost-effective approaches for carcinogen detection and identification. To address this issue, we investigated the utility of high-throughput microarray gene expression and next-generation genome-wide DNA methylation sequencing for the in vitro identification of genotoxic and non-genotoxic carcinogens. Terminally differentiated and metabolically competent human liver HepaRG cells were treated at minimally cytotoxic concentrations of (i) the genotoxic human liver carcinogen aflatoxin B1 (AFB1) and its structural non-carcinogenic analog aflatoxin B2 (AFB2); (ii) the genotoxic human lung carcinogen benzo[a]pyrene (B[a]P) and its non-carcinogenic isomer benzo[e]pyrene (B[e]P); and (iii) the non-genotoxic liver carcinogen methapyrilene for 72 h and transcriptomic and DNA methylation profiles were examined. Treatment of HepaRG cells with the liver carcinogens AFB1 and methapyrilene generated distinct gene-expression profiles, whereas B[a]P had only a slight effect on gene expression. In contrast to transcriptomic alterations, treatment of HepaRG cells with the carcinogenic and non-carcinogenic chemicals resulted in profound changes in the DNA methylation footprint; however, the correlation between gene-specific DNA methylation and gene expression changes was minimal. Among the carcinogen-altered genes, transferrin (TF) emerged as sensitive marker for an initial screening of chemicals for their potential liver carcinogenicity. Potential liver carcinogens (i.e., chemicals causing altered TF gene expression) could then be subjected to gene-expression analyses to differentiate genotoxic from non-genotoxic liver carcinogens. This approach may substantially enhance the identification and assessment of potential liver carcinogens.
Dietary additive ferulic acid alleviated oxidative stress, inflammation, and apoptosis induced by chronic exposure to avermectin in the liver of common carp (Cyprinus carpio)

Abstract:Avermectin (AVM) has been utilized extensively in agricultural production since it is a low-toxicity pesticide. However, the pollution caused by its residues to fisheries aquaculture has been neglected. As an abundant polyphenolic substance in plants, ferulic acid (FA) possesses anti-inflammatory and antioxidant effects. The goal of the study is to assess the FA's ability to reduce liver damage in carp brought on by AVM exposure. Four groups of carp were created at random: the control group; the AVM group; the FA group; and the FA + AVM group. On day 30, and the liver tissues of carp were collected and examined for the detection of four items of blood lipid as well as the activity of the antioxidant enzymes catalase (CAT), glutathione (GSH) and malondialdehyde (MDA) in carp liver tissues by biochemical kits, and the transcript levels of indicators of oxidative stress, inflammation and apoptosis by qPCR. The results showed that liver injury, inflammation, oxidative stress, and apoptosis were attenuated in the FA+AVM group compared to the AVM group. In summary, dietary addition of FA could ameliorate the hepatotoxicity caused by AVM in carp by alleviating oxidative stress, inflammation, apoptosis in liver tissues.

The effect of aflatoxin B1 on the expression of early response genes and transforming growth factor-alpha in CCl4 induced rat liver injury

Abstract:Aflatoxin B1 (AFB1), a fungal toxin produced by Aspergillus flavus, is known to be a possible hepatocarcinogen. But the molecular biologic changes which may occur following exposure to AFB1 are not known and thus the carcinogenesis is not yet understood. This study was performed to examine the expressions of c-myc, c-fos and TGF-alpha genes and to investigate the possible role of those molecular biologic changes in hepatic regeneration and in the development of hepatocellular carcinoma (HCC). Sprague-Dawley rats were divided into 3 groups: Carbon tetrachloride (CCl4) only was administered to group I, AFB1 only was administered to group II and a combination of AFB1 and CCl4 was administered to group III. The animals were sacrificed at 0.5, 1, 2, 6, 12, 24, 48, and 72 hours after treatment. In addition to the examination of the hematoxylin-eosin stained sections, hepatic regeneration and apoptosis were analyzed quantitatively by bromodeoxyuridine (BrdU)-anti-BrdU immunohistochemistry and TUNEL assay utilizing apoptosis kit, respectively. The hepatic expressions of c-myc, c-fos and transforming growth factor-alpha (TGF-alpha) were examined by immunohistochemistry and studied by Western blot. The number of BrdU labelled cells and the degree of necrosis/apoptosis were comparable among the different groups. Livers of the group II rats showed nearly normal histology without regeneration and necrosis/apoptosis. In groups I and III, the number of BrdU- labelled cells showed an increase at 48 hours after treatment, and the increment was significantly higher in group I than in group III. Most BrdU-labelled cells were mature hepatocytes in group I, whereas in group III they appeared to be less mature. In group I, apoptosis showed an increase at around 24 hours, but appeared in group III as early as 12 hours after treatment and persisted through 48 hours. The expression of c-myc and c-fos were also different between the experimental groups. The expression intensity of c-myc in group I was highest at 1 hour and decreased thereafter. In groups II and III, the expressions were much more intense than in group I, except at 1 hour, and the increased intensity persisted throughout the experiment. Group II in particular showed a peak intensity at 30 minutes and at 6 hours after treatment. In group I, c-fos was strongly expressed only at 24 hours, but in group III, there was progressively increased expression with peak intensity at 24 hours. TGF-alpha was expressed in similar intensities in all groups throughout the experiment. These results suggest that AFB1 may evoke an intense and protracted expression of c-myc, provocating the CCl4-induced necrosis of hepatocytes, and a prolonged expression of c-fos, including persistent signals for regeneration which in turn may activate the replication of immature cells. These findings will aid further investigation of molecular biologic and histologic characteristics of the hepatotoxic and hepatocarcinogenic mechanism of AFB1 in rats. And these results in rats, together with clinico-epidemiologic and molecular biologic investigations in humans and other animals, suggest that AFB1 may supply hepatocarcinogenic background in early exposure time in AFB1-contaminated areas of China and Korea.
Effect of aflatoxin B1 on nuclear receptors PXR, CAR, and AhR and their target cytochromes P450 mRNA expression in primary cultures of human hepatocytes

Abstract:Aflatoxin B1 (AFB1), one of the most common mycotoxins found in human foods and animal feed, is principally hepatotoxic and hepatocarcinogenic. The aim of the present study was to explore the effect of AFB1 on messenger RNA (mRNA) expression of pregnane X receptor (PXR), constitutive androstane receptor (CAR), and aryl hydrocarbon receptor (AhR) and some of their target cytochromes using primary cultures of human hepatocytes. Our results showed that AFB1, at noncytotoxic increasing concentrations, caused a significant upregulation of cytochrome P 2B6 (CYP2B6), CYP3A5, and to a lesser extent CYP3A4 and CYP2C9. Pregnane X receptor and CAR mRNA expression increased in the 3 treated livers. Aflatoxin B1 was found also to induce an overexpression of CYP1A1 and CYP1A2 genes accompanied by an increase in AhR mRNA expression. These findings suggest that AFB1 could activate PXR, CAR, and AhR; however, further investigations are needed to confirm nuclear receptor activation by AFB1.
Effects of Ginkgo Biloba Extract on Biomarker Expressions During Aflatoxin B1-induced Hepatocarcinogenesis in Wistar Rats.

Abstract:Objective:To study the effect of Ginkgo biloba extract(EGb761) on metabolism of aflatoxin B1(AFB1)in Wistar rats.Methods:Seventy one Wistar rats were divided into three groups at random: group A(AFB1 group),group B(AFB1+EGb761 group),and group C(control group).The rats in groups A and B were given AFB1(intraperitoneal injection,100-200 μg/ kg body weight,1-3 times/week).The rats in group B were fed the food containing EGb761 while the rats in groups A and C were given normal food.Blood samples were collected and liver biopsy was performed on the 14th,28th and 42nd week.All the rats were sacrificed at the 64th week.The incidence of hepatoma was observed.The hepatic phase Ⅰ drug-metabolizing enzyme CYP450 and phase Ⅱ enzyme GST were detected by spectrometry.The serum AFB1-lysine adduct was determined by high performance liquid chromatography(HPLC).The expression of 8-hydroxydeoxyguanosine(8-OHdG)was measured by immunohistochemistry.Results:The incidence of hepatocellular carcinoma(HCC) in group B was significantly lower than that in group A(26.92% vs 76.00%,P0.001).No hepatocellular carcinoma developed in group C.EGb761 had no effects on the activities of CYP450 and GST in rat liver tissues.The level of AFB1-lysine adduct reached the peak(4 356.01 pg/mg albumin) at the 14th week in group A.EGb761 significantly inhibited the formation of AFB1-lysine adducts in serum by 13.07% at the 14th week(P=0.033),and 73.63% at the 42nd week(P=0.002).The expression of 8-OHdG protein in rat liver tissues in group B was significantly lower than that in group A at the 28th,42nd,and 64th week(P0.05).Conclusion:The main mechanism underlying the effect of EGb761 in blocking hepatogenesis induced by AFB1 may not be fully related with its influence on the activity of liver phase Ⅰ and phase Ⅱ metabolizing enzymes.EGb761 inhibites the production of AFB1-lysine addcuts,decreases the expression of 8-OHdG protein,and finally alleviates the DNA oxidative injury,which may be one of the mechanisms for the effects of EGb761 in inhibiting or delaying hepatogenesis induced by AFB1.

Abstract:This study investigated the tissue-specific accumulation and biotransformation of 6:6 and 8:8 perfluoroalkyl phosphinic acids (PFPiA) in common carp (Cyprinus carpio) during 90 d exposure and 30 d depuration in water in the laboratory. Both 6:6 and 8:8 PFPiAs could quickly accumulate in the carp, and 6:6 PFPiA displayed higher bioaccumulation potential than 8:8 PFPiA. The highest concentrations of PFPiAs were observed in the blood, while the lowest were found in the muscle. The equilibrium dialysis experiment indicated that both PFPiAs had higher binding affinities with the proteins in the fish serum than in liver, which was supported by the molecular docking analysis. The results also indicated that 6:6 PFPiA had higher binding affinities with the serum and liver proteins than 8:8 PFPiA. These results suggested that the tissue-specific distribution of PFPiAs was highly dependent on the binding affinities with the specific proteins. Both in vivo and in vitro experiments consistently indicated that PFPiAs experienced biotransformation and produced perfluoroalkyl phosphonic acids (PFPAs), and biotransformation of 8:8 PFPiA was more active than 6:6 PFPiA. It was worth noting that perfluorohexanonate and perfluorooctanoic acids were identified in fish as metabolites after long-term exposure to PFPiAs for the first time.
